The primary objective of this study was to assess the effectiveness of cold plasma therapy in managing subclinical mastitis in cows caused by Streptococcus agalactiae, Streptococcus uberis and Escherichia coli. After detection of mastitis pathogens, 38 cows were selected for cold plasma therapy for five days. On the fifth day of treatment, the mastitis agents were re-examined and no causative agents were identified. An additional evaluation conducted 28 days later confirmed the absence of mastitis. Cow productivity, milk composition and quality indicators were assessed at the beginning of the experiment and 32 days from the start (28 days after treatment cessation). After the mastitis treatment, the somatic cell count decreased significantly by between 2.89 and 7.09 times, and the milk yield of the cows at the end of the experiment increased from 0.63 kg per day to 2.82 kg per day (P < 0.01). These results highlight the potential of this innovative approach for managing a prevalent disease that causes substantial losses in the dairy industry. Furthermore, they lay the groundwork for expanded research involving larger sample sizes.

Deformed wing virus (DWV) is known as one of the main viruses that affect honeybees’ health all around the world. The virus has two widespread genotypes, DWV-A and DWV-B (VDV-1), transmitted mainly by V. destructor mites. In this study, we collected honeycombs with covered broods from 73 apiaries in eight Lithuanian regions and initially investigated the prevalence of V. destructor mites. Mites were collected from May to the end of July in 2021 from 124 hives. The prevalence of V. destructor infestations in beehives reached 30% and 63% in investigated apiaries. The presence of DWV-A and DWV-B pathogens in mites and broods was examined by RT-qPCR targeting the CRPV-capsid region. The molecular characterization of the virus in mite samples was based on sequence analysis of the RNA-dependent RNA polymerase (RdRp) region. In addition, leader polypeptide (LP), structural protein (Vp3), Helicase, and RdRp genes were used for phylogenetic characterization of dual infection. The prevalences of DWV-B in mites and broods were 56.5% and 31.5%, respectively, while DWV-A was detected in 12.9% of mite samples and 24.7% of brood samples. Some of the examined mite samples harboured dual virus infections. Our findings showed that bee colonies from the same apiary were not always infected by the same viruses. Some bee colonies were virus-free, while others were highly infected. Phylogenetic analysis of 21 sequences demonstrated the presence of highly variable DWV-B and DWV-A genotypes in Lithuania and possible recombinant variants of the virus. This study represents the first molecular characterization of mite-borne pathogens hosted by honeybees (Apis mellifera) in Lithuania.

This study aimed to assess the relationship between the length of the dry period (DL) and the risks of dystocia and stillbirths in Holstein cows. A total of 1072 healthy cows in lactations 1 and ≥2 were categorized based on the DL (mean: 58.07 ± 0.33 days) into three groups: short DL (70 days; 26.9%). Dystocia occurred in 12.4% of cows, while stillbirths accounted for 4.1% of calves. The medium DL group had the highest number of unassisted calvings. The dystocia rates were 11.4% for cows with gestation (GA) ≤ 274 days; 6.0% for cows with GA = 275–282 days; and 19.9% for cows with GA ≥ 283 days (p < 0.001). In season 1 (November–April), dystocia occurred in 15.0% of cases compared to 8.8% in season 2 (May–October) (p < 0.001). The stillbirth rates ranged from 3.6% to 4.0% for cows with medium and long DLs; while those with a short DL had 5.8% (p < 0.05). The stillbirth rates were 5.2% for cows with GA ≤ 274 days; 3.2% for cows with GA = 275–282 days; and 5.1% for cows with GA ≥ 283 days (p < 0.001). Season 1 had a stillbirth rate of 4.8%, while season 2 had a stillbirth rate of 3.3% (p < 0.001). This research provides insights that could assist the dairy industry in making informed decisions to reduce the incidence rates of dystocia and stillbirths in cows.

Babesia vesperuginis is an intraerythrocytic protozoan parasite that circulates among bats and ticks in many countries worldwide. However, the distribution of B. vesperuginis in the Baltic region has not been studied. A total of 86 dead bats from eight different species were collected and screened for Babesia spp. using real-time PCR. Overall, 52.3% (45/86) of the bats were found positive for Babesia spp. The prevalence of Babesia spp. in different organs varied, with the highest prevalence observed in heart tissues (37.0%) and the lowest in liver tissues (22.2%). However, the observed differences in prevalence among organs were not statistically significant. Blood samples from 125 bats of nine different species were also analyzed for Babesia spp. prevalence using real-time PCR and nested PCR. The results showed a prevalence of 35.2% and 22.4%, respectively. Moreover, 28.3% (17/60) of the examined blood samples were confirmed positive for Babesia spp. through blood smear analysis. The total of 32 partial sequences of the 18S rRNA gene derived in this study were 100% identical to B. vesperuginis sequences from GenBank. In eight species of bats, Pipistrellus nathusii, Pipistrellus pipistrellus, Pipistrellus pygmaeus, Vespertilio murinus, Eptesicus nilssonii, Eptesicus serotinus, Myotis daubentonii and Nyctalus noctula, Babesia parasites were identified. In E. nilssonii, Babesia spp. was identified for the first time.

The increasing population of European bison (Bison bonasus) can contribute to the prevalence of zoonotic pathogens. The aim of the present study was to assess the presence of A. phagocytophilum infection in European bison tissues as well as ticks removed from European bison in Lithuania and Poland. A further objective of this work was to compare the detected A. phagocytophilum strains. A total of 85 tissue samples (spleen) of European bison and 560 ticks belonging to two species, Ixodes ricinus (n = 408) and Dermacentor reticulatus (n = 152) were tested. DNA of A. phagocytophilum was detected based on RT-PCR in 40% of the European bison samples, 8.8% of the I. ricinus and 5.9% of the D. reticulatus ticks. Analysis of the obtained partial 16S rRNA gene sequences of A. phagocytophilum revealed the presence of three variants with two polymorphic sites. Furthermore, phylogenetic analysis with partial msp4 gene sequences grouped A. phagocytophilum variants into three clusters. This study revealed that the groEL gene sequences of A. phagocytophilum from European bison and their ticks grouped into ecotype I and only one sequence from Lithuanian European bison belonged to ecotype II. The results of the present study indicated that European bison may play a role as a natural reservoir of A. phagocytophilum.

The use of organic compounds in different spheres of human activity is accompanied by their influx to and accumulation in the environment. The negative impact of those compounds can be one of the reasons for a decline in populations and biodiversity of aboveground invertebrates. Chemical compounds can potentially cause a variety of effects (attractant or repellent) on insects, including species of the Staphylinidae family. In a laboratory experiment, we identified repellent and attractant influence of 40 organic compounds and mixtures of compounds (acids, alcohols, ketones, phenols, aldehydes, aromatic carbohydrates solvents, and vehicle fuels) on Philonthus decorus Gravenhorst, 1802. The ambulatory responses of the males and females to the same chemical compounds most often varied. A strong repellent activity against both sexes of Ph. decorus was caused by oleic acid, while hexane repelled the males. Acetic acid, 1-butanol, and ammonia solution were found to be strongly repellent against females. A moderate (average) repellent activity towards male Ph. decorus was displayed by organic solvents and fuels, some alcohols (isopropanol, isoamyl alcohol, methanol, ethanol), acids (acetic, formic acid), aromatic carbohydrates (toluene, xylene), and formaldehyde. Female Ph. decorus in general were less sensitive to the odors. The list of repellents with moderate activity against the females was much shorter: solvent 646, white spirit, toluene, isopropanol, isoamyl alcohol, citric and oxalic acids, and glycerol. Moderate attractant activity for Ph. decorus was exhibited by some amino acids, alcohols, and fuel mixes: glycine and L-cysteine (for the males), and phenylalanine, methanol, and diesel fuel (for the females). The rest of the 40 chemical compounds we studied caused no ambulatory responses in Ph. decorus. The difficulties we encountered in the interpretation of the results suggest a need for further experimental studies that would expand the knowledge of the chemoecology of insects.

Wild rodents are considered to be one of the most important TBEV-amplifying reservoir hosts; therefore, they may be suitable for foci detection studies. To investigate the effectiveness of viral RNA detection in wild rodents for suspected TBEV foci confirmation, we trapped small rodents (n = 139) in various locations in Lithuania where TBEV was previously detected in questing ticks. Murine neuroblastoma Neuro-2a cells were inoculated with each rodent sample to maximize the chances of detecting viral RNA in rodent samples. TBEV RNA was detected in 74.8% (CI 95% 66.7–81.1) of the brain and/or internal organ mix suspensions, and the prevalence rate increased significantly following sample cultivation in Neuro-2a cells. Moreover, a strong correlation (r = 0.88; p < 0.05) was found between the average monthly air temperature of rodent trapping and the TBEV RNA prevalence rate in cell culture isolates of rodent suspensions, which were PCR-negative before cultivation in cell culture. This study shows that wild rodents are suitable sentinel animals to confirm TBEV foci. In addition, the study results demonstrate that sample cultivation in cell culture is a highly efficient method for increasing TBEV viral load to detectable quantities.

Bat fleas (Insecta: Siphonaptera: Ischnopsyllidae) are highly host-specific ectoparasites distributed worldwide (except Antarctica). The identification of flea species and the detection of flea-borne pathogens plays a significant role not only in veterinary and public health, but also in providing insights into the ecology, behaviour, and geographical distribution of fleas and flea-associated pathogens. The present study aimed to conduct a morphological and molecular identification of fleas collected from Lithuanian bats, as well as to screen for the presence of vector-borne pathogens. A total of 58 flea specimens were collected from bats in Lithuania. Based on morphological analysis, seven flea species were identified (Ischnopsyllus variabilis, I. octactenus, I. simplex, I. elongatus, I. hexactenus, Nycteridopsylla pentactena, and N. eusarca) and screened for the presence of Anaplasma spp., Borrelia spp., Babesia spp., Bartonella spp., Rickettsia spp., and Mycoplasma spp. Molecular analysis of fleas’ COI and 18S rRNA genes showed that the flea species collected from bats are closely genetically related. Bartonella DNA was detected in bat fleas (41.4%). No DNA of Anaplasma, Borrelia, Babesia, Rickettsia, and Mycoplasma was amplified in the tested samples. The Wolbachia sp. was accidentally found in I. elongatus. The phylogenetic analysis of four Bartonella genes (16S-23S rRNA ITS, gltA, rpoB, and groEL) revealed specificity to bats or bats’ ectoparasites. This is the first report about bat flea species and the pathogens in bat fleas in Lithuania. New host records for bat fleas were also reported. This study extends the knowledge about bat fleas and their role as vectors of different pathogens.

Mycoplasma spp. pathogens frequently cause chronic and acute diseases in cats. The aim of the present study was to investigate the presence and genetic diversity of Mycoplasma spp. in cats and their ectoparasites using PCR and sequence analysis of the 16S rRNA gene. Blood samples were collected from 541 domestic and stray cats in Lithuania. Ectoparasites (153 fleas and 321 ticks) were collected from owned domestic cats that live both outdoors and indoors. Mycoplasma spp. were detected in 7.2% of cat blood samples and 4.4% of Ctenocephalides felis fleas. The sequence analysis revealed the presence of Mycoplasma haemofelis in 1.1% of cats and ‘Candidatus Mycoplasma haematominutum’ in 4.8% of cats. Ct. felis fleas harboured M. haemofelis. To the best of the authors’ knowledge, this is the first report on the prevalence and molecular characterisation of Mycoplasma bacteria in cats in Lithuania and cat fleas in the Baltic States.

The present study evaluated genetic diversity of Lithuanian populations of Lythrum salicaria in relation to parameters of riparian environment. Growing along Nemunas, Seaside and Lielupė river basins, 15 populations were examined using amplified fragment length polymorphism markers. Molecular data were related to the river basins, type of land use and cover, natural vice versa regulated fragments of the rivers. Population mean genetic diversity parameters were as follows: percentage of polymorphic loci (57.2), expected heterozygosity (0.183), polymorphismc information content (0.218). Mantel test revealed correlation (R2 = 0.0986, p = 0.01) between genetic and geographic distance of populations. Greater genetic diversity within, rather than among populations (ΦPT = 0.213) was observed. According to the Bayesian clustering, studied populations are admixtures of two gene pools. Analysis of molecular variance revealed significant differentiation between populations belonging to distinct river basins, between populations from natural vs. regulated fragments of the rivers.

Paenibacillus larvae bacterium is known to be the causative agent of American foulbrood (AFB), a widespread, highly contagious and fatal disease in honey bees (Apis mellifera). There are four genotypes of Paenibacillus larvae that are named after their enterobacterial repetitive consensus (ERIC), and a fifth ERIC genotype has recently been found. In this study, a total of 108 independent P. larvae isolates from different geographical regions in Lithuania collected between 2011 and 2021 were investigated by molecular methods. The aims of this study were to detect which enterobacterial repetitive intergenic consensus (ERIC) genotype is the most common in Lithuania apiaries, identify and differentiate subtypes of the defined genotype by using multiple-locus variable number of tandem-repeat analysis (MLVA), and review how bacterial molecular diversity has changed over time in different parts of Lithuania. The obtained molecular analysis results showed that 100% of P. larvae bacterial isolates from Lithuania belong to the ERIC I genotype and can be differentiated to nine different subtypes by using the MLVA and capillary electrophoresis methods.

The family Laelapidae (Dermanyssoidea) is morphologically and ecologically the most diverse group of Mesostigmata mites. Although molecular genetic data are widely used in taxonomic identification and phylogenetic analysis, most classifications in Mesostigmata mites are based solely on morphological characteristics. In the present study, eight species of mites from the Laelapidae (Dermanyssoidea) family collected from different species of small rodents in Lithuania, Norway, Slovakia, and the Czech Republic were molecularly characterized using the nuclear (28S ribosomal RNA) and mitochondrial (cytochrome oxidase subunit I gene) markers. Obtained molecular data from 113 specimens of mites were used to discriminate between species and investigate the phylogenetic relationships and genetic diversity among Laelapidae mites from six genera. This study provides new molecular data on Laelaps agilis, Laelaps hilaris, Laelaps jettmari, Haemogamasus nidi, Eulaelaps stabularis, Hyperlaelaps microti, Myonyssus gigas, and Hirstionyssus sp. mites collected from different rodent hosts and geographical regions in Europe.

Abstract To provide the most comprehensive picture of species phylogeny and phylogeography of European roe deer (Capreolus capreolus), we analyzed mtDNA control region (610 bp) of 1469 samples of roe deer from Central and Eastern Europe and included into the analyses additional 1541 mtDNA sequences from GenBank from other regions of the continent. We detected two mtDNA lineages of the species: European and Siberian (an introgression of C. pygargus mtDNA into C. capreolus). The Siberian lineage was most frequent in the eastern part of the continent and declined toward Central Europe. The European lineage contained three clades (Central, Eastern, and Western) composed of several haplogroups, many of which were separated in space. The Western clade appeared to have a discontinuous range from Portugal to Russia. Most of the haplogroups in the Central and the Eastern clades were under expansion during the Weichselian glacial period before the Last Glacial Maximum (LGM), while the expansion time of the Western clade overlapped with the Eemian interglacial. The high genetic diversity of extant roe deer is the result of their survival during the LGM probably in a large, contiguous range spanning from the Iberian Peninsula to the Caucasus Mts and in two northern refugia.

Data on alien species show that plant invasions are caused by a complex combination of characteristics of invasive species (invasiveness) and characteristics of invaded environment (invasibility). Impatiens parviflora is one of Europe’s top invasive species. The present study aimed to evaluate molecular diversity of populations of highly invasive in Lithuania I. parviflora by applying several DNA markers and relating genetic parameters to abiotic and biotic environment. For sampling, urban forests, riparian forests, and agrarian shrublands were selected. Three different DNA-based techniques, Simple Sequence Repeats (SSR), Inter Simple Sequence Repeat (ISSR) markers, and Random Amplified Polymorphic DNA (RAPD), were used for detecting genetic variation between 21 populations. All population individuals were monomorphic and homozygotic for four loci and heterozygotic for one locus by SSR analysis. Hierarchical analyses of molecular variance at ISSR and RAPD loci revealed significant differentiation of populations depending on geographic zones of the country. Bayesian Structure analyses of molecular data demonstrated existence of many genetic clusters and this finding is in support to multiple introduction of the species. The polymorphism extent at ISSR loci was positively correlated with the total coverage of herbaceous plant species. The coverage by I. parviflora was negatively correlated with the total number of herbaceous species and light in the sites. Our results indicate that Lithuanian sites with I. parviflora might be distinguished by high soil nutrient levels. According to the principal component analysis, the coverage by I. parviflora was a more important variable of populations compared to molecular data or parameters of abiotic environment. In conclusion, complexity of invasibility and invasiveness factors determine the variability of I. parviflora sites, including genetic traits, coverage of invasive species and conditions of environment that were significant and interrelated.

The European bison is a species well known for its low genetic variability due to historical bottleneck incidents. Restoration of the European bison population in Lithuania began at the end of the 20th century, and the Lithuanian population was re-established with ten individuals from the Prioksko-Terrasny reserve in Russia and one free-roaming individual captured in Lithuania. This research focused on the genetic diversity and genetic structure of European bison in Lithuania based on microsatellite markers and mtDNA sequences. The results of the microsatellite analysis revealed that the Lithuanian population (n = 34) has a different genetic structure to individuals in Poland (n = 16), Germany (n = 10), and Sweden (n = 6). The analysis of the mtDNA sequences showed a low level of variation, with two haplotypes observed in the Lithuanian bison population. H1 was more frequent and was also found in Poland, Romania, and Russia. The results of this investigation show that individuals should be continuously genetically monitored, registering their migrations and relocations for the successful management of the species in Lithuania.

Early identification of lameness at all phases of lactation improves milk yield and reduces the incidence of mastitis in the herd. According to the literature we hypothesized that there are associations of electrical conductivity variables of milk flow with lameness in dairy cows. The aim of this study was to determine if blood cortisol and electrical conductivity in the milk flow phases correlate with each other and whether they are related to cow lameness. On one farm, out of 1500 cows, 64 cows with signs of lameness and 56 healthy cows were selected with an average of 2.8 lactations and 60 days in the postpartum period. A local veterinarian who specializes in hoof care treatments identified and scored lameness. During evening milking, the milk flow of all 120 cows was measured using electronic milk flow meters (Lactocorder®, WMB AG, Balgache, Switzerland). Before each milking, two electronic mobile milk flow meters (Lactocorders) were mounted between the milking apparatus and the milking tube to take measurements. We found that the average cortisol concentration in the blood of the studied cows was significantly correlated with the laminitis score. Bartonella bacteria infect the erythrocytes and endothelial cells of mammalians. The spread of the Bartonella infection occurs mainly via bloodsucking arthropod vectors. Studies on Bartonella infection in European bison, the largest wild ruminant in Europe, are lacking. They are needed to clarify their role in the maintenance and transmission of Bartonella spp. The aim of this study was to investigate the presence of the Bartonella pathogen in European bison and their ticks in Lithuania. A total of 38 spleen samples from bison and 258 ticks belonging to the Ixodes ricinus and Dermacentor reticulatus species were examined. The bison and tick samples were subjected to ssrA, 16S–23S rRNA ITS, gltA, and rpoB partial gene fragment amplification using various variants of PCR. Bartonella DNA was detected in 7.9% of the tissue samples of European bison. All tick samples were negative for Bartonella spp. The phylogenetic analysis of 16S–23S rRNA ITS, gltA, and rpoB partial gene fragment revealed that European bison were infected by B. bovis (2.6%) and B. schoenbuchensis (5.3%). This is the first report addressing the occurrence of B. bovis and B. schoenbuchensis in European bison in Europe.

Abstract Background In Lithuania, the first case of canine subcutaneous dirofilariosis was recorded in 2010. Since then, an increasing number of cases of canine dirofilariosis have been documented in different veterinary clinics throughout the country. Human dirofilariosis was diagnosed in Lithuania for the first time in September 2011. However, to the authors’ knowledge, there are no published data on the presence and prevalence of autochthonous dirofilariosis in dogs and humans in the country. The present study provides information about the predominant species and prevalence of Dirofilaria in dogs and describes the cases of human dirofilariosis in Lithuania. It also outlines PCR detection of the bacterial endosymbiont Wolbachia that contributes to the inflammatory features of filarioid infection. Results A total of 2280 blood samples and six adult worms from pet and shelter dogs were collected in the central and eastern regions of Lithuania in 2013–2015. Based on their morphological appearance, morphometric measurements and molecular analysis, all the adult nematodes were identified as Dirofilaria repens. The diagnosis of microfilariae in blood samples was based on blood smear analysis and Knott’s test. The PCR and sequence analysis of the ribosomal DNA ITS2 region and cox1 gene confirmed the presence of D. repens. Overall, 61 (2.7%) of the 2280 blood samples were found to be positive for the presence of D. repens. The infection rate of D. repens was significantly higher in shelter dogs (19.0%; 19/100) than in pet dogs (1.9%; 42/2180) (χ 2 = 100.039, df = 1, P

Anaplasma phagocytophilum and Babesia spp. are causative agents of tick-borne infections that are increasingly considered as a threat to animal and public health. To assess the role of cervids in the maintenance of zoonotic pathogens in Norway, we investigated the prevalence of A. phagocytophilum and Babesia spp. in free-ranging roe deer and red deer. Initial screening of spleen samples of 104 animals by multiplex real-time PCR targeting the major surface protein (msp2) gene and 18S rRNA revealed the presence of A. phagocytophilum infection in 81.1% red deer (Cervus elaphus) and 88.1% roe deer (Capreolus capreolus), and Babesia spp. parasites in 64.9% red deer and 83.6% roe deer, respectively. Co-infections were found in 62.2% red deer and 79.9% roe deer. Nested PCR and sequence analysis of partial msp4 and 18S rRNA genes were performed for molecular characterization of A. phagocytophilum strains and Babesia species. A total of eleven A. phagocytophilum msp4 gene sequence variants were identified: five different variants were 100% identical to corresponding A. phagocytophilum sequences deposited in the GenBank database, while other six sequence variants had unique nucleotide polymorphisms. Sequence analysis of the 18S rRNA gene demonstrated the presence of multiple Babesia species, including Babesia capreoli, Babesia divergens, Babesia venatorum and Babesia odocoilei/Babesia cf. odocoilei. This study is the first report demonstrating the prevalence and molecular characterization of A. phagocytophilum strains and Babesia species in roe deer and red deer in Norway. The high infection and co-infection rates with A. phagocytophilum and Babesia spp. in red deer and roe deer suggest that these cervids may play an important role in the transmission of single and multiple pathogens. The aims of our study were to evaluate the associations between calving difficulty and lameness and their effects on milk yield and quality traits. A total of 4723 calving cases were evaluated for calving difficulty using a 4-point scoring system. Lameness was diagnosed with a visual locomotion score system from 1 to 30 days after calving in 333 fresh dairy cows. Cows were divided into non-lame cows and lame cows. Milk quality traits were registered using Lely Astronaut® A3 milking robots. The normal distribution of all indicators was assessed using the Shapiro–Wilk normality test. Normally distributed milk indicators were expressed as mean ± standard error of the mean. Differences between the mean values of their groups were determined using the Fisher’s least significant difference test. We categorized cows by health status, i.e., lame (LA) and non-lame (HL) cows, and according to calving difficulty (CD) (on a 4-point scale: 1—no problem, 2—slight problem, 3—problems requiring assistance, 4—considerable force and extreme difficulty). In the present study, calving difficulty increased the risk of lameness in cows by 2.09-fold (95% CI = 1.644–2.650, p < 0.001). It was found that the mean standard milk yield in fresh dairy cows with calving difficulty was lower (−6.14 kg, p < 0.001) than in the group where no assistance was required at calving. Similarly, herd affected milk fat (%) and the calving process—herd and the interaction between calving difficulty and herd—and lameness impacted the quantity of milk protein and lactose in cows. We found that severe lameness (3–4 points) (3.88–5.92% of cows) became more prevalent in those cows that had dystocia than those that did not (0.27–2.37% of cows).

Canine babesiosis is an emerging and rapidly expanding tick-borne disease in central and northeast Europe. In the last two decades, the endemic area of Babesia canis has expanded from central Europe to the Baltic region. This study aimed to investigate the genetic diversity of B. canis strains isolated from naturally infected dogs in different regions of Lithuania using PCR-RFLP and sequence analyses based on a partial region of 18S rRNA and Bc28.1 genes. Blood samples from 149 dogs suspected of having babesiosis were collected in Lithuania during 2016–2017. Based on PCR-RFLP profiles and two nucleotide substitutions observed in 18S rRNA gene sequences, three B. canis genotypes were identified in Lithuania—18S rRNA-A, 18S rRNA-B and 18S rRNA-A/B—with the A/B genotype predominating (83.9%). Based on the obtained PCR-RFLP profiles of the Bc28.1 gene, four B. canis genotypes were identified: Bc28.1-B (53.8%), Bc28.1-34 (20.8%), Bc28.1-A (17.9%), and Bc28.1-34/A or B (7.5%). Sequence analysis of the partial Bc28.1 gene revealed eighteen polymorphic sites and thirteen sequence variants among the Lithuanian samples. The B. canis genotypes obtained were detected with varying prevalences in different regions of Lithuania.

We hypothesized that subclinical mastitis detected during the last gestation period can increase the risk of stillbirth in dairy calves. The aim was to investigate the relation of subclinical mastitis detected during the last gestation period and its pathogens with the stillbirth of calves. Cows from the 210th day of pregnancy were selected for the study. They were divided into two groups: the first group—subclinical mastitis was confirmed on the farm by the California mastitis test (CMT); the second group of cows—mastitis was not confirmed by the CMT test. Groups of cows were compared according to the results of their calving—the number of stillborn calves. A stillborn calf was defined as a calf that dies at birth or within the first 24 h after calving, following a gestation period of 260 days. Our results suggest that decreasing the incidence of subclinical mastitis during the last gestation period (from the 210th day of pregnancy) can decrease the risk of stillbirth in dairy calves. Further, it is important to identify the pathogen because the highest risk of stillbirth was found in cows with mastitis caused by Escherichia coli, Staphylococcus aureus, Streptococcus agalactiae, pathogenic Staphylococci and other Streptococci. Cows at the first calving had a 1.38–1.65-times higher risk of having stillborn calves than cows of parity ≥ 2. From a practical point, veterinarians and farmers can consider the effect of subclinical mastitis during late gestation on the risk of stillbirth and it could help for strategies of optimizing reproductive performance in dairy cows.

There is now considerable evidence that in Europe, babesiosis is an emerging infectious disease, with some of the causative species spreading as a consequence of the increasing range of their tick vector hosts. In this review, we summarize both the historic records and recent findings on the occurrence and incidence of babesiosis in 20 European countries located in southeastern Europe (Bosnia and Herzegovina, Croatia, and Serbia), central Europe (Austria, the Czech Republic, Germany, Hungary, Luxembourg, Poland, Slovakia, Slovenia, and Switzerland), and northern and northeastern Europe (Lithuania, Latvia, Estonia, Iceland, Denmark, Finland, Sweden, and Norway), identified in humans and selected species of domesticated animals (cats, dogs, horses, and cattle). Recorded cases of human babesiosis are still rare, but their number is expected to rise in the coming years. This is because of the widespread and longer seasonal activity of Ixodes ricinus as a result of climate change and because of the more extensive use of better molecular diagnostic methods. Bovine babesiosis has a re-emerging potential because of the likely loss of herd immunity, while canine babesiosis is rapidly expanding in central and northeastern Europe, its occurrence correlating with the rapid, successful expansion of the ornate dog tick (Dermacentor reticulatus) populations in Europe. Taken together, our analysis of the available reports shows clear evidence of an increasing annual incidence of babesiosis across Europe in both humans and animals that is changing in line with similar increases in the incidence of other tick-borne diseases. This situation is of major concern, and we recommend more extensive and frequent, standardized monitoring using a “One Health” approach.

Abstract Background Rickettsiae are emerging pathogens causing public health problems in many countries around the world. Rickettsia spp. are found in association with a wide range of arthropods which feed on different species of animals. However, the distribution and natural cycle of Rickettsia species and their association with different arthropod vectors are not fully established. The aim of this study was to investigate the presence and prevalence of Rickettsia spp. in ticks, mites and fleas parasitizing different species of small mammals in Lithuania and to molecularly characterize the Rickettsia spp. obtained from different ectoparasites. Results A total of 1261 ectoparasites (596 Ixodes ricinus ticks, 550 mites of five species and 115 fleas of eight species) collected from 238 rodents in Lithuania during 2013–2014 were investigated for the presence of Rickettsia pathogens. Infection rates were calculated as the maximum likelihood estimation (MLE) with 95% confidence intervals (CI). The infection rate varied among ectoparasites and was found highest in fleas 43.5%, followed by I. ricinus ticks (MLE = 26.5%; 95% CI: 22.2–31.3%) and then mites (MLE = 9.3%; 95% CI: 7.0–12.2%). Sequence analysis of partial gltA and 17kDa genes revealed the presence of Rickettsia helvetica, R. felis, R. monacensis, Rickettsia sp. and rickettsial endosymbionts. Four Rickettsia spp. were identified in fleas, while three Rickettsia spp. were identified in Laelapidae mites and only one (R. helvetica) in I. ricinus ticks. Conclusions To our knowledge, this is the first report of the occurrence and molecular characterization of Rickettsia spp. in 11 species of ectoparasites of small rodents in Lithuania. The present data extend the knowledge on the distribution of Rickettsia spp. and their association with different arthropod vectors. Prior to our study, R. felis had never been identified in Lithuania. To our knowledge, this is also the first report of R. felis in L. agilis and H. microti mites and in Ct. agyrtes and H. talpae fleas, as well as the first detection of R. monacensis in Ct. agyrtes fleas.

The aim of this research was to identify the relationship of calvin ease and level of in-line milk urea (MU) and other milk components, namely milk yield (MY), electrical conductivity (EC), milk fat (MF), milk protein (MP), milk fat/protein ratio (MF/MP), and somatic cell count (SCC) in dairy cows. The cows for the research were selected following such criteria: cows were tested within the period of up to the first 30 days after calving and had had a range of lactation numbers from two to four. Each selected farm housed more than 500 dairy cows and a total of 4712 calving cases from the eight dairy farms were studied and evaluated. The 4-point scale was used for the evaluation of the calving according to the point value meanings where 1 = easy, unassisted; 2 = easy, assisted; 3 = difficult, assisted; 4 = difficult, requiring veterinary intervention. A total of 4712 calving cases were researched and scored. The chemical properties of milk in all research cows were analyzed during the early phase of lactation (from the onset of calving to 30 days past calving) every day, during each milking. Cows were classified into groups according to the level of urea in milk: Group 1 had MU ≤ 15 mg/dL (12.6% of cows), Group 2 had MU 16–30 mg/dL (62.4% of cows), and Group 3 had MU > 30 mg/dL (25.0% of cows). We found that cows with milk urea levels between 16 and 30 mg/dL had the lowest incidence of dystocia, and also the highest concentration of milk lactose (ML ≥ 4.6%), the lowest mean value of milk electrical conductivity, and the lowest value of milk SCC. Dystocia increased the risk of somatic cell growth in cow’s milk above the herd average (OR = 1.364; 95% CI = 1.184–1.571, p < 0.001), and normal urea in milk reduced this risk (OR = 0.749; 95% CI = 0.642–0.869, p = 0.05). In all groups of cows, according to the level of urea in milk, the productivity of cows without dystocia at calving was higher (2.50–5.51 kg) as well as the milk protein % (0.13–0.21%) and milk lactose % (0.07–0.19%). We concluded that, in all groups of cows, according to the level of urea in milk, the productivity and milk lactose concentration of cows without dystocia at calving was higher, and lower somatic cells count and electrical conductivity values were found in the milk compared with cows diagnosed with dystocia at calving. We can state that dystocia has a negative effect on milk urea concentration and can increase the risk of mastitis.

Bartonella are vector-borne parasitic bacteria that cause zoonotic infections in humans. One of the most common infections is cat-scratch disease caused by Bartonella henselae and Bartonella clarridgeiae. Cats are the major reservoir for these two species of bacteria, while cat fleas are vectors for the transmission of infection agents among cats. The aim of the present study was to investigate the presence of Bartonella infections in stray and pet cats and in cat fleas in Lithuania. Blood samples were taken from 163 cats presented in pet clinics and animal shelters. A total of 102 fleas representing two species, Ctenocephalides felis and Ctenocephalides canis, were collected from 12 owned cats that live both outdoors and indoors. Bartonella DNA in samples was detected using a nested PCR targeting the 16S–23S rRNA intergenic spacer (ITS) region. Bartonella DNA was detected in 4.9% (8/163) of the cats and 29.4% (30/102) of the fleas. Sequence analysis of the ITS region showed that the cats and fleas were infected with B. henselae, B. clarridgeiae and Bartonella sp., closely related to B. schoenbuchensis. This study is the first report on the prevalence and molecular characterization of Bartonella spp. in cats and cat fleas in Lithuania.

A reliable surveillance strategy of tick-borne encephalitis virus (TBEV) is necessary to ensure adequate disease control measures. However, current approaches assessing geographical TBEV circulation are ineffective or have significant limitations. In this study we investigated a total of 1363 goat and 312 sheep bulk tank milk samples for the presence of TBEV. Samples were collected from systematically selected farms in Lithuania every 4–5 days from April to November in 2018 and 2019. To validate results, we additionally tested 2685 questing ticks collected in the vicinity of milk collection sites. We found 4.25% (95% CI 3.25–5.47) and 4.48% (95% CI 2.47–7.41) goat and sheep milk samples to be positive for TBEV, respectively. Furthermore, geographical distribution of TBEV in milk samples coincided with the known TBE endemic zone and was correlated with incidence of TBE in humans in 2019. When sampling time coincides, TBEV detection in milk samples is as good a method as via flagged ticks, however bulk milk samples can be easier to obtain more frequently and regularly than tick samples. The minimal infectious rate (MIR) in ticks was 0.34% (CI 95% 0.15–0.64). Therefore, our results confirm that testing milk serves as a valuable tool to investigate the spatial distribution of TBEV at higher resolution and lower cost.

Abstract Background According to targeted treatment (TT), the whole flock is dewormed based on knowledge of the risk, or parameters that quantify the mean level of infection, whereas according to targeted selective treatment (TST), only individual animals within the grazing group are treated, based on parasitological, production and/or morbidity parameters. The aim of this study was to compare two different treatment protocols on sheep farms in Lithuania. The study was conducted from 15 April to 31 October 2014 on three sheep farms. On the TT (the whole flock) and T(S)T (with FECs ≥ 300, respectively) farms all adult animals were treated orally with fenbendazole irrespective of EPG counts before the grazing season. The second treatment was applied with injectable ivermectin on both farms. However, on the TT farm all sheep were also treated on 2nd August regardless of their EPG counts, while on the T(S)T farm only those animals with an EPG ≥ 300 were treated on 1 July using a threshold of ≥ 300 EPG. No treatments were administered on the control farm (n = 1) during the study. Results Spring treatment of ewes significantly reduced nematode faecal egg counts (FEC) both on the TT and T(S)T farms, with the benefit of lowering pasture contamination with infective L3 stage larvae at the start of grazing season, while it remained significantly higher on the control farm. The positive effect of the spring treatment of ewes was reflected by increased body weight gains (BWG) in lambs in the first half of the grazing season. Following the second treatment, the weight gains in lambs on the T(S)T farm were higher compared to lambs on the TT farm, while BWG in the control lambs started to decrease. The difference was also substantiated by the body condition scores (BCS) and dag scores (DS) of lambs, which were highest on the T(S)T farm compared with those on the control and TT farms. Conclusions The results of this study show that both treatment strategies were useful in reducing clinical effects (BCS and DS) of gastrointestinal nematode parasitism and increasing the performance in lambs. Furthermore, on the T(S)T farm some of animals were left in refugia, helping to slow down the development of anthelmintic resistance (AR) in future.

Babesia microti (Aconoidasida: Piroplasmida) (Franca, 1910) is an emerging tick-borne parasite with rodents serving as the considered reservoir host. However, the distribution of B. microti in Europe is insufficiently characterized. Based on the sample of 1180 rodents from 19 study sites in Lithuania, the objectives of this study were: (1) to investigate the presence of Babesia parasites in eight species of rodents, (2) to determine the prevalence of Babesia parasites in rodents from different habitats, and (3) to characterize the detected Babesia strains using partial sequencing of the 18S rRNR gene. Babesia DNA was detected in 2.8% rodents. The highest prevalence of Babesia was found in Microtus oeconomus (14.5%) and Microtus agrestis (7.1%) followed by Clethrionomys glareolus (2.3%), Apodemus flavicollis (2.2%) and Micromys minutus (1.3%). In M.minutus, Babesia was identified for the first time. The prevalence of Babesia-infected rodents was higher in the meadow (5.67%) than in the ecotone (1.69%) and forest (0.31%) habitats. The sequence analysis of the partial 18S rRNA gene reveals that Babesia isolates derived from rodents were 99–100% identical to human pathogenic B. microti ‘Jena/Germany’ strain.

Bovine subclinical mastitis can cause great harm to dairy herds because of its negative impact on milk production and quality and cow health. Improved diagnostic tools are needed to maximise the control of subclinical mastitis distribution and ensure the high quality of milk as an industrial product. Between 2015 and 2020, seventy-two dairy herds were screened for bovine subclinical mastitis causative agents to identify the relationship between seasons, lactose levels and subclinical mastitis infection. The predominant species found in the milk samples were mixed microbiota, coagulase-negative Staphylococcus and Staphylococcus aureus. Yeasts were found exclusively in autumn, while Enterococcus spp. and Escherichia coli were only found in summer and autumn. A negative correlation was detected between milk lactose and number of somatic cells in milk (−0.471; p < 0.001). The lactose levels in milk were closely associated with the prevalence (%) of subclinical mastitis pathogens, such as Streptococcus agalactiae (y = −1.8011x + 10.867, R2 = 0.9298), Staph. aureus (y = −3.5216x + 25.957, R2 = 0.8604) and other Streptococci (y = −0.5956x + 7.6179, R2 = 0.6656). These findings suggest that milk lactose may be used as a biomarker of suspected udder inflammation in modern health prevention programmes for cows to reduce the prevalence of subclinical mastitis pathogens in dairy cattle herds.

The aim of this study was to assess the relationship between temperament and milk performance in cows at different stages of lactation, describing their productivity, metabolic status and resistance to mastitis. This study showed that with increasing lactation, cows’ temperament indicators decreased (p < 0.001) and they became calmer. The highest temperament score on a five-point scale was found in cows between 45 and 100 days of lactation. In the group of pregnant cows, we found more cows (p = 0.005) with a temperament score of 1–2 compared with non-pregnant cows A normal temperament was usually detected in cows with lactose levels in milk of 4.60% or more and when the somatic cell count (SCC) values in cow milk were p < 0.05), this study showed a negative genetic correlation between the temperament of cows and milk yield (p < 0.001). Positive genetic correlations between temperament scores and milk somatic cells (p < 0.001) and milk fat-to-protein ratio (p < 0.05) were found to indicate a lower genetic predisposition in cows with a calmer temperament to subclinical mastitis and ketosis. On the other hand, the heritability of temperament (h2 = 0.044–0.100) showed that only a small part of the phenotypic changes in this indicator is associated with genetic factors.

Background and objectives: Lyme disease is the most common tick-borne infectious disease in Europe, caused by the spirocheta bacteria of Borrelia burgdorferi. Several genospecies of B. burgdorferi are pathogenic to humans. B. burgdorferi sensu stricto, which is prevalent in North America, causes reactive arthritis, whereas B. garinii and B. afzelii, common in Europe, can affect the skin, heart, or nervous system; it has been shown that the clinical symptoms of the disease may be very different. The objective of this study was to identify the baseline characteristics of Lyme disease and to elucidate the frequency of different Lyme disease syndromes in Lithuania. Materials and Methods: Patients who were diagnosed with Lyme disease during an ambulatory visit to the Center of Infectious Diseases, Vilnius University Santaros clinics, from 2014 to 2016, were enrolled in this study. A retrospective material analysis was conducted. Results: In total, 1005 patients were enrolled with the following prevalence of clinical syndromes: erythema migrans (EM), 945 (94.02%); Lyme arthritis, 32 (3.18%); neuroborreliosis, 23 (2.28%); Lyme carditis, 4 (0.39%); and acrodermatitis, 1 (0.09%). Erythema migrans was dominant among middle-aged women, with a rash appearing mainly on the lower extremities. Lyme arthritis mainly manifested among middle-aged women as an oligoarthritis, mostly affecting the knee joint. Neuroborreliosis was seen more often in middle-aged women than men and the main symptom was nervus facialis neuropathy. Lyme carditis, manifested as an atrioventricular block, with a male/female ratio of 3:1, and the median age was 51. Acrodermatitis was diagnosed in a 61-year-old woman, as a painful, red rash on the hand. Conclusions: According to the prevalence of B. garinii and B. afzelii in Europe, previously it was thought that Lyme disease presented as erythema migrans, and less frequently as neuroborreliosis; however, this study revealed that other syndromes may also be seen. In addition, we revealed that the longer it takes for erythema migrans to appear, the greater the likelihood of Lyme arthritis developing.

Background and objective: Lyme disease, also known as Lyme borreliosis (LB), is a tick-borne infectious disease caused by the spirochete bacteria Borrelia. The risk of infection depends on the geographical area, ecological factors, and human behavior. Clinical manifestations of Lyme borreliosis have a wide range, but the most frequent clinical symptom, which is also a diagnostic symptom, is a skin rash called erythema migrans (EM). The disease is very common worldwide. In Lithuania, the disease frequency is 99.9 cases per 100,000 population (Centre for Communicable Diseases and AIDS, Lithuania, 2017). The main aim of this study was to obtain the baseline characteristics of the disease regarding the infected Lithuanian population. Materials and Methods: We analyzed data from the Centre for Communicable Diseases and AIDS about all Lyme disease (A69.2) diagnosed patients over a three-year period (from 2014 to 2016) in Lithuania. Results: In 2014−2016, 7424 (crude incidence rate 85.4) cases with LB were diagnosed in Lithuania. Most of them (4633 (62.4%)) were identified in women. Older people were more likely to suffer from LB. Urban residents were 2.6 times more often affected that those living in villages. Tick bites were primarily observed in high season months, from May to September (90%), with the highest peak in July. There was a higher number of observed tick bites (p = 0.003) in the urban residents. Erythema migrans occurred in 75.6% LB cases, while other symptoms did not exceed a quarter of all LB cases. There were 7353 (99.6%) cases where LB was confirmed via clinical symptoms and/or laboratory tests. Also, 1720 (23.2%) patients were tested for LB immunoglobulins. Conclusions: This study found a high incidence of Lyme disease in Lithuania. We elucidated the baseline characteristics regarding the infected Lithuanian population which may ease medical clinicians’ work on new Lyme diagnoses.

European brown hare, Lepus europaeus, from Central and Eastern European countries (Hungary, Poland, Serbia, Lithuania, Romania, Georgia and Italy) were sampled, and phylogenetic analyses were carried out on two datasets: 1.) 137 sequences (358 bp) of control region mtDNA; and 2.) 105 sequences of a concatenated fragment (916 bp), including the cytochrome b, tRNA-Thr, tRNA-Pro and control region mitochondrial DNA. Our sequences were aligned with additional brown hare sequences from GenBank. A total of 52 and 51 haplotypes were detected within the two datasets, respectively, and assigned to two previously described major lineages: Anatolian/Middle Eastern (AME) and European (EUR). Furthermore, the European lineage was divided into two subclades including South Eastern European (SEE) and Central European (CE). Sympatric distribution of the lineages of the brown hare in South-Eastern and Eastern Europe revealed contact zones there. BAPS analysis assigned sequences from L. europaeus to five genetic clusters, whereas CE individuals were assigned to only one cluster, and AME and SEE sequences were each assigned to two clusters. Our findings uncover numerous novel haplotypes of Anatolian/Middle Eastern brown hare outside their main range, as evidence for the combined influence of Late Pleistocene climatic fluctuations and anthropogenic activities in shaping the phylogeographic structure of the species. Our results support the hypothesis of a postglacial brown hare expansion from Anatolia and the Balkan Peninsula to Central and Eastern Europe, and suggest some slight introgression of individual haplotypes from L. timidus to L. europaeus.

The wild-harvested fruit of Vaccinium oxycoccos (European cranberry) is used medicinally in many European and North American countries; the plant, however, is seldom cultivated. In order to optimize the collection strategy and improve the horticulturally important characters of V. oxycoccos clones, comprehensive investigations of the species are necessary. In the present study we investigated the phenological, morphological and genetic diversity of 29 clones originating from two wild populations growing in two strictly protected Lithuanian reserves, Čepkeliai and Žuvintas. During an ex situ collection at Kaunas Botanical Garden, we observed great phenological variation between the collected V. oxycoccos clones. The following morphological traits most clearly distinguished our study clones: leaf size, berry shape, berry size and fruit colour at full maturity. The genetic variation of V. oxycoccos clones from the two populations was assessed using RAPD and SSR. RAPD analysis conducted with 9 primers resulted in 146 polymorphic loci for the total sample, and SSR analysis with 5 primers revealed 29 alleles for the total sample. A greater degree of polymorphism was demonstrated for the Čepkeliai population than for the Žuvintas population. The study allowed the selection of several clones having promising morphological traits for further testing in the field.
