Physics Beyond Colliders is an exploratory study aimed at exploiting the full scientific potential of CERN's accelerator complex and its scientific infrastructure in the next two decades through projects complementary to the LHC, HL-LHC and other possible future colliders. These projects should target fundamental physics questions that are similar in spirit to those addressed by high-energy colliders, but that require different types of beams and experiments. A kick-off workshop held in September 2016 identified a number of areas of interest and working groups have been set-up to study and develop these directions. All projects currently under consideration are presented including physics motivation, a brief outline of the experimental set-up and the status of the corresponding beam and detector technological studies. The proposals are also put in context of the worldwide landscape and their implementation issues are discussed., Comment: This document (66 pages, 19 figures) is the summary document of the Physics Beyond Colliders (PBC) study. It follows the PBC mandate and draws on a whole set of documents produced in the context of PBC (https://pbc.web.cern.ch), in particular the reports of the QCD and BSM working groups also available at arXiv:1901.04482 and arXiv:1901.09966

7. Improved Determination of the Hadronic Contribution to the Muon (g-2) and to alpha(M_Z**2) Using new Data from Hadronic Tau Decays

We have reevaluated the hadronic contribution to the anomalous magnetic moment of the muon (g-2) and to the running of the QED fine structure constant alpha(s) at s=M_Z**2. We incorporated new data from hadronic tau decays, recently published by the ALEPH Collaboration. In addition, compared to previous analyses, we use more extensive e+e- annihilation data sets. The integration over the total hadronic cross section is performed using experimental data up to 40 GeV and results from perturbative QCD above 40 GeV. The improvement from tau data concerns mainly the pion form factor, where the uncertainty in the corresponding integral could be reduced by more than a factor of two. We obtain for the lowest order hadronic vacuum polarization graph a_mu(had) = (695.0 +/- 15.0) x 10^{-10} and delta(alpha(M_Z**2))(had) = (280.9 +/- 6.3) x 10^{-4} using e+e- data only. The corresponding results for combined e+e- and tau data are a_mu(had) = (701.1 +/- 9.4) x 10^{-10} and delta(alpha(M_Z**2))(had) = (281.7 +/- 6.2) x 10^{-4}, where the latter is calculated using the contribution from the five lightest quarks., Comment: 23 pages, LaTex, 6 figures, Paper submitted to Zeitschrift fuer Physik C

The daily practice in a clinical laboratory of molecular diagnosis in hematology involves using a variety of techniques, based on the amplification of nucleic acids. Current methods for polymerase chain reaction (PCR) use purified genomic DNA, mostly isolated from total peripheral blood cells or white blood cells (WBC). In this paper we describe a real-time fluorescence resonance energy transfer-based method for genotyping directly from blood cells. Our strategy is based on an initial isolation of the WBCs, allowing the removal of PCR inhibitors, such as the heme group, present in the erythrocytes. Once the erythrocytes have been lysed, in the LightCycler® 2.0 Instrument, we perform a real-time PCR followed by a melting curve analysis for different genes (Factors 2, 5, 12, MTHFR, and HFE). After testing 34 samples comparing the real-time crossing point (CP) values between WBC (5×106 WBC/mL) and purified DNA (20 ng/µL), the results for F5 Leiden were as follows: CP mean value for WBC was 29.26±0.566 versus purified DNA 24.79±0.56. Thus, when PCR was performed from WBC (5×106 WBC/mL) instead of DNA (20 ng/µL), we observed a delay of about 4 cycles. These small differences in CP values were similar for all genes tested and did not significantly affect the subsequent analysis by melting curves. In both cases the fluorescence values were high enough, allowing a robust genotyping of all these genes without a previous DNA purification/extraction. MicroRNAs (miRNAs) are small non-coding RNAs that negatively regulate gene expression at the post-transcriptional level, and they have been described as being associated with tumor prognosis. Here, miRNA profiling was planned to explore new molecular prognostic biomarkers in localized intestinal high-risk GIST. Paraffin tumor blocks of 14 and 86 patients were used in the discovery and expansion sets, respectively. GeneChip miRNA v3.0 was employed to identify the miRNAs differentially expressed between relapsed and non-relapsed patient samples, which were validated in the expansion set, by qRT-PCR. RT2 Profiler PCR Array was used for the screening of let-7e targets. Expression levels were correlated with relapse-free survival and overall survival. In the discovery set, 39 miRNAs were significantly deregulated, let-7e and miR-550 being the most underexpressed and overexpressed miRNAs in the relapsed group, respectively. In the expansion set, the underexpression of let-7e or the overexpression of 4 of its target genes ( ACVR1B , CASP3 , COL3A1, and COL5A2 ) were statistically associated with worse relapse-free survival. The expression of let-7e and 4 of its target genes are potential prognostic biomarkers in high-risk localized intestinal GIST. The expression of these genes is a potential molecular tool useful for a more accurate prognosis in this subset of GIST patients.

Located between the on-detector front-end electronics and the global data acquisition system (DAQ), the off-detector electronics of the CMS electromagnetic calorimeter (ECAL) is involved in both detector readout and trigger system. Working at 40 MHz, the trigger part must, within ten clock cycles, receive and deserialize the data of the front-end electronics, encode the trigger primitives using a nonlinear scale, assure time alignment between channels using a histogramming technique and send the trigger primitives to the regional trigger. In addition, it must classify trigger towers in three classes of interest and send this classification to the readout part. The readout part must select the zero suppression level to be applied depending on the regions of interest determined from the trigger tower classification, deserialize front-end data coming from high-speed (800 Mb/s) serial links, check their integrity, apply zero suppression, build the event and send it to the DAQ, monitor the buffer occupancy and send back pressure to the trigger system when required, provide data spying and monitoring facilities for the local DAQ. The system, and especially the data link speed, the latency constraints and the bit-error rate requirements have been validated on prototypes. Part of the system is about to go to production.
