Objective. Enable neural control of individual prosthetic fingers for participants with upper-limb paralysis.Approach. Two tetraplegic participants were each implanted with a 96-channel array in the left posterior parietal cortex (PPC). One of the participants was additionally implanted with a 96-channel array near the hand knob of the left motor cortex (MC). Across tens of sessions, we recorded neural activity while the participants attempted to move individual fingers of the right hand. Offline, we classified attempted finger movements from neural firing rates using linear discriminant analysis with cross-validation. The participants then used the neural classifier online to control individual fingers of a brain-machine interface (BMI). Finally, we characterized the neural representational geometry during individual finger movements of both hands.Main Results. The two participants achieved 86% and 92% online accuracy during BMI control of the contralateral fingers (chance = 17%). Offline, a linear decoder achieved ten-finger decoding accuracies of 70% and 66% using respective PPC recordings and 75% using MC recordings (chance = 10%). In MC and in one PPC array, a factorized code linked corresponding finger movements of the contralateral and ipsilateral hands.Significance. This is the first study to decode both contralateral and ipsilateral finger movements from PPC. Online BMI control of contralateral fingers exceeded that of previous finger BMIs. PPC and MC signals can be used to control individual prosthetic fingers, which may contribute to a hand restoration strategy for people with tetraplegia.

IntroductionBi-directional brain-computer interfaces (BD-BCI) to restore movement and sensation must achieve concurrent operation of recording and decoding of motor commands from the brain and stimulating the brain with somatosensory feedback.MethodsA custom programmable direct cortical stimulator (DCS) capable of eliciting artificial sensorimotor response was integrated into an embedded BCI system to form a safe, independent, wireless, and battery powered testbed to explore BD-BCI concepts at a low cost. The BD-BCI stimulator output was tested in phantom brain tissue by assessing its ability to deliver electrical stimulation equivalent to an FDA-approved commercial electrical cortical stimulator. Subsequently, the stimulator was tested in an epilepsy patient with subcortical electrocorticographic (ECoG) implants covering the sensorimotor cortex to assess its ability to elicit equivalent responses as the FDA-approved counterpart. Additional safety features (impedance monitoring, artifact mitigation, and passive and active charge balancing mechanisms) were also implemeneted and tested in phantom brain tissue. Finally, concurrent operation with interleaved stimulation and BCI decoding was tested in a phantom brain as a proof-of-concept operation of BD-BCI system.ResultsThe benchtop prototype BD-BCI stimulator's basic output features (current amplitude, pulse frequency, pulse width, train duration) were validated by demonstrating the output-equivalency to an FDA-approved commercial cortical electrical stimulator (R 2 > 0.99). Charge-neutral stimulation was demonstrated with pulse-width modulation-based correction algorithm preventing steady state voltage deviation. Artifact mitigation achieved a 64.5% peak voltage reduction. Highly accurate impedance monitoring was achieved with R 2 > 0.99 between measured and actual impedance, which in-turn enabled accurate charge density monitoring. An online BCI decoding accuracy of 93.2% between instructional cues and decoded states was achieved while delivering interleaved stimulation. The brain stimulation mapping via ECoG grids in an epilepsy patient showed that the two stimulators elicit equivalent responses.SignificanceThis study demonstrates clinical validation of a fully-programmable electrical stimulator, integrated into an embedded BCI system. This low-cost BD-BCI system is safe and readily applicable as a testbed for BD-BCI research. In particular, it provides an all-inclusive hardware platform that approximates the limitations in a near-future implantable BD-BCI. This successful benchtop/human validation of the programmable electrical stimulator in a BD-BCI system is a critical milestone toward fully-implantable BD-BCI systems.

Leveraging firsthand experience, BRAIN-funded investigators conducting intracranial human neuroscience research propose two fundamental ethical commitments: (1) maintaining the integrity of clinical care and (2) ensuring voluntariness. Principles, practices, and uncertainties related to these commitments are offered for future investigation.

Bi-directional brain-computer interfaces (BD-BCI) to restore movement and sensation must achieve concurrent operation of recording and decoding of motor commands from the brain and stimulating the brain with somatosensory feedback. Previously we developed and validated a benchtop prototype of a fully implantable BCI system for motor decoding. Here, a prototype artificial sensory stimulator was integrated into the benchtop system to develop a prototype of a fully-implantable BD-BCI. The artificial sensory stimulator incorporates an active charge balancing mechanism based on pulse-width modulation to ensure safe stimulation for chronically interfaced electrodes to prevent damage to brain tissue and electrodes. The feasibility of the BD-BCI system's active charge balancing was tested in phantom brain tissue. With the charge-balancing, the removal of the residual charges on an electrode was evident. This is a critical milestone toward fully-implantable BD-BCI systems.

The goal of this study is to estimate the thermal impact of a titanium skull unit (SU) implanted on the exterior aspect of the human skull. We envision this unit to house the front-end of a fully implantable electrocorticogram (ECoG)-based bi-directional (BD) brain-computer interface (BCI). Starting from the bio-heat transfer equation with physiologically and anatomically constrained tissue parameters, we used the finite element method (FEM) implemented in COMSOL to build a computational model of the SU's thermal impact. Based on our simulations, we predicted that the SU could consume up to 75 mW of power without raising the temperature of surrounding tissues above the safe limits (increase in temperature of 1°C). This power budget by far exceeds the power consumption of our front-end prototypes, suggesting that this design can sustain the SU's ability to record ECoG signals and deliver cortical stimulation. These predictions will be used to further refine the existing SU design and inform the design of future SU prototypes.

Here, a prototype BMI system whose size and power consumption are comparable to those of fully implantable medical devices was designed and implemented, and its performance was tested at the benchtop and bedside.ApproachA prototype of a fully implantable BMI system was designed and implemented as a miniaturized embedded system. This benchtop analogue was tested in its ability to acquire signals, train a decoder, perform online decoding, wirelessly control external devices, and operate independently on battery. Furthermore, performance metrics such as power consumption were benchmarked.Main resultsAn analogue of a fully implantable BMI was fabricated with a miniaturized form factor. A patient undergoing epilepsy surgery evaluation with an electrocorticogram (ECoG) grid implanted over the primary motor cortex was recruited to operate the system. Seven online runs were performed with an average binary state decoding accuracy of 87.0% (lag optimized, or 85.0% at fixed latency). The system was powered by a wirelessly rechargeable battery, consumed  ∼150 mW, and operated for  >60 h on a single battery cycle.SignificanceThe BMI analogue achieved immediate and accurate decoding of ECoG signals underlying hand movements. A wirelessly rechargeable battery and other supporting functions allowed the system to function independently. In addition to the small footprint and acceptable power and heat dissipation, these results suggest that fully implantable BMI systems are feasible.

Fully-implantable, bi-directional brain-computer interfaces (BCIs) necessitate simultaneous cortical recording and stimulation. This is challenging since electrostimulation of cortical tissue typically causes strong artifacts that may saturate ultra-low power (ULP) analog front-ends of fully-implantable BCIs. To address this problem, we propose an efficient hardware-based method for artifact suppression that employs an auxiliary stimulator with polarity opposite to that of the primary stimulator. The feasibility of this method was explored first in simulations, and then experimentally with brain phantom tissue and electrocorticogram (ECoG) electrode grids. We find that the canceling stimulator can reduce stimulation artifacts below the saturation limit of a typical ULP front-end, while delivering only ~10% of the primary stimulator's voltage.

A new synthesis of cerium tetrakis(tropolonate), Ce(trop)4, where trop is deprotonated 2-hydroxy-2,4,6-cycloheptatrienone) or Ce(O2C7H5)4, is developed that results in dark-purple crystals whose X-ray crystal structure shows that the geometry of the eight-coordinate compound closely resembles a D2 d dodecahedron, based on shape parameters. The magnetic susceptibility as a function of the temperature (4-300 K) shows that it is a temperature-independent paramagnet, χ = 1.2(3) × 10-4 emu/mol, and the LIII-edge X-ray absorption near-edge structure spectrum shows that the molecule is multiconfigurational, comprised of a f1:f0 configuration mixture in a 50:50 ratio. Ce(acac)4 and Ce(tmtaa)2 (where acac is acetylacetonate and tmtaaH2 is tetramethyldibenzotetraaza[14]annulene) have similar physical properties, as does the solid-state compound CeO2. The concept is advanced that trop-, acac-, tmtaa2-, cot2-, and O2- are redox-active ligands that function as electron donors, rendering the classification of these compounds according to their oxidation numbers misleading because their magnetic susceptibilities, χ, are positive and their effective magnetic moments, μeff, lie in the range of 0.1-0.7 μB at 300 K.

Metal alkylidenes, which are key organometallic intermediates in reactions such as olefination or alkene and alkane metathesis, are typically generated from metal dialkyl compounds [M](CH2R)2 that show distinctively deshielded chemical shifts for their α-carbons. Experimental solid-state NMR measurements combined with DFT/ZORA calculations and a chemical shift tensor analysis reveal that this remarkable deshielding originates from an empty metal d-orbital oriented in the M-Cα-Cα' plane, interacting with the Cα p-orbital lying in the same plane. This π-type interaction inscribes some alkylidene character into Cα that favors alkylidene generation via α-H abstraction. The extent of the deshielding and the anisotropy of the alkyl chemical shift tensors distinguishes [M](CH2R)2 compounds that form alkylidenes from those that do not, relating the reactivity to molecular orbitals of the respective molecules. The α-carbon chemical shifts and tensor orientations thus predict the reactivity of metal alkyl compounds towards alkylidene generation.

Scandium and yttrium congeneric complexes, supported by a monoanionic PNP ligand, were studied as catalysts for alkene hydrogenation and hydrosilation, and alkyne semihydrogenation and semihydrosilation. The yttrium congener was found to be much more active in all cases, but this greater activity is accompanied by more rapid catalyst decomposition and therefore higher total yields for some of the reactions with the scandium catalyst. Calculations indicate that the reactions may proceed via σ-bond metathesis of the alkyl complexes to form metal hydride intermediates into which alkenes/alkynes insert.

Metallacyclobutanes are an important class of organometallic intermediates, due to their role in olefin metathesis. They can have either planar or puckered rings associated with characteristic chemical and physical properties. Metathesis active metallacyclobutanes have short M-Cα/α' and M···Cβ distances, long Cα/α'-Cβ bond length, and isotropic 13C chemical shifts for both early d0 and late d4 transition metal compounds for the α- and β-carbons appearing at ca. 100 and 0 ppm, respectively. Metallacyclobutanes that do not show metathesis activity have 13C chemical shifts of the α- and β-carbons at typically 40 and 30 ppm, respectively, for d0 systems, with upfield shifts to ca. -30 ppm for the α-carbon of metallacycles with higher d n electron counts (n = 2 and 6). Measurements of the chemical shift tensor by solid-state NMR combined with an orbital (natural chemical shift, NCS) analysis of its principal components (δ11 ≥ δ22 ≥ δ33) with two-component calculations show that the specific chemical shift of metathesis active metallacyclobutanes originates from a low-lying empty orbital lying in the plane of the metallacyclobutane with local π*(M-Cα/α') character. Thus, in the metathesis active metallacyclobutanes, the α-carbons retain some residual alkylidene character, while their β-carbon is shielded, especially in the direction perpendicular to the ring. Overall, the chemical shift tensors directly provide information on the predictive value about the ability of metallacyclobutanes to be olefin metathesis intermediates.

Lu[CH(SiMe3)2]3 reacts with [SiO2-700] to give [(≡SiO)Lu[CH(SiMe3)2]2] and CH2(SiMe3)2. [(≡SiO)Lu[CH(SiMe3)2]2] is characterized by solid-state NMR and EXAFS spectroscopy, which show that secondary Lu···C and Lu···O interactions, involving a γ-CH3 and a siloxane bridge, are present. From X-ray crystallographic analysis, the molecular analogues Lu[CH(SiMe3)2]3-x[O-2,6-tBu-C6H3]x (x = 0-2) also have secondary Lu···C interactions. The (1)H NMR spectrum of Lu[CH(SiMe3)2]3 shows that the -SiMe3 groups are equivalent to -125 °C and inequivalent below that temperature, ΔG(⧧)(Tc = 148 K) = 7.1 kcal mol(-1). Both -SiMe3 groups in Lu[CH(SiMe3)2]3 have (1)JCH = 117 ± 1 Hz at -140 °C. The solid-state (13)C CPMAS NMR spectrum at 20 °C shows three chemically inequivalent resonances in the area ratio of 4:1:1 (12:3:3); the J-resolved spectra for each resonance give (1)JCH = 117 ± 2 Hz. The (29)Si CPMAS NMR spectrum shows two chemically inequivalent resonances with different values of chemical shift anisotropy. Similar observations are obtained for Lu[CH(SiMe3)2]3-x[O-2,6-tBu-C6H3]x (x = 1 and 2). The spectroscopic data point to short Lu···Cγ contacts corresponding to 3c-2e Lu···Cγ-Siβ interactions, which are supported by DFT calculations. Calculated natural bond orbital (NBO) charges show that Cγ carries a negative charge, while Lu, Hγ, and Siβ carry positive charges; as the number of O-based ligands increases so does the positive charge at Lu, which in turns shortens the Lu···Cγ distance. The change in NBO charges and the resulting changes in the spectroscopic and crystallographic properties show how ligands and surface-support sites rearrange to accommodate these changes, consistent with Pauling's electroneutrality concept.

Bright nano objects emitting in the near infrared with a maximal cross section of 41.4 x 103 GM (Goppert Mayer), were prepared by implanting ca. 180 4,4 diethylaminostyryl 2,2 bipyridine (DEAS) Yb(III) complexes on the surface of 12 nm silica nanoparticles. The surface complexes ([DEAS Ln SiO2], Ln =Y,Yb) were characterized using IR, solid state NMR, UV Vis, EXAFS spectroscopies in combination with the preparation and characterization of similar molecular analogues by analytical techniques (IR, solution NMR, UV Vis, X ray crystallography) as well as DFT calculations. Starting from the partial dehydroxylation of the silica at 700 C on high vacuum having 0.8 OH.nm 2, the grafting of Ln(N(SiMe3)2)3 generate ≤SiO Ln(N(SiMe3)2)2, which upon thermal step and coordination of the DEAS chromophore yields (≤SiO)3Ln(DEAS). Surface and molecular analogues display similar properties, in terms of DEAS binding constants absorption maxima and luminescence properties (intense emission band assigned to a ligand centered CT fluorescence and life time) in the solid state, consistent with the molecular nature of the surface species. The densely functionalized nanoparticles can be dispersed via ultra-sonication in small ca. 15-20 nm aggregates (1 to 6 elementary particles) that were detected using two photon microscopy imaging at 720 nm excitation, making them promising nano objects for bio imaging.

Characterization of paramagnetic compounds, in particular regarding the detailed conformation and electronic structure, remains a challenge, and -- still today it often relies solely on the use of X-ray crystallography, thus limiting the access to electronic structure information. This is particularly true for lanthanide elements that are often associated with peculiar structural and electronic features in relation to their partially filled f-shell. Here, we develop a methodology based on the combined use of state-of-theart magnetic resonance spectroscopies (EPR and solid-state NMR) and computational approaches as well as magnetic susceptibility measurements to determine the electronic structure and geometry of a paramagnetic Yb(III) alkyl complex, Yb(III)[CH(SiMe3)2]3, a prototypical example, which contains notable structural features according to X-ray crystallography. Each of these techniques revealed specific information about the geometry and electronic structure of the complex. Taken together, both EPR and NMR, augmented by quantum chemical calculations, provide a detailed and complementary understanding of such paramagnetic compounds. In particular, the EPR and NMR signatures point to the presence of three-centre--two-electron Yb-γ-Me-β-Si secondary metal--ligand interactions in this otherwise tri-coordinate metal complex, similarly to its diamagnetic Lu analogues. The electronic structure of Yb(III) can be described as a single 4f13 configuration, while an unusually large crystal-field splitting results in a thermally isolated ground Kramers doublet. Furthermore, the computational data indicate that the Yb--carbon bond contains some p-character, reminiscent of the so-called a-H agostic interaction. [ABSTRACT FROM AUTHOR]

Addition of CO to [1,2,4-(Me3C)3C5H2]2CeH, Cp'2CeH, in toluene yields the cis (Cp'2Ce)2(mu-OCHCHO), in which the cis enediolate group bridges the two metallocene fragments. The cis enediolate quantitatively isomerizes intramolecularly to the trans-enediolate in C6D6 at 100oC over seven months. When the solvent is pentane, Cp'2Ce(OCH2)CeCp'2 forms, in which the oxomethylene group or the formaldehyde dianion bridges the two metallocene fragments. The cis enediolate is suggested to form by insertion of CO into the Ce-C bond of Cp'2Ce(OCH2)CeCp'2 generating Cp'2CeOCH2COCeCp'2. The stereochemistry of the cis-enediolate is determined by a 1,2-hydrogen shift in the OCH2CO fragment that has the OC(H2) bond anti periplanar relative to the carbene lone pair. The bridging oxomethylene complex reacts with H2, but not with CH4, to give Cp'2CeOMe, which is also the product of the reaction between Cp'2CeH and a mixture of CO and H2. The oxomethylene complex reacts with CO to give the cis enediolate complex. DFT calculations on C5H5 model metallocenes show that the reaction of Cp2CeH with CO and H2 to give Cp2CeOMe is exoergic by 50 kcal mol-1. The net reaction proceeds by a series of elementary reactions that occur after the formyl complex, Cp2Ce(eta-2 CHO), is formed by further reaction with H2. The key point that emerges from the calculated potential energy surface is the bifunctional nature of the metal formyl in which the carbon atom behaves as a donor and acceptor. Replacing H2 by CH4 increases the activation energy barrier by 17 kcal mol-1.

The complexes [Ce(tmtaa)2], [Ce(tmtaa)(tmtaaH)] and [Ce2(tmtaa)3(thf)2] are obtained from Ce[N(SiMe3)2]3 and tmtaaH2, the macrocyclic ligand 6,8,15,17-tetramethyldibenzotetraaza[14]-annulene, depending on the stoichiometry, solvent and temperature. The crystal structure of Ce(tmtaa)2 is isostructural with Zr(tmtaa)2, however magnetic susceptibility measurements in the range 5-300 K show that Ce(tmtaa)2 is not diamagnetic, but is a temperature-independent paramagnet (TIP), similar to Ce(cot)2, cerocene.

The paramagnetic 1:1 coordination complexes of (C5Me5)2Yb with a series of 4,4'-disubstituted bipyridines, bipy-X, where X is Me, tert-Bu, OMe, Ph, CO2Me, and CO2Et have been prepared. All of the complexes are paramagnetic and the values of the magnetic susceptibility as a function of temperature show that these values are less than expected for the cation, [(C5Me5)2Yb(III)(bipy-X)]+, which have been isolated as the cation-anion ion-pairs [(C5Me5)2Yb(III)(bipy-X)]+[(C5Me5)2YbI2]f fnfn where X is CO2Et, OMe and Me. The 1H NMR chemical shifts (293 K) for the methine resonances located at the 6,6' site in the bipy-X ring show a linear relationship with the values of chiT (300 K) for the neutral complexes which illustrates that the molecular behavior does not depend upon the phase with one exception, viz., (C5Me5)2Yb(bipy-Me). Single crystals of the 4,4'-dimethylbipyridine complex undergo an irreversible, abrupt first order phase change at 228 K that shatters the single crystals. The magnetic susceptibility, represented in a delta vs. T plot, on this complex, in polycrystalline form undergoes reversible abrupt changes in the temperature regime 205 - 212 K, which is suggested to be due to the way the individual molec ular units pack in the unit cell. A qualitative model is proposed that accounts for the sub-normal magnetic moments in these ytterbocene-bipyridine complexes.

The uranium metallocenes, [eta5-1,3-(Me3E)2C5H3]2UMe2 (E = C, Si), react with NH3 to give the dimers {[eta5-1,3-(Me3E)2C5H3]2U}2(mu-NH)2 (E = C (1), Si (2)) but with p-toluidine to give the monomeric diamides, [eta5-1,3-(Me3E)2C5H3]2U(NH-p-tolyl)2 (E = C (3), Si (4)). The diamides [eta5-1,3-(Me3E)2C5H3]2U(NH-p-tolyl)2 (E = C (3), Si (4)) do not eliminate p- toluidine but sublime intact at 140oC in vacuum. The uranium metallocene, [eta5-1,2,4-(Me3C)3C5H2]2UMe2, reacts with RNH2 to yield [eta5-1,2,4-(Me3C)3C5H2]2U(NHR)2 (R = Me (8), PhCH2 (9), p-tolyl (10)), which are isolated as crystalline solids. In benzene solution these diamides are in equilibrium with [eta5-1,2,4-(Me3C)3C5H2]2U=NR, which may be isolated pure when R is Me (11) or p-tolyl (12), and the primary amine. The monomeric imide, [eta5-1,2,4-(Me3C)3C5H2]2U=NMe (11), reacts with R'=CR' to yield the cycloaddition products [eta5-1,2,4-(Me3C)3C5H2]2U[N(Me)C(R')=C(R')] (R' = Me (15), Ph (16)), which react with excess MeNH2 to regenerate the diamide [eta5-1,2,4-(Me3C)3C5H2]2U(NHMe)2 (8) and MeN=C(R')CH(R'). The methylimide, [eta5-1,2,4-(Me3C)3C5H2]2U=NMe (11), does not react with Me3SiX reagents; a model is proposed that rationalizes this reactivity pattern.

Reduction of the uranium metallocene, [eta5-1,2,4-(Me3C)3C5H2]2UCl2 (1), Cp'2UCl2, in the presence of 2,2'-bipyridyl and sodium naphthalene gives the dark green metallocene complex, Cp'2U(bipy) (6), which reacts with p-tolylazide or pyridine-N-oxide to give Cp'2U=N(p-tolyl) (7) or Cp'2U(O)(py) (8), respectively. The Lewis acid, BPh3, precipitates Ph3B(py) and gives the base-free oxo, Cp'2UO (10), which crystallizes from pentane. The oxometallocene 10 behaves as a nucleophile with Me3SiX reagents but it does not exhibit cycloaddition behavior with acetylenes, suggesting that the polar resonance structure, Cp'2U+-O- dominates the double bond resonance structure Cp'2U=O.
