This study generated bioactive hydrolysates using the enzyme Alcalase and autolysis from mesopelagic fish, including Maurolicus muelleri and Benthosema glaciale. Generated hydrolysates were investigated for their bioactivities using in vitro bioassays, and bioactive peptides were identified using mass spectrometry in active hydrolysates with cyclooxygenase, dipeptidyl peptidase IV and antioxidant activities. In silico analysis was employed to rank identified peptide sequences in terms of overall bioactivity using programmes including Peptide Ranker, PrepAIP, Umami-MRNN and AntiDMPpred. Seven peptides predicted to have anti-inflammatory, anti-type 2 diabetes or Umami potential using in silico strategies were chemically synthesised, and their anti-inflammatory activities were confirmed using in vitro bioassays with COX-1 and COX-2 enzymes. The peptide QCPLHRPWAL inhibited COX-1 and COX-2 by 82.90% (+/−0.54) and 53.84%, respectively, and had a selectivity index greater than 10. This peptide warrants further research as a novel anti-inflammatory/pain relief peptide. Other peptides with DPP-IV inhibitory and Umami flavours were identified. In the recent years, challenges faced in salmonid farming with regard to deformities have helped to improve our knowledge on skeletal biology and development of bone health in salmonids. Different nutritional, genetic and environmental factors are associated with skeletal deformities in salmonids and other fish species. Minerals are a group of essential nutrients having a vital role to play in skeletal development, growth and remodelling. The knowledge generated thus far on the structural and functional importance of minerals in salmonid bone health is largely restricted to phosphorus. A brief account of dietary phosphorus‐related bone deformities encountered in salmonids, critical life stages for the development of bone deformities, recent developments in the understanding of their aetiology and dietary phosphorus levels required to improve bone health are presented. The effect of increased dietary level of phosphorus in reducing bone health disorders under conditions of improved feed conversion efficiency and use of triploids in salmon farming are illustrated with help of metadata analysis from literature. With regard to the role of other essential minerals (macro‐ and micro‐) impacting bone health in salmonids, data available are extremely limited and hence information on other fish species and mammals is summarized in relation to bone development and incidence of deformities. The need to improve mineral bioavailability, utilization and reduce effluent mineral load is presented in brief with example from Norwegian salmon farming. Refinements in mineral requirement recommendations for salmonids and advanced methodologies for studying aetiology of skeletal anomalies, bone mineral status, skeletal development and deformities are also discussed. The hypothesis of the present study was that Atlantic salmon (Salmo salar) would respond to large variations in supplementation of dietary pro- and antioxidants, and marine lipid, with adjustment of the endogenously synthesised antioxidants, glutathione (GSH) and ubiquinone (UQ). An experiment with 27–3 reduced factorial design (the number of cases reduced systematically from 27 (full design) to 24 (reduced design)) was conducted, where vitamins, minerals and lipid were supplemented in the diet at high and low levels. For the vitamins and minerals the high levels were chosen to be just below anticipated toxic levels and the low levels were just above the requirement (vitamin C, 30 and 1000 mg/kg; vitamin E, 70 and 430 mg/kg; Fe, 70 and 1200 mg/kg; Cu, 8 and 110 mg/kg; Mn, 12 and 200 mg/kg). For astaxanthin, the dietary levels were 10 and 50 mg/kg and for lipid, 150 and 330 g/kg. The experiment was started with post-smolts (148 (sd 17 g)) and lasted for 5 months. The only effect on GSH was a minor increase ( < 10 %) in total concentration in the liver in response to high dietary lipid. GSH redox state was not affected. UQ responded to dietary lipid, astaxanthin and vitamin E, both with regard to total concentration and redox state. Except for an effect of Fe on plasma GSH, the trace elements and vitamin C had no effect on tissue levels and oxidation state of GSH and UQ. This shows that the endogenous redox state is quite robust with regard to variation of dietary pro- and antioxidants in Atlantic salmon. [ABSTRACT FROM PUBLISHER]

Abstract: The chemical composition, content of minerals and the profiles of amino acids and fatty acids were analyzed in fish bones from eight different species of fish. Fish bones varied significantly in chemical composition. The main difference was lipid content ranging from 23 g/kg in cod (Gadus morhua) to 509 g/kg in mackerel (Scomber scombrus). In general fatty fish species showed higher lipid levels in the bones compared to lean fish species. Similarly, lower levels of protein and ash were observed in bones from fatty fish species. Protein levels differed from 363 g/kg lipid free dry matter (dm) to 568 g/kg lipid free dm with a concomitant inverse difference in ash content. Ash to protein ratio differed from 0.78 to 1.71 with the lowest level in fish that naturally have highest swimming and physical activity. Saithe (Pollachius virens) and salmon (Salmo salar) were found to be significantly different in the levels of lipid, protein and ash, and ash/protein ratio in the bones. Only small differences were observed in the level of amino acids although species specific differences were observed. The levels of Ca and P in lipid free fish bones were about the same in all species analyzed. Fatty acid profile differed in relation to total lipid levels in the fish bones, but some minor differences between fish species were observed. [Copyright &y& Elsevier]

Aquaculture produces most of the world's seafood and is a valuable food source for an increasing global population. Low trophic mesopelagic biomasses have the potential to sustainably supplement aquafeed demands for increased seafood production. The present study is a theoretical whole-chain feed and food safety assessment on ingredients from mesopelagic biomass and the resulting farmed fish fed these ingredients, based on analysis of processed mesopelagic biomass. Earlier theoretical estimations have indicated that several undesirable compounds (e.g., dioxins and metals and fluoride) would exceed the legal maximum levels for feed and food safety. Our measurements on processed mesopelagic biomasses show that only fluoride exceeds legal feed safety limits. Due to high levels of fluoride in crustaceans, their catch proportion will dictate the fluoride level in the whole biomass and can be highly variable. Processing factors are established that can be used to estimate the levels of undesirables in mesopelagic aquafeed ingredients from highly variable species biomass catches. Levels of most the studied undesirables (dioxins, PCBs, organochlorine pesticides, brominated flame retardant, metals, metalloids) were generally low compared to aquafeed ingredients based on pelagic fish. Using a feed-to-fillet aquaculture transfer model, the use of mesopelagic processed aquafeed ingredients was estimated to reduce the level of dioxins and PCBs by ~30% in farmed seafood such as Atlantic salmon. Abstract: The intention of this study is to investigate the effect of fish meal soluble protein quality on growth and protein utilisation of Atlantic salmon fed very low fish meal diets. For this purpose stickwater (SW) was separated during herring meal production, fractionated following successive micro- ultra- and nano-filtration steps and reintroduced in the press cake (PC) in order to produce 4 reconstituted fish meals containing different molecular size soluble protein mixes. The SW fractions used for the production of the experimental fish meals were: 1) the micro-filtration (MF) retentate, 2) the ultra-filtration (UF) retentate, 3) the nano-filtration (NF) retentate and 4) the nano-filtration permeate. Four very low fish meal experimental diets were prepared containing 25gkg−1 super prime fish meal and 50gkg−1 diet of one of the 4 experimental fish meals produced. A positive control diet was prepared containing 300gkg−1 diet super prime fish meal. Three more diets were prepared containing 25gkg−1 diet super prime fish meal and 50gkg−1 diet PC. Two of them were supplemented with either crystalline taurine, or crystalline hydroxyproline, up to the levels present in the high fish meal control diet. The experimental diets with the SW fractions contained similar levels of water soluble protein (WSP), which were higher than in the PC diets and lower than in the control. All diets were iso-nitrogenous, iso-lipidic and iso-energetic and were fed to 8 triplicate groups of salmon (80 fish per group of initial body weight 133g), for 69days. High growth rates and low fish in-fish out ratios (FIFO) were achieved in salmon fed all the very low fish meal diets. Feed intake, fish body growth, whole body protein content, protein gain and morphometry were significantly affected by the inclusion of the different SW fractions in the low fish meal diets. The medium size peptides (UF retentate) induced significant increase in feed consumption in salmon and supported higher fish growth compared to the highest (MF retentate) or the lowest molecular weight peptides (NF permeate) where both feed intake and fish growth were reduced. The fish fed the diet containing the NF permeate had significantly lower growth compared to the diets containing the other SW fractions. We did not find any benefits for salmon performance by the supplementation of taurine or hydroxyproline alone to the low WSP PC diets. The present work gives evidence on the multi-dimensional effects of the different marine water soluble components on fish performance. In order to identify novel, high added-value, water soluble marine compounds, advanced large scale separation technologies must be further developed and applied in studies investigating specific functional effects that these compounds may have on fish and other organisms. Abstract: In order to study the short-term effects of dietary P levels during juvenile rearing on mineral status (bone ash content), and its long-term effects on the development of vertebral deformities (radiography and external examination), triplicate groups of Atlantic salmon juveniles (1.3g) were fed blue whiting meal (67%) based diets added 0 (0P), 3 (3P) and 6 (6P) g inorganic P kg−1 (15, 18 or 21gkg−1 total P) for 77days (18.3g), and then followed up on a common commercial diet for 432days (1927g). At the termination of the period on the experimental feeds the vertebrae of the fish fed the 0P diet had a significantly lower ash weight than those fed the 3P and 6P diets, while there was no difference in the occurrence of radiological deformed fish. 252days later, the 0P (31.7±5.5%) dietary group had a significant higher occurrence of radiological deformed fish than the 6P dietary group (9.4±5.6%), while the 3P dietary group displayed an intermediate level (19.7±2.8%). At termination, 432days after the termination of the experimental feeds, the 0P dietary group (5.9±0.7%) had a significantly higher prevalence of externally deformed fish compared to the 3P (3.5±0.9%) and 6P (2.0±0.4%) dietary groups. This was mainly caused by a higher level of deformities in the caudal region (V31–58) of the vertebral column in the 0P group. There were no effects on mortality or growth of the present diets. The results show that inadequate P nutrition in a short period during the juvenile stage can predispose Atlantic salmon to develop vertebral deformities following seawater transfer. The aim of this study was to test the efficacy of a new marine P ingredient from the bone fraction of herring by-products by evaluating the dietary impacts on growth, mineralization and skeletal development in Atlantic salmon fry. Five experimental diets were produced from a fish meal based diet, only differing in the dietary P source and level; low P control (5.5 g/kg soluble P), intermediate P (6.5 g/kg soluble P) and high P (8.0 g/kg soluble P) from either fish bone hydrolysate (FBH1, FBH2) or from NaH 2 PO 4 (NaP1, NaP2). The diets were given for 168 days from start of feeding (0.17 g fish) until 24 weeks of feeding (33 g). Weight and specific growth rate did not reveal diet dependent differences except in the initial 10 week feeding period where fish fed FBH1 showed lower growth as compared to fish fed the Na–P diets. Mineralization of fish evaluated by whole body and bone ash and mineral contents showed diet dependent differences that were related to dietary P level, but not to P source. Fish fed the low P diet showed clear P deficiency signs with significantly reduced tissue ash and mineral content, reduced whole body Ca:P ratio and morphological deviation from the normal. Histological evaluation of the vertebrae after 19 weeks of feeding (15 g) revealed stagnation in cartilage development, with accumulation of mature chondrocytes in fish fed low P control and FBH1 diet. FTIR showed that fish fed FBH1 had lower mineralization and increased cross binding in the vertebral end plates. Fish fed FBH2 resembled the positive NaP1 control. Real time qPCR analyses confirmed the histological results, by showing up-regulating of col10a1 (a marker for mature cartilage) in fish fed low P diet and FBH1. Osteocalcin (a marker for mineralization) was also activated in fish fed the low P diet and FBH1, possibly indicating a compensatory regulation in response to inadequate P. The dietary impacts on histology, FTIR and qPCR analyses in the FBH1 fed fish disappeared at 24 weeks of feeding (33 g). In conclusion, the results showed that P solubilized from bone fraction of herring can be efficiently utilized for growth, mineralization and bone development in Atlantic salmon fry. However, in the initial weeks of feeding, the FBH was a less efficient P source compared to NaP. Overall, the data suggest that P from fish bone hydrolysate is more suited for fish of minimum 15 g size. Statement of relevance The phosphate (P) rock reserve is a limited resource worldwide. Increased efficiency in the utilization of P and improved recycling of P from waste and manures are examples that could reduce the industry's vulnerability to the limited P supply while also reducing the negative environmental impacts. Recycling of P from fishery offal and development of new available P ingredients to farmed fish will significantly improve the sustainability of the aquaculture industry and reduce the environmental loss. Abstract: The effect of dietary inclusion of fish bone and crab by-products on growth performance and feed utilization was investigated in a growth trial with Atlantic cod. High quality fish meal was used as a control. Fish meal was replaced with fish bone meal at 150, 300 and 450 g kg−1, while dietary inclusion level of a crab by-product meal was 54, 115 and 176 g kg−1. Dietary protein and lipid were kept constant at 500 and 180 g kg−1 for all diets. The contents of ash in the diets were 84 g kg−1 for the control and 117, 148 and 180 g kg−1 for the fish bone meal and crab meal based diets. The feeding trial lasted 95 days, and average fish weight increased from 196 g initial weight to a final weight of about 500 g. The growth experiment showed a linear increase in feed intake with increasing dietary ash, resulting in a similar linear increase in growth measured by weight gain, SGR and TGC. The increase in growth was about 10% when the dietary ash level was increased from 84 to 180 g kg−1. For the fish bone meal based diets, no effect was observed on digestibility of protein and lipid, feed efficiency ratio, protein efficiency ratio or protein productive value. The crab meal based diets showed a similar picture, however, cod fed the highest level of crab meal had a lower feed efficiency ratio, protein efficiency ratio and protein productive value compared to the other groups. The results indicate that diets for cod can beneficially contain at least 180 g kg−1 ash from fish bone and 150 g kg−1 from crab meal. This opens the possibility to use unutilized by-products from the fish industry as cost effective and valuable feed ingredients for cod. [Copyright &y& Elsevier]

Atlantic cod of initial mean weight approximately 220 g were fed a control diet and three diets in which fish meal (FM) was replaced with increasing levels of full-fat soybean meal (FFS) supplied at 12, 24, and 36% of dry diet, for 12 weeks. There were no significant differences in final weights, but the specific growth rate (SGR) was significantly higher in fish fed the control (FFS0) diet compared to fish fed the FFS12 and FFS36 diets, and the feed conversion ratio (FCR) was significantly lower in fish fed the FFS0 diet compared to the other three treatments. The fatty acid (FA) compositions of the cod muscle and liver were highly affected by dietary treatment, and linear relationships between dietary and tissue FA concentrations were shown for some of these. Moreover, selective utilization or accumulation in the tissues of specific FA was suggested by the results.
