Several long non-coding RNAs (lncRNAs) have recently emerged as potential biomarkers in cancer biology. Background: Myocardial infarction-associated transcript (MIAT) is a long non-coding RNA (lncRNA), which functions in a variety of disorders, like myocardial infarction and diabetic retinopathy. Moreover, recent reports have established that MIAT is upregulated in several types of malignancies and plays a crucial role in tumorigenesis. Therefore, this research aimed to investigate the expression of MIAT in colorectal cancer (CRC) and further evaluate the impact of its knocking-down on the proliferation and migration of the CRC cell. Method: In this case-control experimental study, we evaluated the expression level of MIAT in a series of CRC and marginal tissues using RT-qPCR. Furthermore, the role of MIAT was assessed employing RNA interference (RNAi)-mediated suppressing strategy in CRC-derived cells. Subsequently, colony formation, cell cycle analysis, migration, apoptosis, and senescence assays were done to decipher the influence of MIAT on initiation and progression of CRC. Results: Our findings revealed that MIAT expression is significantly upregulated in high-grade and vascular invasion tumor tissues. Furthermore, MIAT silencing led to G1 arrest in SW116 and SW48 CRC-derived cells. We also found that MIAT inhibition contributed to the induction of apoptosis/cellular senescence as well as the limitation of colony formation capability and cell migration in CRC cells. The obtained findings also showed that MIAT silencing dysregulated the expression of ATM and CHK2 genes known as DNA damage responsive genes. Conclusion: The results of the present study demonstrated that lncRNA MIAT may control CRC cell proliferation and metastasis through regulating DNA damage-responsive pathway and can be noticed as a potential marker for diagnosis, prognosis, and targeted-therapy of high-grade CRC. To evaluate the responses of some important agronomic traits of 14 bread wheat cultivars, a split plot experiment was conducted based on a randomized complete block design under non-stress, moderate and severe salinity stress conditions. In this study, Backcross and Roshan were identified as the most salinity-tolerant cultivars, while Mihan and Shirudi were considered as the most salinity-sensitive cultivars. The proline content, as well as Δ1-pyrroline-5-carboxylate synthase (P5CS) and vacuolar-type H+-ATPase subunit E (W36) gene expression levels, were examined in these cultivars under normal, moderate and high salinity stress conditions. The proline content and P5CS gene expression level increased with a rise in NaCl concentration. Further, a direct relationship was observed between the proline content and P5CS gene expression in all samples. Our results showed that W36 gene expression in Backcross and Roshan cultivars, as the most resistant ones, strongly increased with elevation of the NaCl concentrations. On the other hand, in the sensitive cultivars such as Mihan and Shirudi, small changes were observed in the gene expression levels with rising salinity levels. Additionally, Backcross and Roshan cultivars had the highest proline content as well as P5CS and W36 genes expression levels under moderate and high salinity stress levels. [ABSTRACT FROM AUTHOR]

Background:Nowadays long non-coding RNAs are known as interesting functional part of the transcriptome. LncRNA SNHG6 was reported to be expressed more in breast cancer tissues than non-tumor ones. As a frequent cancer among women, breast cancer treatment needs applied biomarkers for fast prognosis and diagnosis. SNHG6 RNA and its splice variants could be considered as molecular biomarkers for the breast cancer well-timed treatment. Methods:RNA extraction from 35 breast cancer tissues and their relative non-tumor tissues was done and cDNAs of the RNAs were synthesized and then RT-qPCR was performed. Relative expression of SNHG6 202 and 203 was studied in breast cancer samples. Results:The expression patterns of SNHG6 202 and 203 variants were different. Difference in the expression pattern of SNHG6 202 was significantly remarkable in relation to the HER2 status of tumor samples. SNHG6 203 was expressed in tumor and non-tumor tissues differentially and the expression difference was significant. Also, this transcript exhibited significant expression difference in different stages of the studied breast tumor samples. Conclusion:It could be stated that SNHG6 203 transcript might be considered as a prognostic and staging biomarker in breast cancer studies and treatment. [ABSTRACT FROM AUTHOR]
