Abstract

Background and Objectives: The relationship between hepatitis C virus (HCV) infection and melanoma remains poorly understood. This study aimed to investigate the association between HCV and melanoma, assess outcomes in patients with both conditions, and explore potential molecular mechanisms connecting the two diseases. Materials and Methods: We conducted a retrospective cohort study of 142 melanoma patients, including 29 with HCV-related cirrhosis, and analyzed their clinical outcomes. For external validation, we used the TriNetX Global Collaborative Network database, comprising 219,960 propensity-matched patients per group. An in silico analysis was performed to identify the molecular pathways linking HCV and melanoma. Results: In the retrospective cohort, HCV-positive melanoma patients showed an increased risk of early relapse (41.4% vs. 18.6%, p = 0.014), recurrence (65.5% vs. 39.8%, p = 0.020), and mortality (65.5% vs. 23.0%, p < 0.001) compared to HCV-negative patients. TriNetX data analysis revealed that HCV-positive patients had a 53% lower risk of developing melanoma (RR = 0.470, 95% CI: 0.443–0.498, p < 0.001). However, HCV-positive melanoma patients had higher all-cause mortality (HR = 1.360, 95% CI: 1.189–1.556, p < 0.001). An in silico analysis identified key molecular players, including IL-6 and CTLA4, in the HCV-melanoma network. Conclusions: While HCV infection may be associated with a lower risk of melanoma development, HCV-positive patients who develop melanoma have poorer outcomes. Abstract

Aflatoxicosis is one of the major issues for the public health. Control of aflatoxins is necessary to avoid the health hazards in human and animals. Present study was designed to evaluate the efficacy of Bacillus licheniformis as a novel feed additive for control of aflatoxicosis. A total of 40 rats were equally divided into the four groups. The first group (BD) was kept as control, treated with basal diet. In second group (AF), the rats were treated with aflatoxins (28 μg/kg BW). In the third group (AFBL) the rats were treated with aflatoxins (28 µg/g BW) and Bacillus licheniformis (1 × 108 cfu/mL). In the fourth group (BDBL) the rats were treated with only Bacillus licheniformis (1 × 108 cfu/mL). The parameters studied were, phagocytic response by carbon clearance assay system, lymphoproliferative response to PHA-P, antibody response to SRBCs, total antioxidant capacity and total oxidant status of the rats and interleukin concentrations, along with histopathology of organs. Results of this study confirmed the immunomodulation and antioxidant capacity of Bacillus licheniformis against 28 µg/kg BW of aflatoxins. [ABSTRACT FROM AUTHOR]

Abstract

Hepatitis B virus (HBV) DNA diagnosis plays a critical role in the transmission of HBV infections. Accurate protocols in HBV detection will reduce the spread of the virus from blood donors. Therefore, this research compared the HBV detecting potentials of three different HBV protocols in the virus detection. Blood from 200 donors at the University college blood bank Ibadan were sampled and tested for HBV using the ABON™ HBsAg Hepatitis B Surface Antigen Tests (strip test), Monolisa TM HBsAg ULTRA (ELISA test) and Huruian biology HBV detection kit (qPCR test). HBV positive samples were genotyped by sequencing to ascertain genotype identity. Results showed that of the 200 samples tested, both ABON™ HBsAg Hepatitis B Surface Antigen Tests and Monolisa TM HBsAg ULTRA (for ELISA) did not record any positive sample. However, Huruian biology HBV detection kit via qPCR recorded a total of twenty-three positive samples. Of the twenty-three samples, twenty were of genotype E while three were of genotype B. The results from this study showed that that qPCR is more accurate in HBV virus detection than ELISA technique and also HBV genotype E is the most common genotype isolated in Nigeria.

Breast cancer has been acknowledged as one of the most notorious cancers, responsible for millions of deaths around the globe. Understanding the various factors, genetic mutations, comprehensive pathways, etc., that are involved in the development of breast cancer and how these affect the development of the disease is very important for improving and revitalizing the treatment of this global health issue. The forkhead-box gene family, comprising 19 subfamilies, is known to have a significant impact on the growth and progression of this cancer. This study aimed to investigate the possible anti-protozoal effects of Syzygium aromaticum (clove) oil, Anethum graveolens (dill) seeds oil, Lactobacillus acidophilus LB, and zinc against Cryptosporidium parvum in comparison to NTZ.Methods: Besides the negative control, mice from six experimental groups (T1–T6) were infected with Cryptosporidium parvum oocysts. On the seventh day post-infection (PID), mice from five groups were treated for 8 consecutive days with NTZ, clove oil, dill seed oil, Lactobacillus acidophilus LB, and zinc commercial forms (T2–T5). Oocysts shedding rate, differences of mice body weight, serum IL10, and TNF-α, cryptosporidial antigen, and cd3 at the intestinal mucosa were evaluated at the end of the experiment.Results: The mean of the C. parvum oocysts’ shedding rate was significantly lower in all treated groups than in the non-treated group. The oocysts reduction rate was the highest in zinc-treated mice (98.3%), Lactobacillus acidophilus LB and dill-treated groups (95.77%), and the NTZ-treated group (91.55%). Clove oil was the least effective, with a 74.65% reduction rate. Excluding the clove oil-treated group, immunohistochemical analysis revealed the clearance of the Cryptosporidium antigen in the intestinal tissue in all treated groups.Conclusion: The study has provided a rational basis for using these safe, cheap, and commercially available alternatives in treating cryptosporidiosis combined with NTZ.Keywords: Cryptosporidium parvum, Syzygium aromaticum, Anethum graveolens seeds oil, Lactobacillus acidophilus LB, zinc, nitazoxanide

Abstract

Methicillin-resistant staphylococcal infections are a serious public health concern worldwide, especially in our region, due to the unavailability of accurate therapeutics. The aim of this study was to determine the resistance pattern of Staphylococcus aureusisolated from clinical and asymptomatic specimens and to identify antimicrobial agents against methicillin resistant S. aureusstrains. 47 samples (pus, urine, blood, nose and milk) together from diverse healthcare hospitals in Faisalabad for location of S. strains. Aureus by biochemical examination. The results of these biochemical tests showed that 21 of 47 samples were positive for S. aureus. A resistance study found that 10 of 21 S. aureus isolates were resistant to oxacillin. Twenty-five medicinal plants were collected from different parts of Saudi Arabia, air-dried and extracted with methanol. These plant extracts were then screened for potential against oxacillin-resistant S. aureus. In addition, the semi-maximum inhibitory concentration (IC50) of effective medicinal plant extracts was measured using the micro-dilution method.Methanol extracts from five plants (Centratherum antherminticum, Eucalyptus globulus, Lawsonia inermis, Punica granatumand Rubia cordifolia) have been shown to have anti-bacterial activity against clinically isolated S. oxacillinresistant strains. It was done. The IC50 for these methanol extracts of S. aureusranged from 0.250 to 4.30 mg/mL. Our results suggest that various medicinal plants contain potential anti-bacterial agents against S. aureus, which may contribute to the development of more effective agents against multidrug resistant S. aureusinfections. It might help.

33. Differential effects of methanolic extracts of clove, ginger, garlic and eucalyptus essential oils on anti-parasitic partitivities of G. lambliaand E. histolytica: an in vitro study

Abstract

Plants and essential oils are used in traditional medicine against parasitic intestinal protozoal diseases. The current study revealed the GC–MS analysis and anti-parasitic effects of four medicinal plant species from the Saudi Arabia. All essential oils showed significant variations in chemical constituents. 15 different chemical constituents were present in Eucalyptus camaldulensis, Syzygium aromaticum, Zingiber officinale,except Allium sativumthat showed the presence of diverse 21 chemical compounds. Significant anti-parasitic inhibition was observed against G. lambliaand E. histolyticawhen treated with E. camaldulensis(100%, 100%), S. aromaticum(74.81%, 86.4%), Z. officinale(77.62%, 100%) and A. sativum(98.79%, 74.84%) at a dose rate of 100 µg/mL to control G. lambliaand E. histolyticaparasites, respectively. Low to high levels of inhibition percentages were noticed when concentration of essential oils was increased from 25 to 100 µg/mL. The extracts of essential oils from all plants revealed the presence of mono-terpenes, alkenes, aldehydes, hexanes, sesquiterpenes, alkanes, propenes, phenyls, esters, alcohols, and hydrocarbons. All extracted natural extracts showed cytotoxicity test values over 50% which were considered cytotoxic during their interaction with parasitic cells. Results of current experiment can be lead further for future in vivo trials.
