19. Anticandidal potentiality of biosynthesized and decorated nanometals with fucoidan

Abstract

Candida genus includes many hazardous and risky species that can develop resistance toward various antifungal types. Metals nanoparticles (NPs) possess powerful antimicrobial actions, but their potential human toxicity could limit their practices. The algal polysaccharide fucoidan (Fu) was extracted from the macro-brown algae, Cystoseira barbata, analyzed, and used for biosynthesizing nanoparticles of silver (Ag-NPs) and selenium (Se-NPs). The extracted Fu had elevated fucose levels (58.73% of total monosaccharides) and exhibited the main biochemical characteristic of customary Fu. The Fu biosynthesis of Ag-NPs and Se-NPs was achieved via facile direct protocol; Fu-synthesized NPs had 12.86 and 16.18 nm average diameters, respectively. The ultrastructure of Fu-synthesized NPs emphasized well-distributed and spherical particles that were embedded/capped in Fu as combined clusters. The Fu/Ag-NPs and Fu/Se-NPs anticandidal assessments, against Candida albicans, Candida glabrata, and Candida parapsilosis, revealed that both NPs had powerful fungicidal actions against the examined pathogens. The ultrastructure imaging of subjected C. albicans and C. parapsilosis to NPs revealed that Fu/Ag-NPs and Fu/Se-NPs triggered remarkable distortions, pore formation, and destructive lysis in cell surfaces within 10 h of exposure. The innovative usage of C. barbata Fu for Ag-NP and Se-NP synthesis and the application of their composites as powerful anticandidal agents, with minimized human toxicity, are concluded.

Abstract

A new and innovative rolled graphene oxide (roll‐GO)/poly‐m‐methylaniline (PmMA) core–shell nanocomposite has been successfully synthesized using an in situ polymerization technique. This eco‐friendly and cost‐effective material shows great promise due to its antimicrobial properties. The characterization of the nanocomposite involved X‐ray diffraction and Fourier transform infrared spectroscopy to analyze its structure and functional groups, whereas scanning electron microscopy and transmission electron microscopy (TEM) were utilized to examine its morphology. TEM analysis revealed the formation of roll‐GO, forming multi‐walled tubes with inner and outer diameters of 50 and 70 nm, respectively. Optical analysis demonstrated an enhanced bandgap in the nanocomposite, with bandgap values of 2.38 eV for PmMA, 2.67 eV for roll‐GO, and 1.65 eV for roll‐GO/PmMA. The antibacterial efficacy of the nanocomposite was tested against Gram‐positive bacteria, including Bacillus subtilis and Staphylococcus aureus, as well as Gram‐negative bacteria such as Escherichia coli and Salmonella sp. The well diffusion method was used to determine the inhibition zones, revealing that the nanocomposite demonstrated broad‐spectrum antibacterial activity against all the pathogens tested. The largest inhibition zones were observed for B. subtilis, followed by S. aureus, E. coli, and Salmonella sp. Notably, the inhibition zones increased when the samples were exposed to light compared to dark conditions, with increases of 33 and 18 mm noted for B. subtilis. This enhanced activity under light exposure is attributed to the photocatalytic properties of the nanocomposite. The antibacterial mechanism is based on both adsorption and degradation processes. Moreover, antibacterial activity was found to increase with increasing concentrations of nanoparticles, ranging from 100 to 500 ppm. This suggests that the nanocomposite has potential as an alternative to antibiotics, especially considering the growing issue of bacterial resistance. The promising results obtained from the inhibition zones make these nanocomposites suitable for various applications. Currently, the research team is working on the development of a prototype utilizing these antimicrobial particles within commercial bottles for sterilization purposes in factories and companies. Abstract

Selenium nanoparticles (SeNPs) are reinforced safe forms of the essential micronutrient selenium (Se) which take a lead in countless biotechnological and biomedical applications. The phycosynthesis of SeNPs was successfully investigated using cell-free extract of the microalgae, Spirulina platensis. The phycosynthesized S. platensis-SeNPs (SpSeNPs) were characterized using several characterization techniques such as UV-Visible, transmission electron microscopy, Fourier transform infrared spectroscopy, X-ray diffraction, and energy dispersive X-ray. They were effectually achieved using different concentration from sodium selenite (Na2SeO3) (1, 5, and 10 mM) to give size means of 12.64, 8.61, and 5.93 nm, respectively, with spherical shapes and highly negative zeta potentialities. The infrared analyses revealed the involvement of many phycochemials in SpSeNPs production. The antibacterial properties of SpSeNPs were confirmed, qualitatively and quantitatively, against foodborne microorganisms (Staphylococcus aureus and Salmonella typhimurium); the antibacterial activity was correlated and increased with SeNPs’ size diminution. The scanning micrographs of S. typhimurium cells treated with SpSeNPs indicated the severe action of nanoparticles to destroy bacterial cells in time-dependent manners. The innovative facile phycosynthesis of SeNPs using S. platensis is recommended to generate effectual bioactive agents to control hazardous bacterial species.

Abstract

Selenium (Se) as a bioactive micronutrient could be augmented via transforming into nanoparticles (NPs), especially using biogenic protocols, for usage as an antimicrobial element. The reducing power of costus (Saussurea costus) root extract (SCE) was employed for phytosynthesis of Se-NPs through a simple and rapid protocol that included stirred mixing of 10 mM Na2SeO3 with 1.0% SCE solution for 4 h. The phytosynthesized SCE/Se-NP composite was obtained with a mean diameter of 6.13 nm and a zeta potential of −42.8 mV. Infrared analyses revealed the involvement of many SCE phytogroups in Se-NP synthesis, whereas transmission microscopy displayed well distribution and spherical shapes of the phytosynthesized NPs. The antibacterial assessments against foodborne pathogens (Escherichia coli, Salmonella typhimurium and Staphylococcus aureus) revealed the superior powers of SCE/Se-NPs and the elevated potentialities of SCE and Se-NPs for inhibition of bacterial pathogens. The scanning micrographs indicated that SCE/Se-NPs were attached to bacterial cells and led to their complete lysis/explosion with exposure prolongation. The SCE/Se-NP composites are recommended for the effective control of foodborne bacterial pathogens, applying a simple and eco-friendly phytosynthesis protocol.
