As a focus emerging within the framework of new media, online social networks have taken an active role in the social processes of globalization and have spread communication worldwide. In this regard, it is important to examine the presence of agencies in online social networks as global news providers and to reveal the structural characteristics of online news distribution. This study is about examining the structure of international communication in new media in the context of state and media relations. In the conditions created by the development of the new media, the main preoccupation of the research is to examine the structures of the international news agents, who have had the chance to directly contact the public. The ways of presenting the interventions to the political crisis in Syria on the Twitter accounts of CNN and Sputnik will be examined with the critical discourse analysis method developed by Teun Van Dijk, and the findings will be compared with the intervention arguments developed by the official authorities of the USA and Russia, the countries where the sample news agencies have their headquarters. In this context, it has been determined that the meanings and general discourse in the texts presented by the news agencies are in harmony with the policies of the countries in which they have their headquarters, and that the texts used in the new media are created to protect the state interests structured in economic capital relations., Yeni medya çerçevesinde ortaya çıkan bir odak olarak çevrimiçi sosyal ağlar küreselleşmenin sosyal süreçlerinde etkin bir rol üstlenmiş, iletişimi dünya çapında yaygınlaştırmıştır. Küresel haber sağlayıcılar olarak ajansların çevrimiçi sosyal ağlardaki mevcudiyetlerinin incelenmesi ve çevrimiçi haber dağıtımının yapısal özelliklerinin ortaya çıkarılması bu anlamda önem taşımaktadır. Bu çalışma, devlet ve medya ilişkisi bağlamında uluslararası iletişimin yeni medyadaki yapısının incelenmesini konu almaktadır. Yeni medyanın gelişiminin yarattığı koşullarda kamuoyuyla doğrudan bağlantı kurma imkânı yakalayan uluslararası haber ajanlarının söz konusu mecradaki yapılarının ele alınması araştırmanın esas uğraşıdır. Bu çerçevede, CNN ve Sputnik’in Twitter hesaplarında Suriye’de yaşanan politik krize müdahaleleri sunum biçimleri Teun Van Dijk tarafından geliştirilen eleştirel söylem analizi yöntemi ile incelenecek, elde edilen bulgular örnek haber ajanslarının merkezlerini bulundurdukları ülkeler olan ABD ve Rusya’nın resmi makamlarınca geliştirilen müdahale argümanlarıyla karşılaştırılacaktır. Temel sorunsalımız ise medya ve devlet ilişkisinin yeni medyada ve çevrimiçi sosyal ağlardaki yapısının ele alınması ve bulgular üzerinden bu kaynaşmanın boyutlarının incelenmesidir.

İnsan bedenine karşı yapılan müdahaleleler kişinin rızası dâhilinde hukuka uygunluk sebebi olarak yapılabilmekte olup, bu müdahaleler beden, uzuv, organ ve hatta medikal boyuttan günümüzde genom boyutuna indirgenmeye başlamış ve hastalıkların temelinde en önemli neden olarak genetik bilginin suçlanmaya başlaması ile giderek artan hızla genetik koda yapılabilecek genom düzenleme olarak adlandırılabilecek tıbbi müdahaleler geliştirilmeye başlanmıştır. Müdahalelerin kalıtım materyalinde yapılacak olması da birçok yeni soruyu ve sorunu beraberinde getirmekte ve etik kaygıların yanı sıra gerek somatik hücrelerde gerekse yapılan müdahalenin sonuçlarının nesilden nesile aktarılabileceği eşey hücrelerinde denenmesi ve araştırmaların da artık klinik düzeylere geçmeye başlaması konunun kanunilik bağlamında da hukukta da yer alması gerekliliğini doğurmaktadır. Geliştirilen genom düzenleme teknolojilerinin en sonuncusu olarak CRISPR/cas9 sistemi de önceki yöntemlere göre daha kolay ve ucuz olarak uygulanabilir olması nedeniyle genom düzenlemede uygulamaya hız kazandırmış ve gerek insan genomu gerekse insan dışı canlılarda ve bitkilerde bu konuda hukuki önlem ile düzenlemelerin de ivedilikle yapılması gerekliliğini ortaya koymaktadır. Bu bağlamda uluslararası hukukta var olan düzenlemeler ile kendi hukukumuzda var olan düzenlemeler bu tez kapsamında değerlendirilmiş ve kendi genom kanunumuz için öneriler sunulmuştur. The interventions made against the human body can be made by having consent in compliance with the law as the legimate reason and those interventions today have begun to be reduced from the body, limb, organ and even the medical dimensions to the aspect of the genome. Moreover, since the genetic information has started to be attributed as the most important reason on the basis of diseases, therapeutic interventions targetting the genetic code, known as the genome editing, have begun to be developed. The fact that the interventions are carried out in hereditary material brings many new questions and problems with them as well, and besides ethical concerns, experimenting genome editing in either somatic cells or germ cells where the results of the interventions made in can be transferred to the next generations and further initiation of translation of experiments to the clinical trials has emerged the necessity of legal inferences in the context of law. Since the CRISPR/cas9 system could be applied more easily and cheaply than the previous methods, the genome regulation technology has accelerated the applications in genome editing and necessitates urgent legal regulations either on human genome or non-human living organisms and plants. In this regard, the arrangements existing in international law and those existing in our own law have been assessed within the thesis and suggestions for our own genomic law on behalf of genome editing have been presented. 129

Tarihsel süreç içinde üyeleri Kuzey Asya'dan Anadolu'ya, Kafkasya'dan Avrupa'ya kadar genişleyen bir coğrafyaya dağılmış, köken olarak Kuzey Mezopotamya'ya ait bir din olan Ezidilik hakkında Türkiye'de çok az akademik çalışma yapılmıştır. Bu nedenle, söz konusu bu toplumun kamuoyu tarafından zaten çok sınırlı olan bilinirliğinin kaynağı, çağlar boyunca oluşturulan, çarpıtılmış bilgilerle inşa edilmiş tabulara dayanmaktadır. Kitle iletişim araçları da bu kültür hakkında neredeyse hiç bilgi yada içerik sunmamaktadır. 3 Ağustos 2014 tarihinde IŞİD'in Sincar Bölgesi'ne yönelik saldırıları, Ezidilerin bölgeden kitlesel göçüne sebep olmuş, ''haber değeri'' yüksek bir fenomen olarak görülen bu göç, uluslararası düzeyde bir medya içeriğine dönüştükten sonra Türk ana akım medyasında da çok sınırlı biçimde haberlere konu edilmeye başlanmıştır. Ezidi toplumu, yaşanan büyük göçün sınırlı da olsa medya içeriği haline dönüşmesi sonucu Türkiye kamuoyu tarafından tanınmaya başlanmıştır. Yapılan gözlemler sonucu, alternatif bir medya organı olan İMC TV'nin Ezidi göçüne oldukça yüksek oranda yer verdiği belirlenmiştir. Bu çalışma kapsamında, göç öncesi ve sonrası, Ezidilerin medya içeriğinde nasıl ve ne sıklıkta yer aldığını belirleyebilmek adına, 3 Ağustos 2014 tarihi merkez kabul edilerek, bu tarihin iki ay öncesi ve iki ay sonrası olmak üzere, toplam 4 aylık sürede Ezidiler hakkında İMC TV ana haber bültenlerinde yayınlanan haberler taranmış, elde edilen veriler hem nicel olarak, hem de Teun van Dijk'in eleştirel söylem çözümlemesi çerçevesinde nitel olarak analiz edilmiştir., There has been a very little number of academic studies conducted on Ezidies throughout the history, whose members spread over the geography extending from Northern Asia to Anatolia, from Caucasia to Europe, and which religion found its origin in Northern Mesopotamia. For that reason, the already-scarce sources of there cognition of such society within the public view based on taboos produced down the ages by twisted information. Likewise, mass media does provide nearly no information or content. ISIS attacks on 3 August 2014 targeting "Sincar" territory led mass migration of Ezidi people, such migration being recognized as a newsworthy phenomenon after turning into media content in international level, attracted some limited attention from Turkish main stream media in their news. Ezidi society started being recognized by Turkish public as a result of news on major immigration in Turkish media, even the news presented in such a limited manner. Observations show that IMC TV, which is an alternative media organ, attributed significant importance to the great immigration of Ezidi people. In the context of current study with an aim to understand how and how often Ezidi people took place in media before and after the migration, the date of 3 August 2014 is taken as a reference point, and the period of consecutive 4 months preceding and subsequent to the reference point constitutes our time period in which we had examined news on main news bulletin of IMC TV, then acquired data was analyzed quantitively and according to Teun van Dijk's critical discourse analysis qualitatively.

ABS (Ankaferd Hemostat) has emerged as a novel haemostatic agent of plant origin. Its ability to interact with entire proteins including cell surface receptors and/or blood proteins plus tissue proteins revealed novel functions of ABS to biologic systems especially tumor associated transcription factors. Thus, in our study, we aimed to investigate the effects of ABS on lymphoid neoplastic cells in vitro. We showed that Ankaferd treated B-CLL cells (at doses 0.5, 1 and 2 mu g/mL) ceased to inflate and more than 50% of tumor cells were died compared to 0.1 and 0.25 mu g/mL doses. Additionally, transformation of B-CLL cells to the blastic aggressive lymphoid forms was prevented by addition of Ankaferd to culture medium. ÖZET Lösemik Hematopoezde Anjiotensin Dönüştürücü Enzim (ÂCE) İnsersiyon/Delesyon (I/D) Gen Polimorfizmi Renin-angiotensin sistemi (RAS), yakın döneme kadar sadece kan basıncı ve sıvı-elektrolit dengesini düzenleyen endokrin bir sistem olarak bilinirdi. Günümüzde, RAS'ın spesifik organ sistemlerinde de lokal olarak bulunduğu ve otokrin/parakrin ve hatta mtrakrin işlevlere sahip olduğu belirlenmiştir. Lokal kemik iliği RAS'ı da normal ve neoplastik hematopoeizi etkileyen otokrin/parakrin bir sistem olarak tanıınlanmıştir. Angiotensin dönüştürücü enzim (ACE) etkisiyle oluşan Angiotensin II, Angiotensin II tip 1 reseptörleri aracılığıyla hematopoetik kök hücrelerinde çoğalmayı ve farklılaşmayı uyarmaktadır. Akut lösemili hastaların kemik iliklerinde RAS komponentlerinden olan ACE mRNA düzeyinin artmış olarak bulunması, lokal kemik iliği RAS'ının normal hematopoezin yanısıra neoplastik hematopoezi de uyardığım düşündürmektedir. Kardiyak hastalıklarda yapılan çalışmalarda özellikle ACE İnsersiyon/Delesyon (I/D) gen polimorfizmi hastalıkların prognozu ile ilişkili bulunmuştur. Ayrıca çeşitli neoplastik hastalıklarda da bu genin polimorfizmleri malignansi riskini arttırmıştır. Ancak, lösemili hastalarda ilgili gen polimorfiznılerinin durumu ve bu polimorfizmlerin hastalığın prognozu üzerindeki etkileri bilinmemektedir. Bu çalışmada, literatürde ilk kez ACE I/D geni polimorfiznılerinin neoplastik hematopoez üzerindeki etkileri araştırılmıştır. Çalışmada, Hacettepe Üniversitesi Tıp Fakültesi Hematoloji Ünitesinde tedavi gören AML, ALL, KML, KLL, MM ve MDS tanısı ile takip edilen 46 (yaş için median: 49; 17-81 yaş arasında) lösemili hastaların rutin tanı ve takipleri sırasında alman kemik iliği örnekleri ile 59 (yaş için median: 30; 17-78 yaş arasında) sağlıklı bireyden alınan periferik kan örnekleri çahşılrnıştır. ACE I/D gen polimorfizmi polimeraz zincir reaksiyonu yöntemiyle ilgili primerler kullanılarak Ankara Üniversitesi Tıp Fakültesi Tıbbi Genetik Anabilim Dalı Moleküler Laboratuvannda araştinlmıştir. Hastalarin % 80.4 ünde (n=37) JD/îl genotipi bulunurken bu oran kontrol grubunda % 55.9 (n=33) dur. Insersiyon alleli bulunan (ID/II) bireylerde hastalık riski 3.2 kat artmıştır (Ki Kare Testi, p=0,008; OR:3.2 [C.I. % 95, 1,3-7,9]). Ayrıca, AML, KML, ALL ve KLL lösemi hastalan kontrol grubuna göre karşılaştuıldığmda insersiyon aUelinin varlığında lösemi riski 6.3 kat artmıştır (p=0,001 ve [O.R: 6.3 ; C.I.: % 95; 1,98-20,1]). Ek olarak, 50 yaş ve altı grupta hastaların % 83,3 'ünde (n=20) insersiyon alleli bulunurken bu oran kontrollerde % 54,5 (n=30) olarak saptanmıştır. Bu fark istatistiksel olarak anlamlı bulunmuş ve 4,2 kat artmış lösemi riski ile ilişkili bulunmuştur (p=0,015) [OR:4,2 C.I: %95 1,26- 13,8)]. Bu çalışmada, lokal kemik iliği RAS komponentlerinin gen polimorfiznılerinin lösemik hücre transformasyonu ve proliferasyonu üzerindeki olası elMlerinin ilk delilleri gösterilmiştir. Bu sayede, kemik iliği lokal RAS'ın 50lösemik hastalardaki ekspresyonunun polimorfîk genler nedeniyle arttığının yapılacak ileri çalışmalarla kesin olarak gösterilmesi, lösemi tedavisinde, lokal RAS'ı ve aktive ettiği birçok sinyal iletim yolağını hedef labilecek, yeni tedavi yöntemlerinin gelişmesi için bilimsel zemin teşkil edebilecektir. Anahtar Sözcükler: ACE, ACE I/D gen polimorfizmi, Angiotensin II, lokal renin- angiotensin sistemi, lösemik hematopoez 51 SUMMARY Angiotensin Converting Enzyme (ACE) Insersion/Deletiom (I/D) Gene Polymorphisms in Leukemic Hematopoiesis The Renin-angiotensin system (RAS) has until recently been considered as just an endocrine system that controls the blood pressure and fluid-electrolyte balance. Today, it is known that, beside the circulatory system, RAS is present in specific tissue systems as localized manner having autocrine /paracrine and intractrine functions. Local bone marrow renin-angiotensin system has also defined as an autocrine-paracrine system affecting normal and neoplastic hematopoiesis. Angiotensin II produced by the affect of angiotensin-converting enzyme (ACE), stimulates proliferation and differentiation of hematopoietic stem cells through Angiotensin II type 1 receptors. The finding of increased ACE mRNA level at acute leukemic patients' bone marrow suggested that local bone marrow RAS stimulates neoplastic hematopoiesis in order with normal one. Studies made on cardiac diseases show that especially ACE I/D gene polymorphisms have found to be related with the prognosis of the diseases. Additionally, it's found that gene polymorphisms increased the risk of malignancy in several neoplastic diseases. However, the subset of those gene polymorphisms at leukemic patients and the relationships of them with the prognosis of the disease are not known. In this study, ACE I/D gene polymorphisms and their effects on neoplastic hematopoiesis has been studied for the first time up to the literature. In this study, bone marrow samples of 46 patients (median age: 49; aged 17- 81) diagnosed as AML, ALL, CML, CLL, MM and MDS, taken during their routine investment and diagnosis at Hacettepe University Faculty of Medicine, Hematology Unit, and peripheral blood samples of 59 healthy people (median age: 30; aged 17-78) were studied. ACE I/D gene polymorphism has been investigated using related primers within polymerase chain reaction in the Ankara University Faculty of Medicine, Department of Medical Genetics, Molecular Laboratory. 80.4 % (n=37) of the patients were representing ID/H genotype, where as it was 55.9 % (n=33) in control group. Disease risk has been increased 3.2 fold in insersion allele (Tu/Ü) carrier patients (Chi Square Test, p=0,008; OR: 3.2 [CI. % 95, 1,3-7,9]). Additionally, in the presence of insersion allele leukemia risk has increased as 6.3 times in AML, CML, ALL and CLL patients compared to control group (p=0,001 and [O.R: 6.3 ; C.I.: % 95; 1,98-20,1]). Moreover, in the group of the patients aged 50 or less, 83,3 % (n=20) represented insersion allele, however, it was 54,5 % (n=30) in control group. This discrepancy was statistically significant and has found to be related with 4.2 times increased leukemia risk (p=0,015) [OR: 4,2 CI: %95 1,26-13,8)]. In this study, first evidences of possible affects of gene polymorphisms of the local bone marrow RAS components on leukemic cell transformation and proliferation has been demonstrated. Thus, further studies mat would reveal the increased expression of local bone marrow RAS under the effect of polymorphic genes at leukemic patients will lead scientific background to the investigation of 52novel pharmacologic and/or 'anti-sense' therapeutic applications for leukemia targeting local RAS and its activated signal transition pathways.
