This study aimed to evaluate the influence of sodium monensin on the hepatic accumulation of copper in sheep. Twenty-four Santa Inês crossbred sheep were used and allocated in a 2 × 2 factorial experiment with six repetitions and considering the factors dietary copper (basal and high) and supplementation (with and without sodium monensin). Thus, four homogeneous groups were formed: control (basal diet); monensin (Mon), 30 ppm of monensin; copper (Cu), 10 10 mg/kg BW per day of copper; monensin + copper (MonCu). The experimental period lasted 14 weeks. Liver and bile samples were collected at the beginning and end of the experiment to determine mineral element concentrations, and weekly blood samples for biochemical, hematological, and mineral evaluation. Liver copper concentrations at the beginning of the experiment did not vary between groups, while mean liver copper concentrations at the end of the experiment were higher in the MonCu, Cu, and Mon groups when compared to the control. At the end of the study, hepatic copper concentration was influenced by copper (p = 0.0001) and monensin (p = 0.0003) supplementation. Copper-supplemented groups had reduced liver iron contents (p = 0.0287) and increased copper concentrations in bile. The biochemical evaluation showed increased serum GGT and AST activity (p 0.05) in the Cu and MonCu groups from the eleventh week on compared to the control and Mon groups. The increase in activity of these enzymes was influenced by copper supplementation (p = 0.0340). Monensin interferes positively with the hepatic accumulation of copper and the supplementation of this additive may predispose sheep to copper poisoning.

The effects of feeding, fasting, and re-feeding on the metabolic and ruminal profiles of growing cattle were studied. Blood and urine samples were obtained from 12 crossbred steers weighing approximately 300 kg during the following periods: 11 h of normal feeding (postprandial period), 48 consecutive hours of fasting, followed by 48 h of re-feeding. Compared with the postprandial period, fasting caused the following modifications: moderate hypoglycemia accompanied by remarkable lipolysis detected by the increase in plasma levels of free fatty acids (FFAs); absence of hepatic lipidosis, as there were no changes in aspartate aminotransferase activity or serum cholesterol levels; mild ketogenesis, confirmed by the slight increase of β-hydroxybutyrate (βHB); increased amino acid burn for energy production, verified by the increase in serum urea contents. There were strong positive correlations between the plasma levels of FFAs and βHB (r = 0.68; p < 0.001), fasting duration and FFA concentration (r = 0.92; p < 0.00001), and fasting duration and serum urea (r = 0.52; p < 0.001); there was a negative correlation between fasting duration and blood glucose (r = −0.52; p < 0.0001). During this same period, mild hypovolemia characterized by an increase in intravascular volume deficit was observed. The metabolic condition observed during fasting was completely reversed during re-feeding, except for the temporarily higher proteolysis.

RESUMO Este estudo avaliou a capacidade de um sal mineral rico em molibdênio (Mo) em prevenir a intoxicação cúprica acumulativa (ICA), mediante a análise das variáveis clínicas e dos teores de cobre (Cu) e Mo hepático. Foram utilizados 25 ovinos cruzados da raça Ile-de-France, aleatoriamente distribuídos igualmente em cinco grupos, em que o grupo 1 recebia dieta contendo 80% de volumoso e 20% de concentrado, os grupos 2 e 3 recebiam 50% de volumoso e 50% de concentrado, e os grupos 4 e 5 recebiam a mesma dieta dos grupos 2 e 3, com a adição diária de 150mg de sulfato de Cu. Os grupos 1, 3 e 5 recebiam sal mineral contendo 300ppm de Mo. Foram realizadas três biópsias hepáticas para determinação de Cu, Mo e Zn. Três ovinos do grupo 4 e um do grupo 5 manifestaram ICA. Não houve diferença na frequência de mortalidade entre os grupos (P=0,56). Os teores de Cu hepático nos ovinos com ICA (2450ppm) foram superiores aos que não intoxicaram (1518ppm). Quanto maior a ingestão de Mo na dieta, menor foi o acúmulo de Cu hepático ao término do experimento (r = -0,72). ABSTRACT The aim of this project is to evaluate the capacity of a molybdenum-rich mineral salt in the prevention of cumulative cooper poisoning (CCP) in sheep, through clinical and hepatic copper and molybdenum concentrations. Twenty five crossbreed Ile-de-France sheep were randomly distributed equally into five groups. Group 1 received a 80% forage and 20% concentrate diet, groups 2 and 3 received a 50% forage and 50% concentrate diet, and groups 4 and 5 received the same diet as groups 2 and 3 with a daily supplementation of 150 mg of copper sulfate. Groups 1, 3 and 5 received a mineral salt with 300 ppm of molybdenum. Three times during the experiment a liver biopsy was carried out to evaluate the degree of copper accumulation. Three sheep from group 4 and one sheep from group 5 showed a clinical picture of CCP. There was no difference in the frequency of mortality between groups 4 and 5 (P=0.56). The liver copper concentration was higher in sheep with CCP (2450 ppm) compared to sheep that did not present CCP (1518 ppm). The higher the ingestion of molybdenum in the diet the lower the liver copper concentration at the end of the experiment (r=-0.72).

Objetivou-se conhecer os teores de cobre, zinco, ferro e molibdênio em soro e fígado de pequenos ruminantes criados nos municípios de Casa Nova e Juazeiro, estado da Bahia, e verificar se há carência primária ou secundária de cobre. Foi avaliado um total de 160 amostras de ovinos e caprinos de ambos os sexos, coletados nos períodos seco e chuvoso. O valor de cobre sérico para caprinos foi de 13,8±0,3±mol/L e para ovinos foi de 12,2±0,4±mol/L, enquanto os teores hepáticos médios foram de 220ppm para caprinos e 238ppm para ovinos. O valor de zinco sérico para caprinos foi de 28,3±1,0±mol/L e para ovinos foi de 28,7±0,8±mol/L, enquanto os teores hepáticos médios foram de 99ppm para caprinos e 92ppm para ovinos. O valor de ferro sérico para caprinos foi de 61±3±mol/L e para ovinos foi de 64±2±mol/L, enquanto os teores hepáticos foram de 172ppm para caprinos e 221ppm para ovinos. Os valores médios de cobre não indicam ocorrência de carência deste elemento nos pequenos ruminantes criados nos municípios estudados. Os teores de zinco encontravam-se dentro dos intervalos de normalidade, não sendo verificada a necessidade de suplementação extra deste mineral. Os teores de ferro sérico apresentavam-se bem elevados, contudo sem que houvesse interferência suficiente no metabolismo do cobre de forma a resultar em uma deficiência cúprica. The aim of this research was to know the copper, zinc, iron and molybdenum content in serum and liver of small ruminants raised in the cities of Casa Nova and Juazeiro, state of Bahia, and verify the occurrence of primary or secondary copper deficiency. A total of 160 samples of sheep and goats of both sexes collected in the dry and rainy season were evaluated. The values for serum copper in goats was 13.8±0.3±mol/L and in sheep it was 12.2±0.4 ±mol/L, while the mean liver concentration was 220 ppm for goats and 238 ppm for sheep. The serum zinc value for goats was 28.3±1.0±mol/L and for sheep it was 28.7±0.8±mol/L, while the mean liver concentration was 99 ppm for goats and 92 ppm for sheep. Serum iron value for goats was 61±3±mol/L and for sheep it was 64±2±mol/L, while liver levels were 172 ppm for goats and 221 ppm for sheep. Copper mean values do not indicate the occurrence of copper deficiency in small ruminants raised in the cities studied. Zinc levels were within the normal values and do not require extra supplementation of this mineral. The serum iron levels were elevated, however it did not interfere in copper metabolism in order to result in a cupric deficiency.

The aim of this study was to evaluate whether zinc (Zn) supplementation protects against hepatic copper (Cu) accumulation in copper-loaded sheep. Forty cross-bred lambs were assigned to five experimental groups. These included the control group (C) and four treatment groups that received Cu and/or Zn supplementation (dry matter (DM) basis) over 14 weeks, as follows: Cu (450 mg Cu/kg), Zn-35 (450 mg Cu + 35 mg Zn/kg), Zn-150 (450 mg Cu + 150 mg Zn/kg), and Zn-300 (450 mg Cu + 300 mg Zn/kg). Blood, liver, and bile samples were obtained for mineral determination by inductively coupled plasma optical emission spectrometry (ICP&ndash, OES). The hepatic metallothionein (MT) concentrations were also determined. At the end of the experiment, hepatic Cu concentrations were higher in all Cu-supplemented groups than in C. Hepatic Cu accumulation was lower in the groups receiving the Zn supplementation than in the Cu group, although the difference was only statistically significant (66%) in the Zn-300 group. The MT concentrations tended to be higher (almost two-fold) in the Zn groups (but were not dose related) than in the C and Cu groups, and they were related to hepatic Zn concentrations. Zn supplementation at 300 mg/kg DM is useful for preventing excessive hepatic Cu accumulation in sheep exposed to high dietary concentrations of Cu.

RESUMO Objetivou-se avaliar clinicamente os efeitos da administração intrarruminal de duas quantidades distintas de manga em ovinos. Foram utilizados sete ovinos machos, hígidos, que não receberam carboidratos não fibrosos por, pelo menos, seis meses previamente ao período experimental, quando se avaliou pH ruminal, total de protozoários no suco de rúmen, pH urinário, pH sanguíneo estimado e parâmetros vitais nos tempos zero, 12, 16, 20 e 24 após a administração da manga. Os sete ovinos foram distribuídos aleatoriamente em dois grupos e receberam 0,625% ou 1,875% da MS de manga/kg/PV (M1 e M2, respectivamente), distribuídos em delineamento cross-over, com uma parcela perdida, e 30 dias de washout. Os ovinos M2 apresentaram valores médios para pH ruminal significativamente inferiores a M1 a partir do tempo T12 (5,1 e 6,9, respectivamente), o que indica ocorrência de acidose ruminal. Os animais tratados com M1 não apresentaram alterações sistêmicas, ao passo que os ovinos tratados com M2 apresentaram acidose metabólica leve, detectada por meio do pH urinário ácido (4,8). A ingestão apenas de manga in natura na quantidade de 1,875% da MS de manga/kg/PV mostrou-se capaz de provocar acidose ruminal em ovinos, levando-os a: ligeira depressão, aumento da frequência cardíaca, diminuição dos movimentos ruminais e diarreia em alguns casos. ABSTRACT The aim of this study is to clinically evaluate the effects of intra ruminal administration of two different amounts of mango in sheep. The sample was of seven male healthy sheep, which did not receive non-fibrous carbohydrates for at least six months prior to the trial. Rumen fluid pH, total of protozoa in the rumen fluid, urine pH, estimated blood pH, and vital parameters were evaluated at the following times: zero, 12, 16, 20 and 24 hours after administration of the mango. The seven sheep were randomly divided into two groups and received either 0.625% or 1.875% of the dry matter of the pulp and mango peel per kg body weight (M1 and M2, respectively), distributed in cross-over design with a lost portion and 30 days washout. Sheep treated with M2 showed significantly lower average values for ruminal pH than the M1 since T12 (5.1 and 6.9, respectively), indicating the occurrence of ruminal acidosis. The animals treated with M1 showed no systemic changes, while the sheep treated with M2 had mild metabolic acidosis, detected through the lower urinary pH (4.8). The in natura mango ingestion in the amount of 1.875% of the dry matter of mango per kg of body weight proved to provoke rumen acidosis in sheep, leading these animals to: slight depression, rise in heart rate, diminished rumen movement, and diarrhea in some cases.

RESUMO: Este estudo objetivou determinar a soroprevalência da Babesiose e Anaplasmose em bovinos dos municípios de Ouricuri e Petrolina, estado de Pernambuco, Brasil; e definir os possíveis fatores de risco para a ocorrência dessas doenças. Amostras de sangue foram coletadas para realização de teste sorológico por Imunofluorescência Indireta (RIFI). Questionários epidemiológicos sanitários foram aplicados aos produtores com o objetivo de identificar possíveis fatores de risco. Carrapatos foram coletados, identificados e testados por Reação em Cadeia da Polimerase (PCR) para o diagnóstico da infecção por Anaplasma marginale, Babesia bigemina e Babaesia bovis. O estudo foi conduzido com 861 bovinos, sendo 468 de Petrolina e 393 de Ouricuri. A soroprevalência de A. marginale, B. bigemina e B. bovis em Petrolina foi de 35,0% (164/468), 35,9% (168/468) e 32,3% (151/468), respectivamente; e em Ouricuri foi de 45,5% (179/393), 38,6% (152/393) e 54,9% (216/393), respectivamente. A co-infecção por Anaplasma spp. e Babesia spp. foi observada em 31,6% e 32,1% de amostras de Petrolina e Ouricuri, respectivamente. A detecção de DNA de Babesia spp. por PCR foi possível em 5,8% (8/137) carrapatos, dos quais em 62,5 % (5/8) foi detectada posteriormente infecção por B. bovis, e em 23,3% (32/137) por A. marginale. A presença de carrapatos, o uso de acaricidas, idade, raça, e o município de residência dos animais foram identificados como fatores de risco para TPB pela análise univariável e multivariável. Este estudo permitiu caracterizar os municípios estudados como de instabilidade enzoótica para esses hemoparasitos, e consequentemente, alertar para adoção de medidas adequadas de controle e realização de novos estudos. ABSTRACT: This study aimed to determine the seroprevalence of Babesiosis and Anaplasmosis in cattle from the municipalities of Ouricuri and Petrolina, state of Pernambuco, Brazil, and to define the risk factors for the occurrence of the diseases. Blood samples were collected for serologic testing by Indirect Immunofluorescence Assay (IFA). Sanitary epidemiological questionnaires were applied to the producers aiming to identify possible risk factors. Ticks were collected, identified and tested by Polymerase Chain Reaction (PCR) for the diagnosis of infection by Anaplasma marginale, Babesia bigemina and Babesia bovis. The study was conducted with 861 cattle, being 468 in Petrolina and 393 in Ouricuri. The seroprevalence of A. marginale, B. bigemina and B. bovis in Petrolina was of 35.0% (164/468), 35.9% (168/468) and 32.3% (151/468), respectively; and in Ouricuri was 45.5% (179/393), 38.6% (152/393), and 54.9% (216/393), respectively. Co-infection for Anaplasma spp. and Babesia spp. was observed in 31.6% and 32.1% samples of Petrolina and Ouricuri, respectively. The detection of DNA of Babesia spp. by PCR was possible in 5.8% (8/137) ticks; which 62.5% (5/8) was detected later infection with B. bovis; and 23.3% (32/137) with A. marginale. The presence of ticks, the use of acaricide, age, race, and county of residence of the animals were identified as risk factors for TBD by univariate analysis and multivariate. This study allowed the characterization of the municipalities studied as enzootic instability areas for these hemoparasitic, and consequently alert for adoption of adequate control measures and new studies.

No decorrer de 4h após a convulsão, foram determinados os teores plasmáticos de amônia e glicose, ureia, creatinina, potássio e sódio séricos, volume e gravidade específica da urina, e excreção urinária de amônio e ureia. No momento da convulsão, os teores de amônia plasmáticos foram muito altos e idênticos em todos os tratamentos, mas no 120°min, nos grupos tratados com associação de SSH+SSI+ASE (grupos 4 e 5), houve diminuição desse metabólito. O uso de furosemida (grupos 3 e 5) não aumentou a excreção total de urina. A terapia com associação de SSH+SSI+ASE aumentou ainda o volume urinário e a excreção percentual urinária de ureia e amônia durante o período crítico da 1ª hora de tratamento, mas o uso de SSI+ASE (grupos 2 e 3) teve resultados intermediários. A eficiência do tratamento com SSH+SSI+ASE ou SSI+ASE foi superior ao grupo controle. Embora com efeito menor que o observado com SSH+SSI+ASE, a SSI+ASE promoveu melhora no quadro clínico geral e, ao término do experimento, promoveu também uma adequada desintoxicação da amônia. To test the efficiency of some treatments for ammonia poisoning in cattle, 25 steers were used. Ammonium chloride solution was infused intravenously (iv) in each steer until the onset of convulsive episode. Thereafter, the animals were distributed in one of the five different groups, as follows: 1) control: infusion (iv) of 300mL isotonic saline solution (ISS) throughout the following 4h; 2) infusion (iv) of ISS 30mL kg-1 BW throughout the following 4h and administration of 4L water (W) through stomach tube; 3) the same as group 2, plus a single dose (iv) 2g furosemide kg-1 BW (F); 4) injection of 5mL kg-1 BW hypertonic saline solution (HSS) (7.2%) (iv) throughout the first 30min, followed by 20mL kg-1 BW of ISS and 4L W.; 5) the same as group 4 and a single dose of F. For the next 4h after the convulsion, plasma concentration of ammonia and glucose, serum urea, creatinine, sodium and potassium, urine total volume and specific gravity, and urinary excretion of ammonium and urea were followed. At the convulsion, the ammonia blood levels were high and similar among the groups, but at the 120th min the animals treated with HSS+ISS+W (groups 4 and 5) had lower values than the control. Furosemide (groups 3 and 5) did not prevent an increase in the total excretion of urine. The therapy with HSS+ISS+W still increased the urinary volume and the total urinary excretion of urea and ammonium during the critical 1st h of treatment, while the use of ISS+W had intermediate results. The efficiency of the treatment with HSS+ISS+W or ISS+W was better than the control group. Although with lower efficiency as seen in the treatment with HSS+ISS+W, the ISS+W caused an improvement in the clinical picture and at the end of the experiment generated an adequate ammonia detoxification. Doze novilhos foram experimentalmente intoxicados por administracao intrarruminal com alta dose de ureia (0,5g/kg PV) para estudar o efeito da intoxicacao por ureia sobre o figado e/ou musculatura. Foram coletadas amostras de sangue no decorrer do experimento para determinar o teor de amonia e as atividades de gama glutamiltransferase (gGT), aspartato aminotransferase (AST) e creatina quinase (CK). Onze novilhos apresentaram quadro clinico tipico de intoxicacao por amonia, porem um animal sucumbiu. Animais intoxicados apresentaram alto teor de amonia e elevada atividade de CK e AST. Quanto mais elevados foram os teores de amonia maiores foram as atividades de AST (r=0,59) e CK (0,61). A correlacao entre AST e CK foi alta e significativa (r=0,80), ocorrendo o inverso com AST e gGT (r=0,19). Tanto a atividade de AST como de CK aumentaram significativamente a partir dos episodios convulsivos decorrentes da intoxicacao por amonia. Estes resultados indicam que ocorreu uma expressiva lesao muscular, mas nao hepatica, visto que a CK e uma enzima tipica da musculatura estriada; e o AST encontra-se tanto na musculatura estriada como nos hepatocitos, enquanto que a gGT esta presente somente em celulas hepaticas.

Foram utilizados 25 ovinos da raça Ilede-France, aleatoriamente distribuídos em cinco grupos de cinco animais de cada, sendo que o grupo 1 recebia dieta contendo 80% volumoso e 20% concentrado, os grupos 2 e 3 recebiam 50% volumoso e 50% concentrado, e os grupos 4 e 5 recebiam a mesma dieta dos grupos 2 e 3 com a adição diária de 150 mg de sulfato de Cu até o término do experimento (150 d). Os grupos 1, 3 e 5 recebiam sal mineral contendo 300 ppm de Mo. Foram realizadas três biópsias hepáticas para determinação da concentração de Cu, Mo e Zn neste órgão e três ensaios de retenção aparente de Cu e Mo (0 d, 75 d, 150 d). Quinzenalmente, foi realizado exame clínico e coleta de amostras de sangue e urina. Três ovinos do grupo 4 e um do grupo 5 manifestaram ICA. Não existiu diferença entre a freqüência de mortalidade entre os grupos (P = 0,56). Os teores de Cu hepático nos ovinos com ICA (2450 ppm) foram superiores aos que não intoxicaram (1518 ppm). Quanto maior a ingestão de Mo na dieta menor foi o acúmulo de Cu hepático ao término do experimento (r = -0,72). Ovinos com ICA aumentaram a concentração de Zn hepático nas fases finais da intoxicação. A presença de altas atividades de GGT (>56,5 U/L) e de AST (>120 U/L) indicaram de maneira efetiva a presença de destacado acúmulo de Cu nos tecidos hepáticos (1000 ppm). The aim of this project is to evaluate the capacity of a molybdenum-rich mineral salt in the prevention of cumulative cooper poisoning (CCP) in sheep, through clinical and blood exams and hepatic copper and molybdenum concentrations. Twenty five Ile-de-France sheep were randomly distributed into five groups of five animals each, where group 1 received a 80% forrage and 20% concentrate diet, groups 2 and 3 received a 50% forrage and 50% concentrate diet, and groups 4 and 5 received the same diet as groups 2 and 3 with a daily supplementation of 150mg of copper sulphate until the end of the experiment (150 d). Groups 1, 3 and 5 received a mineral salt with 300 ppm of molybdenum. For three times during the experiment (day zero, 45th and 105th day) a liver biopsy was carried out to evaluate the degree of copper accumulation in this organ. Clinical examination was followed every two weeks, as far as blood and urine samples were withdrawn. Three sheep from group 4 and one sheep from group 5 presented clinical picture of CCP. There was no difference in the frequency of mortality between groups 4 and 5 (P = 0.56). The liver copper concentration was higher in sheep with CCP (2450 ppm) compared to sheep that not presented CCP (1518 ppm). The higher the ingestion of molybdenum in the diet the lower the liver copper concentration at the end of the experiment (r = -0.72). Sheep with CCP had higher liver zinc concentration. The presence of high activity of GGT (> 56.5 U/L) and AST (>120 U/L) indicated effectively the high accumulation of copper in liver (> 1000 ppm).

Summary Fifteen yearling steers were used to study the preventive effect of vitamin E on the protection against free radicals produced by carbon tetrachloride (CCl4). The animals were randomly divided in three equal groups and treated as follows: group A – previously injected (i.m.) with 15 IU/kg BW on the 15th and second day before the trial and drenched with 0.05 ml/kg BW CCl4; group B – only drenched with the same dose of CCl4; group C – drenched with a placebo. Food intake was recorded and blood samples collected daily for 8 days after the CCl4 drenching to compare the activity of aspartate aminotransferase (AST), gamma-glutamyltransferase (γ-GT) and the levels of erythrocyte reduced glutathione (GSH) and serum malonyldialdehyde (MDA). o uso de uréia dietética é comum na suplementação de bovinos criados extensivamente. A ingestão excessiva de uréia freqüentemente é causa de intoxicação por amônia, composto derivado da digestão da uréia. A amônia, em quantidades elevadas no organismo, provoca severo quadro nervoso, pulmonar e digestivo, podendo levar o bovino à morte. A partir de estudos recentes, que demonstraram a importância dos rins na eliminação da amônia do organismo, recomenda-se também que o tratamento desta intoxicação seja realizado por meio de rápida reposição de fluidos e uso de diurético [iv]. A intoxicação é prevenida por uma série de cuidados dietéticos.

Os animais foram monitorados, colhendo-se amostras sanguíneas e todo volume urinário. Os tratamentos O+F+H e O+H promoveram melhora clínica pronunciada, em relação ao grupo H, isto é, permaneceram em estação, retornaram o movimento de rúmen e o apetite e recuperaram-se do edema pulmonar mais rapidamente. Observaram-se, nos grupos O+F+H e O+H, teores plasmáticos mais baixos de amônia e lactato-L, urina mais abundante, excreção pela urina de maior quantidade de amônio e ureia, depuração mais intensa de amônia do sangue e pH urinário mais baixo. Concluiu-se que os tratamentos propostos apresentaram ação efetiva principalmente por diminuírem a hiperamonemia, por meio da maior excreção renal de amônio e maior transformação hepática de amônia em ureia Fifteen yearling steers were used to evaluate treatments for ammonia intoxication. The animals subjected to ammonium chloride infusion were distributed into three groups of five animals each, and treated as following: 1 - control group (C), physiological saline solution (H) infusion; 2 - O+H group, urea cycle aminoacids (O) and H medication; and 3 - O+F+H group, the same protocol of group 2 plus furosemide (F). The animals were monitored by collection of blood samples and the whole urinary volume. O+F+H and O+H treatments promoted pronounced clinical improvement, in attention to H group, i.e., they remained in standing position, returned ruminal movement and appetite, and recovered more quickly from pulmonary edema. In O+F+H and O+H groups, minor plasmatic levels of ammonia and L-lactate, more abundant urine, major ammonium and urea amount excretion by urine, more intensive ammonia depuration from blood, and lower urinary pH were observed. Foram utilizados 10 novilhos mestiços com cânula ruminal, distribuídos em dois grupos: no grupo controle (C; n=4) receberam dieta balanceada para ganho diário de 900g; no grupo tratado com carência pronunciada de energia (CP; n=6), receberam dieta com 30% a menos do nível de mantença em energia. Após 140 dias sob esses regimes de alimentação, foram coletadas amostras do fluido ruminal e urina, para realização de provas bioquímicas e funcionais, antes e às 1, 3, 6 e 9 horas após o fornecimento do alimento. A carência energética resultou em diminuição significativa na quantidade dos protozoários Entodinium, Eodinium, Isotricha, Dasytricha, Eremoplastron, Eudiplodinium, Metadinium, Charonina, Ostracodinium e Epidinium. An outbreak of footrot occurred in a flock of Corriedale sheep; 27 animals were treated with antibiotic and footbathed in a 5% copper sulfate solution. Being deprived of water for17 h, many sheep drank the footbath solution. After 6 h 16 sheep became ill with acute copper poisoning, 10 animals died within 10 h; 6 were severely ill and were sent to Veterinary Hospital, and 4 had mild signs and recovered without treatment. The sick sheep had anorexia, dullness, grinding teeth, moaning, rumen atony, dehydration, dark blue-green diarrheic feces and congested membranes. They were treated with 3.4 mg tetrathiomolybdate/kg body weight and lactated Ringer's solution iv, oral molybdate, sulfate, kaolin and pectin, and drenched with antacids. Two of the 6 sheep died during hospitalization. The ingestion of copper solution caused an intense gastrointestinal injury that resulted in ulcers, petechial and echymotic hemorrhages in the mucosa, mild hemolysis detected by microscopic hemoglobinuria and a lowered packed cell volume, severe hepatic injury that raised the AST and gammaGT blood values, and moderate kidney lesions with increasing serum blood urea and nitrogen creatinine levels.

Doze garrotes Girolando, nunca alimentados com uréia, foram distribuídos em dois grupos de seis animais cada. Ambos os grupos receberam intraruminalmente dose única (0,5 g/kg PV) de uréia extrusada (G1) ou granulada (G2), para induzir quadro de intoxicação por amônia. O quadro clínico exibido pelos garrotes foi acompanhado durante 240 minutos. Além da constatação dos sinais clínicos clássicos ligados a essa intoxicação, o presente trabalho descreve a presença de três novos sinais: desidratação, hipotermia e vasos episclerais ingurgitados. Convulsão, considerada sinal definitivo, ocorreu em cinco de seis animais de cada grupo. Um garrote (G1) exibiu apenas fasciculações, enquanto outro (G2) desenvolveu quadro clínico típico, porém sem convulsão, e recuperou-se espontaneamente sem tratamento. Os surgimentos de tremores musculares, decúbito esternal e episódios convulsivos ocorreram em momentos similares em ambos os grupos, mas quando analisados conjuntamente verificou-se que foram mais tardios no G1 (p < 0,04). O 1º sinal clínico observado foi a fasciculação, seguida por apatia, hiperestesia, apoio em obstáculos, tremores musculares, atonia ruminal, incoordenação motora, decúbito esternal e lateral, desidratação leve ou severa, e convulsão. Maiores freqüências cardíacas foram detectadas na convulsão. Após a convulsão, quatro garrotes de cada grupo apresentaram hipotermia leve. Um garrote do G2 entrou em estado comatoso e sucumbiu subitamente antes que fosse iniciado o tratamento. Apesar da uréia extrusada adiar o surgimento do quadro clínico, os sinais evidenciados foram tão severos quanto os causados por uréia granulada. Ambas formas de uréia, oferecidas em altas doses são perigosas a bovinos que nunca foram alimentados com uréia. Twelve yearling Girolando, rumen-fistulated steers never fed with urea before, were distributed randomly in 2 groups of 6 animals each. Both groups were administered intraruminally a single dose (0.5 g/kg BW) of extruded (G1) or prilled (G2) urea to induce ammonia poisoning. The clinical picture was followed for the next 240 min. Besides the classic signs the present study found 3 new additional sign: dehydration, hypothermia and ingurgitated episcleral veins. Convulsion, considered the definite sign, was seen in 5 out of 6 animals from both groups. One steer (G1) had only fasciculation, while another (G2) developed typical clinical signs, but not convulsion, and recovered spontaneously without treatment. The appearance of clinical signs such as muscle tremors, sternal recumbency and convulsive episode occurred at similar times in both groups, but when analyzed altogether they took place later in G1 (p < 0.04). The 1st sign to show up was fasciculation, followed by apathy, hyperaesthesia, pushing against obstacles, muscle tremor, rumen stasis, incoordination, sternal and then lateral recumbency, mild or severe dehydration, and convulsion. Higher heart rate was detected at the convulsive episodes. After the convulsions, 4 animals from each group had mild hypothermia. One steer from G2 fell down in coma and died suddenly before the beginning of the treatment. Although the extruded urea postponed the clinical picture, the signs were as severe as exhibited by cattle administered prilled urea. Both forms of urea offered at high dose can be harmful to cattle never fed urea.

The intravenous infusion of ammonium chloride was used to induce ammonia (NH3) poisoning in cattle. A 1.5 M ammonium chloride solution, buffered to pH 7.0, was infused at 400 mL/h until a convulsive episode occurred and therapy was initiated. Convulsions occurred with 200 to 1200 mL of ammonium solution. The clinical picture and metabolic effects were similar to the natural poisoning; no side effected occurred. The hypermmoniemia caused hyperglycemia, hyperlactemia, hyperkalemia and Intense metabolic acidosis. After treatment there was a sharp decrease in plasma NH3. Within 110 min all steers stood and recovered appetites. The induction of NH3 poisoning in cattle with ammonium chloride offers many advantages over the administration of high po doses of urea.

Para comparar o risco de intoxicação por uréia granulada (G) e extrusada (E) 24 garrotes, nunca alimentados com uréia, foram distribuídos em quatro grupos de seis animais, onde foi administrada, de uma só vez, (G) ou (E) em duas diferentes doses: alta (A; 0,5 g/kg PV) ou padrão (B; 0,22 g/kg PV). Em seguida, foram acompanhados o pH e os teores de amônia no rúmen, as concentrações sangüíneas de amônia, uréia, creatinina, glicose, lactato-L, potássio, as atividades de gama glutamiltransferase, aspartato aminotransferase e creatina quinase, perfil hemogasométrico e hematócrito, além de acompanhamento quadro clínico no decorrer de 240 min após as administrações de G ou E. Alguns animais dos grupos GB e EB tiveram um discreto quadro de intoxicação, se recuperando sem quaisquer tratamentos. Por outro lado, cinco garrotes de ambos grupos GA e EA tiveram severo quadro tóxico que exigiram tratamento, sendo que um animal GA sucumbiu. A velocidade de hidrólise ruminal da uréia G e E foi semelhante, embora as manifestações clínicas tenha sido iniciadas mais tardiamente no grupo EA. Quanto mais intensa foi a hiperamoniemia mais destacada foi o grau de acidose metabólica, desidratação, a glicólise anaeróbica e a gliconeogênese. Pela análise das atividades enzimáticas comprovou-se que os danos bioquímicos foram intensos na musculatura, mas não nos hepatócitos. Concluiu-se que tanto a uréia G como E quando oferecidas subitamente, em especial em doses altas, podem oferecer igual risco de intoxicação por amônia. To compare the toxicity potential of prilled urea (G) and extruded grain with urea (E), both were administered all at once in two different doses, high (A; 0,5 g/kg BW) or standard (B; 0,22 g/kg BW), to 24 steers divided into four groups of six animals, which had never been fed nonprotein nitrogen compounds. For 240 min after the administration of urea the following variables were determined: rumen fluid pH and ammonia level, blood ammonia, urea, creatinine, glucose, L-lactate, potassium, and activities of gama glutamiltranspeptidase, aspartate aminotransferase and creatine kinase, haemogasometric profile and hematocrit. The clinical picture was also followed. Some steers from groups GB and EB showed slight signs of ammonia toxicity, overcoming it without treatment. On the other hand, five steers from either groups GA and EA showed severe signs of ammonia toxicity that required treatment; even though one animal from group GA succumbed. The speed of ruminal urea hydrolysis of G and E was similar, although the clinical signs started later in group EA. The higher the hyperammonemia, the higher the metabolic acidosis, dehydration, anaerobic glycolysis and gluconeogenesis. The enzymatic profile showed that biochemical damage occurred in the striated muscular tissue, but not in the hepatocytes. The results showed that both G and E had similar potential to cause ammonia poisoning, principally when high doses were administered all at once.

Three hundred sixty-seven male Wistar rats were used to compare the efficiency of urea cycle amino acids (arginine, citrulline and ornithine; Group O), furosemide (Group F), and fluid therapy (saline solution; Group FT) to treat ammonia toxicity. Rats were injected ip initially with an ammonium acetate solution at 99.9% of the lethal dose. Three min later the rats were allocated randomly to Group C (control, received 1.2 mL distilled water). Group O (amino acids listed earlier, 2 mmol/kg bw), Group F (furosemide, 2 mg/kg bw), Group FT (7mL saline), or Groups O+F, O+FT, F+FT, or O+F+FT. All treatments were given ip except for Group F given im injections. Plasma ammonia, urea and creatinine, and hematocrit and pulmonary dry matter were determined. The highest survival rates were obtained with O+FT (57%) and O+F+FT (62.5%); only 6% of the controls survived. Plasma ammonia levels were ten-fold lower in rats treated with O+FT and O+F+FT (p0.0001). Fatally-poisoned rats had higher plasma ammonia, creatinine and hematocrit and exhibited pulmonary edema. Surviving rats had lower plasma urea. Animals given treatments with O, FT and F had (p0.005) higher urea, lower creatinine and less severe pulmonary edema, respectively, than those untreated.

O objetivo deste trabalho consistiu em esclarecer os mecanismos bioquímicos e hormonais da ação da somatotropina recombinante bovina (r-bST), recomendada no tratamento de toxemia da prenhez (TP). Para tal, foram realizados dois experimentos nos quais induziram-se quadros de hipercetonemia ou hiperlipidemia em 39 ovelhas com elevada condição corporal tratadas ou não previamente com 160 mg de um produto comercial de r-bST, pela via subcutânea. A hipercetonemia foi provocada pela infusão IV de 5 mM/kg P.V. de beta hidroxibutirato (BHB) no decorrer de duas horas e a hiperlipidemia com injeção IV de solução 20% de óleo de peixe, perfazendo 0,54g de triacilglicerol/kg P.V. no decorrer de quatro dias. Essas quantidades infundidas lograram provocar os quadros esperados. No primeiro experimento, o tratamento com r-bST promoveu aumento significativo da gliconeogênese por meio da conversão de BHB em glicose, maiores teores de insulina, com consequente aumento da resistência insulínica, incremento na concentração de IGF1 que levou a mobilização de nitrogênio ureico para os depósitos e maior atividade de GGT. No segundo experimento, o rbST gerou destacado aumento da gliconeogênese, pela metabolização dos AGNEs e de BHB em glicose, com maior destaque aos AGNEs, elevação dos teores de insulina e do percentual de resistência insulínica, e maior atividade da AST e GGT e bilirrubinal direta e total. Assim, conclui-se que a atuação farmacológica do r-bST se baseia na vigorosa capacidade do aumento da gliconeogênese, diminuindo concomitantemente os nefastos catabólitos (AGNEs e BHB) no processo da TP. Foi realizado ainda um experimento subsidiário para comparar a capacidade diagnóstica da detecção de BHB, em sangue total, entre o kit diagnóstico enzimático (Randox®), padrão ouro, e tiras reativas comerciais (Precision Xceed®) lidas em dosímetro portátil. Foram obtidas 247 amostras de sangue das ovelhas nas quais tinham sido infundida solução de BHB, no experimento um. Duas faixas de acetonemia foram consideradas: de 0,7 a 1,5 mM caracterizada como cetose inaparente e acima deste valor como cetose clínica. A correlação entre os métodos foi alta (r = 0,98 ; P < 0,001). Na cetose inaparente as tiras reativas apresentaram sensibilidade de 0,97 e especificidade de 0,89 e na cetose aparente sensibilidade de 0,99 e especificidade de 0,75, respectivamente. As tiras reativas podem ser empregadas eficientemente no diagnóstico precoce de casos de TP, com custo baixo e rápida leitura ao pé dos animais. The aim of this study was to clarify the biochemical and hormonal mechanisms of action of recombinant bovine somatotropin (r-bST), recommended in the treatment of pregnancy toxemia (PT). Two experiments were conducted inducing hyperlipidemia or hyperketonemia in 39 high body condition sheep pretreated or no with 160 mg of a commercial product of rbST subcutaneously. The hyperketonemia was caused by IV infusion of 5 mM / kg BW of beta-hydroxybutyrate (BHB) during two hours and hyperlipidemia with IV injection of a 20% fish oil solution corresponding to 0.54 g of triglyceride / kg BW in four days course. These infusions successfully caused the expected effects. In the first experiment, treatment with rbST caused a significant increase of gluconeogenesis by converting BHB into glucose, higher levels of insulin, with consequent increased insulin resistance, increase in the concentration of IGF1 leading to mobilization of urea nitrogen for deposits and increased activity of GGT. In the second experiment, the r-bST greatly increased gluconeogenesis generated by metabolism of non esterified fatty acids (NEFA), BHB and glucose, with emphasis on NEFA, elevated levels of insulin and the percentage of insulin resistance, and increased activity of AST and GGT and bilirrubin direct and total. In conclusion, the pharmacological action of r-bST is based on vigorous increase capacity on gluconeogenesis, concomitantly reducing the adverse catabolites (NEFA and BHB) in the PT process. An additional experiment was performed to compare the diagnostic capability of detecting BHB in whole blood, between the enzymatic diagnostic kit (Randox®), considered gold standard, and commercial test strips (Precision Xceed®) read in hand-held meter. 247 sheep blood samples were obtained in the first experiment in which BHB solution had been infused. Esse trabalho objetivou avaliar o efeito de crescentes quantidades de zinco dietético no metabolismo do cobre e na prevenção de intoxicação cúprica em ovinos. Foram utilizados 40 ovinos, machos, mestiços da raça Santa Inês, distribuídos em 5 grupos experimentais de 8 animais cada, submetidos a diferentes tratamentos com cobre (Cu) e zinco (Zn). Os grupos A, B, C e D receberam diariamente 12 mg de Cu por kg de PV sendo que adicionalmente o grupo A recebeu 1 mg de zinco por kg de PV/dia, o grupo B 4 mg de zinco por kg de PV/dia e o grupo C recebeu 8 mg de zinco por kg de PV/dia. O grupo H (controle), recebeu apenas a dieta basal. O experimento teve duração de 14 semanas. Foi realizada biópsia hepática ao início do estudo e sacrifício dos animais ao término do período experimental para coleta de tecidos e flúidos. Foram determinadas as concentrações de cobre e zinco no fígado, bile e sangue total. Semanalmente foram coletadas amostras de sangue para determinações bioquímicas e hematológicas, bem como realizada pesagem dos animais. Nenhum animal do grupo controle (H) e do grupo C (alto zinco) apresentou manifestação clínica, enquanto um animal dos grupos A, B e D, respectivamente, apresentou quadro clínico característico de intoxicação cúprica acumulativa (lCA). Observou-se uma elevação dos valores de cobre no fígado em todos os grupos experimentais entre o início e o final do experimento, contudo a elevação verificada no grupo H foi marcantemente menor, tendo ocorrido em decorrência de maiores níveis de cobre na dieta basal. Verificou-se ao final do experimento que os grupos A, B, C e D apresentaram maiores concentrações de cobre hepático em relação ao grupo H. O grupo C (alto Zn) teve menor concentração de cobre hepático do que o grupo D, que não recebeu zinco. Os grupos suplementados com zinco apresentaram maiores concentrações de metalotioneína (MT) no fígado em relação aos grupos não suplementados (D e H). Os grupos A e C apresentaram maiores valores de Cu ligado à MT em relação aos grupos D e H. Conseqüentemente houve maior proporção de Cu ligado à MT em relação ao Cu total nos grupos A e C em relação ao grupo D. Não foram verificadas diferenças significativas entre os valores da atividade da yGT entre os grupos suplementados com cobre, havendo apenas uma clara diferença entre o grupo controle e os demais. Ocorreu uma elevação dos valores da atividade da yGT em relação ao tempo zero nos grupos A, B C e D a partir da sétima semana. Observou-se uma elevação dos valores da atividade da AST em relação ao tempo zero nos grupos suplementados a partir da oitava semana do experimento. Não foram verificadas alterações nas variáveis bioquímicas e hematológicas nos grupos experimentais no decorrer do experimento, a exceção dos três animais que apresentaram quadro clínico de ICA. A quantidade de cobre utilizado para desafiar os ovinos foi suficiente para gerar um aumento substancial de cobre hepático e provocar em alguns dos animais quadros característicos de intoxicação cúprica. A adição de altos teores de zinco preveniu a morte de animais no decorrer do experimento; promoveu uma redução na quantidade de cobre acumulado no fígado; estimulou a síntese de metalotioneína hepática; incrementou a quantidade de cobre hepático ligado à metalotioneína; assim como o percentual de cobre hepático ligado a metalotioneína. Essas condições favoreceram a prevenção de quadros de intoxicação por cobre em ovinos. The objective of this study was to evaluate the effects of increasing quantities of dietary zinc on the metabolism of copper and the prevention of copper toxicosis in sheep. Forty male, mixed breed St. Inês, sheep were divided into 5 experimental groups, each containing 8 animais, and were submitled to different doses of copper (Cu) and zinc (Zn). Animais from Groups A, B, C, and D received 12 mg of Cu per kg of body weight daily (BW). Additionally, those in Group A, B, and C were supplemented with increasing levels of zinc: Group A, 1 mg of Znl kg BW; Group B 4 mg of Znl kg BW; and Group C 8 mg of Zn/kg BW. Group H animais served as controls and received only the basic diet. The experiment occurred during 14 weeks during which hepatic biopsies were done at the beginning of the study, and tissue fragments were collected at the end of the experiment after the animais were euthanized. The concentrations of Zn, Cu, bile, and whole blood were determined. Weekly blood samples were obtained to evaluate biochemical and hematological parameters during which the weight of each animal was recorded. Animais used as controls (Group H) and those that supplemented with elevated doses of Zn (Group C) did not demonstrate clinical manifestation of intoxication. However, one member from Group A, B, and D demonstrated clinical manifestations that were consistent with accumulative copper toxicosis (ACC). Elevated values of hepatic Cu occurred in ali groups between the beginning and the end of the experiment, but this elevation was drastically more reduced in Group H animais due to comparatively reduced Cu levels within the basic diet. At the end of the experiment it was verified that Group A, B, C, and D animais demonstrated more elevated concentrations of hepatic Cu relative to their counterparts from Group H. Members from Group C (elevated Zn) demonstrated reduced concentrations of hepatic copper relative to those from Group D, who did not receive any zinco Members from groups that received Zn supplement demonstrated more elevated concentrations of hepatic metallothionein (MT) relative to those from Groups D and H that did not receive any supplementation. Members from Groups A and D demonstrated more reduced values of Cu bound to MT (Cu-MT), relative to those from Groups D and H. Consequently, there was a greater proportion of Cu- MT relative to total Cu in animais from Groups A and C when compared with those from Group D. Significant differences in the values of yGT activity were not verified between the groups that were supplemented with Cu; there was only a distinct difference between members of the control group and the other groups. There was an elevation in the values of yGT activity relative to time 0 on Groups A, B, C and D effective from the eight week. An increase in the values of AST activity relative to time 0 was observed in animais from groups that were supplemented, this being effective from the tenth week of the experiment. Alterations to biochemical and hematological variables were not observed during the experiment; occurring only in those three animais that demonstrated clinical manifestations of ACC. The amount of copper used to challenge the sheep was adequate to induce a substantial increase of hepatic copper and produce the characteristic manifestations of copper toxicosis in some animais. The addition of elevated values of Zn prevented the death of animais during the experiment; promoted a reduction in the quantity of copper accumulated within the liver; stimulated the synthesis of metallothionein; incremented the quantity and the percentage of hepatic copper bound to metallothionein. These conditions favor the prevention of cupper toxicosis in sheep.
