1. Towards Modified Condition/Decision Coverage of Rust

Abstract

Testing is an essential tool to assure software, especially so in safety-critical applications. To quantify how thoroughly a software item has been tested, a test coverage metric is required. Maybe the strictest such metric known in the safety critical systems is Modified Condition/Decision Coverage (MC/DC), which DO-178C prescribes for the highest software assurance level in aviation. In the past, ambiguities in the interpretation of MC/DC have been resolved already, i. e. in CAST-10. However, some central features of the Rust programming language necessitate further clarification. This work investigates aforementioned features, in particular pattern matching, providing a consistent view on how to apply MC/DC to Rust. 2. MetaWearS: A Shortcut in Wearable Systems Lifecycle with Only a Few Shots

Abstract

Wearable systems provide continuous health monitoring and can lead to early detection of potential health issues. However, the lifecycle of wearable systems faces several challenges. First, effective model training for new wearable devices requires substantial labeled data from various subjects collected directly by the wearable. Second, subsequent model updates require further extensive labeled data for retraining. Finally, frequent model updating on the wearable device can decrease the battery life in long-term data monitoring. Addressing these challenges, in this paper, we propose MetaWearS, a meta-learning method to reduce the amount of initial data collection required. Moreover, our approach incorporates a prototypical updating mechanism, simplifying the update process by modifying the class prototype rather than retraining the entire model. We explore the performance of MetaWearS in two case studies, namely, the detection of epileptic seizures and the detection of atrial fibrillation. We show that by fine-tuning with just a few samples, we achieve 70% and 82% AUC for the detection of epileptic seizures and the detection of atrial fibrillation, respectively. Compared to a conventional approach, our proposed method performs better with up to 45% AUC. Furthermore, updating the model with only 16 minutes of additional labeled data increases the AUC by up to 5.3%. Finally, MetaWearS reduces the energy consumption for model updates by 456x and 418x for epileptic seizure and AF detection, respectively.

Abstract

The design of low-power wearables for the biomedical domain has received a lot of attention in recent decades, as technological advances in chip manufacturing have allowed real-time monitoring of patients using low-complexity ML within the mW range. Despite advances in application and hardware design research, the domain lacks a systematic approach to hardware evaluation. In this work, we propose BiomedBench, a new benchmark suite composed of complete end-to-end TinyML biomedical applications for real-time monitoring of patients using wearable devices. Each application presents different requirements during typical signal acquisition and processing phases, including varying computational workloads and relations between active and idle times. Furthermore, our evaluation of five state-of-the-art low-power platforms in terms of energy efficiency shows that modern platforms cannot effectively target all types of biomedical applications. BiomedBench is released as an open-source suite to standardize hardware evaluation and guide hardware and application design in the TinyML wearable domain., Comment: 7 pages, 5 figures. Abstract

Feature selection is a crucial step in building machine learning models. This process is often achieved with accuracy as an objective, and can be cumbersome and computationally expensive for large-scale datasets. Several additional model performance characteristics such as fairness and robustness are of importance for model development. As regulations are driving the need for more trustworthy models, deployed models need to be corrected for model characteristics associated with responsible artificial intelligence. When feature selection is done with respect to one model performance characteristic (eg. accuracy), feature selection with secondary model performance characteristics (eg. fairness and robustness) as objectives would require going through the computationally expensive selection process from scratch. In this paper, we introduce the problem of feature \emph{reselection}, so that features can be selected with respect to secondary model performance characteristics efficiently even after a feature selection process has been done with respect to a primary objective. To address this problem, we propose REFRESH, a method to reselect features so that additional constraints that are desirable towards model performance can be achieved without having to train several new models. REFRESH's underlying algorithm is a novel technique using SHAP values and correlation analysis that can approximate for the predictions of a model without having to train these models. Empirical evaluations on three datasets, including a large-scale loan defaulting dataset show that REFRESH can help find alternate models with better model characteristics efficiently. We also discuss the need for reselection and REFRESH based on regulation desiderata.

Abstract

PURPOSE: The OlympiA randomized phase III trial compared 1 year of olaparib (OL) or placebo (PL) as adjuvant therapy in patients with germline BRCA1/2, high-risk human epidermal growth factor receptor 2-negative early breast cancer after completing (neo)adjuvant chemotherapy ([N]ACT), surgery, and radiotherapy. The patient-reported outcome primary hypothesis was that OL-treated patients may experience greater fatigue during treatment. METHODS: Data were collected before random assignment, and at 6, 12, 18, and 24 months. The primary end point was fatigue, measured with the Functional Assessment of Chronic Illness Therapy-Fatigue scale. Secondary end points, assessed with the European Organization for Research and Treatment of Cancer Quality of Life Questionnaire, Core 30 item, included nausea and vomiting (NV), diarrhea, and multiple functional domains. 8. Avian malaria in a feral-pet pigeon: a case report

Abstract

Abstract Background Avian malaria is caused by diverse parasite species of the genus Plasmodium, and it affects various bird species. The occurrence of this disease in some wild bird species is sparsely documented due to the scarce availability of samples. Hence the pathogenicity in some hosts is not completely known. In addition, feral birds may act as reservoirs bridging the transmission cycle from wild migratory birds to domestic and zoo-kept bird species. Case presentation An owner of pigeons adopted a feral pigeon (Columba livia forma domestica) and housed it together with his other pet-pigeons. The bird died unexpectedly a few weeks after a surgical procedure and necropsy revealed a severely anaemic carcass, with pale organs and hydropericardium. Histopathologic analysis revealed inflammatory infiltrates in the lung and liver, and monocytes and Kupffer cells contained haemozoin pigment indicative of phagocytosis of Plasmodium-infected erythrocytes. A high erythrocytic infection rate of 18% was evident in tissues and blood vessels in various organs. Furthermore, the thyroid had masses classified as thyroid carcinomas. Immunohistochemistry with anti- Plasmodium falciparum HSP70 antibody revealed positive signals in erythrocytes and intravascular leucocytes. Further microscopy analysis using a Hemacolor-stained impression smear revealed a high parasitaemia with an asynchronous infection showing all erythrocytic stages. Molecular diagnosis by PCR identified Plasmodium relictum, lineage GRW11 as the aetiological agent. The bird presented died most likely due to an acute infection as evidenced by the high blood parasitaemia, leading to major erythrocyte destruction. Further analyses of feral pigeons (n = 22) did not reveal any additional cases of Plasmodium infections. Conclusion This study reports the first mortality associated with P. relictum lineage GRW11. The study supports previous studies, suggesting that Plasmodium infections are not frequent in pigeons. Host conditions like immunosuppression due to the tumour may have influenced the infection outcome in this fatal case. Use of anti-P. falciparum HSP70 antibody for detection of P. relictum antigens for immune assays in blood and tissue samples will be a useful tool for future studies.

Abstract

Abstract Background Fluid administration is the first line treatment in intensive care unit (ICU) patients with sepsis and septic shock. While fluid boluses administration can be titrated by predicting preload dependency, the amount of other forms of fluids may be more complex to be evaluated. We conducted a retrospective analysis in a tertiary hospital, to assess the ratio between fluids given as boluses and total administered fluid intake during early phases of ICU stay, and to evaluate the impact of fluid strategy on ICU mortality. Data related to fluid administration during the first four days of ICU stay were exported from an electronic health records system (ICCA®, Philips Healthcare). Demographic data, severity score, norepinephrine dose at ICU admission, overall fluid balance and the percentage of different fluid components of the overall volume administered were included in a multivariable logistic regression model, evaluating the association with ICU survival. Results We analyzed 220 patients admitted with septic shock and sepsis-induced hypotension from 1st July 2021 to 31st December 2023. Fluid boluses and maintenance represented 49.3% ± 22.8 of the overall fluid intake, being balanced solution the most represented (40.4% ± 22.0). The fluid volume for drug infusion represented 34.0% ± 2.9 of the total fluid intake, while oral or via nasogastric tube fluid intake represented 18.0% ± 15.7 of the total fluid intake. Fluid volume given as boluses represented 8.6% of the total fluid intake over the four days, with a reduction from 25.1% ± 24.0 on Day 1 to 4.8% ± 8.7 on Day 4. A positive fluid balance [OR 1.167 (1.029–1.341); p = 0.021] was the most important factor associated with ICU mortality. Non-survivors (n = 66; 30%) received a higher amount of overall inputs than survivors only on Day 1 [2493 mL vs. 1855 mL; p = 0.022]. Conclusions This retrospective analysis of fluids given over the early phases of septic shock and sepsis-induced hypotension showed that the overall volume given by boluses ranges from about 25% on Day 1 to about 5% on Day 4 from ICU admission. Our data confirms that a positive fluid balance over the first 4 days of ICU is associated with mortality.

Abstract

Explainable Artificial Intelligence (XAI) has received widespread interest in recent years, and two of the most popular types of explanations are feature attributions, and counterfactual explanations. These classes of approaches have been largely studied independently and the few attempts at reconciling them have been primarily empirical. This work establishes a clear theoretical connection between game-theoretic feature attributions, focusing on but not limited to SHAP, and counterfactuals explanations. After motivating operative changes to Shapley values based feature attributions and counterfactual explanations, we prove that, under conditions, they are in fact equivalent. We then extend the equivalency result to game-theoretic solution concepts beyond Shapley values. Moreover, through the analysis of the conditions of such equivalence, we shed light on the limitations of naively using counterfactual explanations to provide feature importances. Experiments on three datasets quantitatively show the difference in explanations at every stage of the connection between the two approaches and corroborate the theoretical findings., Comment: Accepted at AIES 2023

13. High C-Rate Performant Electrospun LiFePO4/Carbon Nanofiber Self-Standing Cathodes for Lithium-Ion Batteries

Abstract

In the present study, LiFePO4/CNF self-standing cathodes for LIBs are synthesized by electrospinning. A lower active material amount (12.3 and 34.5 wt%) is used, compared to the conventional tape-casted cathodes (70–85 wt%). The characterization techniques (XRPD, SEM, TEM, EDS, Raman spectroscopy, and thermogravimetry) confirm that the olivine-type structure of LiFePO4 is maintained in the binder-free electrodes, and the active material is homogeneously dispersed into and within the carbon nanofibers. The electrochemical investigation demonstrates that higher Li+ diffusion coefficients (1.36 × 10−11 cm2/s) and improved reversibility are reached for free-standing electrodes, compared to the LiFePO4 tape-casted cathode (80 wt% of active material) appositely prepared for comparison. The 34.5 wt% LiFePO4 self-standing cathode displays a lower capacity fading, good reversibility and stability, enhanced capacity values at C-rates higher than 5C, and a good lifespan when cycled 1000 cycles at 1C and further cycled up to 20C, compared to the tape-casted counterpart. Notably, the improved electrochemical performances are obtained by only the 34.5 wt% of active material. The results evidence the relevant role of the CNF matrix suitable to host LiFePO4, to promote electrolyte permeation and contact with the active material, and to increase the electronic conductivity.

Abstract

Pancreatic cancer is characterized by extensive resistance to conventional therapies, making clinical management a challenge. Here we map the epigenetic dependencies of cancer stem cells, cells that preferentially evade therapy and drive progression, and identify SWI/SNF complex member SMARCD3 as a regulator of pancreatic cancer cells. Although SWI/SNF subunits often act as tumor suppressors, we show that SMARCD3 is amplified in cancer, enriched in pancreatic cancer stem cells and upregulated in the human disease. Diverse genetic mouse models of pancreatic cancer and stage-specific Smarcd3 deletion reveal that Smarcd3 loss preferentially impacts established tumors, improving survival especially in context of chemotherapy. Mechanistically, SMARCD3 acts with FOXA1 to control lipid and fatty acid metabolism, programs associated with therapy resistance and poor prognosis in cancer. These data identify SMARCD3 as an epigenetic modulator responsible for establishing the metabolic landscape in aggressive pancreatic cancer cells and a potential target for new therapies.

Abstract

Abstract The Arctic polar nights bring extreme environmental conditions characterised by cold and darkness, which challenge the survival of organisms in the Arctic. Additionally, multiple anthropogenic stressors can amplify the pressure on the fragile Arctic ecosystems during this period. Determining how multiple anthropogenic stressors may affect the survival of Arctic life is crucial for ecological risk assessments and management, but this topic is understudied. For the first time, our study investigates the complex interactions of multiple stressors, exploring stressor temporal dynamics and exposure duration on a key Arctic copepod Calanus glacialis during the polar nights. We conducted experiments with pulse (intermittent) and press (continuous) exposure scenarios, involving microplastics, pyrene and warming in a fully factorial design. We observed significant effects on copepod survival, with pronounced impacts during later stressor phases. We also detected two‐way interactions between microplastics and pyrene, as well as pyrene and warming, further intensified with the presence of a third stressor. Continuous stressor exposure for 9 days (press‐temporal scenario) led to greater reductions in copepod survival compared to the pulse‐temporal scenario, characterised by two 3‐day stressor exposure phases. Notably, the inclusion of recovery phases, free from stressor exposure, positively influenced copepod survival, highlighting the importance of temporal exposure dynamics. We did not find behaviour to be affected by the different treatments. Our findings underscore the intricate interactions amongst multiple stressors and their temporal patterns in shaping the vulnerability of overwintering Arctic copepods with crucial implications for managing Arctic aquatic ecosystems under the fastest rate of ongoing climate change on earth.

Abstract

Summary: Background: Mucositis is a common and highly impactful side effect of conventional and emerging cancer therapy and thus the subject of intense investigation. Although common practice, mucositis assessment is heterogeneously adopted and poorly guided, impacting evidence synthesis and translation. The Multinational Association of Supportive Care in Cancer (MASCC) Mucositis Study Group (MSG) therefore aimed to establish expert recommendations for how existing mucositis assessment tools should be used, in clinical care and trials contexts, to improve the consistency of mucositis assessment. Methods: This study was conducted over two stages (January 2022–July 2023). The first phase involved a survey to MASCC-MSG members (January 2022–May 2022), capturing current practices, challenges and preferences. These then informed the second phase, in which a set of initial recommendations were prepared and refined using the Delphi method (February 2023–May 2023). Consensus was defined as agreement on a parameter by >80% of respondents. Findings: Seventy-two MASCC-MSG members completed the first phase of the study (37 females, 34 males, mainly oral care specialists). High variability was noted in the use of mucositis assessment tools, with a high reliance on clinician assessment compared to patient reported outcome measures (PROMs, 47% vs 3%, 37% used a combination). The World Health Organization (WHO) and Common Terminology Criteria for Adverse Events (CTCAE) scales were most commonly used to assess mucositis across multiple settings. Initial recommendations were reviewed by experienced MSG members and following two rounds of Delphi survey consensus was achieved in 91 of 100 recommendations. For example, in patients receiving chemotherapy, the recommended tool for clinician assessment in clinical practice is WHO for oral mucositis (89.5% consensus), and WHO or CTCAE for gastrointestinal mucositis (85.7% consensus). The recommended PROM in clinical trials is OMD/WQ for oral mucositis (93.3% consensus), and PRO-CTCAE for gastrointestinal mucositis (83.3% consensus). Interpretation: These new recommendations provide much needed guidance on mucositis assessment and may be applied in both clinical practice and research to streamline comparison and synthesis of global data sets, thus accelerating translation of new knowledge into clinical practice. 31. In Silico Structural Prediction for the Generation of Novel Performant Midi-Dystrophins Based on Intein-Mediated Dual AAV Approach

Abstract

Duchenne Muscular Dystrophy (DMD) is a pediatric disorder characterized by progressive muscle degeneration and premature death, and has no current cure. The current, most promising therapeutic avenue is based on gene replacement mediated by adeno-associated viruses (AAVs) using a shortened, but still functional, version of dystrophin, known as micro-dystrophin (µDys), to fit AAV capacity. The limited improvements observed in clinical trials suggest a sub-optimal performance of µDys in the human context that could be due to the lack of key domains in the protein. Therefore, expressing larger dystrophin proteins may be necessary for a more complete correction of the disease phenotype. In this study, we developed three novel midi-dystrophin constructs using a dual-AAV approach, leveraging split-intein-based protein trans-splicing. The midi-dystrophins include additional domains compared to µDys, such as the central cytoskeleton-binding domain, nNOS and Par1b interacting domains, and a complete C-terminal region. Given the limited capacity of each AAV vector, we strategically partially reduced hinge regions while ensuring that the structural stability of the protein remains intact. We predicted the interactions between the two halves of the split midi-Dys proteins thanks to the deep learning algorithm AphaFold3. We observed strong associations between the N- and C-termini in midi-Dys 1 and 2, while a weaker interaction in midi-Dys 3 was revealed. Our subsequent experiments confirmed the efficient protein trans-splicing both in vitro and in vivo in DBA2/mdx mice of the midi-Dys 1 and 2 and not in midi-Dys 3 as expected from the structural prediction. Additionally, we demonstrated that midi-Dys 1 and 2 exhibit significant therapeutic efficacy in DBA2/mdx mice, highlighting their potential as therapeutic agents for DMD. Overall, these findings highlight the potential of deep learning-based structural modeling for the generation of intein-based dystrophin versions and pose the basis for further investigation of these new midi-dystrophins versions for clinical studies.

Abstract

Convenient, rapid, highly sensitive and on-site iron determination is important for environmental safety and human health. We developed a sensing system for the detection of Fe(III) in water based on 7-mercapto-4-methylcoumarine (MMC)-stabilized silver-coated gold nanostars (GNS@Ag@MMC), exploiting a redox reaction between the Fe(III) cation and the silver shell of the nanoparticles, which causes a severe transformation of the nanomaterial structure, reverting it to pristine GNSs. This system works by simultaneously monitoring changes in the Localized Surface Plasmon Resonance (LSPR) and Surface-Enhanced Raman Spectroscopy (SERS) spectra as a function of added Fe(III). The proposed sensing system is able to detect the Fe(III) cation in the 1.0 × 10−5–1.5 × 10−4 M range, and its selectivity of the GNS@Ag@MMC sensor toward iron has been verified monitoring the LSPR and the SERS response to other cations with a clear selectivity toward Fe(III).

Abstract

Background: Extra virgin olive oil is a fundamental component of the Mediterranean diet. It contains several molecules that sustain human well-being by modulating cellular metabolism and exerting antioxidant, anti-inflammatory, and anti-ageing effects to protect normal tissues, and it can exert anti-angiogenic and pro-apoptotic effects on cancer cells. Metabolites found in different parts of the olive tree, including leaves, also possess properties that might help in cancer prevention and promote wellness in aging. Olive mill wastewater (OMWW), a liquid residue produced during olive oil extraction, represents an environmental issue. However, it is rich in phytochemicals with potential beneficial properties. Dietary supplements based on OMWW can be produced for nutritional supplementation with advantages to the ecology. Purpose: This work aims to measure hematochemical, anthropometric, and metabolomic parameters in volunteers taking an OMWW dietary supplement, Oliphenolia® (OMWW-OL). Methods: The supplementation of OMWW-OL 25 mL twice daily for 30 days was tested on a pilot cohort of volunteers with characteristics close to metabolic syndrome. Hematochemical, anthropometric, serum biomarkers and serum metabolomic parameters were analyzed before the intervention, at 30 days, and 30 days after stopping consumption. Results: A total of 29 volunteers were enrolled, and 23 completed the study. The participants’ parameters at baseline were measured, and then twice daily at 30 days of treatment and 30 days after assumption discontinuation. Although treatment was with an olive derivative, their weight did not increase. Their body mass index, instead of augmenting, slightly decreased, particularly in the women. Also, hydration increased, especially in the women, while blood pressure, glycemia, and insulin decreased. Cholesterol, high-density lipoproteins, and triglycerides were stable, and LDL levels decreased, while vitamin D levels, alongside calcium, perceptibly increased. Albumin also increased. All the values were in support of an equilibrium, with no damaging effects. By mass spectrometry analysis, we also found favorable changes in the vitamin D/histamine and homocysteine/methionine ratios, an increase in a new metabolite of unknown formula, and the vitamin D/unknown metabolite ratio. Conclusions: Supplementation of OMWW-OL has no detrimental effects and might imply the beneficial modulation of several biological parameters. Although this is a small pilot study, with limited potency, it preliminarily suggests that the OMWW extract use could be potentially valuable for people at risk of metabolic syndrome. Some of these parameters could also be relevant in supporting healthy ageing and in cancer prevention.

Abstract

Feature attributions are a common paradigm for model explanations due to their simplicity in assigning a single numeric score for each input feature to a model. In the actionable recourse setting, wherein the goal of the explanations is to improve outcomes for model consumers, it is often unclear how feature attributions should be correctly used. With this work, we aim to strengthen and clarify the link between actionable recourse and feature attributions. Concretely, we propose a variant of SHAP, Counterfactual SHAP (CF-SHAP), that incorporates counterfactual information to produce a background dataset for use within the marginal (a.k.a. interventional) Shapley value framework. We motivate the need within the actionable recourse setting for careful consideration of background datasets when using Shapley values for feature attributions with numerous synthetic examples. Moreover, we demonstrate the efficacy of CF-SHAP by proposing and justifying a quantitative score for feature attributions, counterfactual-ability, showing that as measured by this metric, CF-SHAP is superior to existing methods when evaluated on public datasets using tree ensembles., Comment: Accepted at FAccT '22 (2022 ACM Conference on Fairness, Accountability, and Transparency)

Abstract

Explainable AI (XAI) has been investigated for decades and, together with AI itself, has witnessed unprecedented growth in recent years. Among various approaches to XAI, argumentative models have been advocated in both the AI and social science literature, as their dialectical nature appears to match some basic desirable features of the explanation activity. In this survey we overview XAI approaches built using methods from the field of computational argumentation, leveraging its wide array of reasoning abstractions and explanation delivery methods. We overview the literature focusing on different types of explanation (intrinsic and post-hoc), different models with which argumentation-based explanations are deployed, different forms of delivery, and different argumentation frameworks they use. Abstract

Ocular toxoplasmosis has a known, rare association with acute retinal artery occlusion (RAO). We describe a 21-year-old male who presented with acute focal toxoplasmosis chorioretinitis in the right eye treated with intravitreal clindamycin, intravitreal dexamethasone, and adjunct oral therapy for vision-threatening retinitis with subsequent quiescence. Nine months from his initial presentation, the patient presented with a branch RAO adjacent to an inactive retinal scar in the right eye. Widefield en face structural swept-source optical coherence tomography (SS-OCT) centered on the middle retina showed paracentral acute middle maculopathy (PAMM) in an arteriolar distribution. The patient was started on 81 mg of aspirin daily. Six months later, the en face structural SS-OCT and corresponding B-scans showed resolution of PAMM. Along with a review of the literature on toxoplasmosis-related RAOs, we present the first case of delayed-onset RAO in ocular toxoplasmosis.
