Hepatocellular adenoma is a rare benign liver tumor that occurs predominantly in young women who use exogenous estrogens. We describe a case of a 40-year-old woman on birth control pill who presented with acute right hypochondrial pain and signs of hemodynamical instability triggered by a large bleeding tumor in the right liver lobe. Arterial embolization was performed with cessation of bleeding. To determine etiology, magnetic resonance imaging was conducted with findings suggestive of a hepatocellular adenoma. The tumor was surgically resected, and histologic examination made the definite diagnosis of an inflammatory hepatocellular adenoma. This is a noteworthy case of a rare but potentially fatal complication of liver tumors, whose diagnosis requires a high index of suspicion. Clinicians should consider this diagnosis in young women on oral contraceptive pills who present with severe unexplained abdominal pain and hemodynamical instability. This case aimed to raise awareness of this condition, while reviewing important aspects concerning the management of hepatocellular adenomas.

A unha em pinça é uma deformidade ungueal, hereditária ou adquirida, que se manifesta como uma hipercurvatura transversal do prato ungueal, sobretudo na sua porção distal, o que provoca uma compressão do leito ungueal, podendo causar dor com incapacidade funcional, assim como infecções secundárias. O tratamento conservador associa-se a uma elevada taxa de recorrência, motivo pelo qual a reconstrução cirúrgica é geralmente necessária. Descrevem-se quatro casos clínicos exemplares de diferentes opções corretivas da unha em pinça.

A reconstrução de defeitos que envolvam até 2/3 da extensão dos lábios é desafiadora, levando-se em consideração a importância funcional e estética desta estrutura. Existem diversas técnicas cirúrgicas descritas para esta finalidade, salientando-se o retalho de Abbe-Estlander, o retalho de Karapandzic e o retalho de Bernard-Burrow-Webster. A escolha da melhor abordagem deve levar em conta a dimensão e localização do defeito e as comorbidades do doente. O retalho de Yu constitui uma alternativa a ser considerada em relação a outros métodos, permitindo obter geralmente excelentes resultados cosmético-funcionais.

Traditional farmers’ varieties of tomato grown under extensive farming techniques are considered delicious and healthy foods and are preferred by local consumers. Tomatoes are an important component of a healthy diet, as they provide essential micronutrients, including minerals, which are vital to healthy development, disease prevention, and wellbeing. Given the considerable dietary intake of tomatoes and the scarcity of information about the bioaccessibility of inorganic constituents in this fruit, this study was carried out to evaluate the content and bioaccessibility of minerals (macro- and microelements) in tomato farmers’ varieties widely cultivated in northeastern Portugal homegardens. Among the macroelements, K stood out as the most abundant mineral in the studied varieties, followed by Mg, Ca, and Na. Regarding the microelements, while the yellow tomato had higher concentrations of Fe and Cu, the round tomato had more Zn and Mn. The in vitro bioaccessibility assessment showed that, among the macroelements, Mg was more bioaccessible than Ca and K when all the tomato varieties were considered together. Among the microelements, Cu seemed to be the most bioaccessible. Although the contribution of a 100 g serving of the studied tomato farmers’ varieties to the dietary reference intakes (DRIs) of minerals is relatively low, this food could contribute to reaching these mineral requirements, as it is included in the diet of most of the population, especially in Mediterranean regions.

Background: The role played by Helicobacter pylori in the sinuses, and its association with the same organism’s gastric infection, are still unclear. Methods: In order to compare H.pylori colonization patterns in the nose and stomach we conducted a cohort analysis of 14 patients, eligible for sinus surgery due to chronic medically refractory rhinosinusitis, who were tested for simultaneous presence of H. pylori, by histology, culture and polymerase chain reaction, in pathologic sinus tissue collected during surgery and in gastric mucosa obtained through gastroduodenal endoscopy. Results: H. pylori DNA was found in the sinus mucosa of 15.4% of patients with chronic rhinosinusitis, and all of them showed concurrent H. pylori stomach infection. Sinus colonization was not found without simultaneous gastric colonization, although most patients with gastric infection did not have the bacterial DNA in their sinuses. H. pylori’s presence in the nose was not associated with local inflammatory status, and no cultures could be obtained from any of the sinus tissue samples, including those positive for H. pylori DNA. Conclusions: Only H. pylori DNA, and not the culturable active form of the microorganism, could be found in the sinus mucosa of some patients with H. pylori gastric infection. We could not find evidence, however, that the bacterium’s presence in the nose contributes to local mucosal inflammation.

Ruscus aculeatus L. is a subshrub used in traditional medicine in different parts of the world, namely in Europe and the Iberian Peninsula. According to reported folk knowledge, the aerial parts are mainly used as diuretics and the underground organs are used for the treatment of disorders of the urinary system and as a laxative. In this work, the aerial part and the roots and rhizomes of R. aculeatus were chemically characterized with regard to the content of phenolic compounds and bioactive properties. Aqueous (infusions and decoctions) preparations and hydroethanolic extracts from the two mentioned parts of the plant were prepared. Nine phenolic compounds were detected in all the extracts. Apigenin-C-hexoside-C-pentoside isomer II was the major compound in aqueous extracts and, in the hydroethanolic extract was quercetin-O-deoxyhexoside-hexoside followed by apigenin-C-hexoside-C-pentoside isomer II. All extracts revealed antioxidant activity and potential to inhibit some of the assayed bacteria; aqueous extracts of the aerial part and infusions of roots and rhizomes did not show cytotoxic effects on a non-tumor primary cell culture. This preliminary study provides suggestions of the biological potential associated with the empirical uses and knowledge of this species, in particular its bioactivities.

This study, of theoretical and documentary nature, aims to investigate the role of school councils in Brazilian municipalities. Therefore, we analyzed the construction of the democratic process in Brazil, as well as the progress of popular participation in these bodies, composed by representatives of the school community. In this context, we can highlight the municipality of Frederico Westphalen, Rio Grande do Sul, tracing possibilities and challenges of local social control. It is possible to observe that, despite the creation of the first school councils have taken place in the early 1990s, only in 2003 they became a reality in the city. Many of these boards come up against bureaucracy and lack of knowledge of their mission by its components. Despite the small number of investigations about it, it can be concluded that it is necessary to develop the relationship between theory and practice, so that the school councils are able to carry out their role of bringing the community into the school effectively

Tabebuia impetiginosa (Mart. ex DC.) Standl. has been used in traditional medicine for many centuries, being nowadays marketed as dried plant material (inner bark) for infusions, pills, and syrups. The main objective of the present work was to validate its popular use through the bioactivity evaluation of the inner bark (methanolic extract and infusion) and of two different formulations (pills and syrup) also based on the same plant-material. The antioxidant activity was evaluated by in vitro assays testing free radical scavenging activity, reducing power and inhibition of lipid peroxidation in brain homogenates. The cytotoxicity was determined in four human tumor cell lines (MCF-7, NCI-H460, HeLa and HepG2, and also in non-tumor cells (porcine liver primary cells, PLP2)). Furthermore, the sample was chemically characterized regarding free sugars, organic acids, fatty acids, and tocopherols. Syrup and methanolic extract showed the highest antioxidant activity, related to their highest amount of phenolics and flavonoids. Methanolic extract was the only sample showing cytotoxic effects on the tested human tumor cell lines, but none of the samples showed toxicity in PLP2. Glucose and oxalic acid were, respectively, the most abundant sugar and organic acid in the sample. Unsaturated predominated over the saturated fatty acids, due to oleic, linoleic, and linolenic acids expression. α- and γ-Tocopherols were also identified and quantified. Overall, T. impetiginosa might be used in different phytoformulations, taking advantage of its interesting bioactive properties and chemical composition.

This paper sheds light on the paths of third language (L3) acquisition of Portuguese by Spanish–English speakers whose first language is Spanish (L1 Spanish), English (L1 English), or both in the case of heritage speakers of Spanish (HL). Specifically, it looks at the gradual acquisition of a categorical rule in Portuguese, where some prepositions are invariably contracted with the determiner that follows them. Based on a corpus of 1910 written assignments by Portuguese L3 learners, we extracted 21,879 tokens in obligatory contraction contexts and submitted them to a multivariate analysis. This analysis allowed for the investigation of the impact of linguistic (type of preposition and definite article number and gender) and extra-linguistic factors (course level and learner’s language background), with logistic regression modeling with sum contrasts and individual as a random effect. While results point to some clear similarities across the three language groups—all learners acquired the contractions in a u-shaped progression and used more contractions with the a preposition and fewer with the por preposition—participants acquire contractions at a higher rate when the article is singular than when it is plural, and in the case of HL speakers, more so when the article is masculine than when it is feminine. These results confirm the facilitatory role of a previously acquired language (i.e., Spanish) that is typologically similar to the target language (i.e., Portuguese) in transfer patterns during L3 acquisition.

Advances in gene therapy have been foreshadowing its potential for the treatment of a vast range of diseases involving genetic malfunctioning. However, its therapeutic efficiency and successful outcome are highly dependent on the development of the ideal gene delivery system. On that matter, silica-based vectors have diverted some attention from viral and other types of non-viral vectors due to their increased safety, easily modifiable structure and surface, high stability, and cost-effectiveness. The versatility of silane chemistry and the combination of silica with other materials, such as polymers, lipids, or inorganic particles, has resulted in the development of carriers with great loading capacities, ability to effectively protect and bind genetic material, targeted delivery, and stimuli-responsive release of cargos. Promising results have been obtained both in vitro and in vivo using these nanosystems as multifunctional platforms in different potential therapeutic areas, such as cancer or brain therapies, sometimes combined with imaging functions. Herein, the current advances in silica-based systems designed for gene therapy are reviewed, including their main properties, fabrication methods, surface modifications, and potential therapeutic applications.

Several plants have been used for medicinal applications and have been traditionally consumed as decoctions and infusions. Although some herbs are used alone as a beverage, they are often blended in mixtures to maximize their effects. Herein, the nutritional characterization of six infusions from herbal blends was evaluated using the official methods of analysis (AOAC international). A further characterization of the individual phenolic profile was also performed by HPLC-DAD/ESI-MSn, and finally bioactive potential was determined by evaluating the antioxidant, cytotoxic, anti-inflammatory, and antimicrobial activities of each blend. The wide variety of plants in each sample led to variability in the results for all analyzed parameters. However, blends containing 15% Laurus nobilis L. and 15% Juglan regia L. in their composition showed higher sugar content and energy contribution; higher concentration of phenolic compounds (phenolic acids and flavonoids); greater antioxidant, cytotoxic, and anti-inflammatory capacity; and also better antimicrobial effects against all the tested bacterial and fungal strains. Further studies will be necessary to evaluate the real synergistic effects that these two species show in the presence of other plants, and to evaluate their potential for application in various food, pharmaceutical, and nutraceutical products as infusion preparations.

This article discusses the contributions made by variationist sociolinguistics to the study of languages in contact. After summarizing the development of this subfield and its theoretical and methodological implications, 3rd p.sg. subject pronoun expression is examined in Portuguese and Spanish in contact in Northern Uruguay. Based on comparative sociolinguistics, the results show that this variable use among bilinguals does not converge into a single system. Lastly, it calls for more studies in Portuguese dialects in contact with other languages that follow the framework of variationist sociolinguistics.

Garlic (Allium sativum L.) has been used worldwide not only for its being a subject of dietary interest, but also for medicinal purposes, in prophylaxis, and for the treatment of diverse pathologies. New processing techniques have been developed and placed on the market in recent years to improve the organoleptic and nutritional value of food products. The present work aimed to study bulbils (cloves) of white (commercial and traditionally cultivated samples with different proveniences) and black (processed samples) garlic. All samples were compared with regard to their nutritional composition as well as their antioxidant and antimicrobial activities. Black garlic had the lowest moisture content but the highest total amount of sugars and energetic value. Black garlic also presented the highest antioxidant and antimicrobial (especially against methicillin-resistant Staphylococcus aureus) activities. Thus, black garlic, obtained by processing techniques, can be considered a promising product with high value that will be able to be exploited by the functional food/nutraceutical industry.

The bioactive properties (antioxidant and antitumour activities, and hepatotoxicity) of the infusion and methanolic extracts of Chenopodium ambrosioides L., a plant commonly used in Portuguese folk medicine, were compared. The chemical composition in hydrophilic (sugars, organic acids and phenolic compounds) and lipophilic (fatty acids and tocopherols) fractions were determined. In general, the infusion revealed higher antioxidant activity, while the methanolic extract was the only one showing antitumour effects against colon, cervical and hepatocellular carcinoma cell lines. No toxicity in non-tumour cells was observed either for the infusion or the extract. The studied plant proved to be a good source of natural antioxidants and other bioactive compounds, which may have industrial use. As far as we know, this is the first detailed chemical characterization and bioactivity evaluation of C. ambrosioides methanolic extract and infusion.

Este artigo tem como objetivo discutir alguns fatores sociolinguísticos relevantes ao ensino de português nas comunidades bilíngues em espanhol e português no norte do Uruguai. Nele, destaco duas das características do repertório linguístico dessas comunidades: o bilinguismo e suas consequências nas variedades em contato, como a alternância de códigos e a congruência gramatical; e os padrões de variação interna, tanto do português como do espanhol fronteiriço. Após contribuir com uma descrição do repertório bilíngue e multidialetal nessas comunidades e descartar o mito de monolinguismo e monoestilismo, discuto algumas premissas que devem ser consideradas pelo ensino do português nessas comunidades, explorando algumas sugestões pedagógicas oferecidas no campo de ensino de língua materna e de língua de herança. O objetivo final é contribuir ao desenvolvimento de uma pedagogia sensível às diferenças sociolinguísticas e culturais dos alunos residentes na fronteira.This article is aimed at discussing sociolinguistic factors that are relevant to the teaching of Portuguese in the Spanish-Portuguese bilingual communities of northern Uruguay. It explores two of the main characteristics of these communities' linguistic repertoire: bilingualism and its consequences on the contact varieties, such as code-switching and grammatical convergence and patterns of internal linguistic variation, both in Portuguese and in Spanish. After contributing with a description of these communities' bilingual and multidialectal repertoires and arguing against the idea of monolingual and monostylistic speakers, the text brings a discussion on some premises that should be considered in the teaching of Portuguese in these communities, borrowing some pedagogical suggestions from the fields of first- and heritage-language teaching. Its final objective is to contribute to the development of a set of pedagogical principles that take students' sociolinguistic and cultural differences into account.

This paper, has been elaborated based on ducumental sources,interviews, images and other cultural artifacts, focuses on theproposals of male Catholic Congregations, with special emphasis(focus) on those that promoted rural teachers education, in the midtwentieth century, in the Federal State of Rio Grande do Sul. Itdiscusses the "integral formation of the person" in face of the role of sport in rural normal schools, as well as in reference to the founding principles of the Congregations. It is possible to assert sports practice actuated as a mediating element as well as of integration of the groups and communities, contributing to the evangelizing mission, through the occupation of free time, disciplining the conduct and the conformation to norms an rules. We analize three educational institutions (Rural Normal School of the Archdiocese, Rural Normal School La Salle, and Rural Normal School Murialdo), being demonstrated greater evidence of sports practice by the Rural Normal School La Salle of Cerro Largo. This school, once transformed into a School of Basic Education, abandoned the rural thematics of teacher education, but it kept the preoccupation with sports practice

Esta investigação propõe-se a verificar a ocorrência da Síndrome de Burnout em Agentes Comunitários de Saúde, visto que esses profissionais trabalham diretamente no cuidado a outras pessoas - característica prevalente em tal Síndrome -, bem como as estratégias de enfrentamento utilizadas. Optou-se pelo método descritivo, transversal e quantitativo. Foram aplicados o questionário Maslach Burnout Inventory (MBI) e a Escala Modos de Enfrentamento de Problemas (EMEP). A amostra constituiu-se por 80 ACS, escolhidos aleatoriamente entre aqueles admitidos há pelo menos dois anos no serviço público municipal. Verificou-se que se trata de uma amostra predominantemente do sexo feminino (93,75%), com filhos (68,75%), na faixa etária de 20 a 30 anos (35%) e com companheiro fixo (60%). A renda per capita de 38,75% é de menos de um salário mínimo; 53,04% estão nesse trabalho há mais de seis anos e relatam ter vida sedentária. O MBI revela um sentimento de deterioração da percepção da própria competência dos profissionais e, também, falta de satisfação com o próprio trabalho. Eles demonstram estar emocionalmente esgotados e verifica-se o desenvolvimento incipiente de sentimentos e atitudes negativas, além de cinismo para com as pessoas por eles atendidas. O Modo de Enfrentamento mais utilizado são as estratégias focalizadas no problema, seguidas pela busca de práticas religiosas/pensamento fantasioso. Verificam-se indícios de sofrimento característicos da Síndrome de Burnout, com o esforço de buscar mecanismos que auxiliem no enfrentamento de problemas relacionados ao trabalho. Sugerem-se estratégias de acolhimento a esses profissionais, de forma a auxiliá-los a lidar com o sofrimento no trabalho.This research aims to verify the occurrence of the Burnout Syndrome in Community Health Agents (CHA). These professionals work directly in care delivery to other people, and this is a prevalent characteristic in this Syndrome. Another aim is to verify the coping strategies such professionals use. A descriptive, cross-sectional and quantitative study was carried out. A questionnaire - the Maslach Burnout Inventory (MBI) - and the Ways of Coping Scale (WOCS) were administered. The sample consisted of 80 CHA, randomly chosen among professionals who had been working for at least two years in the municipal public service. The results show that the sample was predominantly female (93.75%), with children (68.75%), belonging to the age group 20-30 years old (35%) and with a steady partner (60%). The per capita income of 38.75% is less than one minimum salary, 53.04% have been working in this job for more than six years and lead a sedentary life. The MBI reveals a feeling of deterioration in the perception of these professionals' own competence and lack of satisfaction with their own work. They present emotional exhaustion, and the incipient development of negative attitudes and cynicism towards the people they deliver care to were noticed. The most used coping strategies are problem-focused ones, followed by the search for religious practices/fantastic thinking. Symptoms of suffering which are typical of the Burnout Syndrome, as well as the effort to seek for mechanisms that help to cope with work-related problems, were observed. Strategies to welcome these professionals are suggested so as to help them deal with suffering at work.

En las últimas décadas, las regiones montañosas de Portugal han sufrido transformaciones económicas y una intensa disminución de la población que conllevaran modificaciones visibles de los agroecosistemas y en consecuencia del paisaje. Un estudio de caso, conducido en un espacio natural protegido de la provincia portuguesa de Trás-os-Montes (2006-2007) - el Parque Natural de Montesinho - y del ámbito del proyecto de investigación ";Etnobotãnica do Nordeste Portugués: saberes, plantas e usos";, pretende comprobar que los conocimientos sobre plantas, sus usos y correspondiente manejo son la expresión de cambios culturales inherentes a estas dinámicas sociales y ecológicas. En un contexto social enmarcado por una ";nueva ruralidad"; (es decir, un modo diferente de encarar y vivir el mundo rural), coexisten conocimientos y usos tradicionales de plantas, muchos de raíz ancestral, y saberes adquiridos más recientemente e incorporados en la cultura local. El análisis de los conocimientos por género, edad, grado de escolaridad, experiencia migratoria de los informantes, permite enfocar y evaluar la pérdida de la memoria social y cultural. Los resultados que se presentan remiten aún para la discusión conceptual de los conocimientos ecológicos y tradicionales. Además, conllevan a una reflexión acerca de la preservación de estos saberes/prácticas y de la conservación de la naturaleza en el contexto de espacios naturales protegidos.

Pterospartum tridentatum (L.) Willk, Gomphrena globosa L., and Cymbopogon citratus (DC.) Stapf are medicinal plants that require a more detailed chemical characterization, given the importance of their consumption as infusions. Therefore, the individual profiles in tocopherols, free sugars, and organic acids were obtained by high performance liquid chromatography (HPLC) coupled to different detectors (fluorescence, refraction index, and photodiode array, resp.). C. citratus revealed the highest content of α-, and total tocopherols, glucose, sucrose, succinic, and ascorbic acids. P. tridentatum presented the highest fructose and total sugars content. Otherwise, G. globosa showed the highest organic acids concentration. As far as we know, this is the first study reporting the mentioned chemical compounds in G. globosa and C. citratus.

This study addresses the effects of gamma irradiation (1, 5 and 8 kGy) on color, organic acids, total phenolics, total flavonoids, and antioxidant activity of dwarf mallow (Malva neglecta Wallr.). Organic acids were analyzed by ultra fast liquid chromatography (UFLC) coupled to a photodiode array (PDA) detector. Total phenolics and flavonoids were measured by the Folin-Ciocalteu and aluminium chloride colorimetric methods, respectively. The antioxidant activity was evaluated based on the DPPH• scavenging activity, reducing power, β-carotene bleaching inhibition and thiobarbituric acid reactive substances (TBARS) formation inhibition. Analyses were performed in the non-irradiated and irradiated plant material, as well as in decoctions obtained from the same samples. The total amounts of organic acids and phenolics recorded in decocted extracts were always higher than those found in the plant material or hydromethanolic extracts, respectively. The DPPH• scavenging activity and reducing power were also higher in decocted extracts. The assayed irradiation doses affected differently the organic acids profile. The levels of total phenolics and flavonoids were lower in the hydromethanolic extracts prepared from samples irradiated at 1 kGy (dose that induced color changes) and in decocted extracts prepared from those irradiated at 8 kGy. The last samples also showed a lower antioxidant activity. In turn, irradiation at 5 kGy favored the amounts of total phenolics and flavonoids. Overall, this study contributes to the understanding of the effects of irradiation in indicators of dwarf mallow quality, and highlighted the decoctions for its antioxidant properties.

Le test ELISA-TÉTANOS (Biosys, France) a été utilisé pour le titrage des anti-corps tétaniques (sensibilité = 0,0025 UI/ml) en sérums humains de donneurs de sang, 566 hommes et 108 femmes, âges de 18 à 58 ans, moyenne de 29 ans, provenant de São Paulo, SP, Brésil. L'OMS, acceptant seulement la séroneutralisation sur souris (NT), la méthode de référence, pour les études sur la protection contre le tétanos, préconise le titre de 0,01 UI/ml comme minimum protecteur. BOURLEAUD & HUET ont proposé la limite de 0,06 UI/ml quand s'emploie le test ELISA, en attendant à une certaine discordance inévitable entre les méthodes. Parmi les 674 sérums étudiés, 178 (26,41%) n'ont pas présenté d'anticorps (< 0,0025 UI/ml); 413 (61,28%) ont présenté des résultats égaux ou supérieurs à 0,01 UI/ml et en 232 (34,42%) les titres ont été égaux ou supérieurs à 0,06 UI/ml. Le pourcentage d'individus protégés a été inversement proportionnel à 1'âge: environ 50% dans le groupe le plus jeune (hommes de 18 à 35 ans et femmes de 18 à 23 ans) contre environ 10% dans le groupe de plus de 42 ans ont présenté des titres sûrement protecteurs (> 0,06 UI/ml).

Realizou-se estudo comparativo de diferentes sorotipos de Leptospira interrogans utilizados na preparação de antígenos empregados no teste ELISA, para a detecção de anticorpos da classe IgM, em amostras de soro na fase precoce e tardia da leptospirose humana. Foram utilizados dez sorotipos, escolhidos entre os que apresentaram maior reatividade na soroaglutinação microscópica (SAM), na cidade de São Paulo. Os cinco sorotipos que apresentaram melhores resultados individualmente no teste ELISA-IgM (canicola, hebdomadis, icterohaemorrhagiae, cynopteri e brasiliensis), foram também estudados em mistura antigênica. Os antígenos não tratados apresentaram maior reatividade do que os antígenos tratados com Triton X - 100 (4%) à temperatura de 50ºC, durante 4 horas. O teste ELISA-IgM empregando os sorotipos não tratados, isoladamente, e em mistura antigênica, mostrou-se altamente sensível, podendo ser empregado como teste de triagem para o diagnóstico precoce da leptospirose humana. Outra aplicação do teste é permitir a detecção do início de situações epidêmicas ou de surtos, possibilitando acionar medidas de vigilância epidemiológica.A comparative study among different serovars of Leptospira interrogans was performed in order to prepare antigens to detect IgM antibodies by ELISA in early and late phase of human leptospirosis. Ten serovars were chosen among the most prevalent detected by microscopic seroagglutination (SAM) in São Paulo city. Using ELISA-IgM five of them showed better results (canicola, hebdomadis, icterohaemorrhagiae, cynopteri and brasiliensis). These ones were also studied in a pool. The non-treated antigens showed higher reactivity than the Triton X-100 (4%/50ºC/4h). ELISA-IgM using individually or pool of non treated antigens proved to be reliable with high sensitivity and should be used for an earlier diagnosis of leptospirosis, as a trial test. Faster diagnostic elucidation can be useful to detect epidemic situations, so, allowing epidemiological surveillance interventions.

Foi empregado o teste imunoenzimático com componentes antigênicos de Cysticercus cellulosae quimicamente ligados a suporte inerte constituído por discos de tecido-resina, ELISA-d, com a finalidade de investigar a entidade neurocisticercose (NC) em líquidos cefalorraquianos (LCR) de pacientes com meningites de etiologia indeterminada. Foram ensaiados 277 LCR de 128 crianças e 149 adultos. A densidade óptica média (DO) obtida para os 22 LCR de pacientes nos quais foi afastada a possibilidade diagnóstica de meningite foi de 0,03. Os 44 LCR de pacientes com meningites determinadas por diversos agentes etiológicos, não cisticercose, apresentaram DO de 0,05. O limiar de reatividade do teste ELISA-d calculado a partir desses dois grupos (controle) foi de 0,13 (DO + 3SD). No grupo de 13 LCR de pacientes com NC comprovada em episódio meningítico por essa causa, foi observada DO de 0,41 (0,10 a 0,91) no teste ELISA-d. Dos 198 LCR de meningites por agente etiológico não identificado pelos métodos usualmente empregados, 23 (11,6%) apresentaram DO acima de 0,13, sugerindo que a possível causa da meningite tenha sido por cisticercose, uma vez que o teste ELISA-d tem apresentado elevadas sensibilidade, especificidade e reprodutibilidade. Em cinco dos 23 LCR a alteração no exame quimiocitológico era às custas do aumento do número de células predominantemente linfomononucleares, em 13 o predomínio era de polimorfonucleares e nos cinco restantes ambos os tipos de células estavam em número aumentado.The enzyme-linked-immunosorbent assay using a new type of support consisting of discs of synthetic fabric-resin (ELISA-d) with antigenic components of Cysticercus cellulosae covalently bound, was employed with the purpose of investigating the presence of specific antibodies to the cysticerci (neurocysticercosis) in the cerebrospinal fluid (CSF) of patients with meningitis without etiological agent determined by the conventional methods. The test was performed in 277 CSF samples (128 children and 149 adults). The mean of optical density values (OD) obtained for 22 CSF normal patients (the diagnostic hyphotesis of meningitis was discarded) was of 0.03. The 44 CSF of patients with meningitis caused by other agents but C. cellulosae, showed OD of 0.05. The cut off determined with these two groups (control group) was 0.13 (OD + 3 SD). The group of 13 CSF of neurocysticercotic meningitis presented OD of 0.41 (0.10 to 0.91). Among the 198 CSF samples of indetermined meningitis, 23 (11.6%) presented OD above the cut off, which suggests the possibility that Cysticercus cellulosae was the etiological agent in this meningitis episode. The ELISA-d test has proved to be efficient for the immunodiagnosis of neurocysticercosis when conducted on CSF samples. The alterations observed in the 23 CSF reactive on ELISA-d were: increased number of lymphomononuclear cells in five, of polymorphonuclear cells in 13 and both cells in five samples.

An enzyme-linked immunosorbent assay (ELISA), employing antigens from Toxocara canis larvae and the absortion of suspected sera with Ascaris lumbricoides extracts was used in a seroepidemiological study performed in five municipalities of São Paulo State, Brazil (São Paulo, Campinas, Santos, Marília and Presidente Prudente) in order to determine the frequency of antibodies to Toxocara. In 2,025 blood samples collected, 806 proceeded from male subjects and 1,219 from females; 483 samples were collected from subjects under 15 years of age and the remaining 1,542 from subjects aged 15 years or over. Among the 2,025 sera investigated, 3.60% had antibodies to Toxocara at significant levels. A moderate predominance of infection with Toxocara among male subjects (3.72%) was observed, although the difference was not statistically significant when this rate was compared with that for female (3.28%). Related to age, a higher frequency of positive results was detected among subjects under 15 years (6.41%) against the older group (2.53%). A trend of more elevated rates of infection was observed in municipalities with high demographic densities (São Paulo, Campinas and Santos). Nevertheless, such findings only appeared to be statistically significant in subjects younger than 15 years.Realizou-se inquérito soroepidemiológico, para detecção de anticorpos anti-Toxocara, em 2.025 soros de indivíduos residentes em cinco municípios do Estado de São Paulo (São Paulo, Campinas, Santos, Marília e Presidente Prudente), através de técnica imunoenzimática (ELISA) e emprego de antígenos obtidos a partir de larvas de 2º - 3º estádio de Toxocara canis. Após absorção com extratos de Ascaris lumbricoides, revelou-se a presença de anticorpos anti-Toxocara, em níveis significativos, em 3,60% dos soros examinados, com predominância de indivíduos com idade inferior a 15 anos (6,41%). Observou-se, ainda, tendência a ocorrerem índices mais elevados de infecção por larvas de Toxocara entre indivíduos residentes em municípios com maior densidade demográfica (São Paulo, Campinas e Santos), especialmente no grupo com menos de 15 anos de idade.

Relata-se a concomitância de leptospirose e síndrome da imunodeficiência adquirida (SIDA) em humanos. Nestes casos não foram observados aspectos clínicos diferentes que possam ser atribuídos à simultaneidade dessas patologias.

Estudamos comparativamente quatro técnicas imunológicas para o diagnóstico da neurocisticercose (NC) utilizando líquido cefalorraquiano (LCR) como espécime clínico: as reações de fixação de complemento (RFC), hemaglutinação passiva (RHA), imunofluorescência indireta (RIF) e teste imunoenzimático ELISA Foram ensaiadas 125 amostras de LCR de pacientes com NC comprovada e 94 amostras de LCR do grupo controle (60 de pacientes com quadros clínicos neurológicos diversos e 34 de pacientes supostamente normais). Os índices de sensibilidade e especificidade obtidos para os testes foram, respectivamente, de 48,0% e 90,4% para a RFC; 88,8% e 96,8% para a RHA; 87,2 e 98,9% para a RIF e 97,6% e 98,9% para o teste ELISA. A diferença significativa (p < 0,05) observada entre os testes permite concluir que o melhor teste para o diagnóstico de NC foi o teste ELISA seguido das reações de HA e IF.

A passive haemagglutination test (PHA) for human neurocysticercosis was standardized and evaluated for the detection of specific antibodies to Cysticercus cellulosae in cerebrospinal fluid (CSF). For the assay, formaldehyde-treated group O Rh-human red cells coated with the cysticerci crude total saline extract (TS) antigen were employed. A total of 115 CSF samples from patients with neurocysticercosis was analysed, of these 94 presented reactivity, corresponding to 81.7% sensitivity, in which confidence limit of 95% probability (CL95%) ranged from 74.5% to 88.9%. Eighty-nine CSF samples derived from individuals of control group presented as nonreactive in 94.4% (CL95% from 89.6% to 99.2%). The positive and negative predictive values were 1.4% and 99.9%, respectively, considering the mean rate of that this assay provide a rapid, highly reproducible, and moderately sensitive mean of detecting specific antibodies in CSF samples.

Foi avaliado o teste imunoenzimático (ELISA), para detecção de anticorpos da classe IgM na leptospirose humana. Nas amostras de soros analisadas, o teste ELISA demonstrou ser mais sensível, específico e precoce, quando comparado ao teste de soroaglutinação microscópica. A análise dos resultados obtidos nesta avaliação demonstra que o teste ELISA permite detectar níveis baixos de anticorpos circulantes, e também anticorpos não aglutinantes. não detectáveis através do teste de soroaglutinação microscópica.

A comparison of two different standardized reagent procedures for the passive haemagglutination test (PHA) in the detection of specific antibody to Cysticercus cellulosae in cerebrospinal fluid (CSF) was carried out. The formaldehyde-treated group O Rh-human red blood cells (HuRBC) and glutaraldehyde-treated sheep red blood cells (SRBC) were the supplies for the reagents preparation and, in the tests, they were designated as PHA-1 and PHA-2, respectively. For both reagents the cells were coated with the cysticerci total saline extract (TS) antigen. PHA-1 and PHA-2 were assessed in a total of 204 CSF from patients with neurocysticercosis, from non-related infections and from healthy individuals. The positivity and specificity indices obtained were respectively 81.7% and 94.4% for PHA-1 and for PHA-2, 88.7% and 96.6%. Since no significant differences were observed between the results provided by two reagents, at level of significance of 0.05, either processes of cell sensitization can alternatively be used according to the own laboratory convenience.

The contact with dogs at home or place of work has been investigated as factors of risk in the occurrence of the visceral larva migrans syndrome caused by Toxocara, in man. Through the E.L.I.S.A. (enzyme-linked immunosorbent assay) technique, the presence of antibodies to Toxocara was searched in the sera of 79 women who have been raising or had raised dogs at home in the last two years and 123 men, who were municipal public employees in charge of the capture and keeping of stray dogs. The control groups were constituted by 205 sera from women who denied domiciliary contact with dogs, at least in the last two years, and 139 sera from men whose occupation did not urge them to contact with dogs. A significant more elevated frequency of antibodies to Toxocara was observed among women with domiciliary contact with dogs; nevertheless, there was not a significant difference in the positive rates in the case of men with occupational contact with dogs.
