3. Qualitative and quantitative analysis of leachables from dental composites under different extraction conditions using liquid chromatography coupled to mass spectrometry.

Dental caries is the most widespread form of disease, affecting over 90% of the global population. Amalgam fillings, which have been in use for nearly two centuries, will face an European Union ban by 2025. Although photocomposite fillings are a suitable alternative, health concerns persist because of potential substance release into the oral cavity. This study aimed to evaluate the release of photoinitiator substances and monomers from dental materials into various solvents at different temperatures over 30 days. Cylindrical specimens of the composite resins were submerged in different extraction solutions and incubated at 37 and 50°C. The findings demonstrated that both the extraction solvent and extraction temperature significantly influenced the quantity of leachables (p < 0.05). Furthermore, most leachables were released within the initial days, although some monomers continued to elute for over 30 days. The estimated daily intake was calculated for the worst‐case scenario, confirming the biocompatibility of the composite fillings. The weight loss of dental materials ranged up to 3.5% after 30 days, regardless of the extraction conditions and dental material (p > 0.05). In conclusion, this study contributes to filling several research gaps in the field by addressing the biocompatibility of various dental materials through quantitative and qualitative analyses supported by statistical evaluation. - Abstract
Bakalářská práce se zabývá problematikou sorpce antibiotik na mikroplasty. Antibiotika i mikroplasty jsou v dnešní době vysoce studovaným tématem, kvůli pozitivním i negativním vlastnostem Pro experiment byly vybrány antibiotika se skupiny sulfonamidů a chinolonů. Ze sulfonamidů byly vybrány sulfamethoxazol a trimethoprim a z chinolonů enrofloxacin a ciprofloxacin, které již byly podnětem pro některé studie s mikroplasty. Jako mikroplast byl využit biologicky odbouratelný polyhydroxybutyrát. V laboratoři byly uskutečněny dva experimenty. První měl za cíl sledovat časovou závislost sorpce antibiotik na polyhydroxybutyrát, druhý pak koncentrační. Koncentrační závislost byla následně vyhodnocena pomocí Freundlichovy a Langmuirovy izotermy. K analýzám byla využita metoda kapalinové chromatografie s hmotnostní spektrometrií., The bachelor thesis deals with the issue of sorption of antibiotics on microplastics. Antibiotics and microplastics are now a highly studied topic, due to the positive and negative properties. Antibiotics with groups of sulfonamides and quinolones were selected for the experiment. From sulfonamides, sulfamethoxazole and trimethoprim were selected, and from the quinolones, enrofloxacin and ciprofloxacin, which have already been the impetus for some microplastic studies. Biodegradable polyhydroxybutyrate was used as the microplastic. Two experiments were performed in the laboratory. The first aimed to monitor the time dependence of antibiotic sorption on polyhydroxybutyrate, the second the concentration. The concentration dependence was subsequently evaluated using the Freundlich and Langmuir isotherms. The method of liquid chromatography with mass spectrometry was used for analyzes.

- Abstract
Cieľom tejto diplomovej práce je analýza obsahu vybraných skupín liečiv vo vypestovanom šaláte. V teoretickej časti sa nachádza kapitola venovaná použitým liečivám, ich použitiu, fyzikálno – chemickým vlastnostiam a ich vplyvu na životné prostredie. Ďalšie kapitoly sa venovali analytickým metódam stanovenia liečiv, hydroponickému pestovaniu šalátu siateho a následne niektorým parametrom ekotoxikologických testov. Experimentálna časť začala optimalizáciou extrakčného postupu QuEChERS. Optimalizovaný postup bol následne aplikovaný na hydroponicky vypestované vzorky koreňov a listov šalátu siateho obsahujúce vybrané skupiny liečiv, ktoré boli následne analyzované pomocou kvapalinovej chromatografie s hmotnostnou spektrometriou. Ďalej sa experimentálna časť zaoberala hydroponickým pestovaním šalátu siateho a vyhodnotením základných ekotoxikologických parametrov., The aim of this thesis is to analyze the content of selected groups of drugs in cultivated lettuce. The theoretical part includes a chapter dedicated to the drugs used, their use, physicochemical properties, and their impact on the environment. Other chapters focused on analytical methods for determining drugs, hydroponic cultivation of lettuce, and some parameters of ecotoxicological tests. The experimental part began with the optimization of the QuEChERS extraction procedure. The optimized procedure was then applied to hydroponically grown samples of roots and leaves of lettuce containing selected groups of drugs, which were subsequently analyzed using liquid chromatography – mass spectrometry. The experimental part also addressed the ecotoxicological impact of different drug concentrations on hydroponically grown lettuce.

8. Assessing Lettuce Exposure to a Multipharmaceutical Mixture under Hydroponic Conditions: Findings through LC-ESI-TQ Analysis and Ecotoxicological Assessments.

- Abstract
The escalating global water scarcity demands innovative solutions, one of which is hydroponic vegetable cultivation systems that increasingly use reclaimed wastewater. Nevertheless, even treated wastewater may still harbor various emerging organic contaminants, including pharmaceuticals. This study aimed to comprehensively assess the impact of pharmaceuticals, focusing on bioconcentration factors (BCFs), translocation factors (TFs), pharmaceutical persistence in aqueous environment, ecotoxicological end points, and associated environmental and health risks. Lettuce ( Lactuca sativa ) was cultivated hydroponically throughout its entire growth cycle, exposed to seven distinct concentration levels of contaminants ranging from 0 to 500 μg·L
-1 over a 35-day period. The findings revealed a diverse range of BCFs (2.3 to 880 L·kg-1 ) and TFs (0.019-1.48), suggesting a high potential of pharmaceutical uptake and translocation by L. sativa . The degradation of 20 pharmaceuticals within the water-lettuce system followed first-order degradation kinetics. Substantial ecotoxicological effects on L. sativa were observed, including increased mortality, alterations in root morphology and length, and changes in biomass weight ( p < 0.05). Furthermore, the estimated daily intake of pharmaceuticals through L. sativa consumption suggested considerable health risks, even if lettuce would be one of the many vegetables consumed. It is hypothetical, as the values were calculated. Moreover, this study assessed the environmental risk associated with the emergence of antimicrobial resistance (AMR) in aquatic environments, revealing a significantly high risk of AMR emergence. In conclusion, these findings emphasize the multifaceted challenges posed by pharmaceutical contamination in aquatic environments and the necessity of proactive measures to mitigate associated risks to both environmental and human health., Competing Interests: The authors declare no competing financial interest., (© 2024 The Authors.
