Mitotic kinase Aurora A (AURKA) diverges from other kinases in its multiple active conformations that may explain its interphase roles and the limited efficacy of drugs targeting the kinase pocket. Regulation of AURKA activity by the cell is critically dependent on destruction mediated by the anaphase-promoting complex (APC/CFZR1) during mitotic exit and G1 phase and requires an atypical N-terminal degron in AURKA called the "A-box" in addition to a reported canonical D-box degron in the C-terminus. Here, we find that the reported C-terminal D-box of AURKA does not act as a degron and instead mediates essential structural features of the protein. In living cells, the N-terminal intrinsically disordered region of AURKA containing the A-box is sufficient to confer FZR1-dependent mitotic degradation. Both in silico and in cellulo assays predict the QRVL short linear interacting motif of the A-box to be a phospho-regulated D-box. We propose that degradation of full-length AURKA also depends on an intact C-terminal domain because of critical conformational parameters permissive for both activity and mitotic degradation of AURKA.

International audience; We describe a slowly progressive myopathy in 7 unrelated adult patients with storage of polyglucosan in muscle fibers. Genetic investigation revealed homozygous or compound heterozygous deleterious variants in the glycogenin-1 gene (GYG1). Most patients showed depletion of glycogenin-1 in skeletal muscle, whereas 1 showed presence of glycogenin-1 lacking the C-terminal that normally binds glycogen synthase. Our results indicate that either depletion of glycogenin-1 or impaired interaction with glycogen synthase underlies this new form of glycogen storage disease that differs from a previously reported patient with GYG1 mutations who showed profound glycogen depletion in skeletal muscle and accumulation of glycogenin-1. Turkey was the fourth and third largest producer of tomato and pepper fromgreenhouses, respectively, according to FAO in 2012. The huge amount of agricultural wastesgenerated from the production of tomato and green pepper are served as renewable sources forenergy production via anaerobic digestion (AD) For efficiently biogas production, it is essential todetermine the operation conditions of AD such as solid loading rate (SLR) for target wastes.The aim of this study is to optimize SLR for optimum biogas production from tomato and greenpepper wastes. For this purpose, biochemical methane potential tests were carried out at 37 °Cwith 7% and 15% of SLRs. For supplying the nutritional requirements of AD process, cow manurewas also added as a 10% of the selected SLR. As the biogas production from tomato wastes wasnearly 75% increased with the increase in SLR from 15% to 7%., the optimum SLR wasdetermined as 7% for anaerobic digestion of tomato wastes. On the other hand, decreasing SLR ofpepper waste from 15% to 7% is resulted with only 11% decrease in biogas production. When theenergy revenue between the 7% and 15% SLRs is compared with disposal cost of nearly two-foldpepper wastes, suitable SLR for pepper wastes can be suggested as 15%. Consequently, theoptimum SLR for anaerobic digestion of tomato and pepper was determined as 7% and 15%,respectively.

Alternatif poliadenilasyon (APA) farklı fizyolojik ve/veya hastalık durumlarında mRNA 3'-UTR'lerinin (translasyon olmayan bölge) kısalmasına neden olabilir. Memeli genlerinin yaklaşık yarısı alternatif kesilme ve poliadenilasyon ile 3'-UTR boyları değişen çeşitli mRNA izoformları oluşturabilirler. Sonuç olarak, mRNA'ların 3'-UTR'lerinin kısalmasının gen ifadesi açısından önemli sonuçları vardır. APA'nın hızlı büyüyen hücrelerde ve kanser hücrelerinde kullanılan bir mekanizma olduğu yakın zamanda anlaşılmıştır. APA ve 3'-UTR kısalması olaylarının kanser hücrelerinde ortaya çıkan birtakım proto-onkogen aktivasyonlarını açıklayabileceği düşünülmektedir. Çalışmamızda, bazı hücrelerin translasyon hızlarını arttırmak amacıyla proksimal poly(A) sitelerini tercih ettiklerine dayanarak, östrojen reseptör pozitif (ER+) meme kanseri hücre hatlarına östrojene (E2) bağlı olarak hızlı büyümeye olanak sağlayacak proksimal poly(A) sitelerinin tercih edilebileceğini düşündük. Bağımsız ifade mikroçiplerinde yapılan öncül prob-tabanlı analizler DNA replikasyonunda önemli bir regülatör olan CDC6'nın (Cell Division Cycle 6) E2 muamelesi sonrası ifadesinde artış ve 3'-UTR'sinde kısalma olduğuna işaret etmiştir. Takip eden deneylerde, CDC6'nın E2 ve ER'ye bağlı ifade artışı ve 3'-UTR kısalması, buna bağlı olarak CDC6 protein seviyesindeki ve BrdU eklenmesindeki artış doğrulanmıştır. Daha sonra yapılan miRNA bağlanma tahminleri ve dual-lusiferaz sonuçları CDC6'nın 3'-UTR'sinin kısalmasının 3'-UTR'a bağlı negatif regülasyonlardan kaçınmak amacıyla olduğuna işaret etmiştir. Sonuç olarak, E2'nin ER+ hücrelerde bölünmeyi tetikleyen etkisinin CDC6 APA'sını da etkilediğini göstermiş, ve de APA'nın komplike regülasyonuna yeni bir bakış açısı sağlanmıştır. Alternative polyadenylation (APA) may cause mRNA 3?-UTR (untranslated region) shortening in different physiological conditions and/or disease states. About half of mammalian genes use alternative cleavage and polyadenylation to generate multiple mRNA isoforms differing in their 3?-UTRs. Consequently, 3?-UTR shortening of mRNAs via APA has important consequences for gene expression. Studies showed that, by preferential use of proximal APA sites and switching to shorter 3?-UTRs, proliferating cells can avoid 3?-UTR dependent negative post-transcriptional regulations. Hence, such APA and 3?-UTR shortening events may explain the basis of some of the proto-oncogene activation cases observed in cancer cells. Based on the fact that certain cell types switch to more proximal poly(A) sites to rapidly increase translation rates by escaping from miRNAs, we hypothesized that upon estradiol (E2) treatment in estrogen receptor positive (ER+) breast cancer cells, such proximal poly(A) sites may be preferred to provide a rapid growth pattern. Initial probe based screen of independent expression arrays suggested upregulation and 3?-UTR shortening of an essential regulator of DNA replication, CDC6 (Cell Division Cycle 6), upon E2 treatment. Further investigations confirmed the E2 and ER dependent upregulation and 3?-UTR shortening of CDC6, which lead to increased CDC6 protein levels and higher BrdU incorporation. Consequently, miRNA binding predictions and dual luciferase assays suggested that 3?-UTR shortening of CDC6 was a likely mechanism to avoid 3?-UTR dependent negative regulations. In summary, we demonstrated CDC6 APA induction by the proliferative effect of E2 in ER+ cells and provided new insights into the complex regulation of APA. E2 induced APA is likely to be an important but previously overlooked mechanism of E2 responsive gene expression. 102

Türkiye'den gerçekleşen yurt dışı işgücü göçünün Türkiye ekonomik büyümesine ve çeşitli ekonomik değişkenlerine döviz transferleri yoluyla etkisinin araştırılması bu çalışmanın ana konusudur. Yurt dışı göçten kaynaklanan döviz transferlerinin göç veren ülkelerin ekonomilerine etkileri üzerine literatür taraması mercek altına alınmıştır.Bu doğrultuda yurt dışı işgücü göçünün Türkiye'nin ekonomik büyümesini etkilediği kabul edilen bazı ekonomik değişkenlere etkisi, regresyon yöntemi ile analiz edilmiştir. Ampirik bulgulara dayalı ekonometrik analiz sonucu göçmen işgücü döviz girdisinin Türkiye'nin ekonomik büyümesi etkisinin istatistiksel olarak anlamsız ve önemsiz olduğu bulgusuna ulaşılmıştır. Buna göre bu tez yaygın olan algılamanın aksine Türkiye'den gerçekleşen göçün Türkiye'nin ekonomisi üzerinde önemli etkileri olmadığını ortaya koyarak halihazırdaki literatürü genişletmektedir.Yurt dışı işgücü göçünü döviz transferlerinin etkilediği çok sayıda ekonomik faktörler arasından (i) GSYİH, (ii) Cari İşlemler Açığı, (iii) Enflasyon, (iv) İşsizlik ve (v) Gini Katsayısı tanımlayıcı ve ekonometrik olarak incelenmiştir. Regresyon analizlerimizin güvenirliğini test etmek için değişkenler Augmented Dickey-Fuller birim kök testi ile sınanmıştır. Bu bağlamda bulduğumuz değerler çerçevesinde regresyon testlerimizin güvenilir olduğunu söyleyebiliriz. Tez çalışması bu hedef doğrultusunda özetle aşağıdaki dört bölümden oluşmaktadır.Tezin birinci bölümünde; öncelikle ekonomik büyümede yurt dışı işgücü göçü ile döviz transferleri faktörüne ve literatürüne genel bir bakıştan sonra, yurt dışı işgücü göçünün tanımı ve sınıflandırılması yapılmıştır. Ayrıca, yurt dışı işgücü göçü yaklaşımları, yurt dışı göç süreci ve tercih modelleri araştırılmıştır. Daha sonra, modelde kullanacağımız değişkenlerden eğitim ve gelir etkisi değişkeni incelenmiş; yurt dışı işgücü göçünün makro ve mikro belirleyicilerine yer verilmiştir.xiiiÇalışmanın ikinci bölümünde; yurt dışı işgücü göçü, işçi döviz transferleri ve ekonomik etkilerine, yurt dışı işgücü göçünün ekonomik büyümedeki rolüne, Türkiye'de yüksek öğretim öğrenci sayısı ve yurt dışı işgücü göçü arasında benzerlik gösteren ilişki ve işgücü göçünün döviz geri dönüşü fonksiyonu ile olduğu öngörülen negatif korelasyon incelenmiştir.Tezin üçüncü bölümünde; yurt dışı işgücü göçünün ekonomik ve ekonometrik analizi yapılmıştır. Bu çerçevede öncelikle işgücü arz ve talep dengesizliği, yurt dışı göç ve ticaret hacmi ilişkisi ve literatürdeki çalışmalar, yurt dışı işgücü döviz transferlerine yönelik yaklaşımların üzerinde durulmuştur. Daha sonra döviz havalelerini etkileyen faktörler olan; sosyo-ekonomik ve mali faktörler ve ayrıca bu ekonomik faktörlerin döviz transferleri ile olan ilişkisi ekonometrik olarak incelenmiştir. Bölümde son olarak; yurt dışı işgücü göçünü açıklayan kuramlara yer verilmiştir.Çalışmanın dördüncü bölümünde; yurt dışı işçi döviz transferlerinin Türkiye'nin çeşitli ekonomik değerlerine etkileri kurulan model ile regresyon yönteminden faydalanılarak analiz edilmiş, hipotezler test edilmiş, çıktılar değerlendirilmiş ve yorumlanmıştır.Çalışmanın ek kısmında, yüz yüze görüşme yöntemi ile üniversite öğrencileriyle yapılan yurt dışı işgücü göçü anketi değerlendirilmiş, çeşitli hipotezler sınanmış ve sonuçlar yorumlanmıştır. The paper uses the time series regression analyses to determine the impact of remittances over the economic growth in Turkey while the sample survey tries to determine potential future out-migration.After an extensive literature screening, this study provides further insight into arguments relating to number of economic factors affecting international labor migration and remittances. The findings of the econometric analysis, which is based on empirical evidence, unveils that migrant labors remittances do not have any statistically significant effect on the economic growth of Turkey. Hence, in contrary to common consensus, this thesis expands the existing literature via revealing that there is not much impact of migration originating from Turkey on the Turkish economy.For Turkey this paper uses cross-section data for GDP, current account deficit, inflation, unemployment and the Gini coefficient which uses descriptive and econometric analyses to test between 1960-2009 data. In order to determine the reliability of our regression analyses this paper uses, unit root variables with time series and the Augmented Dickey-Fuller test. In this context, we find our test results very reliable. This thesis consists of four sections outlined below.The first part of the thesis is an overview of the literature, looking at the definition and classification of international migration. In this context, the paper investigates international labor migration, international migration process and the preferred economic model.xvThe second part of the study investigates the role of the international labor migration over the economic growth in Turkey, in relation to the number of students in higher education and the remittances with their economic impact on receiving countries.In the third section of the thesis; the time series regression findings with the help of the econometric model determines the impact of remittances over the economic growth of the Turkish economy.In the fourth section of the study; tapping into the Saat series regression method used in the model tells us the effects of overseas remittances over Turkish economic growth not to be significant. The time series regression findings show that remittance flow to Turkey have statistically meaningful but have no significant impact on growth.In the annex sample survey results are also assessed in connection with the regression analyses. Aim: An experimental study was performed to evaluate the protective effect of platelet-derivated-growth factor inhibitor (Trapidil) in ischemia reperfusion injury in intussusception. Methods: Sixty Wistar albino rats, weighing 200-250 g in both sexes were enrolled into study. In control group (CG, n:15), 2 cm of intestinal segment, 20 cm proximal to ileoceacal valve was removed after intraperitoneal NaCl infusion. In intussusception group, intussusception (IG, n:15) was performed at 20 cm proximal to ileoceacal valve by pushing the proximal intestinal segment distally through a stile. Two cm of intestinal segment was removed 4 hours after intussusception. Intussusception-reperfusion group (IRG, n:15) was obtained after performing intussusception similar to IG and allowed to perfusion 4 hours after intussusception. In this group, intestinal segment was removed 4 hours after reperfusion. In the last group (TG, n:15), Trapidil (40 mg/kg) was administered intraperitoneally, one hour before reperfusion, after achiving an ischemia/reperfusion injury similiar to IRG. Two cm of intestinal segments, 20 cm proximal to ileoceacal valve, were removed for histopathologic evaluation. The specimens were also evaluated for oxidative injury markers (catalase, glutathione peroxidase-G-px, superoxide dismutase-SOD, and malondialdehyde-MDA). 17q23 amplikon bölgesi meme kanserinde amplifiye olan birçok bölgeden bir tanesidir.Bu tür amplikonlar gen amplifikasyonlarından dolayı aşırı ekspres edilen proto-onkogenleribarındırırlar. Meme kanseri hücre hatlarında ve meme tümörlerinde yapılmış olan kopya sayısıanalizleri amplikon içerisinde birçok birbirinden bağımsız bölgenin amplifiye olduğunubelirlemiştir, bu da bazı genlerin çoğalmak üzere seçilerek tümör oluşumu ve gelişiminekatkıda bulunduğunu göstermektedir.17q23 üzerindeki amplikonları karakterize etmek ve olası onkogenleri belirlemek içinçeşitli çalışmalar yapılmıştır. 17q23 amplikonunun çoğalmasının meme kanserinde rolünüanlamak amacıyla, bu amplikon üzerindeki belirlenmemiş genleri veya EST'leri (ExpressedSequence Tags-İfade edilen Dizi Etiketleri) karakterize etmek çok önemlidir. Bu çalışmada,insan (H.sapiens), şempanze (P.troglodytes) ve fare (M.musculus) genomlarının in silikoanalizleri diziler arsındaki homolijiyi araştırmak ve yeni genleri ortaya çıkarmak amacıylayapılmıştır.Bu çalışmanın amacı, TBX2 ve TBX4 genleri arasındaki 50 kb'lik bölgenin analiziedilmesi ve bu bölgede yeralan T02811 EST'sinin karakterizasyonudur. Bu üç memeliningenomlarının analizleri ve karşılaştırılması, 50 kb'lik genomik DNA dizisi ve bu bölgedeyeralan EST'ler kullanılarak çeşitli biyoinformatik araçlar (BLAST, Pipmaker, Vista,GENSCAN vs.) ile yapılmıştır. Elde edilen sonuçlar göstermektedir ki, 3 tür arasındahazırlanan homoloji haritasına ve EST'lerin pozisyonlarına göre bu bölgede henüztanımlanmamış çeşitli büyüklüklerde yeni genlerin olması mümkündür. Bu 50 kb'lik bölgede,henüz tanımlanmamış bir genin varlığına dair in siliko ve deneysel ipuçları belirledik. T02811transkriptini 313 bp'den 1423 bp'ye uzatmayı başardık.Bu yeni gen potensiyel bir onkogen adayı olup olmadığını bulmak için daha fazlakarakterizasyon çalışmaları yapılmalıdır. Such amplicons harbor proto-oncogenes that may be overexpressed due to geneamplifications. Copy number analysis in breast cancer cell lines and breast tumors identifiedseveral independently amplified regions within the 17q23 amplicon, suggesting that a numberof genes are selected for amplification as they may independently contribute to tumorformation and progression.To characterize distinct amplicons on 17q23 and localize putative oncogenes, variousstudies are done on this region. In order to better understand the role of 17q23 amplification inbreast cancer, characterizing unidentified genes or ESTs (Expressed Sequence Tags) on the17q23 amplicon is crucial. In this study, in silico analysis of human (H.sapiens), chimpanzee(P.troglodytes), and mouse (M.musculus) genomes were performed to examine sequencehomology between these 3 species for the purpose of identifying novel genes.The objective of this study was to analyze a 50 kb region between TBX2 and TBX4genes and characterize the EST T02811 located on that region. Analysis and comparisons ofthese three genomes were established based on genomic sequences and availability of ESTswith gene prediction programs (BLAST, Pipmaker, Vista, GENSCAN etc.). Based on ourresults, we prepared a homology map between these 3 species, including positions of ESTsthat may indicate novel genes. In this 50 kb region, we found in silico and experimantalevidence for the presence of an unidentified gene. We managed to extend the 313 bp ESTT02811 size to 1423 bp which we think is as of yet incomplete.Further studies are needed to characterize this novel gene as a potential oncogenecandidate.
