Montmorillonite (MM) colloid nanoplates have high adsorption capacity due to their large size/thickness ratio, which allows them to be used as carriers for drug delivery. Upon adsorption of the mitochondrial protein cytochrome c (cytC) onto MM plates, the composite cytC–MM particles acquire anticancer properties because of the ability of cancer cells to phagocytize submicron particles (in contrast to the normal cells). In this way, exogenous cytC can be introduced into tumor cells, thereby triggering apoptosis—an irreversible cascade of biochemical reactions leading to cell death. In the present study, we investigated the physicochemical properties of cytC–MM particles as a function of the cytC concentration in the suspension, namely, the electrophoretic mobility, the mass increment of MM monoplates upon cytC adsorption, the ratio of the adsorbed to the free cytC in the bulk, the protein density on the MM’s surface, the number of cytC globules adsorbed on an MM monoplate, the concentration of cytC–MM composite particles in the suspension, and the dependence of cytotoxicity on the cytC–MM particle concentration. For this purpose, we used microelectrophoresis, static and electric light scattering, and a colon cancer cell culture to test the cytotoxic effects of the cytC–MM suspensions. The results show that the cytotoxicity depends linearly on the logarithm of the particle concentration in the cytC–MM suspension reaching 97%.

Изследвани са електричните свойства и адсорбцията на глобуларния хемопротеид цитохром c (cytC) върху глинестия минерал монтморилонит (MM) във водна суспензия и антираковото действие на комплекса cytC-MM. Използвани са три групи методи: физикохимични (светоразсейване в електрично поле, микроелектрофореза и изоелектрично фокусиране), компютърни (белтъчна електростатика) и биологични (клетъчни култури). Установено е, че ММ е във вид на моноламели, при наситена адсорбция cytC се адсорбира свръхеквивалентно, презареждайки положително отрицателните ММ-пластинки, цитохромните глобули образуват монослой и са неподредено разпределени по повърхността, а изоелектричните pH-точки на свободен и адсорбиран cytC и минималната електрическа поляризуемост на cytC-MM съвпадат. Изводите са, че нетният електричен заряд на cytC остава непроменен при адсорбцията, глобулите са ориентирани хаотично с диполния си момент, електричният заряд на ММ-пластинките е напълно екраниран от белтъчните глобули, екранирането е хидродинамично, а не електростатично, и електроориентационният ефект има електрокинетична природа и при адсорбирани глобуларни макройони. Изследван е и антираковият ефект върху клетъчна култура от рак на дебело черво, дължащ се на апоптоза индуцирана от cytC, внесен в раковите клетки чрез фагоцитоза на колоидните cytC-MM частици, и е установено, че клетките почти напълно загиват, а цитотоксичният ефект е с 1/3 по-висок от достигнатия от други автори с различни частици-преносители на cytC. The electric properties and adsorption of the globular protein cytochrome c (cytC) on the clay mineral montmorillonite (MM) in aqueous suspension are investigated. Three groups of methods are employed: physicochemical (electric light scattering, microelectrophoresis and isoelectrofocusing), computer (protein electrostatics) and biological (cell cultures). It is found that MM is in monolamellar form, at saturation the cytC is adsorbed over-equivalently recharging positively the negative MM-plates and forming protein monolayer, the cytC-globules are distributed randomly on the surface, and the isoelectric pH-points of free and adsorbed cytC and the minimal electric polarizability of cytC-MM plates coincide. It is concluded that the net electric charge of cytC remains unaltered at adsorption, its globules are oriented randomly with their dipole moments, the electric charge of MM-plates is shielded completely by the adsorbed protein globules, the shielding is hydrodynamic but not electrostatic, and the electroorientational effect has electrokinetic mechanism in the case of adsorbed globular macroions. The anti-cancer effect on a colon cancer cell culture is also investigated (it is caused by apoptosis induced by introduction of the exogenous cytC by phagocytoses of the colloid cytC-MM particles) and it is found that the cancer cells almost completely die; the cytotoxic effect is 1/3 higher than the reached by other authors by different particles used as bearers of cytC.
