Cells of Aspergillus oryzae IPT-301 rich in fructosyltransferase (FTase) were successfully immobilized by crosslinking with glutaraldehyde and used for the transfructosylation reaction of sucrose. The glutaraldehyde concentration and pH used in the immobilization process were optimized for maximizing the transfructosylation activity (AT) and minimizing the hydrolytic activity (AH). Also, the operational stability and the influence of temperature, pH and sucrose concentration on the enzymatic activities of the free and crosslinked cells were evaluated. Both the maximum AT and minimum AH were obtained for cells immobilized with glutaraldehyde concentration of 2.1% (v/v) and pH 7.9. Crosslinked cells showed considerably higher AT/AH ratio than free cells at several temperatures, pH and sucrose concentrations in the reaction media. Kinetics data suggested that crosslinked cells present higher substrate-enzyme affinity and transfructosylation rate than free cells. Furthermore, after 12 batch reaction cycles the FTase present in the immobilized cell kept 88.9% of its initial AT, demonstrating a considerably higher operational stability than the FTase present in the free cell, which showed 50.3% of its initial AT. These results suggest the potential use of crosslinked cells of Aspergillus oryzae IPT-301 for the large-scale production of fructooligosaccharides (FOS).

Extracellular fructosyltransferase (FTase, E.C.2.4.1.9) from Aspergillus oryzae IPT-301 was immobilized on silica gel by adsorption and biochemically characterized aiming at its application in the transfructosylation reaction of sucrose for the production of fructooligossaccarides (FOS). The transfructosylation activity (AT) was maximized by the experimental design in function of the reaction pHs and temperatures. The AT of the immobilized enzyme showed the kinetics behavior described by the Hill model. The immobilized FTase showed reuse capacity for six consecutive reaction cycles and higher pH and thermal stability than the soluble enzyme. These results suggest a high potential of application of silica gel as support for FTase immobilization aiming at FOS production.

The main commercial FOS production comes from the biotransformation of sucrose and intracellular and extracellular microbial enzymes–fructosyltransferases (FTase). Aspergillus oryzae IPT-301 produces FTase. In order to increase its production, this study focuses on evaluating the effects of different agitation speed and aeration rates which affect yields in a stirred tank bioreactor. Results: Agitation had more influence on cell growth than aeration. The maximum intracellular FTase activity and the volumetric productivity of total intracellular FTase were obtained at 800 rpm and 0.75 vvm, and reached values of 2100 U g−1 and 667 U dm−3 h−1, respectively. The agitation speed had a strong influence on the activity of extracellular FTase produced which reached the maximum amount of 53 U cm−3. The higher value of total activity obtained was 22,831 U dm−3 at 0.75 vvm and 800 rpm. Conclusion: Aeration rates and agitation speed showed strong influence upon the growth and production of fructosyltransferase from Aspergillus oryzae IPT-301 in media containing sucrose as carbon source. Currently, several initiatives aiming to use byproducts rich in arabinan such as pectin and sugar beet pulp as raw material to produce various compounds of interest are being developed. However, it is necessary to use robust enzymes that have an optimal performance under pH and temperature conditions used in the industrial processes. In this work, the first GH93 from the thermophilic fungus Thermothielavioides terrestris (Abn93T) was heterologously expressed in Aspergillus nidulans, purified and biochemically characterized. The enzyme is a thermophilic glycoprotein (optimum activity at 70 °C) with prolonged stability in acid pHs (4.0 to 6.5). The presence of glycosylation affected slightly the hydrolytic capacity of the enzyme, which was further increased by 34% in the presence of 1 mM CoCl2. Small-angle X-ray scattering results show that Abn93T is a globular-like-shaped protein with a slight bulge at one end. The hydrolytic mechanism of the enzyme was elucidated using capillary zone electrophoresis and molecular docking calculations. Abn93T has an ability to produce (in synergism with arabinofuranosidases) arabinose and arabinobiose from sugar beet arabinan, which can be explored as fermentable sugars and prebiotics. BACKGROUND: Fructooligosaccharides (FOS) are sweeteners produced through the use of enzymes called fructosyltransferases (FTases) (EC 2.4.1.9). However, FTase's reuse is meager and the cost of the enzyme production is high, which turns the FOS production economically unfeasible. In order to lessen these issues, the study aims the immobilization of cells from Aspergillus oryzae IPT301, containing FTase enzyme previously glutaraldehydecrosslinked, by means of entrapment in calcium alginate beads. The conditions to obtain the particles, such as alginate type, mycelium mechanical dispersion, degree of crosslinking, size of the beads, and their reflections on transfructosylation activity (AT) were previously evaluated. RESULTS: The highest AT used low viscosity alginate solution (4.0% w/w) crosslinked with calcium chloride (0.2 molL1) and beads mean diameter of 2.15mm. The entrapment conditions were complemented by conducting the reaction under different temperatures, pH values, substrate concentrations and by reusing the beads. The highest transfructosylation activities were obtained under reaction conditions of 50 °C, pH 5.5, and substrate concentration of 400 gL1. CONCLUSION: The AT levels found with the glutaraldehydecrosslinked and entrapped cells in calcium alginate were about 70% higher than the ones obtained with free cells, not glutaraldehydecrosslinked. The anthraquinone dye Remazol Brilliant Blue R is largely used in the textile industry. However, its removal from wastewaters is costly and complex. Many methods have been tested to solve this ecological problem, but there is still a need for efficient methods. We propose here an alternative use of a two-chambered microbial fuel cell (MFC), fuelled with domestic wastewater in the anodic chamber, to degrade a simulated textile dye effluent made of Remazol Brilliant Blue R inoculated with an immobilised fungal strain, Pleurotus ostreatus URM 4809, as a laccase producer, in the cathodic chamber. The MFC showed continuous synthesis of laccase in the cathodic chamber, which, in turn, promoted the rapid decolourisation, of more than 86\% of the textile dye effluent. The yield was further increased by the addition of glycerol. Electrochemical monitoring also indicated an increase in power density and current density. β-Fructofuranosidase production by Aspergillus oryzae IPT301 was maximized in shake flasks. Response Surface Methodology (RSM) involving Small Central Composite Design was adopted to evaluate the fructosyltransferase (FTase) activity by changing three medium component concentrations: sucrose, urea and yeast extract. The optimal set of conditions for maximum fructosyltransferase production was as follows: sucrose 320.5 g/L, urea 7.13 g/L and yeast extract 2.11 g/L. In this optimal condition, the following improvements were achieved: an increase of 48.8% in cell growth, 112% and 62% in micelial and free FTase activities, respectively, 62.8% in the ratio of fructosyltransferase/hydrolytic activities for enzyme linked to mycelium and 67.5% for free enzyme.

Aspergillus oryzae IPT-301, previously reported as a β-fructofuranosidase producing microorganism, was successfully mutated using UV irradiation at 253.7 nm followed by the screening of survivors resistant to certain stress conditions. Strains were first subjected to the β-fructofuranosidase activity assay using a portion from the colony grown in Petri dish as the enzyme source. Seven mutants with fructofuranosidase activity values relative to the parent culture between 140 -190% were selected from survivors grown at temperature of 40ºC or 0.018% (w/v) sodium dodecyl sulfate concentration. They were cultivated on a rotary shaker to characterize mycelium and extracellular fructosyltransferase activities. Three mutants named IPT-745, IPT-746 and IPT-748 showed the highest amount of mycelium activity whose values increased 1.5 -1.8 fold, compared with the parent strain. It was found that more than 55% of total enzyme activity (mycelium- plus extracellular- activity) from these strains was detected in the mycelium fraction. Only one mutant, IPT-747, exceeded the amount of extracellular enzyme exhibited by the parent strain (1.5 times). This mutant also showed the highest value of total fructosyltransferase activity.

Fructooligosaccharides (FOS) are prebiotic substances found in several vegetable or natural foods. The main commercial production of FOS comes from enzymatic transformation of sucrose by the microbial enzyme fructosyltransferase. The development of more efficient enzymes, with high activity and stability, is required and this has attracted the interest of biotechnologists and microbiologists with production by several microorganisms being studied. This article reviews and discusses FOS chemical structure, enzyme characteristics, the nomenclature, producer microorganisms and enzyme production both in solid state fermentation and submerged cultivation.

This work aimed at the study of purification of biopolymers produced by Burkholderia cepacia IPT64, through a chemical attack like an adjuvant procedure through chemical and enzymatic route. Enzymatic lysis using protease was run on an enzyme/cell ratio of 0.02. Chemical attacks were performed as pre or post-treatment for enzymatic attacks, using sodium dodecyl sulphate or hydrogen peroxide. The same chemicals and procedures were used alone as a control. Fenton’s reagent was also tested as a chemical treatment. Using only one of the chemicals H2O2, Fenton’s reagent or SDS, the purity increase achieved values of 14%, 16% or 23%, respectively. When H2O2 or SDS were used as pre-treatment for the enzymatic attack, the results of purity increase achieved values of 58% for H2O2/cell ratio between 0.60 and 1.20, and 57% when SDS/cell ratio at 0.56 was used. In the case when H2O2 or SDS were used as post-treatment for the enzymatic attack, results of purity increase achieved 60% when the H2O2 was used at H2O2/cell ratio ranging between 0.30 and 0.60 and 71% when SDS was used at a ratio SDS/cell of 0.56.

Seventeen different strains of filamentous fungi were grown in batch cultures to compare their abilities for the production of beta-fructofuranosidase. Three of them, Aspergillus oryzae IPT-301, Aspergillus niger ATCC 20611 and strain IPT-615, showed high production with total fructosyltransferase activity higher than 12,500 units l(-1). In addition, the beta-fructofuranosidases of those strains have a high fructosyltransferase activity-to-hydrolytic activity ratio. The temperature and pH effects on the sucrose-beta-fructofuranosidase reaction rate were studied using a 2(2) factorial experimental design. The comparative analysis of the tested variable coefficients shows that the variable pH contributes mostly to the changes in the fructosyltransferase and hydrolytic rates and in the V (t)/V (h) ratio. At 40 and 50 degrees C, there were no significant differences between the fructosyltransferase and hydrolytic velocities of these enzymes.

Polyhydroxybutyrate (PHB) is the most studied among a wide variety of polyhydroxyalkanoates, bacterial biodegradable polymers known as potential substitutes for conventional plastics. This work aimed at evaluating the use of enzymes to recover and purify the PHB produced by Ralstonia eutropha DSM545. Screening experiments allowed the selection of trypsin, bromelain and lysozyme among six enzymes, based on their efficiency in lysing cells of a non-PHB producing R. eutropha strain. Then, process conditions for high efficiency in PHB purification from the DSM545 cells were searched for the enzymes previously selected. The best result was achieved with 2.0% of bromelain (enzyme mass per biomass), equivalent to 14.1 U ml(-1), at 50 degrees C and pH 9.0, resulting in 88.8% PHB purity. Aiming at improving the process efficiency and reducing the enzyme cost, experiments were carried out with pancreatin, leading to 90.0% polymer purity and an enzyme cost three times lower than the one obtained with bromelain. The molecular mass analysis of PHB showed no polymer degradation. Therefore, this work demonstrates the potential of using enzymes in order to recover and purify PHB and bacterial biopolymers in general.

Nas melhores condições testadas, quais sejam: reator de leito empacotado, com suporte de esferas de cerâmica e vazão de 2,3 vvm, foi atingida eficiência de remoção de fenol de aproximadamente 96 %.A process for phenol biodegradation using biofilms was studied, using a synthetic waste to simulate industrial wastewater with high phenol concentrations. The aim of the experiments was to verify the influence of the aeration of 0.7, 1.6, 2.3 vvm in the kinetics of phenol biodegradation. In the best experimented conditions used that were: packed bead reactor, ceramic spheres support and flow rate of 2.3 vvm, was achieved phenol removal efficiency near 96%.

The decrease in viscosity is widely used to estimate the activity of pectinolytic enzymes. This method is shown to be influenced by the production strain and this prevents an accurate comparison between the activities of different microorganisms, especially under different conditions.

No. of bitstreams: 0 Previous issue date: 2017-12-01 The present work had the goal of screening a batch of 20 fungal strains, isolated from sugar cane plantation soil, in order to identify those capable of biosynthesis of silver nanoparticles. These nanoparticles are known to have a large and effective application in clinical microbiology. Four strains were found to be capable of biosynthesis of silver nanoparticles. The biosynthesised nanoparticles were characterised by UV–vis spectroscopy, scanning electron microscopy, EDX, and XRD. They were found to have an average size of 30–100 nm, a regular round shape, and potential antimicrobial activity against Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa. The antimicrobial activity was found to be directly related to the nanoparticles concentration. Mycogenic synthesis of nanoparticles is a green biogenic process preferable to other alternatives. Because fungi are great producers of extracellular enzymes this process makes scaling-up an easier task with high importance for clinical microbiology on the fight against microbial resistance, as well as for other industrial applications. It was the objective of the present study to increase the production of glucoamylase by Aspergillus awamori through solid state fermentation, using wheat bran as the main carbon source and (NH4)2SO4, urea, KH2PO4, glucose, maltose and starch as additional nitrogen, phosphorus, and carbon sources. The production of glucoamylase is strongly influenced by N and C sources. A 100% increase was observed when the (NH4)2SO4 was replaced by urea, with C/N = 4.8, using maltose as the additional carbon source. C/P ratios in a range of 5.1 to 28.7 did not induce glucoamylase production under the studied conditions.

O Programa de Produção e Uso do Biodiesel (PNPB), lançado em 2004, foi a primeira política agroenergética com viés social no Brasil. Desse modo, além da implementação da indústria do biodiesel e de um novo mercado no país, o Programa tem como objetivo a inclusão de agricultores familiares como fornecedores de matéria-prima. O biodiesel, embora não seja uma solução disruptiva para descarbonização dos transportes, é utilizado como substituto parcial do diesel em motores a combustão interna e emite menos Gases do Efeito Estufa (GEEs) em comparação ao seu equivalente fóssil. Em 2008, o governo federal introduziu a mistura compulsória de 2% (B2) de biodiesel ao diesel. Essa porcentagem foi sendo gradualmente acrescida, atingindo 10% (B10) em 2018. Esse trabalho avaliou os impactos do PNPB como política energética nas dimensões tecnológica, econômica, social e ambiental. Foram utilizados dados disponibilizados pelo governo brasileiro e discussões da literatura para traçar os resultados do Programa entre 2008 e 2018 e, com isso, avaliar os avanços e desafios. Foi possível verificar que a expansão da produção e da capacidade instalada acompanhou o aumento da porcentagem da mistura. A consolidação dessa indústria nacional se apoiou em grandes empresas de processamento de soja, o que, por um lado, trouxe a segurança e a garantia do abastecimento, por outro, levou ao fechamento de usinas de pequeno porte e uma distribuição desigual da produção pelo território nacional. Foi constatada uma redução da dependência externa por diesel importado e uma vantagem econômica nesse balanço. Ademais, foram reunidas estimativas das emissões evitadas de gás carbônico equivalente (CO2eq) pelo uso do biodiesel. Na dimensão social, embora a arrecadação média anual das famílias incluídas tenha aumentado, os números da inclusão estão aquém dos objetivos iniciais. Outro desafio remanescente é a diversificação das matérias-primas para o biodiesel. O aumento para 15% de mistura (B15) está previsto até 2025 e as perspectivas diante da nova Política Nacional dos Biocombustíveis, o RenovaBio, apontam que a dimensão ambiental pode orientar novos rumos, incentivando inovações na tecnologia de produção e maior aproveitamento de resíduos. Com isso, a avaliação realizada permitiu uma discussão sobre os principais aspectos do PNPB, apontando, inclusive a falta de transparência em alguns dados. A análise pode ser utilizada em trabalhos futuros para avaliação do setor de biodiesel a partir dos novos impulsos. The Biodiesel Production and Use Program (PNPB), launched in 2004, was the first rural and energy policy with social directives in Brazil. In addition to the implementation of the biodiesel industry and market, the Program aims to include family farmers as suppliers of raw materials. Biodiesel is a partial substitute for diesel in internal combustion engines and emits less Green House Gases (GHG) compared to fossil diesel, although it is not a disruptive solution for transport decarbonisation. In 2008, the federal government introduced the mandatory blend of 2% biodiesel to diesel (B2). This percentage gradually increased, reaching 10% in 2018 (B10). This work evaluated the impacts of PNPB as an energy policy in the technological, economic, social and environmental dimensions. Official data and literature review supported the analysis of the Program\'s results between 2008 and 2018, including the advances and challenges. It was possible to verify that the expansion of production and installed capacity followed the rise of mandatory blends. The consolidation of the national biodiesel industry relied on the large soy processing industries, which, on the one hand, brought security of supply, on the other, led to a closing of small plants and an uneven distribution of the plants throughout the national territory. There was a reduction in external dependence on imported diesel and an economic advantage in this balance. In addition, the PNPB is responsible to avoided CO2eq emissions. In the social dimension, although the average annual income of the families included has increased, the numbers of inclusion are below the initial objectives. Another remain challenge is the diversification of raw materials for biodiesel. The increase to 15% blend (B15) is expected by 2025 and the prospects for the new National Biofuel Policy, RenovaBio, point out that the environmental dimension can guide new directions, encouraging innovations in technology and greater use of waste materials. This work support future analyses of the performance of the biodiesel sector from the new impulses.

The new elephant grass PCEA (P. purpureum x P. purpureum) was specifically developed to meet Brazilian tropical conditions and is the first seed propagated genotype worldwide to be launched by Embrapa Dairy Cattle (CNPGL). The advancement of lignocellulosic bioethanol in Brazil faces numerous challenges, including efficient pretreatments designed for each raw material. In this context, the effect of alkaline pretreatment on PCEA biomass was evaluated to infer about its potential for lignocellulosic bioethanol. The experimental design approached three factors: NaOH %w/v ranging from 1.0 to 3.0 varying on 0.5 %w/v, Temperature (T) from 80 to 120 varying on 20 °C and Reaction Time (RT) from 15 to 35 min varying on 5 min each. The effect of the factors was evaluated considering the response variables of biomass recovery, cellulose and hemicellulose yield, in addition to lignin removal. The untreated biomass and the 17 samples generated from assays were dried and characterized regarding its composition following NREL protocols. The untreated biomass of PCEA presented 31.79±1.55 %wt. of cellulose, 15.51±0.70 %wt. of hemicellulose and 18.23±0.45 %wt. lignin. The observed ash content of 3.94±0.07 %wt. is also important for bioenergy crops, it is desirable on lower values for energy conversion. In addition, the content of 20.54±0.11 %wt. extractives on PCEA indicates further potential to generate biofuels through thermochemical routes. After the alkaline pretreatment, the highest cellulose yield of 58.59 %wt. was achieved in assay 14 at 100 ºC, 25 min of RT and NaOH 3% (w/v). Contrary to initial expectations, only alkali concentration statistically differed at 95% confidence level analysis for the full quadratic model at a coefficient of determination (R2) of 0.78. In contrast, hemicellulose as response for linear with interactions model was significant at 95% confidence level (p-value 0.022), which suggests that the interaction of the three factors altogether had an effect on hemicellulose yield. The highest hemicellulose yield of 25.52 %wt. was reached in assay 9 at 80 ºC, 25 min of RT and 2 %w/v concentration of NaOH. The alkali alone had a p-value of 0.000 (ANOVA at 95% confidence level), which suggests an effect of this factor on biomass recovery. The same was not true for the square and linear with interactions models that presented p-values of 0.231 and 0.353, respectively. The coefficient of determination (R2) for the full quadratic model was 0.92 and the highest recovery of 67.92 %wt. was obtained at the 1.0 %w/v NaOH, at 100 °C and 25 min (assay 13). In conclusion, the optimal condition of the design was established considering efficiency in means of maximal lignin removal without degradation of cellulose, which was achieved at 100 ºC, 25 RT and 3 %w/v NaOH with 99.03 %wt. recovery of cellulose and 73.76 %wt. of lignin removal. These results suggest an exciting potential of the new PCEA for lignocellulosic bioethanol production via alkaline pretreatment to be further explored by the Brazilian agroenergy sector. However, further studies should be carried out with enzymatic hydrolysis to confirm high yield of fermentable sugar and consequently, bioethanol. O novo capim elefante PCEA (P. purpureum x P. purpureum) foi desenvolvido para atender às condições de clima tropical do Brasil, sendo o primeiro genótipo propagado por semente no mundo a ser lançado pela Embrapa Gado de Leite (CNPGL). No Brasil, o avanço do bioetanol lignocelulósico enfrenta inúmeros desafios, incluindo pré-tratamentos eficientes designados para cada matéria-prima. Neste contexto, o efeito do pré-tratamento alcalino foi avaliado para biomassa de PCEA a fim de inferir sobre o seu potencial para bioetanol lignocelulósico. O delineamento experimental analisou três fatores: NaOH %w/v variando de 1,0 a 3,0 em 0,5 %w/v, Temperatura (T) de 80 a 120 variando em 20 °C e Tempo de Reação (RT) de 15 a 35 min variando em 5 min cada. O efeito dos fatores foi avaliado para resposta de recuperação da biomassa, rendimentos de celulose e hemicelulose, além da remoção da lignina. A biomassa não tratada e as 17 amostras geradas a partir dos ensaios foram secas e caracterizadas quanto à sua composição de acordo com protocolos NREL. A biomassa não tratada de PCEA apresentou 31,79±1,55 %wt. de celulose, 15,51±0,70 %wt. de hemicelulose e 18,23±0,45 %wt. de lignina. O teor de cinzas observado foi de 3,94 %wt. (±0,07) deve ser considerado para as culturas bioenergéticas, uma vez que é desejavel em menor quantidade possível para usos energéticos. Além disso, o teor de 20,54±0,11 %wt. de extrativos no PCEA indica um potencial adicional de geração de biocombustíveis através de rotas termoquímicas. Após o pré-tratamento alcalino, o maior rendimento de celulose de 58,59 %wt. foi alcançado no ensaio 14 a 100 ºC, 25 min de RT e NaOH 3% (p/v). Ao contrário das expectativas iniciais, apenas a concentração alcalina diferiu estatisticamente na ANOVA nível de confiança de 95% para o modelo quadrático completo com um coeficiente de determinação (R2) de 0,78. Em contraste, a hemicelulose como resposta para o modelo linear com interações foi significativa ao nível de confiança de 95% (p-valor 0,022), o que sugere que a interação dos três fatores teve efeito sobre o rendimento da hemicelulose. O maior rendimento de hemicelulose de 25,52%wt. foi alcançado no ensaio 9 a 80 ºC, 25 min de RT e 2%w/v de concentração de NaOH. Somente o álcali teve um p-valor de 0,000 (ANOVA a 95% de confiança), o que sugere um efeito deste fator na recuperação da biomassa. O mesmo não ocorreu com os modelos quadrático e linear com interações que apresentaram p-valores de 0,231 e 0,353, respectivamente. O coeficiente de determinação (R2) para o modelo quadrático completo foi de 0,92 e a maior recuperação de 67,92 %wt. foi obtida a 1,0 %w/v NaOH, T of 100 °C e TR of 25 min (ensaio 13). Com isso, a melhor resposta do design considerando a eficiência quanto à máxima remoção de lignina sem degradar a celulose foi observada a 100 ºC, 25 RT e 3 %w/v NaOH com 99,03 %wt. de recuperação da celulose e 73,76 %wt. de remoção da lignina. Estes resultados sugerem um potencial animador do novo PCEA para a produção de bioetanol lignocelulósico através de pré-tratamento alcalino a ser explorado pelo sector agroenergético brasileiro. No entanto, estudos futuros de hidrólise enzimática se fazem necessários, a fim de confirmar o elevado rendimento de açúcares fermentáveis e, consequentemente, de bioetanol.

Segundo a classificação CAZy (Carbohydrate Active enZymes), as glicosil hidrolases (GH) da família 93 são exo-?-1,5-arabinanases que atuam nas ligações ?-1,5 que unem os resíduos de L-arabinofuranosideo que conformam a cadeia principal de arabinana desramificada, liberando arabinose ou arabinobiose. Esta recém descrita família de enzimas, além de ter o potencial de participar junto com outras proteínas na sacarificação de resíduos industriais como a polpa de beterraba, oferece a possibilidade de sintetizar novas moléculas, pois alguns membros desta família podem catalisar reações de transarabinosilação utilizando glicerol, xilitol e sorbitol como aceptores. Embora as GH93 apresentem caraterísticas atrativas para aplicações industriais, até o momento apenas cinco enzimas desta família foram caraterizadas bioquimicamente, sendo que a maioria delas apresentou maior atividade em pHs ácidos e nenhuma com caraterísticas termofílicas. Neste trabalho uma exo-?-1,5-arabinanase abnt do fungo termofílico Thielavia terrestris foi expressa heterólogamente pela primeira vez em fungo filamentoso, utilizando Aspergillus nidulans linhagem A773 transformado com o vetor pEXPYR como sistema de expressão. A enzima foi caraterizada bioquimicamente e sua produção fermentativa avaliada, utilizando dois tipos de indutores em diferentes concentrações e condições de cultivo, escolhendo a melhor condição para o escalonamento. A proteína recombinante de 39,8 KDa mostrou ser uma glicoproteína com uma atividade especifica de 248 U.mg-1 em arabinana desramificada 0,28 mM, pH 5 e temperatura de 70 ?C (valores ótimos para enzima), um Km 0,29 mM, Vmax 0,45 ?mol.min-1, Kcat 115,4 s-1, e eficiência catalítica de 3,9 *105 s-1 M-1, mostrando também um aumento de 34% na liberação de produto quando incubada na presença de CoCl2 1mM. Esta é a primeira exo-arabinanase reportada na literatura com comportamento termofílico e prolongada estabilidade em pHs ácidos. No processo fermentativo a melhor produção de abnt foi atingida usando 3% (m/v) do indutor em condições de cultivo submerso, o escalonamento do processo não afetou a produção enzimática. O sistema de expressão A. nidulans+pEXPYR foi capaz de produzir a mesma quantidade de proteína recombinante utilizando como indutor, um composto cinquenta vezes mais barato que o normalmente utilizado. According to CAZy (Carbohydrate Active enZymes) classification, glycosyl hydrolases (GH) from family 93 are exo-?-1,5-arabinanases acting on ?-1,5 bonds that bind L-arabinofuranose residues in the main chain of arabinan releasing arabinose or arabinobiose. This new family of enzymes was described recently and, besides having the potential to participate along with other proteins in the saccharification of agro industrial wastes such as sugar beet pulp, offers the possibility of synthesize new molecules, since some members of this family can catalyze reactions of transarabinosylation using glycerol, xylitol and sorbitol as acceptors. Although, GH93 presents attractive characteristics for industrial applications, only five enzymes have been characterized biochemically so far, most of them presenting higher activity in acidic pHs and none with thermophilic characteristics. In this work an exo-?-1,5-arabinanase abnt of the thermophilic fungus Thielavia terrestris was heterologously expressed for the first time in a filamentous fungus using Aspergillus nidulans strain A773 transformed with the pEXPYR vector as expression system. The enzyme was characterized biochemically and its fermentative production was evaluated using two types of inducers in different concentrations and conditions of cultivation, setting the best condition for scale up. The recombinant protein of 39.8 KDa showed be a glycoprotein with a specific activity of 248 U.mg-1 in 0.28 mM debranched arabinana at pH 5 and temperature of 70 °C (optimum values for enzyme), a Km 0,29 mM, Vmax 0.45 ?mol.min-1, Kcat 115.4 s-1, and catalytic efficiency of 3.9 * 105 s-1 M-1, also showed an increase of 34% in product release when incubated in the presence of 1 mM CoCl2. This, is the first exo-arabinanase reported in the literature with thermophilic behavior and prolonged stability at acidic pHs. In the fermentation process the best abnt production was achieved using 3% (m/v) of the inducer under submerged culture conditions, and the scaling process did not affect the enzymatic production. The A. nidulans+pEXPYR expression system was able to produce the same amount of recombinant protein using as an inducer, a compound fifty fold less expensive then the normally used.

A busca por uma fonte de matéria prima renovável alternativa aos combustíveis fósseis tem impulsionado o desenvolvimento de novas fontes de biomassa para os biocombustíveis. Uma fonte de biomassa alternativa que vêm ganhando destaque são as microalgas, microorganismos fotossintéticos capazes de capturar o CO2 atmosférico e acumular altos teores de lipídeos em sua biomassa, tornando-os muito atrativos como matéria-prima para a síntese de biodiesel. Nesse sentido, muitos trabalhos têm sidos conduzidos para o desenvolvimento dos mais variados tipos de fotobiorreatores para o cultivo de microalgas. Assim, o presente trabalho propôs um fotobiorreator integrado (coluna de bolhas e tubular), internamente iluminado com luz de LED, para estudar os efeitos dos fatores: cor da iluminação, fotoperíodo, aeração e vazão de circulação de meio, no cultivo foto-autotrófico da microalga marinha Chlorella minutissima em duas fases, utilizando o planejamento estatístico de Taguchi (L9), com vistas à formação de biomassa e ao acúmulo de lipídeos nessa microalga, para a produção de biodiesel. Foi alcançada a máxima concentração de biomassa de 506 mg L -1 e a máxima produtividade volumétrica de biomassa de 62,5 mg L -1 com 20,2 % de lipídeos para o cultivo da Chlorella minutissima no fotobiorreator integrado internamente iluminado com a cor azul, em fotoperíodo de 24 h diárias de iluminação, provido com 0,5 vvm de aeração e 5,1 L min -1 de vazão de recirculação de meio de cultivo. O perfil dos ácidos graxos presentes no extrato lipídico da Chlorella minutissima apresentou predominância de ácidos graxos saturados, com destaque para o ácido palmítico (C 16 : 0), com 59,49 %, 57,28 % e 46,5 %, nos cultivos iluminados com as cores, branco, vermelho e azul, respectivamente. A partir dos lipídeos extraídos, atingiu-se 96,6 % de conversão em biodiesel utilizando etanol via catálise ácida com ácido sulfúrico (10 % m / m) durante 8 h de reação à 80 ?C. The search for a renewable source alternative to fossil fuels has driven the development of new sources of biomass for biofuels. An alternative source of biomass that have come to prominence are microalgae, photosynthetic micro-organisms capable of capturing atmospheric CO2 and accumulating high levels of lipids in their biomass, making them very attractive as a raw material for biodiesel synthesis. In this way, many studies have been conducted for the development of the many types of photobioreactors for the cultivation of microalgae. Thus, the present work proposed an integrated photobioreactor (bubble and tubular column), internally illuminated with LED light, to study the effects of the factors: illumination color, photoperiod, aeration and culture medium recirculation flow rate, in photo-autotrophic cultivation of the Chlorella minutissima, in two phases, using design of the experiment (Taguchi - L9), aiming the biomass production and lipid accumulation, for the biodiesel production. The maximum biomass concentration of 506 mg L -1 and the maximum biomass volumetric productivity of 62.5 mg L-1 with 20.2% of lipids were obtained with the cultivation of Chlorella minutissima in the integrated photobioreactor internally with blue illumination, 24h of photoperiod, 0.5 vvm of aeration and 5.1 L min -1 of culture medium recirculation flow rate. The fatty acids profile present in the lipid extract of Chlorella minutissima showed a predominance of saturated fatty acids, with emphasis on palmitic acid (C 16: 0), with 59.49%, 57.28% and 46.5%, to cultivations illuminated with white, red and blue, respectively. From the extracted lipids, 96.6% were converted in biodiesel via acid catalysis using ethanol with sulfuric acid (10% w / w) for 8 h of reaction at 80 ° C.

Frutose e Ácido Glicônico são produtos importados empregados em diferentes setores nas áreas química, farmacêutica e alimentícia, representando um mercado de dois milhões de dólares (US$ 2,0 milhões) por ano. Por sua vez, a sacarose pode ser empregada como matéria-prima para a obtenção destes produtos através de conversão enzimátiva empregando invertase e glicose-oxidase. O uso de biorreatores com membrana (MBR) mostra-se interessante em processos enzimáticos, pois, ao serem empregados em processos contínuos permitem, simultaneamente, produção e separação dos produtos, reduzindo a formação de subprodutos e, eventual, inibição da enzima por excesso de substrato ou produtos. A sacarose é convertida em xarope de açúcar invertido (solução equimolar de frutose e glicose) pela invertase (Bioinvert®, enzima comercial), seguido pela oxidação da glicose em ácido glicônico pela ação da glicose oxidase (GO). O processo de conversão multi-enzimático da sacarose foi obtido através da alimentação de sacarose (50 mM) em reator com membrana (MBR) contendo invertase (24 U/mL), glicose-oxidase (0,5 U/mL) e catalase (470 U/mL) e operando com vazão específica de 6,0 h-1, 35ºC e pH 5,5. As condições operacionais otimizadas possibilitaram a conversão completa da sacarose (X = 100 %) e da glicose resultante (Y = 100%) com velocidades específicas de reação de 4,2 mmol/U.h, 0,60 mmol/U.h e 0,00062 mmol/U.h, respectivamente, para a invertase, glicose oxidase e catalase. A respeito da oxidação da glicose, a adição de catalase no meio reacional se fez necessária para minimizar os efeitos inibitórios sobre a GO através do peróxido de hidrogênio formado. The fructose and gluconic acid are products of great application in chemical, pharmaceutical and food industry. The actual Brazilian market for these compounds is about US$ 2 millions, here as the sucrose, the raw-material used for their production, represents about 2.4% of the Brazil\'s GNP. This conversion increases the value added to the sugarcane, usually marketed as a commodity, because the fructose and gluconic acid are more valuable products than sucrose. The use of membrane bioreactor (MBR), which operates under mild conditions regarding internal pressure, temperature and pH, has been growing along the years for enzyme catalyzed processes. Moreover, in the MBR the reaction and separation of the products occur simultaneously, avoiding the formation of by-products and the eventual inhibition of the enzyme caused by excess of substrate or products. The sucrose is converted to the inverted syrup (an equimolar solution of fructose and glucose) by invertase (in this work was employed Bioinvert®, a commercial invertase) followed by the oxidation of glucose in gluconic acid by the glucose oxidase (GO). The multi-enzymatic conversion of sucrose was attained when carried out under initial substrate of 50mM and invertase, glucose oxidase and catalase concentrations, respectively, of 24.0 U/mL, 0.5 U/mL and 470 U/mL in a membrane reactor utilizing a dilution rate of 6.0 h-1, 35ºC and pH 5.5. The optimized operational conditions led to a conversion yield of 100% for sucrose hydrolysis and glucose oxidation steps resulting in enzyme productivity of 4.2 mmol/U.h, 0.60 mmol/U.h and 0.00062 mmol/U.h, respectively, to invertase, glucose oxidase and catalase. In regard to the glucose oxidation, the addition of catalase in the reaction medium was necessary, in order to minimize the inhibition of the GO by the hydrogen peroxide formed.

O presente estudo teve como objetivo avaliar a conversão dos açúcares do bagaço de cana em etanol, com foco na integração entre o pré-tratamento com H2SO4 diluído, sacarificação com celulases comerciais e fermentação com Scheffersomyces stipitis DSM- 3651, buscando-se uma configuração simplificada. Previamente aos ensaios de integração, o bagaço foi moído para a redução de sua heterogeneidade granulométrica. Após a moagem, o bagaço foi submetido a fracionamento, por fluxo de ar, separando frações de fibras e finos. Estas frações foram submetidas a analise composicional e observou-se que a fração de finos possui elevados teores de cinzas e extrativos, além de baixo teor de lignina em relação à fração de fibras, entretanto a estratégia de fracionamento do bagaço não foi seletiva. Os experimentos de definição da condição de pré-tratamento mais adequada para a avaliação entre integração entre sacarificação e fermentação foram conduzidos em reator Parr (2 gal) com 2Kg de mistura reacional, incluindo bagaço (10%), água e ácido. Três condições operacionais de pré-tratamento foram avaliadas: 140°C/1%H2SO4/20 min, 150°C/2% H2SO4/30 min e 160°C/3% H2SO4/40 min. Após o pré-tratamento, o slurry foi separado em sólido pré-tratado e hidrolisado hemicelulósico, por filtração. Em seguida o bagaço pré-tratado foi lavado exaustivamente com água destilada e submetido à hidrólise enzimática com celulases (10 FPU/g bagaço) por 72h. Paralelamente, o hidrolisado foi submetido à fermentação com S. stipitis (3 g/L) por 120h. As composições das frações geradas após o pré-tratamento (bagaço pré-tratado, hidrolisado e água de lavagem), quanto aos teores de açúcares e sólidos (solúveis e insolúveis) foram determinadas. As três condições de pré-tratamento levaram a remoção completa de hemicelulose do bagaço, e o aumento da severidade do pré-tratamento acarretou em aumento da eficiência de sacarificação dos sólidos pré-tratados, porém em menor recuperação de glicose após hidrólise enzimática. Por outro lado, o aumento da severidade do pré-tratamento prejudicou a recuperação de açúcares no hidrolisado hemicelulósico, elevando concentrações de ácido acético, furfural e HMF, resultando em inibição completa de fermentação daqueles obtidos nas condições de 150°C e 160°C. A conversão do sólido prétratado (lavado ou não-lavado) + hidrolisado (destoxificado ou não), e do slurry completo (destoxificado ou não) ambos obtidos após pré-tratamento à 140°C, foi avaliada em duas configurações, respectivamente: \"Fermentação do hidrolisado hemicelulósico (FHH), em separado da sacarificação do sólido pré-tratado e da fermentação do hidrolisado celulósico, separadas (SHF)\" e \"Sacarificação do sólido pré-tratado, em separado da co-fermentação dos hidrolisados celulósico e hemicelulósico, integradas (ISHCF)\". Na primeira, observouse que a destoxificação do hidrolisado com lacase (1U/mL) aumentou a produção de etanol, e possibilitou a redução do tempo de conversão em 48h. A sacarificação de sólidos não-lavados foi prejudicada pela presença de compostos solúveis e o condicionamento com lacase não influenciou a eficiência de sacarificação. A eficiência máxima de conversão em configuração de SHF+FHH foi de 28,4%, devido a não lavagem do sólido pré-tratado (13,7%) e destoxificação do HH (14,7%). Em ISHCF, observou-se que a eficiência de conversão de slurry integral destoxificado com lacase é mais alta (38,9%) do que em SHF+FHH, devido a não separação sólido-líquido e maior disponibilidade de açúcares. This study aimed to evaluate the conversion of sugarcane bagasse into ethanol, focusing on integration between pretreatment with dilute H2SO4, saccharification with commercial cellulases and fermentation with Scheffersomyces stipitis DSM-3651, in a simplified design process. Prior to integration assays, the bagasse was milled to reduce its granulometric heterogeneity. After milling, the fractions of bagasse (fiber and pith) were separated by air flow. These fractions had their chemical composition determined and it was observed that pith fraction has higher content of ash, soluble extractives and low lignin content than fiber fraction; however the separation method was not selective. The experiments to define the most suitable pretreatment condition for evaluating the integration between saccharification and fermentation were conducted in a Parr reactor (2 gal) with 2 kg of reaction mixture, including bagasse (10%), water and acid. Three operational conditions of pretreatment were evaluated: 140°C/1% H2SO4/20min, 150°C/ 2% H2SO4/30 min and 160°C/3% H2SO4/40min. After pretreatment, the slurry was separated in pretreated solid and hemicellulosic hydrolyzate, by filtration. The pretreated solid was thoroughly washed with distilled water and submitted to enzymatic hydrolysis with cellulases (10 FPU / g residue) for 72h. In parallel, the hemicelullosic hydrolyzate was submitted to fermentation with S. stipitis (3 g / L) for 120h. The compositions of the fractions generated after pretreatment (pretreated bagasse, hemicellulosic hydrolyzate and washing water), regarding sugars and solids (soluble and insoluble) contents were determined. All pretreatment conditions led to complete removal of hemicellulose from the bagasse, and the increase of pretreatment severity improved the saccharification yield of pretreated solid, but, leaded to low recovery of glucose after enzymatic hydrolysis. On the other hand, the increase of pretreatement severity impaired the recovery of sugars in hemicellulosic hydrolysate besides increasing the concentrations of acetic acid, furfural and HMF, resulting in complete inhibition of fermentation of those obtained under the pretreatement conditions of 150°C and 160°C. The conversion of pretreated solid (washed and non-washed) + hemicellulosic hydrolyzate (with or without detoxification) and of the whole slurry, both obtained at 140°C was evaluated in two process designs, respectively: \"Fermentation of hemicellulosic hydrolyzate (FHH), separated from hydrolysis of pretreated solid and fermentation of cellulosic hydrolyzate, separately (SHF)\" and \"Integrated hydrolysis of pretreated solid separated from co-fermentation of cellulosic and hemicellulosic hydrolysates (ISHCF)\". In the first, it was observed that the detoxification of the hydrolyzate with laccase (1U / ml) improved the ethanol production, particularly for conversion time. The saccharification of non-washed solid was hampered by the presence of soluble compounds and its conditioning treatment with laccase did not influence the saccharification yield. The maximum conversion efficiency in SHF + FHH configuration was 28.4% due to not washing the pretreated solid (13.7%) and detoxification of hemicellulosic hydrolysate (14.7%). In the second process design, it was observed that the conversion efficiency of whole slurry, with laccase detoxification, was higher (38.9%) than in the separate configuration, with a lower conversion time.

A conversão de biomassa vegetal em produtos químicos e energia é essencial a fim de sustentar o nosso modo de vida atual. O bagaço de cana-de-açúcar, matériaprima disponível em abundância no Brasil, poderá tanto ajudar a suprir a crescente demanda pelo etanol combustível como ser empregado para obtenção de produtos de valor agregado, tais como xilitol, além de trazer vantagens econômicas para o setor sucroalcooleiro. O xilitol, um poliol com poder adoçante semelhante ao da sacarose e com propriedades peculiares, como metabolismo independente de insulina, anticariogenicidade e aplicações na área clínica, no tratamento de osteoporose e de doenças respiratórias, é obtido em escala comercial por catálise química de materiais lignocelulósicos. A produção biotecnológica de xilitol como alternativa ao processo químico vem sendo pesquisada e os resultados revelam que a presença de compostos tóxicos nos hidrolisados hemicelulósicos resultantes do processo de hidrólise ácida contribui para sua baixa fermentabilidade. Isto se deve à inibição do metabolismo microbiano causada principalmente por compostos tais como ácidos orgânicos, fenólicos e íons metálicos. No presente trabalho foi avaliado o efeito de diferentes fontes de carbono (xilose, glicose e mistura de xilose e glicose) empregadas no preparo do inóculo de Candida guilliermondii FTI 20037 sobre a bioconversão de xilose em xilitol a partir de fermentações em frascos Erlenmeyer de hidrolisados hemicelulósicos submetidos a procedimentos de destoxificação. A condição de favorecimento deste bioprocesso foi empregada para a avaliação da ampliação de escala em fermentadores de 2,4L para 16L, utilizando como critério de ampliação o KLa (igual a 15h-1). De acordo com os resultados, os máximos valores dos parâmetros fermentativos como fator de conversão de xilose em xilitol e produtividade em xilitol foram alcançados com a utilização de inóculo obtido em xilose durante fermentação do hidrolisado destoxificado por resinas (YP/S = 0,81 g g-1 e QP = 0,60 g L-1 h-1, respectivamente), embora o emprego de carvão ativado tenha gerado valores de rendimento próximos para as diferentes fontes de carbono (YP/S variando de 0,78 a 0,80 g g-1). Considerando o valor de fator de conversão e que o procedimento de destoxificação com carvão ativado é o de menor custo e de mais fácil manipulação em comparação ao processo com resinas, os experimentos de ampliação de escala da produção de xilitol por C. guilliermondii foram realizados nesta condição de destoxificação e empregando-se xilose como fonte de carbono para o inóculo. Nesta etapa ficou evidente a viabilidade de ampliação de escala de produção de xilitol de fermentador de 2,4L para 16L, já que os valores dos parâmetros fermentativos avaliados foram semelhantes entre os fermentadores (valores médios: YP/S ≈ 0,68 g g-1 e QP ≈ 0,28 g L-1 h-1). No entanto, tais valores foram inferiores aos obtidos em frascos Erlenmeyer, possivelmente devido às condições de disponibilidade de oxigênio diferirem nos fermentadores de bancada, uma vez que o oxigênio é o parâmetro mais crítico neste bioprocesso. The conversion of vegetable biomass into chemicals and energy is essential to sustain our current style of life. Sugarcane bagasse, a raw material abundantly available in Brazil, greatly contributes to the supply of the evergrowing demand for ethanol. Furthermore, biomass can be employed for obtaining value-added products, such as xylitol, as well as bring economical advantages for the sugar-ethanol sector. Xylitol, a polyol with sweetener power similar to that of saccharose and peculiar properties such as insulin-independent metabolism, anticariogenic power, and applications in the clinical area, in the treatment of osteoporosis and respiratory diseases, is obtained on a commercial scale by chemical catalysis of lignocellulosic materials. The biotechnological production of xylitol as an alternative to the chemical process has been researched and the results reveal that the presence of toxic compounds in hemicelllosics hydrolysates resulting from acid hydrolysis process contributes to its low fermentability. Such toxicity could be due to the inhibition of microbial metabolism promoted mainly by compounds such as organic acids, phenols and metallic ions. In the present work, the effect of different carbon sources (xylose, glucose and a mixture of xylose and glucose) used in the inoculum preparation of Candida guilliermondii FTI 20037 for the xylose-to-xylitol bioconversion by fermentation of hemicellulosics hydrolysates submitted to detoxification procedures in Erlenmeyer flasks was evaluated. The best condition for this bioprocess was employed to evaluate the scale up from the 2.4L to 16L fermentors, using KLa (equal to 15h-1) as scale-up criteria. According to the results the highest values of fermentative parameters such as xylitol yield and productivity were achieved with the use of inoculum cultivated on xylose during the fermentation of hydrolysate detoxified with resins (YP/S = 0.81 g g-1 and QP = 0.60 g L-1 h-1, respectively), although with the use of charcoal the yield value was similar (YP/S ranging for 0.78 to 0.80 g g-1), regardless of the carbon source employed. Considering the value of xylitol yield and that detoxification with activated charcoal is less expensive and more easily manipulated when compared to detoxification procedure with resins, the experiments for scale up xylitol production by C. guilliermondii were performed in such detoxification condition with xylose as the carbon source for the inoculum. At this stage it was evident the scale up xylitol production from a fermenter of 2.4L to 16L was feasible, since the values of fermentative parameters evaluated were similar to those of the fermentors (medium values YP/S ≈ 0.68 g g-1 e QP ≈ 0.28 g L-1 h-1). However, these values were lower than those obtained in Erlenmeyer flasks, maybe due to conditions of oxygen availability for they differ from those in fermentors, since oxygen is the most critical parameter in this bioprocess.

O trabalho abrange o estudo de biorreduções de cetonas por espécies vegetais, empregando-se homogenatos de células de tecidos de plantas já desenvolvidas (folhas) ou culturas de células de Rauwolfia sellowii e Cereus peruvianus em suspensão. Dentre os objetivos principais do trabalho estão: reduzir cetonas-modelo por culturas de células vegetais, avaliando a eficiência e estereosseletividade do processo. Desenvolver um procedimento de triagem por enzimas vegetais solúveis entre diversas espécies obtidas no campus \"Armando Salles de Oliveira\", através da manipulação de variáveis que sabidamente alteram a atividade enzimática. Este tipo de preparação enzimática é utilizado no estudo de rotas biossintéticas, mas o emprego de homogenatos de células vegetais na triagem por novos biocatalisadores é inovador, tendo sido demonstrado seu potencial como ferramenta na seleção de enzimas vegetais para a biotransformação de xenobióticos. A metodologia é simples e confiável, possibilitando o estudo de biotransformações por enzimas diferentes daquelas expressas em culturas de células e oferecendo maior garantia de que a atividade enzimática observada é originária da espécie vegetal em estudo, e não de microorganismos endofíticos, que podem atuar nas biotransformações em que se utilizam partes de plantas desenvolvidas como biocatalisador The work focuses on the study of the bioreduction of ketones by plant species, using homogenates of already developed plant tissues cells (leaves) or of Rauwolfia sellowii and Cereus peruvianus cell cultures suspensions. Among the main objectives of the study are the evaluation of the efficiency and stereoselectivity of the ketone-reduction process in plant cell cultures. It was employed a screening procedure for soluble enzymes from different plant species found on campus \"Armando Salles de Oliveira\", through the manipulation of variables known to be related to the enzyme activity. This type of enzyme preparation has been already reported in studies of biosynthetic routes, but the use of homogenates of plant cells to the screening for new biocatalysts is innovative. In this work we could demonstrate the potential of this approach as a tool in the selection of plant enzymes for the biotransformation of xenobiotics. The methodology is simple and reliable, allows the study of biotransformations by enzymes different from those expressed in cultured cells, and provides greater assurance that the enzymatic activity observed originated in plant species under study, rather than in endophytic microorganisms, which can act in biotransformations that employ parts of developed plants as biocatalyst.

O principal objetivo deste trabalho foi avaliar o impacto de diferentes condições de processo no preço mínimo de venda do etanol produzido via hidrólise enzimática de bagaço da canade- açúcar. Para alcançar este objetivo o trabalho foi separado em três partes, a primeira para a modelagem das reações envolvidas no processo, a segunda para o dimensionamento dos equipamentos principais e a terceira para a avaliação econômica do projeto. Dessa forma, foi realizada a modelagem cinética das reações de pré-tratamento por explosão a vapor do bagaço de cana, da hidrólise enzimática do bagaço pré-tratado e da fermentação de xilose, os modelos obtidos foram utilizados para o dimensionamento de cada sistema reacional. Os dados experimentais para o pré-tratamento, hidrólise enzimática e fermentação de xilose foram obtidos da literatura. Todos os modelos propostos ajustaram adequadamente os dados experimentais de tal forma que os parâmetros cinéticos calculados foram estatisticamente significativos. O sistema de pré-tratamento foi projetado para operar em regime contínuo em reatores tubulares (PFR) e a hidrólise enzimática para operar em regime contínuo em reatores tipo tanque agitado (CSTR) e tubular em série. O sistema CSTR-PFR para a hidrólise enzimática é promissor para a realização desta reação, pois permite obter elevadas conversões em tempos de reação menores que aqueles necessários para os reatores CSTR. Neste sistema é possível alcançada a conversão de 74,3 % da celulose operando o CSTR e o PFR com tempo espacial de 24 h e 72 h, respectivamente. Foi possível avaliar o impacto das condições de pré-tratamento no dimensionamento do sistema de hidrólise enzimática. Além disso, o efeito da concentração inicial de bagaço e enzimas no rendimento de hidrólise enzimática foi avaliado frente aos resultados econômicos. A avaliação dos custos de capital foi realizada via curvas de custo de equipamentos. As diferentes condições de processo estudadas foram comparadas pela determinação do preço mínimo de venda do etanol (PMVE) obtido através da metodologia do lucro do empreendimento (LE). A avaliação econômica do processo mostrou que as condições que podem minimizar o PMVE são: realização do pré-tratamento a 200 °C por 5 min; realização da hidrólise enzimática com concentração inicial de celulose igual a 9,1 % (m/m) ou aproximadamente 17 % (m/m) de bagaço de cana, tempo espacial nos reatores CSTR e PFR de 24 h e 48 h, respectivamente; dosagem de enzimas igual a 40 FPU/g para celulase e 15 U/g para -glicosidase; capacidade de processamento da planta maior que 500 t/dia de bagaço (base seca) e necessariamente a utilizar da fração hemicelulósica para a produção de etanol. Operando nestas condições para uma planta com capacidade para 1000 t/dia o PMVE foi estimado em R$ 5,79 por litro. Dessa forma, este trabalho atingiu seus objetivos fornecendo subsídios para a escolha das condições mais favoráveis para a redução do PMVE, no entanto para a viabilização deste processo de produção de etanol outras configurações de processo devem ser estudas visando a redução dos custos de investimento e produção. The main objective of this study was to evaluate the impact of different process conditions on the minimum selling price of ethanol via enzymatic hydrolysis of sugarcane bagasse. To achieve this objective the work was separated into three parts, the first to model the reactions involved in the process, the second for the design of major equipment and the third for the economic evaluation of the project. Thus, we performed kinetic modeling of reactions pretreatment by steam explosion of bagasse, the enzymatic hydrolysis of pretreated bagasse and fermentation of xylose, the obtained models were used for the design of each reaction system. The experimental data for the pretreatment, enzymatic hydrolysis and fermentation of xylose were obtained from literature. All the proposed models adequately fitted the experimental data so that the kinetic parameters calculated were statistically significant. The system of pretreatment is designed to operate under a continuous tubular reactor (PFR) and enzymatic hydrolysis to operate in a continuous stirred tank reactor (CSTR) and tubular in series. The CSTR-PFR system for the enzymatic hydrolysis is promising to carry out this reaction, because it offers high conversions in shorter reaction times than those required for CSTR reactors. This system can achieved 74.3% conversion of cellulose operating the CSTR and PFR with space-time 24 h and 72 h, respectively. It was possible to assess the impact of the conditions of pretreatment in the design system of enzymatic hydrolysis. Furthermore, the effect of initial concentration of bagasse and enzymes on yield of enzymatic hydrolysis was evaluated in the face of economic results. The evaluation of capital costs was done through cost curves of equipment. The different process conditions studied were compared by determining the minimum ethanol selling price (MESP) obtained through the methodology of the venture profit (VP). The economic evaluation process showed that the conditions that can minimize the MESP are: completion of pre-treatment at 200 ° C for 5 min; completion of the enzymatic hydrolysis with initial concentration of cellulose of 9.1% (w / w) or about 17% (w / w) of bagasse, time in the reactors CSTR and PFR 24 h and 48 h, respectively, enzyme dosage of 40 FPU / g for cellulase and 15 U / g for -glucosidase; capacity processing plant more than 500 tonnes per day of bagasse (dry basis) and necessarily to use the hemicellulose fraction to ethanol production. Operating in these conditions for a plant with a capacity of 1000 t / day, the MESP was estimated at R$ 5.79 per liter. Thus, this work achieved its objectives by providing subsidies to select the most favorable conditions for the reduction of MESP, but for the viability of this process of ethanol production process of other settings should be studied in order to reduce investment costs and production.

Atualmente, a bioprodução de etanol a partir de materiais lignocelulósicos como o bagaço de cana é de grande interesse nacional e cientifico. Para utilização destes materiais em processos biotecnológicos procedeu-se sua hidrólise em função de sua constituição complexa em celulose, hemicelulose e lignina. Após hidrólise ácida branda obteve-se um hidrolisado hemicelulósico rico em xilose e outros açúcares juntamente com compostos tóxicos a atividade microbiana como ácido acético, furfural, hidroximetilfurfural e compostos fenólicos. Este trabalho avaliou a necessidade da suplementação nutricional bem como a concentração inicial de xilose e células no hidrolisado hemicelulósico de bagaço de cana-de-açúcar sobre os parâmetros fermentativos de sua bioconversão a etanol pela levedura Pichia stipitis UFMG-IMH 43.2. Os ensaios foram realizados de acordo com um planejamento fatorial completo 23 com face centrada tendo como resposta o fator de conversão de açúcares em etanol (YP/S). Na suplementação do hidrolisado avaliou-se a adição dos seguintes nutrientes: MgSO4 (0 a 1,0 g/L), extrato de levedura (0 a 5,0 g/L) e uréia (0 a 5,0 g/L). Neste estudo observou-se que o extrato de levedura (5,0 g/L) exerceu maior influência sobre o processo fermentativo alcançando máximos valores de YP/S (0,13 g/L) e produtividade volumétrica em etanol (0,07 g/L.h) não necessitando da adição dos demais nutrientes. Ao se avaliar a concentração inicial de xilose (30; 52,5; 75 g/L) bem como a concentração celular inicial (0,5; 1,0; 2,0 g/L) no hidrolisado observou-se uma melhoria significativa no valor de YP/S (0,19 g/g) utilizando máxima concentração celular inicial (2 g/L) associada com menor concentração de xilose inicial (30 g/L). Estes resultados indicaram que a levedura Pichia stipitis UFMG-IMH 43.2 favoreceu a conversão em etanol YP/S (0,19 g/g), mostrando que a levedura teve melhor comportamento a baixas concentrações de xilose. Este estudo demonstrou que a levedura P. stipitis UFMG-IMH 43.2 mostrou-se como uma alternativa viável para a produção de bioetanol em hidrolisado hemicelulósico de bagaço de cana-de-açúcar. Currently, ethanol bioproduction from lignocellulosic materials such as sugarcane bagasse is of great national and scientific interest. For utilization of these materials in biotechnological processes according mild acid hydrolysis is required to complex constitution of cellulose, hemicellulose and lignin. After mild acid hydrolysis, a hemicellulosic hydrolyzate rich in xylose and other sugars together with toxic compounds to microbial activity as acetic acid, furfural, hydroxymethylfurfural and phenolics was obtained. The studies on the hydrolysate supplementation evaluated the nutrients addition: MgSO4 (0 to 1.0 g/L), yeast extract (0 to 5.0 g/L) and urea (0 to 5.0 g/L) and effects of initial xylose and cell concentration for ethanol production by Pichia stipitis UFMG-IMH 43.2 yeast were evaluated. The experiments were performed according to factorial design central composite face centered 23 using YP/S as response factor. This study showed that only the addition of 5.0 g/L yeast extract was enough to obtained YP/S equal to 0.13 g/L and productivity of 0.07 g/L-h. The evaluation of initial xylose concentration (30; 52,5; 75 g/L) and initial cell concentration (0,5; 1,0; 2;0 g/L) showed a significant influence on fermentation parameter YP/S. The best conditions were achieved using initial cell concentration of 2.0 g/L and initial xylose concentration of 30 g/L with YP/S equal to 0.19 g/g, showing that this yeast strain had better performance at low xylose concentrations. This study demonstrated that P. stipitis UFMG-IMH 43.2 yeast was a promising alternative for bioethanol production from sugarcane bagasse hemicellulosic hydrolysate.

Os polihidroxialcanoatos (PHAs) são poliésteres acumulados por microrganismos como material de reserva. O conhecimento das vias bioquímicas e enzimas envolvidas na biossíntese e degradação dos PHAs é uma importante ferramenta para auxiliar na produção industrial. A linhagem Burkholderia cepacia IPT64 é capaz de acumular uma blenda composta de P(3HB) e P(3H4PE) a partir de sacarose. Este trabalho está focado em duas das principais enzimas envolvidas na biossíntese de PHAs: a b-cetotiolase (phaA) e a PHA sintase (phaC). A primeira está associada à especificidade pelo substrato, e a segunda é considerada a enzima chave na síntese de PHAs. Neste trabalho a linhagem mutante phaC foi avaliada quanto à atividade enzimática de PHB sintase, que se constatou ter sido perdida. A presença de mais de uma tiolase no genoma de B. cepacia foi detectada. A inativação do gene phaABc identificado anteriormente, bloqueou totalmente a síntese de P(3HB), e não promoveu o aumento da quantidade total de polímero. Este resultado indica que a tiolase identificada é responsável direta do acúmulo de P(3HB). Outra indicação é que não há uma competição das vias de síntese dos dois polímeros P(3HB) e P(3H4PE), já que não houve alteração na quantidade de P(3H4PE) acumulado, mesmo quando P(3HB) deixou de ser acumulado. The polyhydroxyalkanoates (PHAs) are polyesters accumulated by microorganisms as storage compounds. Knowing the biochemistry pathway and enzymes involved in the biosynthesis and degradation of PHAs is an important tool to help industrial production. The Burkholderia cepacia IPT64 strain is able to accumulate a blend of P(3HB) and P(3H4PE) from sucrose. The focus of this work is on the two main enzymes involved in PHA biosynthesis: the b-ketothiolase (phaA) and the PHA synthase (phaC). The first one is associated with substrate specificity, and the second one is considered the key enzyme in PHA synthesis. In this work a mutant strain phaC was evaluated on its PHB synthase enzymatic activity, that was discovered to have been lost. The presence of other thiolases in the B. cepacia genome was detected. The inactivation of phaABc gene identified previously, blocked totally the P(3HB) synthesis, and didnt increase the polymer content. This result indicates that the identified thiolase is directly responsible for P(3HB) accumulation. Another indication is that the synthesis pathways of the two polymers, P(3HB) and P(3H4PE), dont compete with each other, because the content of P(3H4PE) was not altered, even when the P(3HB) was not accumulated.

A glicose 6-fosfato desidrogenase (G6PDH) é a primeira enzima do processo oxidativo da via das pentoses fosfato e apresenta diversas aplicações industriais. Foram avaliadas as influências de algumas variáveis sobre a produção de G6PDH. No Processo Descontínuo variaram-se a concentração da fonte de carbono, relação carbono-nitrogênio, concentração de aminoácidos e nucleotídeos , pH e a vazão de ar fornecida. A variação na atividade enzimática indicou que as condições do meio são importantes na obtenção desta enzima, tendo sido obtida atividade específica máxima de 86 U/g cél e produtividade de 10,5 U/L.h. No Processo Descontínuo-Alimentado variaram-se o sistema de adição e os nutrientes alimentados. A atividade específica máxima obtida foi 72 U/g cél e a produtividade foi 47 U/L.h. Os resultados indicaram o controle da concentração de glicose em 5g/L para uma melhor produtividade e a necessidade de maiores estudos a fim de otimizar o processo. The glucose 6-phosphate dehydrogenase (G6PDH) is the first enzyme of the pentose phosphate pathway, converting glucose-6-phosphate into 6-phosphogluconate. Besides its importance in biochemistry and medical studies, this enzyme is used in several analytical methods, industrial and commercial application. In this work the influence of several variables in the production of Glucose-6-Phosphate Dehydrogenase in batch and fed-batch processes were studied. In batch cultures the variables were the glucose concentration (10 and 20 g/L), the C:N ratio (3.5 and 6.7 g/g), the aminoacids and nucleotides concentrations (10 and 20 mg/L), pH (4.6 and 5.7) and the aeration (0, 0.8, 1.7 and 2.2 vvm). It was observed that the G6PDH activity varied according to the conditions of the culture. Specific Activity value of 86 U/g cell and productivity of 10,5 U/L.h were attained when the test was carried out as follows: 30oC, pH 5.7, 400 rpm, 2.2 vvm, C:N ratio of 6.7, glucose concentration of 10 g/L, aminoacids (Tryptophan and Hystidine) and nucleotides (Adenine and Uracil) concentrations of 20 mg/L. Nevertheless, the fed-batch process was more efficient and productive than the batch process. The productivity obtained in the best fed-batch condition (glucose addition by an increasing exponential mode) was 47 U/L.h, more than four folds the productivity of the batch culture. So that, by maintaining the glucose concentration in the medium below 5 g/L, the productivity should be improved. However, more studies are needed for optimizing the G6PDH production in a Fed-Batch process. In the fed-batch cultures the feeding conditions and kind of feeding were studied. It was observed that the best fed-batch G6PDH specific activity (72 U/g of cell when the glucose was added in a decreasing linear mode) was lower than that attained in the batch cultures.

A transferência de oxigênio para o meio, durante o cultivo de Candida kefyr DSM 70106, mostrou ter grandE influência na produção de inulinase extracelular. O KLa (coeficiente volumétrico de transferência de oxigênio) de 43 h-1 foi o que proporcionou a obtenção de atividade enzimática mais elevada (37,5 U.mL-1). Este parâmetro foi utilizado como critério para ampliação de escala do cultivo em biorreator de 15 litros para 300 litros. O volume de meio obtido foi congelado para utilização nos processos seguintes de separação de células e purificação da inulinase. Para isto foram utilizados três processos diferentes, a saber, filtração (microfiltração, diafiltração e ultrafiltração), extração líquido-líquido por micela reversa e cromatografia de troca iônica em leito fluidizado. Durante a separação de células, a membrana de microfiltração tipo HVLP com 0,45 µm de diâmetro de poro foi a que possibilitou a passagem de maior fluxo de filtrado (324 L.h-1.m-2) quando comparada com outras membranas de poros e materiais diferentes. Associado a este fluxo obteve-se ainda a passagem pela membrana de 99% da enzima presente. Foram também estudados diferentes tipos de módulos de filtração. Os filtros rotativos foram os que apresentaram melhores desempenhos, com destaque para o tipo \"Biodruckfilter\". Fluxo de filtrado de 300 L.h-1.m-2 e transmissão de 99% da enzima foram obtidos neste filtro. Os filtros tipo cassete e os tubulares apresentaram desempenhos, em geral, inferior a 50% quando comparados em termos de fluxo de filtrado. O processo de filtração em escala ampliada, realizada no filtro cassete, apresentou fluxo de filtrado 45% inferior ao obtido na pequena escala. Os estudos de extração líquido-líquido por micela reversa foram feitos utilizando um agente tensoativo catiônico (BDBAC- [N-Benzyl-N-Dodecyl-N bis(2-hydroxyethyl) ammonium chloride] e um aniônico (AOT- sodium-di-2ethyl-hexyl-sulphosuccinate). O BDBAC proporcionou recuperação de 91 % da enzima presente inicialmente. Na ampliação de escala do processo foi verificado um rendimento de 77%, e o aumento da atividade enzimática específica, nos dois casos, foi de aproximadamente 2,8 vezes. Em comparação com os dois processos anteriores, o processo de purificação da inulinase com separação simultânea de células por cromatografia de trocaiônica em leito fluidizado foi o que proporcionou maior aumento da atividade enzimática específica (da ordem de 4,3 vezes). Após adsorção e eluição do leito cromatográfico, 93,1% da enzima foi recuperada. It has been observed that the production of extracellular inulinase by Candida kefyr DSM 70106 was markedly affected by the rate of oxygen transfer to the medium. The highest inulinase activity (37,5 U.Ml-1) was attained at a KLa value (volumetric coefficient of oxygen transfer) equal to 43 h-1. As such, KLa was chosen as a criterion for scaling-up the bioreactor capacity from 15 L to 300 L. Once the fermentation ended, the whole fermented broth was frozen for further cell separation and inulinase purification. For that, three different processes were used, namely filtration (microfiltration, diafiltration and ultrafiltration), liquid-liquid extraction by reversed micelles and ion-exchange chromatography in fluidized bed. Microfiltration membranes differing in the chemical nature and on the pores diameter were used in cell separations. The highest filtrate flow (FF = 324 L.h-1.m-2) occurred when HVLP type membrane (0,45 µm pore diameter) was employed. Moreover, 99% of inulinase passed through the membrane. Several types of filtration modules were also studied. The rotatory filter, such as the \"Biodruckfilter\" (FF=300 L.h-1.m-2 and 99% of inulinase transmission), presented remarkable performance when compared with cassette and tubulartype filters, in which FF was lower than 50%. Furthermore, filtration carried out with a scaled-up cassette module presented a FF 45% lower than that observed on a smaller scale. Studies on liquid-liquid extraction by reverse micelles were made utilizing a cationic surfactant (BDBAC- [N-Benzyl-N-Dodecyl-N-bis(2-hydroxyethyl) ammonium chloride] and an anionic surfactant (AOT-sodium-di-2-ethyl-hexylsulphosuccinate). The use of BDBAC resulted in a recuperation of 91 % of the enzyme initially present, which decreased about 77% when the process was scaled-up. Neverthless, in both cases the increase in specific enzymatic activity was about 2.8 times. In comparison with the aforementioned processes, the ion-exchange chromatography in fluidized bed, in which inulinase purification and cell separation take place simultaneously, provided. an increase in specific enzymatic activity of about 4.3 times. After adsorption and elution of the chromatographic bed about 93% of the inulinase was recovered.

O biodiesel pode ser obtido a partir de óleos vegetais, gorduras animais ou óleos residuais por meio de reação com etanol ou metanol e destaca-se entre as fontes renováveis de energia apontadas como solução para aumentar a segurança no suprimento de energia e minimizar alguns problemas ambientais decorrentes do uso de combustíveis derivados do petróleo.Torna-se necessário então, avaliar as propriedades dos diferentes tipos de biodiesel obtidos a partir de diferentes matérias-primas e processos que convergem ou divergem das especificações requeridas, quais são as desvantagens do ponto de vista ambiental e de desempenho do motor e suas possíveis soluções. Este trabalho teve por objetivo estudar os aspectos técnicos e ambientais relacionados ao seu uso em motores de combustão. The Biodiesel can be produced from vegetal oils, animal fats or residual oils by means of reaction with ethanol or methanol and is detached among the renewable sources of energy as a solution to increase the security in the energy suppliment and to minimize some decurrent environment problems from the use of fossie fuels. Thus, it?s necessary to evaluate the trends for the specifications of fuels, which properties of biodiesel converge or not of the required specifications, which are the disadvantages of the environment and performance engine and its possible solutions. The object of this work had been to study the technical and environment aspects related about its use in combustion engine.
