Background: Numerous polymorphisms in candidate genes coding for haemostatic system proteins have been proposed as risk factors for thrombosis. Methods: We performed a case-control study of consecutive ischaemic stroke survivors aged ≤45 years, treated at our neurology department from 2006 to 2014. Polymerase chain reaction–restriction fragment length polymorphism identified the following polymorphisms: Thr325Ile and Ala147Thr in TAFI, 4 G/5 G in PAI-1, PLA1/A2 in platelet glycoprotein IIb/IIIa, Glu298Asp in eNOS, and C677 T in 5,10-MTHFR. A multivariate logistic regression analysis was performed to evaluate the independent risk of stroke. Results: 204 cases and 204 age- and sex-matched controls were included in the study. Clinical and genetic variables associated with ischaemic stroke were hypertension (P = .03), tobacco use (P = .02), and the polymorphisms Glu298Asp (genotype: P = .001, allele frequency: P = .001) and C677 T (genotype: P = .01); the Ala147Thr, Thr325IIe, 4 G/5 G, and PLA1/A2 mutations were not associated with ischaemic stroke. The 298Asp (P = .03) and T (P = .01) alleles, hypertension (P = .03), tobacco use (P = .01) and family history of stroke (P = .04) were identified as independent risk factors. Conclusion: The polymorphisms Glu298Asp and C677T, affecting the eNOS and 5,10-MTHFR enzymes, respectively, and smoking, hypertension, and family history of stroke were associated with ischaemic stroke in young Mexican patients; this was not the case for the Thr325Ile, Ala147Thr, 4 G/5 G, and PLA1/A2 polymorphisms of the genes coding for fibrinolytic proteins and platelet receptors. Resumen: Introducción: Diversos polimorfismos en genes candidatos que codifican proteínas del sistema hemostático se han propuesto como factores de riesgo para el desarrollo de trombosis. Métodos: Casos y controles, sobrevivientes de enfermedad vascular cerebral (EVC) isquémico idiopático ≤ 45 años de edad del servicio de Neurología incluidos de manera consecutiva del 2006 al 2014. Por PCR-RFLP se identificaron los polimorfismos: Thr325Ile y Ala147Thr del gen de TAFI, 4 G/5 G del gen de PAI-1, PLA1/A2 del gen de la glicoproteína plaquetaria IIb/IIIa, Glu298Asp del gen de eNOS, y C677 T del gen de la 5,10 MTHFR. Se realizó un análisis multivariado de regresión logística para calcular el riesgo independiente de EVC. Resultados: 204 casos y 204 controles pareados por edad y sexo. Se asoció al polimorfismo Glu298Asp (genotipo p=0.001 y frecuencia alélica p=0.001), C677 T (genotipo p=0.01), hipertensión (p=0.03) y tabaquismo (p=0.02) con la presencia de EVC isquémico, no así para los polimorfismos Ala147Thr, Thr325IIe, 4 G/5 G, y PLA1/A2. Se identificó como factor de riesgo independiente al alelo 298Asp (p = 0.03), T (p = 0.01), hipertensión (p = 0.03), tabaquismo (p = 0.01) y AHFEAT (p = 0.04). Conclusiones: Los polimorfismos Glu298Asp y C677 T de los genes que codifican a la enzima eNOS y 5,10 MTHFR, tabaquismo, hipertensión y AHFEAT, se asociaron a la presencia de EVC isquémico en jóvenes mexicanos, no así el Thr325Ile, Ala147Thr, 4 G/5 G, PLA1/A2 en genes que codifican proteínas del sistema de fibrinólisis y receptores plaquetarios.

El Centro Poblado de Mi Perú en la actualidad cuenta con una población de 43,000 habitantes y posee una superficie total de 1.89 Km2. Para esta localidad, una solución de gestión ambiental municipal, es presentada, la cual está orientada a objetivos, y trabaja bajo el enfoque del desarrollo sostenible. La gestión ambiental municipal es considerada como una importante herramienta de gestión para el municipio, por ello es necesario comprender que es realmente y que beneficios se logran, así como también, reconocer la importancia de incorporar variables y factores ambientales en la gestión municipal. En esta investigación se priorizaron tres aspectos: 1) los diagnósticos Ambientales, para determinar el nivel de calidad ambiental en el Centro Poblado Mi Perú; 2) el marco conceptual y teórico de la Gestión Ambiental Municipal, que rodea la gestión ambiental municipal, sus diferentes instrumentos, el área de aplicación del estudio; 3) el Sistema de Gestión Ambiental propuesto, del cual se presenta su arquitectura, funcionamiento y características. Finalmente se presentan los resultados, conclusiones y recomendaciones obtenidas producto del estudio realizado.

Chronic Myeloid Leukemia (CML) originates in a leukemic stem cell that resides in the bone marrow microenvironment, where they coexist with cellular and non-cellular elements. The vascular microenvironment has been identified as an important element in CML development since an increase in the vascularization has been suggested to be related with poor prognosis; also, using murine models, it has been reported that bone marrow endothelium can regulate the quiescence and proliferation of leukemic stem and progenitor cells. This observation, however, has not been evaluated in primary human cells. In this report, we used a co-culture of primitive (progenitor and stem) CML cells with endothelial colony forming cells (ECFC) as an in vitro model to evaluate the effects of the vascular microenvironment in the leukemic hematopoiesis. Our results show that this interaction allows the in vitro maintenance of primitive CML cells through an inflammatory microenvironment able to regulate the proliferation of progenitor cells and the permanence in a quiescent state of leukemic stem cells.

En México hay muy pocas universidades públicas que obtienen certificados de patente y muy pocas de ellas llegan al sector empresarial. El objetivo de este documento es describir y explicar el caso del licenciamiento de una patente académica, aplicando como marco teórico-conceptual la Teoría de Campos de Pierre Bourdieu. El análisis se centra en la diferenciación de perspectivas e intereses entre los actores involucrados: el inventor, la universidad licenciante y el empresario licenciatario. Al final, se reflexiona sobre el impacto social de las invenciones científicas (como la que aquí se presenta) cuando son colocadas en el mercado.

Endothelial cells (ECs) are an important component of the blood coagulation system because it maintains blood fluid. Because in patients with venous thromboembolic disease (VTD) a thrombophilic condition is not found sometimes, we investigated if endothelial colony-forming cells (ECFCs) from these patients have biological and functional abnormalities.Human mononuclear cells (MNCs) were obtained from peripheral blood from patients with VTD and controls to obtain ECFCs. These cells were assayed for their immunophenotype and electron microscopy characteristics and their ability to form capillary-like structures and to produce pro-inflammatory and pro-angiogenic cytokines and reactive oxygen species (ROS).ECFCs appeared at 7 and 21 days of culture in VTD patients and controls, respectively. ECFCs increased 8-fold in patients and emerged 1 week earlier. No differences in the size of the colonies of ECFCs were found. Numbers and time of appearance of ECFCs was different between groups. ECFC-derived ECs (ECFC-ECs) of both groups expressed CD31, CD34, CD146, and CD-309 but none expressed CD45, CD14, or CD90. Interest CD34 was highly expressed in ECFC-ECs from patients. In both groups, ECFC-ECs showed similar capacity to form capillary-like structures but ECFC-ECs from patients had significant abnormalities in the mitochondrial membrane. We found a significant increase in ROS production in ECFC-ECs from patients. There were significant differences in cytokine profiles between VTD patients and controls.We found a dysfunctional state in ECFC from VTD patients resembling some characteristics of dysfunctional ECs. These findings may help to understand some pathophysiological aspects of VTD.

Venous thromboembolic disease (VTD) is a public health problem. We recently reported that endothelial colony-forming cells (ECFCs) derived from endothelial cells (EC) (ECFC-ECs) from patients with VTD have a dysfunctional state. For this study, we proposed that a dysfunctional status of these cells generates a reduction of its proliferative ability, which is also associated with senescence and reactive oxygen species (ROS).Human mononuclear cells (MNCs) were obtained from peripheral blood from 40 healthy human volunteers (controls) and 50 patients with VTD matched by age (20-50 years) and sex to obtain ECFCs. We assayed their proliferative ability with plasma of patients and controls and supernatants of cultures from ECFC-ECs, senescence-associated β-galactosidase (SA-β-gal), ROS, and expression of ephrin-B2/Eph-B4 receptor. Compared with cells from controls, cells from VTD patients showed an 8-fold increase of ECFCs that emerged 1 week earlier, reduced proliferation at long term (39%) and, in passages 4 and 10, a highly senescent rate (30±1.05% vs. 91.3±15.07%, respectively) with an increase of ROS and impaired expression of ephrin-B2/Eph-4 genes. Proliferation potential of cells from VTD patients was reduced in endothelial medium [1.4±0.22 doubling population (DP)], control plasma (1.18±0.31 DP), or plasma from VTD patients (1.65±0.27 DP).As compared with controls, ECFC-ECs from individuals with VTD have higher oxidative stress, proliferation stress, cellular senescence, and low proliferative potential. These findings suggest that patients with a history of VTD are ECFC-ECs dysfunctional that could be associated to permanent risk for new thrombotic events.

The genus Hevea is native to the Brazilian Amazon forest, and H. brasiliensis, popularly known as the rubber tree, is the main source of raw material for the production of natural rubber in the world. To characterize the fungal communities that inhabit the tissues of this species, a total of 550 endophytic fungi were isolated, of which 212 were from native trees located in the state of Acre and 338 in the state of Amazonas. These fungi were grouped into 115 OTUs. The phylum Ascomycota was dominant (93%) and the most abundant isolates belonged to the families Glomerallaceae (genus Colletotrichum), Diaporthaceae (genus Diaporthe), Nectriaceae (genus Fusarium), Hypocreaceae (genus Trichoderma), and Trichocomaceae (genus Penicillium). The diversity and richness of fungi in the state of Acre did not differ significantly between leaves, stems, and roots; however, in the state of Amazonas, the diversity was greater in the stems and roots than in the leaves. In general, the diversity of endophytic fungi was greater in the state of Acre. Comparisons between communities of fungi showed more differences between those that inhabit different tissues than in different states. Furthermore, the fungal communities of roots and stems were similar, and both these were dissimilar from leaves. Isolates identified in this study will be tested for their potential to produce metabolites of interest for industry and for the sustainable development of agribusiness.

Endothelial colony-forming cells (ECFCs) contribute to postnatal vasculogenesis. In venous thromboembolic disease (VTD), they are functionally abnormal and produce high concentrations of TNF-α.To analyze the TNF-α signaling pathway and its relationship with the expression of cell-cycle regulators.Mononuclear cells (MNCs) were collected from the peripheral blood of 20 healthy human volunteers (controls) and 30 patients with VTD matched by age (20-50 years) and sex to obtain ECFCs. We analyzed the relative quantification of the gene transcripts of TNF, NFkB1, PLAU, HMOX1, GSS, eNOS, CDKN1A, and CDKN1B through quantitative RT-PCR (qRT-PCR assays). Identification of NF-κB and activated targets of each pathway: NF-κB (Ser536); IκBα (Ser32/Ser36); p38 (Thr180/Tyr182) JNK (Thr183/Tyr185), p53 and cell-cycle regulators: p16, p18, p21, p27, p57, Cyclin D, Cyclin E, Cyclin A, Cyclin B, CDK2, CDK4; cell-cycle status was determined by KI-67 and 7-AAD. Cells were analyzed with flow cytometry and the FlowJo vX software.In ECFCs from VTD patients, TNF-α receptor and NFkB were overexpressed and hyper-phosphorylated; eNOS and HMOX1 were down-regulated; cell-cycle regulators (p53, p18, p21) were elevated. In addition, the cell cycle was locked in the G2 phase.Our results strongly suggest that these molecular alterations in the pathway of TNF-α and cell cycle regulation induce endothelial dysfunction, reduced proliferation potential and vascular regeneration, and consequently, the occurrence of new thrombotic events.

Thyrotoxicosis and diabetic ketoacidosis (DKA) both may present as endocrine emergencies and may have devastating consequences if not diagnosed and managed promptly and effectively. The combination of diabetes mellitus (DM) with thyrotoxicosis is well known, and one condition usually precedes the other. Furthermore, thyrotoxicosis is complicated by some degree of cardiomyopathy in at least 5% de patients; but the coexistence of DKA, thyroxin (T4) toxicosis, and acute cardiomyopathy is extremely rare. We describe a case of a man, previously diagnosed with DM but with no past history of thyroid disease, who presented with shock and severe DKA that did not improve despite optimal therapy. The patient evolved with acute pulmonary edema, elevated troponin levels, severe left ventricular systolic dysfunction, and clinical and laboratory evidence of thyroxin (T4) toxicosis and thyrotoxic cardiomyopathy. Subsequently, the patient evolved favorably with general support and appropriate therapy for DKA and thyrotoxicosis (hydrocortisone, methimazole, Lugol’s solution) and was discharged a few days later.

No cabe la menor duda que las redes sociales son parte esencial de la vida, gracias a estas se puede contactar con gente alrededor del mundo en segundos. Y no solo se han vuelto el canal más eficiente, y óptimo para poder comunicarse sin importar la distancia; también son el medio de tráfico masivo de información más grande, jamás imaginado desde la creación de internet. Solo basta con contar con un celular con cámara, y señal de internet, para trasmitir algo y que lo puedan ver en cualquier parte del mundo. Compartir todo tipo de información es el pan de cada día de las redes sociales, no solo información privada entre dos personas en un chat en particular; asimismo información pública de toda clase, desde información gubernamental, hasta datos personales, sin autorización previa de la persona, que muchas veces viola las reglas y políticas con las que cuentan las redes sociales. Se puede decir que aquel que no cuente con redes sociales podría tener una gran desventaja frente a los demás, no solo perdería la oportunidad de comunicarse lo más rápido posible con un familiar o amigo que este en otra latitud. Cancer is a major cause of death in children worldwide, with B-lineage cell acute lymphoblastic leukemia (B-ALL) being the most frequent childhood malignancy. Relapse, treatment failure and organ infiltration worsen the prognosis, warranting a better understanding of the implicated mechanisms. Cortactin is an actin-binding protein involved in cell adhesion and migration that is overexpressed in many solid tumors and in adult B-cell chronic lymphocytic leukemia. Here, we investigated cortactin expression and potential impact on infiltration and disease prognosis in childhood B-ALL. B-ALL cell lines and precursor cells from bone marrow (BM) and cerebrospinal fluid (CSF) of B-ALL patients indeed overexpressed cortactin. In CXCL12-induced transendothelial migration assays, transmigrated B-ALL cells had highest cortactin expression. In xenotransplantation models, only cortactinhigh-leukemic cells infiltrated lungs, brain, and testis; and they colonized more easily hypoxic BM organoids. Importantly, cortactin-depleted B-ALL cells were significantly less efficient in transendothelial migration, organ infiltration and BM colonization. Clinical data highlighted a significant correlation between high cortactin levels and BM relapse in drug-resistant high-risk B-ALL patients. Our results emphasize the importance of cortactin in B-ALL organ infiltration and BM relapse and its potential as diagnostic tool to identify high-risk patients and optimize their treatments.

Scoliosis and lordosis are medical conditions, which present an atypical curvature of the spine. These deformations can be easily diagnosed, altogether with the degree of their severity through orthopedic measurement processes and clinical reviews. However, the preformed studies which classify the severity of these deformations are, in their majority, empirical and present great opportunity for improvement. Therefore, they are based on radiographies that offer a clear image of the spine, but present the problem of a constant patient’s exposition to ionizing radiation and as a consequence an increment of potential hazards for health. This also means, results are subject to the physician’s interpretation and expertise. The present work intends to describe and analyze the design and function of a mechanical system that is capable of significantly reducing the radiological exposure of patients with scoliosis and lordosis. The main goal of this device is to diagnose both deformations by obtaining the angles of curvature of the patient’s spine, using a method that does not depend entirely on human interpretation.

- Abstract
This paper describes the implementation of an IoT gateway that allows the exchange of data through different wireless technologies and forwarding of such data to the Internet. The proposed gateway has important advantages such as: supporting for multiprotocol interconnectivity; the remote configuration of wireless nodes for sensor and actuators management; a flexible algorithm to translate the data obtained by sensors into a uniform format for transmission to a cloud server; low energy consumption due to efficient data transfer over the MQTT protocol. In order to demonstrate the usefulness of the developed gateway, a proof-of-concept test was implemented. The implemented scenario consists of 2 wireless nodes responsible for sensing environmental variables and transmitting data to the gateway node through different communication protocols. The obtained results show the feasibility for simultaneous data transmission from the remote wireless nodes to the gateway. Metrics on energy consumption in the devices are also presented., En este artículo se describe la implementación de un gateway IoT que permite el intercambio de datos a través de diferentes tecnologías inalámbricas y el reenvío de dichos datos a Internet. El gateway propuesto incorpora importantes ventajas como: la interconectividad con diferentes protocolos; permite la configuración remota de nodos inalámbricos para la gestión de sensores y actuadores; utiliza un algoritmo flexible para traducir los datos obtenidos por los sensores a un formato uniforme para la transmisión hacia un servidor en la nube; utiliza el protocolo MQTT, el cual permite la difusión de datos de forma simple para minimizar los recursos utilizados del gateway. Con el fin de demostrar el funcionamiento del gateway desarrollado, se implementó una prueba de concepto compuesta por 2 nodos inalámbricos encargados de sensar 6 variables ambientales y transmitiendo dichos datos al gateway a través de diferentes protocolos de comunicación. Los resultados presentados muestran la transmisión de la información de manera simultánea, desde los nodos inalámbricos remotos hacia el gateway. Además, se presentan métricas sobre el consumo de energía en los diferentes dispositivos que componen el escenario desarrollado.

Acute lymphoblastic leukemia (ALL) is the most frequent malignancy of childhood. Substantial progress on understanding the cell hierarchy within ALL bone marrow (BM) has been recorded in the last few years, suggesting that both primitive cell fractions and committed lymphoid blasts with immature stem cell-like properties contain leukemia-initiating cells. Nevertheless, the biology of the early progenitors that initiate the lymphoid program remains elusive. The aim of the present study was to investigate the ability of lymphoid progenitors from B-cell precursor ALL BM to proliferate and undergo multilineage differentiation. By phenotype analyses, in vitro proliferation assays, and controlled culture systems, the lymphoid differentiation potentials were evaluated in BM primitive populations from B-cell precursor ALL pediatric patients. When compared to their normal counterparts, functional stem and progenitor cell contents were substantially reduced in ALL BM. Moreover, neither B nor NK or dendritic lymphoid-cell populations developed recurrently from highly purified ALL-lymphoid progenitors, and their proliferation and cell cycle status revealed limited proliferative capacity. Interestingly, a number of quiescence-associated transcription factors were elevated, including the transcriptional repressor Gfi-1, which was highly expressed in primitive CD34+ cells. Together, our findings reveal major functional defects in the primitive hematopoietic component of ALL BM. A possible contribution of high levels of Gfi-1 expression in the regulation of the stem/progenitor cell biology is suggested.
