Abstract

Gamma-ray spectroscopy and dosimetry are complementary techniques used to locate and identify radioactive sources containing gamma-ray-emitting radioisotopes. Gamma-ray spectroscopy is extensively studied for various applications across multiple fields, including homeland security, environmental radioactivity monitoring, tackling illegal trade of radioiso-topes, and medical sciences. Introducing our newly established startup, Flying DEMon s.r.l., comprised of young researchers, academic professors, and backed by university support. Our venture aims to advance project development, leveraging the grant awarded through the E-TEC2 contest initiated by ENAC. The team will showcase their comprehensive work plan, highlighting the project’s competitiveness and self-sustaining potential. The objective of our startup is to harness cutting-edge technologies in the field of gamma spectroscopy and dosimetry, adaptable for deployment via Unmanned Aircraft Systems (UAS). This innovation holds significant promise for environmental monitoring, facilitating tasks such as pinpointing widespread radioactive sources or identifying concealed and hard-to-reach nuclear waste. Additionally, this advancement holds potential for applications in military, security, and industrial oversight. Our research focus primarily revolves around real-time and rapid gamma-ray analysis in open-field environments. Our group not only supports the core project objectives but also enables its applicability in diverse and non-traditional sectors, such as Agritech.

Abstract

Gamma-ray spectroscopy and gamma-ray imaging are two complementary techniques used for the localization and the identification of radioactive sources containing gamma-ray emitting radioisotopes. The radioactivity monitoring is focused on the detection of both artificial and environmental radioactive sources like Naturally Occurring Radioactive Materials (NORM). This kind of contamination becomes dangerous when the detection of the unwanted substances exhibits a concentration significantly above the environmental radioactive background radiation levels. For this purpose, we have developed, tested and shown a High Efficiency fast-Response GAmma (HERGA) detector useful for the identification of radionuclides and for gamma-ray imaging. A first version of the gamma detector prototype was composed of 16 CsI(Tl) scintillating crystals of 3x3x10 cm3 size, arranged in 4x4 matrix coupled with standard Photomultiplier tubes (PMTs). An image reconstruction of a radioactive gamma emitter source is possible using the coded mask technique, in which a 7x7 mask, made of Plastic and Tungsten tiles, is placed in front of the detector and a pattern recognition algorithm based on classical statistical methods (Kolmogorov Smirnov) is used to reconstruct the source position. The measurements carried out showed a point spread function (PSF) of a few mrad for pointlike sources. The Minimum Detectable Activity (MDA) was also determined in the case of pointlike radioactive sources. In this contribution we will present an update of the HERGA detector prototype in which Silicon Photomultipliers (SiPMs) are used in place of the PMTs. SiPMs provide similar or even better performance compared to the standard PMT sand provide benefits in terms of lower power consumption and reduced cost and compactness. The advantages of the SiPM technology are also characterized by the robustness of the photosensor that makes the new prototype compact, portable, ideal for in-situ and real-time. We will show a comparison between the results obtained with the newest SiPM read-out technology with respect to those obtained with the PMT one, in terms of energy and spatial resolution. The imaging performance is also in phase of testing in order to localize extended radioactive sources such as for example NORM samples or to detect inaccessible or hidden nuclear waste.

Abstract

Recent observations of the light component of the cosmic-ray spectrum have revealed unexpected features that motivate further and more precise measurements up to the highest energies. The Dark Matter Particle Explorer is a satellite-based cosmic-ray experiment that has been operational since December 2015, continuously collecting data on high-energy cosmic particles with very good statistics, energy resolution, and particle identification capabilities. In this work, the latest measurements of the energy spectrum of proton+helium in the energy range from 46 GeV to 464 TeV are presented. Among the most distinctive features of the spectrum, a spectral hardening at 600 GeV has been observed, along with a softening at 29 TeV measured with a 6.6{\sigma} significance. Moreover, the detector features and the analysis approach allowed for the extension of the spectral measurement up to the sub-PeV region. Abstract

More than a century after the performance of the oil drop experiment, the possible existence of fractionally charged particles FCP still remains unsettled. The search for FCPs is crucial for some extensions of the Standard Model in particle physics. Most of the previously conducted searches for FCPs in cosmic rays were based on experiments underground or at high altitudes. However, there have been few searches for FCPs in cosmic rays carried out in orbit other than AMS-01 flown by a space shuttle and BESS by a balloon at the top of the atmosphere. In this study, we conduct an FCP search in space based on on-orbit data obtained using the DArk Matter Particle Explorer (DAMPE) satellite over a period of five years. Unlike underground experiments, which require an FCP energy of the order of hundreds of GeV, our FCP search starts at only a few GeV. An upper limit of $6.2\times 10^{-10}~~\mathrm{cm^{-2}sr^{-1} s^{-1}}$ is obtained for the flux. Abstract

Rationale: Aging in the heart is a gradual process, involving continuous changes in cardiovascular cells, including cardiomyocytes (CMs), namely cellular senescence. These changes finally lead to adverse organ remodeling and resulting in heart failure. This study exploits CMs from human induced pluripotent stem cells (iCMs) as a tool to model and characterize mechanisms involved in aging. Methods and Results: Human somatic cells were reprogrammed into human induced pluripotent stem cells and subsequently differentiated in iCMs. A senescent-like phenotype (SenCMs) was induced by short exposure (3 hours) to doxorubicin (Dox) at the sub-lethal concentration of 0.2 µM. Dox treatment induced expression of cyclin-dependent kinase inhibitors p21 and p16, and increased positivity to senescence-associated beta-galactosidase when compared to untreated iCMs. SenCMs showed increased oxidative stress, alteration in mitochondrial morphology and depolarized mitochondrial membrane potential, which resulted in decreased ATP production. Functionally, when compared to iCMs, SenCMs showed, prolonged multicellular QTc and single cell APD, with increased APD variability and delayed afterdepolarizations (DADs) incidence, two well-known arrhythmogenic indexes. These effects were largely ascribable to augmented late sodium current (INaL) and reduced delayed rectifier potassium current (Ikr). Moreover sarcoplasmic reticulum (SR) Ca2+ content was reduced because of downregulated SERCA2 and increased RyR2-mediated Ca2+ leak. Electrical and intracellular Ca2+ alterations were mostly justified by increased CaMKII activity in SenCMs. Finally, SenCMs phenotype was furtherly confirmed by analyzing physiological aging in CMs isolated from old mice in comparison to young ones. Conclusions: Overall, we showed that SenCMs recapitulate the phenotype of aged primary CMs in terms of senescence markers, electrical and Ca2+ handling properties and metabolic features. Abstract

In June 2019 from the region of Calabria (Italy) our group launched the Stratospheric Balloon MoCRiS (Measurement of Cosmic Ray in Stratosphere) that reached up to 35111 meters. MoCRiS is a didactical project that uses a Light Balloon with a payload of only 2.1 Kg payload box included. The capsule was Lithium-ion batteries operated and fully instrumented with two Hi-Res Camera, one for picture and one for video, GPS Tracker, two Particle detectors, and sensors for temperature and pressure. Two ArduSiPM [6] scintillation detectors, made by the INFN group in Rome, were used as cosmic ray flux meters. The technology used for these types of devices makes them very economical, light (<150 g), and with low power consumption (<1 Watt). These characteristics are optimal for use with the balloon and in space (e.g. in CubeSat), and very attractive for educational use, even on the ground. During the school year, the students of the scientific high school "Stefano Patrizi" of Cariati (CS) Italy, participated in the definition of the project following theoretical and practical lessons on aerospace techniques and particle physics with regard to cosmic ray showers in the atmosphere. The mission was completely successful by experimentally verifying the pressure-to-height relation, the height of the thermal inversion in the atmosphere, reaching and exceeding the maximum Regener-Pfotzer level of the shower development. The instrumental and didactic platform created is easily replicable even with further upgrades. We are expanding the collaboration with other schools for further launches.

Abstract

OCRA - Outreach Cosmic Ray Activities was born in 2018 as a national outreach project of INFN with the aim of collecting, within a national framework, the numerous public engagement activities in the field of cosmic ray physics already present at a local level in the divisions and laboratories. Since spring 2020 OCRA offers also a series of online laboratories on its website https://web.infn.it/OCRA/, designed not only to be used by students individually but also to be offered in the classroom by teachers. The cosmic ray path, present on the website, will be presented together with the online laboratories on the measurement of muons, from the one related to the dependence on the zenith angle made during the International Cosmic Day up to measurements of the flux dependency on the altitude in the atmosphere and in the water. Also, a laboratory allowing to analyze public data of the Pierre Auger Observatory will be presented. In addition, some teaching methods included in the "Teachers' area" of the OCRA website will be described. The developed cosmic ray path was also used to organize an online course for teachers of Italian high schools with the purpose of accompanying teachers when approaching the subject for the first time. Abstract

The Outreach Cosmic Ray Activities (OCRA) project offers a platform for all outreach activities focusing on cosmic rays within the Italian Istituto Nazionale di Fisica Nucleare (INFN). Created in 2018, OCRA now counts 21 of the institute's divisions all over Italy as members. The project's core activity is the participation of all its members in the International Cosmic Day, organized by DESY, and inviting high school students to carry out hands-on measurements of the cosmic ray flux and learn about the related physics background. Two students from each division are then selected to participate in the annual OCRA science camp, a three-day full immersion into the life of a physicist. The national activities are complemented by local initiatives of the OCRA member groups: workshops and tutorship programs, science competitions and the development of new detectors for outreach activities offer a multitude of possibilities for students to engage with our researchers and to explore the world of cosmic rays. Since spring of 2020 OCRA also offers a series of online laboratories on its website https://web.infn.it/OCRA/, designed not only to be used by students individually but also to be used in the classroom by teachers. Here we will give an overview on all activities offered by OCRA with a particular focus on the 2020 online event organized in occasion of the International Cosmic Day, that saw the participation of more than 3000 students.

Abstract

The outreach program “A scuola di Astroparticelle” was proposed in 2016 by the National Institute for Nuclear Physics (INFN - Napoli Division) in collaboration with the Physics Department “Ettore Pancini” of the Federico II University in Napoli, CNR-SPIN and CNR-ISASI Institutes. Its main goal is to engage teachers and students of High Schools in astroparticle physics projects. For the third edition (2018/19), the activities, which are also part of the Italian Educational Program PCTO - “Percorsi per le Competenze Trasversali e per l'Orientamento”, involved 18 schools for a total of 21 projects on several topics. Some projects were strictly related to astroparticles as cosmic rays, while others were more technical, as the development of particle detectors, or cross-disciplinary projects. Students worked for the entire school year and prepared materials for the final event. More than 600 students attended the event and presented their work to a jury with a poster and an oral presentation in plenary sessions. Since 2018, the program is part of OCRA - Outreach Cosmic Ray Activities - a national outreach project of INFN with the aim of collecting, within a common framework, the numerous outreach activities in cosmic-ray field carried out at the local level. The fourth edition (2019-20), in spite of the difficult situation due to the COVID-19 pandemic, has also seen the participation of 22 schools that carried out part of the activities in an online format. The project realized using the open data of the Pierre Auger Observatory will be presented in detail.

Abstract

Human induced pluripotent stem cell (hiPSC)-derived cardiomyocytes (CM) constitute a mixed population of ventricular-, atrial-, nodal-like cells, limiting the reliability for studying chamber-specific disease mechanisms. Previous studies characterised CM phenotype based on action potential (AP) morphology, but the classification criteria were still undefined. Our aim was to use in silico models to develop an automated approach for discriminating the electrophysiological differences between hiPSC-CM. We propose the dynamic clamp (DC) technique with the injection of a specific IK1 current as a tool for deriving nine electrical biomarkers and blindly classifying differentiated CM. An unsupervised learning algorithm was applied to discriminate CM phenotypes and principal component analysis was used to visualise cell clustering. Pharmacological validation was performed by specific ion channel blocker and receptor agonist. The proposed approach improves the translational relevance of the hiPSC-CM model for studying mechanisms underlying inherited or acquired atrial arrhythmias in human CM, and for screening anti-arrhythmic agents.

Abstract

The Outreach Cosmic Ray Activities (OCRA) project offers a platform for all outreach activities focusing on cosmic rays within the Italian Istituto Nazionale di Fisica Nucleare (INFN). Created in 2018, OCRA now counts 21 of the institute's divisions all over Italy as members. The project's core activity is the participation of all its members in the International Cosmic Day, organized by DESY, and inviting high school students to carry out hands-on measurements of the cosmic ray flux and learn about the related physics background. Two students from each division are then selected to participate in the annual OCRA science camp, a three-day full immersion into the life of a physicist. The national activities are complemented by local initiatives of the OCRA member groups: workshops and tutorship programs, science competitions and the development of new detectors for outreach activities offer a multitude of possibilities for students to engage with our researchers and to explore the world of cosmic rays. Since spring of 2020 OCRA also offers a series of online laboratories on its website https://web.infn.it/OCRA/, designed not only to be used by students individually but also to be used in the classroom by teachers. Here we will give an overview on all activities offered by OCRA with a particular focus on the 2020 online event organized in occasion of the International Cosmic Day, that saw the participation of more than 3000 students.

Abstract

Background: Combined treatment with anthracyclines (e.g., doxorubicin; Dox) and trastuzumab (Trz), a humanized anti-human epidermal growth factor receptor 2 (HER2; ErbB2) antibody, in patients with HER2-positive cancer is limited by cardiotoxicity, as manifested by contractile dysfunction and arrhythmia. The respective roles of the two agents in the cardiotoxicity of the combined therapy are incompletely understood. Objective: To assess cardiac performance, T-tubule organization, electrophysiological changes and intracellular Ca2+ handling in cardiac myocytes (CMs) using an in vivo rat model of Dox/Trz-related cardiotoxicity. Methods and Results: Adult rats received 6 doses of either Dox or Trz, or the two agents sequentially. Dox-mediated left ventricular (LV) dysfunction was aggravated by Trz administration. Dox treatment, but not Trz, induced T-tubule disarray. Moreover, Dox, but not Trz monotherapy, induced prolonged action potential duration (APD), increased incidence of delayed afterdepolarizations (DADs) and beat-to-beat variability of repolarization (BVR), and slower Ca2+ transient decay. Although APD, DADs, BVR and Ca2+ transient decay recovered over time after the cessation of Dox treatment, subsequent Trz administration exacerbated these abnormalities. Trz, but not Dox, reduced Ca2+ transient amplitude and SR Ca2+ content, although only Dox treatment was associated with SERCA downregulation. Finally, Dox treatment increased Ca2+ spark frequency, resting Ca2+ waves, sarcoplasmic reticulum (SR) Ca2+ leak, and long-lasting Ca2+ release events (so-called Ca2+ “embers”), partially reproduced by Trz treatment. Conclusion: These results suggest that in vivo Dox but not Trz administration causes T-tubule disarray and pronounced changes in electrical activity of CMs. While adaptive changes may account for normal AP shape and reduced DADs late after Dox administration, subsequent Trz administration interferes with such adaptive changes. Abstract

Reprogramming of adult somatic cells into induced pluripotent stem cells (iPSCs) has revolutionized the complex scientific field of disease modelling and personalized therapy. Cardiac differentiation of human iPSCs into cardiomyocytes (hiPSC-CMs) has been used in a wide range of healthy and disease models by deriving CMs from different somatic cells. Unfortunately, hiPSC-CMs have to be improved because existing protocols are not completely able to obtain mature CMs recapitulating physiological properties of human adult cardiac cells. Therefore, improvements and advances able to standardize differentiation conditions are needed. Lately, evidences of an epigenetic memory retained by the somatic cells used for deriving hiPSC-CMs has led to evaluation of different somatic sources in order to obtain more mature hiPSC-derived CMs.

Abstract

A new generation magnetic spectrometer in space will open the opportunity to inves-tigate the frontiers in direct high-energy cosmic ray measurements and to precisely measure the amount of the rare antimatter component in cosmic rays beyond the reach of current missions. We propose the concept for an Antimatter Large Acceptance Detector In Orbit (ALADInO), designed to take over the legacy of direct measurements of cosmic rays in space performed by PAMELA and AMS-02. ALADInO features technological solutions conceived to overcome the current limi-tations of magnetic spectrometers in space with a layout that provides an acceptance larger than 10 m2 sr. A superconducting magnet coupled to precision tracking and time-of-flight systems can provide the required matter–antimatter separation capabilities and rigidity measurement resolution with a Maximum Detectable Rigidity better than 20 TV. The inner 3D-imaging deep calorimeter, designed to maximize the isotropic acceptance of particles, allows for the measurement of cosmic rays up to PeV energies with accurate energy resolution to precisely measure features in the cosmic ray spectra. The operations of ALADInO in the Sun–Earth L2 Lagrangian point for at least 5 years would enable unique revolutionary observations with groundbreaking discovery poten-tials in the field of astroparticle physics by precision measurements of electrons, positrons, and antiprotons up to 10 TeV and of nuclear cosmic rays up to PeV energies, and by the possible unam-biguous detection and measurement of low-energy antideuteron and antihelium components in cosmic rays., QC 20230227

Abstract

As waves interact with the slopes of coral reefs and other steep bathymetry profiles, plunging breaking usually occurs where the free surface overturns and violent water motion is triggered. Resolving these surf zone processes pose significant challenges for conventional mesh-based hydrodynamic models, due to the rapidly-deforming nature of the free surface and associated flows. Yet the accurate prediction of these surf zone hydrodynamics is critical for predicting a wide range of nearshore processes driven by wave breaking (e.g., wave dissipation and energy transfers; mean water levels and currents; and wave runup). In this study we assess the ability of the mesh-free, Lagrangian particle-based numerical modelling approach Smoothed Particle Hydrodynamics (SPH) based on DualSPHysics, to simulate the fine-scale hydrodynamic processes driven by irregular wave transformation over a fringing reef profile, by comparing results against detailed experimental observations from a physical modelling study. To greatly improve the computational efficiency, the SPH model was coupled to the mesh-based multi-layer nonhydrostatic wave-flow model SWASH. With this coupled approach, SWASH was used to efficiently simulate the evolution of non-breaking waves from the wavemaker up to the fore reef slope, with the SPH model then used to simulate the detailed hydrodynamic processes over the reef from just offshore of the breakpoint to the shoreline. The SPH model was able to accurately reproduce the complex free surface deformations during plunging breaking, the spectral evolution of waves across the reef flat (including nonlinear wave shape), the mean water levels and currents, and wave runup at the shoreline. Abstract

This work presents the results of an investigation on how wave overtopping at a near-vertical seawall at the back of a sandy foreshore is influenced by sequences of erosive storms. The experiments were carried out in the Large Wave Flume (GWK) at Leibniz University, Hannover (Germany). The tested layout consisted of a near-vertical seawall and a sandy foreshore with an initial slope. Three sequences of idealised erosive storms were simulated. Within each storm both the incident wave conditions and still water level were varied in time to represent high and low tide conditions. Each sequence started from a configuration and the beach was not restored in between storms. The measurements included waves, beach profile, wave overtopping volumes. The profile of the beach was measured after each sea state tested. Wave overtopping at each stage of the tested storms was significantly influenced by bed changes. This was linked to the measured evolution of the beach. Measurements showed that a barred profile developed quickly at the start of each sequence, and scour developed at the toe of the structure during high water level conditions, while accretion or partial backfilling developed during low water level conditions. Due to these processes, the position of a sea state in the tested sequence is shown to be an important factor in determining the wave overtopping volume. Remarkably, when a weaker idealised storm followed a more energetic one, nearly the same level of overtopping was recorded. This is explained by the foreshore erosion, leading to increased water depths and wave heights at the toe of the structure. This finding allows to quantify and to explain the variability of wave overtopping in storms following one another at intervals shorter than the recovery time of the foreshore.

Mechanisms that can restore cardiac relaxation improving intracellular Ca2+ dynamics represent a promising therapeutic approach for cardiovascular diseases associated to DD. Istaroxime has the dual properties to accelerate Ca2+ uptake into sarcoplasmic reticulum (SR) through the SR Ca2+ pump (SERCA2a) stimulation and to inhibit Na+/K+ ATPase (NKA). This project aims to characterize istaroxime effects at a concentration (100 nmol/L) marginally affecting NKA, in order to highlight its effects dependent on the stimulation of SERCA2a in an animal model of mild diabetes. Methods and results: Streptozotocin (STZ) treated diabetic rats were studied at 9 weeks after STZ injection in comparison to controls (CTR). Istaroxime effects were evaluated in vivo and in left ventricular (LV) preparations. STZ animals showed 1) marked DD not associated to cardiac fibrosis, 2) LV mass reduction associated to reduced LV cell dimension and T-tubules loss, 3) reduced LV SERCA2 protein level and activity and 4) slower SR Ca2+ uptake rate, 5) LV action potential (AP) prolongation and increased short-term variability (STV) of AP duration, 6) increased diastolic Ca2+, and 7) unaltered SR Ca2+ content and stability in intact cells. Acute istaroxime infusion (0.11 mg/kg/min for 15 min) reduced DD in STZ rats. Accordingly, in STZ myocytes istaroxime (100 nmol/L) stimulated SERCA2a activity and blunted STZ-induced abnormalities in LV Ca2+ dynamics. In CTR myocytes, istaroxime increased diastolic Ca2+ level due to NKA blockade albeit minimal, while its effects on SERCA2a were almost absent. Conclusions: SERCA2a stimulation by istaroxime improved STZ-induced DD and intracellular Ca2+ handling anomalies.
