Although localized prostate cancer is relatively indolent, advanced prostate cancer manifests with aggressive and often lethal variants, including neuroendocrine prostate cancer (NEPC). To identify drivers of aggressive prostate cancer, we leveraged Sleeping Beauty (SB) transposon mutagenesis in a mouse model based on prostate-specific loss-of-function of Pten and Tp53 . Compared with control mice, SB mice developed more aggressive prostate tumors, with increased incidence of metastasis. Notably, a significant percentage of the SB prostate tumors display NEPC phenotypes, and the transcriptomic features of these SB mouse tumors recapitulated those of human NEPC. We identified common SB transposon insertion sites (CIS) and prioritized associated CIS-genes differentially expressed in NEPC versus non-NEPC SB tumors. Integrated analysis of CIS-genes encoding for proteins representing upstream, post-translational modulators of master regulators controlling the transcriptional state of SB -mouse and human NEPC tumors identified sirtuin 1 ( Sirt1 ) as a candidate mechanistic determinant of NEPC. Gain-of-function studies in human prostate cancer cell lines confirmed that SIRT1 promotes NEPC, while its loss-of-function or pharmacological inhibition abrogates NEPC. This integrative analysis is generalizable and can be used to identify novel cancer drivers for other malignancies., Summary: Using an unbiased forward mutagenesis screen in an autochthonous mouse model, we have investigated mechanistic determinants of aggressive prostate cancer. SIRT1 emerged as a key regulator of neuroendocrine prostate cancer differentiation and a potential target for therapeutic intervention.

Introduction: Diagnosing and treating lameness in horses is essential to improving their welfare. In equine orthopedic practice, infrared thermography (IRT) can indirectly detect soreness. Non-steroidal anti-inflammatory drugs can treat painful and inflammatory processes in horses. Using IRT, the efficacy of meloxicam (Maxicam Gel ® ) was evaluated in pre-treating transient synovitis in horses induced by a middle carpal joint injection of lipopolysaccharides (LPS) from E. coli 055:B5 at a dose of 10 endotoxin units., Methods: In a cross-over design, six healthy horses were randomly assigned to receive either 0.6 mg/kg of oral Maxicam Gel ® (MAXVO) or a mock administration (control group, C) following a two-week washout period. IRT of the middle carpal joint, visual lameness assessment and joint circumference were recorded over time. Clinical and hematological evaluations were performed. Synovial fluid aspirates were analyzed for total nucleated cell count, total protein, and prostaglandin E 2 . A mixed effects analysis of variance was performed for repeated measures over time, followed by Tukey's test. A multinomial logistic regression was conducted to determine whether there is a relationship between a thermography temperature change and the lameness score., Results: There were no changes in joint circumference. The MAXVO group showed a lower rectal temperature 4 h after synovitis induction. The C group presented an increase in neutrophils and a decrease in total hemoglobin and hematocrit 8 h after induction. No changes were observed in the synovial fluid between groups. The horses that received meloxicam did not show clinically significant lameness at any time, while the C group showed an increase in lameness 2, 4, and 8 h after synovitis induction., Discussion: IRT indicated that the skin surface temperature of the middle carpal joint was lower in horses who received meloxicam, suggesting a reduction in the inflammatory process induced by LPS. It was observed that the maximum temperature peaks in the dorsopalmar and lateropalmar positions can be utilized to predict the severity of lameness, particularly when the temperature rises above 34°C. In the complex tumor microenvironment (TME), mesenchymal cells are key players, yet their specific roles in prostate cancer (PCa) progression remain to be fully deciphered. This study employs single-cell RNA sequencing to delineate molecular changes in tumor stroma that influence PCa progression and metastasis. Analyzing mesenchymal cells from four genetically engineered mouse models (GEMMs) and correlating these findings with human tumors, we identify eight stromal cell populations with distinct transcriptional identities consistent across both species. Notably, stromal signatures in advanced mouse disease reflect those in human bone metastases, highlighting periostin's role in invasion and differentiation. From these insights, we derive a gene signature that predicts metastatic progression in localized disease beyond traditional Gleason scores. Background: this observational and cross-sectional study aimed to describe training, pharmacological ergogenic aids, dehydration, and nutrition strategies during a peak week in competitive bodybuilders and monitor changes that occurred across this period., Methods: Ten amateur bodybuilders were followed during a peak week phase and one day post-contest. Training, diet, dehydration protocol, anabolic steroid use, and nonsteroidal agents were recorded, prescribed, and supervised by the same coach in charge. Body composition, resting metabolic rate, and circumferences were assessed by the research team., Results: Overall, during the peak week phase, the use of anabolic steroids, diuretics, and other pharmacological aids was high among athletes, and the dose and frequency were similar between competitors. Despite the use of supraphysiological doses of drugs, bodybuilders displayed a statistical reduction of lean mass markers, resting metabolic rate, and fat mass markers, possibly influenced by the performance of high volumes of aerobic exercise combined with dietary energy restriction., Conclusions: The main findings of this study display that the coach in charge of Brazilian bodybuilders applied the same anabolic steroid, diuretic, and other pharmacological protocols for all competitors. Moreover, the protocols employed by bodybuilders did not fully attenuate the loss of lean mass during the peak week period.

Prioritizing treatments for individual patients with cancer remains challenging, and performing coclinical studies using patient-derived models in real time is often unfeasible. To circumvent these challenges, we introduce OncoLoop, a precision medicine framework that predicts drug sensitivity in human tumors and their preexisting high-fidelity (cognate) model(s) by leveraging drug perturbation profiles. As a proof of concept, we applied OncoLoop to prostate cancer using genetically engineered mouse models (GEMM) that recapitulate a broad spectrum of disease states, including castration-resistant, metastatic, and neuroendocrine prostate cancer. Interrogation of human prostate cancer cohorts by Master Regulator (MR) conservation analysis revealed that most patients with advanced prostate cancer were represented by at least one cognate GEMM-derived tumor (GEMM-DT). Drugs predicted to invert MR activity in patients and their cognate GEMM-DTs were successfully validated in allograft, syngeneic, and patient-derived xenograft (PDX) models of tumors and metastasis. Furthermore, OncoLoop-predicted drugs enhanced the efficacy of clinically relevant drugs, namely, the PD-1 inhibitor nivolumab and the AR inhibitor enzalutamide., Significance: OncoLoop is a transcriptomic-based experimental and computational framework that can support rapid-turnaround coclinical studies to identify and validate drugs for individual patients, which can then be readily adapted to clinical practice. This framework should be applicable in many cancer contexts for which appropriate models and drug perturbation data are available. Until mass vaccination befalls, control of the new betacoronavirus-associated severe acute respiratory syndrome pandemic (SARS-CoV-2) is based on decreasing virus circulation by social distancing and blocking transmission foci after diagnosis. Globally adopted SARS-CoV-2 diagnostic criteria embrace viral RNA detection by quantitative reverse-transcription polymerase chain reaction (qRT-PCR) on nasopharynx secretions, which requires healthcare facilities and specialized personnel for sample collection. To develop an alternative protocol, hydrophilic cotton as the material and saliva as the source for biological sample collection in qRT-PCR/RT-endpoint-PCR SARS-CoV-2 diagnostic methods prepared with local consumables were evaluated using 99 archived nasopharynx samples previously diagnosed as positive for SARS-CoV-2 and 111 prospective saliva samples pared with nasopharynx samples from patients attending the local reference ABC Medical School diagnostic laboratory. The kappa agreement coefficient between the SARS-CoV-2 qRT-PCR and RT-endpoint-PCR was k = 0.97 (95 % CI 0.92-1.00) and k = 0.90 (95 % CI 0.81-0.99), respectively, on SARS-CoV-2-positive archived samples, with the initial qRT-PCR C T under 25. The agreement coefficient of the SARS-CoV-2 alternative saliva diagnostic protocol, when used to test the paired nasopharynx samples, was k = 0.79 (95 % CI 0.56-1,00). These data support that the SARS-CoV-2 diagnostic assay based on self-collected saliva on cotton represents an alternative protocol for mass diagnosis and epidemiological studies in low-income regions., (Copyright © 2021 Elsevier B.V. Metabolic and molecular profiles were reported as different for bovine embryos with distinct kinetics during the first cleavages. In this study, we used this same developmental model (fast vs slow) to determine if the relationship between metabolism and developmental kinetics affects the levels of acetylation or tri-methylation at histone H3 lysine 9 (H3K9ac and H3K9me3, respectively). Fast and slow developing embryos presented different levels of H3K9ac and H3K9me3 from the earliest stages of development (40 and 96 hpi) and up to the blastocyst stage. For H3K9me3, both groups of embryos presented a wave of demethylation and de novo methylation, although it was more pronounced in fast than slow embryos, resulting in blastocysts with higher levels of this mark. The H3K9ac reprogramming profile was distinct between kinetics groups. While slow embryos presented a wave of deacetylation, followed by an increase in this mark at the blastocyst stage, fast embryos reduced this mark throughout all the developmental stages studied. H3K9me3 differences corresponded to writer and eraser transcript levels, while H3K9ac patterns were explained by metabolism-related gene expression. To verify if metabolic differences could alter levels of H3K9ac, embryos were cultured with sodium-iodoacetate (IA) or dichloroacetate (DCA) to disrupt the glycolytic pathway or increase acetyl-CoA production, respectively. IA reduced H3K9ac while DCA increased H3K9ac in blastocysts. Concluding, H3K9me3 and H3K9ac patterns differ between embryos with different kinetics, the second one explained by metabolic pathways involved in acetyl-CoA production. So far, this is the first study demonstrating a relationship between metabolic differences and histone post-translational modifications in bovine embryos.

Objective: To compare the use of intravenous and topical tranexamic acid (TXA) in unilateral primary total knee arthroplasty (TKA) in relation to blood loss and complications inherent to the medication., Method: Three groups with 14 patients each were constituted, and all of them were operated using the same surgical technique. In Group 1, usual measures for bleeding control were performed. Group 2 patients received TXA topically on the joint surface. In Group 3, intravenous TXA was used. Hemoglobin (HB), hematocrit (HTC), platelets (PLAT), prothrombin time, activated partial thromboplastin time and volume of blood drained observed 24 hours after arthroplasty were compared to the values of tests found before surgery., Results: There was a decrease in the concentration of HB, HTC and PLAT in all groups in relation to the preoperative, however without significant difference. Group 3 had a lower mean volume of drained blood than the other groups, with statistical significance. No adverse effects or thromboembolic events were observed in the groups that received TXA., Conclusion: This study showed superiority in the use of intravenous TXA in decreasing the volume of bleeding, without increasing the risk of thromboembolic events. Two studies were conducted to determine the effects of a novel Escherichia coli phytase expressed in Pseudomonas fluorescens on growth performance, bone mineralization, and nutrient digestibility in pigs fed corn-soybean meal diets. In experiment 1, 160 nursery pigs (9.79 ± 1.22 kg) were randomly allotted to one of four treatments with 10 pens per treatment and four pigs per pen. Phase I and phase II diets were provided from d 0 to d 14 and d 14 to d 28, respectively. Treatments included: positive control (PC) with all nutrients meeting requirements; negative control (NC) with standardized total tract digestible (STTD) P reduced by 0.15% and 0.14% compared with PC in phase I and phase II, respectively; and NC diets containing 250 or 500 units of phytase (FTU) per kilogram. Results demonstrated that pigs fed PC had greater ( P < 0.01) ADG and G:F for the overall experimental period, and greater ( P < 0.01) bone ash and P concentrations, compared with pigs fed NC or diets with phytase supplementation. Pigs fed diets containing phytase had greater ( P < 0.01) ADG and G:F for the overall experimental period compared with pigs fed the NC diet without phytase, and bone ash and P weights were increased ( P < 0.01) as well. In experiment 2, 63 growing barrows (56.25 ± 2.54 kg) were blocked by BW and randomly allotted to one of seven treatments with nine pens per treatment and one pig per pen. A basal corn-soybean meal diet was formulated to meet nutrient requirements for growing pigs with the exception that STTD P was reduced by 0.18% compared with the requirement, and Ca was included to achieve a Ca:STTD P ratio of 2.15. Six additional diets were formulated by adding 250, 500, 750, 1,000, 1,500, or 2,000 FTU/kg of phytase to the basal diet. Pigs were fed experimental diets for 12 d with 7 d of adaptation and 5 d of fecal sample collection. Results indicated that there was a linear ( P < 0.01) increase in apparent total tract digestibility of ash and ether extract, and STTD of Ca and P also increased (linear, P < 0.05) in response to increasing doses of phytase. Increasing phytase levels in the diets resulted in increase (quadratic, P < 0.05) in apparent ileal digestibility of Arg, His, Ile, Lys, Trp, Asp, and Glu. In conclusion, the novel E. coli phytase was effective in increasing growth performance, bone mineralization, and Ca and P digestibility in pigs fed corn-soybean meal-based diets. In many cell types, epigenetic changes are partially regulated by the availability of metabolites involved in the activity of chromatin-modifying enzymes. Even so, the association between metabolism and the typical epigenetic reprogramming that occurs during preimplantation embryo development remains poorly understood. In this work, we explore the link between energy metabolism, more specifically the tricarboxylic acid cycle (TCA), and epigenetic regulation in bovine preimplantation embryos. Using a morphokinetics model of embryonic development (fast- and slow-developing embryos), we show that DNA methylation (5mC) and hydroxymethylation (5hmC) are dynamically regulated and altered by the speed of the first cleavages. More specifically, slow-developing embryos fail to perform the typical reprogramming that is necessary to ensure the generation of blastocysts with higher ability to establish specific cell lineages. Transcriptome analysis revealed that such differences were mainly associated with enzymes involved in the TCA cycle rather than specific writers/erasers of DNA methylation marks. This relationship was later confirmed by disturbing the embryonic metabolism through changes in α-ketoglutarate or succinate availability in culture media. This was sufficient to interfere with the DNA methylation dynamics despite the fact that blastocyst rates and total cell number were not quite affected. These results provide the first evidence of a relationship between epigenetic reprogramming and energy metabolism in bovine embryos. Likewise, levels of metabolites in culture media may be crucial for precise epigenetic reprogramming, with possible further consequences in the molecular control and differentiation of cells.

A common practice among bodybuilders is the use of carbohydrate loading to improve physical appearance during competition, while limited documented data is available about this issue. The aim of the present study was to evaluate muscle thickness, mood states, gastrointestinal symptoms and subjective silhouette assessment following carbohydrate loading in bodybuilders. Twenty-four male bodybuilders were evaluated at the weighing period following three days of carbohydrate depletion (M1), and 24h of carbohydrate loading leading up to the competition (M2), stratified into: no carbohydrate load (NC, n = 9) and carbohydrate loading (CL, n =1 5). The silhouette scale, Brunel mood scale (BRUMS), muscle thickness (ultrasound), circumferences, and gastrointestinal symptoms (GIS) were evaluated at M1 and M2. The NC displayed no differences in muscle thickness and circumferences between M1 and M2. Body mass, muscle thickness (elbow flexors, a combination of biceps brachii/ brachialis muscle, and triceps brachii) and circumferences (chest, hip, thigh, arm, calves, and forearm) increased significantly (p < 0.05) in the CL at M2. There was a significant increase in photo silhouette scores (p < 0.05) in the CL at M2. There was no significant difference in mood states between groups or time. The most reported GIS was constipation: 7/9 (NC) and 9/15 (CL) during M1 and 6/9 (NC), and 5/15 (CL) at M2 with symptoms described as 'moderate' or 'severe'. Diarrhea was reported by 7/15 CL (4/15 as severe). These data suggest that carbohydrate loading may contribute to an acute increase in muscle volume and physical appearance, however, it needs to be better planned to minimize gastrointestinal symptoms in bodybuilders., (© Journal of Sports Science and Medicine.)

Background: Trained subjects have difficulty in achieving continued results following years of training, and the manipulation of training variables through advanced resistance training (RT) methods is widely recommended to break through plateaus. Objective: The purpose of the present study was to compare the acute effects of traditional RT (TRT) versus two types of sarcoplasma stimulating training (SST) methods on total training volume (TTV), lactate, and muscle thickness (MT). Methods: Twelve trained males (20.75 ± 2.3 years; 1.76 ± 0.14 meters; body mass = 79.41 ± 4.6 kg; RT experience = 4.1 ± 1.8 years) completed three RT protocols in a randomly sequenced order: TRT, SST contraction type (SST-CT), or SST rest interval variable (SST-RIV) with 7 days between trials in arm curl (elbow flexors) and triceps pulley extension (elbow extensors) performed on the same day. Results: The SST groups displayed greater acute biceps and triceps brachii (TB) MT versus the TRT session, with no difference in lactate levels between them. The SST-CT resulted in greater biceps and TB MT versus the SST-RIV session. The TTV was greater for the TRT session versus the SST sessions, except in the case of the elbow flexors (no difference was observed between TRT and SST-CT), and higher for the SST-CT versus the SST-RIV. Conclusion: Trained subjects may benefit from using the SST method as this method may offer a superior MT stimulus and reduced training time, even with a lower TTV.

Purpose: To analyze the early outcomes of viscosupplementation in patients with severe knee osteoarthritis., Method: A randomized, double-blind clinical trial of 143 knees divided into three groups: Group 1 - intra-articular injection of triamcinolone; Group 2 - hylan GF20; and Group 3 - triamcinolone + hylan GF20. Outcomes were evaluated using Lysholm and KSS scores before treatment and after one, three and six months., Results: Within-group comparisons revealed improvements in Lysholm scores in all groups in the one month evaluation relative to pre-treatment levels (p < 0.01). This improvement was maintained in the third month after treatment (p > 0.05). Scores at six months were significantly lower than those observed in the previous follow-up assessments (p < 0.05), but still higher than pre-treatment levels (p < 0.05). KSS scores also improved after one month relative to pre-treatment levels (p < 0.01). This improvement was still present at three and six months after treatment in the corticosteroid group (p > 0.05). Patients treated with hylan GF20 showed lower scores in the last evaluation relative to month one (p < 0.05). No significant differences were observed between the treatment groups (p > 0.05)., Conclusions: Viscosupplementation increased functional scores in patients with severe osteoarthritis of the knee, especially within three months of injection. However, it was not superior to the use of triamcinolone.

Objective: The objective of this study was to evaluate increasing doses of a novel microbial phytase (Cibenza Phytaverse, Novus International, St. Charles, MO, USA) on standardized total tract digestibility (STTD) of P in canola meal (CM), corn, corn-derived distiller's dried grains with solubles (DDGS), rice bran (RB), sorghum, soybean meal (SBM), sunflower meal (SFM), and wheat., Methods: Two cohorts of 36 pigs each (initial body weight = 78.5±3.7 kg) were randomly assigned to 2 rooms, each housing 36 pigs, and then allotted to 6 diets with 6 replicates per diet in a randomized complete block design. Test ingredient was the only dietary source of P and diets contained 6 concentrations of phytase (0, 125, 250, 500, 1,000, or 2,000 phytase units [FTU]/kg) with 0.4% of TiO 2 as a digestibility marker. Feeding schedule for each ingredient was 5 d acclimation, 5 d fecal collection, and 4 d washout. The STTD of P increased (linear or exponential p≤0.001) with the inclusion of phytase for all ingredients., Results: Basal STTD of P was 37.6% for CM, 37.6% for corn, 68.6% for DDGS, 10.3% for RB, 41.2% for sorghum, 36.7% for SBM, 26.2% for SFM, and 55.1% for wheat. The efficiency of this novel phytase to hydrolyze phytate is best described with a broken-line model for corn, an exponential model for CM, RB, SBM, SFM, and wheat, and a linear model for DDGS and sorghum. Based on best-fit model the phytase dose (FTU/kg) needed for highest STTD of P (%), respectively, was 735 for 64.3% in CM, 550 for 69.4% in corn, 160 for 55.5% in SBM, 1,219 for 57.8% in SFM, and 881 for 64.0% in wheat, whereas a maximum response was not obtained for sorghum, DDGS and RB within the evaluated phytase range of 0 to 2,000 FTU/kg. These differences in the phytase concentration needed to maximize the STTD of P clearly indicate that the enzyme does not have the same hydrolysis efficiency among the evaluated ingredients., Conclusion: Variations in enzyme efficacy to release P from phytate in various feedstuffs need to be taken into consideration when determining the matrix value for phytase in a mixed diet, which likely depends on the type and inclusion concentration of ingredients used in mixed diets for pigs. The use of a fixed P matrix value across different diet types for a given phytase concentration is discouraged as it may result in inaccurate diet formulation.

Profilin controls actin nucleation and assembly processes in eukaryotic cells. Actin nucleation and elongation promoting factors (NEPFs) such as Ena/VASP, formins, and WASP-family proteins recruit profilin:actin for filament formation. Some of these are found to be microtubule associated, making actin polymerization from microtubule-associated platforms possible. Microtubules are implicated in focal adhesion turnover, cell polarity establishment, and migration, illustrating the coupling between actin and microtubule systems. Here we demonstrate that profilin is functionally linked to microtubules with formins and point to formins as major mediators of this association. To reach this conclusion, we combined different fluorescence microscopy techniques, including superresolution microscopy, with siRNA modulation of profilin expression and drug treatments to interfere with actin dynamics. Our studies show that profilin dynamically associates with microtubules and this fraction of profilin contributes to balance actin assembly during homeostatic cell growth and affects micro-tubule dynamics. Objectives: in this study, two research tools were validated to study the impact of technological influence on health professionals' care practice., Methods: the following methodological steps were taken: bibliographic review, selection of the scales, translation and cultural adaptation and analysis of psychometric properties., Results: the psychometric properties of the scale were assessed based on its application to a sample of 341 individuals (nurses, physicians, final-year nursing and medical students). The validity, reliability and internal consistency were tested. Two scales were found: Caring Attributes Questionnaire (adapted) with a Cronbach's Alpha coefficient of 0.647 and the Technological Influence Questionnaire (adapted) with an Alpha coefficient of 0.777., Conclusions: the scales are easy to apply and reveal reliable psychometric properties, an additional quality as they permit generalized studies on a theme as important as the impact of technological influence in health care.

Large profit losses in the swine industry can be attributed to morbidity and mortality of piglets before weaning, especially in the low birth weight (LBW) piglet. Recent evidence suggests sow's milk contains insufficient concentrations of Arg to support optimal growth and health of piglets. Therefore, our objective was to assess global metabolomic profiles and the potential for Arg supplementation to promote growth of LBW (≤0.9 kg BW) and average birth weight (ABW; 1.3 to 1.5 kg BW) piglets. Piglets were selected in littermate pairs at processing to receive either Arg or an isonitrogenous control (Ala) and weighed daily to assess growth rate, and blood was collected at approximately 16 d of age for metabolomics analysis. In terms of growth, LBW and ABW piglets supplemented with Arg weighed 22.3 and 12.7% less, respectively, at d 16 compared with Ala-supplemented piglets of the same birth weight group. Overall, differences ( < 0.05) were observed among treatments for metabolic pathways involving energy (i.e., tricarboxylic acid cycle intermediates), AA, nucleotides, and fatty acids. Increased nucleotide turnover, indicative of an increase in DNA damage and cell death, was particularly noted in the LBW piglet. However, Arg supplementation reduced these effects to levels comparable to those observed in ABW piglets. Moreover, changes in glucose metabolism suggested a compromised ability to extract energy from dietary sources may have occurred in the LBW piglet, but these effects were partially recovered by Arg supplementation. We conclude that a reduction in the growth potential of LBW piglets may be associated with alterations in multiple metabolic pathways, and further reduction due to Arg supplementation may have resulted from perturbations in multiple metabolic pathways.

There exists a conspicuous gap of knowledge about the organization of life at mesoscopic levels. Ultra-fast coherent diffractive imaging with X-ray free-electron lasers can probe structures at the relevant length scales and may reach sub-nanometer resolution on micron-sized living cells. Here we show that we can introduce a beam of aerosolised cyanobacteria into the focus of the Linac Coherent Light Source and record diffraction patterns from individual living cells at very low noise levels and at high hit ratios. We obtain two-dimensional projection images directly from the diffraction patterns, and present the results as synthetic X-ray Nomarski images calculated from the complex-valued reconstructions. We further demonstrate that it is possible to record diffraction data to nanometer resolution on live cells with X-ray lasers. Extension to sub-nanometer resolution is within reach, although improvements in pulse parameters and X-ray area detectors will be necessary to unlock this potential.

Two experiments were conducted to determine DE and ME and the apparent ileal digestibility (AID) and the standardized ileal digestibility (SID) of CP and AA in corn and corn coproducts (high-fat corn germ [HFCG], corn bran, liquid corn extractives [LCE], and a mixture of corn germ meal and LCE [CGM-LCE]) fed to growing pigs. In Exp. 1, 40 growing barrows (initial BW: 33.4 ± 5.77 kg) were housed individually in metabolism cages and randomly allotted to 1 of 5 diets. A corn-based diet (97.4% corn) and 4 diets that contained both corn and each of the corn coproducts were formulated. Each diet was fed to 8 pigs. Feces and urine samples were collected using the marker to marker method with 5-d adaptation and 5-d collection periods. The DE and ME were calculated using the difference procedure. The concentrations of DE and ME in HFCG, corn bran, LCE, and CGM-LCE were less (P < 0.05) than in corn. Among corn coproducts, the concentration of DE in HFCG was greater (P < 0.05) than in corn bran, but the DE in corn bran was not different from DE values in LCE and CGM-LCE. No differences were observed in the ME concentrations among corn coproducts. In Exp. 2, 6 growing barrows (initial BW: 96.6 ± 1.16 kg) with a T-cannula in the distal ileum were randomly allotted to a 6 × 6 Latin square design with 6 diets and 6 periods. A N-free diet and 5 diets that contained corn, HFCG, corn bran, LCE, or CGM-LCE as the sole source of CP and AA were formulated. Each period lasted 7 d and ileal digesta were collected on d 6 and 7 of each period. The SID of CP and all indispensable AA was greater (P < 0.05) in corn than in all corn coproducts with the exception that the SID of Lys in corn was not different from the SID of Lys in HFCG, and the SID of Trp in corn was also not different from the SID of Trp in CGM-LCE. Among corn coproducts, the SID of CP, Arg, and Lys were greater (P < 0.05) in HFCG and CGM-LCE than in corn bran, the SID of Lys and Val was greater (P < 0.05) in LCE than in corn bran, and the SID of Arg was greater (P < 0.05) in HFCG and CGM-LCE than in LCE, but for the remaining indispensable AA, no differences among corn coproducts were observed. In conclusion, the corn coproducts used in these experiments contain less ME and have reduced SID of most AA compared with corn, but there are no differences in ME among corn coproducts and only few differences in the SID of indispensable AA among HFCG, corn bran, LCE, and CGM-LCE.

Two experiments were conducted to investigate if adjustments in diet formulations either based on total analysed amino acids or standardized ileal digestible (SID) amino acids may be used to eliminate negative effects of including heat-damaged soybean meal (SBM) or heat-damaged corn distillers dried grains with solubles (DDGS) in diets fed to weanling pigs. In Experiment 1, four corn-SBM diets were formulated. Diet 1 contained non-autoclaved SBM (315 g/kg), and this diet was formulated on the basis of analysed amino acid concentrations and using SID values from the AminoDat® 4.0 database. Diet 2 was similar to Diet 1 in terms of ingredient composition, except that the non-autoclaved SBM was replaced by autoclaved SBM at 1 : 1 (weight basis). Diet 3 was formulated using autoclaved SBM and amino acid inclusions in the diet were adjusted on the basis of analysed total amino acid concentrations in the autoclaved SBM and published SID values for non-autoclaved SBM (AminoDat® 4.0). Diet 4 also contained autoclaved SBM, but the formulation of this diet was adjusted on the basis of analysed amino acids in the autoclaved SBM and SID values that were adjusted according to the degree of heat damage in this source of SBM. Pigs (160; initial BW: 10.4 kg) were allotted to the four treatments with eight replicate pens per treatment in a randomized complete block design. Diets were fed to pigs for 21 days. The gain to feed ratio (G : F) was greater (P<0.05) for pigs fed Diet 1 compared with pigs fed the other diets and pigs fed Diet 4 had greater (P<0.05) G : F than pigs fed Diet 2. In Experiment 2, 144 pigs (initial BW: 9.9 kg) were allotted to four diets with eight replicate pens per diet. The four diets contained corn, SBM (85 g/kg) and DDGS (220 g/kg), and were formulated using the concepts described for Experiment 1, except that heat-damaged DDGS, but not heat-damaged SBM, was used in the diets. Pigs fed Diet 1 had greater (P<0.05) G : F than pigs fed Diet 2, but no differences were observed for G : F among pigs fed diets containing autoclaved DDGS. Results demonstrate that the negative effects of heat damage of SBM or DDGS may be ameliorated if the reduced concentration and digestibility of amino acids in heat-damaged SBM or DDGS is taken into account in diet formulation. Further research is needed to improve the prediction of the ileal digestibility of amino acids in heat-processed ingredients used in practical diet formulations.

Production of crystalline l-Thr results in the generation of a Thr biomass that contains more than 80% CP, 5.20% Lys, 5.10% Val, 4.52% Thr, 4.15% Ile, and 1.06% Trp. This Thr biomass product can possibly be used as a feed ingredient in diets fed to weanling pigs, but there is little information about the nutritional value of this product. The objective of this work was to determine the AA digestibility and energy concentration in Thr biomass and to compare these values to values obtained for fish meal in diets fed to pigs. The apparent ileal digestibility (AID) and the standardized ileal digestibility (SID) of CP and AA were determined in Exp. 1. Nine pigs (initial BW: 13.4 ± 2.5 kg) were equipped with a T-cannula in the distal ileum and allotted to a triplicated 3 × 3 Latin square design with 3 diets and 3 periods in each square. One diet contained 20.0% Thr biomass as the sole source of AA, and a second diet contained 25.0% fish meal as the sole source of AA. The last diet was a N-free diet that was used to measure basal endogenous losses of AA and CP. Results indicated that the AID and SID of all AA except Trp, Gly, and Pro were greater (P < 0.05) in Thr biomass than in fish meal. In Exp. 2, 24 pigs (initial BW: 18.1 ± 3.5 kg) were placed in metabolism cages and randomly allotted to 3 diets. The first diet contained 96.4% corn, the second diet contained 79.3% corn and 17.0% Thr biomass, and the third diet contained 75.3% corn and 24.0% fish meal. Total collection of feces and urine was performed for 5 d after a 5-d adaptation period, and all samples of ingredients, diets, feces, and urine were analyzed for GE. Digestible energy and ME were then calculated. The DE in the Thr biomass was greater (P < 0.05) than in fish meal and corn (4,935 vs. 3,938 and 3,939 kcal DE/kg DM, respectively), and the ME in the Thr biomass was also greater (P < 0.05) than in fish meal and corn (4,335 vs. 3,508 and 3,839 kcal ME/kg DM, respectively). Results from these experiments indicate that Thr biomass is an excellent source of AA and ME. Therefore, the Thr biomass is a viable ingredient that can be used as an alternative to fish meal and possibly other animal proteins in diets for pigs.

The objective of this experiment was to determine the standardized ileal digestibility (SID) of amino acids (AA) in 3 sources of distillers dried grains with solubles (DDGS) with different concentrations of fat. Twelve growing barrows (initial body weight: 76.1 [Formula: see text] 6.2 kg) were randomly allotted to a replicated 6 × 4 Youden square design with 6 diets and 4 periods. The fat content of the 3 sources of DDGS were 11.5, 7.5, and 6.9% respectively. Diets contained 60% DDGS and fat concentration of the diets were 7.5, 5.2, and 5.2%, respectively. Two additional diets containing the 2 sources of DDGS with 7.5 and 6.9% fat were also formulated, and corn oil was added to these diets to increase the concentration of fat in the diets to levels that were calculated to be similar to the diet containing conventional DDGS with 11.5% fat. A N-free diet was also formulated to calculate endogenous losses of crude protein (CP) and AA from the pigs. Pigs were fed experimental diets during four 7-d periods. The first 5 d of each period were an adaptation period and ileal digesta were collected on d 6 and 7 of each period. The apparent ileal digestibililty (AID) and SID of CP and all indispensable AA, except AID Pro and SID of Trp, were greater (P < 0.01) in conventional DDGS than in the 2 sources of DDGS with reduced fat. Adding oil to the diets containing the 2 sources of DDGS with reduced fat did not consistently increase SID of AA. In conclusion, conventional DDGS has greater SID values for most AA compared with DDGS that contains less fat and inclusion of additional oil to diets containing low-fat DDGS does not increase AID or SID of AA. The lower AA digestibility in low-fat DDGS could not be overcome by the inclusion of additional fat to the diets.

Two experiments were conducted to determine the effects of heat damage, achieved by autoclaving, on the nutritional composition and on the standardized ileal digestibility (SID) of AA in sunflower meal (SFM) and cottonseed meal (CSM) fed to growing pigs. The second objective was to establish a relationship between the concentration of SID AA in SFM and CSM and the concentration of fiber components, reducing sugars, and AA. In Exp. 1, 10 growing pigs (initial BW: 23.1 ± 1.3 kg) were surgically equipped with a T-cannula in the distal ileum and allotted to a replicated 5 × 5 Latin square design with 5 diets and 5 periods in each square. A common source of SFM was separated into 4 batches that were either not autoclaved or autoclaved at 130°C for 20, 40, or 60 min. Four diets (approximately 14.5% CP) that contained each of the 4 batches of SFM were formulated, and SFM was the only source of CP and AA in the diets. A N-free diet that was used to determine the basal endogenous losses of CP and AA from pigs was also formulated. Each period consisted of 5 d of adaptation to the diets followed by 2 d of ileal digesta collection. The SID of Lys in SFM was reduced (linear, P < 0.05) from 83.2% in nonautoclaved SFM to 63.5% in SFM autoclaved for 60 min at 130°C. The concentrations of total Lys and reducing sugars in SFM may be used as predictors (R(2) = 0.85) of the concentration of SID Lys in SFM. In Exp. 2, 10 growing pigs (initial BW: 35.0 ± 1.5 kg) were surgically equipped with a T-cannula in the distal ileum and allotted to a replicated 5 × 5 Latin square design with 5 diets and 5 periods in each square. A source of CSM was separated into 4 batches that were either not autoclaved or autoclaved at 130°C for 15, 35, or 60 min. Four diets (approximately 13.4% CP) containing CSM as the only source of CP and AA were formulated. A N-free diet was also formulated and used as described for Exp. 1. The SID of Lys in nonautoclaved CSM (66.2%) was greater (P < 0.05) than in autoclaved (60 min at 130°C) CSM (54.1%). The equation (R(2) = 0.68) that best predicted the concentration of SID Lys in CSM included the concentration of ADIN. In conclusion, heat damage reduces the SID of AA in SFM and CSM. For SFM, the concentration of SID Lys may be predicted from the concentrations of total Lys and reducing sugars. The concentration of ADIN may be used to predict the concentration of SID Lys in CSM.

Dehydroepiandrosterone (DHEA) and the dehydroepiandrosterone sulfate (DHEA-S) are steroids produced mainly by the adrenal cortex. There is evidence from both human and animal models suggesting beneficial effects of these steroids for obesity, diabetes mellitus, hypertension, and osteoporosis, conditions associated with the post-menopausal period. Accordingly, we hypothesized that DHEA supplementation in ovariectomized (OVX) female rats fed a high-fat diet would maintain glucose-induced insulin secretion (GSIS) and pancreatic islet function. OVX resulted in a 30% enlargement of the pancreatic islets area compared to the control rats, which was accompanied by a 50% reduction in the phosphorylation of AKT protein in the pancreatic islets. However, a short-term high-fat diet induced insulin resistance, accompanied by impaired GSIS in isolated pancreatic islets. These effects were reversed by DHEA treatment, with improved insulin sensitivity to levels similar to the control group, and with increased serine phosphorylation of the AKT protein. These data confirm the protective effect of DHEA on the endocrine pancreas in a situation of diet-induced overweight and low estrogen concentrations, a phenotype similar to that of the post-menopausal period.

Introduction: This paper proposes a new methodology to simultaneously select the most relevant SNPs markers for the characterization of any measurable phenotype described by a continuous variable using Support Vector Regression with Pearson Universal kernel as fitness function of a binary genetic algorithm. The proposed methodology is multi-attribute towards considering several markers simultaneously to explain the phenotype and is based jointly on statistical tools, machine learning and computational intelligence., Results: The suggested method has shown potential in the simulated database 1, with additive effects only, and real database. In this simulated database, with a total of 1,000 markers, and 7 with major effect on the phenotype and the other 993 SNPs representing the noise, the method identified 21 markers. Of this total, 5 are relevant SNPs between the 7 but 16 are false positives. In real database, initially with 50,752 SNPs, we have reduced to 3,073 markers, increasing the accuracy of the model. In the simulated database 2, with additive effects and interactions (epistasis), the proposed method matched to the methodology most commonly used in GWAS., Conclusions: The method suggested in this paper demonstrates the effectiveness in explaining the real phenotype (PTA for milk), because with the application of the wrapper based on genetic algorithm and Support Vector Regression with Pearson Universal, many redundant markers were eliminated, increasing the prediction and accuracy of the model on the real database without quality control filters. The PUK demonstrated that it can replicate the performance of linear and RBF kernels.

Two experiments were conducted to determine the standardized ileal digestibility (SID) of AA (Exp. 1), the standardized total tract digestibility (STTD) of P, and the concentration of DE and ME (Exp. 2) in hydrolyzed feather meal (FM) fed to growing pigs. Eight samples of FM were obtained from 4 different processing plants (FM1, FM2, FM3, and FM4). Each plant provided samples of FM without and with added blood. In Exp. 1, 10 barrows (initial BW: 24.0 ± 0.8 kg) were prepared with a T-cannula in the distal ileum and allotted to a 10 × 10 Latin square design with 10 diets and ten 7-d periods. A N-free diet, a diet based on soybean meal (SBM), and 8 diets based on a combination of SBM and each of the 8 sources of FM were formulated. Values for the SID of CP and AA in each source of FM were calculated using the difference procedure. The SID of CP and all AA was different (P < 0.001) among sources of FM. Among the indispensable AA, the overall effect of addition of blood was statistically significant (P < 0.05) for the SID of Ile, Leu, Lys, Phe, and Val, but for some sources, the SID of these AA was increased by addition of blood, whereas for other sources, the SID was reduced or not changed (interaction, P < 0.05). As an example, the SID of Lys in FM3 and FM4 with added blood were greater (P < 0.05) than in FM3 and FM4 without blood, but no difference in the SID of Lys was observed for FM1 and FM2 without and with blood (interaction, P < 0.01). In Exp. 2, 72 growing barrows (initial BW: 13.3 ± 1.5 kg) were used with 9 diets and 8 replicate pigs per diet. A corn-diet consisting of 98.4% corn was formulated and 8 additional diets were formulated by mixing corn with 25% FM. The STTD of P and the DE and ME of each source of FM were calculated using the difference procedure. The STTD of P tended (P = 0.09) to be different among FM sources and the STTD of P in FM with added blood was less than in FM without blood. On a DM basis, DE and ME values were affected (P < 0.05) by both source of FM and addition of blood. However, an interaction (P < 0.05) between source of FM and addition of blood for ME was observed because addition of blood to FM3 and FM4 reduced (P < 0.05) the ME, whereas addition of blood to FM1 and FM2 had no impact on ME. In summary, the SID of AA, STTD of P, and the energy concentration vary among sources of FM and the effects of adding blood to FM is not consistent among sources.

Background and Objectives: Few longitudinal studies have examined the long-term effect on deferral for low haematocrit (Hct) or haemoglobin, indicators of presence of anaemia. This study retrospectively analysed 11 years of donation history to examine predictors related to such deferrals among repeat blood donors., Materials and Methods: We included 385,357 donors with at least two visits to the blood centre between January 1996 and December 2006 who were not deferred due to haematocrit at their first visit. We evaluated variables related to the development of low Hct (LHct-below 38% for females and 39% for males) after whole blood donations., Results: Over the 11-year period, 3,850 (1.5%) of the 252,301 males and 18,104 (13.6%) of the 133,056 females were deferred due to LHct at some point after their first donation. Genders, age, baseline Hct, Hct at the visit immediately before deferral due to LHct, and interval between donations, were associated with higher rates of development of LHct in repeat donors., Conclusion: Our analysis showed that deferral due to low Hct levels in repeat blood donors is highly prevalent in Brazil. Assigning longer donations intervals based on the Hct levels at the qualifying donation or supplementing iron to donors at risk may decrease deferral rate of donors with low Hct., (Copyright © 2013 Elsevier Ltd. Objective: To assess the prevalence of low body weight/thinness, overweight and obesity in a representative sample of children and adolescents from a Brazilian region with low economic development., Methods: A total of 982 girls and 986 boys, aged seven to 17 years old and assisted by Segundo Tempo Program, from Montes Claros, Minas Gerais, Brazil, were included in the study. Low body weight/thinness, overweight and obesity were defined based on body mass cut-off indexes recommended by the International Obesity Task Force. The prevalence of the nutritional status according to sex and age was compared by chi-square test., Results: In girls, the frequency of low body weight/thinness, overweight and obesity was 4.1, 18.4 and 3.8%, respectively; in boys, these percentages were 6.3, 13.2 and 2.9%, respectively. The low body weight/thinness for girls raised from 2.7% (7-10 years old) to 5.5% (15-17 years old); the body weight excess (overweight and obesity) decreased from 30.1 to 16.2% for the same age groups. In boys, the corresponding trends were from 3.2 to 9.4% for low body weight/thinness, and from 23.4 to 9.2%, for body weight excess., Conclusions: The data indicate that, even in a region with low economic status, the body weight excess was the main problem associated with nutritional health. The high overweight and obesity prevalence rates indicate the need of public policies for promoting healthy feeding behaviors and physical activity.

The primary objective of this experiment was to determine the effects of heat treatment on the standardized ileal digestibility (SID) of amino acids (AA) in corn distillers dried grains with solubles (DDGS) fed to growing pigs. The second objective was to develop regression equations that may be used to predict the concentration of SID AA in corn DDGS. A source of corn DDGS was divided into 4 batches that were either not autoclaved or autoclaved at 130°C for 10, 20, or 30 min. Four diets containing DDGS from each of the 4 batches were formulated with DDGS being the only source of AA and CP in the diets. A N-free diet also was formulated and used to determine the basal endogenous losses of CP and AA. Ten growing pigs (initial BW: 53.5 ± 3.9 kg) were surgically equipped with a T-cannula in the distal ileum and allotted to a replicated 5 × 4 Youden square design with 5 diets and 4 periods in each square. The SID of CP decreased linearly (P < 0.05) from 77.9% in non-autoclaved DDGS to 72.1, 66.1, and 68.5% in the DDGS samples that were autoclaved for 10, 20, or 30 min, respectively. The SID of lysine was quadratically reduced (P < 0.05) from 66.8% in the non-autoclaved DDGS to 54.9, 55.3, and 51.9% in the DDGS autoclaved for 10, 20, or 30 min, respectively. The concentrations of SID Arginine, Histidine, Leucine, Lysine, Methionine, Phenylalanine, or Threonine may be best predicted by equations that include the concentration of acid detergent insoluble N in the model (r2 = 0.76, 0.68, 0.67, 0.84, 0.76, 0.73, or 0.54, respectively). The concentrations of SID Isoleucine and Valine were predicted (r2 = 0.58 and 0.54, respectively) by the Lysine:CP ratio, whereas the concentration of SID Tryptophan was predicted (r2 = 0.70) by the analyzed concentration of Tryptophan in DDGS. In conclusion, the SID of AA is decreased as a result of heat damage and the concentration of SID AA in heat-damaged DDGS may be predicted by regression equations developed in this experiment.

Long-chain fatty acids are capable of inducing alterations in the homoeostasis of glucose-stimulated insulin secretion (GSIS), but the effect of medium-chain fatty acids (MCFA) is poorly elucidated. In the present study, we fed a normoenergetic MCFA diet to male rats from the age of 1 month to the age of 4 months in order to analyse the effect of MCFA on body growth, insulin sensitivity and GSIS. The 45% MCFA substitution of whole fatty acids in the normoenergetic diet impaired whole body growth and resulted in increased body adiposity and hyperinsulinaemia, and reduced insulin-mediated glucose uptake in skeletal muscle. In addition, the isolated pancreatic islets from the MCFA-fed rats showed impaired GSIS and reduced protein kinase Ba (AKT1) protein expression and extracellular signal-related kinase isoforms 1 and 2 (ERK(1/2)) phosphorylation, which were accompanied by increased cellular death. Furthermore, there was a mildly increased cholinergic sensitivity to GSIS. We discuss these findings in further detail, and advocate that they might have a role in the mechanistic pathway leading to the compensatory hyperinsulinaemic status found in this animal model.

RhoD is a member of the classical Rho GTPases and it has an essential role in the regulation of actin dynamics. Furthermore, RhoD also localizes to early endosomes and recycling endosomes, indicating additional roles in the regulation of endosome trafficking. A yeast two-hybrid screen identified Zipper-Interacting Protein Kinase (ZIPK) as a RhoD target. We found that RhoD interacts with ZIP kinase in a GTP dependent manner and modulates actin and focal adhesion reorganization. Interestingly, while ectopic expression of ZIPK in fibroblasts induces actin reorganization and actomyosin contraction seen as stress fiber bundling and membrane blebbing, the concomitant expression of active RhoD suppressed this phenotype. Previously, RhoD has been associated with focal adhesion regulation, and in line with this notion, we observed that ZIPK resulted in reorganization of focal adhesion and increased adhesion size. Importantly, the RhoD activity suppressed ZIPK-dependent effects on FAK activity, indicating a functional interplay between RhoD and FAK in the focal adhesion dynamics., (Copyright © 2013 Elsevier Inc. In 2 experiments, 48 weanling (initial BW: 13.5 ± 2.4 kg, Exp. 1) and 24 growing pigs (initial BW: 36.2 ± 4.0 kg, Exp. 2) were used to determine effects of a novel bacterial 6-phytase expressed in Aspergillus oryzae on the apparent total tract digestibility (ATTD) of phosphorus and calcium in corn-soybean meal diets fed to weanling and growing pigs. In Exp. 1 and 2, pigs were randomly allotted to 6 dietary treatments using a randomized complete block design and a balanced 2 period changeover design, respectively. In both experiments, 6 diets were formulated. The positive control diet was a corn-soybean meal diet with added inorganic phosphorus (Exp. 1: 0.42 and 0.86% standardized total tract digestible phosphorus and total calcium, respectively; Exp. 2: 0.32 and 0.79% standardized total tract digestible phosphorus and total calcium, respectively). A negative control diet and 4 diets with the novel phytase (Ronozyme HiPhos, DSM Nutritional Products Inc., Parsippany, NJ) added to the negative control diet at levels of 500, 1,000, 2,000, and 4,000 phytase units (FYT)/kg were also formulated. In Exp. 1, the ATTD of phosphorus was greater (P < 0.01) for the positive control diet (60.5%) than for the negative control diet (40.5%), but increased (linear and quadratic, P < 0.01) as phytase was added to the negative control diet (40.5% vs. 61.6%, 65.1%, 68.7%, and 68.0%). The breakpoint for the ATTD of phosphorus (68.4%) was reached at a phytase inclusion level of 1,016 FYT/kg. In Exp. 2, the ATTD of phosphorus was greater (P < 0.01) for the positive control diet (59.4%) than for the negative control diet (39.8%) and increased (linear and quadratic, P < 0.01) as phytase was added to the negative control diet (39.8% vs. 58.1%, 65.4%, 69.1%, and 72.8%). The breakpoint for the ATTD of phosphorus (69.1%) was reached at a phytase inclusion level of 801 FYT/kg. In conclusion, the novel bacterial 6-phytase improved the ATTD of phosphorus and calcium in both weanling and growing pigs. The optimum level of inclusion for this phytase is 800 to 1,000 FYT/kg of complete feed to maximize ATTD of phosphorus and calcium in weanling and growing pigs.

Diabetes mellitus (DM) is a great public health problem, which attacks part of the world population, being characterized by an imbalance in body glucose homeostasis. Physical exercise is pointed as a protective agent and is also recommended to people with DM. As pancreatic islets present an important role in glucose homeostasis, we aim to study the role of physical exercise (chronic adaptations and acute responses) in pancreatic islets functionality in Wistar male rats. First, animals were divided into two groups: sedentary (S) and aerobic trained (T). At the end of 8 weeks, half of them (S and T) were submitted to an acute exercise session (exercise until exhaustion), being subdivided as acute sedentary (AS) and acute trained (AT). After the experimental period, periepididymal, retroperitoneal and subcutaneous fat pads, blood, soleus muscle and pancreatic islets were collected and prepared for further analysis. From the pancreatic islets, total insulin content, insulin secretion stimulated by glucose, leucine, arginine and carbachol were analyzed. Our results pointed that body adiposity and glucose homeostasis improved with chronic physical exercise. In addition, total insulin content was reduced in group AT, insulin secretion stimulated by glucose was reduced in trained groups (T and AT) and insulin secretion stimulated by carbachol was increased in group AT. There were no significant differences in insulin secretion stimulated by arginine and leucine. We identified a possible modulating action on insulin secretion, probably related to the association of chronic adaptation with an acute response on cholinergic activity in pancreatic islets.

An experiment was conducted to determine the influence of adding graded levels of microbial phytase to corn, distillers dried grains with solubles (DDGS), high-protein distillers dried grains (HP-DDG), and corn germ on the standardized total tract digestibility (STTD) of P. A second objective was to develop regression equations to predict the response of adding phytase to each of these ingredients. Four corn-based diets, 4 DDGS-based diets, 4 HP-DDG-based diets, and 4 corn germ-based diets were formulated. The 4 diets with each ingredient were formulated to contain 0, 500, 1,000, or 1,500 phytase units (FTU)/kg. A P-free diet was also formulated to determine basal endogenous losses of P. A total of 102 pigs (initial BW: 18.2 ± 2.1 kg) were individually housed in metabolism cages equipped with a feeder, a nipple drinker, and a screen floor that allowed for total collection of feces. Pigs were allotted to the 17 diets in a randomized complete block design with 6 replicate pigs per diet. Pigs were fed their respective diets for 12 d, and feces were collected quantitatively from d 6 to 11. Supplementation with 500, 1,000, or 1,500 FTU of microbial phytase/kg increased (linear, P < 0.01; quadratic, P < 0.05) the STTD of P in corn from 40.9 to 67.5, 64.5, and 74.9%, respectively, tended to increase (linear, P = 0.07) the STTD of P in DDGS from 76.9 to 82.9, 82.5, and 83.0%, respectively, increased (linear, P < 0.01; quadratic, P < 0.05) the STTD of P in HP-DDG from 77.1 to 88.0, 84.1, and 86.9%, respectively, and increased (linear and quadratic, P < 0.01) the STTD of P in corn germ from 40.7 to 59.0, 64.4, and 63.2%, respectively. Regression equations were developed to calculate the STTD of P in corn and corn germ, and R(2) values were 0.63 and 0.79, respectively. However, for DDGS and HP-DDG, the R(2) values were only 0.20 and 0.36, respectively, and these equations were, therefore, not considered adequate to predict the STTD of P. In conclusion, the increase in the STTD of P in corn and corn germ that is a result of microbial phytase can be predicted by regression equations, but microbial phytase has much less of an effect on the STTD of P in DDGS and HP-DDG and responses to addition of graded levels of phytase to these ingredients can, therefore, not be accurately predicted by regression equations.

Physical inactivity leads to the accumulation of visceral fat and, consequently, to the activation of a network of inflammatory pathways which may promote development of insulin resistance, atherosclerosis, neurodegeneration, and tumour growth. These conditions belong to the "diseasome of physical inactivity". In contrast, the protective effect of regular exercise against diseases associated with chronic inflammation may to some extent be ascribed to an anti-inflammatory effect. The so called "acute exercise threshold", the complex mixture of several variables involved in exercise, such as type, volume, frequency, and intensity range is capable of inducing positive physiological adaptations and has been specifically addressed in the recent literature. The major concern is related to the level of the threshold: "exercise training shifts from a therapeutic adaptive intervention to one with potential pathological consequences". Nonetheless, if the mechanical stimulus is too weak to disrupt cellular homeostasis, training adaptations will not occur. Answering these questions could present practical applications, especially during inflammatory diseases associated with detrimental muscle effects and could theoretically constitute a "new" therapeutic approach to treat/improve an inflammatory state. This paper aims to describe specific data from the literature regarding the effects of exercise on inflammatory diseases in order to promote a more sophisticated perspective on the anti-inflammatory effects of exercise.

The objectives of this experiment were to determine the apparent ileal digestibility and the standardized ileal digestibility (SID) of CP and AA in bakery meal, corn gluten meal, corn gluten feed, corn germ meal, and hominy feed and to compare these values with the apparent ileal digestibility and SID of CP and AA in corn and distillers dried grains with solubles (DDGS). Eight growing barrows (initial BW: 82.5 ± 5.5 kg) were randomly allotted to an 8 × 8 Latin square design with 8 diets and 8 periods. Diets contained corn, DDGS, bakery meal, corn gluten meal, corn gluten feed, corn germ meal, or hominy feed as the sole source of protein and AA. An N-free diet was used to measure basal endogenous losses of AA and protein. Pigs were fed experimental diets for eight 7-d periods, with ileal digesta being collected on d 6 and 7 of each period. Results indicated that the SID of Lys in corn gluten meal (78.7%) was greater (P < 0.01) than in DDGS, bakery meal, corn germ meal, and hominy feed (46.0, 48.4, 68.4, and 58.8%, respectively). The SID of all indispensable AA except Arg, Leu, and Met in bakery meal were not different from those in DDGS. The SID of Arg, His, Leu, and Met in corn gluten feed were less (P < 0.01) than in corn, but the SID of all other indispensable AA in corn gluten feed were not different from those in corn. However, for most indispensable AA, the SID in corn gluten feed was not different from the SID in DDGS. The SID of all indispensable AA in corn germ meal, except Arg, His, Leu, and Met, were not different from the SID in corn. Likewise, the SID of all indispensable AA in corn germ meal, except Arg and Leu, were not different from those in DDGS. The SID of Ile, Met, Phe, and Val in hominy feed were less (P < 0.01) than in corn, but the SID of the remaining indispensable AA in hominy feed were not different from the SID of indispensable AA in corn. All indispensable AA in hominy feed also had SID values that were not different from the SID values of AA in DDGS, except for Arg and Lys, which had greater (P < 0.01) SID than in DDGS. In conclusion, bakery meal had SID values of most AA that were less than in corn, but corn gluten meal had SID values for most AA that were greater than the SID of AA in corn, bakery meal, and corn coproducts.

Three experiments were conducted to test the hypotheses that pigs fed diets that are equal in digestible P will perform equally regardless of the concentration of total P in the diets, and that the addition of microbial phytase, distillers dried grains with solubles (DDGS), or a combination of phytase and DDGS will result in a reduction in P excretion. In Exp. 1, a P-free diet and 6 diets containing corn, soybean meal (SBM), or DDGS without or with microbial phytase (500 phytase units per kg) were formulated. Diets were fed for 12 d to 42 pigs (initial BW = 13.5 +/- 3.9 kg) housed in metabolism cages that allowed for total collections of feces. Basal endogenous P losses were determined to be 199 mg/kg of DMI for pigs fed the P-free diet. Addition of phytase increased (P < 0.01) the standardized total tract digestibility (STTD) of P in corn (64.4 vs. 26.4%) and SBM (74.9 vs. 48.3%), but there was no effect (P > 0.10) of the addition of phytase on the STTD of P in DDGS (75.5 vs. 72.9%). In Exp. 2, a total of 160 pigs (initial BW = 11.25 +/- 1.95 kg; 4 pigs/pen) were allotted to 4 corn- and SBM-based diets with 2 amounts of phytase (0 or 500 phytase units per kg) and 2 amounts of DDGS (0 or 20%) in a 2 x 2 factorial arrangement of treatments. All diets were formulated to contain 0.32% STTD of P according to the STTD values determined in Exp. 1. Diets were fed for 21 d and results indicated that inclusion of phytase in the diet containing no DDGS tended (P < 0.10) to decrease G:F, but inclusion of 20% DDGS in the diets tended (P < 0.10) to increase ADG, ADFI, and final BW. In Exp. 3, the diets used in Exp. 2 were fed to 24 pigs (initial BW = 14.6 +/- 1.4 kg) that were placed in metabolism cages individually. Feces and urine were collected for 5 d. Phytase and DDGS increased (P < 0.01) the apparent total tract digestibility of P in the diets. Absorption of P was greater (P < 0.05) in pigs fed corn-SBM-DDGS diets than pigs fed corn-SBM diets, and phytase, DDGS, or the combination of phytase and DDGS reduced (P < 0.01) P excretion. In conclusion, the addition of phytase increased the STTD of P in corn and SBM, but had no effect on the STTD of P in DDGS. Diets may be formulated based on STTD values without compromising pig performance, and dietary phytase, DDGS, or the combination of phytase and DDGS will reduce P excretion by growing pigs.

Exercise performed before infections has been linked to improvement of the immune response against infections. The purpose of this study was to evaluate the influence of preinfection moderate-intensity treadmill training on acute Trypanosoma cruzi infection in mice. Ninety-nine female BALB/c mice were divided into 4 groups, as follows: training + infection (T+I) (n = 41); no training + infection (NT+I) (n = 38); training + no infection (T+NI) (n = 10); and no training + no infection (NT+NI) (n = 10). The exercise program for trained groups was carried out on a motorized treadmill for 8 weeks. Infected groups were inoculated with the Y strain of T. cruzi. Infectivity, prepatent period, patent period, parasitemia peak, mortality, survival time, weight, food intake, tumor necrosis factor-alpha serum levels, and peritoneal macrophage hydrogen peroxide production were evaluated. We found that preinfection training induced statistically significant reductions in parasitemia peak (p < 0.03) and weight loss (p < 0.04). However, no statistically significant differences were found for the other parameters evaluated when trained and nontrained infected groups were compared. We conclude that preinfection aerobic training induces some improvement in the immune response to T. cruzi infection in female BALB/c mice.
