The aim of this study is to make a differential diagnosis and prognosis of the ampullary adenocarcinoma subtypes. We also investigated the role of prognostic markers PD-1 and PD-L1, and epidermal growth factor receptor (EGFR).Local or locally advanced stage ampullary adenocarcinoma patients who had undergone pancreaticoduodenectomy at the time of diagnosis were included. MUC1,MUC2, MUC5AC, CDX2, CK7, CK20, PD-1, and PDL-1 were analysed immunohistochemically, and EGFR was analysed by real-time polymerase chain reaction.According to histopathological and immunohistochemical evaluation, we found27 patients as pancreatobiliary type and 56 patients as intestinal type adenocarcinoma. The median survival of patients with intestinal and pancreatobiliary typeadenocarcinoma was 23 months and 76 months (p = 0.201), respectively.When the survival of PD1-positive (n = 23) and PD-L1-positive (n = 18) patientswere compared with the patients with negative staining (n = 60, n = 65), no significant difference was found. Epidermal growth factor receptor mutation was detected in a total of 6 patients, and 5 of these 6 mutations were shown in intestinaltype tumours and one in a pancreatobiliary type tumour. A significant differencewas determined in terms of overall survival for the patients with EGFR mutationscompared to those without (p = 0.008). In conclusion, we could reveal the prognostic significance of EGFR mutation, which is also a target molecule.

Purpose: Determination of proteomic differences plays an important role in biomarker investigations. Dueto its heterogenic molecular background, identification of certain biomarkers is still a demand both fordiagnosis and prognosis of neuroblastoma. In this study, it is aimed to identify markerproteins/mechanisms that may play role in neuroblastoma prognosis.Material and Methods: Real-time PCR analyses were performed for 2p24.3, 11q23, 1p36 and 17q25status from tumor samples of the patients to determine the risk groups. A proteomic approach was usedfor different risk groups of the disease by using matrix-assisted laser desorption ionization–time of flight(MALDI-TOF/TOF) approach. Mononuclear cell pools from blood samples of patients for different riskgroups were constructed and protein expression changes for different groups were identified.Results: Manganese-superoxide dismutase (SOD2) protein was found to significantly increase in highrisk group of neuroblastoma patients.Conclusion: Our results showed that SOD2 may play an important role in neuroblastoma progressionand be a candidate prognostic peripheral blood marker for neuroblastoma patients.

Boron and their derived molecules have prevention or treatment potential against prostate cancer. In this study, we aim to investigate the effects of Boric acid (BA) and Disodium Pentaborate Dechydrate (DPD) in metastatic prostate cancer cells such as DU-145 which is brain metastatic prostate cancer, and PC3 which is bone metastatic prostate cancer. Metastatic human prostate cancer cell lines, PC-3 and DU-145, were used to show whether inhibition effects of BA and DPD on prostate cancer cells in this study. BA and DPD were applied for 24 hours to the cells. Cell viability determination was performed using WST-1 assay. Apoptotic cell death was evaluated with Annexin-V/PI flow cytometric analysis and caspase-3 expression immunohistochemically. A wound healing assay was also used to measure cancer cell migration after exposure to BA and DPD. Applying BA and DPD made inhibition of cell proliferation in both BA (1 mM) and DPD (7 mM) at 24 h. The results of Annexin-V/PI showed that DPD induced higher levels of apoptosis than BA in both prostate cancer cells. Caspase-3 expressions were also higher than BA with DPD in both metastatic prostate cancer cells. We evaluated cell migration using a wound healing assay and the result showed that cell migration was inhibited with BA and DPD in both cells. Both BA and DPD inhibited the cell viability of metastatic prostate cancer cells. Apoptotic cell death with applying DPP had a higher rate than BA treatment. Moreover, BA and DPD inhibited cell migration in both cells when we compared them with control. This study’s results showed that BA and DPD of boron derivates significantly induced cells to apoptosis and the migration was inhibited by the derived form of boron in metastatic prostate cancer cells.

Background: Tumor heterogeneity describesthe differences between cancer cells in the same tumor sample.Neuroblastoma(NB) is a type of cancer where tumor heterogeneity complicates its treatment.This study aims to explore the role of molecular heterogeneity detected by routinemolecular tests in NB.Methods: Seventy-one patients were included in the study. NBsamples were chosen among 1300 NB samples that were evaluated using moleculartests between 2012-2020 according to the guidelines of Turkish PediatricOncology Group protocol. Molecularinvestigations were performed (total 142 samples) obtained from two differentareas of the tumor (synchronous) or at two different times (metachronous).Heterogeneity was questioned for five tests: MYCN amplification, 1p36LOH, 11q23 deletion and 17q25 gain (identifiedwith real-time PCR) and DNA ploidy (identified with flow cytometry). Results: Heterogeneity was observed for MYCN in 22.53%, for 1p36LOH in 36.62%, for11q23del in 29.58%, and for 17q25 gain in 40.85% of cases, while DNA ploidy was heterogeneous in 36.4% of cases. Molecular heterogeneity did not show statisticaldifference among metachronous and synchronous cases. High-risk cases more frequently displayed molecularheterogeneity without any statistically significant difference between bothgroups.Conclusions: Our findings support the fact that molecularheterogeneity either exists in different areas of a tumor or seen in the sametumor at different times. It will be beneficial to perform more than onemolecular analysis on the tumor tissue specimens. In addition, recurrences or re-biopsy specimensfrom metachronous metastases shall be re-evaluated using molecular tests in casesof NB. &nbsp

Giriş:Nöroblastom, tüm çocukluk çağıkanserlerinin %8-10’unu oluşturan çocukluk çağının en sık görülenekstrakraniyal solid tümörüdür. Retinoik asid (RA) nöroblastom tedavisindekullanılan diferansiye edici ajandır. Nöroblastomun ana kemoterapötik ajanı isesisplatindir (CDDP). Eksozomlarve içerdikleri mikrornalar (miRNA) ideal bir biyobelirteç yanında tedavi ajanıolarak kullanımları da mümkündür. miRNA’lar küçük kodlamayan tek iplikliRNA’lar olup, 6-8 adet genin ekspresyonunu düzenleyen epigenetik moleküllerdir.Bu çalışmanın amacı; nöroblastomhücrelerinde diferansiasyon süreçlerinde etkili olduğu bildirilen miRNA’ların,retinoik asit, sisplatin ve retinoik asit ile sisplatin kombinasyonu ileeksozom aracılı olarak ekspresyonlarının değişip değişmediğininbelirlenmesiydi.Yöntem:İnsan SH-SY5Y nöroblastom hücrelerine RA (1uM-5gün)uygulaması ile diferansiye olmaları sağlandı. Diferansiasyon sonrasında hücrelereCDDP (20uM-24saat) eklenmesi ile RA-CDDP kombine tedavi modeli uygulandı.Kontrol, RA, CDDP ve RA-CDDP uygulanmış hücrelerden eksozomlar ve onlardan damiRNA’lar izole edildi. Hücrelerden elde edilen eksozom aracılı miRNA(miR-132-5p, miR-490-3p, miR-34a ve miR-323a-5p) ekpsresyonları RT-PCR yöntemiile çalışıldı. Sonuç:CDDP uygulaması ile hücrelerde miR-323a-5p ekspresyonukontrole göre 100 kat artış gösterirken, RA grubuna göre 9 kat ve RA-CDDPgrubuna kıyasla da 3 kat artış gösterdi. miR-323a-5p ekspresyonu RA grubundakontrole göre 5 kat, RA-CDDP grubuna göre ise 30 kat ekspresyonu arttı. RAuygulaması ile miR-34a, miR-132-5p ve miR-490-5p ekspresyonları değişmedi. CDDPve RA-CDDP grubunda miR-34a ekspresyonu kontrole göre 1 kat arttı. miR-490-5p ekspresyonu CDDP ile kontrolegöre değişmezken, RA-CDDP ile 7 kat artış gösterdi.Tartışma:NB hücrelerinin RAaracılı diferansiasyonu ve CDDP ile tedavide ve tek başlarına uygulamalarındada hücrelerden eksozom aracılı olarak miRNA ekspresyon düzeylerini değiştirdiğisaptandı. Çalışma sonuçları nöroblastomun tedavi süreçlerinde eksozom aracılıolarak miRNA ekspresyon değişimi yolu ile birçok hedef genin ekspresyonunun dakontrol edilebileceğine işaret etmektedir. Nöroblastomda diferansiasyonsürecinde etkili olan miRNA’ların eksozom aracılı belirlenmesi tedaviyanıtlarının değerlendirilmesi açısından kullanışlı olabilir. Kliniktenöroblastom tedavisinin uygulanması ve izleminde eksozom aracılı mikroRNA genekspresyon paternlerinin belirlenmesi özellikle epigenetik modifikasyonlarayeni bakış açısı getirebilecektir.&nbsp

Sisplatin, birçok çocukluk çağı kanserinin tedavisinde kullanılan bir kemoterapötik ajandır. DNA platinizasyonu üzerinden etkiliolan bu alkilleyici ajanın en önemli yan etkileri nefrotoksisite, ototoksisite ve nörotoksisitedir. Sisplatin ototoksisitesi yaygın olarak sensörinöral, bilateral, geri dönüşümsüz ve ilerleyici işitme kaybına neden olmaktadır. Sisplatin kaynaklı işitme kaybı özellikle çok küçükçocuklarda, dil ediniminde bozulmaya, öğrenme ve psikolojik gelişimde zorluklara ve hayatlarının geri kalanında onları etkileyecek olansosyal işlevsellikte azalmaya yol açtığı için özellikle önemlidir. Bu çalışmanın amacı, çocukluk çağı kanserlerinde sisplatin ototoksisitesini önceden öngörmemizi sağlayabilecek olan biyobelirteçlerin araştırılmasıdır.

AIM: It was aimed to evaluate in-vivo whether darbepoetin-alpha (DPO) application has a preventive role in the cisplatin-induced ototoxic effect.MATERIALS and METHODS: In the study, four groups were formed using 28 Wistar albino rats. Group 1 (intraperitoneal) IP saline given control group, Group 2 cisplatin (16mg/kg IP single dose), Group 3 DPO (25 μg/kg IP), and Group 4 is the group in which a single dose of 25 μg/kg IP DPO was administered 24 hours before and half an hour after cisplatin administration. Distortion product otoacoustic emission (DPOAE) and brainstem auditory evoked potentials (BAEP) were performed on rats before agent administration and on the 7th day of the experiment. After hearing measurements, the rats were sacrificed. Apoptotic cell death in ear tissue, caspase-3, -8, -9 expression, Inducible Nitric Oxide Synthase (iNOS), Neuronal Nitric Oxide Synthase (nNOS) expression levels, antioxidant Nrf2 (Nuclear factor (erythroid-derived 2)) - like 2) and related Heme oxygenase-1 (HO-1), NAD(P)H quinone oxidoreductase 1 (NQQ1) were studied by immunohistochemical method. Serum glutathione (GSH) and anti-inflammatory TNF-α and IL-1β protein levels were determined using ELISA kits.RESULTS: The hearing loss was detected at all frequencies showing ototoxic effects compared to control in hearing measurements with cisplatin application. In the examinations performed at the immunohistochemical tissue level, structural changes in the inner ear, necrosis, and necroptosis in the brain and nerve tissues were detected in the cisplatin-administered group. The preventive effects of darbepoetin on the inner ear and brain damage induced by cisplatin are detected by apoptotic protein expressions and oxidative stress-related markers, iNOS and nNOS, Nrf-2 and related HO-1, NQO1, serum GSH, and TNF-α and IL-1β proteins.CONCLUSION: In this study, the protective effects of DPO in ototoxicity caused by cisplatin were demonstrated by reducing apoptosis, increasing antioxidant Nrf-2 and target proteins and glutathione levels, and through anti-inflammatory proteins., Amaç: Darbepoetin-alfa (DPO) uygulamasının sisplatine bağlı ototoksik etkiyi önleyici rolü olup olmadığının in-vivo olarak değerlendirilmesi amaçlandı. Materyal metod: Çalışmada 28 adet wistar albino rat kullanılarak 4 grup oluşturuldu. Grup 1 (intraperitoneal) IP salin verilen kontrol grubu, Grup 2 sisplatin (16mg/kg IP tek doz), Grup 3 DPO (25 μg/kg IP), Grup 4 sisplatin verilmeden 24 saat önce ve verildikten yarım saat sonra 25 μg/kg tek doz IP DPO uygulanan grup. Ratlara ajan uygulamalarından önce ve deneyin 7. gününde distorsiyon ürünü otoakustik emisyon (DPOAE) ve işitsel uyarılmış beyin sapı potansiyelleri (ABR) yapıldı. İşitme ölçümleri sonrasında ratlar sakrifiye edildi. Apoptotik hücre ölümü kulak dokusunda kaspaz-3, -8, -9 ve Nrf2 ve Hem oksijenaz-1 (HO-1), NQQ1, Glutatyon (GST) gibi Nrf2 ilişkili antioksidan ve iNOS, nNOS ekspresyon düzeyleri immünohistokimyasal yöntemle çalışıldı. Serum glutatyon (GSH) ve anti-inflamatuar TNF-α ve IL-1 protein düzeyleri ELISA kitleri kullanılarak belirlendi. Bulgular: Sisplatin uygulaması ile işitme ölçümlerinde kontrole göre ototoksik etkileri gösteren tüm frekanslarda işitme kaybı saptandı. İmmunohistokimyasal doku düzeyinde gerçekleştirilen incelemelerde sisplatin uygulanan grupta iç kulakta strüktürel değişiklikler, beyin ve sinir dokularında nekroz ve nekroptoz saptandı. Darbepoetinin sisplatinin ortaya çıkardığı iç kulak ve beyin hasarlarını önleyici etkileri hem apoptotik protein ekspresyonları hem de oksidatif stres ile ilişkili belirteçler olan Nrf-2 ve ilişkili HO-1, NQO1, iNOS ve nNOS, GST ve anti-inflamatuar proteinleri üzerinden saptandı.Sonuç: Bu çalışmada sisplatinin oluşturduğu ototoksisitede DPO’in koruyucu etkileri apoptozun azaltılması, antioksidan Nrf-2 ve hedef proteinler ve glutatyon düzeylerinin artışı yanında anti-inflamatuar proteinler üzerinden geliştiği gösterilmiştir.

RNA-SequencingTechnologies for Gaining New Insights About Pediatric Cancers

Evaluation of Yap-1 in Risk Classes and Prognosis inNeuroblastoma

Objectives: The present study aims to evaluate the relationship between microribonucleic acid (miRNA) and target gene expressions with clinical and histopathological data in ovarian cancer. Methods: We evaluated 96 archival samples of paraffin-embedded tissue. Some potentially significant miRNA and target gene expressions were evaluated in different histopathological characteristics. These were quantified using realtime-polymerase chain reaction (RT-PCR) in tumor and normal tissue. In miRNA expressions, twofold changes are accepted as significant. Results: According to histopathological groups, 38 (39.6%) were endometroid adenocarcinoma, 11 (11.5%) were borderline serous, 29 (30.2%) were serous, and 18 (18.8%) were mucinous carcinoma. When evaluated according to their stages, 26 (27.1%) patients were stage 1A/1B. A relationship was found between miR200a and miR200c and histopathologic groups, between miR141 and estrogen receptors, between CXCL1 and survival status, and between KEAP1 and ki67. Additionally, miR200a in endometrial and miR200c in mucinous adenocarcinoma were overexpressed. When the relationship between all miRNAs and histopathological groups was evaluated, a significant change was found only in miR200c expression. It was significantly higher in serous than endometrial tumors and significantly higher in mucinous than endometroid tumors. Conclusion: These suggested that miR200a and 200c expressions might be useful for the evaluation of histopathological subgroups of ovarian cancer.

Amaç:İdiyopatik normal basınçlı hidrosefali (iNBH), yaşlı erişkinlerde majör nörodejeneratif bozukluklarla benzer klinikbulguları paylaşan ve bu majör nörodejeneratif bozukluklara eşlik eden, bilişsel ve yürüme bozukluğunun önde gelengeri dönüşümlü bir nedenidir. Bu çalışma ile iNBH ayırıcı tanısında Alzheimer hastalığı (AH)-beyin omurilik sıvısı (BOS)biyobelirteçlerinin yararlı olup olmayacağını araştırmayı amaçladık.Gereç ve Yöntem:Amiloid-beta (Aß) 42 ve 40, toplam tau (t-tau), fosforile tau (p-tau) düzeyi, 192 ardışık BOS örneğinde (iNPH (n=80),AH ( n=48), frontotemporal demans (FTD) (n=34), Parkinson hastalığı demansı ve Lewy cisimcikli demanstan oluşanLewy cisim hastalıkları (LBH) (n=30)) ELISA ile ölçüldü.Bulgular:Çalışma popülasyonunun yaş ortalaması 75.6±7.7 yıl olup, %54.2si kadındı. iNBH hastalarında, AH ve LCH hastalarınagöre BOS Aß42 seviyeleri anlamlı olarak daha yüksekti ve p-tau ve t-tau seviyeleri daha düşüktü. Ek olarak, iNBHhastaları, FTD hastalarına göre anlamlı ölçüde daha yüksek t-tau seviyelerine sahipti. Yaş ve cinsiyete göre ayarlanmışmultinominal regresyon analizi, Aß42 seviyesi bir standart sapma (SD) arttığında iNBHye göre AH olma ihtimalinin%37 azaldığını ve LCHa sahip olma ihtimalinin de %47 azaldığını ortaya koydu. iNPHye göre LCHa sahip olmaolasılığının, p-tau seviyesi 1SD arttığında %76 artttığı gösterildi. Bir hastanın t-tau değeri 1SD arttığında NBHye göreLCH’a sahip olma olasılığı 2.5 kat, AH’ye sahip olma olasılığı 2.1 kat daha fazladır.Sonuç:Sonuçlarımız, özellikle azalmış BOS t-tau düzeyi başta olmak üzere, BOS Aβ42, p-tau ve t-tau düzeylerinin, iNBHyiLCHlardan ayırt etmede potansiyel değere sahip olduğunu göstermektedir. Ayrıca sonuçlarımız iNBHde, AHye göret-tau düzeyinin daha düşük ve Aβ42 düzeyinin daha yüksek olduğunu bildiren önceki çalışmaları doğrulamaktadır.

Giriş: Pediatrik Burkitt lenfoma (pBL) çocuklarda en sık gözlemlenennon-Hodgkin lenfoma türüdür. Oldukça hızlı büyüyen ve agresif karakteristiğesahip pBL’nin tanısının hızlı biçimde konulup tedaviye kısa süredebaşlanması gerekmektedir. pBL tanısının konulabilmesi için uygulananve tümör biyopsi temini içeren invazif yöntemlerin yerini alabilecek noninvaziftanı yöntemlerine ihtiyaç vardır. miRNA’ların kanser hastalarınınplazma örneklerinde yüksek ifade edilmesi durumunda non-invazif tanıyıgeliştirme potansiyelleri bulunmaktadır. pBL hasta plazmalarındaki miRNA’larınifade profilleri ve pBL tanısını geliştirme potansiyelleri öncedenincelenmemiştir. Bu çalışmada pBL hasta plazmalarında ifadesi artan dolaşımsalserbest ve ekzozomal miRNA’lar miRNA-seq ile belirlenerek miRNAseviyelerinin non-invazif (invazif olmayan) tanı biyobelirteç potansiyellerideğerlendirilmiştir.Yöntem: Tanı esnasında 17 pBL olgusuna ait tümör doku ve çevreselkan örnekleri yanı sıra kontrol grubu olarak 15 obstrüktif uyku apnesi(OSA) olgusuna ait tonsil doku ve çevresel kan örnekleri temin edildi.FFPE tümör, dolaşımsal serbest ve ekzozomal miRNA izolasyonları uygunQiagen kitleriyle gerçekleştirildi. OSA tonsil dokularında IgD−/CD38+/CD77+ fenotipli sentroblast hücreleri FACS ile ayrıştırıldıktan sonra totalmiRNA örnekleri miRNeasy Micro Kit (Qiagen, Almanya) kullanılarak izoleedildi. Agilent 2100 BioAnalyzer cihazıyla miRNA örneklerinin boyutve konsantrasyonları değerlendirilerek NUGEN Ovation® SoLo RNA-SeqSystem ile miRNA-seq kütüphaneleri hazırlandı. NovaSeq 6000 yeni nesildizileme sistemi aracılığıyla tek uçlu, 50 bç uzunluğunda ve her bir miRNAörneği için ~10 milyon adet NGS okuması (İng. reads) elde edildi. Kontrolgrubuna kıyasla istatistiksel olarak anlamlı bir farkla ifade edilen miRNA’larmiRDeep2 programını içeren biyoinformatik iş akışı ile belirlendi. İfadesiartan miRNA’ların tanısal potansiyelleri easyROC (sürüm: 1.3.1) programıylaalıcı işletim karakteristiği (İng. receiver operating characteristic[ROC]) eğrisi altındaki alan (AUC) belirlenerek değerlendirildi. miRNA-seqsonuçlarının çapraz teyidi seçili miRNA’ların qRT-PCR deneyleri ve doğrusalregresyon analizleriyle gerçekleştirildi.Sonuçlar: pBL olgularındaki total dolaşımsal serbest miRNA konsantrasyonlarıkontrol grubu örneklere kıyasla anlamlı biçimde yüksekti. miRNAseqanalizleriyle tümör doku örneklerinde toplam 409 adet miRNA’nınifadesinin istatistiksel olarak anlamlı (log2FC>1, FDRbelirlendi. Plazma örneklerinde ise kontrol grubuna kıyasla 35 dolaşımsalserbest ve 128 dolaşımsal ekzozomal miRNA’nın ifadesinde artış gözlemlendi.pBL tümör dokularında ifadesi yüksek olan miRNA’ların ROC analizlerihsa-miR-143-5p, hsa-miR-654-3p, ve hsa-miR-10400-5p ifade seviyelerininpBL için tanısal biyobelirteçler olabileceğini gösterdi. pBL plazmalarındayüksek ifadeli dolaşımsal serbest ve ekzozomal miRNA’ların tekveya entegre ROC analizleri, özellikle dolaşımsal ekzozomal miRNA’larınbazı üçlü kombinasyonlarının non-invazif tanı geliştirme potansiyeline(AUC>0.9) sahip olduğunu gösterdi (Şekil 1). Seçili miRNA’lar kullanılarakgerçekleştirilen qRT-PCR deneyleriyle miRNA-seq analiz sonuçlarının hemtümör doku miRNA hem de plazma miRNA seviyeleri için büyük ölçüdeörtüştüğü gözlemlendi.Tartışma: Sonuç olarak, bu projede pediatrik Burkitt lenfomanın invazifolmayan tanısını geliştirme potansiyeli bulunan ve plazma örneklerindeifadeleri artan miRNA’lar miRNA-seq aracılığıyla belirlenmiştir. NTEGRATIVE ANALYSIS OF FEN1 EXPRESSION AND PROGNOSTIC IMPACT IN NEUROBLASTOMA

Background and AimsRelapsed or refractory (R/R) high-risk (HR) neuroblastoma (NB) has a dismalprognosis. Anti-GD2-mediated chemo-immunotherapy has a considerable anti-tumoractivity in patients with R/R HR-NB. In the present study, we purposed toinvestigate the impacts and adverse effects of the combination of immunotherapywith dinutuximab beta (DB) and chemotherapy in patients with R/R HR-NB.MethodsPatients of over 12 months with documentation of a HR-NB diagnosis wereeligible at relapse or designation of refractory disease status. Inclusioncriteria were as follows: relapsed or refractory, measurable bycontrast-enhanced magnetic resonance imaging (MRI) and/or computed tomography(CT) or metaiodobenzylguanidine (MIBG)/ fluorodeoxyglucose (FDG) positron emissiontomography (PET)/CT evaluable disease and/or demonstrated by bone marrowaspiration and biopsy. Chemotherapy scheme was irinotecan (IV, 50 mg/m² perdose, on Days 0-4) and temozolomide (PO, 100 mg/m² per dose, on days 0-4).Dinutuximab beta was administered intravenously for 10 days through continuousinfusion with 10 mg/m2 per day (on Days 1-10). The patients received 2 to 12successive cycles with duration of 28 days each. Disease assessment wasperformed after cycles 2, 4, and 6 and every 2 to 3 cycles thereafter.ResultsBetween January 2020 and January 2022, nineteen (n=19) patients received atotal of 116 cycles of DB+CT. Objective (complete or partial) responses wereachieved in 12/19 (63%) patients, including complete remission (CR) in 6/19 andpartial response (PR) in 6/19. Stable disease was observed in two patients. Theremaining five patients developed bone/bone marrow and soft tissue progressionafter 2-4 cycles of treatment. The most common side effect was fever, which wasmore common in the first cycles of treatment. Grade ≥3 toxicities wereleukopenia (62%), thrombocytopenia (27%), hypertransaminasemia (25%), fever(14%), and rash/itching (11%), respectively.ConclusionsDB-based chemo-immunotherapy is suitable leading to an encouraging responserate in patients with R/R HR-NB

Objectives: Thymoquinone a bioactive component extracted from the seeds of Nigella sativa, has been proved to have antiinflammatory, antioxidantproperties anti antitumor efficiency against breast, prostate, bladder, and renal cell cancer. We aimed to investigate the synergistic cytotoxic and apoptoticeffects of thymoquinone in combination with nivolumab, an immuno-control inhibitor, and its mechanism in renal cell cancer models.Materials and Methods: In vitro studies: Caki-1 and Renca renal cancer cell lines with mononuclear cells were cocultured and thymoquinone andnivolumab effect were analyzed by MTT and Annexin V + PI. In vivo studies: Renal cell cancer model was provided by subcutaneous injection of RENCAcells to C57BL/6, male, 8 weeks old mice in four groups. Control group, thymoquinone (1,4,7,14days,20mg/kgIP), nivolumab administered (1,6,13days10mg/kgIP), thymoquinone and nivolumab. Seven animals was randomized in each experimental group. Apoptosis, necrosis, CD4,CD8,C20,CD34,CD31,VEGF-A byimmunohistochemistry, FGFR-1,PDGFRbeta,VEGFR-2,c-kit,RET,c-Met,AXL,Flt-3,TIE2, ICAM-1,VCAM-1 and E-selectin mRNA levels by quantitative real-time-PCR were evaluated on tumor tissue. Liver, kidney, brain, lung and heart tissues was evaluated for side effects. Plasma biochemistry tests were performed.Statistical analysis was be performed with Mann-Whitney U test with p Results: Thymoquinone showed antitumor effect in vitro and in vivo. It also showed increase in immune cell death of nivolumab. Tumor tissue showedprominent necrosis or complete response to therapy in combination group.Conclusions: We conclude that thymoquinone has antitumor effect on renal cell carcinoma and increases the effect of nivolumab immune, cell deatheffects.
