Many studies have been conducted to evaluate the association between air pollution and adverse health effects using a wide variety of methods to assess exposure. However, the assessment of individual long-term exposure to ambient air pollution is a challenging task and has not been evaluated in a large autopsy study. Our goal was to investigate whether exposure to urban air pollution is associated to the degree of lung anthracosis, considering modifying factors such as personal habits, mobility patterns and occupational activities. We conducted a study in Sao Paulo, Brazil from February 2017 to June 2018, combining epidemiological, spatial analysis and autopsy-based approaches. Information about residential address, socio-demographic details, occupation, smoking status, time of residence in the city and time spent commuting was collected via questionnaires applied to the next-of-kin. Images of the pleura surface from upper and lower lobes were used to quantify anthracosis in the lungs. We used multiple regression models to assess the association between the amount of carbon deposits in human lungs, measured by the fraction of pleural anthracosis (FA), and potential explanatory variables. We analyzed 413 cases and our data showed that for each additional hour spent in daily commuting, the ratio FA/(1-FA) is multiplied by 1.05 (95% confidence interval: [1.02; 1.08]). The estimated coefficient for daily hours spent in traffic was not considerably affected by the inclusion of socio-demographic variables and smoking habits. We estimate a tobacco equivalent dose of 5 cigarettes per day in a city where annual PM2.5 concentration oscillates around 25 μg/m3. Pleural anthracosis is a potential index of lifetime exposure to traffic-derived air pollution.

Although myocardial growth corresponds to an adaptive response to maintain cardiac contractile function, the cardiac hypertrophy is a condition that occurs in many cardiovascular diseases and typically precedes the onset of heart failure. Different endocrine factors such as thyroid hormones, insulin, insulin-like growth factor 1 (IGF-1), angiotensin II (Ang II), endothelin (ET-1), catecholamines, estrogen, among others represent important stimuli to cardiomyocyte hypertrophy. Thus, numerous endocrine disorders manifested as changes in the local environment or multiple organ systems are especially important in the context of progression from cardiac hypertrophy to heart failure. Based on that information, this review summarizes experimental findings regarding the influence of such hormones upon signalling pathways associated with cardiac hypertrophy. Understanding mechanisms through which hormones differentially regulate cardiac hypertrophy could open ways to obtain therapeutic approaches that contribute to prevent or delay the onset of heart failure related to endocrine diseases.

The cardiac growth process (hypertrophy) is a crucial phenomenon conserved across a wide array of species and is critically involved in the maintenance of cardiac homeostasis. This process enables an organism to adapt to changes in systemic demand and occurs due to a plethora of responses, depending on the type of signal or stimuli received. The growth of cardiac muscle cells in response to environmental conditions depends on the type, strength and duration of stimuli, and results in adaptive physiological responses or non-adaptive pathological responses. Thyroid hormones (TH) have a direct effect on the heart and induce a cardiac hypertrophy phenotype, which may evolve to heart failure. In this review, we summarize the literature on TH function in the heart by presenting results from experimental studies. We discuss the mechanistic aspects of TH associated with cardiac myocyte hypertrophy, increased cardiac myocyte contractility and electrical remodeling, as well as the associated signaling pathways. In addition to classical crosstalk with the sympathetic nervous system (SNS), emerging work pointing to the new endocrine interaction between TH and the renin-angiotensin system (RAS) is also explored. Given the inflammatory potential of the angiotensin II peptide, this new interaction may open the door for new therapeutic approaches which target the key mechanisms responsible for TH-induced cardiac hypertrophy.

Background Cardiac fibrosis occurs because of disruption of the extracellular matrix network leading to myocardial dysfunction. Angiotensin II (Ang II ) has been implicated in the development of cardiac fibrosis. Recently, micro RNA s have been identified as an attractive target for therapeutic intervention in cardiac pathologies; however, the underlying mechanism of micro RNA s in cardiac fibrosis remains unclear. Next‐generation sequencing analysis identified a novel characterized microRNA, miR‐1954, that was significantly reduced in AngII‐infused mice. The finding led us to hypothesize that deficiency of miR‐1954 triggers cardiac fibrosis. Methods and Results A transgenic mouse was created using α‐MHC (α‐myosin heavy chain) promoter and was challenged with AngII infusion. AngII induced cardiac hypertrophy and remodeling. The in vivo overexpression of miR‐1954 showed significant reduction in cardiac mass and blood pressure in AngII‐infused mice. Further analysis showed significant reduction in cardiac fibrotic genes, hypertrophy marker genes, and an inflammatory gene and restoration of a calcium‐regulated gene (Atp2a2 [ATPase sarcoplasmic/endoplasmic reticulum Ca2+ transporting 2]; also known as SERCA 2), but no changes were observed in apoptotic genes. THBS 1 (thrombospondin 1) is indicated as a target gene for miR‐1954. Conclusions Our findings provide evidence, for the first time, that miR‐1954 plays a critical role in cardiac fibrosis by targeting THBS 1. We conclude that promoting the level of miR‐1954 would be a promising strategy for the treatment of cardiac fibrosis.

We have previously demonstrated that calcium-binding protein S100A8 and myeloid differentiation factor-88 (MyD88) are important mediators of nuclear transcription factor kappa-B (NF-κB) activation in cardiomyocytes and that signalling molecules are involved in the hypertrophic response that is stimulated by thyroid hormones (TH). Angiotensin II (Ang II), the main active peptide of the renin-angiotensin system (RAS), binds to type 1 Ang II receptor (AT1R) and subsequently promotes cardiac hypertrophy and the inflammatory response with NF-κB activation underlying the cardiovascular effects. Considering the amount of evidence that RAS is an important mediator of TH actions on the cardiovascular system, we aimed to investigate whether cardiac expression of NF-κB and upstream associated molecules could be altered in hyperthyroidism, as well as whether AT1R could mediate the effects of TH on cardiac tissue and in cardiomyocytes in culture. Wistar rats were subjected to hyperthyroidism with or without the AT1R blocker losartan. The TH serum levels, haemodynamic parameters and cardiac mass were assessed to confirm the hyperthyroid status. The S100A8, MyD88 and nuclear NF-κB expression levels were increased in the hearts of the hyperthyroid rats, and the losartan treatment attenuated these TH effects. In addition, the cultured cardiomyocytes that had been stimulated with losartan exhibited blunted S100A8 upregulation and NF-κB activation compared with the TH-treated cells. Together, our results suggest that AT1R participates in TH-induced cardiac hypertrophy partly by mediating S100A8, MyD88 and NF-κB activation via TH. These findings indicate the important crosstalk between TH and RAS, highlighting the participation of AT1R in the triggered mechanisms of TH that contribute to the cardiac hypertrophy response.

Background Disruption of endoplasmic reticulum (ER) homeostasis is a common feature of cardiac diseases. However, the signaling events involved in ER stress-induced cardiac dysfunction are still elusive. Here, we uncovered a mechanism by which disruption of ER homeostasis impairs cardiac contractility. Methods/results We found that ER stress is associated with activation of JNK and upregulation of BNIP3 in a post-myocardial infarction (MI) model of cardiac dysfunction. Of interest, 4-week treatment of MI rats with the chemical ER chaperone 4-phenylbutyrate (4PBA) prevented both activation of JNK and upregulation of BNIP3, and improved cardiac contractility. We showed that disruption of ER homeostasis by treating adult rat cardiomyocytes in culture with tunicamycin leads to contractile dysfunction through JNK signaling pathway. Upon ER stress JNK upregulates BNIP3 in a FOXO3a-dependent manner. Further supporting a BNIP3 mechanism for ER stress-induced deterioration of cardiac function, siRNA-mediated BNIP3 knockdown mitigated ER stress-induced cardiomyocyte dysfunction by reestablishing sarcoplasmic reticulum Ca2+ content. Conclusions Collectively, our data identify JNK-dependent upregulation of BNIP3 as a critical process involved in ER stress-induced cardiomyocyte contractile dysfunction and highlight 4PBA as a potential intervention to counteract ER stress-mediated BNIP3 upregulation in failing hearts.

O presente estudo avaliou aspectos relacionados ao contexto inflamatório na hipertrofia cardíaca induzida pelos hormônios tiroideanos (HT) e o possível envolvimento do sistema renina-angiotensina (SRA) nesse processo, utilizando análises in vivo e com enfoque maior na abordagem in vitro. Os resultados mostraram algumas alterações em citocinas circulantes e cardíacas de animais tratados com HT. Além disso, as expressões de S100A8 e MyD88 foram aumentadas no coração de ratos submetidos ao hipertiroidismo e em cardiomiócitos em cultura estimulados com HT. S100A8 e MyD88 mediaram a ativação do fator nuclear NF-κB pelos HT, tendo papel crucial para o crescimento hipertrófico de cardiomiócitos tratados com HT. Por fim, a ação dos HT modulando a expressão de S100A8 e NF-κB foi mediada pelo SRA. Estes dados contribuem com o entendimento das bases moleculares da ação dos HT e da relação deste com o SRA na hipertrofia cardíaca. The present study evaluated inflammation related aspects in cardiac hypertrophy induced by thyroid hormones (TH) and the possible involvement of the renin-angiotensin system (RAS) in this process, by using in vivo and in vitro analysis. The results showed alterations in circulating and cardiac cytokines from TH treated animals. The expression of S100A8 and MyD88 were increased in the heart of hyperthyroid rats and in cultured cardiomyocytes stimulated with TH. S100A8 and MyD88 mediated the nuclear factor NF-κB activation by TH and these factors presented crucial role to the hypertrophic growth of TH-treated cardiomyocytes. Finally, the action of TH on S100A8 and NF-κB expression was mediated by RAS. These data contribute to the knowledge of molecular basis of TH action and the relationship between TH and RAS in cardiac hypertrophy.

Recent studies have evidenced the involvement of inflammation-related pathways to the development of cardiac hypertrophy and other consequences on the cardiovascular system, including the calcium-binding protein S100A8. However, this has never been investigated in the thyroid hormone (TH)-prompted cardiac hypertrophy. Thus, we aimed to test whether S100A8 and related signaling molecules, myeloid differentiation factor-88 (MyD88) and nuclear factor kappa B (NF-қB), could be associated with the cardiomyocyte hypertrophy induced by TH. Our results demonstrate that the S100A8/MyD88/NF-қB signaling pathway is activated in cardiomyocytes following TH stimulation. The knockdown of S100A8 and MyD88 indicates the contribution of those molecules to cardiomyocyte hypertrophy in response to TH, as evaluated by cell surface area, leucine incorporation assay, and gene expression. Furthermore, S100A8 and MyD88 are crucial mediators of NF-қB activation, which is also involved in the hypertrophic growth of TH-treated cardiomyocytes. Supporting the in vitro data, the contribution of NF-қB for TH-induced cardiac hypertrophy is confirmed in vivo, by using transgenic mice with cardiomyocyte-specific suppression of NF-қB. These data identify a novel pathway regulated by TH that mediates cardiomyocyte hypertrophy. However, the potential role of this new pathway in short and long-term cardiac effects of TH remains to be further investigated.Inflammation-related signaling is activated by T3 in cardiomyocytes. The angiotensin II type 1 receptor (AT1R) is involved in the development of cardiac hypertrophy promoted by thyroid hormone. Recently, we demonstrated that triiodothyronine (T3) rapidly increases AT1R mRNA and protein levels in cardiomyocyte cultures. However, the molecular mechanisms responsible for these rapid events are not yet known. In this study, we investigated the T3effect onAT1RmRNA polyadenylation in cultured cardiomyocytes as well as on the expression of microRNA-350 (miR-350), which targetsAT1RmRNA. The transcriptional and translational actions mediated by T3onAT1Rlevels were also assessed. The total content of ubiquitinated proteins in cardiomyocytes treated with T3was investigated. Our data confirmed that T3rapidly raised AT1R mRNA and protein levels, as assessed by real-time PCR and western blotting respectively. The use of inhibitors of mRNA and protein synthesis prevented the rapid increase in AT1R protein levels mediated by T3. In addition, T3rapidly increased the poly-A tail length of theAT1RmRNA, as determined by rapid amplification of cDNA ends poly-A test, and decreased the content of ubiquitinated proteins in cardiomyocytes. On the other hand, T3treatment increased miR-350 expression. In parallel with its transcriptional and translational effects on theAT1R, T3exerted a rapid posttranscriptional action onAT1RmRNA polyadenylation, which might be contributing to increase transcript stability, as well as on translational efficiency, resulting to the rapid increase in AT1R mRNA expression and protein levels. Finally, these results show, for the first time, that T3rapidly triggers distinct mechanisms, which might contribute to the regulation of AT1R levels in cardiomyocytes.

In order to get further insight upon the effects of Thyroid hormone (T3) in the heart, we have used a microarray approach to identify responsive genes in rats submitted to T3 injections for 12, 24h and 7 days. Firstly, we have compared 5 different algorithms to analyze microarray data and found MAS5 to be the best fit in our model. Although it is well known that the thyroid hormone (T3) is an important positive regulator of cardiac function over a short term and that it also promotes deleterious effects over a long term, the molecular mechanisms for such effects are not yet well understood. Because most alterations in cardiac function are associated with changes in sarcomeric machinery, the present work was undertaken to find novel sarcomeric hot spots driven by T3 in the heart. A microarray analysis indicated that the M-band is a major hot spot, and the structural sarcomeric gene coding for the M-protein is severely down-regulated by T3. Real-time quantitative PCR-based measurements confirmed that T3 (1, 5, 50, and 100 physiological doses for 2 days) sharply decreased the M-protein gene and protein expression in vivo in a dose-dependent manner. Furthermore, the M-protein gene expression was elevated 3.4-fold in hypothyroid rats. Accordingly, T3 was able to rapidly and strongly reduce the M-protein gene expression in neonatal cardiomyocytes. Deletions at the M-protein promoter and bioinformatics approach suggested an area responsive to T3, which was confirmed by chromatin immunoprecipitation assay. Functional assays in cultured neonatal cardiomyocytes revealed that depletion of M-protein (by small interfering RNA) drives a severe decrease in speed of contraction. Interestingly, mRNA and protein levels of other M-band components, myomesin and embryonic-heart myomesin, were not altered by T3. We concluded that the M-protein expression is strongly and rapidly repressed by T3 in cardiomyocytes, which represents an important aspect for the basis of T3-dependent sarcomeric deleterious effects in the heart.

Previous studies have indicated that AMP-activated protein kinase (AMPK) plays a critical role in the control of cardiac hypertrophy mediated by different stimuli such as thyroid hormone (TH). Although the classical effects of TH mediating cardiac hypertrophy occur by transcriptional mechanisms, recent studies have identified other responses to TH, which are more rapid and take place in seconds or minutes evidencing that TH rapidly modulates distinct signaling pathway, which might contribute to the regulation of cardiomyocyte growth. Here, we evaluated the rapid effects of TH on AMPK signaling pathway in cultured cardiomyocytes and determined the involvement of AMPK in T3-induced cardiomyocyte growth. We found for the first time that T3 rapidly activated AMPK signaling pathway. The use of small interfering RNA against AMPK resulted in increased cardiomyocyte hypertrophy while the pharmacological stimulation of AMPK attenuated this process, demonstrating that AMPK contributes to regulation of T3-induced cardiomyocyte growth.

Hyperthyroidism promotes cardiac hypertrophy and the Angiotensin type 1 receptor (AT1R) has been demonstrated to mediate part of this response. Recent studies have uncovered a potentially important role for the microRNAs (miRNAs) in the control of diverse aspects of cardiac function. Then, the objective of the present study was to investigate the action promoted by hyperthyroidism on β-MHC/miR-208b expression and on α-MHC/miR-208a expression, as well as the possible contribution of the AT1R in this event. The findings of this study confirmed that AT1R is a key mediator of the cardiac hypertrophy induced by hyperthyroidism. Additionally, we demonstrated that like β-MHC, miR-208b was down-regulated in the hyperthyroid group. Similarly, like the expression of its host gene, α-MHC, miR-208a expression was up-regulated in response to hyperthyroidism. Finally, our data suggest for the first time that AT1R mediates the hyperthyroidism-induced increase on cardiac miRNA-208a/α-MHC levels, while does not influence on the reduction of miRNA-208b/β-MHC levels.

Estudos recentes demonstram que o Hormônio Tiroideano (HT) é capaz de modular rapidamente o estado de fosforilação de proteínas quinases relacionadas ao processo de hipertrofia cardíaca. Evidências experimentais indicam que a proteína quinase ativada por AMP (AMPK) seja um alvo importante no controle do crescimento hipertrófico, uma vez que a ativação desta enzima determina ampla variedade de efeitos siológicos, incluindo o controle de proteínas relacionadas à síntese protéica. Dessa forma, os objetivos deste estudo foram os de avaliar os efeitos do HT sobre a modulação da via de sinalização da AMPK, além de verificar o possível envolvimento desta quinase no modelo de hipertrofia in vitro induzida pelo HT. Os resultados obtidos mostraram que rapidamente este hormônio ativa a AMPK e proteínas relacionadas a esta sinalização. Além disso, a estimulação farmacológica da AMPK atenua a hipertrofia de cardiomiócitos induzida pelo HT. Estes dados sugerem que a AMPK seja uma possível ferramenta terapêutica em doenças cardiovasculares como a hipertrofia cardíaca. Some studies have shown that Thyroid Hormones (TH) are also able to rapidly modulate the phosphorylation state of protein kinases related to cardiac hypertrophy process by non-genomic actions. In this sense, experimental evidences indicate that AMP-activated protein kinase (AMPK) is an important target in the control of hypertrophic growth, since the activation of this enzyme determines wide variety of physiological effects, including the control of enzymes related to protein synthesis. Thus, the objective of this study was to evaluate the effects of TH on the modulation of AMPK signaling pathway and to check the possible involvement of this kinase in vitro model of hypertrophy induced by TH. The results showed that this hormone rapidly activates AMPK and related proteins to this signaling. Furthermore, pharmacological stimulation of AMPK attenuated cardiomyocyte hypertrophy induced by TH. These data suggest that AMPK may correspond to a possible therapeutic tool in cardiovascular disease.

The kallikrein-kinin and renin-angiotensin systems interact at multiple levels. In the present study, we tested the hypothesis that the B1 kinin receptor (B1R) contributes to vascular hypertrophy in angiotensin II (ANG II)-induced hypertension, through a mechanism involving reactive oxygen species (ROS) generation and extracellular signal-regulated kinase (ERK1/2) activation. Male Wistar rats were infused with vehicle (control rats), 400 ng/Kg/min ANG II (ANG II rats) or 400 ng/Kg/min ANG II plus B1 receptor antagonist, 350 ng/Kg/min des-Arg(9)-Leu(8)-bradykinin (ANGII+DAL rats), via osmotic mini-pumps (14 days) or received ANG II plus losartan (10 mg/Kg, 14 days, gavage - ANG II+LOS rats). After 14 days, ANG II rats exhibited increased systolic arterial pressure [(mmHg) 184 ± 5.9 vs 115 ± 2.3], aortic hypertrophy; increased ROS generation [2-hydroxyethidium/dihydroethidium (EOH/DHE): 21.8 ± 2.7 vs 6.0 ± 1.8] and ERK1/2 phosphorylation (% of control: 218.3 ± 29.4 vs 100 ± 0.25]. B1R expression was increased in aortas from ANG II and ANG II+DAL rats than in aortas from the ANG II+LOS and control groups. B1R antagonism reduced aorta hypertrophy, prevented ROS generation (EOH/DHE: 9.17 ± 3.1) and ERK1/2 phosphorylation (137 ± 20.7%) in ANG II rats. Cultured aortic vascular smooth muscle cells (VSMC) stimulated with low concentrations (0.1 nM) of ANG II plus B1R agonist exhibited increased ROS generation, ERK1/2 phosphorylation, proliferating-cell nuclear antigen expression and [H3]leucine incorporation. At this concentration, neither ANG II nor the B1R agonist produced any effects when tested individually. The ANG II/B1R agonist synergism was inhibited by losartan (AT1 blocker, 10 µM), B1R antagonist (10 µM) and Tiron (superoxide anion scavenger, 10 mM). These data suggest that B1R activation contributes to ANG II-induced aortic hypertrophy. This is associated with activation of redox-regulated ERK1/2 pathway that controls aortic smooth muscle cells growth. Our findings highlight an important cross-talk between the DABK and ANG II in the vascular system and contribute to a better understanding of the mechanisms involved in vascular remodeling in hypertension.

Cannabis sativa (maconha) é a droga ilícita mais conhecida e utilizada em todo o mundo. Recentemente, o uso recreativo da droga por jovens aumentou muito, especialmente entre as gestantes. Estudos indicam que seu uso na gestação causa efeitos fetotóxicos adversos, no entanto, poucos estudos avaliam esses efeitos durante este período crítico sobre o desenvolvimento do sistema imunológico. Neste estudo, avaliamos o impacto da exposição gestacional à fumaça de maconha resultante da queima de cigarros sobre o desenvolvimento do baço e do timo de proles de camundongos BalbC. Utilizamos um modelo que se aproxima das reais condições de uso humano (inalação) sob os aspectos de dose e média exposição. Camundongos fêmeas gestantes (n = 20) foram expostas ao fumo da maconha ou ao ar filtrado (grupo controle) do 5,5° ao 17,5° dia gestacional (DG) por 5 minutos diariamente. Para isso, foi utilizado um aparelho para inalação da fumaça desenvolvido em nosso laboratório, onde os cigarros de maconha (200mg de Cannabis sativa) foram queimados e a fumaça conduzida para a câmara de exposição dos animais (exposição somente inalatória). Os baços e timos das proles foram coletados no 18,5° dia gestacional (DG), 20° e 60° dias pós-natais (DPN) para a realização das análises histológica e histoquímica. Avaliações morfológicas e quantitativas dos órgãos foram realizadas utilizando-se métodos estereológicos e por meio da semi quantificação de fibras do sistema colágeno. Os resultados indicam um aumento no peso corporal dos animais com 60 DPN cujas mães foram expostas ao fumo de Cannabis sativa durante a gravidez. Os machos com 60 DPN expostos apresentaram redução no volume total do timo, assim como em seus principais compartimentos, medula e córtex, onde também foi observado um aumento de fibras do sistema colágeno, quando comparados aos animais controles. Além disso, o baço desses animais também apresentou redução significativa em seu volume, porém não houve redução significativa no volume total da polpa vermelha. Machos e fêmeas com 60 DPN do grupo exposto apresentaram redução no volume total da polpa branca e aumento de fibras do sistema colágeno na região das trabéculas do parênquima do baço. Em suma, a exposição gestacional à maconha causa efeitos fetotóxicos que podem ser observados por alteração no baço e no timo da prole (sendo os efeitos distintos para machos e fêmeas) e estas alterações se tornam mais marcantes com o avanço da idade do animal, com potencial comprometimento da resposta imune nesses indivíduos. Cannabis sativa (marijuana) is one of the most well-known and used illicit drugs worldwide. Recently, recreational use of that drug among young people has increased greatly, especially among pregnant women. Studies indicate that its use in pregnancy causes adverse fetotoxic effects, however there are few studies evaluating the effects of marijuana use during this critical period on the developing immune system. In this study, we evaluated the impact of gestational exposure to smoke marijuana resulting from burning cigarettes on the spleen and thymus development of BalbC mice offspring. We have used a model that approximates the real conditions of human use (inhalation) under the dose and medium exposure aspects. Pregnant mice (n=20), were exposed to marijuana smoke or filtered air (control group) from 5.5° to 17.5° gestational day (GD) for 5 minutes daily. Thus, we used a smoke inhalation apparatus developed in our laboratory where the marijuana cigarettes (200mg of Cannabis sativa) were burned and conducted to exposure chamber reaching the animals (nose-only exposure). Spleen and thymus of the offspring were collected on the 18.5° gestational day (GD), 20° and 60° postnatal day (PND) for the histological and histochemical analysis. Morphological and quantitative evaluations of these organs were performed using stereological methods and semi quantification of collagen fibers. The results indicate an increase in the mice body weight at PND 60 whose mothers were exposed to Cannabis sativa smoke during pregnancy. The PND 60 males from exposed group showed lower thymus total volume, as well as its main compartments such as medulla and cortex, where collagen fibers was also observed, when compared with males from the control group. Also, the spleen of these animals also presented a significant reduction, however no significant reduction was showed in the total volume of the red pulp. In addition, males and females from the exposed group at PND 60 presented reduction in total volume in the white pulp and increased of collagen fibers in the trabeculae region of the spleen. In summary, gestational exposure to marijuana causes fetotoxic effects that can be observed due to alteration in the spleen and thymus of the offspring (the effects being different for males and females), and these changes being marked with the advancement of the age, with potential impairment of the immune response in these individuals.

A Cannabis sativa, genericamente conhecida por maconha é uma das drogas ilícitas mais utilizadas no mundo. Por ser considerado inofensiva em alguns países seu uso recreativo é liberado, o que tem aumentado em grandes proporções do uso desta substância, incluindo mulheres gestantes. A gestação é um período em que ocorrem alterações dinâmicas fisiológicas consideráveis na mãe, placenta e feto e com isso, os processos de absorção, distribuição e eliminação de drogas utilizadas pode ser muito variável. O que torna a utilização de desta droga mais perigosa nesse período. Diversos desfechos gestacionais negativos estão associados a seu uso, porém pouco se sabe sobre os mecanismos envolvidos nesta associação. Assim, neste estudo avaliamos experimentalmente utilizando um modelo animal, se os desfechos gestacionais negativos podem estar associados a alterações placentárias. Resumidamente, fêmeas gestantes de camundongos foram expostas a fumaça proveniente da queima da Cannabis diariamente durante a gestação via inalação, mimetizando o uso por humanos. No 18,5° dia da gestação (a termo) foram eutanasiadas e as placentas coletadas apara avaliação estereológica. Os resultados mostram que a o consumo da cannabis em baixa dose pode provocar alterações na estrutura da placenta. Há um aumento significativo em seu volume total decorrente do aumento no volume do labirinto, além disto o volume de capilares fetais está aumentados e a barreira inter-hemal fica mais fina. Estas alterações apesar de sugerirem um mecanismo compensatório, o crescimento fetal normal não é mantido Cannabis sativa, commonly known as marijuana or pot, is one of the most used ilicit drug in the world. In some countries its recreational use is legal because this drug is not considered harmful to health. Legalization of its use has increased the number of users of this drug including pregnant women. Gestation is a period of physiological dynamic changes in the mother, fetus and placenta, thus the process of absorption, metabolism and excretion is different. Association between negative gestational and fetal outcomes and gestational exposure to marijuana is described in the medical literature, however little is known about the mechanisms involved. Taking this into account, in this study we investigated experimentally, using in a animal model that mimics human use, if the negative gestational outcomes could be associated to placental structural and functional changes. Briefly, pregnant mice were exposed during gestation to either filtered or marijuana smoke by inhalation and at gestational Day 18 (at term) they were euthanized and placentas collected for stereological analysis. Our preliminary results indicate that the use of cannabis sativa during pregnancy is associated to structural changes in the placenta. Placental total volume is increased and is due to increases in the volume of the labyrinth. In the labyrinth the volume of fetal capillaries and their surface area are increased and the interhemal membrane thickness decreased. These changes suggest a compensatory mechanism however, they fail to support the fetal normal growth.
