Magnesium alloys present a compelling prospect for absorbable implant materials in orthopedic and trauma surgery. This study evaluates an ultra-high purity, lean magnesium–calcium alloy (X0), both with and without plasma electrolytic oxidation (PEO) surface modification, in comparison to a clinically utilized WE43 magnesium alloy. It is shown that the mechanical properties of X0 can be tuned to yield a high-strength material suitable for bone screws (with an ultimate tensile strength of 336 MPa) or a ductile material appropriate for intraoperatively deformable plates (with an elongation at fracture of 24 %). Four plate-screw combinations were implanted onto the pelvic bones of six sheep without osteotomy for 8 weeks. Subsequent analysis utilized histology, micro-computed tomography, and light and electron microscopy. All implants exhibited signs of degradation and hydrogen-gas evolution, with PEO-coated X0 implants demonstrating the least volume loss and the most substantial new-bone formation on the implant surface and surrounding cancellous bone. Furthermore, the osteoconductive properties of the X0 implants, when uncoated, exceeded those of the uncoated WE43 implants, as evidenced by greater new-bone formation on the surface. This osteoconductivity was amplified with PEO surface modification, which mitigated gas evolution and enhanced osseointegration, encouraging bone apposition in the cancellous bone vicinity. These findings thus indicate that PEO-coated X0 implants hold substantial promise as a biocompatible and absorbable implant material.

Biodegradable magnesium is a highly desired material for fracture fixation implants because of its good mechanical properties and ability to completely dissolve in the body over time, eliminating the need for a secondary surgery to remove the implant. Despite extensive research on these materials, there remains a dearth of information regarding critical factors that affect implant performance in clinical applications, such as the in vivo pH and mechanical loading conditions. We developed a measurement system with implantable strain, temperature, pH and motion sensors to characterize magnesium and titanium plates, fixating bilateral zygomatic arch osteotomies in three Swiss alpine sheep for eight weeks. pH 1–2 mm above titanium plates was 6.6 ± 0.4, while for magnesium plates it was slightly elevated to 7.4 ± 0.8. Strains on magnesium plates were higher than on titanium plates, possibly due to the lower Young's modulus of magnesium. One magnesium plate experienced excessive loading, which led to plate failure within 31 h. This is, to our knowledge, the first in vivo strain, temperature, and pH data recorded for magnesium implants used for fracture fixation. These results provide insight into magnesium degradation and its influence on the in vivo environment, and may help to improve material and implant design for future clinical applications.

The article analyzes the activities of innovative development institutions in Russia. The key problems that hinder innovation processes and the development of innovation infrastructure are identified. Key performance indicators of development institutions are proposed, as well as a set of measures that ensure the effectiveness of their functioning in modern Russian.

Theoretical and practical significance of the work done consists in the formation and presentation to the scientific and business community of promising conceptual directions of sustainable development of economic entities through the development of adaptive strategy; deployment of industry clusters; introduction and commercialization of innovative technologies; differentiated application of elements of scientific management; formation and development of social capital, taking into account the requirement of protection from cyber threats in the era of industries 4.0 and 5.0.

Skeletal muscle (SM) is essential for movement, stability, and overall body function, and it readily adapts to changes in energy demand. Myogenesis is energy-intensive and involves complex molecular and cellular events. We recently demonstrated that the absence of lysosomal acid lipase (LAL) in vivo significantly impacts the SM phenotype, primarily by disrupting energy homeostasis and reducing ATP production. As systemic LAL deficiency affects multiple organs, we hypothesized that the altered SM phenotype resulted from systemic rather than SM-specific loss of LAL activity.To distinguish between systemic and cell-intrinsic effects, we used primary myoblasts isolated from Lal-deficient (-/-) mice as well as C2C12 cells treated with the pharmacological inhibitor of LAL, Lalistat-2. We found a significant accumulation of cholesteryl esters in both models studied, highlighting the central role of LAL in lipid catabolism in the SM. However, lipid accumulation was absent under lipoprotein-deficient culture conditions. Neither genetic loss nor pharmacological inhibition of LAL affected myofiber formation or mitochondrial function in vitro, in contrast to what we observed in SM isolated from Lal-/- mice. Tracing [13C6]-labeled glucose in both cell culture models revealed only minor changes in tricarboxylic acid cycle metabolites. These results suggest that although LAL plays an essential role in lipid metabolism, its impact on the processes involved in muscle differentiation and cellular energy production is minor. We conclude that the cell-intrinsic effects of Lal-/- SM are unlikely to drive the SM phenotype observed in vivo.

Background and Aims: Recent studies showed that patients suffering from lysosomal acid lipase deficiency (LAL-D) benefit from enzyme replacement therapy; however, liver histopathology improved in some but not all patients. We hypothesized that the pan-peroxisome proliferator-activated receptor agonist lanifibranor may have beneficial effects on liver inflammation in LAL knockout (Lal−/−) mice based on its promising results in alleviating liver inflammation in patients with metabolic dysfunction-associated steatohepatitis. Methods: Female Lal−/− mice were daily gavaged with lanifibranor or vehicle for 21 days. The effects of the treatment were assessed by measuring body and organ weights, plasma lipids and lipoproteins, as well as hematological parameters, followed by liver proteomics and metabolomics. Results: Lanifibranor treatment slightly altered organ weights without affecting the total body weight of Lal−/− mice. We observed major changes in the proteome, with multiple proteins related to lipid metabolism, peroxisomal, and mitochondrial activities being upregulated and inflammation-related proteins being downregulated in the livers of treated mice. Hepatic lipid levels and histology remained unaltered, whereas plasma triacylglycerol and total cholesterol levels were decreased and the lipoprotein profile of lanifibranor-treated Lal−/− mice improved. Conclusion: Lanifibranor treatment positively affected liver inflammation and dyslipidemia in Lal−/− mice. These findings suggest the necessity of a further combined study of lanifibranor with enzyme replacement therapy in Lal−/− mice to improve the phenotype. Moreover, there is a compelling rationale for conducting clinical trials to assess the efficacy of lanifibranor as a potential treatment option for LAL-D in humans.

Abstract Background Matrix metalloproteinase 12 (MMP12) is a macrophage-secreted protein that is massively upregulated as a pro-inflammatory factor in metabolic and vascular tissues of mice and humans suffering from cardiometabolic diseases (CMDs). However, the molecular mechanisms explaining the contributions of MMP12 to CMDs are still unclear. Methods We investigated the impact of MMP12 deficiency on CMDs in a mouse model that mimics human disease by simultaneously developing adipose tissue inflammation, insulin resistance, and atherosclerosis. To this end, we generated and characterized low-density lipoprotein receptor (Ldlr)/Mmp12-double knockout (DKO) mice fed a high-fat sucrose- and cholesterol-enriched diet for 16–20 weeks. Results DKO mice showed lower cholesterol and plasma glucose concentrations and improved insulin sensitivity compared with LdlrKO mice. Untargeted proteomic analyses of epididymal white adipose tissue revealed that inflammation- and fibrosis-related pathways were downregulated in DKO mice. In addition, genetic deletion of MMP12 led to alterations in immune cell composition and a reduction in plasma monocyte chemoattractant protein-1 in peripheral blood which indicated decreased low-grade systemic inflammation. Aortic en face analyses and staining of aortic valve sections demonstrated reduced atherosclerotic plaque size and collagen content, which was paralleled by an improved relaxation pattern and endothelial function of the aortic rings and more elastic aortic sections in DKO compared to LdlrKO mice. Shotgun proteomics revealed upregulation of anti-inflammatory and atheroprotective markers in the aortas of DKO mice, further supporting our data. In humans, MMP12 serum concentrations were only weakly associated with clinical and laboratory indicators of CMDs. Conclusion We conclude that the genetic deletion of MMP12 ameliorates obesity-induced low-grade inflammation, white adipose tissue dysfunction, biomechanical properties of the aorta, and the development of atherosclerosis. Therefore, therapeutic strategies targeting MMP12 may represent a promising approach to combat CMDs.

The review focuses on the means of personal protection of people from blood-sucking arthropods, mainly from Ixodidae ticks. Individual (personal) protection of humans is an important component of nonspecific prevention of natural-focal vector-borne diseases. The paper considers statistical data describing the level of significance and relevance of the information about the infections transmitted by ticks, for instance, tick-borne viral encephalitis, tick-borne borreliosis etc. Presented are the characteristics of different groups of personal protection means: insectoacaricidal, repellent, and insectoacaricidal-repellent ones. Indicators of their effectiveness are assessed; their practical use, as well as the features of special protective clothing and the necessity of its application is described. The toxicity of protective clothing is reduced through a lining fabric, local inserts of fabric with insectoacaricidal impregnation and the use of underwear. Different interpretation of the term “repellent” in the domestic and foreign literature, and also two different approaches (“Eastern” and “Western”) to the design and manufacture of personal protection means for people against attacks of blood-sucking arthropods, including Ixodidae ticks, in Russia and Western Europe countries and the USA is discussed. The paper highlights the effect of different pyrethroid groups on the behavioral reactions of ticks. It is shown that in the Russian Federation the use of permethrin is prohibited for the treatment of protective clothing against blood-sucking arthropods, in particular against the taiga tick Ixodes persulcatus, which is the main vector of dangerous infections across the major part of the country (in the east of the European part, the Urals, Siberia and the Far East).

The article deals with the case of intestinal obstruction due to the formation of a giant fecal stone in the sigmoid colon. Coprolites occur in individuals suffering from severe chronic constipation. In most cases, fecal stones can be removed with the help of conservative measures, however, in this case, there were absolute indications for urgent surgery for acute intestinal obstruction.

Frequent development SARS-CoV-2 in patients with a surgical profile of a planned and emergency order indicate the need to study the features of the course of the postoperative period and histological examination of autopsy material of lung tissues in patients with coronavirus infection in these individuals.

The urgency of the problem stated in the article stems from the fact that in modern conditions the study of the psycho-diagnostic component of the teacher's activities is relevant in practical terms, since the functions of these activities affect the efficiency of pedagogical activity and the educational process itself, including the effectiveness of students' learning activities. The purpose of the article is to examine the role of teacher's psycho-diagnostic activity in the school educational system. The leading method of studying the problem is an empirical method of research (surveys, testing, content analysis), methods of mathematical and statistical processing, allowing a complex consideration of teachers' diagnostic activity characteristics. The study involves 100 teachers and 420 secondary school students, 14-15 years of age. The article reveals that the psycho-diagnostics, carried out by teachers, is characterized by haphazardness, and does not rely on the psychological and pedagogical technologies. The teachers carry out activity diagnostics more successfully, identifying its goals and methods. Psycho-pedagogical technologies of forming an ability to carry out personal diagnostics improve teachers' overall psychological culture. The study deepens the understanding of the psycho-diagnostic component, contributes to the development of the teacher's professional activity, expands teacher's psycho-diagnostic opportunities, and determines the levels of the teaching staff's psychological culture development. The practical significance of the study results is in their direct use for improving teachers' psychological training and other kinds of professional training in the "man-to-man" sphere.

Lysosomal acid lipase (LAL) is the only known enzyme that degrades cholesteryl esters and triglycerides at an acidic pH. In LAL deficiency (LAL-D), dysregulated expression of matrix metalloproteinase 12 (MMP-12) has been described. The overexpression of MMP-12 in myeloid lineage cells causes an immune cell dysfunction resembling that of Lal knockout (Lal KO) mice. Both models develop progressive lymphocyte dysfunction and expansion of myeloid-derived suppressor (CD11b+ Gr-1+) cells. To study whether MMP-12 might be a detrimental contributor to the pathology of LAL-D, we have generated Lal/Mmp12 double knockout (DKO) mice. The phenotype of Lal/Mmp12 DKO mice closely resembled that of Lal KO mice, while the weight and morphology of the thymus were improved in Lal/Mmp12 DKO mice. Cytological examination of blood smears showed a mildly reversed lymphoid-to-myeloid shift in DKO mice. Despite significant decreases in CD11b+ Ly6G+ cells in the peripheral blood, bone marrow, and spleen of Lal/Mmp12 DKO mice, the hematopoietic bone marrow progenitor compartment and markers for neutrophil chemotaxis were unchanged. Since the overall severity of LAL-D remains unaffected by the deletion of Mmp12, we conclude that MMP-12 does not represent a viable target for treating the inflammatory pathology in LAL-D. Magnesium‐based alloys are excellent materials for temporary medical implants, but understanding and controlling their corrosion performance is crucial. Most nanoscale corrosion studies focus on the surface, providing only 2D information. In contrast, macro‐ and microscale X‐ray tomography offers representative volume information, which is, however, comparatively low in resolution and rather qualitative. Here a new mesoscale approach overcomes these drawbacks and bridges the scale gap by combining 3D measurements using ptychographic X‐ray computed tomography (PXCT) with electron microscopy. This combination allows to observe the corrosion progress non‐destructively in 3D and provides high‐resolution chemical information on the corrosion products. A medical Mg–Zn–Ca alloy is used and compared the same sample in the pristine and corroded states. With PXCT an isotropic resolution of 85 and 123 nm is achieved for the pristine and corroded states respectively, which enables to distinguish nanoscale Mg2Ca precipitates from the matrix. The corroded state in best approximation to the in situ conditions is imaged and reveals the complexity of corrosion products. The results illustrate that the corrosion‐layer is dense and defect‐free, and the corrosion of the material is grain‐orientation sensitive. Promoting financial stability is one of the main priorities for the governments of countries seeking to achieve sustainable economic growth. The article aims to assess and model the impact of macroeconomic factors on the financial stability of the Russian Federation in the period 2010-2030 using ADF, OLS, VAR, ARCH, VECM and other techniques. In addition, the linear causal relationship between a group of 6 macroeconomic indicators and the financial stability of the Russian Federation was studied. The results of this research show that Russia's financial stability depends mainly on exports of crude oil and natural gas, price stability, volume of government debt, deficits and surpluses of the state's budget, exchange rate stabilization of national currency, and effectiveness of the banking system. Additionally, events taking place in Eastern Europe, the Middle East and the African continent may negatively affect Russia's financial security if it fails to take the necessary preventive measures. Bone is a dynamic tissue composed of cells, an extracellular matrix, and mineralized portion. Osteoblasts are responsible for proper bone formation and remodeling, and function. These processes are endergonic and require cellular energy in the form of adenosine triphosphate (ATP), which is derived from various sources such as glucose, fatty acids, and amino acids. However, other lipids such as cholesterol have also been found to play a critical role in bone homeostasis and can also contribute to the overall bioenergetic capacity of osteoblasts. In addition, several epidemiological studies have found a link between elevated cholesterol, cardiovascular disease, an enhanced risk of osteoporosis, and increased bone metastasis in cancer patients. This review focuses on how cholesterol, its derivatives, and cholesterol-lowering medications (statins) regulate osteoblast function and bone formation. It also highlights the molecular mechanisms underlying the cholesterol–osteoblast crosstalk.

Herein, we report for the first time a study dedicated to acidic gases’ solubility in ionic liquids with sterically hindered bulky anion, namely bis(2-ethylhexyl) sulfosuccinate ([doc]), experimentally evaluated at low pressures. The effect of cation change (imidazolium, pyridinium, and pyrrolidinium) on the thermophysical properties and sorption capacities was also discussed. The densities and the activation energies of the tested ILs exhibited minor differences. Furthermore, the COSMO-RS model was used to predict the free volumes of ILs aiming to investigate its influence on gas solubilities. The conducted calculations have revealed an antibate correlation between the fractional free volume (FFV) and Henry’s law constant. In particular, the lowest FFV in 1-methylimidazolium [doc] corresponded to the minimal sorption and vice versa. In addition, it was shown that the presence of protic cation results in a significant reduction in CO2 and H2S solubilities. In general, the solubility measurement results of the synthesized ILs have shown their superiority compared to fluorinated ILs based on the physical absorption mechanism.
