2. Effect of Neopterin on Splenic Erythropoiesis in Mice

Abstract

Neopterin is produced by monocytes and is a biomarker for inflammation. We recently found that neopterin induces stromal cells to produce cytokines that suppress erythropoiesis in the bone marrow of mice. Despite this suppression, the peripheral hematocrit values are not decreased. The goal of the present study was to determine whether the maintenance of hematocrit values is caused by a neopterin-induced acceleration of extramedullary erythropoiesis. We examined the effects of intravenously injected neopterin on splenic erythropoiesis in C57BL/6J mice and found a marked increase (259% of the pretreatment level) in the number of splenic erythroid progenitor cells (BFU-E and CFU-E) after a slight temporal decrease in the number of erythroid progenitor cells. These results suggest that neopterin increases splenic erythropoiesis to compensate for the suppression of erythropoiesis in the bone marrow, resulting in the maintenance of hematocrit levels in peripheral blood.

Abstract

Neopterin is produced by monocytes and a useful biomarker of inflammatory responses. We found recently in mice that neopterin suppressed erythropoiesis but enhanced myelopoiesis possibly by activating stromal cells, the same phenomenon observed in inflammation. In this study, we investigated whether neopterin affects the proliferation and differentiation of immature erythroid progenitor cells (BFU-E) by stimulating bone-marrow stromal cells in culture. Neopterin decreased the number of colonies of BFU-E formed by unfractionated bone-marrow cells in a semisolid culture system, but increased the number of granulocyte-macrophage progenitor cells (CFUGM). In contrast, it did not affect numbers of BFU-E or CFU-GM among a lineage-negative (fractionated) bonemarrow- cell population cultured without stromal cells. These results suggest that neopterin decreased the number of colonies of BFU-E by activating bone-marrow stromal-cell function.

Abstract

Neopterin is produced by monocytes and is a useful biomarker for inflammation. We recently found that neopterin is biologically active against hematopoiesis during inflammatory processes. Namely, neopterin stimulates bone marrow myelopoiesis but suppresses B-lymphopoiesis and erythropoiesis. Neopterin also enlarges the splenic mast cell progenitor pool. These hematopoietic responses are not due to a direct effect of neopterin on hematopoietic stem cells; rather, they are an indirect effect mediated by the stromal cell production of cytokines. These hematopoietic responses are compatible with the hematopoietic response that occurs during inflammatory processes such as infection. These findings suggest that neopterin reinforces the hematopoietic response during inflammation and contributes to the potential upregulation of the inflammatory process as a host defense. Thus, neopterin acts as a potent hematopoietic regulatory factor by activating stromal cell functions, although further study is needed to elucidate the mechanism of action.

Abstract

Neopterin is produced by monocytes and is a useful biomarker of inflammatory responses. We found that neopterin enhances granulopoiesis, but suppresses B-lymphopoiesis triggered by the positive- and negative regulations of cytokines produced by stromal cells in mice. Furthermore, neopterin suppressed the colony formation of mast cell progenitor (CFU-mast) from bone marrow cells in in vitro culture system. In this study, neopterin was also found to regulate the proliferation and differentiation of splenic CFU-mast in vitro as observed in that from bone marrow, which was confirmed in the mouse model of senescent stromal-cell impairment (SCI). In non- SCI mice (=less senescent stage of SCI mice), neopterin also decreased the number of colonies of interleukin-3 (IL-3)-dependent mast-cell progenitor cells (CFU-mast) from unfractionated spleen cells, but not that from the lineage-negative (fractionated) spleen cell population without stromal cells in a semisolid in vitro culture system. In contrast, in a case of SCI mice, the treatment with neopterin did not decrease the number of colonies of IL-3- dependent mast-cell progenitor cells (CFU-mast) from unfractionated spleen cells. These results suggest that, firstly, neopterin decrease the number of colonies of IL-3-dependent CFU-mast by stimulating splenic stromal cells, and secondly, such neopterin function becomes declined during senescence because of an impaired stromalcell function.

9. In vivo Stimulatory Effects of Neopterin on Hematopoiesis

Abstract

The pteridine neopterin (NP) was shown to be produced by monocytes and has been known as a useful maker of immunological activation, although, the biological activity of this agent is still unclear. To elucidate the biological function of NP, the changes in the number of blood leukocyte, bone marrow and spleen hemopoietic progenitor cells after intraperitoneal administration of NP into mice were investigated. Administration of NP increased the number of blood leukocytes about 2 fold higher than that of the control at days 7 and 14. Blood films, made by smearing samples of peripheral blood, showed the increment of granulocytes in blood of NP treated animals. The number of granulocyte-macrophage progenitor cells (CFU-GM) was also increased in both bone marrow and spleen in mice with NP administration. Extensive study showed that NP stimulated the hematological recovery in bone marrow transplanted animals. All these findings suggest that NP has a stimulating activity on hemopoiesis by affecting the proliferation and differentiation of hemopoietic progenitor cells in vivo.

Abstract

We previously reported that macrolide antibiotics, such as clarithromycin (CAM), blocked autophagy flux, and simultaneous proteasome and autophagy inhibition by bortezomib (BTZ) plus CAM resulted in enhanced apoptosis induction in multiple myeloma (MM) cells via increased endoplasmic reticulum (ER) stress loading. However, in actual therapeutic settings, cell adhesion-mediated drug resistance between bone marrow stromal cells (BMSC) and MM cells has been known to be a barrier to treatment. To investigate whether CAM could enhance BTZ-induced cytotoxicity in MM cells under direct cell adhesion with BMSC, we established a co-culture system of EGFP-labeled MM cells with BMSC. The cytotoxic effect of BTZ on MM cells was diminished by its interaction with BMSC; however, the attenuated cytotoxicity was recovered by the co-administration of CAM, which upregulates ER stress loading and NOXA expression. Knockout of NOXA in MM cells canceled the enhanced cell death by CAM, indicating that NOXA is a key molecule for cell death induction by the co-administration of CAM. Since NOXA is degraded by autophagy as well as proteasomes, blocking autophagy with CAM resulted in the sustained upregulation of NOXA in MM cells co-cultured with BMSC in the presence of BTZ. Our data suggest that BMSC-associated BTZ resistance is mediated by the attenuation of ER stress loading. However, the addition of CAM overcomes BMSC-associated resistance via upregulation of NOXA by concomitantly blocking autophagy-mediated NOXA degradation and transcriptional activation of NOXA by ER stress loading.
