Abstract Background Circular RNAs (circRNAs) are capable of affecting breast cancer (BC) development. However, the role and underneath mechanism of circFKBP8 (also known as hsa_circ_0000915) in BC remain largely unknown. Methods Expression analyses were performed using quantitative real-time polymerase chain reaction (qRT-PCR), western blot, and immunohistochemistry (IHC) assays. Effects on cell functional phenotypes were determined by assessing cell proliferation, migratory capacity, invasion, and stemness in vitro. The relationship between microRNA (miR)-432-5p and circFKBP8 or E2F transcription factor 7 (E2F7) was examined by RNA pull-down, dual-luciferase reporter, and RNA immunoprecipitation (RIP) assays. Xenograft assays were used to identify the function of circFKBP8 in vivo. Results CircFKBP8 was presented at high levels in BC tissues and cells. High circFKBP8 expression was associated with worse overall survival in BC patients. CircFKBP8 suppression inhibited BC cell proliferation, migratory capacity, invasion and stemness in vitro. CircFKBP8 suppression blocked xenograft tumor growth in vivo. Mechanistically, circFKBP8 functioned as a miR-432-5p sponge to modulate E2F7 expression. CircFKBP8 modulated BC cell malignant behaviors by miR-432-5p, and miR-432-5p affected these cell phenotypes through E2F7. Conclusion Our observations prove that circFKBP8 promotes BC malignant phenotypes through the miR-432-5p/E2F7 cascade, offering a promising therapeutic and prognostic target for BC.

In the context of the growing research interest in multi-component alloys (MAs) and their exceptional performance under extreme environments, the high-temperature oxidation resistance and applications of MAs have attracted significant attention in the field of metallic materials. While the cost-effective and mechanical properties of Co-free MAs are of great importance, their oxidation resistance remains insufficiently understood. In this work, we designed multiple heterogeneous structures within a cast dual-phase Cr30Fe30Ni30Al5Ti5 MA by tailoring the Al and Ti ratio, which consists of body-centered-cubic (BCC) grains reinforced by multi-scale nanoprecipitates (i.e., L21, B2, and η phase) and an L12-strengthened face-centered cubic (FCC) skeleton. Isothermal oxidation experiments at 800 °C, 1000 °C, and 1200 °C with varying exposure durations were conducted. The oxidation kinetics at 800 °C and 1000 °C followed a parabolic law, while both low weight increment and oxidation rate confirm remarkable oxidation resistance. At 800 °C, the oxides mainly consist of Cr2O3 and Al2O3, while are dominated by (TiO2 + Cr2O3) and the mixed oxides of Al2O3, TiO2, and Ti2O above 1000 °C. Importantly, the inability to form a continuous Al2O3 oxide scale at higher temperatures led to a deterioration in oxidation resistance. These findings offer valuable insights into underlying mechanisms contributing to oxidation resistance for Co-free MAs.

Currently, there is an urgent need for new antibacterial and antifungal agents to combat the growing challenge of antibiotic resistance. As the largest ecosystem on Earth, the marine ecosystem includes a vast array of microorganisms (primarily bacteria and fungi), plants, invertebrates, and vertebrates, making it a rich source of various antimicrobial compounds. Notably, terpenoids, known for their complex structures and diverse bioactivities, are a significant and promising group of compounds in the battle against bacterial and fungal infections. In the past five years, numerous antimicrobial terpenoids have been identified from marine organisms such as bacteria, fungi, algae, corals, sea cucumbers, and sponges. This review article provides a detailed overview of 141 terpenoids with antibacterial and/or antifungal properties derived from marine organisms between 2019 and 2024. Terpenoids, a diverse group of natural organic compounds derived from isoprene units, are systematically categorized based on their carbon skeleton structures. Comprehensive information is provided about their names, structures, biological sources, and the extent of their antibacterial and/or antifungal effectiveness. This review aims to facilitate the rapid identification and development of prospective antimicrobials in the pharmaceutical sector.

Lithium-ion battery State of Health (SOH) estimation is an essential issue in battery management systems. In order to better estimate battery SOH, Extreme Learning Machine (ELM) is used to establish a model to estimate lithium-ion battery SOH. The Swarm Optimization algorithm (PSO) is used to automatically adjust and optimize the parameters of ELM to improve estimation accuracy. Firstly, collect cyclic aging data of the battery and extract five characteristic quantities related to battery capacity from the battery charging curve and increment capacity curve. Use Grey Relation Analysis (GRA) method to analyze the correlation between battery capacity and five characteristic quantities. Then, an ELM is used to build the capacity estimation model of the lithium-ion battery based on five characteristics, and a PSO is introduced to optimize the parameters of the capacity estimation model. The proposed method is validated by the degradation experiment of the lithium-ion battery under different conditions. The results show that the battery capacity estimation model based on ELM and PSO has better accuracy and stability in capacity estimation, and the average absolute percentage error is less than 1%.

High-entropy alloys (HEAs) have attracted considerable attention for their exceptional microstructures and properties. Discovering new HEAs with desirable properties is crucial, but traditional design methods are laborious and time-consuming. Fortunately, the emerging Machine Learning (ML) offers an efficient solution. In this study, composition-microhardness data pairs from various alloy systems were collected and expanded using a Generative Adversarial Network (GAN). These data pairs were converted into empirical parameter-microhardness pairs. Then Active Learning (AL) was employed to screen the Al-Co-Cr-Cu-Fe-Ni system and identify the eXtreme Gradient Boosting (XGBoost) as the optimal ML master model. Millions of data training iterations employing the XGBoost sub-model and accuracy evaluations using the Expected Improvement (EI) algorithm establish the relationship between HEA compositions and microhardness. To better assess the practical value and avoid potential risks of the traditionally medicinal and edible basidiomycete Schizophyllum commune, which may arise from undescribed metabolites, a combination of elicitors was introduced for the first time to discover products from cryptic and low-expressed gene clusters under laboratory cultivation. Treating S. commune NJFU21 with the combination of five elicitors led to the upregulated production of a class of unusual linear diterpene-derived variants, including eleven new ones (1–11), along with three known ones (12–14). The structures and stereochemistry were determined by 1D and 2D NMR, HRESIMS, ECD, OR and VCD calculations. Notably, the elongation terminus of all the diterpenes was decorated by an unusual butenedioic acid moiety. Compound 1 was a rare monocyclic diterpene, while 2–6 possessed a tetrahydrofuran moiety. The truncated metabolites 4, 5 and 13 belong to the trinorditerpenes. All the diterpenes displayed approximately 70% scavenging of hydroxyl radicals at 50 μM and null cytotoxic activity at 10 μM. In addition, compound 1 exhibited potent antifungal activity against the plant pathogenic fungi Colletotrichum camelliae, with MIC values of 8 μg/mL. Our findings indicated that this class of diterpenes could provide valuable protectants for cosmetic ingredients and the lead compounds for agricultural fungicide development.

Terpenoid alkaloids are recognized as a class of compounds with limited numbers but potent biological activities, primarily derived from plants, with a minor proportion originating from animals and microorganisms. These alkaloids are synthesized from the same prenyl unit that forms the terpene skeleton, with the nitrogen atom introduced through β-aminoethanol, ethylamine, or methylamine, leading to a range of complex and diverse structures. Based on their skeleton type, they can be categorized into monoterpenes, sesquiterpenes, diterpenes, and triterpene alkaloids. To date, 289 natural terpenoid alkaloids, excluding triterpene alkaloids, have been identified in studies published between 2019 and 2024. These compounds demonstrate a spectrum of biological activities, including anti-inflammatory, antitumor, antibacterial, analgesic, and cardioprotective effects, making them promising candidates for further development. This review provides an overview of the sources, chemical structures, and biological activities of natural terpenoid alkaloids, serving as a reference for future research and applications in this area.

Sepsis-induced acute lung injury (ALI), characterized by widespread lung dysfunction, is associated with significant morbidity and mortality due to the lack of effective pharmacological treatments available clinically. Small-molecule compounds derived from natural products represent an innovative source and have demonstrated therapeutic potential against sepsis-induced ALI. These natural small molecules may provide a promising alternative treatment option for sepsis-induced ALI. This review aims to summarize the pathogenesis of sepsis and potential therapeutic targets. It assembles critical updates (from 2014 to 2024) on natural small molecules with therapeutic potential against sepsis-induced ALI, detailing their sources, structures, effects, and mechanisms of action.

Artabotrys, a pivotal genus within the Annonaceae family, is renowned for its extensive biological significance and medicinal potential. The genus’s sesquiterpene compounds have attracted considerable interest from the scientific community due to their structural complexity and diverse biological activities. These compounds exhibit a range of biological activities, including antimalarial, antibacterial, anti-inflammatory analgesic, and anti-tumor properties, positioning them as promising candidates for medical applications. This review aims to summarize the current knowledge on the variety, species, and structural characteristics of sesquiterpene compounds isolated from Artabotrys plants. Furthermore, it delves into their pharmacological activities and underlying mechanisms, offering a comprehensive foundation for future research.

Diterpenes represent one of the most diverse and structurally complex families of natural products. Among the myriad of diterpenoids, grayanane diterpenes are particularly notable. These terpenes are characterized by their unique 5/7/6/5 tetracyclic system and are exclusive to the Ericaceae family of plants. Renowned for their complex structures and broad spectrum of bioactivities, grayanane diterpenes have become a primary focus in extensive phytochemical and pharmacological research. Recent studies, spanning from 2018 to January 2024, have reported a series of new grayanane diterpenes with unprecedented carbon skeletons. These compounds exhibit various biological properties, including analgesic, antifeedant, anti-inflammatory, and inhibition of protein tyrosine phosphatase 1B (PTP1B). This paper delves into the discovery of 193 newly identified grayanoids, representing 15 distinct carbon skeletons within the Ericaceae family. The study of grayanane diterpenes is not only a deep dive into the complexities of natural product chemistry but also an investigation into potential therapeutic applications. Their unique structures and diverse biological actions make them promising candidates for drug discovery and medicinal applications. The review encompasses their occurrence, distribution, structural features, and biological activities, providing invaluable insights for future pharmacological explorations and research.

Nb2O5 nanoparticles with an average particle size of 10 nm supported on a rhombic dodecahedral metal organic framework (MOF) were successfully synthesized by a facile one-pot hydrothermal reaction and subsequent calcination process. Experimental results demonstrated that the prepared catalyst drastically improved the hydrogen storage behavior of MgH2. 7 wt% Nb2O5@MOF doped MgH2 started to desorb hydrogen at 181.9 °C and 6.2 wt% hydrogen could be released within 2.6 min and 6.3 min at 275 °C and 250 °C, respectively. The fully dehydrogenated composite also displayed excellent hydrogenation by decreasing the onset absorption temperature to 25 °C and taking up 4.9 wt% and 6.5 wt% hydrogen within 6 min at 175 °C and 150 °C, respectively. Moreover, the corresponding activation energy was calculated to be 75.57 ± 4.16 kJ mol−1 for desorption reaction and 51.38 ± 1.09 kJ mol−1 for absorption reaction. After 20 cycles, 0.5 wt% hydrogen capacity was lost for the MgH2+7 wt% Nb2O5@MOF composite, much lower than 1.5 wt% of the MgH2+7 wt% Nb2O5 composite. However, the addition of Nb2O5@MOF had limited effect on reducing the dehydrogenation enthalpy of MgH2. Microstructure analysis revealed that Nb2O5 particles were uniformly distributed on surface of the MgH2 matrix and synergistically improved the hydrogen storage property of MgH2 with MOF.

ABSTRACT The gut microbiota develops specialized local communities along the gastrointestinal (GI) tract of the host. Among these microbes, intestinal bacteria and fungi interact closely with the host and play vital roles in their health. However, the gut biogeography of bacteria and fungi has rarely been studied simultaneously, and their correlations have not been reported. In this study, we collected the intestinal luminal contents from different gastrointestinal regions (the ileum, caecum, and colon) of wild-type female cynomolgus monkeys and performed 16S and 18S rRNA gene sequencing to determine the gut biogeography of bacterial and fungal communities. The richness and diversity of bacteria and fungi increased gradually from the small to the large intestine. The bacterial and fungal composition of ileum differed significantly from that of the colon. Specifically, Bacteroidia and Spirochaetia were abundant in the caecum and colon, while Actinobacteria and Cyanobacteria were enriched in the ileum. As for fungal taxa, the genera Aspergillus, Wallemia, and Cryptococcus_f_Tremellaceae were enriched in the colon, while the genus Fusarium was abundant in the caecum. Consequently, the bacterial and fungal functions of the ileum differed from those of the caecum and colon. Notably, we characterized the correlations between intestinal bacteria and fungi at different gastrointestinal regions of cynomolgus monkeys using Spearman correlation analysis. In summary, our results contribute to a more comprehensive understanding of the composition and function of intestinal bacteria and fungi in non-human primates (NHPs) and reveal their correlations along the longitudinal axis of the intestine. IMPORTANCE Gut microbiota varies along the gastrointestinal (GI) tract and exerts profound influences on the host’s physiology, immunity, and nutrition. Given that gut microbes interact with the host closely and the gastrointestinal function differed from the small to the large intestine, it is essential to characterize the gut biogeography of the microbial community. Here, we focused on intestinal bacteria and fungi in cynomolgus monkeys and determined their spatial distribution along the GI tract by performing 16S and 18S rRNA gene sequencing. The composition and function of bacterial and fungal communities differed significantly at different biogeographic sites of the intestine, and the site-specific correlations between intestinal bacteria and fungi were revealed. Thus, our studies characterized the gut biogeography of bacteria and fungi in NHPs and revealed their site-specific correlations along the GI tract.

Pain is often debilitating, and current treatments are neither universally efficacious nor without risks. Transient receptor potential (TRP) ion channels offer alternative targets for pain relief, but little is known about the regulation or identities of endogenous TRP ligands that affect inflammation and pain. Here, transcriptomic and targeted lipidomic analysis of damaged tissue from the mouse spinal nerve ligation (SNL)-induced chronic pain model revealed a time-dependent increase in Cyp1b1 mRNA and a concurrent accumulation of 8,9-epoxyeicosatrienoic acid (EET) and 19,20-EpDPA post injury. Production of 8,9-EET and 19,20-EpDPA by human/mouse CYP1B1 was confirmed in vitro, and 8,9-EET and 19,20-EpDPA selectively and dose-dependently sensitized and activated TRPA1 in overexpressing HEK-293 cells and Trpa1-expressing/AITC-responsive cultured mouse peptidergic dorsal root ganglia (DRG) neurons. TRPA1 activation by 8,9-EET and 19,20-EpDPA was attenuated by the antagonist A967079, and mouse TRPA1 was more responsive to 8,9-EET and 19,20-EpDPA than human TRPA1. This latter effect mapped to residues Y933, G939, and S921 of TRPA1. Intra-plantar injection of 19,20-EpDPA induced acute mechanical, but not thermal hypersensitivity in mice, which was also blocked by A967079. Similarly, Cyp1b1-knockout mice displayed a reduced chronic pain phenotype following SNL injury. These data suggest that manipulation of the CYP1B1–oxylipin–TRPA1 axis might have therapeutic benefit.
