Context: Cellular abnormalities in the ducts and breast tissue cause breast cancer to invade nearby tissues like the liver. The fibroblast growth factor (FGF) signals are sent to its receptor and regulate various cellular processes to maintain liver homeostasis. However, this condition has few and inefficient treatments. Breast cancer is characterized by the development of cellular abnormalities within the ducts and breast tissue, leading to the invasion of adjacent tissues, such as the liver. Aims: To examine the impact of Rhodomyrtus tomentosa administration on the expression of the FGF family in cancer animal models, with a specific focus on the liver. Methods: Cancer model rats were administered at dosages of 100, 200, and 300 mg/kg BW for 30 days. Liver tissue and blood serum samples were extracted from the rat. Liver tissue was stained for immunohistochemistry using FGF1, FGF15, FGF19, and FGF21, and blood samples were collected for ELISA analysis. Results: The study found that DMBA in hepatocyte cells degrades parenchyma, making it hydrophilic and necrosating. Hepatocyte cell function improved with the greatest dose of R. tomentosa. The elevation was associated with an area around the portal vein where recently dividing hepatocytes form clusters. Conclusions: The promise of R. tomentosa as a candidate for the development of hepatoprotective medicines in cancer treatment arises from its ability to influence the expression of crucial liver health markers, including FGF1, FGF15, FGF19, and FGF21.

Objective: Matrix metalloproteinase (MMP-1 and −9), Epidermal Growth Factor (EGF), and Transforming Growth factor (TGF)-β are expressed in the oral ulcer wound-healing process. The Adipose mesenchymal stem cell metabolites (AdMSCMs) may accelerate wound-healing. This study aimed to investigate the expression of MMP-1, MMP-9, EGF, and TGF-β in the oral mucosa ulcer rat model treated with topical AdMSCMs. Materials and Methods: Oral ulcer models were created in twenty healthy male Wistar rats (Rattus novergicus) divided into AdMSCMs and control groups. The oral ulcer model was treated topically using AdMSCMs oral gel three times daily for 3 and 7 days. The expression of MMP-1, MMP-9, EGF, TGF-β was evaluated through histological examination using the immunohistochemistry method. Independent t-test was used to compare the mean of expression of MMP-1, MMP-9, EGF, TGF-β between control and treatment groups (AdMSCMs), and paired t-test was used to analyze the mean between day 3 and day 7 of each group. Results: A lower expression of MMP-1, MMP-9 in AdMSCMs group and higher expression EGF and TGF-β in AdMSCMs group compared to the control group in day 3 and day 7. Independent t-test results showed a significant difference in the expression of MMP-1, MMP-9, EGF between the control and AdMSCMs group in day 3 and day 7. Orthodontic relapse, the return to a pre-treatment position after orthodontic correction, is driven by the RANKL/TNF-α-mediated ROS/Keap1/Nrf2 signaling axis. This mechanism triggers aseptic inflammation and oxidative stress, influencing bone resorption and formation. Antioxidants can mitigate oxidative stress, potentially improving post-orthodontic outcomes. This study explores the efficacy of antioxidant compounds derived from green seaweed/algae in managing orthodontic relapse. Green seaweed/algae extracts were prepared via sonication, and bioactive compounds were identified using ultra-performance liquid chromatography-electrospray ionization-tandem mass spectrometry (UPLC-ESI-MS/MS) analysis. Compounds underwent bioactivity prediction, toxicity assessment, and drug-likeness evaluation, revealing significant therapeutic potential. Network pharmacology and molecular docking identified key proteins associated with orthodontic relapse, including IL-1β, STAT3, ESR1, MAPK1, JAK2, and HMOX1. Molecular docking simulations indicated favorable binding energies for green seaweed compounds, particularly the alkaloids adenosine (ΔG −6.9 to −7.3 kcal/mol) and lycopodine (ΔG −6.3 to −8.5 kcal/mol), against targeted proteins, matching or outperforming standard drugs such as s-ibuprofen (ΔG −6.7 kcal/mol). In vitro assays confirmed the antioxidant activity of these compounds, with EC50 dose of 52.2–54.2 μg/mL for ABTS radical scavenging capacities. Protein expression analysis in tibial-femoral bone marrow cells further demonstrated the potential of green seaweed/algae compounds to suppress osteoclastogenesis by modulating the RANKL/TNF-α-mediated ROS/Keap1/Nrf2 pathway. This research highlights the promise of green seaweed-derived antioxidants in reducing oxidative stress and managing orthodontic relapse, providing a foundation for future therapeutic developments.

Background: Dental caries is frequently observed in children. Increasing the awareness of oral health among school-age children is anticipated to decrease the occurrence of dental caries. Purpose: The aim of this study was to determine the prevalence of dental caries among students in Medowo III elementary school using dmft/DMFT index. Methods: A cross-sectional descriptive study was carried out among the entire student at Medowo III elementary school. The dental caries of each student were assessed using the dmft and DMFT indices. The assessment of variable distribution was conducted using the Shapiro-Wilk test. The dmft/DMFT values were compared between different age groups and genders using either the Student's t-test or the Mann-Whitney test. The data was processed using a significance level of p < 0.05. Results: The study included a total of 43 students. The average caries index in primary dentitions was markedly greater than in permanent dentitions. According to WHO, the prevalence of dental caries was very high in the primary dentition of students aged 7-9 years. No notable disparities were detected between female and male students in relation to the caries index of both primary and permanent teeth. However, the average caries index of primary teeth was considerably higher in middle-childhood students compared to preadolescence students. Conclusion: The dental caries prevalence among students in Medowo III elementary school were still high. Therefore, oral health prevention program was needed in order to enhance the community's awareness toward dental caries.

Coffee is a kind of daily beverage, and its correlation with cardiovascular disease and atherosclerosis is still debatable. The aim of this study is to investigate the effects of Amstirdam coffee extract (ACE) on lipoprotein-associated phospholipase A2 (Lp-PLA2) and the inflammatory response in atherosclerosis mouse models. The study used 25 Swiss male mice for five groups (n = 5): healthy mice fed a normal diet (N); mice fed a high-fat, high-fructose diet (HFFD); mice fed HFFD and treated with ACE at doses of 104 (D1), 520 (D2), and 5200 mg/kg BW (D3). The levels of Lp-PLA2, regulatory T cells (Tregs) (CD4+CD25+CD62L+, CD4+CD25+IL-10+, CD4+CD25+TGF-+), IL-10 (CD4+IL-10+), and TGF-B (CD4+TGF+) were analyzed using a flow cytometer. Histological analysis of the mouse aorta was done by hematoxylin and eosin (HE) staining. This study indicated a significant increase in total cholesterol (TC), triglyceride (TG), LDL, and Lp-PLA2 levels in the HFFD group. HFFD also reduced HDL, IL-10, and TGF produced by CD4 and Tregs compared with the normal group. ACE at all doses significantly reduced Lp-PLA2 levels compared with the HFFD group (p < 0.05). Interestingly, the administration of 520 mg/kg BW ACE (D2) increased the production of IL-10 significantly compared to other doses (p < 0.05). The D3 group possessed a high TGF- production and Treg expression level significantly different between groups (p < 0.05). Foam cells were mostly found in the aorta of the HFFD group compared to the normal and ACE treatment groups. This study suggested that ACE could reduce Lp-PLA2 enzyme activity and foam cell formation through the immunosuppressive activity of IL-10 and TGF cytokines.

This study evaluated the physicochemical properties of toothpaste from combined Caulerpa racemosa and Thunnus fish bone (Toothpaste Caulerpa and Thunnus or TCT) and its antibacterial activity towards the colonization of selected cariogenic and periodontal bacterias. Four forms of toothpaste which contained C. racemosa extract and calcium carbonate or bone isolates of tuna and control (F1 (1.5:45); F2 (3:45); F3 (4.5:45); F4 (0:45)) were compared and analyzed for antioxidant activity (DPPH assay), organoleptic (sensory), homogeneity, viscosity, pH, and foamability. Antibacterial activity tests were conducted on Streptococcus mutans, Staphylococcus aureus, and Porphyromonas gingivalis. The antioxidant activity of the group’s (F1, F2, F3, F4) p=0.0001 differed considerably (CI 95%). F3 was the most antioxidant-active formula, with 27.46 ± 3.09%. Background: Nutritional insufficiency and stunting during infant growth and development also causes a delay in the ossification centers and may lead to poor skeletal and dental forms resulted in malocclusion. Early interceptive orthodontics can eliminate or reduce the severity of a developing malocclusion, the complexity of orthodontic treatment, the overall treatment time, and the cost. Proper oral and dental health knowledge and a favorable attitude toward oral and dental health are critical in preventing numerous dental issues such as malocclusion. Purpose: the aim of this community empowerment is to enhance the knowledge of preventive orthodontic treatment for malocclusion and stunting in elementary school children. Methods: The data was collected at Miftahul Ulum Melirang's Madrasah Ibtidaiyah, or elementary school, in Melirang Village, Bungah District, Gresik, East Java. For the community empowerment program assessment, participants were requested to fill out a Google form for the pre- and post-test to identify the enhancement of knowledge about or preventative orthodontic treatment for malocclusion and stunting in primary school children. Results: The majority of the community empowerment participants understood the presentation on oral and dental health empowerment. The post-test percentage was greater (91.5%) than the pre-test rate (53.75%). Conclusion: The dental and oral health empowerment program about preventive orthodontics, malocclusion, and stunting was effectively completed, enhancing the knowledge level of elementary school children.

The prevalence of breast cancer among patients in Indonesia is significant. Indonesian individuals maintain the belief that cancer cannot be cured alone by pharmaceuticals and treatment; herbal remedies must be used in conjunction. Rhodomyrtus tomentosa, also known as Haramonting, is an indigenous Indonesian medicinal plant renowned for its copious antioxidant properties. The objective of study was to assess the impact of haramonting on breast cancer by examining the expression of various biomarker proteins associated with breast cancer. Haramonting was administered to breast cancer model mice at different doses over a period of 30 days. Subsequently, blood and breast samples were obtained for immunohistochemistry and enzyme-linked immunosorbent assays (ELISA). Authors have discovered that there has been a notable rise in the proliferation of epithelial cells in the duct lobes, resulting in the formation of ducts and lobules. Additionally, the researchers discovered that the breasts exhibited distinct clinical and histological alterations. Haramonting possesses the capacity to restore the concentrations of malondialdehyde (MDA) and superoxide dismutase (SOD) to normal levels in the blood serum of rats afflicted with cancer. The histopathological analysis of the breast tissue revealed elevated levels of Her2, IL33, EGFR, and MUC1. The authors also discovered a notable increase in the growth of epithelial cells, with two or more layers of cells reaching towards the centre of the duct. The size of the epithelial cells exhibits variability; however, this state ameliorates with the administration of a dosage of 300 mg/kgBW of this botanical specimen. Inhaling Allethrin (C19H26O3) may induce oxidative stress in lung cells by causing the formation of free radi-cals. Interleukins (IL) are a group of secreted cytokines or proteins and signaling molecules initially produced as an immune response by leukocytes. Rhodomyrtus tomentosa (Aiton) Hassk. (haramonting) contains antioxidants that may prevent lung damage induced by allethrin-containing electric mosquito repellents. In this study, six groups of rats were exposed to allethrin via an electric mosquito repellent, including positive, negative, and comparison control groups and three groups were administered Rhodomyrtus tomentosa (Aiton) Hassk at 100 mg/kg BW, 200 mg/kg BW, and 300 mg/kg BW. After 30 days, the pulmonary tissue and the blood were taken for immunohisto-chemical and ELISA analysis. The accumulation of inflammatory cells causes the thickening of the alveolar wall structures. Injuries were more prevalent in the A+ group than in the other groups. The connection between the alveoli and blood capillaries, which can interfere with alveolar gas exchange, is not regulated, and the lu-minal morphology is aberrant, causing damage to the alveolar epithelial cells. Exposure to electric mosquito coils containing allethrin can increase the expression of interleukin-1, interleukin-8, interleukin-9, and interleu-kin-18 in blood serum and tissues while decreasing the expression of interleukin-6 and interleukin-10. Like the Vitamin C group, Rhodomyrtus tomentosa can increase alveolar histological alterations by decreasing the ex-pression of IL-1β, IL-8, IL-9, and IL-18 while increasing IL-6 and IL-10. So that this plant can be developed in the future as a drug to prevent lung harm from exposure.

Background: Mumps virus (MuV) can trigger severe infections, such as parotitis, epididymo-orchitis, and meningitis. The effectiveness of MuV vaccine administration has been proven, but current outbreaks warrant the development of antivirals against MuV. Zingiber officinale var. Roscoe or ginger is often used as an alternative remedy. Currently, there are no known in vitro or in vivo studies that investigate ginger as an MuV antiviral. Purpose: This study aims to evaluate the antiviral potency of the bioactive compounds in Zingiber officinale var. Roscoe against MuV. Methods: Antiviral activity screening was conducted by druglikeness analysis, antiviral probability, molecular docking, and molecular dynamic simulation. Results: As an antiviral, 6-shogaol from Zingiber officinale var. Roscoe has potency against MuV. It has a good binding affinity and can establish interactions with the binding domain of the target protein by forming hydrogen, Van der Waals, and alkyl bonds. Conclusion: The complex of 6-shogaol_NP was predicted to be volatile but stable for triggering inhibitory activity. However, these results must be proved by in vivo and in vitro approaches to strengthen the scientific evidence.

Background: Peri-implantitis is one of many factors that can cause implant failure, with common cases ranging from 1%-47% and the highest incidence ranging from 10.7%- 47.2%. Mechanical debridement (MD) is currently the standard for peri-implantitis treatment. However, MD has limitations in the removal of infected tissue. Moreover, the rough texture of the implant’s surface and bacteria adhesion and colonization increases the difficulty in performing MD. To overcome these limitations, adjunct therapy is needed to increase peri-implantitis treatment effectiveness. One of those adjunct therapies, photodynamic therapy (PDT), is used to destroy bacterial cells and significantly reduce inflammatory cell infiltration around the implant. Purpose: To describe the effectiveness of PDT as an adjunct therapy to MD in periimplantitis treatment through narrative review. Review: PDT is effective in reducing the number of bacteria, plaque index (PI), bleeding on probing (BOP), probing depth (PD), crestal bone loss (CBL), and excessive proinflammatory cytokines (IL-6, IL-1β, TNF-α) in patients. However, the effectiveness of PDT can be influenced by several factors, including patients’ conditions, such as diabetes and smoking habits, types of photosensitizers used, and exposure time. Conclusion: PDT is an effective adjunctive therapy to MD in peri-implantitis treatment since it can improve clinical parameter values, significantly reduce P. gingivalis, and decrease proinflammatory cytokines.

Background: Maxillary canines play a critical point in creating an aesthetic smile as they support the part of facial muscle. Canines are also the second most frequently impacted teeth after the third molars. In some cases, it is possible to retract canines into their correct position by orthodontics treatment. Purpose: This article highlighted a treatment option with surgical exposure, in which a maxillary canine was impacted. It presented gradual steps from pre-surgical to post-surgical orthodontic treatment. Case: A 16 years old female patient referred to Orthodontic Department by Oral Surgeon for pre-surgical orthodontic treatment of her impacted maxillary canine. Case Management: After pre-surgical orthodontic treatment, the impacted canine was surgically exposed by closed technique since open surgical exposure might need excessive removal of the surrounding bone. Traction was given through a gold chain which attached to the palatal surface of the impacted canine. The tooth was ideally positioned with fixed orthodontic appliances. The permanent right maxillary canine was successfully positioned into proper alignment with the remaining teeth. Aesthetic smile was improved. Conclusion: Malocclusion with impacted canine was successfully treated with MBT orthodontic prescription combined with gold chain that can retract the impacted canine into its physiological position and the patient was satisfied with the aesthetic result.

Context: Orthodontic tooth movement (OTM) changes the periodontal tissue and increases the incidence of root resorption (OIRR). Caffeic acid phenethyl ester (CAPE), an antioxidant and anti-inflammatory chemical generated from honey propolis, might be useful in controlling inflammation during OTM and so reducing the risk of OIRR. Aims: To evaluate if CAPE supplementation has an anti-inflammatory impact on tumor necrosis factor-α (TNF-α) and nuclear transcription factor κB (NF-κB) during experimental OTM in male Wistar rats (Rattus novergicus). Methods: Forty-eight healthy male Wistar rats were divided into positive control group (OTM 10 g/mm2 force application) and experimental group (OTM application and CAPE administration). Each groups were observed for 3, 7, 14 days. Context: Microvesicle is a cell micro molecule that may play a role in the process of osteosarcoma stem cell apoptosis. Aims: To investigate the activity of peripheral blood mononuclear cells (PBMCs) through the secretion of interleukin-2 (IL-2), interleukin-6 (IL-6), interleukin-10 (IL-10), C-X-C Motif Chemokine Ligand 13 (CXCL13) and tissue inhibitor of metalloproteinases-3 (TIMP-3) on co-cultivation of peripheral blood mononuclear cells (PBMCs) sensitized by mesenchymal stem cell secretome (MSCS) co-cultivated with osteosarcoma stem cells (OS-SCs). Methods: This study was true experimental with a post-test only control group design. This was in vitro study PBMSCs sensitized by MSCS as then samples were divided into 4 treatment groups, respectively: Zero-day treatment (P0) PBMCs were co-cultivated with OS-SCs for 0 hours; First treatment (P1) PBMCs were co-cultivated with OS-SCs for 1 hour; Second treatment (P2) PBMCs were co-cultivated with OS-SCs for 2 days; Third treatment (P3) PBMCs were co-cultivated with OS-SCs for 4 days. The examination method used in this study was flow cytometry and indirect enzyme-linked immunosorbent assay (ELISA). The data were statistically analyzed with analysis of variance (ANOVA) with a p≤0.05 considered a significant difference. Results: There was a tendency for a significant increase in extravesicular secretion in the secretion of IL-2, IL-6, IL-10, CXCL13, TIMP3, in the microvesicle PBMCs when sensitized by MSCs secretome co-cultivated with OS-SCs environment co-cultivated after the fourth day with significantly different between groups (p≤0.05). Conclusions: PBMSCs’ microvesicle such as IL-2, IL-6, IL-10, CXCL13, TIMP3 was significantly sensitized by MSCS and co-cultivated with OS-SCs after the fourth day of in vitro.

Context: Modified polyether ether ketone (PEEK) by adding nano-hydroxyapatite (HA) material on its fixture for mini-implant fabrication may increase resistance force through osseointegration. Aims: To analyze the binding molecular docking of PEEK incorporated with HA as a biomaterial candidate for orthodontic mini-implant fabrication through a bioinformatic approach, an in silico study. Methods: 3D ligand structure consisting of HA, PEEK and target proteins consisting of osteopontin, osteocalcin, osteonectin, bone morphogenetic protein 4 (BMP4), bone morphogenetic protein 2 (BMP2), bone morphogenetic protein 7 (BMP7), alkaline phosphatase (ALP), runt-related transcription factor 2 (RUNX2), Insulin growth factor-1 (IGF-1), osterix, tartrate-resistant acid phosphatase (TRAP), collagen alpha-1 (COL1A1) obtained from RCSB-PDB. It was analyzed the binding affinity of a single HA, PEEK, and HA + PEEK complex to twelve target proteins related to osseointegration. The types of chemical interactions produced by the ligands in the target protein domain consisted of Van der Waals, hydrogen, hydrophobic, pi, and alkyl. Results: The blind docking simulation succeeded in identifying the most negative binding affinity; it was found in the HA + PEEK molecular complex compared to HA and PEEK in the single condition. The type of chemical interaction formed consisted of hydrogen, van der Waals, pi, and alkyl. HA+PEEK showed the most negative binding affinity with ALP and IGF-1, as much as -8.7 binding affinity. Conclusions: The molecular docking of PEEK with HA exhibited a prominent binding affinity with osteogenic markers like ALP and IGF-1 in silico, allowing it to have a higher potential than nano-HA or PEEK as a single biomaterial for osseointegration as the fabrication of mini-implants that may support orthodontic treatment.

Context: Metal allergy is a general term to describe allergic diseases due to the release of metal ion reactions in the body which are mediated by T cells and involve inflammatory cytokines that can cause morbidity and mortality. Molecular docking is an analysis that can be used to assess the interaction of ligand bonds with target proteins that are used to predict metal allergies caused by metal ions that stimulate cytokines. Aims: To analyze the binding sites of Cu2+, Fe2+, Mn2+, Mn3+, Fe3+, CrO42-, Si4+, and Hg+ ions on cytokines to predict dental metal allergy through a bioinformatics approach, in silico. Methods: Metal ion particles consisting of Cu2+, Fe2+, Mn2+, Mn3+, Fe3+, CrO42-, Si4+, and Hg+ were predicted to bind tumor necrosis factor-α (TNF-α), interferon-γ (IFN-γ), interleukin (IL) IL-1b, IL-2, IL-4, IL-10, IL-13, IL-17, IL-23, and IL-33 act as target proteins were examined. Results: The blind docking simulation succeeded in identifying the comparison of the binding activity of metal ion particles on cytokines target proteins. The docking simulation results show that the metal ion with the most negative binding affinity value binds to the IL-17 protein. Conclusions: Metal ion particles consisting of Cu2+, Fe2+, Mn2+, Mn3+, Fe3+, CrO42-, Si4+, and Hg+ have the most negative binding affinity values for binding to IL-17 protein, which can cause allergic reactions predicted by molecular docking, in silico.

Background: Utilizing the bioactive compounds found in pigmented rice might significantly reduce the risk of breast cancer. This study aims to systematically review existing literature on the benefit of Asian pigmented rice bioactive compounds and their implication in breast cancer. Methods: Searches of the literature were conducted in two databases (Scopus and PubMed) for a systematic review. The keywords resulted in a total of 407 articles, consisting of 103 PubMed and 304 Scopus articles. 32 manuscripts were excluded because the article was over 10 years old. After excluding book chapters and non-English languages, we had 278 potential articles to be reviewed. After checking and screening the title and abstract and eliminating duplicate articles, then 66 articles were obtained. After the selection and elimination of the full-text manuscripts, finally 10 of them which met the inclusion criteria. Result: The included studies in this review were entirely based in Asia. The year of publication ranged from 2013 to 2020. Half of included studies used black rice extract, two used red jasmine rice extracts, and three used Korean rice extracts (black, red, dark purple and brown rice). All studies were conducted in vitro and three studies were compared with in vivo tests on female mice. The pigmented rice is mainly black, red, and dark purple rice, and contains a variety of peonidin-3-glucoside, cyanidin-3-glucoside, γ-oryzanol, γ-tocotrienol, proanthocyanidin, cinnamic acid, and anthocyanins that may act as pro-apoptotic, anti-proliferative, and anti-metastasis of the breast cancer cells. Conclusion: Pigmented rice is a beneficial food which possessed bioactive compounds that may have significant potential concerning a breast cancer.

Context: Oral manifestations that arose from COVID-19 infection often causes morbidity and systemic drug administration is less effective. Roselle flower (Hibiscus sabdariffa) is one of the plants that is often used in infusion as it gives health benefits. Hence, H. sabdariffa may benefit from adjuvant therapy to treat oral manifestation due to COVID-19. Aims: To investigate the potential of H. sabdariffa anthocyanins, tartaric acid, and ascorbic acid chemical compounds as antiviral, anti-inflammatory, antioxidant, and increasing tissue regeneration in oral manifestation due to COVID-19 infection in silico. Methods: Chemical compounds consisted of anthocyanins, (+)-tartaric acid, and ascorbic acid beside target proteins consisted of ACE2-spike, Foxp3, IL-10, IL6, IL1β, VEGF, FGF-2, HSP70, TNFR and MDA-ovalbumin were obtained from the database, ligand samples were selected through absorption, distribution, metabolism, excretion and toxicology analysis, then molecular docking simulations, identification of protein-ligand interactions, and 3D visualization were performed. Results: Anthocyanins, tartaric acid, and ascorbic acid are the active compounds in H. sabdariffa, which act as antioxidants. The activity of anthocyanin compounds is higher than other compounds through value binding affinity, which is more negative and binds to specific domains of target proteins by forming weak binding interactions that play a role in biological responses. Anthocyanins have the most negative binding energy compared to tartaric-acid and ascorbic acid. Conclusions: Anthocyanins act as antioxidants; this mechanism increases heat shock protein-70 (HSP70), which may play an important role in increasing wound regeneration of oral manifestation in severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) as documented in silico.

Background: Dental caries is the most common dental and oral health condition. Children's growth and development may be hampered by dental cavities. Furthermore, it may raise the risk of stunting owing to a lack of dietary intake, which may have an impact on quality of life. Oral and dental health empowerment is required to increase dental health knowledge and understanding of dental disorders, particularly during the corona virus disease-19 (COVID-19) pandemic. Objective: to describe oral and dental health knowledge improved following dental health empowerment at Madrasah Ibtidaiyah Miftahul Ulum Elementary School in Melirang, Bungah Distric, Gresik City, East Java. Material and Methods: Three surveyors were responsible for data collecting. This study's sample size was 150 people ranging from fourth to sixth grade. The seminar covered dental and oral health and was provided via teledentistry. The participants were asked to fill out a Google form for the pre- and post-test to determine the improvement of oral and dental health knowledge for the dental health empowerment evaluation. Result: The majority of oral and dental health participants understood the presentation on oral and dental health empowerment. The post-test percentage was higher (93.59%) than the pre-test rate (56.81%). Conclusion: The program findings demonstrated that oral and dental health empowerment via teledentistry was an effective medium for increasing elementary school children's understanding of dental and oral health status.

The aim of this systematic review is to describe and identify the prospects of β-Tricalcium Phosphate (β-TCP) as an alveolar bone grafting (ABG) material in cleft lip/palate (CL/P) or alveolar bone cleft defects. A systematic review protocol based on the Preferred Reporting Items for Systematic Reviews and Meta-Analyses 2020 (PRISMA 2020) was drafted. The literature search was conducted using MEDLINE/PubMed, Web of Science/ISI Web of Knowledge, Scopus, and the Cochrane Library, with English as the inclusion criterion and no publication year limits. The keywords yielded a total of 5824 publications. After removing duplicates and non-English articles, there were 3196 suitable articles available for evaluation. Subsequently, 1315 studies remained after reviewing titles and abstracts. Furthermore, 85 full articles were assessed for eligibility. After reading the complete texts of those papers, 20 were eventually selected that matched the inclusion requirements. Thirteen out of the twenty studies included in this systematic review were deemed to have a low risk of bias; one had a high risk of bias; and six had a moderate risk of bias due to not reporting randomization. β-TCP, when used as an ABG material, is biocompatible, visible, practical, offers a less invasive procedure, and does not interfere with orthodontic treatment. Synthetic β-TCP for ABG can be an alternative to autologous bone grafts under certain terms and conditions. The efficacy of β-TCP for ABG in CL/P or alveolar bone cleft defects can be enhanced through a tissue engineering approach that combines β-TCP with growth factors, mesenchymal stem cells, or other graft materials, along with modifications to β-TCP’s physical properties.

Background: The elementary school period is a golden age for instilling the values of clean and healthy living behaviour (CHLB) and has the potential as an agent of change to promote CHLB in schools, families and communities so as to create quality human resources in the future. Currently children are very vulnerable to dental and oral health, because at that age there is a transition or change of teeth, namely from deciduous teeth to permanent teeth, for that special attention needs to be given to dental and oral health so that the growth and development of teeth can be maintained properly. Purpose: to enhance knowledge of CHLB and screening of dental malocclusion in an effort to improve the quality of life related to dental and oral health. Methods: 116 participants from grades 3-5 elementary school students. Data was collected by means of intraoral examination and filling out pre-test and post-tests questionnaires. Furthermore, an analysis of the frequency distribution data was carried out by mean of statistical analysis. Results: male students had higher caries (59 people) and those who were not caries (4 people), compared to girls (47 people) and those who were not caries (6 people). CHLB knowledge level is good (86.8%), moderate (8.2) low (5%). Conclusion: caries in boys is higher than in girls and all respondents’ knowledge of CHLB is good.

Context: The most common biomaterial used for dental implants is titanium. However, the release of metal ions and the risk of allergic reactions to metals that may occur in some patients cannot be avoided. Hydroxyapatite-polymethylmethacrylate (HA-PMMA) composite biomaterials are proposed to have potential as dental implant biomaterials due to their mechanical, chemical, and biological properties. HA-PMMA may induce osseointegration, biocompatible, less allergic reactions, and no metal ions released. In addition, HA-PMMA can be obtained from Indonesia’s abundant natural resources. Aims: To explore HA-PMMA composites through molecular docking as a biomaterial candidate for dental implants in silico. Methods: Structure data format (sdf), molecular weight, and identity number (CID) of HA-PMMA ligand samples were obtained from PubChem database and minimized through OpenBabel. 3D structure, selection method, resolution, atom count, weight, sequence length, and ID protein BMP2, BMP4, BMP7, alkaline phosphatase (AP), osteonectin, osteopontin, and osteocalcin on RCSB-PDB native ligand and water sterilization on PyMol were carried out with the aim of to maximize the formation of binding affinity during molecular docking simulations. Results: HA-PMMA composites can enhance the activity of proteins associated with osseointegration such as BMP-2/4/7, AP, osteocalcin, osteonectin, and osteopontin in silico. HA-PMMA composites have the strongest binding to osteonectin and are predicted to enhance the AP activity in silico. Conclusions: HA-PMMA composites are potential candidates for dental implant biomaterials with the osteointegration ability through binding with BMP-2/4/7, AP, osteocalcin, osteonectin, and osteopontin in silico.

Context: Human immunodeficiency virus (HIV) antiretrovirals that target the binding of viral enzyme are chosen as the lead solution in the treatment of HIV-1 infection, such as non-catalytic site integrase inhibitor (NCINI), nevirapine, and darunavir. There are natural compounds from specific plants that can be effective in treating HIV-1 infection such as tea catechin. Tea catechin administration causes a decrease in viral load and inhibition of entry mechanisms and an increased effect of apoptosis in infected cells. Aims: To identify the triple inhibitor mechanism in tea catechins against the three HIV-1 enzymes and apoptosis agonists through in silico approach as an innovation in handling HIV-1 infection. Methods: The 3D structure of tea catechin compounds from the database was examined, and then all target compounds were analyzed for drug-likeness, molecular docking, pathway prediction, and molecular interactions to determine the potential of tea catechin compounds as antiviral HIV-1 in silico. Results: Tea catechin compounds have the potential to serve as antiviral against HIV-1 through apoptosis agonist and triple inhibitor mechanisms. Apoptosis occurs due to the interaction of tea catechins with pro-apoptotic proteins in cells, and the epigallocatechin gallate (EGCG) compound is a class of tea catechins with the same binding position as control. Conclusions: The binding of the EGCG molecule complex results in low binding energy. Therefore, it allows EGCG acts as a triple inhibitor in HIV-1 infection.

Context: Bifidobacterium bifidum as a beneficial probiotic may regulate the inflammation and induce the bone remodeling by enhancing osteoblasts and basic fibroblast growth factor (bFGF) expression in the tension side of alveolar bone during orthodontic tooth movement (OTM). Aims: To evaluate the expression of bFGF and the number of osteoblasts during OTM after B. bifidum probiotic supplementation in male Wistar rats. Methods: Wistar rats (n = 42) were divided into 6 groups (n = 7) accordingly as follow: OTM and phosphate buffer saline (PBS) for 3 days (K3); OTM and PBS for 7 days (K7); OTM and PBS for 14 (K14); OTM and B. bifidum for 3 days (P3); OTM and B. bifidum for 7 days (P7); OTM and B. bifidum for 14 days (P14). OTM was established by NiTi close coil spring with 10 g force-placed between the first incisor and the first maxillary molar of Wistar rat. The samples were then terminated on days 3, 7, and 14. The maxillary tissue was isolated for the immunohistochemical examination and hematoxylin-eosin staining. All data were analyzed by using an independent t-test (p0,05). Herbal active compound with immunoregulator ability is considered a potential therapy for COVID-19 oral manifestation by downregulating pro-inflammatory cytokine storm. Meanwhile, anthocyanin and ternatin are the active compounds in Clitoria ternatea, which may act as a potential immunoregulator for COVID-19 therapy. The intention of this investigation was to investigate anthocyanin and ternatin as active compounds in C. ternatea that may be able to increase anti-inflammatory cytokine and inhibit pro-inflammatory cytokine and key proteins of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2). This study implemented bioinformatic approach to analyze anthocyanin and ternatin as active compounds in C. ternatea with anti- and pro-inflammatory cytokines and antiviral examination history through blind molecular docking study (in silico). Moreover, anthocyanins and ternatin were obtained from PubChem database by minimizing ligand structure in PyRx software to increase the flexibility. RCSB database was employed for preparing the protein samples consisting of interleukin (IL)-6, SARS-CoV-2-ACE2 glycoprotein complex, tumor necrosis factor-α receptor (TNF-αR), matrix metalloproteinase-9 (MMP-9), IL-6, IL-10, and human beta defensin-2 (HBD-2). In addition, The PyMol sofware was used to sterilize the protein samples to obtain the molecular docking optimization. This investigation found that, in the molecular docking simulation, the anthocyanin and ternatin showed producing the negative binding affinity to the ACE2 domain which interacted with RBD glycoprotein SARS-CoV-2. Anthocyanin and ternatin were then predicted to be able to influence any inhibitory activity of TNF-αR, MMP-9, and IL-6; increase IL-10; and increase HBD2 binding affinity values negatively. It can be predicted through molecular docking that anthocyanin and ternatin as the active compounds in C. ternatea contribute as a potential agent for COVID-19 oral manifestation therapy.

Objective: To show the cytotoxicity of Porphyromonas gingivalis lipopolysaccharide (LPS) on human umbilical cord mesenchymal stem cells (HUCMSCs) to better understand the characteristics for its application in regenerative procedures under periodontopathogen LPS influence. Material and Methods: Ultrapure Porphyromonas gingivalis LPS was used in this study. This research used a frozen stock HUCMSCs, previously confirmed by flow cytometry. The biological characteristics, such as cell morphology, proliferation, and protein expression, were screened. To check the cytotoxicity, HUCMSCs were cultured and divided into two groups, the control group and LPS group with various concentrations from 25 to 0.39 µg/mL. MTT assay was done and the cells were observed and counted. The significance level was set at 5%. Results: The percentage of living HUCMSCs on LPS group were not significantly different among concentrations (p>0.05) from 25 to 0.39 µg/mL, even though there were slight mean decrease between groups, but they were not significant. The duration of 24 hours of exposure of LPS does not significantly lower HUCMSCs viability. Conclusion: LPS does not affect the viability of HUCMSCs. The lower the concentration of LPS, the higher the viability of HUCMSCs.

Objective: To examine the cytotoxicity of calcium hydroxide on human umbilical cord mesenchymal stem cells (HUCMSC) to understand the characteristics for use in regenerative dentistry procedures especially regenerative endodontics. Material and Methods: HUCMSC was isolated, cultured, and confirmed by flow cytometry. The biological characteristics, such as cell morphology, proliferation, and protein expression, were screened. To check the cytotoxicity, HUCMSC was cultured and divided into two groups, the control group (cultured in minimum essential medium (MEM) alpha) and calcium hydroxide group (cultured in MEM alpha and calcium hydroxide). Methyl-thiazole-tetrazolium (MTT) assay was done on different concentrations of calcium hydroxide (0.39 to 25 µg/mL) and the cells were observed and counted. One-way ANOVA test was used with a significance level set at 5%. Results: Flow cytometric analysis confirmed positive of CD73, CD90, CD105, negative of CD45 and CD34. A significant difference was found between the concentration of 6.25 and 3.125 µg/mL (p=0.004). There was no significant difference among 6.25, 12.5 and 25 µg/mL concentrations. There was also no significant difference among 0.39, 0.78, 1.56, and 3.125 µg/mL concentrations. Conclusion: Even though calcium hydroxide is a medicament of choice in clinical endodontics, it decreases the viability of HUCMSC. The lower the concentration of calcium hydroxide, the higher the viability of HUCMSC.

This study aimed to analyze the distribution patterns and levels of platelet-derived growth factor (PDGF), insulin-like growth factor (IGF), vascular endothelial growth factor (VEGF), and transforming growth factor-β (TGF-β) in PRF incorporated with SMSCs. Materials and Methods: This study employed a true experiment (in vitro) design with post-test only performed in the control group alone. Background: An immunoinformatic approach may be useful to investigate the conserved region in the spike glycoprotein of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) Indonesia isolates. The aim of this study was to investigate Indonesian SARS-CoV-2 isolates based on B cell epitopes by targeting the conserved regions in the spike glycoprotein to trigger increased multi-variant virus neutralization and memory response for the development of vaccine seed candidates. Methods: SARS-CoV-2 spike glycoprotein gene sequences originating from Indonesia were compared with Wuhan (China), the United Kingdom, South Africa, India, the United States, and Brazil isolates obtained from the NCBI and GISAID databases. The recognition of antigens was carried out directly using B cells through the B cell receptor (BCR). An indirect B cell activation by Cluster of Differentiation (CD)4+ T cells and major histocompatibility complex (MHC)-II was predicted through the binding with human leukocyte antigen (HLA) based on IC50 value. In addition, vaccine allergenicity and toxicity were investigated. During the molecular complex examination, the 3D peptide structure was investigated and the lowest amount of energy formed when the vaccine candidate peptide bound to BCR and MHC-II was calculated. Results: As a result, the spike glycoprotein sequences of Indonesian SARS-CoV-2 isolates had conserved regions which were very similar to reference countries such as China, the United Kingdom, South Africa, India, the United States, and Brazil. Conclusion: It was predicted that the conserved regions could be identified as the epitope of B and T CD4+ cells that produced the peptides for vaccine candidate with antigenic, non-allergen, and non-toxic properties.

Such tests using chorionic villus or amniocentesis may increase the risk of harm to both mother and foetus. In the present day, a prenatal paternity test using circulating cell-free fetal Deoxyribonucleic Acid (DNA) is one alternative method due to it being non-invasive and safe for both mother and foetus. Aim The aim of this study is to detect Short Tandem Repeat (STR) at 5 Loci (vWA, TH01, D13S317, D18S51, and D21S11) and amelogenin genes in circulating cell-free fetal DNA in paternity tests. Materials and methods Forty-one samples of maternal blood were obtained from pregnant woman. Circulating free fetal DNA was subsequently extracted. A paternity test was conducted using an STR test at loci vWA, TH01, D13S317, D18S51, D21S11 in circulating free fetal DNA. An analysis of the paternity test between loci > 200 bp and 200 bp. Conclusion Circulating free fetal DNA can be used as an alternative sample for prenatal paternity tests because of its similarity with maternal DNA.

Background: Alveolar bone defect regeneration has long been problematic in the field of dentistry. Gingival stromal progenitor cells (GSPCs) offer a promising solution for alveolar bone regeneration. In order to optimally differentiate and proliferate progenitor cells, growth factors (GFs) are required. Platelet rich fibrin (PRF) has many GFs and can be easily manufactured. Core-binding factor subunit-α1 (CBF-α1) constitutes a well-known osteogenic differentiation transcription factor in SPCs. Sox9, as a chondrogenic transcription factor, interacts and inhibits CBF-α1, but its precise role in direct in vitro osteogenesis remains unknown. GSPCs cultured in vitro in PRF to optimally stimulate osteogenic differentiation has been largely overlooked. The aim of this study was to analyze GSPCs cultured in PRF osteogenic differentiation predicted by CBF-α1/Sox9. Methods: This study used a true experimental with post-test only control group design and random sampling. GPSCs isolated from the lower gingiva of four healthy, 250-gram, 1-month old, male Wistar rats (Rattus Novergicus) were cultured for two weeks, passaged every 4-5 days. GSPCs in passage 3-5 were cultured in five M24 plates (N=108; n=6/group) for Day 7, Day 14, and Day 21 in three different mediums (control negative group: αModified Eagle Medium; control positive group: High Glucose-Dulbecco’s Modified Eagle Medium (DMEM-HG) + osteogenic medium; Treatment group: DMEM-HG + osteogenic medium + PRF). Background: Utilizing the bioactive compounds found in pigmented rice might significantly reduce the risk of breast cancer. This study aims to systematically review existing literature on the benefit of Asian pigmented rice bioactive compounds and their implication in breast cancer. Methods: Searches of the literature were conducted in two databases (Scopus and PubMed) for a systematic review. The keywords resulted in a total of 407 articles, consisting of 103 PubMed and 304 Scopus articles. 32 manuscripts were excluded because the article was over 10 years old. After excluding book chapters and non-English languages, we had 278 potential articles to be reviewed. After checking and screening the title and abstract and eliminating duplicate articles, then 66 articles were obtained. After the selection and elimination of the full-text manuscripts, finally 10 of them which met the inclusion criteria. Result: The included studies in this review were entirely based in Asia. The year of publication ranged from 2013 to 2020. Half of included studies used black rice extract, two used red jasmine rice extracts, and three used Korean rice extracts (black, red, dark purple and brown rice). All studies were conducted in vitro and three studies were compared with in vivo tests on female mice. The pigmented rice is mainly black, red, and dark purple rice, and contains a variety of peonidin-3-glucoside, cyanidin-3-glucoside, γ-oryzanol, γ-tocotrienol, proanthocyanidin, cinnamic acid, and anthocyanins that may act as pro-apoptotic, anti-proliferative, and anti-metastasis of the breast cancer cells. Conclusion: Pigmented rice is a beneficial food which possessed bioactive compounds that may have significant potential concerning a breast cancer.

The current study is aiming to systematically review the impact of coffee consumption on periodontal health status. Methods: An article search was carried out in two electronic databases (PUBMED and Web of Sciences). All type of experimental and observational studies were included. The assessment of the included articles were conducted using Joanna Briggs Institute (JBI) critical appraisal tool. Data were analyzed qualitatively. Result: A total of 10 articles were included in this study. Most (5) of the studies discovered a negative correlation between coffee intake and periodontal health, while 4 other studies found the protective effect of daily coffee consumption against alveolar bone loss. Last, only one study found that coffee intake did not relate with periodontitis. Conclusion: The effect of coffee consumption on periodontal health was fragmented since coffee has complex components that may give either beneficial effects or negative impact on periodontal health.

Background: A chronic inflammatory process can cause disorders on the cardiovascular system. It is caused by an enhancement of inflammatory cytokines that can decrease the heart working-function. This article aims to review inflammatory cytokines that can cause the cardiovascular system disease. Methods: The review process began by taking articles from two databases, namely PubMed and SpringerLink without using the publication year limit. The reviewed article was a research article using human samples, which analyzed the impact of inflammatory cytokines on cardiovascular disease or the risk level of cardiovascular disease. Result: From a total of 3926 articles originating from two databases, 21 obtained articles have matched with the inclusion criteria for the review process. Of those 21 articles, 17 reported an increased effect of inflammatory cytokines on cardiovascular disease, while four articles showed no association between increased inflammatory cytokines and cardiovascular disease. There were six inflammatory cytokines that could affect cardiovascular disease, namely: TNF-α, IL-1β, IL-2, IL-6, IL-8, and IL-17 where cytokine IL-6 is confirmed by as many as 14 articles, TNF-α as many as six articles, and IL-1 β as many as three articles as inflammatory cytokines that could affect cardiovascular function. Conclusion: The increased level of inflammatory cytokines can be a trigger for decreased function and cause the disease in the cardiovascular system.

Background: Stem cells from human exfoliated deciduous teeth (SHED) seeded in carbonate apatite scaffold (CAS) may have multiple functions that could be used to regenerate the alveolar bone defects. The purpose of this study is to examine the ability of SHED and CAS in alveolar bone defects using an immunohistochemical analysis. Methods: ten three-month-old healthy male Wistar rats (R. novergicus) that weighed between 150–250 grams (g) were used as animal models. A simple blind random sampling method was used to select the sample that was assigned to the study group for CAS and SHED seeded in CAS (n=5). The animal study model of the alveolar bone was established by extracting the anterior mandible teeth. Rodent anesthesia was applied to relieve the pain during the procedure for all test animals. Immunohistochemistry was performed after seven days to facilitate the examination of the receptor activator of NF-κβ ligand (RANKL), osteoprotegrin (OPG), transforming growth factor-β (TGF-β), vascular endothelial growth factor (VEGF), runt-related transcription factor 2 (RUNX2), alkaline phosphatase (ALP), osteocalcin, and osteopontin expression.
