Zusammenfassung Gegenstand und Ziel Nach bereits publizierten Ergebnissen dieser prospektiven Studie zu Vorkommen, Vorbericht, klinischem Bild der Neuropathie des N. tibialis (Tibialis-Neuropathie, TN) war es Ziel auf Behandlung und Prognose dieser Erkrankung einzugehen. Material und Methoden Von 88 Deutsch-Holstein-Kühen mit ein- (ETN, n = 71) und beidseitiger (BTN, n = 17) TN konnten Daten von 68 Kühen (56 ETN, 12 BTN) ausgewertet werden. Sie wurden retrospektiv 4 Gruppen zugeteilt: ohne Behandlung – spontane Ausheilung innerhalb 48 h (Spontan, 5 ETN), kein Verband (0Cast, 8 ETN, 3 BTN) bzw. mit Behandlung mit Antiphlogistika und Stützverband (StV, 3 ETN) oder Kunstharzverband (Cast, 40 ETN, 9 BTN). Zu 5 Zeitpunkten (14, 21, 28, 42, 56 d nach Erstvorstellung) erfolgten Kontrollen der behandelten Kühe. Zur letzten Kontrolle erfolgte bei 29 Kühen, wie bereits zur Erstvorstellung, eine Bestimmung der Plasma-Kreatinkinase-Aktivität. Ergebnisse Bei behandelten Kühen mit ETN wurde eine deutlich höhere Ausheilungsrate beobachtet als bei unbehandelten Kühen (Cast 98 % und StV 100 % vs. 0Cast 62 %). Im Vergleich dazu war der beobachtete Unterschied zwischen behandelten und unbehandelten Kühen mit BTN nicht so deutlich (78 % vs. 67 %). Die ermittelte Überlebensdauer der Kühe nach Ausheilung der TN war höher als ohne Ausheilung (545 d vs. 100 d). Die initial erhöhten Plasma-Kreatinkinase-Aktivitäten näherten sich nach Behandlung wieder dem Referenzbereich (434 U/l vs. 152 U/l). Schlussfolgerungen Insbesondere für die ETN ist nach Therapie mit Antiphlogistika und stabilisierendem Verband die Prognose als sehr gut anzusehen. Bei BTN war die Prognose abhängig von Art und Grad der Primärschädigung. Hinweise für eine schlechte Prognose waren insbesondere Ausfälle der Hautsensibilität. Bei guter Prognose ist die Therapie der TN aus wirtschaftlicher Sicht zu empfehlen, da in dieser Studie die Kühe nach Ausheilung im Mittel bis mindestens zur darauffolgenden Laktation gehalten werden konnten. Klinische Relevanz Die Studie verdeutlicht die Vorteile der Behandlung mit einem stabilisierenden Verband im Ausheilungsverlauf der TN. Bei der TN scheint im Vergleich zu Neuropathien anderer peripherer Nerven die Schädigung der Muskulatur von besonderer Bedeutung für das Krankheitsgeschehen zu sein.

The liver plays a central role in the postpartum (PP) energy metabolism of the transition dairy cow; however, studies describing the liver metabolome during this period were lacking. The aim of the presented study was therefore to compare the alterations in the liver and blood metabolome of transition dairy cows. For this purpose, an on-farm trial with 80 German Holstein cows (mean lactation number: 3.9; range: 2–9) was performed, with thorough documentation of clinical traits and clinical chemistry, as well as production data. Liver biopsies and blood samples were collected at d 14 (mean: 12 d, range: 1–26 d) antepartum (AP), d 7 (7, 4–13) and 28 (28, 23–34; mean, earliest-latest) PP for targeted mass spectroscopy-based metabolomics analysis using the AbsoluteIDQ p180 kit (Biocrates Life Sciences). Statistical analysis was performed using multivariate (partial least squares discriminant analysis) as well as univariate methods (linear mixed model). Multivariate data analysis of the liver metabolome revealed 3 different metabotypes (A = medium, B = minor, C = large alterations in the liver metabolome profile between AP and PP). In metabotype C, an increase of almost all acylcarnitines, lysophosphatidylcholines (lysoPC), sphingomyelins, and some phosphatidylcholines (PC, mainly at 7 d PP) was observed after calving. In contrast to metabotype C, the clinical data of the metabotype B animals indicated a higher PP lipomobilization and occurrence of transition cow diseases. The liver metabolome profile of these animals most likely mirrors a failure of adaptation to the PP state. This strong occurrence of metabotypes was much less pronounced in the blood metabolome. Additionally, differences in metabolic patterns were observed across the transition period when comparing liver and blood matrices (e.g., in different biogenic amines, acylcarnitines and sphingolipids). In summary, the blood samples at 7 d PP showed lower acylcarnitines and PC, with minor alterations and a heterogeneous pattern in AA, biogenic amines, and sphingomyelins compared with 14 d AP. In contrast to 7 d PP, the blood samples at 28 PP revealed an increase in several AA, lysoPC, PC, and sphingomyelins in comparison to the AP state, irrespective of the metabotype. In the liver biopsies metabotype B differed from metabotype C animals ante partum by following metabolites: higher α aminoadipic acid, lower AA, serotonin, taurine, and symmetric dimethylarginine levels, lower or higher concentrations of certain acylcarnitines (higher: C2, C3, C5, C4:1; lower: C12:1, C14:1-OH, C16:2), and lower lysoPC (a C16:0, C18:0, C20:3, C20:4) and hexose levels. In blood samples, fewer differences were observed, with lower serotonin, acylcarnitine C16:2, lysoPC (a C16:0, C17:0, C18:0 and C18:1), PC aa C38:0, and PC ae C42:2. The results show that the use of only the blood metabolome to assess liver metabolism may be hampered by the fact that blood profiles are influenced by the metabolism of many organs, and metabolomics analysis from liver biopsies is a more suitable method to identify distinct metabotypes. Future studies should investigate the stability and reproducibility of the metabotype and phenotypes observed, and the possible predictive value of the metabolites already differing AP between metabotype B and C.

The aims of this study were to evaluate histopathologic changes during the transition period, describe the histopathological features of the metabotypes identified in Part I ( Scharen et al., 2021b ), and investigate effects of a metaphylactic treatment with butaphosphan and cyanocobalamin (BCC) on the liver parenchyma. Eighty German Holstein cows (mean 305-d production: 10,957 kg, range: 6,480–15,193 kg; mean lactation number: 3.9, range: 2–9) from a commercial dairy farm in Saxony, Germany, were enrolled in a randomized, prospective, triple-blinded study. Two groups received a treatment with BCC (5 or 10 mL/100 kg of body weight 10% butaphosphan and 0.005% cyanocobalamin, Catosal, Bayer Animal Health, n = 20 each) and one group a placebo treatment (NaCl 0.9%, n = 40). Liver biopsy specimens were collected 14 d antepartum (AP) and 7, 28, and 42 d postpartum (PP), routinely processed for histologic examination, and stained with hematoxylin and eosin, Sudan III, periodic acid-Schiff, and picrosirius red stains. The sections were assessed for fat and glycogen content and degenerative, inflammatory, fibrotic, and proliferative changes. The statistical analysis included the effects of the sampling day, the lactation number, the treatment, and the metabotype (A = medium, B = minor, C = large alterations in the liver metabolome profile between AP and PP status). There was mild to moderate fat infiltration in the liver of 37% of cows in the last 2 wk AP, and moderate to severe fat infiltration in 66% of cows in the first days PP. The degree of fat infiltration increased from 2 wk AP until the end of the first week PP, and then decreased until the end of the study period, at which time about 25% of cows had moderate to severe fatty infiltration. Lipidosis was positively correlated with the severity of liver cell degeneration, and negatively correlated with the degree of glycogen deposits. Complete glycogen depletion of hepatocytes was not observed in cows, even in the presence of severe hepatic lipidosis. Moderate to severe lymphocytic hepatitis was seen in 39% of cows throughout the study period, and cows with lactation numbers 5 or greater had perisinusoidal fibrosis more often than younger cows. Severe fibrosis and cirrhosis of the liver did not occur. Metabotype B animals exhibited a higher chance of fatty infiltration, lower glycogen storage, and perisinusoidal fibrosis and for this metabotype positive correlations were calculated between increased fat deposition in the liver and marked glycogen depletion, and increased degenerative, inflammatory, fibrotic, and proliferative changes of hepatic tissue. For the treatment with BCC, no significant effect was observed. In summary, during the transition period, the liver of dairy cows is characterized by fat accumulation and glycogen depletion and histologic signs of hepatitis and hepatocyte degeneration. These histomorphologic changes were accentuated in animals exhibiting little alterations in their liver metabolome profile across the transition period (metabotype B) and support the assumption of a decreased grass silage quality as a causative factor.

As a follow-up to a previous study on the incidence, history and clinical findings of tibial neuropathy (TN), the present work aimed at describing the treatment and prognosis of this disease.Of 88 German Holstein dairy cows with unilateral (UTN, n = 71) or bilateral (BTN, n = 17) TN, 68 (56 UTN, 12 BTN) with a complete data set were analyzed. They were retrospectively assigned to one of four groups: no treatment - spontaneous healing within 48 h (Spontaneous, 5 UTN), no bandage (0Cast, 8 UTN, 3 BTN) or treatment with anti-inflammatory drugs and support bandage (StV, 3 UTN) or fiberglass cast (Cast, 40 UTN, 9 BTN). Treated cows were re-examined five times (14, 21, 28, 42 and 56 days after the first presentation). The plasma activity of creatine kinase was measured at the last re-examination in 29 cows similar to measurement at day 0.The observed overall success rate of treatment of cows with UTN was considerably higher compared with untreated cows (Cast 98 % and StV 100 % vs. 0Cast 62 %). By comparison, the observed difference between treated and untreated cows with BTN was not so clear (78 % vs. 67 %). Recovering cows exhibited a calculated longer median survival time than cows that did not recover (545 d vs. 100 d). Plasma creatine kinase activities were increased initially and returned within the reference interval (434 U/l and 152 U/L) following treatment.Cows with ETN have an excellent prognosis provided that treatment with anti-inflammatory drugs and stabilizing bandage is administered. In cows with BTN, the prognosis depended on the type and degree of the primary injury. Loss of skin sensitivity indicated a poor prognosis. From an economic standpoint, treatment of TN is indicated provided that the prognosis is good. In cows that had healed clinically, the average survival time extended into the following lactation.This study highlights the advantages of a support bandage for the treatment of cows with TN. Zusammenfassung Ziel Das vermehrte Auftreten von Ketose in der Transitphase bei Milchkühen verursacht hohe ökonomische Verluste. Die frühzeitige Identifizierung betroffener Tiere bereits im subklinischen Stadium ermöglicht, die Erkrankungshäufigkeit zu reduzieren und Verluste zu vermeiden. Ziel der Studie war, die Eignung eines mobilen Testgeräts zur Analyse der Betahydroxybutyrat-Konzentration (BHB-Konzentration) in der praktischen Anwendung zu prüfen. Darüber hinaus sollten die mit diesem Gerät gemessenen BHB-Konzentrationen mit denen verglichen werden, die mit einer Laborreferenzmethode aus dem Blut der V. jugularis ermittelt wurden. Material und Methoden Bei 81 Milchkühen wurde zu 7 Zeitpunkten (14 und 7 Tage a. p. sowie 7, 14, 21, 28, 42 Tage p. p.) Blut für die Analyse der BHB-Konzentration entnommen. Zur Messung im Labor mit einer kinetisch-enzymatischen Methode diente Blut aus der V. jugularis und V. oder A. coccygea mediana, zur Messung mit dem Gerät WellionVet BELUA Blut aus der V. oder A. coccygea mediana sowie Kapillarblut (gewonnen am Übergang der Haut zur Schleimhaut an der Vulva). Ergebnisse Anfängliche Anwendungsfehler bei der Bedienung des Geräts konnten mit Training abgestellt werden. Bei den Analyseergebnissen im Labor lagen die BHB-Konzentrationen aus dem Blut der V. jugularis um 0,07 mmol/l niedriger als die in Blut aus der V. oder A. coccygea mediana. Die mit dem Testgerät ermittelten BHB-Konzentrationen in Proben der V. oder A. coccygea mediana lagen um 0,13 mmol/l niedriger und die im Kapillarblut um 0,12 mmol/l niedriger als die im Labor aus dem Blut der V. jugularis gemessenen. Die mit dem Testgerät gemessenen BHB-Konzentrationen differierten zwischen den beiden Entnahmelokalisationen nicht signifikant. Schlussfolgerung Für die Kapillarblutentnahme eignet sich die Lokalisation am Übergang von Haut zu Schleimhaut an der Vulva. Das Gerät WellionVet BELUA liefert sofort Ergebnisse mit guter Übereinstimmung zwischen den Messwerten in Proben von verschiedenen Blutentnahmeorten wie auch im Vergleich zur Laboranalyse. Das Testsystem ist zur Anwendung in Milchviehbetrieben geeignet. Als Matrix kann sowohl Kapillarblut als auch Blut aus der V. oder A. coccygea mediana empfohlen werden.

The objective of this study was to test the effects of essential fatty acids (EFA), particularly α-linolenic acid (ALA), and conjugated linoleic acid (CLA) supplementation on metabolic and endocrine traits related to energy metabolism, including the somatotropic axis, in mid-lactation dairy cows. Four cows (126 ± 4 d in milk) were used in a dose-escalation study design and were abomasally infused with coconut oil (CTRL; 38.3 g/d; providing saturated fatty acids), linseed and safflower oils (EFA; 39.1 and 1.6 g/d; n-6:n-3 FA ratio = 1:3), Lutalin (CLA; cis-9,trans-11 and trans-10,cis-12 CLA, 4.6 g/d of each), or EFA and CLA (EFA+CLA) for 6 wk. The initial dosage was doubled twice after 2 wk, resulting in 3 dosages (dosages 1, 2, and 3). Each cow received each fat treatment at different times. Cows were fed with a corn silage-based total mixed ration providing a low-fat content and a high n-6:n-3 fatty acid ratio. Plasma concentrations of metabolites and hormones (insulin-like growth factor-binding proteins only on wk 0 and 6) were analyzed at wk 0, 2, 4, and 6 of each treatment period. Liver biopsies were taken before starting the trial and at wk 6 of each treatment period to measure hepatic mRNA abundance of genes linked to glucose, cholesterol and lipid metabolism, and the somatotropic axis. The changes in the milk and blood fatty acid patterns and lactation performance of these cows have already been published in a companion paper. The plasma concentration of total cholesterol increased with dosage in all groups, except CLA, reaching the highest levels in EFA+CLA and CTRL compared with CLA. The high-density lipoprotein cholesterol plasma concentration increased in CTRL and was higher than that in EFA and CLA, whereas the concentration of low-density lipoprotein cholesterol increased in a dose-dependent manner in EFA and EFA+CLA, and was higher than that in CLA. Hepatic mRNA expression of 3-hydroxy-3-methyl-glutaryl-CoA synthase 1 was upregulated in all groups but was highest in EFA+CLA. Expression of sterol regulatory element-binding factor 1 tended to be lowest due to EFA treatment, whereas expression of long chain acyl-CoA-synthetase was lower in EFA than in CTRL. Hepatic mRNA expression of GHR1A tended to be higher in EFA+CLA than in CTRL. The plasma concentration of insulin-like growth factor I increased in CLA, and the plasma IGFBP-2 concentration was lower in EFA+CLA than in CTRL at wk 6. The plasma concentration of adiponectin decreased in EFA+CLA up to dosage 2. Plasma concentrations of albumin and urea were lower in CLA than in CTRL throughout the experimental period. Supplementation with EFA and CLA affected cholesterol and lipid metabolism and their regulation differently, indicating distinct stimulation after the combined EFA and CLA treatment. The decreased IGFBP-2 plasma concentration and upregulated hepatic mRNA abundance of GHR1A in EFA+CLA-supplemented cows indicated the beneficial effect of the combined EFA and CLA treatment on the somatotropic axis in mid-lactation dairy cows. Moreover, supplementation with CLA might affect protein metabolism in dairy cows.

Zusammenfassung Gegenstand und Ziel Die Versorgung von Kühen mit Zitzenverletzungen stellt unter den Bedingungen der modernen Milchproduktion eine große Herausforderung dar. Ihre Behandlung und das Management während der Heilungsphase behindern den Schlüsselprozess der Milchwirtschaft, das Melken. Auf Basis der Auswertung des Patientenguts der Klinik für Klauentiere der Universität Leipzig werden therapeutisches Vorgehen und postoperatives Management präsentiert. Material und Methode In einer retrospektiven Untersuchung wurden die Patientendaten von Mai 2012 bis Juli 2017 analysiert. Die Datenerhebung umfasste Lokalisation und Art der Verletzung, initiale und postoperative Medikation sowie Behandlung und deren Erfolg. Ergebnisse Insgesamt wurden bei 116 Kühen 120 Zitzenverletzungen behandelt. Als häufigste Lokalisation traten Zitzenverletzungen unter Einbeziehung des Verschlussapparats (n = 85; 70,8 %) auf. Gedeckte Zitzenverletzungen stellten die häufigste Verletzungsart dar (n = 73; 60,8 %). Insgesamt waren 65 (54,2 %) der vorgestellten Läsionen gedeckt und bezogen den Verschlussapparat ein. Diese wurden zeitnah nach Vorstellung, unabhängig von anderen Diagnosen (Thelitis [außer akut, hochgradig], Zisternitis oder Mastitis) fast ausschließlich lateralendoskopisch geführt operiert. Insgesamt 98 (87,4 %) der behandelten Zitzen waren nach der Ausheilung melkbar, 7,2 % (n = 8) wurden amputiert und die übrigen ließen sich nach einer weiteren Operation uneingeschränkt melken. Die nach mindestens 700 Tagen erfolgte Nachfrage zum Verbleib der Tiere ergab, dass 23 (21,5 %) der chirurgisch und 2 (50 %) der konservativ versorgten Tiere wegen Problemen mit der betroffenen Zitze oder dem zugehörigen Viertel geschlachtet wurden. Schlussfolgerung und klinische Relevanz Zitzenverletzungen sind für die Rinderpraxis relevante Erkrankungen. Bei unverzüglicher, konsequenter Versorgung der Läsion und der intensiven, kompetenten Überwachung des Heilungsverlaufs mit angepasster postoperativer Therapie sind bezüglich Wiederherstellung der Euterfunktion die besten Behandlungsergebnisse zu erwarten.

Zusammenfassung Ziel Überprüfung, ob sich die Änderung der Kortisolkonzentration im Blut (KoB) von Milchkühen während eines akuten Stressreizes durch die Kortisolkonzentration im Speichel (KoS), in der Tränenflüssigkeit (KoTr) und in der Milch (KoM) und die Kortisolmetabolitenkonzentration im Kot (KoK) nachvollziehen lässt. Material und Methoden Bei 10 gesunden Deutsch-Holstein-Kühen diente eine simulierte Klauenbehandlung (sKB) im Durchtreibestand als Modell für eine akute Stresssituation. KoB, KoS, KoTr, KoM sowie KoK wurden einmal täglich über 10 Tage gemessen. Bei der an Tag 4 durchgeführten sKB wurden die KoB und KoTr (Minute 0, 15, 25, 30, 40, 50, 60, 80) und die KoK (Minute 480, 540, 600, 660) bestimmt. Ergebnisse Während der sKB stiegen die KoB und KoTr (Maximum zu Minute 60) und fielen anschließend ab. Nach der sKB kam es zu einem Anstieg der KoK (Maximum zu Minute 660). Während der sKB korrelierten KoB und KoTr sowie KoK und KoTr signifikant und KoK und KoB tendenziell. Im gesamten Verlauf der sKB (Area under the Curve [AUC], Minute 0–80) zeigten KoB und KoTr eine signifikante Korrelation (p = 0,04). Die KoB fiel von Tag 1 zu Tag 4 signifikant ab (p < 0,01). An Tag 5 waren KoB (p = 0,03) und KoK (p < 0,01) signifikant höher. Der Verlauf der KoS und KoTr (Tag 1–10) spiegelte den KoB-Verlauf gut wider. Der Verlauf der KoK wies Parallelen, der von KoM einige Unterschiede zum Verlauf der KoB auf. Deutlich positive Korrelationen ergaben sich für den Zeitraum Tag 1–10 im Vergleich von KoB und KoS (p = 0,002) sowie von KoB und KoTr (p = 0,002). Schlussfolgerungen und klinische Relevanz Die Gewinnung von Tränenflüssigkeit und Speichel stellt bei der Bestimmung der Kortisolkonzentration eine Alternative zur invasiven Blutentnahme dar. Ein ruhiger Umgang mit den Kühen führt zu einer geringeren Stressreaktion und kann somit das Tierwohl verbessern.

Zusammenfassung Gegenstand und Ziel Die thermische Enthornung von Kälbern stellt einen mit Schmerz und Stress verbundenen Eingriff dar. Verschiedene Verfahren der Schmerzausschaltung sowie die unmittelbaren Auswirkungen auf physiologische, biochemische und ethologische Prozesse wurden bereits ermittelt. Ziel der Studie war zu evaluieren, ob die Enthornung selbst, das Schmerzmanagement oder das Alter der Kälber zum Zeitpunkt des Eingriffs die Gesundheit der Tiere beeinflusst. Material und Methode Die Studie wurde als randomisierte, dreifach verblindete, prospektive Untersuchung an 337 weiblichen Deutsch-Holstein-Kälbern durchgeführt. Die Enthornung der Tiere erfolgte zwischen dem 4. und 10. Lebenstag oder zwischen dem 15. und 28. Lebenstag. Jedes Kalb wurde dabei einer von 9 Behandlungsgruppen zugelost, die sich in Art und Umfang der analgetischen Komponenten (Sedation, Leitungsanästhesie, Antiphlogese vs. Plazebo) unterschieden. Alle Kälber, bis auf die aus der Gruppe der scheinenthornten, wurden thermisch enthornt und sämtliche Tiere unterlagen einer klinischen Überwachung beginnend vor bis 4 Wochen nach dem Eingriff. Die erhobenen Befunde wurden als Boniturnoten unter Verwendung des Schwellenwertkonzepts mit dem Statistikprogramm SAS ausgewertet. Ergebnisse Die Enthornung beeinflusste die Gesundheit des Respirationsapparats der Kälber. Tiere, bei denen die Enthornung lediglich simuliert wurde, zeigten die niedrigsten Erkrankungsraten. Am häufigsten erkrankten Kälber, bei denen die Enthornung ohne Analgesie erfolgte. Die Varianten des Schmerzmanagements senkten das Risiko respiratorischer Erkrankungen in unterschiedlichem Maß. Die zu einem früheren Zeitpunkt enthornten Kälber wiesen seltener eine abweichende innere Körpertemperatur auf als ältere Kälber. Schlussfolgerung Die Enthornung stellt ein Risiko für die Gesundheit des Respirationsapparats von Kälbern dar. Sind Hornloszüchtung oder die Haltung horntragender Tiere nicht möglich, kann durch ein adäquates Schmerzmanagement das Risiko respiratorischer Erkrankungen gesenkt werden. Die Enthornung zu einem frühen Zeitpunkt ist möglich und sinnvoll. Klinische Relevanz Die Studie belegt die Bedeutung eines adäquaten Schmerzmanagements zur Enthornung für die Kälbergesundheit.

Resection of the distal interphalangeal joint (DIJ) is a claw-preserving method for treatment of septic arthritis. This study presents the radiographic and histological findings of the surgically-treated area after resection of the DIJ using a plantar approach in seven German Holstein cows. Histological postmortem evaluation 439 to 710 days after surgery showed that there was extensive proliferation of connective tissue rich in collagen fibres in the space left after resection in three cows; this was classified as fibrous ankylosis. In the remaining four cows, histological evaluation 1010 to 1756 days after surgery showed extensive new bone formation in the joint cavity consistent with osseous ankylosis. Radiographs of the resected DIJ region obtained at the time of histological examination revealed no osseous ankylosis in two cows, partial ankylosis in one cow and complete osseous ankylosis in four cows. Formation of complete osseous ankylosis after resection of the DIJ did take longer than 1–2 years in three of our specimens indicating a longer time span compared to earlier studies.

Recently, dose reference levels (DRLs) have been defined in Germany for auxiliary low-dose CT scans in hybrid SPECT/CT and PET/CT examinations, based on data from 2016/17. Here, another survey from 2020 was evaluated and compared with the new DRLs as well as with similar surveys from foreign countries.The survey, which had already been conducted in the Nordic countries, queried for various examinations including the following values: patient weight and height, volume CT dose index (CTDIData from 15 SPECT/CT and 13 PET/CT systems from 15 nuclear medicine departments were collected. For the following examinations datasets from more than 10 systems were submitted: SPECT lung VQ, SPECT bone, SPECTPET cardiac, PET brain, PET oncology. Especially for examinations of the thorax and heart, the new DRLs are very strict compared to this study. The CTDIFor certain examination types, there is a need for dose optimization at some clinics and devices in order to take into account the new DRLs in Germany in the future.ZIEL: Seit Kurzem gibt es in Deutschland für auxiliäre Niedrigdosis-CT-Aufnahmen bei SPECT/CT- und PET/CT-Untersuchungen Dosisreferenzwerte (DRW), welche auf Daten von 2016/17 basieren. Hier wurde eine weitere Umfrage aus dem Jahr 2020 ausgewertet und mit den neuen DRW sowie mit ähnlichen Umfragen aus anderen Ländern verglichen.Die Umfrage, welche bereits in den nordischen Ländern durchgeführt wurde, erfasste für unterschiedliche Einsatzbereiche u.a. folgende Werte: Patientengewicht/-größe, volumenbezogener CT-Dosisindex (CTDIDaten von insgesamt 15 SPECT/CT- und 13 PET/CT-Geräten aus 15 Zentren wurden erhoben. Hierbei beinhalteten die Daten z.B. folgender Untersuchungen mehr als 10 Geräte: Lungen-SPECT, Knochen-SPECT, kardiologisches SPECTPET, Hirn-PET, onkologisches PET. Besonders für Untersuchungen des Thorax und des Herzens sind die neuen DRW im Vergleich zu dieser Studie sehr streng. Die CTDIFür einzelne Untersuchungstypen besteht bei einigen Kliniken und Geräten Dosisoptimierungsbedarf, um die neuen DRW in Deutschland zukünftig zu berücksichtigen.

Matrices that can be collected non-invasively for quantification of a stress response in sheep have received little attention in the veterinary literature. This study examines the suitability of blood, tears and saliva for determining a stress response in sheep undergoing sham foot trimming on a tilt table. The cortisol concentration of blood, tears and saliva and the concentration of cortisol metabolites in faeces were measured in 13 healthy Meat Merino ewes once a day for six days. Sham foot trimming on a tilt table was used as the stressor and was done during a one-hour period on day 4; cortisol concentrations of blood and tears were measured at 0, 10, 20, 30, 40 and 60 minutes. Cortisol concentrations of blood (maximum at 30 minutes) and tears (maximum at 40 minutes) increased during the procedure and then decreased. There were significant correlations between cortisol concentrations of blood and tears (p = 0,04) during sham foot trimming (area under the curve, 0 to 60 minutes). Vascular changes play an important role in the pathogenesis of claw horn disruption lesions in cattle. The aim of the study was to measure arterial blood flow in the hind limbs of German Holstein cows with claw horn disruption lesions. A 10-MHz linear transducer was used to assess blood flow in the interdigital artery in the dorsal pastern region in the hind limbs of 11 non-lame and 33 lame German Holstein cows in which lameness was scored clinically. Qualitative and quantitative blood flow parameters were compared in affected limbs and unaffected contralateral hind limbs in lame cows and in the hind limbs of lame cows and non-lame cows. A pulsed-wave Doppler signal suitable for analysis was obtained in 78 of 88 limbs (33 affected and contralateral limbs, 22 limbs of control cows). Blood flow curve types 1 and 2 were predominant in the hind limbs of lame cows. Vessel diameter, end-diastolic velocity, and blood flow rate were significantly greater in lame cows than in non-lame cows and were numerically greater in moderately lame cows than in mildly lame cows. The differences in the qualitative and quantitative parameters between lame and non-lame cows were most likely caused by inflammation of the pododerm. The role of weight distribution between the paired hind limbs and the existence of claw horn disruption appeared to have an effect on the differences in local circulation in the affected and unaffected contralateral hind limbs in lame cows.

Zusammenfassung Ziel Das Schmerzmanagement beim Tier spielt aus der rechtlichen und ethischen Verpflichtung des Menschen heraus eine wichtige Rolle für das Tierwohl. Für Eingriffe an der distalen Gliedmaße beim Schaf ist die retrograde intravenöse Stauungsanästhesie (RIVA) eine beschriebene Methode zur Betäubung der Zehe. Bisher hat sie beim Schaf keine breite Anwendung in der Praxis gefunden. Ziel der Studie war, Durchführbarkeit, Sicherheit und Wirksamkeit der RIVA zu überprüfen. Weiterhin sollte ermittelt werden, ob das Schmerzmanagement durch die Kombination aus RIVA und Sedation verbessert werden kann und ob sich diese Methode positiv auf das postoperative Wohlergehen der Tiere auswirkt. Methoden Bei 36 Merinofleischschafen mit Dermatitis interdigitalis contagiosa und 12 gesunden Kontrolltieren wurde das Verhalten während der Behandlung der Läsionen unter verschiedenen Methoden des Schmerzmanagements bzw. während der Klauenpflege und postoperativ in der Herde erfasst. Die Merkmale wurden mit Scores bewertet und die Ausprägung bei den Tieren in den 4 Studiengruppen (RIVA, Sedation mit Xylazinhydrochlorid + RIVA, Plazebo, Kontrolle) verglichen. Ergebnisse Die RIVA konnte erfolgreich durchgeführt werden. Lokale Veränderungen im Bereich der Applikationsstelle und des Stauschlauchs bei 2 Schafen heilten vollständig ab. Eine deutliche Reduktion der Abwehrbewegungen während des schmerzhaften Eingriffs belegt die Wirksamkeit der RIVA. Es ist davon auszugehen, dass die RIVA-Tiere aufgrund des Handlings in Rückenlage vergleichbar viel Stress empfanden haben wie die Plazebo-Tiere, denn stressassoziierte Merkmale (Kopfschlagen, Leerkauen) traten in den Gruppen ähnlich häufig auf. Die Sedation führte zusätzlich zur reduzierten Ausprägung schmerz- und stressassoziierter Merkmale (Abwehrbewegungen, Entlastungshaltung, Lautäußerungen, u. a.). Die Xylazinhydrochlorid-RIVA-Tiere entlasteten die betroffene Gliedmaße postoperativ seltener und zeigten häufiger Futteraufnahme sowie Wiederkauen als die Tiere der anderen Behandlungsgruppen. Schlussfolgerung Die RIVA ist eine einfach durchführbare und sichere Methode zur lokalen Schmerzausschaltung. Eine zusätzliche Sedation reduziert die Stress- und Schmerzantwort. Dieses Schmerz- und Stressmanagement hat zudem einen positiven Effekt auf das postoperative Wohlergehen der Tiere.

The aim of this study was to document the occurrence, history, and clinical findings of tibial neuropathy in German Holstein dairy cows in order to yield information regarding the importance and etiology of this disorder in dairy herds in Central Germany.This prospective study was undertaken between January 2013 and October 2017 and included 88 German Holstein dairy cows with unilateral (UTN, n = 71) or bilateral (n = 17, BTN) tibial neuropathy. Data were collected from the history as well as the clinical and laboratory examinations. Three grades of paresis were defined.The percentage of cows with tibial neuropathy among all German Holstein dairy cows presented with disorders of the locomotor system in the study period amounted to 2.2 %. UTN was seen predominantly following dystocia or as a result of prolonged lateral recumbency in consequence to a primary disease. The majority of cases of BTN occurred in association with the resumption of cyclicity following calving. These were accompanied by clinical signs of a cauda equina syndrome. Increased plasma activities of creatine kinase (UTN 98 %; BTN 100 %) and aspartate aminotransferase (UTN 89 %; BTN 100 %) were observed in nearly all affected cows.Tibial neuropathy is a regularly occurring disorder in dairy herds in Central Germany. The condition results in pain and locomotor impairment and therefore constitutes a welfare concern. UTN and BTN have different causes, and the history combined with clinical signs (severity of paresis) provides etiological information. Increased enzymatic activities suggest a muscular involvement in the disease process.ZIEL: Die prospektive Studie hatte zum Ziel, Vorkommen, Vorbericht und klinisches Bild der Neuropathie des N. tibialis (Tibialis-Neuropathie) bei Deutsch-Holstein-Kühen zu dokumentieren und damit Hinweise auf Bedeutung und Ätiologie dieser Erkrankung in mitteldeutschen Milchviehbetrieben zu gewinnen.Bei 88 Deutsch-Holstein-Kühen mit einseitiger (ETN, n = 71) oder beidseitiger (BTN, n = 17) Tibialis-Neuropathie wurden zwischen Januar 2013 und Oktober 2017 Daten zum Vorbericht sowie klinische und labordiagnostische Befunde erhoben. Für die Ausprägung der Parese wurden 3 Grade definiert.Im Untersuchungszeitraum wiesen 2,2 % der aufgrund einer Erkrankung des Bewegungsapparats vorgestellten Deutsch-Holstein-Kühe eine Tibialis-Neuropathie auf. Eine ETN trat vorwiegend post partum nach Dystokie auf oder wenn sich Tiere aufgrund von Grunderkrankungen vermehrt in Seitenlage positioniert hatten. Kühe mit BTN wurden hauptsächlich mit Erkrankungsbeginn im Zeitraum der ersten Brunst nach der Abkalbung vorgestellt und wiesen Anzeichen einer „zentralen“ Nachhandparese auf. Bei fast allen Tieren fanden sich im Blutplasma erhöhte Aktivitäten der Kreatinkinase (ETN: 98 %; BTN: 100 %) und Aspartataminotransferase (ETN: 89 %; BTN: 100 %).Die Tibialis-Neuropathie kommt in mitteldeutschen Milchviehbetrieben regelmäßig vor. Bewegungseinschränkung und Schmerzen beeinträchtigen das Wohlbefinden der Kühe. Die Ätiologie von ETN und BTN sollte differenziert betrachtet werden. Vorbericht und klinisches Bild (vorliegender Paresegrad) liefern Hinweise auf die Ätiologie. Die labordiagnostischen Befunde legen nahe, dass auch die Muskulatur der betroffenen Gliedmaße geschädigt wird.

Sufficient glucose availability is crucial for exploiting the genetic potential of milk production during early lactation, and endocrine changes are mainly related to repartitioning of nutrient supplies toward the mammary gland. Long-chain fatty acids, such as essential fatty acids (EFA) and conjugated linoleic acid (CLA), have the potential to improve negative energy balance and modify endocrine changes. In the present study, the hypothesis that combined CLA and EFA treatment supports glucose metabolism around the time of calving and stimulates insulin action and the somatotropic axis in cows in an additive manner was tested. Rumen-cannulated German Holstein cows (n = 40) were investigated from wk 9 antepartum (AP) until wk 9 postpartum (PP). The cows were abomasally supplemented with coconut oil (CTRL, 76 g/d); 78 g/d of linseed and 4 g/d of safflower oil (EFA); Lutalin (CLA, isomers cis-9,trans-11 and trans-10,cis-12 CLA, each 10 g/d); or the combination of EFA+CLA. Blood samples were collected several times AP and PP to determine the concentrations of plasma metabolites and hormones related to glucose metabolism and the somatotropic axis. Liver tissue samples were collected several days AP and PP to measure glycogen concentration and the mRNA abundance of genes related to gluconeogenesis and the somatotropic axis. On d 28 AP and 21 PP, endogenous glucose production (eGP) and glucose oxidation (GOx) were measured via tracer technique. The concentration of plasma glucose was higher in CLA than in non-CLA-treated cows, and the plasma β-hydroxybutyrate concentration was higher in EFA than in non-EFA cows on d 21 PP. The eGP increased from AP to PP with elevated eGP in EFA and decreased eGP in CLA-treated cows; GOx was lower in CLA than in CTRL on d 21 PP. The plasma insulin concentration decreased after calving in all groups and was higher in CLA than in non-CLA cows at several time points. Plasma glucagon and cortisol concentrations on d 21 PP were lower in CLA than non-CLA groups. The glucagon/insulin and glucose/insulin ratios were higher in CTRL than in CLA group during the transition period. Plasma IGF-I concentration was lower in EFA than non-EFA cows on d 42 AP and was higher during the dry period and early lactation in CLA than in non-CLA cows. The IGF binding protein (IGFBP)-3/-2 ratio in blood plasma was higher in CLA than in non-CLA cows. Hepatic glycogen concentration on d 28 PP was higher, but the mRNA abundance of PC and IGFBP2 was lower in CLA than non-CLA cows on d 1 PP. The EFA treatment decreased the mRNA abundance of IGFBP3 AP and PCK1, PCK2, G6PC, PCCA, HMGCS2, IGFBP2, and INSR at several time points PP. Results indicated elevated concentrations of plasma glucose and insulin along with the stimulation of the somatotropic axis in cows treated with CLA, whereas EFA treatment stimulated eGP but not mRNA abundance related to eGP PP. The systemic effects of the combined EFA+CLA treatment were very similar to those of CLA treatment, but the effects on hepatic gene expression partially corresponded to those of EFA treatment.

Clinical ketosis is common during the dairy cows' transition period and is responsible for considerable economic loss. Early identification of cows with subclinical ketosis is the first step for maintaining the health and productivity of dairy cows. The goals of the study were two-fold: The first was to examine the usefulness of a mobile test device as a cow-side test; and the second was to compare BHB concentrations measured by the ketometer using capillary blood and blood collected from the coccygeal vessels with values determined by a reference method in the laboratory using jugular blood.Blood samples were collected from a jugular vein or the coccygeal vessels in 81 dairy cows at 7 time points (14 and 7 days pre-partum and 7, 14, 21, 28, and 42 days post-partum) for kinetic enzymatic measurement of BHB concentration in the laboratory. Blood samples were concurrently collected from the coccygeal vessels or by pricking the vulvar lip at the transition of the skin to the mucosa (capillary blood) to determine BHB concentration using the WellionVet BELUA ketometer (MED TRUST GmbH, Marz, Austria).Initial errors in operating the ketometer were quickly eliminated with experience. BHB concentrations of jugular blood measured in the laboratory were 0.07 mmol/l lower than those measured in coccygeal blood. The mean BHB concentration measured in coccygeal and capillary blood using the WellionVet BELUA ketometer did not significantly differ but were 0.13 and 0.12 mmol/l respectively, lower than the mean jugular vein concentrations measured in the laboratory.The WellionVet BELUA ketometer is useful for determination of BHB concentration in cows provided that the manufacturer's specifications are followed. Capillary blood is best collected at the transition from the vulvar skin to its mucosa. The device generates rapid results that correlate well with BHB concentrations determined in the laboratory and with the results obtained from different blood collection sites. It is ideally suited for monitoring dairy cows for subclinical ketosis using capillary or coccygeal blood.ZIEL: Das vermehrte Auftreten von Ketose in der Transitphase bei Milchkühen verursacht hohe ökonomische Verluste. Darüber hinaus sollten die mit diesem Gerät gemessenen BHB-Konzentrationen mit denen verglichen werden, die mit einer Laborreferenzmethode aus dem Blut der V. jugularis ermittelt wurden.Bei 81 Milchkühen wurde zu 7 Zeitpunkten (14 und 7 Tage a. p. sowie 7, 14, 21, 28, 42 Tage p. p.) Blut für die Analyse der BHB-Konzentration entnommen. Die mit dem Testgerät ermittelten BHB-Konzentrationen in Proben der V. oder A. coccygea mediana lagen um 0,13 mmol/l niedriger und die im Kapillarblut um 0,12 mmol/l niedriger als die im Labor aus dem Blut der V. jugularis gemessenen. Die mit dem Testgerät gemessenen BHB-Konzentrationen differierten zwischen den beiden Entnahmelokalisationen nicht signifikant.Für die Kapillarblutentnahme eignet sich die Lokalisation am Übergang von Haut zu Schleimhaut an der Vulva. Das Gerät WellionVet BELUA liefert sofort Ergebnisse mit guter Übereinstimmung zwischen den Messwerten in Proben von verschiedenen Blutentnahmeorten wie auch im Vergleich zur Laboranalyse. Das Testsystem ist zur Anwendung in Milchviehbetrieben geeignet. Als Matrix kann sowohl Kapillarblut als auch Blut aus der V. oder A. coccygea mediana empfohlen werden.

We studied the prevalence of extended-spectrum beta-lactamase (ESBL)-producing Enterobacteriaceae in dairy calves as part of a routine health check protocol. In addition, data regarding antimicrobial use (AMU), farm hygiene, and farm management were collected in order to identify possible risks for ESBL occurrence. Ten farms participated in the study with a median of 781 milking cows (319-1701). All calves investigated were younger than two weeks with an average age of 6.8 (±3.9) days. The farms were visited and samples were collected twice at an interval of 7-11 months. Faecal samples diluted 1:10, were plated onto BrillianceTM ESBL agar in duplicates. After 24 hours at 37°C, colonies were counted and total colony forming units (cfu)/ml calculated. Bacteria species were identified biochemically. ESBL-production was phenotypically confirmed using the MICRONAUT-S β-Lactamases system. Additionally, antimicrobial susceptibility was tested using VITEK® 2 technology. Phylotyping of E. coli isolates and screening for bla genes was performed by PCR. ESBL-producing enterobacteria were detected on all farms and 96.5% of calves investigated shed ESBL-positive bacteria. Of all ESBL-producing isolates, the majority were E. coli (92.9%), followed by Enterobacter cloacae (5.1%) and Klebsiella pneumoniae subsp. pneumoniae (2.0%). The majority of E. coli isolates was clearly assigned to phylogroup C (25.0%), followed by phylogroups A (15.2%) and E (14.1%). CTX-M group 1 was most frequently detected (80.4%). E. cloacae contained blaCTX-M and blaTEM or blaSHV. K. pneumoniae harboured blaSHV only. Besides resistance to penicillins and cephalosporins, the majority of isolates was also resistant to one or more antibiotic classes, with a high proportion being resistant against fluoroqinolones. 52.5% of isolates were further characterised as threefold multidrug resistant gram-negative bacteria (3MDR-GNB) according to the German Commission for Hospital Hygiene and Infection Prevention. None of the isolates were 4MDR-GNB, i.e. none revealed carbapenem-resistance. Penicillins were the most frequently administered antibiotics to calves on most farms and were the predominant substance class at herd level on all farms. Overall, the number of calves treated prior to sampling was rather low (11.7%). Analyses of data regarding the farm management identified weaknesses in biosecurity and cleaning and disinfection. Besides beta-lactam antibiotics being the most commonly used antibiotics no other risk factors could be identified. In summary, the prevalence of ESBL-carriers in dairy calves was exceptionally high and should be motivation to develop strategies for the reduction of multidrug-resistant bacteria in farm animals.

Hot iron disbudding of calves is a stressful and painful procedure. Previous parts of an ongoing comprehensive study on disbudding in dairy calves dealt with various types of pain management and the direct effects of the procedure on physiological, biochemical and behavioral processes. The goal of this study part was to investigate the effects of the disbudding procedure per se, pain management and the age of the calf at the time of disbudding on the health status of the calf.A total of 337 German Holstein heifer calves were used for this prospective, randomized and triple-blinded study. The calves were disbudded at 4-10 days or at 15-28 days of age. Each calf was randomly assigned to one of 9 treatment groups, which differed with respect to pain management (sedation, local anesthesia, nonsteroidal antiinflammatory drugs, placebo). All but the sham-disbudded calves underwent hot iron disbudding and all calves were monitored clinically before and until 4 weeks after the procedure. The findings were analyzed using valuation scores and a threshold model of the SAS software.Disbudding exhibited adverse effects on respiratory health. Calves undergoing sham disbudding experienced the lowest incidence of respiratory disease and calves that did not receive any anesthesia or pain medication displayed the highest frequency. All treatments showed mitigating effects on the incidence of respiratory tract diseases, however the effects varied with the type of pain management. Fever was less common in calves that were disbudded early in life than calves that underwent disbudding at an older age.Disbudding of young calves poses a risk to respiratory health. If breeding of polled offspring is not possible or when farming of horned cattle is not feasible, the risk of respiratory disease induced by disbudding can be minimized by adequate pain management. Disbudding of very young calves is feasible and recommended.The present study underlines the importance of optimal pain management during disbudding of dairy calves.Die thermische Enthornung von Kälbern stellt einen mit Schmerz und Stress verbundenen Eingriff dar. Verschiedene Verfahren der Schmerzausschaltung sowie die unmittelbaren Auswirkungen auf physiologische, biochemische und ethologische Prozesse wurden bereits ermittelt. Ziel der Studie war zu evaluieren, ob die Enthornung selbst, das Schmerzmanagement oder das Alter der Kälber zum Zeitpunkt des Eingriffs die Gesundheit der Tiere beeinflusst.Die Studie wurde als randomisierte, dreifach verblindete, prospektive Untersuchung an 337 weiblichen Deutsch-Holstein-Kälbern durchgeführt. Die Enthornung der Tiere erfolgte zwischen dem 4. und 10. Lebenstag oder zwischen dem 15. und 28. Lebenstag. Jedes Kalb wurde dabei einer von 9 Behandlungsgruppen zugelost, die sich in Art und Umfang der analgetischen Komponenten (Sedation, Leitungsanästhesie, Antiphlogese vs. Plazebo) unterschieden. Alle Kälber, bis auf die aus der Gruppe der scheinenthornten, wurden thermisch enthornt und sämtliche Tiere unterlagen einer klinischen Überwachung beginnend vor bis 4 Wochen nach dem Eingriff. Die erhobenen Befunde wurden als Boniturnoten unter Verwendung des Schwellenwertkonzepts mit dem Statistikprogramm SAS ausgewertet.Die Enthornung beeinflusste die Gesundheit des Respirationsapparats der Kälber. Tiere, bei denen die Enthornung lediglich simuliert wurde, zeigten die niedrigsten Erkrankungsraten. Am häufigsten erkrankten Kälber, bei denen die Enthornung ohne Analgesie erfolgte. Die Varianten des Schmerzmanagements senkten das Risiko respiratorischer Erkrankungen in unterschiedlichem Maß. Die zu einem früheren Zeitpunkt enthornten Kälber wiesen seltener eine abweichende innere Körpertemperatur auf als ältere Kälber.Die Enthornung stellt ein Risiko für die Gesundheit des Respirationsapparats von Kälbern dar. Sind Hornloszüchtung oder die Haltung horntragender Tiere nicht möglich, kann durch ein adäquates Schmerzmanagement das Risiko respiratorischer Erkrankungen gesenkt werden. Die Enthornung zu einem frühen Zeitpunkt ist möglich und sinnvoll.Die Studie belegt die Bedeutung eines adäquaten Schmerzmanagements zur Enthornung für die Kälbergesundheit.

The objective of this study was to determine whether changes in cortisol concentration measured in blood serum (KoB) of cows exposed to an acute stressor shows a correlation to cortisol concentrations in saliva (KoS), tears (KoT) and milk (KoM) as well as the concentration of cortisol metabolites in feces (KoK).In 10 healthy German Holstein cows, sham foot trimming (sKB) including the movement/fixation of the cows through/in a foot trimming chute was used as a model for acute stress. KoB, KoS, KoT, KoM and KoK were measured once a day for 10 days. During sKB, performed on day 4, KoB and KoT were measured at the initiation of foot trimming (minute 0) as well as 15, 25, 30, 40, 50, 60 and 80 minutes later. Additionally, KoK was measured 480, 540, 600 and 660 minutes after the start of sKB.KoB and KoT increased during sKB and reached a maximum at 60 minutes followed by a decrease. KoK increased after sKB and reached a maximum at 660 minutes. There was a significant correlation between KoB and KoT, KoK and KoB and a trend towards a significant correlation between KoK and KoT during sKB. KoB and KoT were significantly correlated (area under the curve, minute 0-10, p = 0.04). KoB decreased significantly from day 1 to day 4 (p0.01). On day, 5 KoB (p = 0.03) and KoK (p0.01) were significantly higher. KoS and KoT served as good proxies for KoB throughout the study, and KoK and KoB exhibited similar profiles. There were several differences between the profiles of KoM and KoB. During the 10-day measurement period, a significant positive correlation was detected between KoB and KoS (p = 0.002) as well as between KoB and KoT (p = 0.002).The collection of tear fluid and saliva for determination of cortisol concentrations are non-invasive alternatives to blood sampling. Calm handling of cows may reduce their stress reaction and thus improve animal welfare.ZIEL: Überprüfung, ob sich die Änderung der Kortisolkonzentration im Blut (KoB) von Milchkühen während eines akuten Stressreizes durch die Kortisolkonzentration im Speichel (KoS), in der Tränenflüssigkeit (KoTr) und in der Milch (KoM) und die Kortisolmetabolitenkonzentration im Kot (KoK) nachvollziehen lässt.Bei 10 gesunden Deutsch-Holstein-Kühen diente eine simulierte Klauenbehandlung (sKB) im Durchtreibestand als Modell für eine akute Stresssituation. KoB, KoS, KoTr, KoM sowie KoK wurden einmal täglich über 10 Tage gemessen. Bei der an Tag 4 durchgeführten sKB wurden die KoB und KoTr (Minute 0, 15, 25, 30, 40, 50, 60, 80) und die KoK (Minute 480, 540, 600, 660) bestimmt.Während der sKB stiegen die KoB und KoTr (Maximum zu Minute 60) und fielen anschließend ab. Nach der sKB kam es zu einem Anstieg der KoK (Maximum zu Minute 660). Während der sKB korrelierten KoB und KoTr sowie KoK und KoTr signifikant und KoK und KoB tendenziell. Im gesamten Verlauf der sKB (Area under the Curve [AUC], Minute 0–80) zeigten KoB und KoTr eine signifikante Korrelation (p = 0,04). Die KoB fiel von Tag 1 zu Tag 4 signifikant ab (p0,01). An Tag 5 waren KoB (p = 0,03) und KoK (p0,01) signifikant höher. Der Verlauf der KoS und KoTr (Tag 1–10) spiegelte den KoB-Verlauf gut wider. Der Verlauf der KoK wies Parallelen, der von KoM einige Unterschiede zum Verlauf der KoB auf. Deutlich positive Korrelationen ergaben sich für den Zeitraum Tag 1–10 im Vergleich von KoB und KoS (p = 0,002) sowie von KoB und KoTr (p = 0,002).Die Gewinnung von Tränenflüssigkeit und Speichel stellt bei der Bestimmung der Kortisolkonzentration eine Alternative zur invasiven Blutentnahme dar. Ein ruhiger Umgang mit den Kühen führt zu einer geringeren Stressreaktion und kann somit das Tierwohl verbessern.

Background Recumbent cows are a diagnostic challenge because of a wide range of differential diagnoses, which include trauma, neurological and metabolic disorders, malnutrition and mineral deficiencies. This case report describes recumbent suckler cows that presented as a herd problem. In addition to weakness due to inanition, Cu and Se deficiencies were considered as possible aetiologies of the recumbency. Furthermore, Trypanosoma (T.) theileri, a blood parasite of unknown importance in Germany, was detected in the blood of some cows. Case presentation Three recumbent cows were referred to the Clinic for Ruminants and Swine, Faculty of Veterinary Medicine of the University of Leipzig. They were unable to rise and had low body condition scores and rough hair coats. Haematological and serum biochemical analyses showed neutrophilia, electrolyte imbalances, increased activities of muscle and liver enzymes and decreased concentrations of trace elements, especially Copper (Cu) and Selenium (Se). T. theileri was detected in a routine blood smear from one cow. The cows did not respond to an intensive care protocol, which included intravenous fluids and electrolytes, mineral substitution, non-steroidal anti-inflammatories and antibiotics, and were therefore euthanized or died. Postmortem examination showed cachexia, subcutaneous and scleral oedema and muscular dystrophy, especially in the hind limbs. Follow-up examination of the herd of origin produced similar findings including the detection of T. theileri in a large proportion of the herd. Ration analysis revealed considerable undersupply of several nutrients. Conclusions Based on all findings, an aetiological diagnosis of trace mineral and nutrient deficiency with possible involvement of T. theileri was made.

Dairy cows in modern production systems are at risk to develop metabolic disorders during the transition period. Reasons for individual differences in susceptibility, as well as the underlying pathomechanisms, are still only partially understood. The development of metaphylactic treatment protocols is needed. In this context, an on-farm prospective 3-fold blinded randomized study involving 80 German Holstein cows was performed throughout 1 yr. The trial involved a thorough recording of the production and clinical traits, clinical chemistry, and liver biopsies and blood and urine sampling at d 14 (mean: 12 d, range: 1-26 d) antepartum (AP), and d 7 (7, 4-13) and 28 (28, 23-34) postpartum (PP) for metabolomics analyses. Two groups received a treatment with butaphosphan and cyanocobalamin (BCC) at either the dosage recommended by the manufacturer or the double dosage (5 or 10 mL/100 kg of body weight 10% butaphosphan and 0.005% cyanocobalamin (Catosal, Bayer Animal Health), n = 20 in each group, parity: 4.2 ± 2.0 and 3.4 ± 1.3, respectively (mean ± SD)] and one group a placebo treatment (NaCl 0.9%, n = 40, parity: 4.0 ± 1.9). The animals were treated at 6 time points (7, 6, and 5 d AP, and 1, 2, and 3 d PP) via intravenous injection. Mass spectroscopy-based targeted metabolomics analysis of blood plasma and liver samples were performed using the AbsoluteIDQ p180 kit (Biocrates Life Sciences), whereas the urine samples were analyzed by nuclear magnetic resonance spectroscopy. Statistical analysis was performed using multivariate [partial least squares discriminant analysis (PLS-DA)] and univariate methods (linear mixed model). Multivariate data analysis (PLS-DA plots) of the liver metabolome revealed 3 different metabotypes (A = medium, B = minor, C = large alterations in liver metabolome profile between AP and PP status). Metabotype B animals were characterized by higher PP lipomobilization (stronger PP body condition decrease and higher blood bilirubin, fatty acids, gamma-glutamyltransferase, and triglyceride levels) and a higher occurrence of transition cow diseases, compared with the animals in metabotype C. Analysis of the feeding data showed that the period of metabotype B animals (calving in a distinct time frame) was characterized by a decreased grass silage quality. The PP liver metabolome of the metabotype C animals was characterized by higher concentrations of AA, acylcarnitines, lysoPC and sphingomyelins compared with metabotype B. For the metaphylactic treatment with BCC a dose-dependent effect was confirmed, differing between the metabotypes. In all matrices and metabotypes at various time points significant treatment effects were observed, with different profiles in clinical chemistry and as well in metabolomics data. The most clear-cut treatment effect was observed in metabotype B in the liver at 7 d PP, characterized by an increase in several acylcarnitines and phosphatidylcholines, indicating a more efficient influx and oxidation of fatty acids in mitochondria and thereby an increase in energy supply and more efficient triglyceride export in the liver. The results from the liver metabolomics analysis support the application of an indication-based metaphylactic treatment with BCC.

The aim was to investigate the frequency of squamous cell carcinoma and the outcome after surgical treatment in cows admitted to the clinic between 2012 and 2018. The study included six cows with squamous cell carcinoma (confirmed in 5 cattle and suspected in 1). All cattle underwent clinical examination and additional testing to confirm the diagnosis. Breed, age, gender, localization of neoplasm and treatment were recorded. In a telephonequ estionnaire owners were asked about survival, culling, integration into the herd after discharge from the clinic, wound healing and neoplasm recurrence. Five ocular squamous cell carcinomas were diagnosed during the study period. Another neoplasm on the dorsolateral thoracic wall of a cow could not be diagnosed with certainty; however, it had characteristics of squamous cell carcinoma. Two cows with ocular squamous cell carcinomawere euthanised. The prognosis and outcome of squamous cell carcinoma depend on early detection of clinical signs, the presence of metastases and the timing of tumour removal. Surgical removal of a squamous cell carcinoma at or near the eye or at other locations of the body appears to have a good prognosis.

Objective The assessment of pain in sheep is a clinical challenge, because being a prey species, they tend to mask it. Since pain in animals cannot be measured directly, various methods are applicable to detect its effects on physiological, biochemical and ethological processes. Clinicians experienced in behavioural assessment are able to reliably determine the level of pain endured by an animal. The objective of this study was to identify behaviours that reflect acute and chronic pain as well as stress in sheep. Methods The behaviour of 36 Meat Merino ewes with contagious footrot (contagious interdigital dermatitis) was analysed during normal activities in the flock and during footrot treatment, which included claw trimming to remove abnormal horn. The behaviour during treatment was compared with that of 12 healthy control sheep of the same breed during routine foot trimming. Grading of the observed behaviours was made using a numerical score. The occurrence of footrot was defined as, and equated with, chronic pain, and treatment was equated with acute pain. For the assessment of stress induced by the manipulative treatment, the data of the healthy sheep were taken as a comparison. Results The strongest indications of chronic pain were non-weight bearing lameness of an affected limb, shifting of weight and abnormal gait patterns. Additionally, sheep with footrot urinated more frequently than healthy animals. Stress-related bruxism occurred in healthy and ill sheep during foot trimming, but not during normal activities in the flock. Tail wagging and strong defensive movements of all limbs commonly occurred during footrot treatment. Head shaking appeared to be primarily stress-related in association with dorsal recumbency on the tilt table, but was exacerbated by pain elicited by hoof trimming. Conclusion The behaviours selected in this study were useful for the identification of chronic and acute pain in sheep; however, correct interpretation of behavioural changes requires an experienced individual.
