Introduction: Heat stress caused by global warming adversely affects wheat yield through declining most nutritional quality attributes in grains, excluding grain protein content. Methods: This research investigated the biochemical, physiological, and antioxidant responses of wheat plants under heat stress, focusing on the role of plant growth-promoting bacteria (Micromonospora sp.). Two wheat genotypes were studied: one heat-sensitive and one heat-tolerant, examining their responses to heat stress with and without bacterial inoculation. Results: Under heat stress, the sensitive cultivar experienced significant reductions in photosynthesis rate, chlorophyll content, and RuBisCO activity (57-61%), while the tolerant cultivar had milder reductions (24-28%). Micromonospora sp. treatment notably improved these parameters in the sensitive cultivar (+48-78%), resulting in a substantial increase in biomass production (+43-53%), which was not seen in the tolerant cultivar. Additionally, oxidative stress markers (H2O2 and MDA) were elevated more in the sensitive cultivar (82% and 90% higher) compared to the tolerant one. Micromonospora sp. treatment effectively reduced these markers in the sensitive cultivar (-28% and -27%). Enhanced activity of antioxidant enzymes and ASC-GSH pathway enzymes was particularly evident in Micromonospora sp.-treated sensitive plants. Carbohydrate metabolism shifted, with increased soluble sugars and significant rises in sucrose content in Micromonospora sp.-treated plants under stress. Discussion: The higher soluble sugar levels facilitated amino acid synthesis, contributing to biosynthesis of secondary metabolites, including flavonoids, polyphenols, and anthocyanins. This was reflected in increased activity of phenylalanine ammonia-lyase, cinnamate (CA) 4-hydroxylase, and chalcone synthase enzymes, indicating the activation of phenylpropanoid pathways. Overall, the findings suggest that Micromonospora sp. can mitigate heat stress effects by enhancing photosynthetic efficiency, antioxidant defense, and metabolic adaptations in heat-sensitive wheat cultivars. Soil remediation for cadmium (Cd) toxicity is essential for successful tobacco cultivation and production. Melatonin application can relieve heavy metal stress and promote plant growth; however, it remains somewhat unclear whether melatonin supplementation can remediate the effects of Cd toxicity on the growth and development of tobacco seedlings. Herein, we evaluated the effect of soil-applied melatonin on Cd accumulation in tobacco seedlings, as well as the responses in growth, physiological and biochemical parameters, and the expression of stress-responsive genes. Our results demonstrate that melatonin application mitigated Cd stress in tobacco, and thus promoted plant growth. It increased root fresh weight, dry weight, shoot fresh weight and dry weight by 58.40%, 163.80%, 34.70% and 84.09%, respectively, compared to the control. Physiological analyses also showed significant differences in photosynthetic rate and pigment formation among the treatments, with the highest improvements recorded for melatonin application. In addition, melatonin application alleviated Cd-induced oxidative damage by reducing MDA content and enhancing the activities of enzymatic antioxidants (CAT, SOD, POD and APX) as well as non-enzymatic antioxidants (GSH and AsA). Moreover, confocal microscopic imaging confirmed the effectiveness of melatonin application in sustaining cell integrity under Cd stress. Scanning Electron Microscopy (SEM) observations illustrated the alleviative role of melatonin on stomata and ultrastructural features under Cd toxicity. The qRT-PCR analysis revealed that melatonin application upregulated the expression of photosynthetic and antioxidant-related genes, including SNtChl, q-NtCSD1, NtPsy2 and QntFSD1, in tobacco leaves. Together, our results suggest that soil-applied melatonin can promote tobacco tolerance to Cd stress by modulating morpho-physiological and biochemical changes, as well as the expression of relevant genes. Panicum maximum is planted extensively in tropical and subtropical areas, due to its high-quality forage and high biomass yield. This study aims to assess the varied metabolic dynamics of P. maximum subject to different pollution-related wastewater levels, thus providing information for sustainable agriculture and soil restoration. We analyzed the primary and secondary metabolites in P. maximum subject to two different types of polluted wastewater (WW), compared to a control group. The alterations observed in the metabolite profiles were affected by several factors, including nutrient imbalances and oxidative stress induced by heavy metal accumulation. Initially, the increased nutrient availability stemming from wastewater treatment promoted plant growth; however, this positive effect was later diminished by the adverse impacts of heavy metals, which generated oxidative stress, resulting in metabolic disturbances and a decrease in the plant biomass. Importantly, the substantial increase in antioxidant enzymes, related to primary (e.g., sugars) and secondary metabolites (e.g., phenolics and flavonoids), underscores plants' adaptive strategies to cope with stress. The increased biosynthesis of flavonoids and phenolic compounds is a protective mechanism against oxidative stress, which also improves the antimicrobial activity, following the activation of key biosynthetic pathways involved in their synthesis. These complex interactions among diverse metabolites suggest that plants exposed to polluted wastewater use various biochemical strategies to increase both their survival and defenses against pathogens. Collectively, these findings emphasize the significance of understanding how wastewater management practices can affect plant health, metabolic responses, and the broader implications for food safety and ecosystem stability. Amaranth is a nutritionally valuable crop, as it contains phenolic acids and flavonoids, yielding diverse plant secondary metabolites (PSMs) like phytosterol, tocopherols, and carotenoids. This study explored the variations in the contents of seventeen polyphenolic compounds within the leaves of one hundred twenty Amaranthus accessions representing nine Amaranthus species. The investigation entailed the analysis of phenolic content across nine Amaranthus species, specifically A. hypochondriacus, A. cruentus, A. caudatus, A. tricolor, A. dubius, A. blitum, A. crispus, A. hybridus, and A. viridis, utilizing ultra performance liquid chromatography with photodiode array detection (UPLC-PDA). The results revealed significant differences in polyphenolic compounds among accessions in which rutin content was predominant in all Amaranthus species in both 2018 and 2019. Among the nine Amaranthus species, the rutin content ranged from 95.72 ± 199.17 μg g−1 (A. dubius) to 1485.09 ± 679.51 μg g−1 (A. viridis) in 2018 and from 821.59 ± 709.95 μg g−1 (A. tricolor) to 3166.52 ± 1317.38 μg g−1 (A. hypochondriacus) in 2019. Correlation analysis revealed, significant positive correlations between rutin and kaempferol-3-O-β-rutinoside (r = 0.93), benzoic acid and ferulic acid (r = 0.76), and benzoic acid and kaempferol-3-O-β-rutinoside (r = 0.76), whereas gallic acid showed consistently negative correlations with each of the 16 phenolic compounds. Wide variations were identified among accessions and between plants grown in the two years. The nine species and one hundred twenty Amaranthus accessions were clustered into six groups based on their seventeen phenolic compounds in each year. These findings contribute to expanding our understanding of the phytochemical traits of accessions within nine Amaranthus species, which serve as valuable resources for Amaranthus component breeding and functional material development. Background: The chickpea pod borer Helicoverpa armigera (Hübner) is a significant insect pest of chickpea crops, causing substantial global losses. Methods: Field experiments were conducted in Central Punjab, Pakistan, to investigate the impact of biotic and abiotic factors on pod borer population dynamics and infestation in nine kabuli chickpea genotypes during two cropping seasons (2020-2021 and 2021-2022). The crops were sown in November in both years, with row-to-row and plant-to-plant distances of 30 and 15 cm, respectively, following a randomized complete block design (RCBD). Results: Results showed a significant difference among the tested genotypes in trichome density, pod wall thickness, and leaf chlorophyll contents. Significantly lower larval population (0.85 and 1.10 larvae per plant) and percent damage (10.65% and 14.25%) were observed in genotype Noor-2019 during 2020-2021 and 2021-2022, respectively. Pod trichome density, pod wall thickness, and chlorophyll content of leaves also showed significant variation among the tested genotypes. Pod trichome density and pod wall thickness correlated negatively with larval infestation, while chlorophyll content in leaves showed a positive correlation. Additionally, the larval population positively correlated with minimum and maximum temperatures, while relative humidity negatively correlated with the larval population. BACKGROUND RESULTS CONCLUSION It is imperative to enhance the quality of sprouts since they are a rich source of various primary and secondary metabolites. The objective of this work was to examine how multiwalled carbon nanotubes (MWCNTs) priming at various concentrations affected the nutritional qualities of four horticultural plants (T. foenum‐graecum, L. grandiflorum, L. sativum and A. graveolens) and their sprouting processes.Among the four applied concentrations (10–60 mgL−1), MWCNTs at 10 and 40 mg L⁻¹ induced the highest biomass accumulation in L. grandiflorum and T. foenum‐graecum, respectively, while 60 mg L⁻¹ was most effective for L. sativum and A. graveolent. MWCNTs induced growth by enhancing photosynthesis, as shown by increased chlorophyll content and rubisco activity, which rose by 27%, 17%, 23% and 12% in T. foenum‐graecum, L. grandiflorum, L. sativum, and A. graveolens, respectively. Enhanced photosynthesis by MWCNTs improved sugar metabolism as indicated by increased activity of sugar metabolic enzymes such as amylase, starch synthase and invertase. This also supplied the carbon necessary for the production of primary (amino acids, fatty acids and organic acids) and secondary (flavonoids and polyphenols) metabolites. There was consistently higher activity of antioxidant enzymes (catalase and peroxidase). • Amaranthus retroflexus L. is one of the most prevalent plant species in a contaminated area. • A. retroflexus was able to modify metabolic processes. • The synthesis of polyamines and amino strategies can cause tolerance in the plants to heavy metal stress. • Antioxidant defense strategy emerged as a reliable protection mechanism for plants in contaminated soils. One of the most prevalent plant species in the contaminated area around a sewage dumping lake at Khulais (Jeddah, Saudi Arabia) in the Arabian desert eco-region was found to be Amaranthus retroflexus L. This plant has the capacity to bioaccumulate and bioremediate heavy metals. The current study, therefore, aimed at investigating the plant's defense mechanisms by conducting metabolic and biochemical assessments. To this end, A. retroflexus plants were collected from the sewage dumping lake of Khulais, across five sites at varying distances from the lake, each exhibiting different level of heavy metal. The results indicated an increase in antioxidant defense system emerged as a protection strategy for A. retroflexus plants against soil contaminations. This, for instance, included the increased synthesis of polyamines (e.g., putrescine, spermidine, and spermine by +9–63%), flavonoids (e.g., naringenin, kaempferol, luteolin, and rutin by +10–146%), and phenolic acids (e.g., chlorogenic acid, gallic acid, caffeic acid, p-coumaric acid, chicoric acid, rosmarinic acid, and protocatechuic acid), increased by 10–254%. Additionally, plants adjusted their metabolic processes by synthesizing various low molecular weight amino acids, including proline (+19–299%), phenylalanine (+98–240%), glutamate (+34–492%), arginine (+10–64%), and ornithine (+51–210%). The activity of the metabolic enzymes involved in metabolism of these amino acids was enhanced accordingly. It can be concluded that the synthesis of polyamines and amino acids can be coordinated and complemented to improve the tolerance of A. retroflexus to cope with heavy metal accumulation in contaminated soils.
