The blood-brain barrier (BBB) poses a significant challenge for drug delivery to the brain. Therefore, the development of safe methods for an effective delivery of medications to the brain can be a revolutionary step in overcoming this limitation. Using a quantum-dot-based 1267 nm laser (photosensitiser-free generation of singlet oxygen), we clearly show the photostimulation of lymphatic delivery of bevacizumab (BMZ) to the brain tissues and the meninges. These pilot findings open promising perspectives for photomodulation of a lymphatic brain drug delivery bypassing the BBB, and potentially enabling a breakthrough strategy in therapy of glioma using BMZ and other chemotherapy drugs.

The quercetin content in aqueous EtOH extracts of covering leaves of anthocyanin Purple Saratov diploid variety of Zea mays L. grown under dry conditions reached 39.36 ± 0.41 mg/100 g of dry extract and significantly exceeded the value (26.03 ± 0.36 mg/100 g of dry extract) for maize grown under normal wetting conditions. The large amounts of polyphenolic compounds in anthocyanin Purple Saratov diploid variety showed good prospects for using it as a source of them.

The deposition of amyloid-β (Aβ) in the brain is a risk factor for Alzheimer’s disease (AD). Therefore, new strategies for the stimulation of Aβ clearance from the brain can be useful in preventing AD. Transcranial photostimulation (PS) is considered a promising method for AD therapy. In our previous studies, we clearly demonstrated the PS-mediated stimulation of lymphatic clearing functions, including Aβ removal from the brain. There is increasing evidence that sleep plays an important role in Aβ clearance. Here, we tested our hypothesis that PS at night can stimulate Aβ clearance from the brain more effectively than PS during the day. Our results on healthy mice show that Aβ clearance from the brain occurs faster at night than during wakefulness. The PS course at night improves memory and reduces Aβ accumulation in the brain of AD mice more effectively than the PS course during the day. Our results suggest that night PS is a more promising candidate as an effective method in preventing AD than daytime PS. These data are an important informative platform for the development of new noninvasive and nonpharmacological technologies for AD therapy as well as for preventing Aβ accumulation in the brain of people with disorder of Aβ metabolism, sleep deficit, elderly age, and jet lag.

Detection and study of biologically active compounds seems a promising area of research in cancer diagnostics and therapies. The glycoprotein and polysaccharide fractions showing high cytotoxicity towards several human and animal cancer cell lines: A549, Hep-2, HeLa, С6 and SPEV-2 were isolated from basidiomycete Lentinus edodes vegetative mycelium and fruiting body and further characterized. It was found that water-soluble glycoprotein fractions caused the most significant, 70-100% inhibition of metabolic activity of SPЕV-2, А549 and С6 cell lines. The effective concentrations of glycoprotein fractions reducing the viability of cancer cell lines were determined. The protein and subunit composition of fractions was studied; the highly active galactose-specific lectins were found to be present in these fractions. Comparative analysis of transcriptomes of L. edodes vegetative mycelium, fruiting body and primordium revealed the presence of carbohydrate-binding glycoproteins (lectins) specific for each stage of basidiomycete morphogenesis. Histological examination revealed some morphological indicators of immune system activation and the absence of toxic effect on gastro-intestinal mucosa of animals at peroral administration of fungal glycoprotein fractions. Fungal protein and, in particular, lectin preparations derived from L. еdodes vegetative mycelium might be considered as novel prospective tools in cancer diagnostics and therapies.

The lymphatic system of the brain meninges and head plays a crucial role in the clearance of amyloid-β protein (Aβ), a peptide thought to be pathogenic in Alzheimer’s disease (AD), from the brain. The development of methods to modulate lymphatic clearance of Aβ from the brain coild be a revolutionary step in the therapy of AD. The opening of the blood-brain barrier (OBBB) by focused ultrasound is considered as a possible tool for stimulation of clearance of Aβ from the brain of humans and animals. Here, we propose an alternative method of non-invasive music-induced OBBB that is accompanied by the activation of clearance of fluorescent Aβ (Fαβ) from the mouse brain. Using confocal imaging, fluorescence microscopy and magnetic resonance tomography, we clearly demonstrate that OBBB by music stimulates the movement of Fαβ and Omniscan in the cerebrospinal fluid and lymphatic clearance of Fαβ from the brain. We propose the extended detrended fluctuation analysis (EDFA) as a promising method for the identification of OBBB markers in the electroencephalographic (EEG) patterns. These pilot results suggest that music-induced OBBB and the EDFA analysis of EEG can be a non-invasive, low cost, labelling free, clinical perspective and completely new approach for the treatment and monitoring of AD.

We report here a pilot identification of the presence of the lumenized Lyve-1/Prox-1-expressing vessels with distinct walls composed of a single endothelial layer in the unaffected brain and with intraventricular hemorrhages. These lymphatic vessels (LVs) have valves and an undulating shape in the distal region that is the classical characteristic of lymphatic precollectors. Furthermore, we identified Lyve-1/Prox-1-expressing lymphatic elements in the enlarged perivascular spaces. These pioneering results might be a revolutionary step in our understanding of anatomy and physiology of the cerebral lymphatics and stimulate a reassessment of basic assumptions in the aetiology of brain diseases associated with lymphatic dysfunction. Intraventricular hemorrhage (IVH) is the most fatal form of brain injury in adults and infants. However, therapy of IVH is very limited, and new strategies are needed to reduce hematoma expansion. For the first time, in our experiments on mice with IVH, we clearly demonstrate that transcranial low-level infra-red photostimulation (PS) accelerates the red blood cells (RBCs) evacuation from the ventricles that is associated with reducing the mortality in 1.57 times, improving emotional status and recovery of intracranial pressure. We revealed the mechanisms of PS-modulation of lymphatic contractility and tone of the lymphatic vessels underlying the lymphatic clearing functions. Our findings strongly suggest that PS-mediated stimulation of lymphatic clearance of RBCs can be a novel non-invasive, bedside, readily applicable, and commercially viable technology for the treatment of IVH.

The blood-brain barrier (BBB) poses a significant challenge for drug delivery to the brain. The limitations of our knowledge about the nature of BBB explains the slow progress in the therapy of brain diseases and absence of methods for drug delivery to the brain in clinical practice. Here we show that the BBB opens for high molecular weight compounds after exposure to loud sound (100 dB 370 Hz) in rats. The role of stress induced by loud sound and the systemic and molecular mechanisms behind it are discussed in the framework of the BBB opening as an informative platform for a novel fundamental knowledge about the nature of BBB and for the development of a non-invasive brain drug delivery technology.

Here, we demonstrate the therapeutic effects of transcranial photobiomodulation (tPBM, 1267 nm, 32 J/cm2, a 9-day course) in mice with the injected model of Alzheimer’s disease (AD) associated with accumulation of beta-amyloid (Aβ) in the brain resulting in neurocognitive deficit vs. the control group (CG) (the neurological severity score (NNS), AD 3.67 ± 0.58 vs. CG 1.00 ± 0.26%, p < 0.05) and mild cerebral hypoxia (AD 72 ± 6% vs. CG 97 ± 2%, p < 0.001). The course of tPBM improved neurocognitive status of mice with AD (NNS, AD 2.03 ± 0.14 vs. CG 1.00 ± 0.26, vs. 2.03 ± 0.14, p < 0.05) due to stimulation of clearance of Aβ from the brain via the meningeal lymphatic vessels (the immunohistochemical and confocal data) and an increase in blood oxygen saturation of the brain tissues (the pulse oximetry data) till 85 ± 2%, p < 0.05. These results open breakthrough strategies for non-pharmacological therapy of AD and clearly demonstrate that tPBM might be a promising therapeutic target for preventing or delaying AD based on stimulation of oxygenation of the brain tissues and activation of clearance of toxic molecules via the cerebral lymphatics.

A scheme of a statically determinate planar truss with two additional supports duplicating the main ones is proposed. The formula for the dependence of the lower estimate of the first natural frequency on the number of panels is obtained. The solution is compared with the numerical one. Determination of the forces in the rods by the method of cutting out the nodes and with all the transformations performed in the Maple computer mathematics system. The high accuracy of the result is shown with a large number of panels.

Music plays a more important role in our life than just being an entertainment. For example, it can be used as an anti-anxiety therapy of human and animals. However, the unsafe listening of loud music triggers hearing loss in millions of young people and professional musicians (rock, jazz and symphony orchestra) owing to exposure to damaging sound levels using personal audio devices or at noisy entertainment venues including nightclubs, discotheques, bars and concerts. Therefore, it is important to understand how loud music affects us. In this pioneering study on healthy mice, we discover that loud rock music below the safety threshold causes opening of the blood-brain barrier (OBBB), which plays a vital role in protecting the brain from viruses, bacteria and toxins. We clearly demonstrate that listening to loud music during 2 h in an intermittent adaptive regime is accompanied by delayed (1 h after music exposure) and short-lasting to (during 1–4 h) OBBB to low and high molecular weight compounds without cochlear and brain impairments. We present the systemic and molecular mechanisms responsible for music-induced OBBB. Finally, a revision of our traditional knowledge about the BBB nature and the novel strategies in optimizing of sound-mediated methods for brain drug delivery are discussed.

The extract of G. officinalis was separated by five major fractions and the antitumor activity of hydrophobic fractions has been established. For the most active fractions spectral investigations of the main component were performed using UV-Vis electronic spectra, circular dichroism (CD), Fourier-transform infrared spectroscopy (FTIR) and nuclear magnetic resonance (NMR) spectroscopy. On the basis of spectral characteristics, it was demonstrated that the main components of the fractions 4 and 5 of G. officinalis providing its biological activity, highly likely, are of flavonoid nature. Based on the NMR data we propose the structure of the aglycon moiety of the main component from fraction 4 as 4,2’,5’-trihydroxychalcone. The fraction 5 is the most active, the antitumor effect of which is implemented through cytotoxic, cytostatic, apoptotic and autophagosomal activity in respect to human kidney cancer tumor cells.

Transcranical photobiomodulation (tPBM, 1267 nm, 32 J/cm2) is effective non-invasive method for clearance of beta-amyloid from the brain in mice with Alzheimer’s disease and for improvemen of their neurological status.

In human data we observed the presence of products of degraded blood (the iron/hemosiderin) in the meningeal lymphatic vessels (MLVs) and in the deep cervical lymph node (dcLN) in newborns who died after intracranial hemorrhages. Here we demonstrate novel data related to transcranial photobiomodulation (tPBM) of clearance of blood products (the iron/hemosiderin) from the brain via MLVs in newborn rats after stress-induced intracranial hemorrhages (SIH). Experiments were carried out in 73 newborn rats (5 g) with SIH. All procedures were performed in accordance with the “Position of the American Heart Association on Research Animal Use” (Circulation 1985; 71:849A). The new generation laser 1268 nm (30 nm, 7 min) was used for tPBM of lymphatic clearance of blood products that was analyzed in vivo (two-photon, fluorescent, optical coherent tomography imaging) and ex vivo (immunohistochemical, confocal data) using markers of cerebral and lymphatic endothelium (CD31, LYVE-1/LYVE-1 conjugated with Alexa 488 and Prox-1). The results clearly demonstrated that 30 min after SIH the iron/hemosiderin (detected by atomic absorption spectroscopy and histological method) accumulated in the dcLN and observed in the MLVs. The tPBM caused stimulation of lymphatic drainage function that was accompanied by decrease in the iron/hemosiderin level in the brain tissues with an appropriate increase of their level in the dcLN. The tPBM (one time a day) during one week contributed a better recovery after SIH vs. the control group (without tPBM) that was confirmed by analysis of inflammatory factors (C-reactive protein, IL-6 and lipoprotein-associated phospholipase A2, immunoassay analysis), macrophages infiltration (confocal imaging), edema, necrotic and ischemic events (histological data). In sum, our results suggest that tPBM by stimulation of endogenous processes of cerebral lymphatic drainage and clearing function might be a novel non-invasive approach for brain recovery after neonatal intracranial hemorrhages. This seems to be the time to gain new knowledge about the meningeal lymphatic system and a deeper understanding of its anatomy and physiology. Although it is known that the meningeal lymphatics present in the layers of the brain, limited information is available about the role of this system in brain function. Here, for the first time we clearly demonstrate that the meningeal lymphatic pathway is involved in brain clearing from the blood after intracranial hemorrhage associated with hypoxia and forms a connective bridge between interstitial, cerebral spinal fluid and peripheral lymphatics. We also show that the development of methods to stimulate meningeal lymph flow after hemorrhagic evidence in the brain might be a neuroprotective strategy for effective recovery of the brain after a cerebrovascular catastrophe.

In a series of previous studies, we demonstrated that the photodynamic therapy (PDT), as a widely used tool for treatment of glioblastoma multiforme (GBM), also site-specifically opens the blood&ndash, brain barrier (BBB) in PDT-dose and age-related manner via reversible disorganization of the tight junction machinery. To develop the effective protocol of PDT-opening of the BBB, here we answer the question of what kind of photosensitizer (PS) is the most effective for the BBB opening. We studied the PDT-opening of the BBB in healthy mice using commercial photosensitizers (PSs) such as 5-aminolevulenic acid (5-ALA), aluminum phthalocyanine disulfonate (AlPcS), zinc phthalocyanine (ZnPc) and new synthetized PSs such as galactose functionalized ZnPc (GalZnPc). The spectrofluorimetric assay of Evans Blue albumin complex (EBAC) leakage and 3-D confocal imaging of FITC-dextran 70 kDa (FITCD) extravasation clearly shows a revisable and dose depended PDT-opening of the BBB to EBAC and FITCD associated with a decrease in presence of tight junction (TJ) in the vascular endothelium. The PDT effects on the BBB permeability, TJ expression and the fluorescent signal from the brain tissues are more pronounced in PDT-GalZnPc vs. PDT-5-ALA/AlPcS/ZnPc. These pre-clinical data are the first important informative platform for an optimization of the PDT protocol in the light of new knowledge about PDT-opening of the BBB for drug brain delivery and for the therapy of brain diseases.

There is a hypothesis that augmentation of the drainage and clearing function of the meningeal lymphatic vessels (MLVs) might be a promising therapeutic target for preventing neurological diseases. Here we investigate mechanisms of photobiomodulation (PBM, 1267 nm) of lymphatic drainage and clearance. Our results obtained at optical coherence tomography (OCT) give strong evidence that low PBM doses (5 and 10 J/cm2) stimulate drainage function of the lymphatic vessels via vasodilation (OCT data on the mesenteric lymphatics) and stimulation of lymphatic clearance (OCT data on clearance of gold nanorods from the brain) that was supported by confocal imaging of clearance of FITC-dextran from the cortex via MLVs. We assume that PBM-mediated relaxation of the lymphatic vessels can be possible mechanisms underlying increasing the permeability of the lymphatic endothelium that allows molecules transported by the lymphatic vessels and explain PBM stimulation of lymphatic drainage and clearance. These findings open new strategies for the stimulation of MLVs functions and non-pharmacological therapy of brain diseases.

Adsorption of the vital dye Congo red suppresses swarming of Azospirillum brasilense in a semiliquid medium, and the bacteria become able to spread with the formation of microcolonies. By using direct and stereoscopic light microscopy, the patterns of the front of Azospirillum spreading in a semiliquid medium containing the dye were analyzed. It was found that in a medium with Congo red, small motile colonies were formed among the individual cells, and once formed, they left the boundaries of the swarming front. The microcolonies produced by azospirilla in the presence of the dye were ordered bacterial structures, rather than random cell aggregates. Transmission electron microscopy revealed that the cells grown without the dye had polar flagella, whereas the cells from the medium with Congo red had no flagella and were covered with a layer of fibrillike material. Immunochemical data for the cell surface changes resulting from interaction with the dye make it possible to consider Azospirillum lipopolysaccharide as a probable Congo red receptor.

The genus-specific surface protein antigens of Azospirillum brasilense strains were visualized immunochemically. The procedure used for cell sample preparation was optimized to ensure that the surface protein structures were detected on cells in situ. Gold and gold-silver nanoparticles were conjugated to antibodies raised against the flagellin of A. brasilense type strain Sp7, against the lipopolysaccharide of A. brasilense Sp245, and against the genus-specific protein determinants of A. brasilense Sp7. Electron microscopic analysis using nanoparticle-labeled antibodies revealed antigenic determinants of the polar flagellum on the A. brasilense Sp245 cell surface, which in these bacteria are normally screened from the surroundings by a lipopolysaccharide sheath. Pili-like structures were detected on the Sp245 wild-type strain and on its Fla– Swa– Omegon-Km mutant SK048, which are presumably involved in microcolonial spreading in these bacteria.

The chemical composition of a new bioactive extract from immortelle sandy (Helichrysum arenarium (L.) Moench.) was investigated. Naringin, its soluble aggregate, prunin, quercetin, apigenin, naringenin, apigenin 5-O-glucoside, and isosalipurposide were found among the flavonoids. The tested immortelle sandy extract was shown to contain known dimers, trimers, or more complex aggregates. The compounds had similar spectral but substantially different chromatographic characteristics, which can be used for their identification based on the corresponding data on flavonoids glycosylated at different positions and/or carbohydrates of various complexities, which are also characterized by similar absorption spectra and different retention times. It was found by molecular absorption spectroscopy that the immortelle extract contained 73.48 mg of flavonoids relative to rutin or 17.94 mg relative to quercetin per 1 g of dry extract weight, which corresponded to 20.99 and 5.13%, respectively. The immortelle extract obtained by the method proposed displayed antitumor activity against the transplanted sarcoma 45 and had a beneficial effect on animals in general.

Glycosylated flagellin of the polar flagellum of the plant growth-promoting rhizobacteria Azospirillum brasilense Sp245 was for the first time isolated and characterized by biochemical and bioinformatics methods. Using the amino acid sequence taken from the NCBI database of bacterial whole-genome DNA sequencing, the secondary and tertiary structures of the protein part of this glycoprotein were determined by template-based molecular modeling. With the use of a set of predictors, regions of its intrinsic structural disorder were identified, and binding sites of carbohydrate fragments to the surface of the molecule were determined. A positive effect of the polar flagellum flagellin on the root meristem of wheat seedlings was for the first time revealed for associative bacteria. The effect was manifested in an increase in the division rate of plant cells - a significant increase in the mitotic index. Thus, the induction of specific responses of plants to their interactions with flagellin of the associative bacteria may probably be considered as a demonstration of its elicitor properties.

Paenibacillus polymyxa 92, isolated from wheat roots, produced large amounts (38.4 g L−1) of exopolysaccharide (EPS) in a liquid nutrient medium containing 10 % (w/v) sucrose. The EPS was precipitated from the culture broth with cold acetone and was purified by gel filtration and anion-exchange chromatography. The molecular mass of the EPS was 2.29–1.10 × 105 Da. Diffuse reflectance infrared Fourier transform and nuclear magnetic resonance spectra showed that the EPS was a linear β-(2→6)-linked fructan (levan). Aqueous EPS solutions showed pseudoplastic behavior when shear stress was applied at different temperatures. By using the Ostwald–de Waele model, the rheological characteristics of the EPS solution were ascertained. The sorption capacity of the EPS for Zn(II), Cd(II), Pb(II), and Cu(II) was investigated. Sorption was maximal (q = 481 mg g−1) for Cu(II) ions. In model experiments, treatment of wheat seeds with EPS solution significantly increased the length of seedling roots and shoots.

The role of sound, sound-induced stress and molecular mechanisms behind is discussed in the framework of BBB opening as an informative platform for a novel fundamental knowledge about the nature of BBB and for the development of a non-invasive brain drug delivery technology.Social aspects of music/sound-induced opening of BBB provide completely new information about noise and healthy life conditions that will stimulate new research in this field.

Alzheimer&rsquo, s disease (AD) is an incurable pathology associated with progressive decline in memory and cognition. Phototherapy might be a new promising and alternative strategy for the effective treatment of AD, and has been actively discussed over two decades. However, the mechanisms of therapeutic photostimulation (PS) effects on subjects with AD remain poorly understood. The goal of this study was to determine the mechanisms of therapeutic PS effects in beta-amyloid (A&beta, )-injected mice. The neurological severity score and the new object recognition tests demonstrate that PS 9 J/cm2 attenuates the memory and neurological deficit in mice with AD. The immunohistochemical assay revealed a decrease in the level of A&beta, in the brain and an increase of A&beta, in the deep cervical lymph nodes obtained from mice with AD after PS. Using the in vitro model of the blood-brain barrier (BBB), we show a PS-mediated decrease in transendothelial resistance and in the expression of tight junction proteins as well an increase in the BBB permeability to A&beta, These findings suggest that a PS-mediated BBB opening and the activation of the lymphatic clearance of A&beta, from the brain might be a crucial mechanism underlying therapeutic effects of PS in mice with AD. These pioneering data open new strategies in the development of non-pharmacological methods for therapy of AD and contribute to a better understanding of the PS effects on the central nervous system.

Here we show the photodynamic treatment (PDT) causes significant increase in the permeability of the blood-brain barrier (BBB) in healthy mice. Using different doses of laser (635 nm, 10-40 J/cm2) and photosensitizer (5-aminole-vulinic acid – 5-ALA, 20 and 80 mg/kg, i.v.), we found the optimal PDT for the reversible opening of BBB that is 15 J/cm2 and 5-ALA, 20 mg/kg, when the brain tissues recover 3 days later. Further increase in the laser or 5-ALA doses has not amplifying effect on the BBB but associated with severe damages of brain tissues. These results can be good informative platform for the further studies of new strategies in brain drug delivery and for the better understanding of mechanisms underlying cerebrovascular effects of PD-related fluorescence guided resection of brain tumor.

The structure of potential bacteriophage receptors located on cell walls of Gram-negative bacteria deposited at Belarusian collection of microorganisms was investigated. Studies by 1D and 2D 1H and 13C NMR spectroscopy enabled to elucidate the structure of the O-specific polysaccharides (OPS) constituting lipopolysaccharide (LPS) of some Pseudomonas species. The capacity of bacteriophage to adsorb to LPS molecules was tested.

A new application of the photodynamic treatment (PDT) is presented for the opening of blood-brain barrier (BBB) and the brain clearing activation that is associated with it, including the use of gold nanoparticles as emerging photosensitizer carriers in PDT. The obtained results clearly demonstrate 2 pathways for the brain clearing: (1) using PDT-opening of BBB and intravenous injection of FITC-dextran we showed a clearance of this tracer via the meningeal lymphatic system in the subdural space; (2) using optical coherence tomography and intraparenchymal injection of gold nanorods, we observed their clearance through the exit gate of cerebral spinal fluid from the brain into the deep cervical lymph node, where the gold nanorods were accumulated. These data contribute to a better understanding of the cerebrovascular effects of PDT and shed light on mechanisms, underlying brain clearing after PDT-related opening of BBB, including clearance from nanoparticles as drug carriers.

Malignant gliomas are highly invasive tumors that use the cerebral vessels for invasion due to high vascular fragility of the blood–brain barrier (BBB). On one hand, glioma is characterized by the BBB disruption, on the other hand, drug brain delivery via the BBB is a big challenge in glioma therapy. The limited information about vascular changes associated with glioma growth is a reason of slow progress in prevention of glioma development. Here, we present in vivo and ex vivo study of the BBB disruption and glioma cells (GCs) migration in rats using fluorescence and confocal microscopy. We uncovered a local breach in the BBB in the main tumor mass but not within the border of normal and malignant cells, where the BBB was impermeable for high weight molecules. The migration of GCs were observed via the cerebral vessels with the intact BBB that was associated with macrophages infiltration. The mechanisms underlying glioma progression remain unknown but there is an evidence that the sympathetic nervous system (SNS) via activation of vascular beta2-adrenoreceptors (B2-ADRs) can play an important role in tumor metastasis. Our results clearly show an increase in the expression of vascular B2-ADRs and production of the beta-arrestin-1 — co-factor of B2-ADRs signaling pathway in rats with glioma. Pharmacological blockade of B2-ADRs reduces the BBB disruption, macrophages infiltration, GCs migration and increases survival rate. These data suggest that the blockade of B2-ADRs may be a novel adjuvant therapeutic strategy to reduce glioma progression and prevent metastasis.
