Abstract: Acceptability and relevance of nanoparticles in the society is greatly improved using a safer-by-design strategy. However, this is difficult to implement when too late in the development process or when nanoparticles are already on the market (e.g., TiO2). We employ this strategy for emerging nanoparticles of lanthanide oxysulfide of formula (Gd,Ce)2O2S, relevant for photocatalysis as well as for multimodal imaging, as the bandgap of the nanoparticles, related to their Ce content, impacts their ability to absorb visible light. As a first step, we investigated the production of reactive oxygen species (ROS) as a function of cerium content, in abiotic conditions and in vitro using murine macrophage RAW 264.7 cell line. We demonstrate that, at sub-lethal doses, Ce-containing oxysulfide nanoparticles are responsible for a higher ROS intracellular formation than cerium-free Gd2O2S nanoparticles, although no significant inflammatory response or oxidative stress was measured. Moreover, there was no significant loss of cerium as free cation from the nanoparticles, as evidenced by X-ray fluorescence mapping. Based on these results, we propose a risk analysis for lanthanide oxysulfide nanoparticles, leading to a technology assessment that fulfills the safer-by-design strategy.

Abstract: The increasing number of patents suggests that the use of nanoparticles (NPs) in agriculture will continue to grow in the future. Properties associated with nanosized objects might be desirable, but may pose a threat to the environment since the fate of nanomaterials is poorly understood. Seed treatment is an extensive practice to provide micronutrients and disease protection. This study investigates the effect of Ag2S NPs, Ag0 NPs, and AgNO3 (1, 100, and 1000 mg Ag per L) on the germination of Phaseolus vulgaris seeds and the mechanism of interaction in the seed coat. Ag treatments did not affect the germination rate. On the other hand, the development of seedlings was significantly improved by Ag2S NPs while AgNO3 showed a negative effect compared to the control (water). Silver from different sources was accumulated and biotransformed along the seed coat. Ag from the AgNO3 treatment was found as AgCl and bound to oxygen, and Ag0 NPs underwent oxidative dissolution forming thiolate complexes. On the other hand, Ag2S NPs did not show any detectable chemical changes. Silver distribution and speciation were investigated in the bean testa using μ-XRF and μ-XANES, while the macromolecular composition was assessed by ATR-FTIR. The Ag biotransformation depended on the source. For the FTIR results, it is possible to verify perturbations in the carbohydrate region as well as changes in the protein region. The combination of these results can contribute to building another step in the understanding of how nanoparticles can improve agricultural inputs and benefit food crops.

Abstract: International audience; ABSTRACT: Identifying engineered nanomaterials (ENMs) made from earth-abundant elements in soils is difficult because soil also contains natural nanomaterials (NNMs) containing similar elements. Here, machine learning models using elemental fingerprints and mass distributions of three TiO2 ENMs and Ti-based NNMs recovered from three natural soils measured by single-particle inductively coupled plasma time-of-flight mass spectrometry (spICP-TOFMS) was used to identify TiO2 ENMs in soil. Synthesized TiO2 ENMs were unassociated with other elements (>98%), while 40% of Ti-based ENM particles recovered from wastewater sludge had distinguishable elemental associations. All Ti-based NNMs extracted from soil had a similar chemical fingerprint despite the soils being from different regions, and >60% of Ti-containing NNMs had no measurable associated elements. A machine learning model best distinguished NNMs and ENMs when differences in Ti-mass distribution existed between them. A trained LR model could classify 100 nm TiO2 ENMs at concentrations of 150 mg kg−1 or greater. The presence of TiO2 ENMs in soil could be confirmed using this approach for most ENM-soil combinations, but the absence of a unique chemical fingerprint in a large fraction of both TiO2 ENMs and Ti-NNMs increases model uncertainty and hinders accurate quantification.

Abstract: Despite the increasing attention given to the impacts of nanoplastics in terrestrial environments, there is limited data about the effects on plants, and the quantitative information on uptake. In the present study, wheat plants grown in hydroponics were exposed to Pd-doped nanoplastics. This allowed us to quantify nanoplastics uptake and translocation to the shoots. Visualization of nanoplastics in roots was performed with synchrotron micro X-ray fluorescence (µXRF). Nanoplastics accumulated on the root epidermis, especially at the root tip and in root maturation zones. A close relationship between plant roots, rhizodeposits and nanoplastics behaviour was shown. Reinforcement of the cell wall in roots was evidenced using Fourier transform infrared spectroscopy (FTIR) and synchrotron-computed microtomography (µCT). Synchrotron-computed nanotomography (nanoCT) evidenced the presence of globular structures but they could not be identified as nanoplastics since they were observed both in the control and treated roots. By utilizing the inorganic tracer in the doped-nanoplastics, this study paves the road for elucidating interactions in more complex systems by using an integrative approach combining classical phytotoxicity markers with advanced nanometrology techniques. Abstract: International audience; Sulfidation is a key process for silver nano-particles released from consumer products in the environment. This study focuses on the impact of a model soil microorganism , Bacillus subtilis, on the fate of pristine and already sulfidized Ag-NPs. The nanoparticles were incubated with the initial growth medium, isolated secretome, and living bacteria, and characterized for their size and morphology, agglomeration state, structure, and Ag speciation. No Ag internalization or sorption on the cell wall was detected. A partial sulfidation, leading to an Ag−Ag 2 S core−shell structure, was observed in the presence of the secretome, and the rate limiting step of the reaction was the oxidation of Ag 0 , and it was favored near the crystal dislocations. The sulfidation was complete in the presence of the living bacteria and followed an indirect pathway. Both crystalline Ag 2 S and amorphous Ag 2 S and/or Ag-thiol were identified. At the opposite, the bacteria had no impact on Ag 2 S. These results suggest that microorganisms participate in the sulfidation of Ag-NPs in aerobic systems such as unsaturated soils, and thus affect the bioavailability of Ag. It is important to take these transformations into account during exposure experiments, since they drastically change the exposure conditions. Finally, the secretome of B. subtilis might be used for the green synthesis of Ag−Ag 2 S core−shell nanoparticles.

Abstract: Phosphorus (P) resource availability is declining and the efficiency of applied nutrients in agricultural soils is becoming increasingly important. This is especially true for P fertilizers from recycled materials, which often have low plant availability. Specific co-fertilization with ammonium can enhance P plant availability in soils amended with these P fertilizers, and thus the yield of plants. To investigate this effect, we performed a pot experiment with maize in slightly acidic soil (pH 6.9) with one water-soluble (triple superphosphate [TSP]) and two water-insoluble (sewage sludge-based and hyperphosphate [Hyp]) P fertilizers and an ammonium sulfate nitrate with or without a nitrification inhibitor (NI). The dry matter yield of maize was significantly increased by the NI with the Hyp (from 14.7 to 21.5 g/pot) and TSP (from 40.0 to 45.4 g/pot) treatments. Furthermore, P uptake was slightly increased in all three P treatments with the NI, but not significantly. Olsen-P extraction and P K-edge micro-X-ray absorption near-edge structure (XANES) spectroscopy showed that apatite-P of the water-insoluble P fertilizers mobilized during the plant growth period. In addition, novel nitrogen (N) K-edge micro-XANES spectroscopy and the Mogilevkina method showed that the application of an NI increased the fixation of ammonium in detectable hot spots in the soil. Thus, the delay in the nitrification process by the NI and the possible slow-release of temporarily fixed ammonium in the soil resulted in a high amount of plant available ammonium in the soil solution. This development probably decreases the rhizosphere pH due to release of H+ by plants during ammonium uptake, which mobilizes phosphorus in the amended soil and increases the dry matter yield of maize. Abstract: Significance Silver nanowires are a type of novel fiber that are likely to have wide application in consumer electronics but that can also carry risk for cell internalization and toxicity. Here, we show that silver nanowire toxicity can be greatly diminished by reducing nanowire diameter without affecting device performance. X-ray microscopy and supporting studies revealed that endolysosome membranes can mechanically crumple thin silver nanowires, the likely mechanism for diminishing toxicity., Fibrous particles interact with cells and organisms in complex ways that can lead to cellular dysfunction, cell death, inflammation, and disease. The development of conductive transparent networks (CTNs) composed of metallic silver nanowires (AgNWs) for flexible touchscreen displays raises new possibilities for the intimate contact between novel fibers and human skin. Here, we report that a material property, nanowire-bending stiffness that is a function of diameter, controls the cytotoxicity of AgNWs to nonimmune cells from humans, mice, and fish without deterioration of critical CTN performance parameters: electrical conductivity and optical transparency. Both 30- and 90-nm-diameter AgNWs are readily internalized by cells, but thinner NWs are mechanically crumpled by the forces imposed during or after endocytosis, while thicker nanowires puncture the enclosing membrane and release silver ions and lysosomal contents to the cytoplasm, thereby initiating oxidative stress. This finding extends the fiber pathology paradigm and will enable the manufacture of safer products incorporating AgNWs.

Abstract: International audience; Increasing evidence indicates the presence of engineered nanoparticles (ENPs) in sewage sludge derived from wastewater treatment. Land application of sewage sludge is, therefore, considered as an important pathway for ENP transfer to the environment. The aim of this work was to understand the effects of sewage sludge containing nano-TiO2 on plants (tomato) when used as an amendment in agricultural soil. We assessed developmental parameters for the entire plant life cycle along with metabolic and bio-macromolecule changes and titanium accumulation in plants. The results suggest that the sewage sludge amendment containing nano-TiO2 increased plant growth (142% leaf biomass, 102% fruit yield), without causing changes in biochemical responses, except for a 43% decrease in leaf tannin concentration. Changes in elemental concentrations (mainly Fe, B, P, Na, and Mn) of plant stem, leaves and, to a lesser extent fruits were observed. Fourier-transformed infrared analysis showed maximum changes in plant leaves (decrease in tannins and lignins and increase in carbohydrates) but no change in fruits. No significant Ti enrichment was detected in tomato fruits. In conclusion, we evidenced no acute toxicity to plants and no major implication for food safety after one plant life cycle exposure.

Abstract: This work aims to investigate the nature and the specific mechanisms by which polycarboxylic compounds participate in the tolerance of Silene vulgaris to Cr with special attention given to the rhizosphere system. This knowledge is important to use this species in the implementation of phytoremediation technologies in Cr-polluted soils. According to the results, chromium is chelated and mobilized by the citric and malic acids in plant tissues, while oxalic acid might participate in the reduction and chelation of Cr in the rhizosphere. At the applied doses, the response of both exudation rate and root exudate composition (total polyphenols and quercitin) seems to involve a rearrangement in the lignification of the plant cell wall to immobilize Cr. Quercetin-3-dirhamnosyl-galactoside and apiin (apigenin-7-O-apiosyl-glucoside) have been identified as the major polyphenols in the root exudates of S. vulgaris. The increments found in the apiin concentration in root exudates seem to be related to the protection against Cr toxicity by chelation of Cr or by free radical scavenging. Abstract: Dust deposition is an important source of phosphorus (P) to many ecosystems. However, there is little evidence of dust-derived P-containing minerals in soils. Here we studied P forms along a well-described climatic gradient on Hawaii, which is also a dust deposition gradient. Soil mineralogy and soil P forms from six sites along the climatic gradient were analyzed with bulk (X-ray diffraction and P K-edge X-ray absorption near edge structure) and microscale (X-ray fluorescence, P K-edge X-ray absorption near edge structure, and Raman) analysis methods. In the wettest soils, apatite grains ranging from 5 to 30 µm in size were co-located at the micro-scale with quartz, a known continental dust indicator suggesting recent atmospheric deposition. In addition to co-location with quartz, further evidence of dust-derived P included backward trajectory modeling indicating that dust particles could be brought to Hawaii from the major global dust-loading areas in central Asia and northern Africa. Although it is not certain whether the individual observed apatite grains were derived from long-distance transport of dust, or from local dust sources such as volcanic ash or windblown fertilizer, these observations offer direct evidence that P-containing minerals have reached surface layers of highly-weathered grassland soils through atmospheric deposition., Geoderma, 381, ISSN:0016-7061, ISSN:1872-6259

Abstract: Nanosized TiO2 is one of the most produced nanomaterials. Agricultural soils are a major compartment of accumulation of TiO2-NPs after release from consumer products into the sewer system, due to the use of sewage sludge as a soil amendment. TiO2 is naturally present in soils, and distinguishing between anthropogenic and natural TiO2 in soils is thus important to assess the risks associated with the increased use of nano-TiO2. Methods to distinguish these materials in complex matrices such as soils are currently lacking. The purpose of this study was, therefore, to search for characteristic physical and chemical properties of natural and engineered nano-TiO2, based on a combination of bulk, micro and nanofocused X-ray fluorescence and X-ray absorption spectroscopy, transmission electron microscopy, X-ray diffraction and chemical analyses. Digested sewage sludge, agricultural soil and sludge-amended soil were studied by these techniques. The particle size distribution was not a relevant criterion since the sludge and the sludge-amended soil contained a variety of nanometer- and micrometer sized Ti-containing particles. Both the sludge and the soil contained a mixture of rutile and anatase, with a minor proportion of amorphous TiO2. In the sludge, there was no trend relating particle size and Ti mineralogy. The morphology of the TiO2 particles proved to be different in the two matrices, with smooth faceted particles in the sludge and rough irregular ones in the soil. In addition, natural TiO2 particles were included in micro and macroaggregates of the soil and formed intricate assemblages with minerals and organic compounds. In the sludge, TiO2 formed homo and heteroaggregates of simpler structure, richer in organic matter. Thus, the study of the morphology of TiO2 particles and their status in unperturbed mineral–organic assemblages may provide some insights into their origin. The observed differences may attenuate over time, due to the incorporation of the sludge material within the soil structure. Abstract: The extensive use of Cr compounds in industry has led to the disposal of this element in soil and water, with consequent hazards to the environment and human health. Phytomanagement of Cr contaminated sites could be a sustainable alternative to obtain a profit from otherwise useless land. The objective of this work was to evaluate the influence of Cr speciation on root exudation in two genotypes of the metallophyte Silene vulgaris, which have shown different tolerance to Cr(VI). Plant growth has been related to oxidative stress by malondialdehyde (MDA) measures. No toxicity symptoms were observed at the applied dose of Cr(III), whereas biomass was significantly reduced by exposure to Cr(VI). Shoots have been found to be more sensitive to Cr stress than roots. Cr accumulation in plants was correlated to exudate release rates and induced changes in their composition. The genotype SV21 showed less biomass reduction and oxidative stress than the less tolerant SV38. The increment of the exudation rate in the genotype SV21 under Cr(VI) exposure suggested that exudation could be one of the mechanisms implicated in the tolerance of this species.

Abstract: Phytomanagement could be a viable alternative in areas polluted with wastes from chromium-using industries. This study investigated the ability of Silene vulgaris to take up Cr(III) and Cr(VI) with special attention on the mechanism used by this species to tolerate high doses of Cr(VI). Plants were grown semihydroponically with different concentrations of either Cr(III) or Cr(VI). A combination of synchrotron X-ray spectroscopic techniques, scanning electron and light microscopy and infrared spectroscopy were used to determine the distribution and speciation of Cr. S. vulgaris accumulated more Cr when grown with Cr(VI) resulting in an overall reduction in biomass. Starch accumulation in leaves may be attributed to an impartment between carbon utilization and assimilation resulted from stunted plant growth but not the complete inhibition of photosynthesis indicating that S. vulgaris possess tolerance mechanisms that allows it to survive in Cr(VI) rich environments. These primary tolerance mechanisms are (a) the total reduction of Cr(VI) to Cr(III) in the rhizosphere or just after uptake in the fine lateral root tips and (b) chelation of Cr(III) to the cell wall both of which reduce metal interference with critical cell functions. These mechanisms make S. vulgaris suitable for in situ remediation of Cr polluted soils.

Abstract: International audience; Agricultural soils are major sinks of silver nanoparticles in the environment, and crops are directly exposed to these emerging contaminants. A clear picture of their chemical transformations, uptake and transport mechanisms, and phytotoxic impacts is still lacking. In this work, wheat plants were exposed to pristine metallic (Ag-NPs) and sulfidized (Ag 2 S-NPs) silver nanoparticles and ionic Ag. Data on Ag distribution and speciation, phytotoxicity markers, and gene expression were studied. A multi-technique and multi-scale approach was applied, combining innovating tools at both the laboratory and synchrotron. Various chemical transformations were observed on the epidermis and inside roots, even for Ag 2 S-NPs, leading to an exposure to multiple Ag forms, which likely evolve over time. Genes involved in various functions including oxidative stress, defense against pathogens, and metal homeostasis were impacted in different ways depending upon the Ag source. This study illustrates the complexity of the toxicity pattern for plants exposed to Ag-NPs, the necessity of monitoring several markers to accurately evaluate the toxicity, and the interest of interpreting the toxicity pattern in light of the distribution and speciation of Ag.

Abstract: International audience; The ID21 beamline (European Synchrotron Radiation facility, France) is a multi micro-analytical platform combining X-ray and infrared micro-probes, for characterization of elements, species, molecular groups and crystalline structures in complex materials. Applications are mainly in the fields of cultural heritage, life science, environmental and earth sciences, materials sciences. Here, we first present the status of instruments: (i) the scanning micro-spectroscopy end-station, operating from 2.0 to 9.2 keV, under vacuum and offering cryo conditions, for the acquisition of 2D micro X-ray fluorescence (μXRF) maps, single point micro X-ray Absorption Near Edge Structure (μXANES) spectra and speciation maps with sub-micrometric resolution; (ii) the XANES full-field end-station, operating in the same vacuum and energy conditions, for the acquisition of hyper-spectral radiographs of thin concentrated samples, resulting in speciation maps with micrometric resolution and millimetric field of view; (iii) the scanning micro-X-ray diffraction (μXRD)/μXRF end-station, operating at 8.5 keV, in air, for the acquisition of 2D crystalline phase maps, with micrometric resolution; and (iv) the scanning infrared microscope, operating in the mid-infrared range for the acquisition of molecular maps and some structural maps with micrometric resolution. Recent hardware and software developments are presented, as well as new protocols for improved sample preparation of thin sections. Secondly, a review of recent applications for the study of cultural heritage is presented, illustrated by various examples: determination of the origin of the color in blue Chinese porcelains and in brown Sèvres porcelains; detection of lead in ink on Herculaneum papyri; identification and degradation of modeling materials used by Auguste Rodin and of chrome yellow pigments used by Vincent van Gogh. Cryo capabilities are illustrated by the analysis of plants exposed to chromate solutions. These examples show the variety of materials analyzed, of questions tackled, and particularly the multiple advantages of the ID21 analytical platform for the analysis of ancient and artistic materials.

Abstract: International audience; Nanotechnology is the new industrial revolution of our century. Its development leads to an increasing use of nanoparticles and thus to their dissemination. Their fate in the environment is of great concern and especially their possible transfer in trophic chains might be an issue for food safety. However, so far our knowledge on this topic has been restricted by the lack of appropriate techniques to characterize their behavior in complex matrices. Here, we present in detail the use of cutting-edge beam-based techniques for nanoparticle in situ localization, quantification and speciation in a crop plant species (Lactuca sativa). Lettuce seedlings have been exposed to TiO2 and Ag nanoparticles and analyzed by inductively coupled plasma spectrometry, micro-particle induced X-ray emission coupled to Rutherford backscattering spectroscopy on nuclear microprobe, micro-X-ray fluorescence spectroscopy and X-ray absorption near edge structure spectroscopy. The benefits and drawbacks of each technique are discussed, and the types of information that can be drawn, for example on the translocation to edible parts, change of speciation within the plant, detoxification mechanisms, or impact on the plant ionome, are highlighted. Such type of coupled approach would be an asset for nanoparticle risk assessment.

19. Transformation of Silver Nanoparticles in Sewage Sludge during Incineration

Abstract: Silver nanoparticles (Ag-NP) discharged into the municipal sewer system largely accumulate in the sewage sludge. Incineration and agricultural use are currently the most important strategies for sewage sludge management. Thus, the behavior of Ag-NP during sewage sludge incineration is essential for a comprehensive life cycle analysis and a more complete understanding of the fate of Ag-NP in the (urban) environment. To address the transformation of Ag-NP during sewage sludge incineration, we spiked metallic Ag(0)-NP to a pilot wastewater treatment plant and digested the sludge anaerobically. The sludge was then incinerated on a bench-scale fluidized bed reactor in a series of experiments under variable conditions. Complementary results from X-ray absorption spectroscopy (XAS) and electron microscopy-energy dispersive X-ray (EM-EDX) analysis revealed that Ag(0)-NP transformed into Ag2S-NP during the wastewater treatment, in agreement with previous studies. On the basis of a principal component analysis and subsequent target testing of the XAS spectra, Ag(0) was identified as a major Ag component in the ashes, and Ag2S was clearly absent. The reformation of Ag(0)-NP was confirmed by EM-EDX. The fraction of Ag(0) of the total Ag in the ashes was quantified by linear combination fitting (LCF) of XAS spectra, and values as high as 0.8 were found for sewage sludge incinerated at 800 °C in a synthetic flue gas atmosphere. Low LCF totals (72% to 94%) indicated that at least one relevant reference spectrum was missing in the LCF analysis. The presence of spherical Ag-NP with a diameter of50 nm extending into the sub-nm range was revealed by electron microscopy analyses. The rapid formation of Ag(0)-NP from Ag2S during sewage sludge incineration, as demonstrated in this study, needs to be considered in the life cycle assessment of engineered Ag-NP.

Abstract: International audience; The objective of this work was to investigate the fate of silver nanoparticles (Ag-NPs) in a sludge-amended soil cultivated with monocot (Wheat) and dicot (Rape) crop species. A pot experiment was performed with sludges produced in a pilot wastewater treatment plant containing realistic Ag concentrations (18 and 400 mg kg(-1), 14 mg kg(-1) for the control). Investigations focused on the highest dose treatment. X-ray absorption spectroscopy (XAS) showed that Ag2S was the main species in the sludge and amended soil before and after plant culture. The second most abundant species was an organic and/or amorphous Ag-S phase whose proportion slightly vaned (from 24% to 36%) depending on the conditions. Abstract: [EN] Background Reactive oxygen species (ROS) are normally produced in respiratory and photosynthetic electron chains and their production is enhanced during desiccation/rehydration. Nitric oxide (NO) is a ubiquitous and multifaceted molecule involved in cell signaling and abiotic stress. Lichens are poikilohydrous organisms that can survive continuous cycles of desiccation and rehydration. Although the production of ROS and NO was recently demonstrated during lichen rehydration, the functions of these compounds are unknown. The aim of this study was to analyze the role of NO during rehydration of the lichen Ramalina farinacea (L.) Ach., its isolated photobiont partner Trebouxia sp. and Asterochloris erici (Ahmadjian) Skaloud et Peksa (SAG 32.85 = UTEX 911). Results Rehydration of R. farinacea caused the release of ROS and NO evidenced by the fluorescent probes DCFH2-DA and DAN respectively. However, a minimum in lipid peroxidation (MDA) was observed 2 h post-rehydration. The inhibition of NO in lichen thalli with c-PTIO resulted in increases in both ROS production and lipid peroxidation, which now peaked at 3 h, together with decreases in chlorophyll autofluorescence and algal photobleaching upon confocal laser incidence. Trebouxia sp. photobionts generate peaks of NO-endproducts in suspension and show high rates of photobleaching and ROS production under NO inhibition which also caused a significant decrease in photosynthetic activity of A. erici axenic cultures, probably due to the higher levels of photo-oxidative stress. Conclusions Mycobiont derived NO has an important role in the regulation of oxidative stress and in the photo-oxidative protection of photobionts in lichen thalli.
