The German Association for Psychiatry and Psychotherapy (DGPPN) has committed itself to establish a prospective national cohort of patients with major psychiatric disorders, the so-called DGPPN-Cohort. This project will enable the scientific exploitation of high-quality data and biomaterial from psychiatric patients for research. It will be set up using harmonised data sets and procedures for sample generation and guided by transparent rules for data access and data sharing regarding the central research database. While the main focus lies on biological research, it will be open to all kinds of scientific investigations, including epidemiological, clinical or health-service research.

The development and screening of floating-type bait formulations designed to improve the activity of bacterial toxins against larval Anopheles is described. Floating and spreading abilities of carrier particles (wheat flour) were compared using corn oil, lecithin, and two products yielding surface films on water (Arosurf and Liparol). Mixtures containing 1 or 5% Arosurf showed the best spreading abilities on a water surface, but strongly inhibited the ingestion of wheat flour by Anopheles albimanus larvae. Corn oil and lecithin improved spreading satisfactorily at a concentration of 5% and inhibited larval feeding by only 6-25%. To select a suitable concentration of active ingredient in formulations, Bacillus thuringiensis var. israelensis (B.t.i.) primary powder in concentrations ranging from 0.01 to 0.5% was mixed with wheat flour/corn oil mixtures and tested in quantities exceeding the gut volumes of treated larvae. Complete mortality was obtained with concentrations of 0.1% (Anopheles stephensi), 0.2% (An. albimanus), or 0.3% (An. quadrimaculatus) B.t.i. When in 175-liter containers the activity of formulations (5% corn oil, 0.2% B.t.i.) and of toxin suspensions was compared by conventional dosage-mortality regression, formulations were more active by a factor of 68 against An. stephensi, 39 against An. albimanus and 67 against An. quadrimaculatus.

Feeding rates of fourth instar larvae of Aedes vexans were compared by counting substrate filled gut segments after exposure to food or inert particles. Food particles (wheat flour, fishmeal or yeast) were ingested approximately 3 times faster than inert particles (kaolin, pumice or synthetic cellulose). Aqueous fishmeal extract accelerated ingestion of inert particles to the level of ingestion of food particles, demonstrating gustatory stimulation of larvae. Absolute amounts of ingested materials were calculated on the basis of dry weights of guts completely filled with test substrates. At 21 degrees C, 46-74 micrograms of food particles, and 15-33 micrograms of inert particles were ingested per larva during 10 min of exposure. In the subsequent time span, ingestion rates of food particles decreased continuously with satiation of larvae.

Larvae of Culex quinquefasciatus are much more susceptible to the toxin of Bacillus sphaericus than are larvae of Aedes aegypti. In the present study, the rate of ingestion, dissolution, and the cleavage by midgut proteases of the B. sphaericus toxin were compared in larvae of these species to determine whether these factors account for the differences in susceptibility. During filter feeding, larvae of both species removed significant quantities of B. sphaericus toxin from suspensions. Filtration rates for 1 hr, the time at which C. quinquefasciatus exhibited marked intoxication, were higher for A. aegypti (576-713 microliters/larva/hr) than for C. quinquefasciatus (446-544 microliters/larva/hr). Within 24 hr of exposure, A. aegypti larvae ingested 97-99% of the toxin particulates and suffered not more than 10% mortality in suspensions which induced complete mortality in C. quinquefasciatus within 2 hr of exposure. Quantification of the particulate toxin present in larvae after exposure to B. sphaericus suspensions revealed that larvae of both species contained only minor amounts of the toxin, suggesting the larvae had been able to solubilize the toxin after ingestion. Proteases recovered from the feces of larvae cleaved at 43-kDa protein isolated from B. sphaericus toxin extract to 40 kDa in both species. Thus, differences in susceptibility to the B. sphaericus toxin between A. aegypti and C. quinquefasciatus are not due to differences in rates of ingestion, dissolution, or the specificity of proteases.
