3. Rastreio audiológico no concelho da Amadora

Introdução: O concelho da Amadora localiza-se na Área Metropolitana de Lisboa e a população com origem em Cabo-Verde, Angola e Guiné-Bissau, contribui para a grande densidade populacional deste concelho, 7.195 habitantes por km2 (CMA, 2023). No início da aprendizagem formal da leitura e da escrita é necessária uma audição clara para que a associação fonema-grafema-fonema seja aprendida de modo correto (Serrano et al., 2018). Objetivos: Identificar alterações audiológicas, ainda numa fase assintomática, em crianças do 1º ano do 1º ciclo que frequentavam as 27 escolas do ensino básico do Concelho da Amadora. Material e Métodos: Realizou-se um rastreio audiológico a 1148 crianças com idades compreendidas entre os 5 e os 14 anos (922 com 6 anos) sendo 50,8 % do sexo masculino. Todas as crianças tinham o consentimento do responsável legal para a realização do rastreio. No rastreio foi realizado a otoscopia, o timpanograma e o ouve/não ouve nas frequências de 1, 2 e 4 kHz 20dB. Resultados: Das 1148 crianças não se conseguiu rastrear 5 crianças e 17 não se conseguiu completar todos os testes que compõem o rastreio, tendo-se considerado que passavam o rastreio sempre que os testes conseguidos estavam dentro da normalidade. Das crianças rastreadas, 17,24% das crianças (197) não passaram o rastreio, sendo encaminhadas para o médico ORL 10,24% das crianças e para o médico de família 7%. As alterações que mais motivaram o encaminhamento foram as alterações bilaterais do timpanograma com 32,48% de crianças encaminhadas, seguidas das alterações bilaterais do timpanograma em simultâneo com alterações no rastreio de audição com 17,09% de crianças encaminhadas. Conclusões: Este rastreio permitiu a 17,24% das crianças o encaminhamento correto de modo a proceder ao diagnóstico e intervenção precoces, antes mesmo da sua manifestação clínica. Possibilitou, ainda, a sensibilização da comunidade académica, da autarquia e da sociedade em geral para a necessidade da promoção da saúde auditiva com a implementação de programas de rastreio audiológico em idade escolar e pré-escolar de modo a reduzir o impacto da perda auditiva no desenvolvimento global da criança com destaque para os processos de ensino/aprendizagem.

Entrepreneurship education, as an emerging scientific field, has undergone significant evolution at the conceptual and praxis levels. The concept of entrepreneurship was considered from a broad perspective as the capacity to act upon opportunities and ideas, thereby creating social, cultural, or financial value in diverse contexts. The study and cultivation of this competence are highly relevant to enhancing employability and equipping young people with the skills, knowledge, and attitudes necessary within an entrepreneurial culture for the exercise of active citizenship. This study aims to consolidate the concept and analyse the pedagogical approaches in entrepreneurship education, particularly focusing on experiential learning. It also explores the most frequently used instructional methods in higher education programmes. The research is based on a comprehensive literature review, complemented by a case study of the entrepreneurship education programme implemented at the University of Lisbon, in Portugal, enriched by data collected from one of its curricular units. Experiential learning emerges as a valid approach in entrepreneurship teaching, especially when teachers effectively combine practical experience with theory while learners assume the shared responsibility of learning from experience. As the most recommended instructional methods, one can point to collaborative pedagogical models, namely problem-based learning, project-based learning, peer assessment, design thinking, formative feedback, service learning, and active methodologies that integrate digital technologies.

5. Pyruvate dehydrogenase complex deficiency: updating the clinical, metabolic and mutational landscapes in a cohort of Portuguese patients

Abstract Background The pyruvate dehydrogenase complex (PDC) catalyzes the irreversible decarboxylation of pyruvate into acetyl-CoA. PDC deficiency can be caused by alterations in any of the genes encoding its several subunits. The resulting phenotype, though very heterogeneous, mainly affects the central nervous system. The aim of this study is to describe and discuss the clinical, biochemical and genotypic information from thirteen PDC deficient patients, thus seeking to establish possible genotype–phenotype correlations. Results The mutational spectrum showed that seven patients carry mutations in the PDHA1 gene encoding the E1α subunit, five patients carry mutations in the PDHX gene encoding the E3 binding protein, and the remaining patient carries mutations in the DLD gene encoding the E3 subunit. These data corroborate earlier reports describing PDHA1 mutations as the predominant cause of PDC deficiency but also reveal a notable prevalence of PDHX mutations among Portuguese patients, most of them carrying what seems to be a private mutation (p.R284X). The biochemical analyses revealed high lactate and pyruvate plasma levels whereas the lactate/pyruvate ratio was below 16; enzymatic activities, when compared to control values, indicated to be independent from the genotype and ranged from 8.5% to 30%, the latter being considered a cut-off value for primary PDC deficiency. Concerning the clinical features, all patients displayed psychomotor retardation/developmental delay, the severity of which seems to correlate with the type and localization of the mutation carried by the patient. The therapeutic options essentially include the administration of a ketogenic diet and supplementation with thiamine, although arginine aspartate intake revealed to be beneficial in some patients. Moreover, in silico analysis of the missense mutations present in this PDC deficient population allowed to envisage the molecular mechanism underlying these pathogenic variants. Conclusion The identification of the disease-causing mutations, together with the functional and structural characterization of the mutant protein variants, allow to obtain an insight on the severity of the clinical phenotype and the selection of the most appropriate therapy.

6. Digital technologies integration in teacher education: the active teacher training model

The objective of this research was to identify the theoretical and practical bases that contribute to a model that will allow the implementation of an innovative teaching-learning model for the integration of digital technologies in teacher education. This model of teacher training, based on identified pedagogical trends, was characterised by a flexible approach to the training process, including active training strategies that encourage the acquisition of diversified skills, including digital. This approach can also transfer to students skills which enable them to take responsibility for their learning and creation of their own knowledge. The research method used was two-fold: i) action research in the development of training workshops in an in-service research training project and ii) a case study in a pre-service teacher education study, in Portugal. It was found in this study that the participating teachers were able to develop skills and integrate digital technologies in their own teaching-learning process and could change their teaching practices, which will support the development of online education in the future.

Technological skills development is a central issue for a country’s educational and social policies. Throughout their school career, from primary to secondary education and later in higher education, students have the opportunity to build and develop various skills, including technological. Considering the different needs and different uses that these skills encompass, these will certainly be necessary and useful in students’ academic and professional life. This study reports on an investigation of technological skills development in higher education. It aims to analyze the relevance of technologies integration, which technological skills are built and developed by higher education students, and what their perception about the importance of technological skills is. Based on a literature review, an online questionnaire was designed and applied to 217 students from three public higher education institutions located in the North, Center, and South of Portugal. This intended to verify which areas of technological skills (from the European Digital Competence Framework for Citizens) are most developed and to understand respective repercussions. It is concluded that the balanced development of students’ technological skills in higher education is crucial for their personal, social, and professional future and consequently, for their quality of life, with the integration of digital technologies being relevant in the change of the academic work organization, in the relations between learners, teachers, and institutions, and in the new ways of teaching and learning.

Eosinophilic esophagitis is an emerging chronic oesophageal inflammatory disease, increasingly recognized both in children and in adults. It is presently accepted that this entity has a chronic course with persistent or relapsing symptoms and a wide range of clinical manifestations at different age groups: in children it is responsible for feeding disorders, vomiting or abdominal pain; in teenagers and adults it causes dysphagia and oesophageal food impaction. The etiopathogenesis of eosinophilic esophagitis has not been yet well established, but immuno-allergic mechanisms associated with a Th2 immune response have been proposed. Diagnosis is based both on clinical and histological grounds, requiring that oesophageal mucosal biopsy specimens obtained from different oesophageal locations contain more than 15 intraepithelial eosinophils per high power field and the exclusion of gastroesophageal reflux disease. Currently proposed effective treatments rely on eviction of putative allergens through dietary exclusion and on treatment with swallowed steroids. We describe two cases (a young child and a teenager) illustrative of the clinical spectrum and course variability of eosinophilic esophagitis at pediatric age. A brief review of current knowledge concerning this puzzling clinical entity is presented, emphasizing specific diagnostic dilemmas and questions concerning its long-term follow-up.

The Mediterranean basin constitutes one of the best areas to analyse Neanderthal populations and lifestyles in South-Western Europe. In this context, new excavations conducted in the Middle Palaeolithic site of Cova del Puntal del Gat expand the information available regarding this rich region. In this study, new results are reported, including detailed studies on stratigraphy, lithic technology, anthracology, carpology, and zooarchaeology and taphonomy of macro and micromammals, with the final objective of characterizing the Neanderthals’ subsistence strategies and occupational patterns. These results are framed within a broader regional study perspective that includes MIS 5 and 4 sites. Chronostratigraphic review has enabled us to reorganize many sites that were originally included in MIS 3, towards older stages belonging to the end of MIS 4 and throughout MIS 5. Le bassin méditerranéen constitue l’une des meilleures zones pour analyser les populations et les modes de vie néandertaliens dans le sud-ouest de l’Europe. Dans ce contexte, les nouvelles fouilles menées dans le site paléolithique moyen du Cova del Puntal del Gat élargissent les informations disponibles pour cette riche région. Nous rapportons ici les nouveaux résultats, y compris des études détaillées sur la stratigraphie, la technologie lithique, l’anthracologie, la carpologie, ainsi que la taphonomie et la zooarchéologie de macro- et micro-restes de mammifères, dans le but de caractériser les stratégies de subsistance des Néandertaliens et leurs modes d’occupation. Ces résultats s’inscrivent dans une perspective d’étude régionale plus large qui inclut des sites MIS 5 et 4. La révision chronostratigraphique nous a permis de réorganiser de nombreux sites qui étaient initialement inclus dans le MIS 3, vers des stades plus anciens appartenant à la fin du MIS 4 et tout au long du MIS 5. A asma é uma doença prevalente nos ambulatórios e consultórios de Pneumologia, atingindo faixas etárias variadas. O controle da doença visa melhorar a qualidade de vida do portador e evitar complicações, sendo feito através do uso de medicações anti-inflamatórias e broncodilatadoras e do controle de fatores ambientais, possíveis gatilhos das crises. No entanto, pouco se fala a respeito da influência das condições psicológicas sobre esse controle. O transtorno de ansiedade e o transtorno depressivo são muito prevalentes na atualidade e existem diversas evidências de que exercem forte influência no controle da asma. Com o objetivo de avaliar a prevalência de sintomas depressivos e de ansiedade em pacientes portadores de asma e analisar o impacto destes no controle da doença, foi realizado um estudo transversal, observacional, descritivo no período de abril de 2019 a março de 2020 envolvendo asmáticos atendidos no ambulatório de Pneumologia da Faculdade Ciências Médicas de Minas Gerais, em Belo Horizonte (MG). Participaram do estudo 40 pacientes, que responderam a dois questionários validados (HADS - Hospital Anxiety and Depression Scale e ACT - Asthma Control Test) e dos quais foram coletados dados demográficos e espirométricos. Os resultados mostraram a existência de associação entre o controle da asma e os transtornos psiquiátricos. A prevalência de transtornos de ansiedade e/ou depressão na totalidade da amostra foi de 75%, sendo o de ansiedade o mais prevalente. Entre os portadores de asma não controlada, os transtornos estavam presentes em 92%, ao passo que entre aqueles com asma controlada a prevalência foi de 47%. Não houve, no entanto, associação significativa entre idade, sexo, alterações espirométricas e o controle da asma. Ao final do estudo, não foi possível definir claramente a relação causa-consequência entre controle da asma e os transtornos psiquiátricos, apesar da evidente associação encontrada, sendo necessário, portanto, novos estudos para tal.

É notória, ao longo das últimas décadas, a necessidade de criar interações entre o sector académico e o sector industrial de modo a maximizar o valor socio-económico dos avanços científicos. É este um dos motes do quadro de financiamento científico europeu Horizon 2020, que promove o desenvolvimento de determinada área científica através do estabelecimento de redes de colaboração com vários stakeholders para além do meio académico, entre eles o sector industrial. Ao mesmo tempo que é promovido pela Comissão Europeia a criação de diálogo entre a academia e a indústria, parece haver desconhecimento por parte dos coordenadores e gestores de projetos em como abordar estas interações, em parte devido ao não conhecimento de quem são os intervenientes do lado da indústria, como estabelecer os primeiros contactos e que ferramentas de comunicação usar. Igualmente, o meio académico parece não valorizar ou compreender a utilidade em estabelecer uma colaboração próxima com a indústria, o que leva muitas vezes os beneficiários dos projetos do quadro Horizon 2020 a não priorizarem este diálogo, apesar das obrigações dos contractos de financiamento. Assim, muitas das ações organizadas neste sentido acabam por não ser frutíferas, uma vez que são desprovidas de um objetivo específico e com uso inadequando dos recursos materiais e humanos. Então como podem os coordenadores de projetos Horizon 2020 corresponder às exigências europeias no que toca ao estabelecimento de relações com a indústria? Através da análise da bibliografia, de projetos Horizon 2020 coordenados por instituições portuguesas e de entrevistas a elementos de institutos de investigação portugueses, foi possível criar um guia prático no qual são identificados e enumerados exemplos de bons procedimentos e considerações, que têm como objetivo servir como linhas orientadoras para futuros coordenadores e gestores de projeto no desenho estratégico dos seus planos de comunicação com potenciais colaboradores do sector industrial. In the last two decades, it has become clear the need to establish an interaction between the academia and industry to maximize the socio-economic value of the scientific advances. This is the main goal of the Horizon 2020 finding program, which aims to promote the development of a specific research area through establishment of networks with different stakeholders outside of the academia, such as the industrial partners. If in one hand the European Commission foster the establishment of a dialogue between the academia and industry, on the other the project coordinators and managers are not aware on how to promote this network, as they are not aware who to contact, how to establish those first contacts and are unable to identify which communication tools should be used. Likewise, the academia does not seem to value or understand the relevance of establishing a collaborative network with industry, although it is required by the grant agreement obligations. Several of the organized actions are devoid of meaning, as they lack a welldefined goal and clear assessment of the human and material resources available. As such, how can the Horizon 2020 project coordinators properly correspond to the contractual obligations when it comes to fostering a academia-industry collaborations, if they are not perceived as relevant? Through bibliographic analysis, evaluation of Horizon 2020 funded-projects coordinated by Portuguese institutes, and interviews to keys players of Portuguese R&D institutes, it was possible to develop a guideline that highlights good practices and relevant considerations, that will assist future project coordinators and managers in the design of a strategic communication plan to further foster the academia-industry collaborations.

- Abstract
School Principals lead the school management and share it with family members in early childhood schools in Brazil. The Scientific knowledge produced about the role of the school principal demands time to reach society. Also, the literature points out flaws in the education of the school principal as well. It suggests a gap between the literature recommendations regarding school management and: the practice of the school principal; and the family knowledge in school management. Therefore, the study aims to identify the school principals', as well the students' perspectives, regarding the role of the school principal. To this end, a qualitative approach was used, based on interviews with informants about the principal's role in school management. The main results were: the principals understand that all areas of school management influence the students' success; pedagogical management is focused exclusively on the student; in the financial administrative field, there is a resonance with what the literature advocates, but there is difficulty in the execution of bureaucracy; in democratic management, there is a withdrawal of families in the decision-making process and an approximation for the execution of actions established by the management.

A contribution to the discussion about Vila Nova de São Pedro (VNSP) as a production centre of symbolic lithic artefacts, the origin of raw materials, as well the comparison with similar artefacts from other Chalcolithic sites are the major goals. For the first time, non-destructive analytical methods were employed to obtain a detailed chemical characterization of these remarkable artefacts. The results point to its production mainly with local raw material. This work enhances the understanding of the trade routes of raw materials and objects from the “production centre” of VNSP during the Chalcolithic.

The Tinto and Odiel rivers (SW Spain) drain from a vast sulfide mining district and join at a 20-km-long estuary that enters the Atlantic Ocean. In this work, the contents of rare earth elements (REE) and fractionation in Neogene–Holocene sediment cores from La Fontanilla cove (Tinto estuary) were studied. The sediments were collected from a depth of 18 m at different distances from the recent river flow and were analyzed for new information on the temporal development of the REE load in the sediment column. Results show that the å REE is higher in the finer sediments and during periods of mining activity from prehistoric to recent times. Marine influence appears to increase the light REE (LREE) relative to the heavy REE (HREE). The REE patterns of these estuarine sediments show convex curvatures in the MREE relative to the LREE and HREE, indicating the presence of acid-mixing processes between the fluvial waters affected by acid mine drainage (AMD) and seawater, as well as the precipitation of poorly crystalline mineral phases. Significant positive Eu anomalies were found in ebb-tide channels and marsh deposits, which can reflect the mineralogical composition and/or a strong localized salinity gradient combined with organic matter degradation. O trabalho de projeto, enquadrado nas metodologias ativas de ensino-aprendizagem de base construtivista, permite a construção de competências transversais, atualmente potenciado pela utilização das tecnologias digitais. Esta metodologia assume na contemporaneidade uma renovada importância, designadamente devido: à complementaridade com o ensino online, onde os espaços e os tempos se diluem, e à ponte que permite estabelecer entre o ensino e a sociedade. O objetivo deste estudo é analisar de que forma a metodologia de trabalho de projeto contribuiu para a construção de competências pelos estudantes, particularmente de autonomia e de literacia digital. A abordagem será qualitativa com recurso a um estudo de caso, recorrendo a observação participante e análise de conteúdo de reflexões escritas. Como resultados espera-se contribuir para uma melhor compreensão do processo de implementação da metodologia de trabalho de projeto, com recurso intensivo à integração de tecnologias digitais na comunicação e no processo de ensino, avaliação e aprendizagem.

Action research as a qualitative research methodology in the educational context can contribute to a reflective and collective questioning with a view to improving pedagogical practices, to the understanding of these practices and situations in which they were developed and, consequently, to the professional development of teachers. In data collection and processing, content analysis is a privileged method in the analysis of qualitative data which includes a set of techniques that allow a more rigorous and objective compliance with the scientific criteria and validation of knowledge construction. This article, through a literature review and a case analysis, aims to understand how the action research methodology can be applied in research studies in the educational context, specifically in teacher education, and to verify the importance of the content analysis method in the qualitative research quality. The case presented consists of a research-training project developed in an educational community with the objective of promoting the innovation of methodologies and pedagogical strategies through the application of a program of active teacher education with the integration of digital technologies. This project confirmed the development of a more digital culture in this educational community, fostering the professional development of teachers in the context of the future knowledge society, confirming the appropriateness of the proposed research methodology. A investigação-ação enquanto metodologia de investigação qualitativa no contexto educacional pode contribuir para um questionamento reflexivo e coletivo com vista ao melhoramento das práticas pedagógicas, para a compreensão sobre essas práticas e situações nas quais estas foram desenvolvidas e, consequentemente, para o desenvolvimento profissional docente. Na recolha e tratamento dos dados, a análise de conteúdo apresenta-se como um método privilegiado na análise de dados qualitativos que inclui um conjunto de técnicas que permitem cumprir de forma mais rigorosa e objetiva os critérios de cientificidade e a validação da construção do conhecimento. Este artigo, através de uma revisão de literatura e análise de um caso, pretende compreender como pode ser aplicada a metodologia de investigação-ação em estudos de investigação no contexto educacional, especificamente na formação de professores, e verificar a importância que assume o método de análise de conteúdo na qualidade de uma investigação qualitativa. O caso apresentado constitui-se por um projeto de investigação-formação desenvolvido numa comunidade educativa com o objetivo de promover a inovação de metodologias e estratégias pedagógicas através da aplicação de um programa de formação ativa de professores com integração das tecnologias digitais. Neste projeto confirmou-se o desenvolvimento de uma cultura mais digital nesta comunidade educativa, propiciadora do desenvolvimento profissional docente no contexto da futura sociedade do conhecimento, constando-se a adequação da metodologia de investigação proposta. La investigación-acción como metodología de investigación cualitativa en el contexto educativo puede contribuir a un cuestionamiento reflexivo y colectivo con vistas a mejorar las prácticas pedagógicas, a la comprensión de estas prácticas y de las situaciones en las que se desarrollaron y, en consecuencia, al desarrollo profesional de los docentes. En la recogida y tratamiento de datos, el análisis de contenido es un método privilegiado en el análisis de datos cualitativos que incluye un conjunto de técnicas que permiten un cumplimiento más riguroso y objetivo de los criterios de cientificidad y validación de la construcción del conocimiento. Este artículo, a través de una revisión bibliográfica y del análisis de un caso, pretende comprender cómo la metodología de la investigación-acción puede aplicarse en estudios de investigación en el contexto educativo, concretamente en la formación de profesores, y comprobar la importancia del método de análisis de contenido en la calidad de una investigación cualitativa. El caso presentado consiste en un proyecto de investigación-formación desarrollado en una comunidad educativa con el objetivo de promover la innovación de metodologías y estrategias pedagógicas mediante la aplicación de un programa de formación activa del profesorado con la integración de tecnologías digitales. Paroxysmal nocturnal hemoglobinuria (PNH) is an acquired life-threatening disorder that is extremely rare in the pediatric age. Patients with PNH may present a wide range of clinical manifestations. In most cases, the severity is associated with the size of the PNH clones; yet, some patients can be asymptomatic with a large clone. Thrombosis occurs less frequently as a presenting symptom, but it is considered the leading cause of mortality. In this article, we describe a case of PNH in a pediatric patient, complicated with venous thrombosis and with portal hypertension, associated with gastroesophageal varices at the time of diagnosis.

Portuguese museums and private collectors gather a variety of Italian glazed terracotta sculptures attributed to the della Robbia workshop, particularly active between the 15th and 16th centuries. Compositional studies of the paste using INAA, PGAA and XRD and enamel using PIXE, and luminescence protocols, were used as a tool to ascertain the attribution of the sculptures to della Robbia workshop. Geochemical and mineralogical patterns were defined enabling to express the production recipe. The luminescence ages were found to be between 1380 and 1640 in some cases with high uncertainties. The compositions of the blue glazes were proved to be similar to other measured on della Robbia sculptures from French and Italian collections based on cobalt pigments. Arsenic behaviour, which is associated to cobalt in most cobalt-minerals, is rather peculiar, as it was already found in other della Robbia sculptures enabling a chronological framework, pointing to a production after 1520. Based on the results, most of the analysed sculptures implies a della Robbia production, and only a few point to a composition and chronology not related with that workshop. As Instituicoes de Ensino Superior (IES) assumem um papel importante no crescimento economico e desenvolvimento regional com um contributo ao nivel da melhoria do capital humano, inovacao e transferencia de conhecimento, que se repercute consequentemente na qualidade de vida da populacao. Este trabalho insere-se no projeto de investigacao U-Value [1] , que tem como objetivo estudar o impacto que as IES tem na qualidade de vida das regioes, quer ao nivel qualitativo quer quantitativo, bem como a eficiencia das mesmas na transformacao de fundos publicos, fundos proprios e diferentes tipos de capital humano, social e organizacional em outputs com influencia na qualidade de vida das populacoes e sustentabilidade das suas regioes. Neste artigo, suportado numa revisao de literatura, pretendemos analisar alguns indicadores em Portugal, do lado da procura com base nas despesas ( inputs ), ao nivel dos estudantes e ID e do lado da oferta com base no conhecimento ( outputs ) relacionados com o capital humano, inovacao e transferencia de conhecimento, como forma de analisar as praticas e a responsabilidade das IES ao nivel da inovacao e transferencia de conhecimento para as empresas e a sociedade. Para quantificar os indicadores a serem analisados, serao utilizadas as bases de dados publicas portuguesas. Trata-se de um levantamento de dados preliminar de suporte a investigacao, pretendeu-se analisar algumas relacoes estabelecidas com base nos indicadores referidos.

The genesis and development of calcretes are discussed by using nuclear and related methods of analyses. In the studied calcrete, smectite is the main clay mineral associated with palygorskite. Chemical elements proportions, particularly V, U, Mn, Cu, Ba and REE, indicate a change to more oxidizing conditions and the contribution of bio-mediated processes in the oxalate-carbonate pathway, also confirmed by the presence of needle-fibre calcite. The proposed pathways leading to the studied calcrete development comprise both abiotic and biogenic genesis, emphasizing the polygenetic character due to different agents and processes and the paleoenvironmental trend towards aridification, with a transition between lacustrine and palustrine environments.

A diverse orchard with fruit fly hosts may provide information about trophic relationships, including new insights into beneficial insects. We evaluated the composition of the fruit fly complex to provide information on tephritid species, parasitoids and multitrophic interactions for the southern region of Minas Gerais, Brazil. Sampling was carried out using traps and by collecting fruits from plants and/or the ground according to availability/the fruiting period. Occurrences of Anastrepha amita Zucchi and A. punctata Hendel were recorded for the first time in the state of Minas Gerais, and new trophic associations were obtained for A. bahiensis Lima, A. bistrigata Bezzi, A. fraterculus (Wiedemann), A. obliqua (Macquart) and Ceratitis capitata (Wiedemann). Ten tephritid species were obtained from trap sampling, with C. capitata, A. fraterculus and A. obliqua being the most abundant. Five species of fruit flies and seven species of parasitoids were obtained from fruits. The braconid Doryctobracon areolatus (Szépligeti) was the most frequently collected among the parasitoid species. Pitanga (Eugenia uniflora L.) and purple guava (Psidium myrtoides O. Berg) fruits were classified as repositories of fruit fly parasitoids.

Tese de doutoramento em Medicina, As doenças de foro respiratório são altamente prevalentes a nível mundial, encontrando-se no grupo das doenças mais fatais em países desenvolvidos. Epitélio pulmonar gerado in-vitro a partir de células estaminais pluripotentes humanas tem aplicações em medicina regenerativa, no estabelecimento de novos modelos de doença, ensaios farmacológicos pré-clínicos e estudo do desenvolvimento humano. Neste trabalho descrevemos uma estratégia para gerar células da via aérea e alveolares maturas a partir de células estaminais pluripotentes humanas. Seguimos os paradigmas do desenvolvimento para gerar progenitores pulmonares (PPs) a partir das células estaminais e descobrimos que a glycogen synthase kinase 3 (GSK3) desempenha um papel central no balanço entre expansão de progenitores e a sua maturação em multi-linhagem. É importante realçar que a maioria das células geradas através do nosso método exibia características similares a células pulmonares pós-natais. Para além disso, o nosso modelo permitiu inferir um novo mecanismo envolvido no desenvolvimento pulmonar humano. Demonstramos que o efeito da inibição da maturação pela GSK3 no seu estado inibido é em parte devido à regulação do ciclo celular. Em culturas libertadas da inibição da GSK3, a via de sinalização NOTCH favorece a maturação dos PPs em populações epiteliais distais, inibindo as mais proximais, enquanto que a via WNT promove a maturação de células de Clara, implicando desta forma ambas as vias de sinalização no processo de especificação proximo-distal do epitélio pulmonar. Finalmente, demonstramos que o modelo desenvolvido é passível de ser utilizado com plataforma para desenvolvimento de modelos de doença respiratória humana e que poderá ser uma ferramenta importante para estudos sobre a fisiologia e importância de populações epiteliais pulmonares raras., Respiratory diseases are highly prevalent worldwide and consistently rank within the group of the most fatal diseases in developed countries. Lung epithelium generated in-vitro from human pluripotent stem cells (hPSCs) has tremendous potential for applications in regenerative medicine, disease modeling, pre-clinical drug screenings and the study of human development. Here we describe a strategy to derive mature airway and alveolar lung epithelium from hPSCs. We followed developmental cues to derive hPSCs towards lung progenitors (LPs) and found that glycogen synthase kinase 3 (GSK3) plays a central role in the balance between progenitor expansion and multilineage maturation of LPs. Importantly, the majority of the cells generated by our method exhibited characteristics similar to those found in postnatal human lungs. Our model offers novel mechanistic insight into human lung development. We show that the maturation inhibiting effects of GSK3 inhibition (GSK3i) in our cultures is partly due to cell cycle regulation. In addition, upon release of GSK3i, NOTCH signaling induces a distal cell fate at the expense of proximal and ciliated cell fates, whereas WNT signaling promotes a proximal club cell fate, implicating both signaling pathways in proximodistal specification of the human lung. 46. Pyruvate dehydrogenase complex deficiency: updating the clinical, metabolic and mutational landscapes

Background : Pyruvate dehydrogenase complex (PDC) catalyzes the irreversible decarboxylation of pyruvate into acetyl-CoA which ultimately generates ATP. PDC deficiency can be caused by alterations in any of the genes encoding its several subunits, and the resulting phenotype, though very heterogeneous, mainly affects the neuro-encephalic system. The aim of this study is to describe and discuss the clinic, metabolic and genotypic profiles of thirteen PDC deficient patients, thus seeking to establish possible genotype-phenotype correlations. Results : The mutational spectrum revealed that seven patients (54 %) carry mutations in the PDHA1 gene , encoding the E1α subunit, five patients (38 %) carry mutations in the PDHX gene, encoding the E3 binding protein, and the remaining patient (8 %) harbors mutations in the DLD gene, encoding the E3 subunit. These data corroborate PDHA1 mutations as the predominant cause of PDC deficiency, though revealing a notable prevalence of PDHX mutations among Portuguese patients, most of them carrying a seemingly private mutation (p.R284X). The biochemical analyses revealed high lactate and pyruvate plasma levels whereas de ratio L/P was under 16; enzymatic activities, when compared to control values, revealed to be independent from the genotype and ranged from 8.5% to 30% which may be considered a cut-off value for primary PDC deficiency. Concerning the clinical features, all patients displayed developmental delay/psychomotor retardation, the severity of which seems to correlate with the type and localization of the mutation carried by the patient. The therapeutic options essentially go through the administration of a ketogenic diet and supplementation with thiamine, although arginine aspartate intake revealed to be beneficial in some patients. Moreover, the in silico analysis of the missense mutations present in this PDC deficient population allowed to understand the molecular mechanism underlying these pathogenic variants. Conclusion : The identification of the disease-causing mutations, together with the functional and structural characterization of the mutant protein variants, allows to get insight on the severity of the clinical phenotype and the selection of the most appropriate therapy.
