Background and Aim: Stainless steel crown (SSC) is the most durable and reliable restoration for primary teeth with extensive caries but its metalic appearance has always been a matter of concern. With advances in restorative materials and metal bonding processes, composite veneer has enhanced esthetics of these crowns in clinic. The aim of this study was to evaluate the shear bond strength of SSC to composite resin using different surface treatments and adhesives. Materials and Methods: In this experimental study, 90 stainless steel crowns were selected. They were mounted in molds and divided into 3 groups of 30 each (S, E and F). In group S (sandblast), buccal surfaces were sandblasted for 5 seconds. In group E (etch) acidic gel was applied for 5 minutes and in group F (fissure bur) surface roughness was created by fissure diamond bur. Each group was divided into 3 subgroups (SB, AB, P) based on different adhesives: Single Bond, All Bond2 and Panavia F. Composite was then bonded to specimens. Cases were incubated in 100% humidity at 37°C for 24 hours. Shear bond strength was measured by Zwick machine with crosshead speed of 0.5 mm/min. Data were analyzed by ANOVA test with p0.05) so the two variables were studied separately. No significant difference was observed in mean shear bond strength of composite among the three kinds of adhesives (P>0.05). Similar results were obtained regarding surface treatments (P>0.05). Conclusion: Based on the results of this study, treating the SSC surface with bur and using single bond adhesive and composite can be used successfully to obtain esthetic results in pediatric restorative treatments.

Statement of Problem: In recent years, the prevalence of temporomandibular disorders among children and adolescents has increased, however, no study on the correlation between occlusal relations and temporomandibular disorders, in Mashhad adolescents, has been conducted yet. Purpose: The aim of this study was to survey the relationship between occlusal indices and temporomandibular disorders (TMD) among Mashhad adolescents, with the age range of 11-14 years old. Methods and Material: In this descriptive cross-sectional study, 1066 students (533 males and 533 females), age ranging from 11-14 years old (12.620.96 years), from seven Mashhad educational regions, were selected. Temporomandibular joints were thoroughly examined for all subjects. All occlusal indices including dentition system, angles classification, types of malooclusion such as cross-bite, deep-bite as well as types of lateral occlusion, premature contacts in lateral and protrusive movements were investigated. Data were analyzed statistically using, Chi-Square and logestic regression tests. Results: The prevalence of TMD was 23.5%. There was no significant relation between TMD and type of dentition, angles classification and type of occlusion in lateral movements, however, statistically significant relation was found between TMD and deep over bite (P

The immune system is known to critically regulate the central nervous system during development, homeostasis, as well as during injury and repair. More recently, much work has been undertaken to characterize the role of immune cells in the cochlea. Macrophages are the resident innate immune cells of the cochlea. Several studies on mice have reported that mac-rophages mediate drug uptake via the stria vascularis, promote synaptic repair after noise exposure, and participate in tissue remodeling after cochlear implantation. In other organs macrophages can be heterogenous. Whether there are sub-populations of macrophages in the cochlea and the function of such subgroups are unknown. Here we report molecular heterogeneity among cochlear macrophages which include a population resembling microglia, the primary innate immune cells of brain and two molecularly distinct subpopu-lations of macrophages resembling two recently identified macrophage population in peripheral nerve. To begin to ascertain the function of one of these populations we examined FIRE mice (Csf1rΔFIRE/ΔFIRE), a mouse model that is entirely microglia- deficient but retains other macrophages which lacks embryonic macrophages and microglia We examined cochlear histology at postnatal days 5, 14, 6 weeks and 6 months of age. Surprisingly, FIRE mice had lower auditory brainstem response thresholds and higher wave I amplitudes when compared to aged-matched control mice at both 6 weeks and 6 months, suggesting less aged-related hearing loss. However, the FIRE mice number,organization of inner and outer hair cells and inner hair cell synapse were similar to aged-matched control C57/Bl6 mice. Taken together, these findings suggest that microglia are dispensable for cochlear development but may play a role in age-related hearing loss, a role which requires further investigation. Statement of Problem: Congenital missing tooth is the condition of having genetically one or more missing teeth which cannot be observed clinically or in radiographic images. This is one of the most prevalent anomalies in dental development that occurs either individually or as symptoms of a syndrome. Moreover, for permanent teeth, it is common with a reported prevalence of between 1.6-9.6%. Purpose: The aim of this study was to assess the prevalence rate and the pattern of congenital missing teeth in adolescents referring to Mashhad School of Dentistry. Materials and Method: In this cross-sectional study, a total of 600 panoramic radiographs related to the subjects aged 9-14 years (351 girls and 249 boys) were analyzed. The data were recorded in the related forms, and then analyzed using Chi-square and Exact tests. Results: Among 600 panoramic radiographs, 94 teeth were found to be missing. The most and the least frequent missing teeth were the mandibular second premolars and the maxillary central incisors (observed in only 1 subject), respectively. The most commonly absent teeth were the mandibular second premolars, the maxillary lateral incisors, the mandibular central incisors, and the maxillary second premolars in order of the frequency. Conclusion: This study revealed that the frequency of missing tooth in girls is higher than that in boys. Thus, due to the girls' more aesthetic sensibility and also intricate treatment of such anomaly, accurate and frequent examination of adolescents' particularly girls' teeth for on-time diagnosis is crucial. Context: Hypodontia is defined as the congenital absence of one or a few teeth, and is also the most common anomaly in dental development. This condition occurs either individually or as part of the symptoms of a syndrome, and it is more common in permanent teeth than in deciduous teeth, reporting a prevalence of between 1.6 and 9.6%. Aims: The objective of this study is to investigate the prevalence of hypodontia for permanent teeth in nine- to 14-year-old children who attended the Mashhad School of Dentistry in 2007. Setting and Design: We conducted this descriptive, analytical, cross-sectional study, to determine the mentioned aims. Materials and Methods: In this descriptive, cross-sectional study, panoramic radiographs belonging to 600 children (351 girls and 249 boys), aged nine to 14 years, were available for examination. All related findings were recorded in the respective forms. Statistical Analysis Used: The data were processed using Exact and Chi-square tests. Results: The prevalence of hypodontia in the girls was 9.2%, in the boys 8.8%, and in both sexes combined 9%. The most and the least frequent cases of absent teeth were the mandibular second premolars and the maxillary central incisor (only one child), respectively. The most commonly absent teeth were the mandibular second premolars, the maxillary lateral incisors, the mandibular central incisor, and the maxillary second premolars, in that order. Conclusions: This study showed a high frequency of hypodontia among the understudied population. Thus, due to the complicated treatment, accurate examination of children for on-time diagnosis of this developmental anomaly is crucial.

Introduction: Microglia, the principal immune cells of the Central Nervous System (CNS), are exquisitely sensitive to CNS injury and disease, fueling a reactive state called microgliosis. We have used parabiosis, a surgical procedure that allows the creation of peripheral blood chimeras without transplantation, to show that unlike most other tissue-resident macrophages that rely on circulating blood-borne precursors for their replacement, microglia are capable of self-renewal within the CNS. In contrast, certain inflammatory pathological conditions such as multiple sclerosis and its murine model, experimental autoimmune encephalomyelitis (EAE), are known to lead to the recruitment of inflammatory monocytes to the CNS. Is the entry of these cells, normally excluded from the CNS, a causal factor in disease progression Methods: To address this question we developed a new experimental model, based on parabiosis and differential bone marrow irradiation, which allows the precise distinction between peripheral blood- derived monocyte/macrophages and resident microglia, thus enabling us to investigate the kinetics of microglia activation, blood born monocytes entry in the CNS and their differentiation into macrophages during EAE progression. Results: Our data reveals a dynamic interplay between macrophages and microglia and strongly supports a causal link between myelomonocytic cell invasion and disease progression. In summary, our data identified the invasion of circulating monocytes into the CNS parenchyma as a major adverse event in EAE progression, supporting therapeutic strategies specifically aimed at inhibiting the migration of myelomonocytic cells rather than that of leukocytes in general. Background: The eye is a highly specialized sensory organ which encompasses the retina as a part of the central nervous system, but also non-neural compartments such as the transparent vitreous body ensuring stability of the eye globe and a clear optical axis. Hyalocytes are the tissue-resident macrophages of the vitreous body and are considered to play pivotal roles in health and diseases of the vitreoretinal interface, such as proliferative vitreoretinopathy or diabetic retinopathy. However, in contrast to other ocular macrophages, their embryonic origin as well as the extent to which these myeloid cells might be replenished by circulating monocytes remains elusive., Results: In this study, we combine transgenic reporter mice, embryonic and adult fate mapping approaches as well as parabiosis experiments with multicolor immunofluorescence labeling and confocal laser-scanning microscopy to comprehensively characterize the murine hyalocyte population throughout development and in adulthood. We found that murine hyalocytes express numerous well-known myeloid cell markers, but concomitantly display a distinct immunophenotype that sets them apart from retinal microglia. Embryonic pulse labeling revealed a yolk sac-derived origin of murine hyalocytes, whose precursors seed the developing eye prenatally. Finally, postnatal labeling and parabiosis established the longevity of hyalocytes which rely on Colony Stimulating Factor 1 Receptor (CSF1R) signaling for their maintenance, independent of blood-derived monocytes., Conclusion: Our study identifies hyalocytes as long-living progeny of the yolk sac hematopoiesis and highlights their role as integral members of the innate immune system of the eye. As a consequence of their longevity, immunosenescence processes may culminate in hyalocyte dysfunction, thereby contributing to the development of vitreoretinal diseases. Therefore, myeloid cell-targeted therapies that convey their effects through the modification of hyalocyte properties may represent an interesting approach to alleviate the burden imposed by diseases of the vitreoretinal interface., (© 2024. Cerebrospinal fluid (CSF) matrix biomarkers have become increasingly valuable surrogate markers of neuropsychiatric diseases in research and clinical practice. In contrast, CSF cells have been rarely investigated due to their relative scarcity and fragility, and lack of common collection and cryopreservation protocols, with limited exceptions for neurooncology and primary immune-based diseases like multiple sclerosis. the advent of a microfluidics-based multi-omics approach to studying individual cells has allowed for the study of cellular phenotyping, intracellular dynamics, and intercellular relationships that provide multidimensionality unable to be obtained through acellular fluid-phase analyses. challenges to cell-based research include site-to-site differences in handling, storage, and thawing methods, which can lead to inaccuracy and inter-assay variability. In the present study, we performed single-cell RNA sequencing (10x Genomics) on fresh or previously cryopreserved human CSF samples from three alternative cryopreservation methods: Fetal Bovine Serum with Dimethyl sulfoxide (FBS/DMSO), FBS/DMSO after a DNase step (a step often included in epigenetic studies), and cryopreservation using commercially available Recovery© media. In comparing relative differences between fresh and cryopreserved samples, we found little effect of the cryopreservation method on being able to resolve donor-linked cell type proportions, markers of cellular stress, and overall gene expression at the single-cell level, whereas donor-specific differences were readily discernable. We further demonstrate the compatibility of fresh and cryopreserved CSF immune cell sequencing using biologically relevant sexually dimorphic gene expression differences by donor. Our findings support the utility and interchangeability of FBS/DMSO and Recovery cryopreservation with fresh sample analysis, providing a methodological grounding that will enable researchers to further expand our understanding of the CSF immune cell contributions to neurological and psychiatric disease., (© 2024. Amyotrophic lateral sclerosis (ALS) is a fatal disease affecting upper and lower motor neurons. Microglia directly interact with motor neurons and participate in the progression of ALS. Single-cell mass cytometry (CyTOF) analysis revealed prominent expression of α5 integrin in microglia and macrophages in a superoxide dismutase-1 G93A mouse model of ALS (SOD1 G93A ). In postmortem tissues from ALS patients with various clinical ALS phenotypes and disease duration, α5 integrin is prominent in motor pathways of the central and peripheral nervous system and in perivascular zones associated with the blood-brain barrier. In SOD1 G93A mice, administration of a monoclonal antibody against α5 integrin increased survival compared to an isotype control and improved motor function on behavioral testing. Together, these findings in mice and in humans suggest that α5 integrin is a potential therapeutic target in ALS.

Microglial research has advanced considerably in recent decades yet has been constrained by a rolling series of dichotomies such as "resting versus activated" and "M1 versus M2." This dualistic classification of good or bad microglia is inconsistent with the wide repertoire of microglial states and functions in development, plasticity, aging, and diseases that were elucidated in recent years. New designations continuously arising in an attempt to describe the different microglial states, notably defined using transcriptomics and proteomics, may easily lead to a misleading, although unintentional, coupling of categories and functions. To address these issues, we assembled a group of multidisciplinary experts to discuss our current understanding of microglial states as a dynamic concept and the importance of addressing microglial function. Background: Dental trauma injuries are the second most common traumatic injuries among children and adolescents. An incorrect diagnosis or a delayed treatment of traumatic injuries may lead to irreparable damages. Thus, proper management provided by physicians and dentists is crucial for patients affected by these injuries.This study aimed to evaluate the impact of dentoalveolar trauma management training on improving physicians' and dentists' knowledge in different emergency wards in Mashhad, Iran. Methods: In this educational interventional study, a total number of 60 residents and interns selected from the following categories: maxillofacial surgery residents, emergency medicine residents, ear-nose-throat (ENT) residents, and medical interns were included, who were randomly selected. We gathered information using a valid and reliable questionnaire. Afterward, we provided the participants with booklets about dental traumas. By passing one month, the above-mentioned questionnaire was used to re-evaluate the target groups' knowledge. The results were statistically analyzed using Chi-square, One-way ANOVA and paired sample T-test with statistically significant differences defined for p < 0.05. Results: Prior to the training, the interns had the lowest knowledge, while the maxillofacial surgery residents had the highest level of knowledge in comparison to interns, ENT residents, and emergency medicine residents, respectively ( p < 0.001). After completing the course, the maxillofacial surgery residents once again represented the highest knowledge, followed by emergency medicine residents, ENT residents, and medical interns, respectively. A statistically significant difference was noticed in pre- and post-course knowledge evaluation. Conclusion: A comparison of the results before and after the training course demonstrates a significant lack of knowledge on dentoalveolar traumas among physicians and also reveals that providing this education is necessary because it significantly increases their knowledge in this regard., Competing Interests: The authors declare that they have no competing interests., (© 2022 Iran University of Medical Sciences.)

The innate immune landscape of the central nervous system (CNS), including the brain and the retina, consists of different myeloid cell populations with distinct tasks to fulfill. Whereas the CNS borders harbor extraparenchymal CNS-associated macrophages whose main duty is to build up a defense against invading pathogens and other damaging factors from the periphery, the resident immune cells of the CNS parenchyma and the retina, microglia, are highly dynamic cells with a plethora of functions during homeostasis and disease. Therefore, microglia are constantly sensing their environment and closely interacting with surrounding cells, which is in part mediated by soluble factors. One of these factors is Osteopontin (OPN), a multifunctional protein that is produced by different cell types in the CNS, including microglia, and is upregulated in neurodegenerative and neuroinflammatory conditions. In this review, we discuss the current literature about the interaction between microglia and OPN in homeostasis and several disease entities, including multiple sclerosis (MS), Alzheimer's and cerebrovascular diseases (AD, CVD), amyotrophic lateral sclerosis (ALS), age-related macular degeneration (AMD) and diabetic retinopathy (DR), in the context of the molecular pathways involved in OPN signaling shaping the function of microglia. As nearly all CNS diseases are characterized by pathological alterations in microglial cells, accompanied by the disturbance of the homeostatic microglia phenotype, the emergence of disease-associated microglia (DAM) states and their interplay with factors shaping the DAM-signature, such as OPN, is of great interest for therapeutical interventions in the future.

Neuroinflammation and neurodegeneration may represent two poles of brain pathology. Brain myeloid cells, particularly microglia, play key roles in these conditions. We employed single-cell mass cytometry (CyTOF) to compare myeloid cell populations in the experimental autoimmune encephalomyelitis (EAE) model of multiple sclerosis, the R6/2 model of Huntington's disease (HD) and the mutant superoxide dismutase 1 (mSOD1) model of amyotrophic lateral sclerosis (ALS). We identified three myeloid cell populations exclusive to the CNS and present in each disease model. Blood-derived monocytes comprised five populations and migrated to the brain in EAE, but not in HD and ALS models. Single-cell analysis resolved differences in signaling and cytokine production within similar myeloid populations in EAE compared to HD and ALS models. Moreover, these analyses highlighted α5 integrin on myeloid cells as a potential therapeutic target for neuroinflammation. Together, these findings illustrate how neuropathology may differ between inflammatory and degenerative brain disease.

Objective: Type I interferon (IFN) signaling is a central pathogenic pathway in systemic lupus erythematosus (SLE), and therapeutics targeting type I IFN signaling are in development. Multiple proteins with overlapping functions play a role in IFN signaling, but the signaling events downstream of receptor engagement are unclear. This study was undertaken to investigate the roles of the type I and type II IFN signaling components IFN-α/β/ω receptor 2 (IFNAR-2), IFN regulatory factor 9 (IRF-9), and STAT-1 in a mouse model of SLE., Methods: We used immunohistochemical staining and highly multiplexed assays to characterize pathologic changes in histology, autoantibody production, cytokine/chemokine profiles, and STAT phosphorylation in order to investigate the individual roles of IFNAR-2, IRF-9, and STAT-1 in MRL/lpr mice., Results: We found that STAT-1(-/-) mice, but not IRF-9(-/-) or IFNAR-2(-/-) mice, developed interstitial nephritis characterized by infiltration with retinoic acid receptor-related orphan nuclear receptor γt-positive lymphocytes, macrophages, and eosinophils. Despite pronounced interstitial kidney disease and abnormal kidney function, STAT-1(-/-) mice had decreased proteinuria, glomerulonephritis, and autoantibody production. Phosphospecific flow cytometry revealed shunting of STAT phosphorylation from STAT-1 to STAT-3/4., Conclusion: We describe unique contributions of STAT-1 to pathology in different kidney compartments in a mouse model, and provide potentially novel insight into tubulointerstitial nephritis, a poorly understood complication that predicts end-stage kidney disease in SLE patients., (© 2016, American College of Rheumatology.)

Objective: The aim of this study was to evaluate the effect of increased exposure time and light intensity on microhardness of cured composite through different thicknesses of tooth structure in primary teeth., Materials and Methods: One hundred and seventy cylindrical resin composite specimens were prepared. All specimens were divided into 17 experimental and control groups. "Light-emitting diode" light curing unit (LCU) applied directly or through 1, 2, and 3 mm thicknesses tooth slices for experimental groups. The irradiation protocols were 25 and 50 s at 650 mW/cm(2) and 15 and 30 s at 1100 mW/cm(2). The "quartz-tungsten-halogen" LCU (400 mW/cm(2)) for 40 s was used in control group. Microhardness was measured by the Vickers hardness test., Results: Indirectly cured specimens and those cured through a 1 mm thick tooth structure, an increase in intensity caused hardness drop. In the specimens cured through 2 and 3 mm thick tooth structures, increased intensity and/or exposure time did not show any appropriate changes on microhardness., Conclusion: Irradiation through a 1.0 mm thick tooth slice resulted in reduced microhardness although it was still within the clinically acceptable level. The hardness values of the specimens cured through 2 or 3 mm thick tooth slices fell below the clinically acceptable level even after doubling the exposure time and/or light intensity.

The specific function of microglia, the tissue resident macrophages of the brain and spinal cord, has been difficult to ascertain because of a lack of tools to distinguish microglia from other immune cells, thereby limiting specific immunostaining, purification, and manipulation. Because of their unique developmental origins and predicted functions, the distinction of microglia from other myeloid cells is critically important for understanding brain development and disease; better tools would greatly facilitate studies of microglia function in the developing, adult, and injured CNS. Here, we identify transmembrane protein 119 (Tmem119), a cell-surface protein of unknown function, as a highly expressed microglia-specific marker in both mouse and human. We developed monoclonal antibodies to its intracellular and extracellular domains that enable the immunostaining of microglia in histological sections in healthy and diseased brains, as well as isolation of pure nonactivated microglia by FACS. Using our antibodies, we provide, to our knowledge, the first RNAseq profiles of highly pure mouse microglia during development and after an immune challenge. We used these to demonstrate that mouse microglia mature by the second postnatal week and to predict novel microglial functions. Together, we anticipate these resources will be valuable for the future study and understanding of microglia in health and disease.

Background and aims. Dental trauma is an important oral health problem in children that requires immediate and appropriate management for long term success. The aim of this study was to evaluate the knowledge of elementary school teachers about emergency management of traumatic dental injuries in children. Materials and methods. A total of 163 teachers from 21 elementary schools from Mashhad, Iran participated in this study. Data were collected using a two-part questionnaire comprised of questions regarding demographic data and participants' knowledge about dental trauma. Results. The level of the teachers' knowledge was moderate (53.3%). Of the 163 participants, 104 (63.8%) were females and 59 (36.2%) were males. Among several predictors that were surveyed in this study, only the age of respondents signifi-cantly impacted the teachers' knowledge (P = 0.004). Conclusion. The results of this study showed the moderate level of knowledge of teachers regarding emergency manage-ment of traumatized teeth. Adding dental trauma management courses in the teacher training curriculum and requiring teachers to be updated in regards to this issue can improve their knowledge and thus led to better management of traumatic dental injuries in children.

Background and aims . In the presence of certain systemic diseases, oral microflora may aggravate the condition of the disease. Microbial population in the oral cavity especially with heart disease can increase the risk of bacterial endocarditis. The aim of this study was to evaluate the rate of oral Streptococcus mutansand the rate of caries in children suffering from heart disease. Materials and methods. In this cross-sectional research, 66 children with congenital or acquired heart disease and 50 healthy children were selected. Children were orally examined and decayed, missing, and filled teeth (DMFT) index was recorded for each subject. Saliva samples were taken from all subjects, and cultured on a special laboratory media and another specific media for S. mutans (sorbitoll +manitol). Bacterial counts were recorded, and for statistical analysis, chi square, Pearson's, and Exact Fisher tests were performed using SPSS 16 software. Results. The rate of S. mutans in children with congenital heart disease was significantly higher than the rates in childrenwith acquired heart disease and healthy control subjects. The mean DMFT in children with acquired heart disease who tookpenicillin as prophylaxis monthly was significantly lower than the other groups. Conclusion . The results revealed lower oral bacteria counts and comparatively lower caries rates in children with heart diseases, probably because of an effect of the regular prophylactic antibiotic regimen.

Background and aims. Tooth loss may lead to mastication disability, which in turn has important impact on individual's quality of life. Social and psychological factors have been shown to be associated with tooth loss. This study aimed to investigate the social determinants and prevalence of tooth loss, and presence of functional dentition among female heads of household under support of Welfare Organization in Mashhad, Iran. Materials and methods. In current study 556 participants aged 16-76 years were recruited. Sociodemographic characteristics (age, level of education, family size, and monthly income) were collected using interviewer-led questionnaire. Data about number of teeth and functional dentition were obtained by oral examination. The data were analyzed using Chi-square, Kruskal-Wallis, T-test and binary logistic regression analysis. Results. Four percent of participants were edentulous. Tooth loss was significantly associated with level of education, age and family size (P < 0.001). There was no significant association between level of income and tooth loss (P = 0.88). Only 37.5% of dentate subjects had functional dentition (anterior and premolar teeth). Women older than 40 years were 0.63 times less likely to have functional dentition than those younger than 40 years. Females with at least a high-school diploma were six times more likely to have functional dentition than their illiterate counterparts. Conclusion. Social determinants of functional dentition should be taken into account when planning oral health promoting programs for female heads of household. For reducing oral health inequalities access to dental services should be facilitated.

Activation of TLR9 by direct injection of unmethylated CpG nucleotides into a tumor can induce a therapeutic immune response; however, Tregs eventually inhibit the antitumor immune response and thereby limit the power of cancer immunotherapies. In tumor-bearing mice, we found that Tregs within the tumor preferentially express the cell surface markers CTLA-4 and OX40. We show that intratumoral coinjection of anti-CTLA-4 and anti-OX40 together with CpG depleted tumor-infiltrating Tregs. This in situ immunomodulation, which was performed with low doses of antibodies in a single tumor, generated a systemic antitumor immune response that eradicated disseminated disease in mice. Further, this treatment modality was effective against established CNS lymphoma with leptomeningeal metastases, sites that are usually considered to be tumor cell sanctuaries in the context of conventional systemic therapy. These results demonstrate that antitumor immune effectors elicited by local immunomodulation can eradicate tumor cells at distant sites. We propose that, rather than using mAbs to target cancer cells systemically, mAbs could be used to target the tumor infiltrative immune cells locally, thereby eliciting a systemic immune response.

Dental practitioners are susceptible to Musculoskeletal Disorders (MSDs). The symptoms may begin from education courses. The aim of this study was to assess prevalence of musculoskeletal pain reports and correlated factors among the Mashhad Dental School students, Iran. A total of 177 undergraduate and postgraduate dental students, who were involved in educational clinical training, completed a questionnaire focusing on pain reports of different body anatomical regions. Variables such as gender, academic grade, academic year, clinical working hour, regular exercise times and also pain characteristics including pain duration intensity and frequency were evaluated. As results 82% of undergraduate students and 90% of postgraduate students reported body pain in at least one region. The most prevalent pain locations were: chest/shoulder (46.9%), head/neck (41.8%), middle back (33.9%) and right hand (25.4%). Severity of reported pain was increased due to performing dental work, increased working stress and working fatigue. Regular exercise was associated with alleviated some pain characteristics. As conclusions musculoskeletal pain reports were highly prevalent among dental students. Attention to prevention of musculoskeletal disorders should be considered as priority in dental schools. For dental students exercise training courses for strengthening muscles of shoulder/chest, neck and back are necessary.

Background: Dental unit waterline system is considered potential source for contamination with Legionella species. The aim of this study was to determine if contamination of a dental unit water line system by Legionella pneumophila serogroup1 in the Mashhad School of Dentistry occurred in 2009., Methods: A total of 52 dental units were selected from all clinical departments of the Mashhad School of Dentistry. Samples of water were collected from outlets of water/air spray, high-speed dental hand pieces and water cup fillers. Samples were tested via the ELISA method., Results: At the beginning of the work day, a total of 36.1 percent of dental units were contaminated by Legionella pneumophila serogroup 1., Conclusion: Infection control of the dental unit water line system regarding legionella in the Mashhad School of Dentistry is a challenge and engineering controls should be used in contaminated clinics.

In multiple sclerosis and the experimental autoimmune encephalitis (EAE) mouse model, two pools of morphologically indistinguishable phagocytic cells, microglia and inflammatory macrophages, accrue from proliferating resident precursors and recruitment of blood-borne progenitors, respectively. Whether these cell types are functionally equivalent is hotly debated, but is challenging to address experimentally. Using a combination of parabiosis and myeloablation to replace circulating progenitors without affecting CNS-resident microglia, we found a strong correlation between monocyte infiltration and progression to the paralytic stage of EAE. Inhibition of chemokine receptor-dependent recruitment of monocytes to the CNS blocked EAE progression, suggesting that these infiltrating cells are essential for pathogenesis. Finally, we found that, although microglia can enter the cell cycle and return to quiescence following remission, recruited monocytes vanish, and therefore do not ultimately contribute to the resident microglial pool. In conclusion, we identified two distinct subsets of myelomonocytic cells with distinct roles in neuroinflammation and disease progression.

Microgliosis is a common response to multiple types of damage in the CNS. However, the origin of the cells involved in this process is still controversial and the relative importance of local expansion versus recruitment of microglia progenitors from the bloodstream is unclear. Here, we investigated the origin of microglia using chimeric animals obtained by parabiosis. We found no evidence of microglia progenitor recruitment from the circulation in denervation or CNS neurodegenerative disease, suggesting that maintenance and local expansion of microglia are solely dependent on the self-renewal of CNS resident cells in these models.

Amyotrophic lateral sclerosis (ALS) is associated with increased numbers of microglia within the central nervous system (CNS). However, it is unknown whether the microgliosis results from proliferation of CNS resident microglia, or recruitment of bone marrow (BM)-derived microglial precursors. Here we assess the distribution and number of BM-derived cells in spinal cord using transplantation of green fluorescent protein (GFP)-labeled BM cells into myelo-ablated mice over-expressing human mutant superoxide dismutase 1 (mSOD), a murine model of ALS. Transplantation of GFP+ BM did not affect the rate of disease progression in mSOD mice. Mean numbers of microglia and GFP+ cells in spinal cords of control mice were not significantly different from those in asymptomatic mSOD mice and showed no change with animal age. The number of GFP+ cells and microglia (F4/80+ and CD11b+ cells) within the spinal cord of mSOD mice increased compared to age-matched controls at a time when mSOD mice exhibited disease symptoms, continuing up to disease end-stage. Although we observed an increase in the number of GFP+ cells in spinal cords of mSOD mice with disease symptoms, mean numbers of GFP+ F4/80+ cells comprised less than 20% of all F4/80+ cells and did not increase with disease progression. Furthermore, the relative rates of proliferation in CD45+GFP- and CD45+GFP+ cells were comparable. Thus, we demonstrate that the microgliosis present in spinal cord tissue of mSOD mice is primarily due to an expansion of resident microglia and not to the recruitment of microglial precursors from the circulation., (Copyright 2006 Wiley-Liss, Inc.)
