We present a detailed discussion of how to obtain precise stellar photometry in crowded fields using images from multi-conjugate adaptive optics (MCAO) systems, with the intent of informing the scientific development of this key technology for the Extremely Large Telescopes. We use deep J and K_s exposures of NGC 1851 taken with the Gemini Multi-Conjugate Adaptive Optics System (GeMS) on Gemini South to quantify the performance of the instrument and to develop an optimal strategy for stellar photometry using PSF-fitting techniques. We judge the success of the various methods we employ by using science-based metrics, particularly the width of the main sequence turn-off region. We also compare the GeMS photometry with the exquisite HST data in the visible of the same target. We show that the PSF produced by GeMS possesses significant spatial and temporal variability that must be accounted for during the analysis. We show that the majority of the variation of the PSF occurs within the "control radius" of the MCAO system and that the best photometry is obtained when the PSF radius is chosen to closely match this spatial scale. We identify photometric calibration as a critical issue for next generation MCAO systems such as those on TMT and E-ELT. Our final CMDs reach K_s~22---below the main sequence knee---making it one of the deepest for a globular cluster available from the ground. Theoretical isochrones are in remarkable agreement with the stellar locus in our data from below the main sequence knee to the upper red giant branch., Comment: Accepted for publication in AJ

We discuss images of the star clusters GLIMPSE C01 (GC01) and GLIMPSE C02 (GC02) that were recorded with the Subaru IRCS. Distortions in the wavefront were corrected with the RAVEN adaptive optics (AO) science demonstrator, allowing individual stars in the central regions of both clusters -- where the fractional contamination from non-cluster objects is lowest -- to be imaged. In addition to J, H, and K' images, both clusters were observed through a narrow-band filter centered near 3.05um; GC01 was also observed through two other narrow-band filters that sample longer wavelengths. Stars in the narrow-band images have a FWHM that is close to the telescope diffraction limit, demonstrating that open loop AO systems like RAVEN can deliver exceptional image quality. The near-infrared color magnitude diagram of GC01 is smeared by non-uniform extinction with a dispersion +/- 0.13 magnitudes in A_K. The Red Clump is identified in the K luminosity function (LF) of GC01, and a distance modulus of 13.6 is found. The K LF of GC01 is consistent with a system that is dominated by stars with an age > 1 Gyr. As for GC02, the K LF is flat for K > 16, and the absence of a sub-giant branch argues against an old age if the cluster is at a distance of ~ 7 kpc. Archival SPITZER [3.6] and [4.5] images of the clusters are also examined, and the red giant branch-tip is identified., Comment: To appear in the Astronomical Journal

Aims: for the first time the astrometric capabilities of the Multi-Conjugate Adaptive Optics (MCAO) facility GeMS with the GSAOI camera on Gemini-South are tested to quantify the accuracy in determining stellar proper motions in the Galactic globular cluster NGC 6681. Methods: proper motions from HST/ACS for a sample of its stars are already available, and this allows us to construct a distortion-free reference at the epoch of GeMS observations that is used to measure and correct the temporally changing distortions for each GeMS exposure. In this way, we are able to compare the corrected GeMS images with a first-epoch of HST/ACS images to recover the relative proper motion of the Sagittarius dwarf spheroidal galaxy with respect to NGC 6681. Results: we find this to be (\mu_{\alpha}cos\delta, \mu_{\delta}) = (4.09,-3.41) mas/yr, which matches previous HST/ACS measurements with a very good accuracy of 0.03 mas/yr and with a comparable precision (r.m.s of 0.43 mas/yr). Conclusions: this study successfully demonstrates that high-quality proper motions can be measured for quite large fields of view (85 arcsec X 85 arcsec) with MCAO-assisted, ground-based cameras and provides a first, successful test of the performances of GeMS on multi-epoch data., Comment: 5 pages, 4 figures. Near-infrared (NIR) spectra that have an angular resolution of ~ 0.15 arcsec are used to examine the stellar content of the central regions of the nearby elliptical galaxy Maffei 1. The spectra were recorded at the Subaru Telescope, with wavefront distortions corrected by the RAVEN Multi-Object Adaptive Optics science demonstrator. The Ballick-Ramsey C_2 absorption bandhead near 1.76 microns is detected, and models in which 10 - 20% of the light near 1.8 microns originates from stars of spectral type C5 reproduce this feature. Archival NIR and mid-infrared images are also used to probe the structural and photometric properties of the galaxy. Comparisons with models suggest that an intermediate age population dominates the spectral energy distribution between 1 and 5 microns near the galaxy center. This is consistent not only with the presence of C stars, but also with the large HBeta index that has been measured previously for Maffei 1. The J-K color is more-or-less constant within 15 arcsec of the galaxy center, suggesting that the brightest red stars are well-mixed in this area., Comment: To appear in The Astrophysical Journal

The Extremely Large Telescopes currently under construction have a collecting area that is an order of magnitude larger than the present largest optical telescopes. For seeing-limited observations the performance will scale as the collecting area but, with the successful use of adaptive optics, for many applications it will scale as $D^4$ (where $D$ is the diameter of the primary mirror). Central to the success of the ELTs, therefore, is the successful use of multi-conjugate adaptive optics (MCAO) that applies a high degree correction over a field of view larger than the few arcseconds that limits classical adaptive optics systems. In this letter, we report on the analysis of crowded field images taken on the central region of the Galactic globular cluster NGC 1851 in $K_s$ band using GeMS at the Gemini South telescope, the only science-grade MCAO system in operation. We use this cluster as a benchmark to verify the ability to achieve precise near-infrared photometry by presenting the deepest $K_s$ photometry in crowded fields ever obtained from the ground. We construct a colour-magnitude diagram in combination with the F606W band from HST/ACS. As well as detecting the "knee" in the lower main sequence at $K_s\simeq20.5$, we also detect the double subgiant branch of NGC 1851, that demonstrates the high photometric accuracy of GeMS in crowded fields., Comment: Accepted for publication in ApJL (3 Sep 2015). The Thirty Meter Telescope (TMT) first light instrument IRIS (Infrared Imaging Spectrograph) will complete its preliminary design phase in 2016. The IRIS instrument design includes a near-infrared (0.85-2.4 micron) integral field spectrograph (IFS) and imager that are able to conduct simultaneous diffraction-limited observations behind the advanced adaptive optics system NFIRAOS. The IRIS science cases have continued to be developed and new science studies have been investigated to aid in technical performance and design requirements. In this development phase, the IRIS science team has paid particular attention to the selection of filters, gratings, sensitivities of the entire system, and science cases that will benefit from the parallel mode of the IFS and imaging camera. We present new science cases for IRIS using the latest end-To-end data simulator on the following topics: Solar System bodies, the Galactic center, active galactic nuclei (AGN), and distant gravitationally-lensed galaxies. We then briefly discuss the necessity of an advanced data management system and data reduction pipeline.

Using five independent analytic and Monte Carlo simulation codes, we have studied the performance of wide field ground layer adaptive optics (GLAO), which can use a single, relatively low order deformable mirror to correct the wavefront errors from the lowest altitude turbulence. GLAO concentrates more light from a point source in a smaller area on the science detector, but unlike traditional adaptive optics, images do not become diffraction-limited. Rather the GLAO point spread function (PSF) has the same functional form as a seeing-limited PSF, and can be characterized by familiar performance metrics such as Full-Width Half-Max (FWHM). The FWHM of a GLAO PSF is reduced by 0.1" or more for optical and near-infrared wavelengths over different atmospheric conditions. For the Cerro Pachon atmospheric model this correction is even greater when the image quality is worst, which effectively eliminates "bad-seeing" nights; the best seeing-limited image quality, available only 20% of the time, can be achieved 60 to 80% of the time with GLAO. This concentration of energy in the PSF will reduce required exposure times and improve the efficiency of an observatory up to 30 to 40%. These performance gains are relatively insensitive to a number of trades including the exact field of view of a wide field GLAO system, the conjugate altitude and actuator density of the deformable mirror, and the number and configuration of the guide stars., Comment: 16 pages, 19 figures, accepted for publication in PASP

Systematic effects on HI and Halpha long-slit observations make a measurement of the inner slope of the dark matter density distribution difficult to determine. Halos with constant density cores and ones with r^-1 profiles both appear consistent with the data, although constant density cores generally provide better fits. High-resolution, two-dimensional velocity fields remove most of the systematic effects, yet as a result of noncircular and random motions the inner slopes still cannot be accurately measured. Halo concentration parameters provide a more useful test of cosmological models because they are more tightly constrained by observations. We present a high resolution, SparsePak two-dimensional velocity field for the center of the low surface brightness (LSB) galaxy DDO 39. These data are a significant improvement on previous HI or Halpha long slit data, yet the inner rotation curve is still uncertain due to significant noncircular and random motions. These intrinsic uncertainties, probably present in other LSB galaxies too, result in a wide range of inner slopes being consistent with the data, including those expected in cold dark matter (CDM) simulations. The halo concentration parameter provides a more useful test of cosmological models than the inner slope as it is more tightly constrained by observations. Background Molecular skin profiling techniques, typically performed on skin samples taken by punch biopsy, have enhanced the understanding of the pathophysiology of atopic dermatitis (AD), thereby enabling the development of novel targeted therapeutics. However, punch biopsies are not always feasible or desirable, and novel minimally invasive methods such as skin tape stripping have been developed. Aim To develop, optimize and validate a novel tape stripping method guided by noninvasive in vivo skin imaging to sample atopic skin in children. Methods Skin tape stripping-based procedures were compared and optimized using data from 30 healthy controls (HCs: 5 adults, 25 children) and 39 atopic children. Evaluations were guided by high-resolution photography, reflectance confocal microscopy, optical coherence tomography and transepidermal water loss measurements. We assessed and compared adverse events (AEs), the time needed to perform the sampling and the cDNA levels obtained from the tapes. Results Tape stripping methods based on previously described protocols resulted in erosions in all participants and required a median time of 65 min to perform (range 60–70 min), but provided good cDNA yield. Shorter durations appeared less invasive but provided lower cDNA yield. The final optimized tape stripping protocol, using 11 tapes of 22 mm in diameter, each applied twice for 5 s with 90° rotation, did not produce significant AEs, was completed within a median time of 7 min (range 5–15 min) and provided good cDNA yield both in HCs and atopic children. Conclusion Our minimally invasive method is safe and reliable, and provides reproducible acquisition of cDNA in atopic children. Threatened species with restricted ranges are at risk from habitat fragmentation and loss, which amplifies genetic bottleneck and impacts of small changes to their environments. Ecological models including population viability analyses (PVAs) can predict the trajectory of populations in a way that is not invasive or detrimental to the study species. They can therefore be a vital tool in modeling populations for conservation purposes. Although habitat suitability models have been used in studies to suggest areas for protected area designation, PVAs are generally not used in this regard. Dryophytes suweonensis and Dryophytes flaviventris are two threatened treefrog species endemic to the Korean Peninsula. The two species face threats of habitat loss and degradation and predation by invasive species among others. We used an integrated modeling approach combining ecological niche, connectivity, and PVAs in Vortex to determine the likelihood of extinction of each species under baseline and protected area designation scenarios. Designation scenarios were simulated in Vortex through halting future reduction in carrying capacity (halting future degradation to sites through protected area status), reducing effects of catastrophes (mitigating the effects of drought), and reducing mortality rates (controlling invasive predator populations and ex situ raising of tadpoles to maturity). We classified the combination of these management efforts as "active management" as opposed to "no management," which is currently being practiced. We additionally used a stepwise approach to determine designation priority of individual patches. Under current conditions (no management), the resulting effective metapopulations after 100 years were 167 ± 325 individuals with an 86.5% extinction probability for D. suweonensis and 165 ± 200 individuals with a 90.3% extinction probability for D. flaviventris. Under active management of all sites (93 sites covering 426.9 km2), the extinction probability was 0% for both species with significantly increased metapopulation sizes, 15 910 ± 2855 for D. suweonensis, and 4400 ± 874 for D. flaviventris. Determining designation priority can inform the regulatory bodies on which habitat to designate and whether active or passive management should be applied. Without intervention, these species will be likely to face imminent extinction. In addition to being useful for government‐imposed conservation management, our study can be followed by future studies as a methodology for prioritizing sites for protected area designation. The Extremely Large Telescope and the Thirty Meter Telescope will use state of the art multiconjugate adaptive optics (MCAO) systems to obtain the full D 4 advantage that their apertures can provide. However, to reach the full astrometric potential of these facilities for on-sky science requires understanding any residual astrometric distortions from these systems and find ways to measure and eliminate them. In this work, we use Gemini multiconjugate adaptive optic system (GeMS) observations of the core of NGC 6723 to better understand the on-sky astrometric performance of MCAO. We develop new methods to measure the astrometric distortion fields of the observing system, which probe the distortion at the highest possible spatial resolution. We also describe methods for examining the time-variable and static components of the astrometric distortion. When applied to the GeMS Gemini South Adaptive Optics Imager (GSAOI) data, we are able to see the effect of the field rotator at the subpixel level, and we are able to empirically derive the distortion due to the optical design of GeMS/GSAOI. We argue that the resulting distortion maps are a valuable tool to measure and monitor the on-sky astrometric performance of future instrumentation. Our overall astrometry pipeline produces high-quality proper motions with an uncertainty floor of ∼45 μas yr−1. We measure the proper motion dispersion profile of NGC 6723 from a radius of ∼10″ out to ∼1′ based on ∼12,000 stars. We also produce a high-quality optical–near infrared color–magnitude diagram, which clearly shows the extreme horizontal branch and main-sequence knee of this cluster.

Background Molecular skin profiling techniques, typically performed on skin samples taken by punch biopsy, have enhanced the understanding of the pathophysiology of atopic dermatitis (AD), thereby enabling the development of novel targeted therapeutics. However, punch biopsies are not always feasible or desirable, and novel minimally invasive methods such as skin tape stripping have been developed. Aim To develop, optimize and validate a novel tape stripping method guided by noninvasive in vivo skin imaging to sample atopic skin in children. Methods Skin tape stripping-based procedures were compared and optimized using data from 30 healthy controls (HCs: 5 adults, 25 children) and 39 atopic children. Evaluations were guided by high-resolution photography, reflectance confocal microscopy, optical coherence tomography and transepidermal water loss measurements. We assessed and compared adverse events (AEs), the time needed to perform the sampling and the cDNA levels obtained from the tapes. Results Tape stripping methods based on previously described protocols resulted in erosions in all participants and required a median time of 65 min to perform (range 60–70 min), but provided good cDNA yield. Shorter durations appeared less invasive but provided lower cDNA yield. The final optimized tape stripping protocol, using 11 tapes of 22 mm in diameter, each applied twice for 5 s with 90° rotation, did not produce significant AEs, was completed within a median time of 7 min (range 5–15 min) and provided good cDNA yield both in HCs and atopic children. Conclusion Our minimally invasive method is safe and reliable, and provides reproducible acquisition of cDNA in atopic children. - Abstract
Background: After acute coronary syndrome, diabetes conveys an excess risk of ischaemic cardiovascular events. A reduction in mean LDL cholesterol to 1·4–1·8 mmol/L with ezetimibe or statins reduces cardiovascular events in patients with an acute coronary syndrome and diabetes. However, the efficacy and safety of further reduction in LDL cholesterol with an inhibitor of proprotein convertase subtilisin/kexin type 9 (PCSK9) after acute coronary syndrome is unknown. We aimed to explore this issue in a prespecified analysis of the ODYSSEY OUTCOMES trial of the PCSK9 inhibitor alirocumab, assessing its effects on cardiovascular outcomes by baseline glycaemic status, while also assessing its effects on glycaemic measures including risk of new-onset diabetes. Methods: ODYSSEY OUTCOMES was a randomised, double-blind, placebo-controlled trial, done at 1315 sites in 57 countries, that compared alirocumab with placebo in patients who had been admitted to hospital with an acute coronary syndrome (myocardial infarction or unstable angina) 1–12 months before randomisation and who had raised concentrations of atherogenic lipoproteins despite use of high-intensity statins. Patients were randomly assigned (1:1) to receive alirocumab or placebo every 2 weeks; randomisation was stratified by country and was done centrally with an interactive voice-response or web-response system. Alirocumab was titrated to target LDL cholesterol concentrations of 0·65–1·30 mmol/L. In this prespecified analysis, we investigated the effect of alirocumab on cardiovascular events by glycaemic status at baseline (diabetes, prediabetes, or normoglycaemia)—defined on the basis of patient history, review of medical records, or baseline HbA1c or fasting serum glucose—and risk of new-onset diabetes among those without diabetes at baseline.
