Myelin is an essential component of the nervous system and myelin damage causes demyelination diseases. Myelin is a sheet of oligodendrocyte membrane wrapped around the neuronal axon. In the fluorescent images, experts manually identify myelin by colocalization of oligodendrocyte and axonal membranes that fit certain shape and size criteria. Because myelin wriggles along x-y-z axes, machine learning is ideal for its segmentation. However, machinelearning methods, especially convolutional neural networks (CNNs), require a high number of annotated images, which necessitates expert labor. To facilitate myelin annotation, we developed a workflow and a software for myelin ground truth extraction from multi-spectral fluorescent images. Additionally, we shared a set of myelin ground truths annotated using this workflow.

Merkezi sinir sisteminde bulunan oligodendrositler, nöronlara trofik destek sağlayan ve elektrik sinyallerini daha hızlı ve daha verimli iletmelerini kolaylaştıran miyelin kılıfının oluşumundan sorumludur. Bu nedenle, miyelin kılıfı sağlıklı bir sinir sisteminin gelişiminde önemli bir rol oynar. Fare embriyonik kök hücre bazlı bir in vitro miyelinasyon sistemi geliştirilmiştir ve bu sistem SARAPE adı verilen miyelizasyon modelini oluşturmak için kullanılmıştır. Bu modelde, miyelinazyonun çapa atma evresi nöron ve oligodendrosit membran arasındaki ilk temastır. Caspr proteinin nöronların yüzeyindeki Contactin'e bağlandığı ve daha sonra, miyelinasyon sürecinin başından sonuna kadar kırılmayan bir etkileşim oluşturmak için oligodendrosit membranı üzerindeki NF155 proteinine bağlandığı bilinmektedir. Ana amacımız, bu proteinleri kullanarak uzun süreli canlı görüntülemeye uygun moleküler araçlar ve protokoller geliştirerek, miyelin zarının oluşumunun arkasındaki ayrıntılı mekanizmayı anlamaya yardımcı olmaktır. Bu amaçla NF155, Caspr ve Contactin proteinlerinin floresan etiketli versiyonu, lentiviral plazmitler içerisindeki oligodendrosit ve nörona spesifik promotörler altına klonlanmış ve lentivirüsler elde edilmiştir. Daha sonra, zenginleştirme işlemi yapılarak NPH'ler de kalıcı Caspr ifadesi sağlanmış ve hücreler canlı görüntülemeye uygun hale getirilmiştir. Myelin, produced by oligodendrocytes in the central nervous system and schwann cells in the peripheral nervous system, is one of the main components required for the efficient functioning of the nervous system. By virtue of the myelin membrane, the speed and efficiency of signal transmission increases, trophic support is provided to neurons and it is easier for them to survive. Therefore, myelin sheath plays essential role in the development of a healthy nervous system. A mouse embryonic stem cell-based in vitro myelination system is developed and used to create a myelization model called SARAPE. In this model, anchoring phase of the myelination is the first contact between neuron and oligodendrocyte membrane. It has been studied that Caspr bind to Contactin on the surface of neurons. Then, they interact with their binding target on the oligodendrocyte membrane, NF155, to form an interaction which is never broken from the beginning to the end of the myelination process. Our main aim is to help understanding the detailed mechanism behind the formation of the myelin membrane by generating molecular tools and protocols using these proteins, compatible with long-term live imaging. For this purpose, fluorescent tagged version of NF155, Caspr and Contactin proteins were cloned under the oligodendrocyte and neuron specific promoters in lentiviral constructs and by using cloned constructs, lentivirus particles were obtained. Next, by performing enrichment process, NPCs with permanent Caspr expression were generated. Finally, these cells were shown to compatible for live imaging.

Zaman içerisinde teknolojinin gelişimi ile beraber yapı malzemeleri ve oluşturdukları sistemlerde gelişmiştir. Bu gelişme hızlandıkça ortaya çıkan her yeni malzeme ve sisteme, mimarların tam anlamıyla hâkim olması güçleşmiştir. Bu durum cephe için de geçerlidir. Cephe de malzemeler ve sistemler ile beraber gelişmiş ve değişmiştir. Başarılı bir cephe için malzemeleri iyi tanımak, doğru sistemi seçmek ya da tasarlamak, doğru uygulama yapmak şarttır. Alınan yanlış kararlar kusurlara, kusurlar ise hasarlara neden olabilir. Hasar oluşma riskini en aza indirmek için kararların belirli standartlar dahilinde alınması en doğru yöntemdir. Bu standartlar projelerin teknik şartnamelerinde verilir.Kaplamalı cephe sistemleri ile tasarlanan mimari projelerin teknik şartnameleri incelenmek istendiğinde bazı projelere ait teknik şartnamelerin olmadığı anlaşılmıştır. Şartnamesi olan projelerde ise, cephe sistemi bileşenlerinin üretim ve yapım standartlarında eksiklikler, sistem tasarım ve performans standartlarının verilmediği anlaşılmıştır. Çoğunlukla bileşenler için marka isimlerine ya da malzemenin boyutları hakkında kısa bilgilere yer verilmiştir. Bu duruma teknik şartnamelere önem verilmemesi veya şartnameleri hazırlayan kişilerin yeterli bilgiye sahip olmaması neden olabilir. Türkiye'de cephe sistemi bileşen üretim ve yapım, sistem tasarım ve performans standartlarını bir arada ortaya koyan bir kaynak bulunmamaktadır. Yürütülen tez çalışmasıyla kaplamalı cephe sistemi bileşen üretim ve yapım, sistem tasarım ve performans standartlarını bir arada ortaya koyan kontrol listeleri önerilmiştir. Şartnamede eksikliklerden dolayı oluşacak hasar oluşma riskini en aza indirmek amaçlanmıştır.Tez çalışması kapsamında kaplamalı cephe sistemi bileşen malzemeleri olarak çekirdek için gaz beton blok, kil tuğla, ısı yalıtım için taş yünü ve cam yünü, dış kaplama için alüminyum kompozit panel ve seramik karo seçilmiştir. Çalışma dört ana aşama takip edilerek geliştirilmiştir.İlk aşamada kaplamalı cephe sistem bileşenleri, seçilen bileşenlerin üretimi ve yapımı hakkında bilgilere ulaşılmıştır. Devamında kaplamalı cephe sisteminden beklenen performanslar tanımlanmıştır. Beklenen performansa bağlı olarak ilgili bileşenler belirlenmiştir.Sonraki aşamada ulusal ve uluslararası standartlar ile sivil toplum kuruluşlarının yayınları taranmıştır. Standart taraması için ulusal standartlardan Türk Standartları Enstitüsünün (TSE), uluslararası standartlardan American Society for Testing and Materials (ASTM), European Committee for Standardization (CEN), International Organization for Standrdization (ISO), German Institute for Standardization (DIN), British Standards Institution (BSI), Standards Council of Canada (SCC), Singapore Standards Council (SSC), Standards New Zealand / Standards Australia (NZS/AS) standartları taranmıştır. Kaplamalı cephe sistem bileşenleri için seçilen malzemelerin üretim ve yapım, sistem tasarım ve performans standartları tespit edilerek tablolar oluşturulmuştur.Çalışmanın üçüncü aşamasında Türkiye'de ki mevcut durum tespit edilmiştir. Kaplamalı cephe sistem bileşenleri için üretici firmalar ile görüşülmüştür. Yapımı içinse üretici ve yüklenici firmalar ile görüşülmüştür. Görüşmelerde kullandıkları standartlar öğrenilerek tablolar oluşturulmuştur.Son aşamada Türkiye'de yapılacak kaplamalı cephe sistemi için bileşen üretim ve yapım sistem tasarım ve performans standartlarını içeren kontrol listeleri önerilmiştir. Bu listeler literatür taraması ve firma görüşmeleri sonucu oluşturulan tabloların karşılaştırmalı olarak incelenmesi sonucu oluşturuluştur. İncelemeler sonucu TSE standartlarında eksikliği görülen ASTM, EN, ISO, DIN, BSI, SCC, SSC ve NZS/AS standartları ve sivil toplum kuruluşlarının yayınladığı standartlar ve rehberler tespit edilmiştir. Kaplamalı cephe sistem bileşeni için seçilen malzemelerin üretimi ve yapımı, sistem tasarım ve performansı için ayrı ayrı listeler hazırlanmıştır. Building materials and systems have been developed, as a result of development of technology. Facade has been developed with materials and systems as well. It becomes difficult for architects to know every new material and system that emerged. For a successful facade, it's compulsory to know necessary materials, selection of system and correct construction. Making wrong decisions can cause defects which can cause damage. It is the most accurate method to take decisions within standards to minimize the risk of damage. These standards are given in the technical specifications of the architectural projects.When technical specifications of architectural project which designed with cladding facade system, has reviewed, it has been figured out that some projects did not have technical specifications. Existing technical specifications did not include enough production and construction standards of the cladding facade system components. They just include brand names or sizes of materials and there is no information about design and performance standards of the system. The reason for this situation is not paying attention for technical specifications or lack of knowledge. The thesis is aimed to minimize the risk of damage caused by lacking of the standards. There are no sources that combine facade system component production and construction standards, system design and performance standards in Turkey. Also with the thesis study, this problem has been tried to solve by suggesting checklists which put together the cladding system components' production and construction standards, system design and performance standards.In the scope of the thesis, the cladding facade system components which are included aerated concrete block, clay brick as core material, rock wool and glass wool as thermal insulation and aluminum composite panel and ceramic tile as external covering material, have been handled. The study was developed by following seven chapter.The first chapter include problems, aims, scope of the thesis, method of study and hypothesis. By using the checklist, a successful cladding facade system will be obtained that will reduce the risk of damage, provide both external and internal comfort conditions, and have a long service life.The second chapter starts with definitions of terms related to thesis such as; enclosure, facade, external wall, curtain wall, cladding and cladding facade system. In the thesis, the cladding facade system is defined formed by the addition of the external covering component in front of the core and plaster components of the exterior wall, can be suspended or fitted to the load bearing system. The chapter continues with history and classification of facade system.After classification of facade system performance, performance requirements and related components are defined. End of the chapter definition of standard and standard types are given. Also in this part of chapter governmental and nongovernmental organizations which published national, international standards and publications are given. Method of searching standards and publications are explained.The third chapter is about literature study of cladding facade system components, expected performances from cladding facade system. Firstly, components of cladding facade system and tehcnical properties are given. After that information about selected materials which are chosen aerated concrete block, clay brick as core material, rock wool and glass wool as thermal insulation and aluminum composite panel and ceramic tile as external covering, are given. The chapter continues with production process and construction methods of selected materials. Last part of this chapter is performance requirements which expected from cladding facade system. Relevant components are identified depending on expected performance. So that natioanal, international standards and publications are searhed for in this direction.The fourth chapter includes result of searching national, international standards and publications of non-governmental organization. The Turkish Standards Institute (TSE), the American Society for Testing and Materials (ASTM), the European Committee for Standardization (CEN), the International Organization for Standardization (ISO) Institution (BSI), Standards Council of Canada (SCC), Singapore Standards Council (SSC), Standards New Zealand / Standards Australia (NZS / AS) standards have been reviewed. Tables were created by determining existing production and construction standards of the cladding facade system components, design and performance standards of the system, respectively.In the fifth chapter, the current situation in Turkey has been identified. The existing standards were compiled by talking face to face with aerated concrete block, clay brick, rock wool, glass wool, aluminum composite panel and ceramic tile manufacturers for production standards, manufacturer and contractors for construction standards also design office and facade consultants for system design and performance standards. Production process and production standards which are used in production process, were asked to manufacturers during the interview. Also aerated concrete block and aluminium composite panel' production process were observed at the factory. Design offices and facade consultants were asked about standards they use for design and performance of cladding facade system, manufacturers and constractors were asked about the standards they use for consruction of cladding facade system components. Tables were created for the standards which are used by these companies, that has learned in the interviews.In the sixth chapter, checklists have been suggested, including production and construction standards, system design and performance standards for cladding facade system to be constructed in Turkey. With the scope of the thesis, the suggestion checklists has been prepared for each cladding facade system components' production and construction, system design and performance standards, respectively. These suggestion checklists were made the result of comparison literature review and the company interviews. After review, The American Society for Testing and Materials (ASTM), The European Committee for Standardization (CEN), The International Organization for Standardization (ISO) Institution (BSI), The Standards Council of Canada (SCC), The Singapore Standards Council (SSC), The Standards New Zealand/Standards Australia (NZS / AS) standards and civil society organizations' publications which are missing in The Turkish Standards Institute (TSE) standards, have been detected. The results were put together to create suggestion checklists that architects should use.Last chapter includes of results and evaluation of the study are given for each part, separately. End of the chapter, suggestions are presented for the future studies. 135

VERGİ MÜKELLEFİNİN SUSMA HAKKINA YÖNELİK İHLALLER BAKIMINDAN ANAYASA MAHKEMESİNE BİREYSEL BAŞVURU HAKKIÖZETHukuk devleti, kişilere nazaran birçok üstün yetki ile donatılmış idare karşısında kişi temel hak ve özgürlüklerinin güvenceye kavuşturularak korunmasını sağlar. Anayasa Mahkemesine Bireysel Başvuru Hakkının iç hukukumuzda tanınması da bu amaca hizmet eder. Vergi idaresi, vergilendirme yetkisini kullanırken, mükellefin temel hak ve özgürlüklerini ihlal edebilir. Bu çalışmada, vergi mükellefinin Anayasa Mahkemesine bireysel başvuru hakkı ve bireysel başvuru konusu olarak vergi mükellefinin susma hakkı ele alınmıştır. Vergi incelemesi esnasında, mükellefin defter ve belgelerini ibraz etmemesi nedeniyle hürriyeti bağlayıcı cezaya çarptırılmasının, vergi mükellefinin susma hakkını ihlal edeceği sonucuna ulaşılmıştır. Bu nedenle çalışmanın amacı, defter ve belgeleri gizleme fiilinin suç olarak kabulünün anayasaya aykırılığına dikkat çekmektir.Çalışmada öncelikle ‘bireysel başvuru’; kavramsal, terminolojik yönleriyle açıklanmış, bireysel başvurunun tarihçesi ve uluslararası hukuktaki modelleri ele alınmıştır. Anayasa Mahkemesine bireysel başvuru, yasal dayanakları ile ele alındıktan sonra kurum vergi hukuku bakımından irdelenmiştir. Mükellefin bireysel başvuruda bulunabilmesi için taşıması gereken maddi ve usule ilişkin koşullar açıklanmıştır. Son olarak susma hakkı ve ‘defter ve belgeleri gizleme suçu’ anlatılmıştır. Netice itibariyle mevcut olağan iç hukuk yollarının, yükümlünün susma hakkına bir güvence sağlayamadığı bulgusuna ulaştıktan sonra, etkili hukuksal koruma için Anayasa Mahkemesine bireysel başvuru yolu önerilmiştir. CONSTITUTIONAL COMPLAINT REGARDING VIOLATIONS ON A TAXPAYER'S RIGHT TO SILENCEABSTRACTState of law protects fundamental rights and liberties vis-à-vis administration granting authorization on many aspects compared to persons. A recognition of constitutional complaint procedure is in the scope of this aim. Tax Administration may violate a taxpayer's basic rights and freedoms while exercising its taxation power. In this paper, the right to apply to the Constitutional Court and the right to silence of a taxpayer as a constitutional complaint are examined. During a tax audit, if a taxpayer does not submit its records & documents and this is because liberty binding punishment is inflicted on him/ her, it is claimed that these may violate the right to silence of a taxpayer. Therefore, the main aim of this paper is showing a secretion of records & documents as a crime is against the Constitution. Firstly in this paper, constitutional complaint is explained regarding its notional and terminological aspects. Then, the history of constitutional complaint and its different models in international law are discussed. After an examination of constitutional complaint's legal basis, this institution is addressed in terms of tax law. Substantive and moral conditions for a taxpayer on an application to constitutional complaint are clarified. Finally, the right to silence and a secretion of records & documents as a crime are presented. Consequently, it is concluded that existing domestic remedies on the right to silence does not provide a taxpayer with an assurance. This is because, constitutional complaint is suggested for an effective protection.

8. Oligodendrocyte interactome in healthy and diseased nervous system.

Objective: Myelin is essential for a healthy nervous system. Myelin formed by oligodendrocytes, accelerates impulse propagation and supports neuronal survival. Demyelination leads to neurodegeneration. In multiple sclerosis (MS) immune attack results in demyelination. Our goal is to dissect interactions among oligodendrocytes, neurons, and immune cells to improve our understanding of myelination and the demyelinating diseases. We aim to discover new targets for remyelination therapies. Methods: We are building tools for analyzing protein-protein and cell-to-cell interactions. To identify genes involved in myelination and MS, we developed a bioinformatics-based strategy, interactome analysis, which combines proteome and gene expression methodologies. Identified genes are evaluated in peripheral blood of MS patients and in mouse models. To accelerate drug discovery, 23 machine learning-based methodologies were assessed for myelin identification in fluorescent microscopy images. Results: Interactome analysis identified hundreds of proteinprotein interactions between pairs of oligodendrocytes, neurons, macrophages, microglia, and T cells. Most significant interactions are further analyzed in vivo and in vitro. Our customizedconvolutional neural network and Boosted Tree methods segmented myelin at over 98% accuracy. Identified myelin can be quantified and visualized in 3D. Conclusion: The interactome analysis yielded novel genes that are likely to be linked to MS. Machine learning-based methodologies are very effective in accelerating myelin quantification and thus, drug screens against demyelinating diseases such as MS. Overall, our innovative analysis strategies employing computer assistance produced novel avenues of exploration for myelination and demyelinating diseases. OBJECTIVES: Skull base is an important and a challenging area for surgeons. Success in skull base surgery depends on various factors such as pre-operative evaluation, appropriate surgical technique, anesthesia duration, intraoperative neuromonitorization and wound care. MATERIALS and METHODS: This study was performed in the Anatomy dissection laboratory of M.U. Medical Faculty (Ethical committee approval number 2010-103). Twelve fetuses between 17-33 gestational weeks fixed with formaldehyde were enrolled to the study. RESULTS: This study was planned to investigate the cross sectional areas of the sigmoid sinus in three levels to compare the right-left sides and the probable relationship among the levels in fetuses to further delineate the developmental factors on jugular foramen asymmetry. The cross-sectional measurements of sigmoid sinus lumen were done on 3 levels which are described as A1 level; sinodural angle, A2 level; the midpoint between the sinodural angle and endocranial orifice and A3 level as the entrance (endo-cranial orifice) of the jugular foramen. There is a strong positive correlation between left (L) A1 and L A2 and also the same for L A1 and right (R) A2. These strong and positive correlations are all valid between L A2-L A3, L A2-R A2, L A2-R A3, L A3-R A3, R A1-R A2. CONCLUSION: Multicenter studies would be beneficial to investigate the topic with greater number of fetuses also on the different regions for genetic differences. [ABSTRACT FROM AUTHOR]

OBJECTIVE: There is still ongoing research on the relationship of arcuate eminence (AE) and superior semicircular canal (SSC). We aimed to evaluate the precision of predictability of SSC through the morphology of AE via radiological means. Computed tomography (CT) assessment is performed with 0.5-mm-thin sections temporal bone algorithm on dry skulls which were marked with fixated copper wire by scotch tapes on the most prominent part of the middle fossa floor assuming the location of AE. The data are reformatted on the workstation with vitrea 2.0. The distances of the determined three points including lateral (A), apical (B), and medial (C) of the SSC and the copper wire are measured radiologically. Also, the height between the most apical part of the SSC to the floor of the skullbase (H) is measured. The angles between the placed copper wires and the SSC (E) are calculated. The angle between SSC and the midpoint of the IAC (F) and SSC to the sulcus of the greater GSPN (G) were measured. The nearest distance was measured between the most posterior part of the SSC and the point marked by the perpendicular line drawn from the medial border of the petrous bone to the most posterior part of the internal auditory canal (IAC) (D). RESULTS: The right and left A, B, and C distances are 2.54+/- 2.75, 3.67+/-3.16, 5.85+/-3.77; 2.92+/-2.24, 3.68+/-2.93, 6.09+/-3.40, respectively. We could not find any statistical significance when the right A, B, and C distances were compared with the left values. Examination of the values revealed that C distance is greater than the A distance of the same side both for right (p=0.040) and left (p=0.022) measurements. The calculated left and right E angles are 30.313+/-12.838, and 35.558+/-18.437 degrees, respectively. Statistical significance was not found between the right and left angles. The right and left F, G angles were 53.17, 47.25; 93.58, 100.92 degrees; and D distances are 8.01, 8.13 millimeters, respectively. Statistical significance was not found when right and left E, F, G angles and D distances were compared. Among 12 left and 12 right sides, the copper wire was found to be nearly overlapping to SSC in two in the right and only one in the left. CONCLUSION: This study reveals that there is a great variability predicting the exact location of SSC through the prominence of AE. Complementary studies are needed with greater number of dry skulls and cadavers. Comparison of different hypothesis including the effect of temporal lobe sulcus is to be discussed to better enlighten the exact relationship of the aforementioned anatomical structures. OBJECTIVE: This study planned to evaluate the effects of vitamin A (VA) and steroid treatment on nerve healing. Study design: we investigated electrophysiological, electron microscopic (EM), immunohistochemical, and biochemical findings in rat sciatic nerve model. MATERIALS and METHODS: In total, 112 female rats were divided into 16 groups, each consisting of 7 animals. Subjects were evaluated according to compound muscle action potential (CMAP) findings, ultrastructural staging of myelinated axons with EM, nitric oxide (NO) and malondialdehyde (MDA) findings in serum and tissue, and immunohistochemical findings in the first week and first month. RESULTS: NO and MDA levels in serum and tissue were found to be statistically lower after high-dose methylprednisolone (MP), normal-dose MP, high-dose MP+VA, normal-dose, and MP+VA treatment modalities compared to controls (p<0.05). The most similar EM findings compared to control animals were seen in the normal-dose MP and high-dose MP+VA treatment groups. The best amplitude values were seen in the high-dose MP and continuous normal-dose MP therapy groups in the first month. In the normal-dose MP and high-dose MP+VA treatment groups, intense staining was found compared to the control and sham groups according to the sciatic nerve immunohistochemical examination. CONCLUSION: This study revealed noticeable VA effects on the inflammatory process during the healing of traumatic nerve paralysis by electron microscopy and immunohistochemistry. However, similar findings were not established by electrophysiology. Therefore, more experimental studies are needed to further investigate the efficacy and the mechanism of action of VA on steroid administration and the inflammatory process. [ABSTRACT FROM AUTHOR]
