Arinosaster patriciae gen. nov. et sp. nov. is the second continental sponge registered for a subterranean environment (cave habitat) in Brazil and the Neotropical Region. The sponges were recorded and collected in a 5m depth technical dive in a sinkhole of Rio Claro, tributary of Rio Arinos, Tapajós system, Amazon Basin (-13.8170386, -56.6914225) at the locality of Sumidouro do Rio Claro, Municipality of Diamantino, state of Mato Grosso, central western Brazil. The cave is placed in sandstone rocks of the Parecis Group (Upper Cretaceous). “In situ” photographs of colonies, of living specimens, SEM illustrations of dissociated spicules as well as of the skeletal structure, are presented. The occurrence of euaster microscleres of the type spherasters are for the first time reported for continental sponges but, also new, is the occurrence of spongin fibers, composing with fibers of silicious spicules in the skeletal arrangement. The absence of gemmules in the studied material and the fact that new specimens remain undetected call for the proposition of a new monospecific genus to be retained as Incertae Sedis until new and gemmuliferous colonies are found.

The characid genus Oligosarcus consists of 20 described species distributed throughout most of South American river basins below 14º south latitude. This study focus on the phylogenetic relationships of the species of Oligosarcus based on the analysis of osteological characters to provide data to discuss the biogeographic history of the genus. The analysis resulted in a single most parsimonious tree with 152 steps (CI= 0.355 and RI= 0.600). The 18 included Oligosarcus species were hierarchically organized into 17 clades. A minimal age of 15 Ma for the genus is suggested based on the putatively cladogenetic event represented by the continued shortening of the Eastern Cordillera that established the eastern boundary of the modern central Andean plateau and was responsible for cladogenesis between the common ancestor of O. schindleri + O. bolivianus versus the remaining congeners. There is a pronounced disjunction in the upland species distribution by the lowland areas of the Chaco-Pantanal basin. This indicates that upland habitats (headwater streams) are preferential habitats for a set of species including O. argenteus, O. bolivianus, O. brevioris, O. paranensis, O. perdido, O. pintoi, O. planaltinae, O. brevioris, and O. schindleri. Fragmentation of populations of O. pintoi and O. perdido are at least 2.5 Ma old, since the origin of the upper Paraguay depression clearly promoted the present-day observed disjunction in the distribution of these species. The lowland Oligosarcus species are all included in a single clade but the obtained results suggest that fragmentation of upland versus lowland components of the genus was not causally related to a single vicariant event. The available fossil record of Oligosarcus indicates that the genus already occurred in the coastal plain at about 2.3 to 1.25 Ma. The sister-group relationship between O. hepsetus and O. longirostris provided additional evidences of the so-called historical affinities between the SE Brazilian coastal plain and the Rio Iguaçu drainage basin.

Microglanis cottoides é amplamente distribuída em bacias hidrográficas ao longo da costa Atlântica brasileira que estão isoladas entre si, bem como das grandes drenagens do interior, pelas montanhas escarpadas da margem oriental do escudo cristalino brasileiro, conferindo a esta espécie qualidades apropriadas para estudos filogeográficos. Além disso, existe a hipótese de ocorrência de espécies crípticas e sinônimos em M. cottoides, tornando o estudo desta espécie de grande importância para a sistemática e o entendimento da evolução deste grupo de peixes. Identificação e descrição de espécies, tradicionalmente, são feitas com base em caracteres morfológicos, mas na atualidade ferramentas moleculares têm sido utilizadas, em conjunto com a morfologia, para o estudo da diversidade biológica. A proposta deste trabalho foi (1) testar as relações filogenéticas entre diferentes populações de M. cottoides e (2) analisar a estrutura genética, demografia histórica e padrões temporais de divergência nas populações de M. cottoides, ao longo de sua área de distribuição, além da relação filogenética entre seus congêneres. As relações filogenéticas e filogeográficas, a estrutura genética e a demografia histórica foram analisadas utilizando uma combinação de marcadores morfológicos e moleculares (microssatélites e DNAmt-COI). A alta divergência genética encontrada entre os indivíduos da bacia do rio Uruguai e demais bacias costeiras sugere que a espécie designada como M. cottoides para a bacia do rio Uruguai, seja uma espécie nova de Microglanis que se enquadra no critério de espécie críptica. Por outro lado, as populações de M. cibelae apresentaram baixas divergências genéticas entre M. cottoides das demais bacias costeiras, o que é compatível para indivíduos de uma mesma espécie. Assim, M. cottoides, como atualmente diagnosticado, forma um grupo não monofilético, pois inclui uma espécie morfológica e geneticamente distinta da bacia do rio Uruguai e exclui uma espécie morfológica e geneticamente similar das bacias dos rios Tramandaí e Mampituba (M. cibelae). Para as análises filogeográficas, a distribuição de M. cottoides restringiu-se às drenagens costeiras, considerando as populações das bacias dos rios Tramandaí e Mampituba, previamente identificadas como M. cibelae, como pertencentes a esta espécie. Os níveis de variação genética observados em M. cottoides estão dentro do esperado quando comparados com outras espécies de Siluriformes e os loci investigados mostraram um alto nível de polimorfismo. De forma geral, a diversidade genética encontrada nas populações de M. cottoides foi alta e semelhante ao que geralmente é encontrada em populações de peixes selvagens. No entanto, observou-se forte estruturação genética principalmente entre as amostras de Paranaguá, Guaratuba e Itapocu e menor estruturação entre Araranguá e Mampituba, e entre Tramandaí e Patos, indicando maior possibilidade de fluxo gênico entre essas populações e, consequentemente, maior similaridade entre elas. Contudo, considerando a distância geográfica e principalmente o isolamento das bacias costeiras que, provavelmente, ocorreu entre 6.000 a 10.000 anos antes do presente, a similaridade entre as populações sugere fluxos gênicos históricos, refletindo um período em que as bacias estavam conectadas. Paleocanais fluviais presentes na plataforma continental podem indicar que estas drenagens tinham comunicação com outras drenagens de bacias próximas, permitindo a ampla distribuição de M. cottoides em drenagens que hoje estão isoladas. Entretanto, nota-se que as divergências genéticas entre os indivíduos das bacias costeiras aumentam com a distância, e observa-se que entre o sul de Santa Catarina e o norte do Rio Grande do Sul há uma quebra filogeográfica de cerca de 1,9 Ma, causando a divisão das populações de M. cottoides em dois filogrupos. As análises filogeográficas e filogenéticas demonstraram que as espécies litorâneas, M. parahybae e M. cottoides, são filogeneticamente mais relacionadas e divergiram-se há aproximadamente 5 Ma. Já M. garavelloi da bacia do Alto Paraná e Microglanis sp. da bacia do rio Uruguai divergiram dessas espécies do litoral há pelo menos 8 Ma, coincidindo com a época de diversificação da maior parte da ictiofauna Neotropical, que ocorreu entre 3 e 10 Ma. Portanto, tudo indica que a história filogeográfica de M. cottoides envolveu eventos vicariantes de origem tectônica como a formação das drenagens costeiras a partir de episódios orogenéticos e, mais recentemente, por eventos climáticos pleistocênicos que isolaram várias populações. Porém, em períodos de glaciação, quando as drenagens voltam a se encontrar, as populações de M. cottoides possivelmente trocam informações genéticas e por isso mantêm sua unidade específica. Microglanis cottoides is widely distributed in basins along the Brazilian Atlantic coast that are isolated from each other and from major drainages of the interior, by rugged mountains of the east bank of the Brazilian crystalline shield, giving to this species appropriate qualities for phylogeographic studies. Furthermore, there is a chance of occurrence of cryptic species and synonyms in M. cottoides, making this kind of study of great importance for the systematic and for understanding of the evolution of this group of fish. Identification and description of species are traditionally made based on morphological characteristics, but currently molecular tools have been used in conjunction with morphology for the study of biological diversity. The purpose of this study was (1) to test the phylogenetic relationships between different populations of M. cottoides and (2) to analyze the genetic structure, historical demography and temporal patterns of divergence among populations of M. cottoides, throughout its distribution area in addition to the phylogenetic relationship among congeners. The phylogenetic and phylogeographic relationships, genetic structure and the historical demographics were analyzed using a combination of morphological and molecular markers (microsatellites and mtDNA-COI). The high genetic divergence found among specimens of the Uruguay River basin and other coastal basins suggests that the species identified as M. cottoides from the Uruguay River basin, is a new species of Microglanis that fits in the criterion of cryptic species. On the other hand, populations of M. cibelae showed low genetic differences between M. cottoides from remaing coastal basins, which is compatible for individuals of the same species. Thus, M. cottoides, as currently diagnosed, form a non-monophyletic group because it includes a morphologically and genetically distinct species from the Uruguay river basin and excludes a morphological species and genetically similar species from the Tramandaí and Mampituba rivers basins (M. cibelae). For phylogeographical analysis, the distribution of M. cottoides was restricted to the coastal streams, considering the populations of Tramandaí and Mampituba basins, previously identified as M. cibelae, as belonging to this species. The levels of genetic variation observed in M. cottoides are as expected when compared to other species of Siluriformes and the investigated loci showed a high level of polymorphism. Overall, the genetic diversity found in populations of M. cottoides was high and similar to what is usually found in wild fish populations. However, there was a strong genetic structure mainly between samples of Paranaguá, Guaratuba and Itapocu and lower structure between Araranguá and Mampituba, and between Tramandaí and Patos, indicating a higher possibility of gene flow between these populations, and consequently, greater similarity between them. However, given the geographical distance and especially the isolation of coastal basins that probably occurred between 6,000 to 10,000 years before present, the similarity between the populations suggests historic gene flows, reflecting a time when the basins were connected. River paleochannels present on the continental platform may indicate that these drainages had communication with other drainages of nearby basins, allowing widespread distribution of M. cottoides in drainage that are isolated today. However, it notes that the genetic differences between individuals of coastal watersheds increase with the distance, and it is observed that between the south of Santa Catarina and northern Rio Grande do Sul there is a phylogeographic break of about 1.4 My, causing the division of populations of M. cottoides in two phylogroups. The phylogeographic and phylogenetic analysis showed that the coastal species, M. parahybae and M. cottoides are phylogenetically more related and diverged at about 3 My. Since M. garavelloi from Alto Paraná basin and Microglanis sp. from the rio Uruguay basin diverged from these coastal species for at least 5 My, coinciding with the time of diversification of most of the Neotropical fish fauna, which occurred between 3 and10 My. Therefore, it seems that the phylogeographic history of M. cottoides involves vicariant events of tectonic origin like the formation of coastal drainages from orogenic events and, more recently, by Pleistocene climatic events that isolated various populations. However, in glaciation periods, when the drainage meet again, populations of M. cottoides possibly exchange genetic information and therefore maintain their specific unit.

O continente sul-americano é a única região biogeográfica onde podemos encontrar um grupo estenohalino de elasmobrânquios: as arraias da família Potamotrygonidae. Entre 19 e 21 espécies de potamotrigonídeos estão taxonomicamente organizadas em três gêneros, distribuídas pelas bacias dos rios Atrato, Magdalena, Maracaibo, Orinoco, Essequibo, Amazonas e do Prata, os quais desembocam no Oceano Atlântico e no Mar do Caribe. Diversos cenários biogeográficos postulados para explicar a presença de linhagens predominantemente marinhas nos sistemas fluviais da América do sul foram propostos. O mais aceito para a origem e diversificação de Potamotrygonidae é a de invasão de um ancestral caribenho no sistema fluvial sul-americano durante as ingressões marinhas do Mioceno inferior no noroeste do continente, seguido de isolamento por alteração dos padrões de drenagens do Orinoco e formação dos Andes. A fauna parasitária de Potamotrygonidae está representada predominantemente por membros de Cercomeromorpha, um clado composto por linhagens de platelmintos membros de Cestoda e Monogenoidea. Dois gêneros de monogenóideos parasitas branquiais podem ser encontrados em potamotrigonídeos: Potamotrygonocotyle (Monocotylidae), para o qual 12 espécies são conhecidas, e a monotípica Paraheteronchocotyle amazonense (Hexabothriidae). O objetivo desta tese é propor uma hipótese de relacionamento filogenético baseada em dados moleculares para as espécies de Potamotrygonocotyle, e discutir padrões de evolução da espécie P. aramasae e seu hospedeiro Paratrygon aiereba, distribuídos em diferentes rios amazônicos. Sequências nucleotídicas do gene mitocondrial cox1 e nuclear ITS1 foram utilizadas simultaneamente em uma busca de uma hipótese filogenética para todas as espécies conhecidas de Potamotrygonocotyle. Os resultados recuperaram o status monofilético de cinco espécies, sugeriram complexos de linhagens crípticas para três espécies nominais e revelaram relacionamentos de grupo-irmão não detectados na hipótese filogenética mais recente para o gênero, baseada em dados morfológicos. Adicionalmente, os resultados apresentados suportam decisões taxonômicas recentes envolvendo sinonímias entre espécie nominais de Potamotrygonocotyle. Finalmente, os relacionamentos filogenéticos de haplótipos de diferentes populações de P. aramasae e seu hospedeiro P. aiereba foram acessados pela análise cladística de múltiplos marcadores mitocondriais, com o objetivo de se detectar possíveis padrões filogeográficos e testá-los sob predições filogenéticas de três hipóteses de diversificação amazônica (Hidrogeológica, de Museu e Paleogeográfica). Sequências dos genes ribossomal 16S e citocromo oxidase 1 foram obtidas para espécimes de parasitas de seis sub-bacias amazônicas. Os hospedeiros foram amostrados em 10 sub-bacias amazônicas e na sub-bacia do rio Essequibo, para os quais foram obtidas sequências nucleotídicas dos genes mitocondriais citocromo oxidase 1, citocromo b e ATPase. Os resultados sugerem a ausência de padrões filogeográficos para o parasita, cujos resultados indicam um modelo de alta dispersão. Em contraste, P. aireba é caracterizada por populações bem estruturadas, de acordo com as sub-bacias amostradas. O padrão filogeográfico geral desta espécie é concordante com as predições da Hipótese de Museu (linhagens mais antigas em rios dos escudos pré-cambrianos; linhagens apicais na bacia sedimentar amazônica). South America is the only biogeographical region in the world where is possible to find an exclusive stenohalin group of elasmobranchs: the Potamotrygonidae stingrays. Between 19 and 21 species of potamotrygonid stingrays are taxonomically organized in three genera, distributed in the Atrato, Magdalena, Maracaibo, Orinoco, Essequibo, Amazonas e la Plata river Basins, discharging in both Atlantic and Caribbean waters. Distinct biogeographical scenarios have been proposed to explain the presence of predominantly marine lineages in the south American fluvial systems. The most accepted theory for Potamotrygonidae is the invasion of a Caribbean ancestor in the south-American freshwater system during the marine ingression of the early Miocene in the northwest of the continent, followed by its isolation due alterations of Orinoco rivers drainage patterns, as well as formation of Andes. The parasitic fauna of potamotrygonids is predominantly represented by members of Cercomeromorpha, a clade composed by members of Cestoda and Monogenoidea. Two genera of gill parasites monogenoidean might be found in potamotrygonids: Potamotrygonocotyle (Monocotylidae), for which 12 species are currently recognized, and the monotypic Paraheteronchocotyle amazonense (Hexabothriidae). The goal of this study is to propose a phylogenetic hypothesis based in molecular data for members of Potamotrygonocotyle, as well as to discuss evolutionary patterns of the species P. aramasae and its host Paratrygon aiereba, distributed through different amazon rivers. Nucleotide sequences of the mitochondrial gene cox1, and the nuclear gene ITS1 were simultaneously used to infer phylogenetic sister-group relationships among allrecognized species of Potamotrygonocotyle. The results recovered the monophyletic status of five species, suggested cryptic lineages for three species complexes, and revealed sister-groups relationships not detected previously by the phylogenetic analysis of morphological data. Additionally, the results supports recent taxonomic decisions involving synonyms among nominal species within the genus. The phylogenetic relationships of haplotypes from distinct populations of the parasite P. aramasae, as well as from its host Paratrygon aiereba, were accessed by the cladistic analysis of mitochondrial markers, in an attempt of detecting possible phylogeographic patterns, and to test them against phylogenetic predictions of three amazonian bio-diversification hypotheses (Hydrogeology, Museum, Paleogeography). Sequences of the ribosomal 16S gene and cytochrome oxidase 1 were obtained from parasite specimens collected in six amazon sub-basins. The hosts were sampled in 10 amazon sub-basins, as well as in the Essequibo sub-basin, for which cythocrome oxidase 1, cythocrome b, and ATPase sequences were obtained. No clear phylogeographic patterns were revealed for theparasite haplotypes, and the results suggest a high dispersion model. In contrast, P. aiereb is characterized by strongly structured populations, according to the sub-basin sampled. The general phylogeographic pattern recovered for this species is in agreement with the Museum Hypothesis predictions (older lineages in rivers of the pre-cambrian shields; apical lineages inhabiting the sedimentary amazon basin).
