Terpinols are monoterpene compounds, and biological effects like anticancer, anticonvulsant and antiulcer have been demonstrated in many studies due to their antioxidant properties. The aim of the research was to investigate the anti-inflammatory and antiapoptotic biofunctions of α-terpineol in LPS induced inflammation model of liver cells. HepG2 (ATCC® HB-8065) cells of human origin were selected as material. LPS and α-terpineol were applied to cells at different concentrations for 24 hours, and their effective concentrations were determined by cell viability tests (MTT). Afterwards, gene expression levels of TNF-α, IL-1β, IL-10, Caspase 3, Bax and Bcl-2 were investigated by qRT-PCR method. LPS at 50 ng/ml, caused 11.5% cell loss and was chosen for modeling. However, α-terpineol at 10 μM concentration prevented 2.12% of cell loss. In the study, LPS increased TNF-α and IL-1β gene expressions, and α-terpineol application reversed this situation. Again, while IL-10 was suppressed by high LPS concentration, it was significantly stimulated by α-terpineol. Besides, LPS increased caspase 3 and Bax gene expressions, but α-terpineol suppressed this stimulation. Bcl-2 gene expressions were suppressed by LPS and stimulated by α-terpineol. In summary, short-term and low proliferative concentration of α-terpineol can be an alternative treatment agent, especially in pathogen-induced liver disorders., Terpinoller monoterpen bileşikler olup, birçok çalışmada antioksidan özelliklerinden dolayı antikanser, antikonvülsant ve antiülser gibi biyolojik etkileri ortaya konulmuştur. Araştırmanın amacını, LPS’in karaciğer hücrelerine uygulanarak oluşturulan yangı modelinde, α-terpineol’ün antiinflamatuar ve antiapoptotik biyofonksiyonlarının araştırılması oluşturmuştur. Denemede insan orijinli HepG2 (ATCC® HB-8065) hücreleri materyal olarak seçilmiştir. LPS ve α-terpineol, hücrelere farklı konsantrasyonlarda 24 saat uygulanmıştır ve etkin konsantrasyonları, hücre canlılık testleriyle (MTT) gerçekleştirilmiştir. Sonrasında, TNF-α, IL-1β, IL-10, Kaspaz 3, Bax ve Bcl-2 gen ekspresyon düzeyleri tam zamanlı kantatif polimeraz zincir reaksiyonuyla (qRT-PCR) araştırılmıştır. LPS’in 50 ng/ml konsantrasyonu %11,5 oranında hücre kayıplarına neden olduğu belirlenmiş ve modelleme için bu konsantrasyon seçilmiştir. Ancak, 10 μM konsantrasyonda α-terpineolün hücre kayıplarını % 2.12 oranında önlediği bulunmuştur. Çalışmada, LPS’in TNF-α ve IL-1β gen ekspresyonlarını arttırdığı, α-terpineol uygulamasının ise bu durumu tersine çevirdiği tespit edilmiştir. Yine, IL-10 gen ekspresyonu yüksek LPS konsantrasyonu ile baskılanırken, α-terpineol tarafından anlamlı düzeyde uyarıldığı da ortaya konulmuştur. Bununla birlikte, LPS’in kaspaz 3 ve Bax gen ekspresyonlarını arttırdığı, ancak α-terpineol’ün bu stimülasyonu baskıladığı tespit edilmiştir. Bcl-2 gen ekspresyonları ise LPS tarafından baskılanırken, α-terpineol tarafından uyarıldığı bulunmuştur. Özetle, α-terpineol’ün kısa süreli ve düşük olan proliferatif konsantrasyonunun özellikle patojen kaynaklı karaciğer rahatsızlıklarında alternatif bir tedavi ajanı olabileceği görülmüştür.

This study, it was aimed to determine the effects of murt volatile oil added to the diet on Lipopolysaccharide (LPS) induced inflammation on body weight, feed consumption, feed conversion ratio, slaughter characteristics, some blood and organ parameters in quails. The study groups were formed as follows: control (commercial chick starter feed), M group (commercial chick starter + 200 mg/kg Murt volatile oil), MLPS group (commercial chick starter + 1 mg/kg-17th day and 7.5 mg/kg- 24th day intrabdominal LPS administered group) and LPS (commercial chick starter feed + 1 mg/kg 17th day and 7.5 mg/kg - 24th day intraabdominal LPS administered group). The study, 4 experimental groups consisting of 40 (4x10) quail chicks were formed and the fattening period lasted for 36 days. The study, blood and liver OSI (Oxidative stress index) values were found to be lower in the M and MLPS groups with murt supplementation than in the control groups without murt addition. During the study, the body weight value was determined lower in the groups with Murt added than in the groups without Murt addition. In addition, the feed conversion ratio was significantly better in the M group with murt supplementation between the 5th and 33rd days than in the other groups. Significant differences were determined between the groups in terms of gizzard ratio and chest+back+neck+wing ratio values. In the study, macroscopic and histopathological hyperemia was not observed in the small intestines, there was an increase in cell infiltration in other groups with added murt compared to the control group, and degeneration and necrosis of the epithelium were detected more prominently. In the study, it was determined that the addition of murt to quail compound feed had a positive effect on growth performance, feed consumption and feed efficiency.

Çalışma, subklinik mastitisli ineklerde süt ve süt hücrelerinde Vitamin C düzeylerinin belirlenmesi amacıyla yapılmıştır. Süt örneklerinde somatik hücre sayımı yapıldı ve örnekler kontrol (1-87 x 1000 hücre), mastitli 1. grup (154-380 x 1000 hücre), 2. grup (418-812 x 1000 hücre), 3. grup (914-1928 x 1000 hücre) ve 4. grup (2614-8050 x 1000 hücre) olacak şekilde gruplandırıldı (n=12). Süt hücrelerinde ve süt serumunda (yağı ve hücreleri alınmış) Vitamin C düzeyleri belirlendi ve Vitamin C ve süt somatik hücre sayısı arasındaki korelasyonlar hesaplandı. Ayrıca alınan süt numunelerinden mikrobiyolojik ekim yapılarak, etken izolasyonu ve identifikasyonu yapıldı. Oksidatif stresin çeşitli nörodejeneratif hastalıkların patogenezinde önemli bir rol oynadığı bilinmektedir. Diyet polifenolleri ve diğer doğal antioksidanlar nörodejenerasyonun giderilmesi amacıyla yapılan klinik testlerde en popüler bileşiklerdir. Yapılan güncel çalışmalarda, bir çeşit yemiş olan Zizyphus jujuba'nın birçok önemli biyolojik aktivitelere sahip olduğu rapor edilmiştir. Bu çalışma, insan glioblastoma hücreleri üzerinde Zizyphus jujuba'nın antioksidan etkinliğinin araştırılması amaçlamaktadır. Hücre canlılığı, doza bağlı kolorimetrik viyabilite testleri ile belirlenmiştir. Hücreler, 1 saat 100 µM Ziziphus jujuba'nın uçucu yağları ile ön-muamele edilip, daha sonra 100 µM H2O2 12 saat boyunca hücrelere ilave edildi. Uygulamalar sonunda hücre homojenatları uzaklaştırılarak taze besiyeri ile değiştirildi. Total oksidan kapasite (TOS) ve total antioksidan kapasite (TAS) düzeyleri spesifik kolorimetrik yöntemler kullanılarak tayin edilmiştir. Oksidatif stress indeksi (OSİ) TOS ve TAS değerlerinin oranlamasıyla elde edilmiştir. It has been demonstrated in recent studies that fruit of Zizyphus jujuba possesses several vital biological activities. This study intends to evaluate antioxidant activity of Zizyphus jujuba on human glioblastoma cells. Cell survival was quantified by colorimetric viability assay with dose response. Cells were pretreated with 100 µM Ziziphus jujuba essential oil for 1h, then 100 µM HO was added to the cells for 12 hours. After that, the cell medium were taken after treatment period and replaced with fresh medium. Total oxidant capacity (TOS) and total antioxidant capacity (TAS) levels were estimated using specific colorimetric methods. Oxidative stress index (OSI) was calculated from the ratio of TOS and TAS values. Many researches have been reported that the essential oil from seeds helps to prevent the oxidative stress induced neuronal diseases. The antioxidant potential of Ziziphus jujuba may be attributed to the presence of flavonoids and the other constituents present therein. Bu çalışma iskemi-reperfüzyon (I/R) hasarlı rat beyinlerinde fosfodiesteraz 4 (PDE4) mRNA izoenzimleri, oksidant ve antioksidant savunma sistemi üzerine kafeik asit fenetil ester (KAFE)’in nöroprotektif etkilerini araştırmak amacıyla yapıldı. Yirmi bir adet rat rastgele üç eşit gruba ayrıldı. Sham-kontrol, iskemi/reperfüzyon (I/R) ve I/R+KAFE. Sham-kontrol grubundaki ratlara bilateral common carotid arter oklüzyonu yapılmaksızın sadece cerrahi müdahalede bulunuldu. İskemi/reperfüzyon (I/R) bilateral common carotid arterlerin atravmatik klempler ile 30 dakika oklüzyonu ve takiben arter klempleri açılarak reperfüzyonu ile sağlandı. I/R+KAFE grubu I/R grubu ile aynı cerrahi usüle tabi tutuldu fakat oklüzyondan 1 saat önce ve reperfüzyondan 12 saat sonra iki defa 15 µmol kg -1 dozunda intraperitoneal KAFE verildi. Ratlar iskemi/ reperfüzyondan 24 saat sonra sakrifiye edildi. Beyin korteksindeki cAMP düzeyi ELISA ile, PDE4 mRNA izoenzim transkripsiyonları ise qRT-PCR ile değerlendirildi. KAFE iskemi ile uyarılan beyin korteksindeki NO üretimini önemli oranda azalttı. I/R grubu ile karşılaştırıldığında SOD, CAT ve XO aktivitelerini KAFE anlamlı düzeyde değiştirmezken, GSH-Px aktivitesini önemli oranda arttırdı. KAFE cAMP düzeyini değiştirmeksizin PDE4A ve PDE4B düzeyini önemli oranda azalttı. İskemi ile uyarılan nörolojik hasar skorları KAFE tarafından azaltıldı. Bu sonuçlar KAFE’nin global beyin iskemi/reperfüzyon hasarı sırasında rat beyinlerinde antioksidant savunma sistemini ve NO salınımını hafifce dengelediğini önerir. Ayrıca KAFE bazı PDE4 izoenzim düzeylerini azaltarak nöroprotektif etki sağlar., The aim of this study was to investigate the neuroprotective efects of cafeic acid phenethyl ester (CAPE) on phosphodiesterase 4 (PDE4) mRNA isoenzymes, oxidant and antioxidant defence in ischemia/reperfusion (I/R) injured rat brains. Twenty-one rats were randomly divided into three equal groups: sham-control, ischemia/reperfusion (I/R) and I/R+CAPE. Rats in sham-control group underwent only surgical intervention without bilateral common carotid artery occlusion. Ischemia/reperfusion was induced by bilateral common carotid artery occlusion with atraumatic clips for 30 min, followed by artery reopening. The I/R+CAPE group was subjected to the same surgical procedure as I/R group, but CAPE was administered intraperitoneally at the dose of 15 µmol kg -1 twice, 1 h before occlusion and at 12 th h of reperfusion. The rats were sacrificed 24 h after I/R. The cAMP concentration was analyzed by ELISA and PDE4 isozyme mRNA transcriptions were evaluated by qRT-PCR methodology in the brain cortex. Ischemia-induced NO production was significantly attenuated by CAPE in the cerebral cortex. CAPE significantly enhanced GSH-Px activity, while SOD, CAT and XO activities non-significantly changed, as compared to the I/R group. CAPE significantly decreased PDE4A and PDE4B transcripts, without changing cAMP levels compared to I/R group. Ischemia-induced neurologic deficit scores were reduced by CAPE. These results suggest that CAPE slightly modulates the antioxidant defense system and NO release in rat brain during global cerebral ischemia/reperfusion injury. In addition, CAPE treatments produce the neuroprotective efect by reducing the levels of some PDE4 transcriptions.

Bu araştırma, oksijen ile muamele edilerek biyoaktif özellik kazandırılmış doğal kireç taşı içeren bir yem katkı maddesinin etlik piliçlerde performans üzerine etkisinin belirlenmesi amacıyla yürütüldü. Ayrıca bu yem katkı maddesinin protein, lipit ve glikoz metabolizması üzerine etkileri, bazı serum biyokimyasal parametreler analiz edilerek belirlendi. Araştırmada toplam 225 adet günlük yaşta etlik civciv her biri 3’er alt gruptan oluşan 3 gruba tesadüfî olarak ayrıldı. Kontrol grubu yemine ilave yapılmadı. Deneme grupları yemlerine 200 (Bio200) ve 400 (Bio400) mg/kg oranında oksijen ile işlem görmüş kireç taşı içeren katkı maddesi ilave edildi. Hayvanlara 5 haftalık deneme süresince yem ve su ad libitum verildi. Araştırma sonunda grupların ortalama canlı ağırlık, canlı ağırlık artışları, yemden yararlanma oranları Biyoaktif katkı maddesinden etkilenmezken (P>0.05), yem tüketiminde düşüş kaydedildi (P?0.05). Gruplar arasında serum total protein, glikoz, trigliserit, LDL, HDL ve total kolesterol ile serum kalsiyum ve fosfor düzeyi bakımından istatistiksel bir farklılık bulunmadı (P>0.05). Biyoaktif katkı maddesinin belirlenen biyokimyasal parametreler üzerine olumsuz bir etkisinin bulunmaması ve canlı ağırlık artışını düşürmeksizin yem tüketimini azaltması, yemden yararlanma etkinliğinin iyileştirilmesinde potansiyel bir değer taşıdığını gösterdi. Sonuç olarak, verim artırmaya yönelik uygulamalarda kullanım olanağı bulabilecek bu yeni ürün ile ilgili özellikle etki mekanizmasına ilişkin ileri araştırmaların yapılıp, kullanımının yaygınlaştırılmasının etlik piliç yetiştiriciliğinde ve organik hayvancılıkta ekonomik üretim olanakları sağlayabileceği kanaatine varıldı., The current study was conducted to evaluate the effects of natural feed additives containing oxygen-activated chalk on performance of broilers. The certain biochemical parameters were also analyzed to assess the influence of the feed additive on protein, lipid and glucose metabolisms of the broilers. A total of 225 unsexed, one-day old broiler chicks were randomly divided into 3 groups comprising 3 subgroups with 75 chicks of each. The chicks in control group were fed with un- supplemented basal diet. Bioactive feed additive containing oxygen-activated chalk was added to the diet of those groups Bio200 and Bio400 at the levels of 200 and 400 mg/kg, respectively. Feed and water were given to the animals ad libitum for 5 weeks. Feed consumptions and body weights of the birds were recorded and body weight gain and feed efficiency were calculated weekly intervals to assess the growth performance of groups. At the end of the trial, while body weight, body weight gain and feed efficiency were not affected (P>0.05), feed consumption significantly decreased by the addition of the bioactive supplement to the diet (P≤0.05). The serum total protein, glucose, triglyceride, LDL and HDL and total cholesterol levels of the groups which may reflect the status of the protein, lipid and glucose metabolisms were not affected by supplementation diet with the feed additive (P>0.05). Serum calcium and phosphor levels also were not different between the groups (P>0.05). The results of present study indicates that the bioactivated chalk can be considered as a potential feed additive to promote the growth in broilers due to positive effects on feed consumption without compromising the reduction in live weight gain. Consequently, it has been needed to carry out further research on the product as an alternative feed additive could provide more economic production possibility in broilers and organic animal production.
