Lichens are a symbiosis between a fungus and one or more photosynthetic microorganisms that enables the symbionts to thrive in places and conditions they could not compete independently. Exchanges of water and sugars between the symbionts are the established mechanisms that support lichen symbiosis. Herein, we present a new linkage between algal photosynthesis and fungal respiration in lichen Flavoparmelia caperata that extends the physiological nature of symbiotic co-dependent metabolisms, mutually boosting energy conversion rates in both symbionts. Measurements of electron transport by oximetry show that photosynthetic O 2 is consumed internally by fungal respiration. At low light intensity, very low levels of O 2 are released, while photosynthetic electron transport from water oxidation is normal as shown by intrinsic chlorophyll variable fluorescence yield (period-4 oscillations in flash-induced Fv/Fm). The rate of algal O 2 production increases following consecutive series of illumination periods, at low and with limited saturation at high light intensities, in contrast to light saturation in free-living algae. We attribute this effect to arise from the availability of more CO 2 produced by fungal respiration of photosynthetically generated sugars. We conclude that the lichen symbionts are metabolically coupled by energy conversion through exchange of terminal electron donors and acceptors used in both photosynthesis and fungal respiration. Algal sugars and O 2 are consumed by the fungal symbiont, while fungal delivered CO 2 is consumed by the alga.

Antibiotic fermentation residues is a hazardous waste due to the existence of residual antibiotics and antibiotic resistance genes (ARGs), probably leading to the induction and spread of antibiotic resistant bacteria (ARB) in the environment, which could pose potential harm to the ecosystem and human health. It is urgent to develop an effective technology to remove the residual antibiotics and ARGs. In this study, the anaerobic digestion combined with gamma irradiation was applied for the disposal and utilization of cephalosporin C fermentation residues. The experimental results showed that the antibacterial activities of cephalosporin C against Staphylococcus aureus were significantly decreased after anaerobic digestion. The removal of tolC, a multidrug resistant gene, was improved up to 100% by the combination of gamma irradiation and anaerobic digestion compared to solely anaerobic digestion process, which may be due to the changes of microbial community structures induced by gamma irradiation., (Copyright © 2019 Elsevier B.V. Low oxygen conditions occur in grass sites due to high and frequent precipitation, poor soil quality, and over-irrigation followed by slow drainage. Three warm-season and one cool-season grass were analyzed at metabolic level during a time-course experiment performed in a controlled anoxic environment. Prolonged oxygen depletion proved detrimental by leading to premature death to all the species, with the exception of seashore paspalum. Moreover, the anoxia tolerance observed in these grasses has been associated with slow use of carbohydrates, rather than with their relative abundance, which was more important than their antioxidant capacity. Further physiological characterization of eight seashore paspalum genotypes to anoxia was also performed, by examining the variation in photosystem II (PSII) efficiency and gas exchange during post-anoxia recovery. Multivariate analysis highlighted the presence of three main clusters of seashore paspalum genotypes, characterized by different ability to restore the PSII photochemistry during recovery after one day of anoxia. Taken together, our data demonstrate that the analysis of post-anoxia recovery of fluorescence and gas exchange parameters can represent a fast and reliable indicator for selecting species and cultivars more able to acclimate their photosynthetic apparatus., (© 2017 Scandinavian Plant Physiology Society.)

Huge amount of aging biogas slurry is in urgent need to be treated properly. However, due to high NH 3 -N concentration and low C/N ratio, this aging biogas slurry is refractory for traditional methods. Its denitrification has become a big challenge. In this paper, photosynthetic bacteria (PSB) were employed to handle this problem. The results showed denitrification of aging biogas slurry by PSB treatment was promising. The highest removal efficiency of NH 3 -N reached 99.75%, much higher than all other treatments. The removal of NH 3 -N followed pseudo zero order reaction under dark-aerobic condition. The better inoculation rate for NH 3 -N removal was 30%; and aerobic condition was more beneficial for NH 3 -N removal than anaerobic condition because of different metabolic pathways., (Copyright © 2017 Elsevier Ltd. Comparative analyses of bacterial and archaeal community structures and dynamics in three biogas digesters during start-up and subsequent operation using microwaved, ultrasonicated or untreated waste activated sludge were performed based on 454 pyrosequencing datasets of part of 16S ribosomal RNA sequences and quantitative PCR. The pre-treatment increased the solubility, and thus the availability of the substrate for microbial degradation and significantly affected the succession of the anaerobic community structure over the course of the digestion. Bacteroidetes, Proteobacteria and Firmicutes were the dominant phyla in all digesters throughout operation. Proteobacteria decreased in relative abundance from 23-26 % to 11-13 % in association with enhanced substrate availability. Negative correlations between relative abundance of Alpha-, Beta- and Gammaproteobacteria and the substrate availability and/or biogas production were disclosed in statistical analyses. Clostridiales was the dominant order in Firmicutes, and Clostridiales, Clostridia and Firmicutes relative abundance and richness were shown to positively correlate with substrate availability and biogas generation. Methanogenic communities had a fairly restricted structure, highly dominated by Methanosaeta and Methanobrevibacter phylotypes. A gradual decline in Methanobrevibacter and increased representation of Methanosaeta concilii over time were particularly apparent in the digester receiving untreated waste activated sludge, whereas more diversified archaeal communities were maintained in the pre-treatment digesters. The quantitative PCR analyses revealed a methanogenic community distribution that coincided with the 454 pyrosequencing data.

Background: Oxygen is generally considered essential for lethal photosensitisation by photodynamic processes. The oral anaerobes, Prevotella intermedia and P. nigrescens are known to be photosensitive, but are also extremely sensitive to the cytotoxic effects of oxygen., Methods: The Prevotellaceae were exposed to two 405 nm light sources for different exposure times in an anaerobic chamber. Viable counts of the light exposed samples were compared to light-free controls to determine the proportion of bacteria killed., Results: Lethal photosensitivity was demonstrated against P. intermedia and P. nigrescens. Anoxygenic phototrophic bacteria are a diverse collection of organisms that are defined by their ability to grow using energy from light without evolving oxygen. The dominant groups are purple sulfur bacteria, purple nonsulfur bacteria, green sulfur bacteria, and green and red filamentous anoxygenic phototrophic bacteria. They represent several bacterial phyla but they all have bacteriochlorophylls and carotenoids and photochemical reaction centers which generate ATP and cellular reductants used for CO 2 fixation. They typically have an anaerobic lifestyle in the light, although some grow aerobically in the dark. Some of them oxidize inorganic sulfur compounds for light-dependent CO 2 fixation; this ability can be exploited for photobiological removal of hydrogen sulfide from wastewater and biogas. The anoxygenic phototrophic bacteria also perform bioremediation of recalcitrant dyes, pesticides, and heavy metals under anaerobic conditions. Finally, these organisms may be useful for overexpression of membrane proteins and photobiological production of H 2 and other valuable compounds.

The model green microalga Chlamydomonas reinhardtii is frequently subject to periods of dark and anoxia in its natural environment. Here, by resorting to mutants defective in the maturation of the chloroplastic oxygen-sensitive hydrogenases or in Proton-Gradient Regulation-Like1 (PGRL1)-dependent cyclic electron flow around photosystem I (PSI-CEF), we demonstrate the sequential contribution of these alternative electron flows (AEFs) in the reactivation of photosynthetic carbon fixation during a shift from dark anoxia to light. At light onset, hydrogenase activity sustains a linear electron flow from photosystem II, which is followed by a transient PSI-CEF in the wild type. By promoting ATP synthesis without net generation of photosynthetic reductants, the two AEF are critical for restoration of the capacity for carbon dioxide fixation in the light. Our data also suggest that the decrease in hydrogen evolution with time of illumination might be due to competition for reduced ferredoxins between ferredoxin-NADP(+) oxidoreductase and hydrogenases, rather than due to the sensitivity of hydrogenase activity to oxygen. Finally, the absence of the two alternative pathways in a double mutant pgrl1 hydrogenase maturation factor G-2 is detrimental for photosynthesis and growth and cannot be compensated by any other AEF or anoxic metabolic responses. Several batch culture studies were carried out to evaluate an anaerobically treated effluent as a low-cost growth medium for the microalga Chlamydomonas acidophila and to study the effectiveness of the microalga in removing NH4-N from the effluent. An initial decrease in the effluent pH to 3 was required for adequate growth of C. acidophila and removal of NH4-N. Growth of the microalgae was inhibited at high light intensity (224μmolphotonsm(-2)s(-1) at the surface of the vessels). However, the growth was not greatly affected by the high solid content and turbidity of the effluent. The microalga was able to grow in media containing NH4-N at concentrations of up to 1000mgL(-1) (50% of effluent) and to remove 88mg of NH4-NL(-1) in 10days. The effects of simulated outdoor seasonal climatic conditions on Nannochloropsis salina (N. salina) grown using nutrients from anaerobic digestion (AD) effluent were evaluated in this study. Under various light exposure (LE) and temperature (10-30 °C) conditions, N. salina specific growth rate (μ) was strongly affected by LE. Light availability (LA) was observed to be crucial for biomass production, with μ values of 0.038±0.013 d(-1), 0.093±0.013 d(-1), and 0.151±0.021 d(-1) for 6-h, 12-h, and 24-h LA conditions, respectively. Temperature (10-25 °C) was not significant in affecting the light dependent growth coefficient (μ/LE), indicating the suitability of culturing this strain in the Ohio climate. Cultures exposed to low illumination had significantly higher unsaturated fatty acid content than those under high illumination, with nearly 29% higher eicosapentaenoic acid (C20:5) content. Background: Lethal photosensitization has been previously demonstrated in Porphyromonas gingivalis, but oxygen is considered to be essential to this process. However, since P. gingivalis is a periodontal pathogen which grows in the low oxygen conditions found in the subgingival crevice, it was considered prudent to study its photosensitivity in anaerobic conditions., Methods: A series of experiments were undertaken to attempt to induce lethal photosensitization in P. gingivalis (ATCC 33277) under strict anaerobic conditions using two different 405 nm light sources. Samples of P. gingivalis were grown on a blood-containing, solid growth medium before being suspended in saline and then exposed to 405 nm light delivered by either a hand-held light source (Toothcare™) (11.4 mW/cm(2)) or a laser pointer (328.5 mW/cm(2)). With the exception of the adjustment of the P. gingivalis suspensions to a fixed optical density, the experiments were carried out in their entirety within an anaerobic chamber., Results: The lowest Toothcare light dose tested (0.34 J/cm(2); 30s) yielded a statically significant kill of 63.4% which increased to 94.1% kill at higher light doses (3.42 J/cm(2); 300 s). The laser pointer similarly achieved kills of 90.2% at the lower light dose tested (9.86 J/cm(2); 30s) and 94.5% kill at the highest light dose (98.55 J/cm(2); 300 s)., Conclusions: Lethal photosensitization can be instigated in planktonic suspensions of P. gingivalis at 405 nm delivered by hand-held devices under anaerobic conditions. The solid-state photo-CIDNP (photochemically induced dynamic nuclear polarization) effect is studied in photosynthetic reaction centers of Heliobacillus mobilis at different magnetic fields by (13)C MAS (magic-angle spinning) NMR spectroscopy. Two active states of heliobacterial reaction centers are probed: an anaerobic preparation of heliochromatophores ("Braunstoff", German for "brown substance") as well as a preparation of cells after exposure to oxygen ("Grünstoff", "green substance"). Braunstoff shows significant increase of enhanced absorptive (positive) signals toward lower magnetic fields, which is interpreted in terms of an enhanced differential relaxation (DR) mechanism. In Grünstoff, the signals remain emissive (negative) at two fields, confirming that the influence of the DR mechanism is comparably low.

Microbial pathogens in municipal sewage sludges need to be inactivated prior to environmental disposal. The efficacy of high energy (10 MeV) e-beam irradiation to inactivate a variety of selected microbial pathogens and indicator organisms in aerobically and anaerobically digested sewage sludge was evaluated. Both bacterial and viral pathogens and indicator organisms are susceptible to e-beam irradiation. However, as expected there was a significant difference in their respective e-beam irradiation sensitivity. Somatic coliphages, bacterial endospores and enteric viruses were more resistant compared to bacterial pathogens. The current US EPA mandated 10 kGy minimum dose was capable of achieving significant reduction of both bacterial and viral pathogens. Electrogenic activity of oxygenic photo-bioelectrocatalytic fuel cell (PhFCOX) under mixotrophic mode was evaluated using atmospheric CO2 and domestic wastewater as carbon sources for harnessing bioelectricity with mixed microalgae as anodic biocatalyst. PhFCOX operation showed good electrogenic activity (3.55 μW/m(2)) associated with higher biomass growth (2.87 g/l) and chlorophyll content (5.12 mg/l). Electrogenic activity was relatively higher during the day time (46 mV; 0.6 mA) compared to the night (6 mV; 0.01 mA). Performance of PhFCOX undergoing oxygenic photosynthesis (DO; 3.5 mg/l) was compared with the mixotrophic fuel cell (PhFCAX) with photosynthetic bacteria as biocatalyst under anoxygenic conditions (DO; 0.45 mg/l). The dissolved oxygen produced during photolysis of water in oxygenic photosynthesis is a major limiting factor affecting the electrogenic activity. Ag(2)O/TNBs were fabricated by depositing Ag(2)O nanoparticles on the surface of TiO(2) nanobelts (TNBs). The disinfection activities of Ag(2)O/TNBs on two representative bacterial types: Gram-negative Escherichia coli ATCC15597 and Gram-positive Bacillus subtilis, were examined under both dark and visible light conditions. Ag(2)O/TNBs exhibited stronger bactericidal activities than Ag(2)O nanoparticles and TNBs under both dark and light conditions. For both cell types, disinfection effects of Ag(2)O/TNBs were greater under light conditions relative to those under dark conditions. The bactericidal mechanisms of Ag(2)O/TNBs under both dark and light conditions were explored. Ag(+) ions released from Ag(2)O/TNBs did not contribute to the bactericidal activity of Ag(2)O/TNBs under dark conditions, whereas the released Ag(+) ions showed bactericidal activity under visible light irradiation conditions. Active species (H(2)O(2), O(2)(-)·, and e(-)) generated by Ag(2)O/TNBs played important roles in the disinfection processes under both dark and visible light irradiation conditions. Photolysis of some polycyclic aromatic hydrocarbons (PAHs) on soil surfaces may play an important role in the fate of PAHs in the environment. Photolysis of PAHs on soil surfaces under UV irradiation was investigated. The effects of oxygen, irradiation intensity and soil moisture on the degradation of the three PAHs were observed. The results showed that oxygen, soil moisture and irradiation intensity enhanced the photolysis of the three PAHs on soil surfaces. The degradation of the three PAHs on soil surfaces is related to their absorption spectra and the oxidation-half-wave potential. The photolysis of PAHs on soil surfaces in the presence of oxygen followed pseudo first-order kinetics. The photolysis half-lives ranged from 37.87 days for benzo[a]pyrene to 58.73 days for phenanthrene. The results indicate that photolysis is a successful way to remediate PAHs-contaminated soils.

Wheat (Triticum aestivum L.) seedlings contain four alanine aminotransferase (AlaAT) homologues. Two of them encode AlaAT enzymes, whereas two homologues act as glumate:glyoxylate aminotransferase (GGAT). To address the function of the distinct AlaAT homologues a comparative examination of the changes in transcript level together with the enzyme activity and alanine and glutamate content in wheat seedlings subjected to low oxygen availability, nitrogen and light deficiency has been studied. Shoots of wheat seedlings were more tolerant to hypoxia than the roots as judging on the basis of enzyme activity and transcript level. Hypoxia induced AlaAT1 earlier in roots than in shoots, while AlaAT2 and GGAT were unaffected. The increase in AlaAT activity lagged behind the increase in alanine content. Nitrogen deficiency has little effect on the activity of GGAT. In contrast, lower activity of AlaAT and the level of mRNA for AlaAT1 and AlaAT2 in wheat seedlings growing on a nitrogen-free medium seems to indicate that AlaAT is regulated by the availability of nitrogen. Both AlaAT and GGAT activities were present in etiolated wheat seedlings but their activity was half of that observed in light-grown seedlings. Exposure of etiolated seedlings to light caused an increase in enzyme activities and up-regulated GGAT1. It is proposed that hypoxia-induced AlaAT1 and light-induced peroxisomal GGAT1 appears to be crucial for the regulation of energy availability in plants grown under unfavourable environmental conditions. Key message In young wheat seedlings, both AlaAT and GGAT are down-regulated by nitrogen deficiency, whereas AlaAT1 is upregulated by hypoxia and GGAT1 by light.

Electrogenic activity of photo-bioelectrocatalytic /photo-biological fuel cell (PhFC) was evaluated in a mixotrophic mode under anoxygenic microenvironment using photosynthetic consortia as biocatalyst. An acetate rich wastewater was used as anolyte for harnessing energy along with additional treatment. Mixotrophic operation facilitated good electrogenic activity and wastewater treatment associated with biomass growth. PhFC operation documented feasible microenvironment for the growth of photosynthetic bacteria compared to algae which was supported by pigment (total chlorophyll and bacteriochlorophyll) and diversity analysis. Pigment data also illustrated the association between bacterial and algal species. The synergistic interaction between anoxygenic and oxygenic photosynthesis was found to be suitable for PhFC operation. Light dependent deposition of electrons at electrode was relatively higher compared to dark dependent electron deposition under anoxygenic condition. PhFC documented for good volatile fatty acids removal by utilizing them as electron donor. The photocatalytic degradation of two phenolic compounds, p-coumaric acid and caffeic acid, was performed with a suspended mixture of TiO(2) and powdered activated carbon (PAC) (at pH=3.4 and 8). Adsorption, direct photolysis and photocatalytic degradation were studied under different pH and UV light sources (sunlight vs. 365nm UV lamps). The potential for reusing this catalyst mixture in sequential photocatalytic runs was examined as well. Quantum yields for the direct photolysis of caffeic acid under solar and artificial 365nm light were calculated (for the first time) as 0.005 and 0.011, respectively. A higher removal rate of contaminants by either adsorption or photocatalysis was obtained at a low pH (pH 4). Furthermore, the addition of PAC increased the removal efficiency of the phenolic compounds. Fast removal of the pollutants from the solution over three sequential runs was achieved only when both TiO(2) and PAC were present. This suggests that at medium phenolic concentrations, the presence of PAC as a co-sorbent reduces surface poisoning of the TiO(2) catalyst and hence improves photocatalysis degradation of phenolic pollutants. The adsorption equilibrium of caffeic acid or p-coumaric acid on TiO(2), PAC and the combined mixture of TiO(2) and PAC follows the Langmuir isotherm model. Experiments with PAC TiO(2) mixture and olive mill wastewater (anaerobically treated and diluted by a factor of 10) showed higher removal of polyphenols than of chemical oxygen demand (COD). 87% removal of total polyphenols, compared to 58% of COD, was achieved after 24h of exposure to 365nm irradiation (7.6W/m(2)) in the presence of a suspended mixture of TiO(2) and PAC, indicating "self-selectivity" of polyphenols., (Copyright © 2011 Elsevier Ltd. The effect of substrate (glucose) concentration on the stability and yield of a continuous fermentative process that produces hydrogen was studied. Four anaerobic fluidized bed reactors (AFBRs) were operated with a hydraulic retention time (HRT) from 1 to 8 h and an influent glucose concentration from 2 to 25 g L(-1). The reactors were inoculated with thermally pre-treated anaerobic sludge and operated at a temperature of 30 °C with an influent pH around 5.5 and an effluent pH of about 3.5. The AFBRs with a HRT of 2 h and a feed strength of 2, 4, and 10 g L(-1) showed satisfactory H(2) production performance, but the reactor fed with 25 g L(-1) of glucose did not. The highest hydrogen yield value was obtained in the reactor with a glucose concentration of 2 g L(-1) when it was operated at a HRT of 2 h. The maximum hydrogen production rate value was achieved in the reactor with a HRT of 1 h and a feed strength of 10 g L(-1). The AFBRs operated with glucose concentrations of 2 and 4 g L(-1) produced greater amounts of acetic and butyric acids, while AFBRs with higher glucose concentrations produced a greater amount of solvents.

Perfluorooctanoic acid (PFOA) is a new-found hazardous persistent organic pollutant, and it is resistant to decomposition by hydroxyl radical (HO*) due to its stable chemical structure and the high electronegativity of fluorine. Photocatalytic reduction of PFOA with beta-Ga2O3 in anoxic aqueous solution was investigated for the first time, and the results showed that the photoinduced electron (e(cb-)) coming from the beta-Ga2O3 conduction band was the major degradation substance for PFOA, and shorter-chain perfluorinated carboxylic acids (PFCAs, CnF2n+i1COOH, 1 < or = n < or = 6) were the dominant products. Furthermore, the concentration of F- was measured by the IC technique and defluorination efficiency was calculated. After 3 hr, the photocatalytic degradation efficiency was 98.8% and defluorination efficiency was 31.6% in the presence of thiosulfate and bubbling N2. The degradation reaction followed first-order kinetics (k = 0.0239 min(-1), t1/2 = 0.48 hr). PFCAs (CnF2n+1COOH, 1 < or = n < or = 7) were detected and measured by LC-MS and LC-MS/MS methods. It was deduced that the probable photocatalytic degradation mechanism involves e(cb-) attacking the carboxyl of CnF2n+1COOH, resulting in decarboxylation and the generation of CnF2n+1*. The produced CnF2n+1* reacted with H2O, forming CnF2n+1OH, then CnF2n+1OH underwent HF loss and hydrolysis to form CnF2n+1COOH.

A dual irradiation process involving aerobic thermophilic irradiation pretreatment (ATIP) and intermittent irradiation anaerobic digestion was developed to improve the digestion of waste-activated sludge. First, the effect of ATIP on further anaerobic digestion of activated sludge in batch mode was investigated. When exposed to ATIP for 24 h, the digestion reactor gave the highest methane yield, removed the most dissolved organic carbon (DOC) and showed the most effective reduction of VS compared to other irradiation times. This process was further enhanced by using an anaerobic fluidised-bed reactor packed with carbon felt in semi-continuous mode for digesting the pretreated activated sludge under intermittent irradiation conditions. Dual irradiation for 24 h followed by 60 min of anaerobic irradiation processing per day turned out to be optimal. D-Arginine dehydrogenase (DADH) catalyzes the flavin-dependent oxidative deamination of D-arginine and other D-amino acids to the corresponding imino acids. The 1.07 Å atomic-resolution structure of DADH crystallized with D-leucine unexpectedly revealed a covalent N(5) flavin adduct, instead of the expected iminoleucine product in the active site. This acyl adduct has been successfully reproduced by photoreduction of DADH in the presence of 4-methyl-2-oxopentanoic acid (ketoleucine). This study investigated the effects of light (visible light - 5800 lux, 24h) or dark regime and aerobic or anaerobic condition on the decay of added oxolinic acid (OA) at 5, 10 and 20 mg L(-1) in eel pond sediment. An asymptotic decaying exponential model C(t)=C(min)+C(o) × exp (-k × t) was used to facilitate quantitative approach to OA transformation, where C(t) is the concentration of OA after t days, C(min) the estimated level-off concentration of OA residue, C(o) the concentration of added OA and k the decaying coefficient. OA decayed faster under light (C(min)=4.6 mg L(-1)) than under dark (C(min)=7.8 mg L(-1)) and also decayed faster under aerobic (C(min)=4.0 mg L(-1)) than under anaerobic condition (C(min)=8.5 mg L(-1)). In this paper, we quantify the effect of heating pretreatment on the degree of solubilization of waste activated sludge. The pretreatment process was carried out using a lab-scale industrial microwave unit (2450 MHz frequency). Response surface analysis was applied to determine the combination of output power (400-1600 W), target temperature (60-120 degrees C), and total solid concentration (1-3% total solid (TS)). The power, temperature, and TS concentration significantly affected the solubilization degree of sludge. Within the design boundaries, the conditions predicted to maximize the solubilization degree of 17.9% were determined to be 400 W, 102 degrees C, and 2.3% TS.

Commonly used pretreatment method of alkaline solubilization (using NaOH) and a relatively new technology of microwave (MW) irradiation (160 degrees C) were combined as a pretreatment method of waste activated sludge (WAS) in this study. First alkaline and MW pretreatment methods were examined separately, then their combination for different conditions was investigated in terms of their effect on COD solubilization, turbidity and capillary suction time (CST). For combined pretreatments, soluble COD to total COD ratio (SCOD/TCOD) of WAS increased from 0.005 (control) to 0.18, 0.27, 0.34 and 0.37 for combined methods of MW and pH-10, 11, 12 and 12.5, respectively. Deteriorated dewaterability due to alkaline pretreatment was also improved due to the incorporation of MW irradiation. Further, with small scale batch anaerobic reactors, pH-10, pH-12, MW (alone), MW+pH-10 and MW+pH-12 pretreated WAS samples were anaerobically digested. Highest total gas and methane productions were achieved with MW+pH-12 pretreatment with 16.3% and 18.9% improvements over control reactor, respectively. Finally the performance of MW+pH-12 pretreatment was examined with 2L anaerobic semi-continuous reactors for 92 days and compared to that of the control reactors. These reactors were operated at an SRT of 15 days. After steady state, 43.5% and 55% improvements were obtained in respective daily total gas and methane productions. TS, VS and TCOD reductions were improved by 24.9%, 35.4% and 30.3%, respectively based on a relative calculation with respect to control reactors. This way combined alkaline-microwave treatment proved to be an effective sludge minimization method. Pretreated digested sludge had 22% improved dewaterability than unpretreated digested sludge. Higher SCOD and NH(3)-N concentrations were measured in the pretreated digested sludge supernatant; however, PO(4)-P concentration did not increase much.

Effect of microwave pretreatment (MW) high temperature (175 degrees C) and MW intensity to waste activated sludge digested with acclimatized inoculum in single- and dual-stage semi-continuous mesophilic anaerobic digesters at different sludge retention times (SRTs) (20, 10 and 5 days) were investigated. MW pretreatment led to similar sludge stabilization at low SRTs (5 and 10 days). Although lowering MW intensity slightly improved sludge solubilization, it had a negative effect on digestion at low SRTs. Single-stage digesters with MW pretreatment surpass dual-stage digesters performances.

Halobacterium salinarum swims with the help of a polarly inserted flagellar bundle. In energized cells, the flagellar motors rotate continuously, occasionally switching the rotational sense. Starving cells become immotile as the energy level drops. Presumably, there is a threshold of energy required for flagellar rotation. When starved, immotile cells are energized by exposure to light, the speed of flagellar rotation increases gradually to its steady state over several minutes. Since the light-driven proton pump bacteriorhodopsin energizes the cell membrane to the maximal level within a fraction of a second, the delay in reaching the maximal swimming speed suggests that the halobacterial flagellar motor may not be driven directly by proton motive force. Swimming cells, which obtain their energy exclusively through light-driven proton pumping, become immotile within 20 min when treated with N,N'-dicyclohexylcarbodiimide (DCCD), an inhibitor of the proton translocating ATP synthase. However, flagellar motility in DCCD-treated cells can be restored by the addition of L-arginine, which serves as a fermentative energy source and restores the cytoplasmic ATP level in the presence of DCCD. This suggests that flagellar motor rotation depends on ATP, and this is confirmed by the observation that motility is increased strongly by L-arginine at zero proton motive force levels. The flagellar motor may be driven either by ATP directly or by an ATP-generated ion gradient that is not coupled directly to the proton gradient or the proton motive force of the cell.

Purple non-sulfur bacteria (Rhodospirillaceae) have been extensively employed for studying principles of photosynthetic and respiratory electron transport phosphorylation and for investigating the regulation of gene expression in response to redox signals. Here, we use mathematical modeling to evaluate the steady-state behavior of the electron transport chain (ETC) in these bacteria under different environmental conditions. Elementary-modes analysis of a stoichiometric ETC model reveals nine operational modes. Most of them represent well-known functional states, however, two modes constitute reverse electron flow under respiratory conditions, which has been barely considered so far. We further present and analyze a kinetic model of the ETC in which rate laws of electron transfer steps are based on redox potential differences. Our model reproduces well-known phenomena of respiratory and photosynthetic operation of the ETC and also provides non-intuitive predictions. As one key result, model simulations demonstrate a stronger reduction of ubiquinone when switching from high-light to low-light conditions. This result is parameter insensitive and supports the hypothesis that the redox state of ubiquinone is a suitable signal for controlling photosynthetic gene expression.

It has been shown that coherent electromagnetic irradiation (EMI) of extremely high frequency (45-53 GHz) or millimeter waves (wavelength 5.6-6.7 mm) of low intensity (flux capacity 0.06 mW/cm2) of Escherichia coli K12, grown under anaerobic conditions during the fermentation of sugar (glucose) for 30 min or 1 h, caused a decrease in their growth rate, the maximum inhibitory effect being achieved at a frequency of 51.8 or 53 GHz. This effect depended on medium pH when the maximal action was determined at pH 7.5. In addition, separate 30-min of 1-h irradiation (frequency 51.8 or 53 GHz) of doubly distilled water or some inorganic ions contained in Tris-phosphate buffer where the cells were transferred induced oppositely directed changes in further growth of these bacteria under anaerobic conditions; irradiation of water caused a decrease in the growth rate of bacteria. A significant change in pH of water (0.5-1.5 unit) was induced by a 30-irradiation at a frequency of 49, 50.3, 51.8, or 53 GHz, when the initial pH value was 6.0 or 8.0, but not 7.5. These results indicate the changes in the properties of water and its role in the effects of EMI of extremely high frequency. The marked effect of EMI on bacteria disappeared upon repeated irradiation for 1 h at a frequency of 51.8 or 53 GHz with an interval of 2 hours. This result indicates some compensatory mechanisms in bacteria.

Alpha-anomeric 2'-deoxynucleosides (alphadN) are one of the products formed by ionizing radiation (IR) in DNA under anoxic conditions. Alpha-2'-deoxyadenosine (alphadA) and alpha-thymidine (alphaT) are not recognized by DNA glycosylases, and are likely removed by the alternative nucleotide incision repair (NIR) pathway. Indeed, it has been shown that alphadA is a substrate for the Escherichia coli Nfo and human Ape1 proteins. However, the repair pathway for removal of alphadA and other alphadN in yeast is unknown. Here we report that alphadA when present in DNA is recognized by the Saccharomyces cerevisiae Apn1 protein, a homologue of Nfo. Furthermore, alphaT is a substrate for Nfo and Apn1. Kinetic constants indicate that alphadA and alphaT are equally good substrates, as a tetrahydrofuranyl (THF) residue, for Nfo and Apn1. Using E. coli and S. cerevisiae cell-free extracts, we have further substantiated the role of the nfo and apn1 gene products in the repair of alphadN. Surprisingly, we found that bacteria and yeast NIR-deficient mutants are not sensitive to IR, suggesting that DNA strand breaks with terminal 3'-blocking groups rather than alphadN might contribute to cell survival. We propose that the novel substrate specificities of Nfo and Apn1 play an important role in counteracting oxidative DNA base damage.

Repair under non-growth conditions of DNA double-stranded breaks (DSBs) and S1 nuclease-sensitive sites (SSSs; e.g. DNA damage which is processed by in vitro treatment with S1 nuclease to DSBs) induced by [60Co]-gamma-rays (200 Gy; anoxic conditions) was monitored in a diploid repair-competent strain of Saccharomyces cerevisiae. We used pulsed-field gel electrophoresis (PFGE), which allows the separation of chromosome-sized yeast DNA molecules, to determine the number of DSBs and SSSs in individual chromosome species of yeast. Our results indicate that SSSs which have been regarded as clusters of base damage in opposite DNA strands are repaired efficiently in a repair-proficient diploid strain of yeast. The time course of SSS repair is comparable to the one of DSB repair, indicating similarities in the molecular mechanism. Both types of repair kinetics are different for different chromosome species.

Diploid cells of the wild-type yeast Saccharomyces cerevisiae, mutants homozygous with respect to rad2 and rad54 loci as well as a double mutant with both these loci in homozygous state were used to demonstrate the previously observed (in other yeast strains) genetic determination of radiosensitivity modification of hypoxic cells by oxygen and electron-affinic compounds. It was shown that both oxygen effect and the effect of hypoxic sensitizers depended on the activity of repair systems. A possible mechanism of participation of postradiation recovery in modification of yeast cell radiosensitivity is discussed.

Chinese hamster V79 spheroids were stained with a nontoxic fluorescent stain, Hoechst 33342, which penetrates slowly into the spheroids. Single cells from these spheroids were then sorted by a fluorescence-activated cell sorter according to staining intensity (and therefore position in the spheroids). Flow cytometry characterization of the various cell subpopulations indicated that the innermost cells were more radiosensitive than expected on the basis of cell cycle position or cell thiol content. However, comparison of the radiosensitivities of cells sorted from equivalent depths from completely aerobic or anoxic V79 spheroids indicated that the oxygen enhancement ratio remained remarkably constant at 2.7 +/- 0.2 through the spheroid.

A rhodium compound, Rh(NH3)3Cl3, does not sensitize the spores of Bacillus megaterium to X-rays. However, it is a very effective sensitizer of vegetative cells of Staphylococcus aureus, raising the sensitivity four times in O2 and over 100 times in anoxia. The inhibition by oxygen of the sensitizing action of Rh(III), which operates over a wide range of [O2], is noteworthy. These experiments were performed in saline-phosphate buffer using 50 kVp X-rays. The results are discussed in terms of the known radiation chemistry of this compound.

Three months after whole-body irradiation of mice with a sublethal dose of 5 Gy a study was made of some indices of energy metabolism like tissue respiration, oxidative phosphorylation, and formation of lactic acid in the survived brain homogenate. Revealed were the diminution of coupling of tissue respiration of oxidative phosphorylation, the rate of oxygen consumption and the level of cyano-resistant respiration being constant, the increase in the rate of glycolysis in anaerobic and particularly, in aerobic conditions, and reduction of the Pasteur and Crabtree effects. The above mentioned changes in the brain energy metabolism seem to be a manifestation of the process of the reduced metabolism formation in the nervous tissue at the remote times after irradiation.

A first attempt to impair a biological function by the synergistic effects of HpD and nitroimidazoles (metronidazole or misonidazole) under anaerobic conditions is reported. A function tightly linked to a membrane has been chosen as model and the Ca2+ movements through the inner mitochondrial membrane have been studied. When isolated mitochondria (1 mg/cm3) are irradiated in oxic condition with 25 micrograms/cm3 HpD and 1 mmol dm-3 nitroimidazole in the medium, Ca2+ uptake is stopped after 15 s of irradiation. When irradiated in anoxic conditions with 25 micrograms/cm3 HpD alone, addition of ATP triggers a normal Ca2+ uptake even after 10 min of irradiation. In contrast Ca2+ uptake is stopped by 1 min irradiation after addition of 1 mmol dm-3 misonidazole to HpD and by 2 min irradiation after addition of 1 mmol dm-3 metronidazole. Thus, under these experimental conditions, the enhancement ratio of the HpD action (defined as the ratio of the durations of irradiation necessary to obtain the cessation of Ca2+ uptake before and after addition of a component other than HpD) is of the order of 40, 10 and 5 for oxygen, misonidazole and metronidazole, respectively. Misonidazole is still efficient at a concentration of 0.1 mmol dm-3.

A study was made of Ehrlich ascites tumor growth and life span of mice bearing hemoblastosis La after inoculation of tumor cells subjected, in anaerobic conditions, to the effect of gamma-radiation and/or metronidazole and isometronidazole. It was shown that the cytotoxic effect of isometronidazole was less manifest than that of metronidazole the radiosensitizing effect, with a reference to anoxic tumor cells in vitro, being nearly the same.

Radiation and gynecologic oncologists agree that pelvic irradiation in the presence of pelvic inflammatory disease, parametritis, or pyometra can be catastrophic. A temperature elevation in a patient having intrauterine radiation is very perplexing. This study was designed to investigate one of the three reported etiologies for this condition. Some investigators have suggested an exacerbation of a quiescent infection as a possible etiology because of the necrotic condition of many cancerous cervices. Aerobic, anaerobic, and fungus cultures were performed on 32 patients prior to intrauterine administration of cesium. The organisms cultured were similar to those identified as the normal cervical flora. The frequency of the organisms seemed independent of the amount of irradiation. The number of organisms cultured did not vary with the use or nonuse of cleansing vaginal suppositories. This study indicated no increase in bacterial growth in these cancerous necrotic cervices; also it did not indicate the growth of any unusual pathologic bacteria.

The effect of radiosensitization of hypoxic bacterial cells by 9 nitroimidazoles was measured in the bacterial strains E. coli AB 1157 and S. lactis 712. Seven of these compounds were similar to misonidazole in their redox properties, but differed widely in their lipophilicites. The dependence of sensitization enhancement on reduction potential was similar to that reported in mammalian cells. The efficiency of sensitization was similar for compounds of low lipophilicity, but increased if the octanol: water partition coefficients of the compounds were higher than about 3.5. With one compound, otherwise similar to misonidazole, the increased lipophilicity led to about one order of magnitude lower concentration achieving the same degree of radiosensitization.

Gamma-ray-irradiated medium is found to produce an immediate slowing of respiration in cells of Escherichia coli 15 T-L-. It also introduces a lag in cell division that can be shortened by the presence of methylene blue and potassium cyanide. The lag is absent if the cells are grown anaerobically. These results suggest that the peroxides produced by irradiating medium block the electron transport system. Since cells concentrate peroxides, the loss of this function induced by bombardment was used to deduce the target molecular weight of the enzyme responsible. It is 73,000.
