Superconductivity, discovered by Kamerlingh Onnes in 1911, continues to be a fascinating subject of condensed matter physics today. Much interest has been devoted to the study of the superconductivity induced in a metal which by itself is not superconducting but is in electrical contact with a superconductor. As the carriers of superconductivity, the Cooper pairs, diffuse across the contact into the metal they remain correlated, although the pairing mechanism is lifted; we call this the proximity effect. The observation of these superconducting correlations has come within the reach of experiments in the last decade. With state-of-the-art fabrication techniques mesoscopic samples have been produced which are small and clean enough for the quantum mechanical coherence of the electrons to be preserved over the sample size. This theoretical thesis focuses on the variety of signatures of single-particle physics that appear in the electrical transport and the magnetic screening properties of these systems. We study the resonance structure in the finite voltage conductance and shot noise of NS junctions. Mapping out the H-T phase diagram of a NS hybrid layer we describe the diamagnetic screening including non-locality and impurities, the magnetic breakdown of the Meissner expulsion, and discover a paramagnetic instability towards spontaneous magnetic moments. We trace back the double critical-current-flux periodicity in ballistic SNS junctions to non-locality. We study the current in a clean superconductor-normal-metal-superconductor junction of length d and width w in the presence of an applied magnetic field H. We show that both the geometrical pattern of the current density and the critical current as a function of the total flux in the junction, depend on the ratio of the Josephson vortex distance a_0 and the range r of the nonlocal electrodynamics. In particular, the critical current has the periodicity of the superconducting flux quantum only for r

6. Magnetic breakdown in a normal-metal - superconductor proximity sandwich

We study the magnetic response of a clean normal-metal slab of finite thickness in proximity with a bulk superconductor. We determine its free energy and identify two (meta-)stable states, a diamagnetic one where the applied field is effectively screened, and a second state, where the field penetrates the normal-metal layer. We present a complete characterization of the first order transition between the two states which occurs at the breakdown field, including its spinodals, the jump in the magnetization, and the latent heat. The bistable regime terminates at a critical temperature above which the sharp transition is replaced by a continuous cross-over. - Abstract
The purposes of meat inspection have been formulated for more than 100 years as (a) protecting health of consumers, (b) maintain the reputation of the meats in home and export markets, and (c) detecting communicable diseases of animals before they have spread beyond easy control. Today, one would add to protect animal welfare, clarify that protecting consumer health includes both chemical and biological hazards, and add food fraud to the issues of reputation. To transform the scientific knowledge into modern meat safety assurance systems (MSAS), the risk managers need to understand the social capital in the meat value chain to align the behaviors of farmers, food business operators and competent authorities with technical knowledge. The meat value chain could be perceived as a commons – a material or immaterial property held jointly by the members of a community, whom may govern access to and use of the property through social structures, traditions, and/or formal rules i.e. social capital. The social capital and food safety culture amongst farmers and food business operators is a key driver for successful meat safety while information asymmetry increases risks for a tragedy of commons scenario. Ostrom's core design principles for stable commons could inform the design of MSASs. Tools for reducing the information asymmetry and building trust and social capital between all stakeholders within the meat value chain include the food safety culture, food chain information, use of health epidemiological indicators, sensors and block chains, industry/private standards, and the applying system approach from farm to fork.

Abstract Visual attention is an intrinsic part of intra- and inter-specific interactions. Its structure (e.g. short glances vs. long gazes) depends on the species, type and expected outcome of the interaction. The outcomes of earlier repeated interactions determine the resulting valence of the relationships. Human-animal relationships rely upon species-specific cognitive abilities and attentional characteristics for associative learning and memory formation. Visual attention is crucial in cognitive processing and relationships construction, but its structure may depend on species-specific characteristics and relationships’ quality. Dolphins are renowned for their cognitive abilities and in human-care settings they have regular interactions with caretakers. Dolphins’ visual attention structure and laterality both differ depending on the visual stimulus to which the animal is attending (species-specific, non-biological). We hypothesized that visual attentional characteristics and associated behaviours, in a group of 9 captive bottlenose dolphins (Tursiops truncatus), would differ according to past experience and familiarity with humans. Human motionless tests showed a role of experience (time spent) in the facility on play and vocal production near humans regardless of familiarity. However, familiarity influenced dolphin presence near humans and visual laterality: they stayed longer and used their right eye more to glance at familiar humans. These findings provide new insights on how dolphins cognitively process stimuli according to familiarity and past experiences, as well as on how human – animal relationships evolve in captive settings.

10. Optimized partial freezing protocol enables 10-day storage of rat livers

Abstract Preserving organs at subzero temperatures with halted metabolic activity holds the potential to prolong preservation and expand the donor organ pool for transplant. Our group recently introduced partial freezing, a novel approach in high-subzero storage at -15 °C, enabling 5-day storage of rodent livers through precise control over ice nucleation and unfrozen fraction. However, increased vascular resistance and tissue edema suggested a need for improvements to extend viable preservation. Here, we describe an optimized partial freezing protocol with key optimizations, including an increased concentration of polyethylene glycol (PEG) to enhance membrane stability while minimizing shear stress during cryoprotectant unloading with an acclimation period and a maintained osmotic balance through an increase in bovine serum albumin (BSA). These approaches ensured the viability during preservation and recovery processes, promoting liver function and ensuring optimal preservation. This was evidenced by increased oxygen consumption, decreased vascular resistance, and edema. Ultimately, we show that using the optimized protocol, livers can be stored for 10 days with comparable vascular resistance and lactate levels to 5 days, outperforming the viability of time-matched static cold stored (SCS) livers as the current gold standard. This study represents a significant advancement in expanding organ availability through prolonged preservation, thereby revolutionizing transplant medicine.

Antimicrobial resistance (AMR) surveillance systems involve multiple stakeholders and multilevel standard operating procedures, which increase in complexity with further integration of the One Health (OH) concept. AMR is a OH challenge. It is crucial for the success of an AMR surveillance system to evaluate its performance in meeting the proposed objectives, while complying with resource restrictions. The OH-EpiCap tool was created to evaluate the degree of compliance of hazard surveillance activities with essential OH concepts across there dimensions: organization, operational activities, and impact of the OH surveillance system.To present feedback on the application of the OH-EpiCap from a user’s perspective, the tool was used to evaluate nine national AMR surveillance systems, each with different monitoring contexts and objectives. The OH-EpiCap tool was assessed using the updated CoEvalAMR methodology. This methodology evaluates the content themes and functional aspects of the tool in a standardized way, while it also captures the user’s subjective experiences in using the tool via a strengths, weaknesses, opportunities, and threats (SWOT) approach.The results of the evaluation of the OH-EpiCap are presented and discussed. The OH-EpiCap is an easy-to-use tool, which can facilitate a fast macro-overview of the application of the OH concept to a surveillance activity, when used by specialists in the matter, serving as a basis for the discussion of possible adaptations of AMR surveillance activities, or targeting areas that may be further investigated using other pre-established tools.

Science - Abstract
Abstract Drug-recalcitrant infections are a leading global-health concern. Bacterial cells benefit from phenotypic variation, which can suggest effective antimicrobial strategies. However, probing phenotypic variation entails spatiotemporal analysis of individual cells that is technically challenging, and hard to integrate into drug discovery. In this work, we develop a multi-condition microfluidic platform suitable for imaging two-dimensional growth of bacterial cells during transitions between separate environmental conditions. With this platform, we implement a dynamic single-cell screening for pheno-tuning compounds, which induce a phenotypic change and decrease cell-to-cell variation, aiming to undermine the entire bacterial population and make it more vulnerable to other drugs. We apply this strategy to mycobacteria, as tuberculosis poses a major public-health threat. Our lead compound impairs Mycobacterium tuberculosis via a peculiar mode of action and enhances other anti-tubercular drugs. This work proves that harnessing phenotypic variation represents a successful approach to tackle pathogens that are increasingly difficult to treat.

Abstract Dietary restriction promotes resistance to surgical stress in multiple organisms. Counterintuitively, current medical protocols recommend short-term carbohydrate-rich drinks (carbohydrate loading) prior to surgery, part of a multimodal perioperative care pathway designed to enhance surgical recovery. Despite widespread clinical use, preclinical and mechanistic studies on carbohydrate loading in surgical contexts are lacking. Here we demonstrate in ad libitum-fed mice that liquid carbohydrate loading for one week drives reductions in solid food intake, while nearly doubling total caloric intake. Similarly, in humans, simple carbohydrate intake is inversely correlated with dietary protein intake. Carbohydrate loading-induced protein dilution increases expression of hepatic fibroblast growth factor 21 (FGF21) independent of caloric intake, resulting in protection in two models of surgical stress: renal and hepatic ischemia-reperfusion injury. The protection is consistent across male, female, and aged mice. In vivo, amino acid add-back or genetic FGF21 deletion blocks carbohydrate loading-mediated protection from ischemia-reperfusion injury. Finally, carbohydrate loading induction of FGF21 is associated with the induction of the canonical integrated stress response (ATF3/4, NF-kB), and oxidative metabolism (PPARγ). Together, these data support carbohydrate loading drinks prior to surgery and reveal an essential role of protein dilution via FGF21.

Abstract Background The objective of this study was to assess the likelihood of acute appendicitis (AA) in children presenting with abdominal symptoms at the emergency department (ED), based on their prior primary care (PC) consultation history. Methods Between February and June 2021, we prospectively enrolled all children presenting at the ED with acute abdominal pain indicative of possible acute appendicitis (AA). Subsequently, they were categorized into three groups: those assessed by a PC physician (PG), those brought in by their family without a prior consultation (FG), and those admitted after a PC consultation without being assessed as such. The primary objective was to assess the probability of AA diagnosis using the Pediatric Appendicitis Score (PAS). Secondary objectives included analyzing PAS and C-reactive protein (CRP) levels based on the duration of pain and final diagnoses. Results 124 children were enrolled in the study (PG, n = 56; FG, n = 55; NG, n = 13). Among them, 29 patients (23.4%) were diagnosed with AA, with 13 cases (23.2%) from the PG and 14 cases (25.4%) from the FG. The mean PAS scores for AA cases from the PG and FG were 6.69 ± 1.75 and 7.57 ± 1.6, respectively, (p = 0.3340). Both PAS scores and CRP levels showed a significant correlation with AA severity. No cases of AA were observed with PAS scores

Introduction: Endovascular aneurysm sealing (EVAS) appeared to be an innovative alternative to conventional endovascular abdominal aortic aneurysm repair. However, high rates of midterm failure of EVAS led to withdrawal of the device from the market. The study aim was to report midterm outcomes of patients treated with EVAS alone or associated with chimneys (Ch-EVAS) and the management of their complications. Methods: In this single centre study, all consecutive Nellix implants between 2013 and 2016 were included. The primary endpoint was device failure: (1) a triad of caudal migration of the Nellix stents >5 mm, separation of the endobags (>5 mm), and sac enlargement (>5 mm), with or without visible endoleak, (2) secondary aneurysm rupture, (3) surgical explant of the graft, or (4) any intervention for a type I endoleak. Overall mortality, aneurysm related mortality, and re-intervention rates were analysed. Results: Fifty patients (male n = 43, female n = 7) were included. Median follow-up was 3.05 years (interquartile range [IQR] 0.52, 4.63) and follow up index was 0.51 (IQR 0.10, 0.88). Device failures occurred in 17 patients (34%). Overall and aneurysm related mortality rates during the follow up period were 30% and 13%. Fourteen (28%) patients required re-interventions. Five EVAS patients (17%) presented with complications. Type Ia endoleaks were managed by device explantation for three patients, and endovascular aneurysm repair in Nellix for two patients. Type Ib endoleaks were managed with an iliac branched device and limb extension. Nine Ch-EVAS patients (42.9%) presented with complications. Type Ia endoleaks were was managed by Nellix stent prolongation and renal extension, two multibranched thoraco-abdominal devices, and two device explantations. Type Ib endoleaks were managed by limb extension and stent complications by stent angioplasty and iliorenal bypass. Conclusion: The midterm outcome of EVAS is poor. All patients who underwent EVAS implantation must be informed and should undergo frequent surveillance. Open repair and device explantation should be considered as the primary treatment.

Abstract Yawning is undeniably contagious and hard to resist. Interestingly, in our species, even the mere sound of a yawn can trigger this contagious response, especially when the yawner is someone familiar. Together with humans, one other mammal species is known to produce loud and distinct vocalisations while yawning, Theropithecus gelada. Geladas are known for their complex social interactions and rich vocal communication, making them intriguing subjects for studying yawning behaviour. To explore the contagious effect of yawn sounds on geladas, we conducted playback experiments in a zoo-housed colony with animals living in two groups. We exposed them to yawn sounds (Test) or affiliative grunts (Control) produced by males from either their own group or the other one. The results were remarkable, as simply hearing yawn sounds led to yawn contagion in geladas, with multiple responses observed when the yawns came from members of their own group. This finding adds a significant contribution to the research on mimicry and behavioural contagion in primates. Moreover, it raises intriguing questions about the involvement of sensory modalities beyond visual perception in these phenomena.

It has been argued that domestication explains the ability of domestic animals to use human cues, but similar abilities exist in wild animals repeatedly exposed to humans. Little is known on the importance of the developmental stage of this exposure for developing such abilities. Orphancy and subsequent hand-rearing constitute a quasi-experimental situation for investigating this question. Cheetahs (Acinonyx jubatus) are interesting as they present a two-step development, spending their first two months of life in a den with occasional visits from the mother, and then accompanying their mother outdoors. Mother loss may occur at any of these stages, and the orphaned cheetahs are then taken under human care. We tested the importance of the stage of development on their long-term perception of humans by comparing the reactions of adult wild-born cheetahs, orphaned at “den-age” (Early-Orphaned, 0–2 months) or at a later stage (Late-Orphaned, 2–6 months), to motionless humans (familiar vs. unfamiliar). The results showed that Early-Orphaned, but not Late-Orphaned cheetahs, produced affiliative calls (purrs) towards humans and discriminated familiar and unfamiliar humans. Taken together, these results suggest that the timing in which an emotional bond is created with the primary caregiver (humans in the case of Early-Orphaned cheetahs and the mother in the case of Late-Orphaned cheetahs) is crucial in determining the reaction towards the species of the early caregiver throughout life.

Abstract Objectives Certain low‐level immune‐related adverse events (irAEs) have been associated with survival benefits in patients with various solid tumors on immune checkpoint inhibitors (ICIs). We aimed to investigate the association between irAEs and response to neoadjuvant ICIs in patients with head and neck squamous cell carcinoma (HNSCC) and to identify differences in circulating cytokine levels based on irAE status. Methods This was a retrospective cohort study including three neoadjuvant clinical trials from July 2017 to January 2022: NCT03238365 (nivolumab ± tadalafil), NCT03854032 (nivolumab ± BMS986205), NCT03618654 (durvalumab ± metformin). The presence and type of irAEs, pathologic treatment response, and survival were compared. Canonical linear discriminant analysis (LDA) was performed to identify combinations of circulating cytokines predictive of irAEs using plasma sample multiplex assay. Results Of 113 participants meeting inclusion criteria, 32 (28.3%) developed irAEs during treatment or follow‐up. Positive p16 status was associated with irAEs (odds ratio [OR] 2.489; 95% CI 1.069–6.119; p = 0.043). irAEs were associated with pathologic treatment response (OR 3.73; 95% CI 1.34–10.35; p = 0.011) and with higher OS in the combined cohort (HR 0.319; 95% CI 0.113–0.906; p = 0.032). Patients with irAEs within the nivolumab cohort had significant elevations of select cytokines pre‐treatment. Canonical LDA identified key drivers of irAEs among all trials, which were highly predictive of future irAE status. Conclusions irAEs are associated with response to neoadjuvant ICI therapy in HNSCC and can serve as clinical indicators for improved clinical outcomes. irAEs can be predicted by concentrations of several circulating cytokines prior to treatment.

Introduction In light of the burden of traumatic brain injury (TBI) in children and the excessive number of unnecessary CT scans still being performed, new strategies are needed to limit their use while minimising the risk of delayed diagnosis of intracranial lesions (ICLs). Identifying children at higher risk of poor outcomes would enable them to be better monitored. The use of the blood-based brain biomarkers glial fibrillar acidic protein (GFAP) and ubiquitin carboxy-terminal hydrolase-L1 (UCH-L1) could help clinicians in this decision. The overall aim of this study is to provide new knowledge regarding GFAP and UCH-L1 in order to improve TBI management in the paediatric population.Methods and analysis We will conduct a European, prospective, multicentre study, the BRAINI-2 paediatric study, in 20 centres in France, Spain and Switzerland with an inclusion period of 30 months for a total of 2880 children and adolescents included. To assess the performance of GFAP and UCH-L1 used separately and in combination to predict ICLs on CT scans (primary objective), 630 children less than 18 years of age with mild TBI, defined by a Glasgow Coma Scale score of 13–15 and with a CT scan will be recruited. To evaluate the potential of GFAP and UCH-L1 in predicting the prognosis after TBI (secondary objective), a further 1720 children with mild TBI but no CT scan as well as 130 children with moderate or severe TBI will be recruited. Finally, to establish age-specific reference values for GFAP and UCH-L1 (secondary objective), we will include 400 children and adolescents with no history of TBI.Ethics and dissemination This study has received ethics approval in all participating countries. Results from our study will be disseminated in international peer-reviewed journals. All procedures were developed in order to assure data protection and confidentiality.Trial registration number NCT05413499.

Background. Brief normothermic machine perfusion is increasingly used to assess and recondition grafts before transplant. During normothermic machine perfusion, metabolic activity is typically maintained using red blood cell (RBC)–based solutions. However, the utilization of RBCs creates important logistical constraints. This study explored the feasibility of human kidney normothermic perfusion using William’s E–based perfusate with no additional oxygen carrier. Methods. Sixteen human kidneys declined for transplant were perfused with a perfusion solution containing packed RBCs or William’s E medium only for 6 h using a pressure-controlled system. The temperature was set at 37 °C. Renal artery resistance, oxygen extraction, metabolic activity, energy metabolism, and histological features were evaluated. Results. Baseline donor demographics were similar in both groups. Throughout perfusion, kidneys perfused with William’s E exhibited improved renal flow (P = 0.041) but similar arterial resistance. Lactic acid levels remained higher in kidneys perfused with RBCs during the first 3 h of perfusion but were similar thereafter (P = 0.95 at 6 h). Throughout perfusion, kidneys from both groups exhibited comparable behavior regarding oxygen consumption (P = 0.41) and reconstitution of ATP tissue concentration (P = 0.55). Similarly, nicotinamide adenine dinucleotide levels were preserved during perfusion. There was no evidence of histological damage caused by either perfusate. Conclusions. In human kidneys, William’s E medium provides a logistically convenient, off-the-shelf alternative to packed RBCs for up to 6 h of normothermic machine perfusion.
