In a study to domesticate the species Calotropis procera, for wool production in drylands, investigations were done to determine the effects of seed transfer distance on the growth of the species in a typical farm setting. The objectives of the research were to determine the growth of three provenances of C. procera and to determine the influence of seed transfer distance on the growth parameters. Diameter and height data were used to generate horizontal and vertical growth curves respectively and further subjected to Duncan Multiple Range Test (DMRT) to isolate existence of significant differences across the three provenances. Crop landraces have unique local agroecological and societal functions and offer important genetic resources for plant breeding. Recognition of the value of landrace diversity and concern about its erosion on farms have led to sustained efforts to establish ex situ collections worldwide. The degree to which these efforts have succeeded in conserving landraces has not been comprehensively assessed. Here we modelled the potential distributions of eco-geographically distinguishable groups of landraces of 25 cereal, pulse and starchy root/tuber/fruit crops within their geographic regions of diversity. We then analysed the extent to which these landrace groups are represented in genebank collections, using geographic and ecological coverage metrics as a proxy for genetic diversity. We find that ex situ conservation of landrace groups is currently moderately comprehensive on average, with substantial variation among crops; a mean of 63% ± 12.6% of distributions is currently represented in genebanks. Breadfruit, bananas and plantains, lentils, common beans, chickpeas, barley and bread wheat landrace groups are among the most fully represented, whereas the largest conservation gaps persist for pearl millet, yams, finger millet, groundnut, potatoes and peas. Geographic regions prioritized for further collection of landrace groups for ex situ conservation include South Asia, the Mediterranean and West Asia, Mesoamerica, sub-Saharan Africa, the Andean mountains of South America and Central to East Asia. The plant Chasmanthera dependens (Hochst) have been used to manage many diseases including diabetes mellitus without scientific evaluation of its efficacy and safety. The main objective of the project was to determine the antidiabetic activity of Chasmanthera dependens plant in a rat model. Experimental design was used where the stem of the plant was collected, dried, crashed into fine powder, extracted with distilled water at 60°C and lyophilized using a Freeze Dryer, packaged in air tight container and stored at -20°C ready for use. The extract was orally and intraperitoneally screened in alloxan induced diabetic mice for its hypoglycemic activity at doses of 25, 48.4, 93.5, 180.9 and 350 mg per kg body weight. Diabetes in mice was induced using 186.9 mg/kg of alloxan monohydrate. Negative controls included normal and diabetic rats orally and intraperitoneally administered with physiological saline while positive controls included diabetic rats administered with glibenclamide as oral and insulin as intraperitoneal reference drug. The results revealed hypoglycemic activity in Chasmanthera dependens at the five different doses when given either orally or intraperitoneally. In conclusion, the medicinal plant at the various doses demonstrated significant (ρ ≤ 0.05) hypoglycemic activity. The author’s recommendation is continued use of the plant extracts at low therapeutic doses. Consideration should be made to carry out the same studies using higher animals including man.

The World Agroforestry (ICRAF) in Kenya plays a key role in conserving tree genetic diversity, thereby contributing to the delivery of ecosystem services in tree-based production systems. This study explored the benefits of using the two most popular fodder tree species among smallholder farmers, sourced from the ICRAF genebank: Calliandra calothyrsus (Calliandra) and Gliricidia sepium (Gliricidia). Through a survey of key informants and genebank users, we examined the benefits derived from the adoption of Calliandra and Gliricidia and the unique role of the ICRAF genebank as the main source of tree germplasm for Kenyan smallholders. The constraints to germplasm access could limit protein fodder supply and the intensity of fertilization in farmers’ fields, which in turn could affect productivity in livestock and maize sectors in Kenya. We find that improved food security, higher incomes, increased milk production, reduced vulnerability to drought, reduced soil erosion, and enhanced soil fertility are identified as the main farmer-perceived benefits linked to the use of Calliandra and Gliricidia. The findings demonstrate the importance of agroforestry in the delivery of ecosystem services, in the light of climate change and heightened pressure for sustainable agricultural practices, and the crucial role of the genebank in conserving and distributing unique, high quality tree germplasm.

Genebanks underpin global food security, conserving and distributing agrobiodiversity for use in research and breeding. The CGIAR collections include >700,000 seed accessions, held in trust as global public goods. However, the role of genebanks in contributing to global food security can only be realized if collections are effectively managed. Examination of the historical viability monitoring data from seven CGIAR genebanks confirmed that high seed viability was maintained for many decades for the various crops and forage species. However, departures from optimum management procedures were revealed, and there were insufficient data gathered to derive reliable estimates of longevity needed to better forecast regeneration requirements, estimate the size of seed lots that should be stored, and optimize accession monitoring intervals.

Societal Impact Statement The increasing occurrence of native and non‐native pests and diseases of trees in Africa is impacting rural livelihoods, economic development, and biodiversity across the continent. Here we provide an updated and comprehensive overview of insect pests and pathogens targeting natural and planted forests in Africa, and raise awareness of this growing emergency. Data availability and limited biosecurity of some countries are limiting the ability to tackle this invasion. An improved knowledge of taxonomy, distribution, and damage caused by these organisms will be crucial for developing continent‐wide strategies to manage this emergency and enhance country‐level intervention capacity. Summary An increasing multitude of insect pests and pathogens is targeting indigenous trees of natural forests, agroforestry systems, and exotic trees in planted forests in Africa. This is raising major concerns for a continent already challenged by adaptations to climate change, as it threatens a vital resource for food security of rural communities, economic growth, and ecosystem conservation. The accidental introduction through trade of non‐native species in particular is accelerating, and it adds to the damage to tree‐based landscapes by native pests and diseases. Old‐time and new invaders heavily impact planted forests of exotic eucalypts, pines, and acacias, and are spreading quickly across African regions. But many non‐native pathogens are recently found affecting important indigenous trees. We describe the threat to African trees by providing an overview of highly relevant insect pests and diseases of indigenous and exotic trees in Africa, and discuss implications for management and future research. The implementation of an integrated and globally coordinated approach based on improved biosecurity, biological control, and tree resistance would contribute to mitigate the potentially devastating impact of these invasions on African natural resources.

Baobab (Adansonia digitata L., Malvaceae) seed oil contains Fatty Acids (FAs) with commercial and cosmeceutical value. The influence of provenance and geographical region on FAs profile and content of seed oil from baobab wild populations in sub-Saharan Africa (SSA) was assessed in this study. The FA profiles were determined as fatty acid methyl esters (FAME) by Gas Chromatography (GC). Generally, the seed oil contained 17–22% saturated fatty acids (SFA), 32–38% monounsaturated fatty acids (MUFA) and 22–26% polyunsaturated fatty acids (PUFA). The African Orphan Crops Consortium (AOCC) successfully initiated the ambitious genome sequencing project of 101 African orphan crops/trees with 6 genomes sequenced, 6 near completion, and 20 currently in progress. Addressing stunting, malnutrition, and hidden hunger through nutritious, economic, and resilient agri-food system is one of the major agricultural challenges of this century. As sub-Saharan Africa harbors a large portion of the severely malnourished population, the African Orphan Crops Consortium (AOCC) was established in 2011 with an aim to reduce stunting and malnutrition by providing nutritional security through improving locally adapted nutritious, but neglected, under-researched or orphan African food crops. Foods from these indigenous or naturalized crops and trees are rich in minerals, vitamins, and antioxidant, and are an integral part of the dietary portfolio and cultural, social, and economic milieu of African farmers. Through stakeholder consultations supported by the African Union, 101 African orphan and under-researched crop species were prioritized to mainstream into African agri-food systems. The AOCC, through a network of international-regional-public-private partnerships and collaborations, is generating genomic resources of three types, i.e., reference genome sequence, transcriptome sequence, and re-sequencing 100 accessions/species, using next-generation sequencing (NGS) technology. Furthermore, the University of California Davis African Plant Breeding Academy under the AOCC banner is training 150 lead African scientists to breed high yielding, nutritious, and climate-resilient (biotic and abiotic stress tolerant) crop varieties that meet African farmer and consumer needs. To date, one or more forms of sequence data have been produced for 60 crops. Reference genome sequences for six species have already been published, 6 are almost near completion, and 19 are in progress.

The nutrient-rich tubers of the greater yam Dioscorea alata L. provide food and income security for millions of people around the world. Despite its global importance, however, greater yam remains an “orphan crop.” Here we address this resource gap by presenting a highly-contiguous chromosome-scale genome assembly of greater yam combined with a dense genetic map derived from African breeding populations. The genome sequence reveals an ancient lineage-specific genome duplication, followed by extensive genome-wide reorganization. Using our new genomic tools we find quantitative trait loci for susceptibility to anthracnose, a damaging fungal pathogen of yam, and several tuber quality traits. Genomic analysis of breeding lines reveals both extensive inbreeding as well as regions of extensive heterozygosity that may represent interspecific introgression during domestication. These tools and insights will enable yam breeders to unlock the potential of this staple crop and take full advantage of its adaptability to varied environments.

The inherent ability of seeds (orthodox, intermediate, and recalcitrant seeds and vegetative propagules) to serve as carriers of pests and pathogens (hereafter referred to as pests) and the risk of transboundary spread along with the seed movement present a high-risk factor for international germplasm distribution activities. Quarantine and phytosanitary procedures have been established by many countries around the world to minimize seed-borne pest spread by screening export and import consignments of germplasm. The effectiveness of these time-consuming and cost-intensive procedures depends on the knowledge of pest distribution, availability of diagnostic tools for seed health testing, qualified operators, procedures for inspection, and seed phytosanitation. This review describes a unique multidisciplinary approach used by the CGIAR Germplasm Health Units (GHUs) in ensuring phytosanitary protection for the safe conservation and global movement of germplasm from the 11 CGIAR genebanks and breeding programs that acquire and distribute germplasm to and from all parts of the world for agricultural research and food security. We also present the challenges, lessons learned, and recommendations stemming from the experience of GHUs, which collaborate with the national quarantine systems to export and distribute about 100,000 germplasm samples annually to partners located in about 90 to 100 countries. Furthermore, we describe how GHUs adjust their procedures to stay in alignment with evolving phytosanitary regulations and pest risk scenarios. In conclusion, we state the benefits of globally coordinated phytosanitary networks for the prevention of the intercontinental spread of pests that are transmissible through plant propagation materials.

A systematic approach to tree planting and management globally is hindered by the limited synthesis of information sources on tree uses and species priorities. To help address this, the authors ‘mined’ information from 23 online global and regional databases to assemble a list of the most frequent tree species deemed useful for planting according to database mentions, with a focus on tropical regions. Using a simple vote count approach for ranking species, we obtained a shortlist of 100 trees mentioned in at least 10 of our data sources (the ‘top-100’ species). A longer list of 830 trees that were mentioned at least five times was also compiled. Our ‘top-100’ list indicated that the family Fabaceae (syn. Leguminosae) was most common. The information associated with our mined data sources indicated that the ‘top-100’ list consisted of a complementary group of species of differing uses. These included the following: for wood (mostly for timber) and fuel production, human nutrition, animal fodder supply, and environmental service provision (varied services). Of these uses, wood was most frequently specified, with fuel and food use also highly important. Many of the ‘top-100’ species were assigned multiple uses. The majority of the ‘top-100’ species had weediness characteristics according to ‘attribute’ invasiveness databases that were also reviewed, thereby demonstrating potential environmental concerns associated with tree planting that need to be balanced against environmental and livelihood benefits. Less than half of the ‘top-100’ species were included in the OECD Scheme for the Certification of Forest Reproductive Material, thus supporting a view that lack of germplasm access is a common concern for trees. A comparison of the ‘top-100’ species with regionally-defined tree inventories indicated their diverse continental origins, as would be anticipated from a global analysis. However, compared to baseline expectations, some geographic regions were better represented than others. Our analysis assists in priority-setting for research and serves as a guide to practical tree planting initiatives. We stress that this ‘top-100’ list does not necessarily represent tree priorities for the future, but provides a starting point for also addressing representation gaps. Indeed, our primary concern going forward is with the latter.

The international collections of plant genetic resources for food and agriculture (PGRFA) hosted by 11 CGIAR Centers are important components of the United Nations Food and Agriculture Organization&rsquo, s global system of conservation and use of PGRFA. They also play an important supportive role in realizing Target 2.5 of the Sustainable Development Goals. This paper analyzes CGIAR genebanks&rsquo, trends in acquiring and distributing PGRFA over the last 35 years, with a particular focus on the last decade. The paper highlights a number of factors influencing the Centers&rsquo, acquisition of new PGRFA to include in the international collections, including increased capacity to analyze gaps in those collections and precisely target new collecting missions, availability of financial resources, and the state of international and national access and benefit-sharing laws and phytosanitary regulations. Factors contributing to Centers&rsquo, distributions of PGRFA included the extent of accession-level information, users&rsquo, capacity to identify the materials they want, and policies. The genebanks&rsquo, rates of both acquisition and distribution increased over the last decade. The paper ends on a cautionary note concerning the potential of unresolved tensions regarding access and benefit sharing and digital genomic sequence information to undermine international cooperation to conserve and use PGRFA.

The African Orphan Crops Consortium promotes the strategic, genome-enabled improvement of under-researched crops for biodiversity-based, nutritious food solutions in Africa. The African Plant Breeding Academy empowers the continent’s plant breeders to apply advanced genetic approaches and shared genetic solutions to the task of tailoring the immense diversity of underutilized crops to the needs of Africa’s producers, processors and consumers.

Parasitic weeds pose a severe problem in agricultural production, causing massive crop losses in many regions of the world and especially Africa. One example to be considered the most significant biological constraint to food production in sub-Saharan Africa (SSA) is Striga hermonthica, the most widespread among the Striga species in the semi-arid tropical African zones. The wide geographical distribution set conditions for genetically structured populations. The genetic variations among the weed populations allow for the quick breakdown of resistance in crops hence making control of the weed difficult. Efficient and effective control of S. hermonthica demands knowledge on inherent genetic variability within local and regional races of the weed. However, the genetic diversity and virulence of S. hermonthica ecotypes in Kenya and Uganda on selected sorghum varieties remain unknown. This study aimed at evaluating the genetic diversity among seven S. hermonthica populations from locations in Kenya and Uganda using 5 primer sets of Expressed Sequence Tags-Simple Sequence Repeats (EST-SSR). The genetic diversity was moderate as shown by the Nei’s genetic distance values. AMOVA measured low genetic differentiation among the populations. This study also investigated twelve sorghum varieties for their response to S. hermonthica infection. The result demonstrated that the varieties resistance responses to S. hermonthica varied widely. The phenotype of resistant interaction was characterized by the inability of the weeds haustoria to penetrate the sorghums root endodermis due to severe necrosis and in rare cases the parasites radicle growing away from the host root. The resistant sorghum varieties were the Asareca W2, Asareca AG3, N13 and the Wild-type which had low mean number of S. hermonthica plantlets growing on their roots, while the most susceptible varieties were Sap 027, Epurpur which had the highest mean number of S. hermonthica plantlets growing on their roots. There was a highly significant difference in the means of the number of Striga growing on the roots of sorghum varieties, Striga dry biomass and S. hermonthica length between the susceptible and resistant ones. This knowledge holds unique potentiality since resistant sorghum germplasm tested will be sourced and targeted to the seven specific geographical areas where virulence of the particular S. hermonthica populations was characterized.

Allanblackia parviflora A. Chev. is an indigenous tree species which is found in West African rainforest zones. It is an underutilized fruit tree species that has been targeted for improvement as part of efforts to domesticate high-value indigenous multi-purpose trees for fruit and seed production in Africa. Allanblackia has several benefits, such as providing shade, timber, and medicine; however, the production of edible oil from its seeds is the economically most important use. There is evidence that the Allanblackia seed oil, which is used for cooking, the production of margarine and the manufacturing of ointments and soap, is being developed as a new agri-business in Ghana, Nigeria, Cameroon, and Tanzania. Despite the nutritional and socio-economic importance of A. parviflora, it is still at the early stages of its domestication process. Even though several researchers have explored the biology of this species, there is very limited scientific information available on its morphological and genetic diversity and silvicultural management in West Africa. Therefore, this systematic review presents an up-to-date overview on the uses, seed chemical composition, and morphological and genetic diversity of this fruit tree species, and proposes a way forward for future research towards improvement and domestication.

Baobab (Adansonia digitata L.) is an indigenous fruit tree associated with the Savannah drylands of sub‐Saharan Africa. Local communities mainly utilize the leaves, pulp, and seeds of baobab as a source of food and for income generation. Despite the highly economic value of tea in Africa, its genetic and geographic origins remain largely unexplored. Here we address this by collecting 439 samples across 11 countries in Africa and Asia to investigate the origin and genepool composition of African tea based on 23 nuclear microsatellites loci (nSSRs) and three cpDNA intergenic spacer regions. Our results indicated that the African tea represents a potpourri originating from multiple introductions over time. The nSSR analysis revealed that the majority (79%) of tea accessions collected in Africa belong to Indian Assam tea which have likely originated from India and/or Sri Lanka. The patterns of nSSR variation also showed that Chinese Assam tea is genetically distinct from Indian Assam tea, and has rarely been used in African tea breeding efforts since only 4% of the African tea accessions possessed this genotype. We found a total of 22 cpDNA haplotypes, which grouped into three main geographic clades that were concordant with the distribution of microsatellite genotypes. Several private cpDNA haplotypes were identified in Chinese Assam tea in Southern Yunnan province of China. Therefore Chinese Assam tea will be important for the enrichment of African tea gene pools. Our results is a useful guide in future tea breeding programmes in Africa.

Wagio RS, Runo S, Muchugi A. 2019. Genetic diversity of Solanum nigrum cultivated in Kenya. Asian J Agric 2: 63-71. Solanum nigrum L., sometimes known as black nightshade, is a worldwide weed found on arable land, in gardens, and garbage dumps. It has been utilized in the pharmaceutical industry to produce analgesics, ointments, and vasodilators. Using morphological and Simple Sequence Repeats (SSR) markers, this study aimed to assess the genetic diversity of S. nigrum populations growing in specific regions of Kenya. One hundres-twenty (120) samples from four populations were evaluated in total. Morphological characterization utilized four aerial characters. leaf surface (smooth and hairy), leaf margin (whole and sinuous), leaf form (ovate, lanceolate, and diamond), fruit color (red, purple small, and purple large). Tukey's HSD test demonstrated substantial variation among the evaluated attributes (P?0.05). Four SSR primers produced 63 polymorphic bands, ranging from 14 to 17 per primer set. The range of Polymorphic Information Content was 0.1429 to 0.1855, with a mean value of 0.1704. Heterozygosity (using SSR markers) averaged out to be He=0.1370. Axis 1 and 2 of the PCoA explained 44.65% of the population distribution variance. The analysis of multi-level variance (AMOVA) showed that there was only 14% variation between populations and 86% variation inside the populations. Nei's genetic distance varied between 0.010 (Kipkaren and Matanya) and 0.020 (Makuyu and Mauche). Using DARwin 6.0.5 and Unweighted Neighbor Joining with 1000 bootstraps, a dissimilarity study was conducted. The absence of morphological and genetic distinctions in the dendrogram is indicative of the absence of genetic divergence between groups. Variance in the Agro-Ecological Zone is not indicative of variation in the S. nigrum cultivars. Comparing morphological and molecular data revealed leaf surface and fruit color grouping with genetic data. Using the same data, however, there was no grouping between leaf shape and leaf margin. The observed physical distinctions are primarily the effect of human selection. This study demonstrates that the S. nigrum complex displays limited genetic diversity but considerable morphological heterogeneity. Low genetic distance and the absence of clustering patterns indicate that S. nigrum is a closely related group of plants. The results of this study will have far-reaching implications for future breeding efforts of the Solanum complex, as they will be utilized to produce markers connected to crucial agronomic features. The findings will contribute further to resolving the taxonomic ambiguity surrounding S. nigrum.

Two of the most economically important plants in the Artocarpus genus are jackfruit (A. heterophyllus Lam.) and breadfruit (A. altilis (Parkinson) Fosberg). Both species are long-lived trees that have been cultivated for thousands of years in their native regions. Today they are grown throughout tropical to subtropical areas as an important source of starch and other valuable nutrients. There are hundreds of breadfruit varieties that are native to Oceania, of which the most commonly distributed types are seedless triploids. Jackfruit is likely native to the Western Ghats of India and produces one of the largest tree-borne fruit structures (reaching up to 45 kg). To-date, there is limited genomic information for these two economically important species. Here, we generated 273 Gb and 227 Gb of raw data from jackfruit and breadfruit, respectively. The high-quality reads from jackfruit were assembled into 162,440 scaffolds totaling 982 Mb with 35,858 genes. Similarly, the breadfruit reads were assembled into 180,971 scaffolds totaling 833 Mb with 34,010 genes. A total of 2822 and 2034 expanded gene families were found in jackfruit and breadfruit, respectively, enriched in pathways including starch and sucrose metabolism, photosynthesis, and others. The copy number of several starch synthesis-related genes were found to be increased in jackfruit and breadfruit compared to closely-related species, and the tissue-specific expression might imply their sugar-rich and starch-rich characteristics. Overall, the publication of high-quality genomes for jackfruit and breadfruit provides information about their specific composition and the underlying genes involved in sugar and starch metabolism.

BackgroundThe African eggplant (Solanum aethiopicum) is a nutritious traditional vegetable used in many African countries, including Uganda and Nigeria. It is thought to have been domesticated in Africa from its wild relative, Solanum anguivi. S. aethiopicum has been routinely used as a source of disease resistance genes for several Solanaceae crops, including Solanum melongena. A lack of genomic resources has meant that breeding of S. aethiopicum has lagged behind other vegetable crops.ResultsWe assembled a 1.02-Gb draft genome of S. aethiopicum, which contained predominantly repetitive sequences (78.9%). We annotated 37,681 gene models, including 34,906 protein-coding genes. Expansion of disease resistance genes was observed via 2 rounds of amplification of long terminal repeat retrotransposons, which may have occurred ∼1.25 and 3.5 million years ago, respectively. By resequencing 65 S. aethiopicum and S. anguivi genotypes, 18,614,838 single-nucleotide polymorphisms were identified, of which 34,171 were located within disease resistance genes. Analysis of domestication and demographic history revealed active selection for genes involved in drought tolerance in both “Gilo” and “Shum” groups. A pan-genome of S. aethiopicum was assembled, containing 51,351 protein-coding genes; 7,069 of these genes were missing from the reference genome.ConclusionsThe genome sequence of S. aethiopicum enhances our understanding of its biotic and abiotic resistance. The single-nucleotide polymorphisms identified are immediately available for use by breeders. The information provided here will accelerate selection and breeding of the African eggplant, as well as other crops within the Solanaceae family.

Warburgia ugandensis Sprague (Canellaceae) occurs in East and Central Africa and is an important multipurpose tree species. Over-exploitation of natural forests for medicinal purposes and clearance for farming threaten the species survival. Cultivation of the tree species would ensure sustainable medicinal source and its conservation. However, on-farm genetic diversity of the species is currently unknown. The genetic diversity of the on-farm W. ugandensis populations and their proximate natural populations were analyzed using the amplified fragment length polymorphism (AFLP). Four primer combinations produced a total of 223 polymorphic bands. Both the natural and on-farm populations had high genetic diversity ranging from H = 0.2892 to H = 0.1278. Principal co-ordinates analysis and dendrogram separated the ten populations into two major groups corresponding to Kenyan and Tanzanian populations, respectively. Ugandan populations were shared between the two major groups; this is probably because Uganda is believed to be the centre of diversity for W. ugandensis. Close genetic relationships between the on-farm and their proximate natural population were revealed. Analysis of molecular variance (AMOVA) revealed that a total of 54% AFLP variation resided within populations with 46% reside among populations. The high genetic diversity of W. ugandensis on-farm populations could be useful in germplasm collection and conservation strategies.
