Background: Babesia spp. are protozoan parasites that infect the red blood cells of domesticated animals, wildlife and humans. A few cases of giant pandas (a flagship species in terms of wildlife conservation) infected with a putative novel Babesia sp. have been reported. However, comprehensive research on the morphological and molecular taxonomic classification of this novel Babesia sp. is still lacking. This study was designed to close this gap and formally describe this new Babesia sp. infecting giant pandas. Methods: Detailed morphological, molecular and phylogenetic analyses were conducted to characterise this Babesia sp. and to assess its systematic relationships with other Babesia spp. Blood samples from giant pandas infected with Babesia were subjected to microscopic examination. The 18S ribosomal RNA (18S rRNA), cytochrome b (cytb) and mitochondrial genome (mitogenome) of the new Babesia sp. were amplified, sequenced and assembled using DNA purified from blood samples taken from infected giant pandas. Based on the newly generated 18S rRNA, cytb and mitogenome sequences, phylogenetic trees were constructed. Results: Morphologically, the Babesia sp. from giant pandas exhibited various forms, including round to oval ring-shaped morphologies, resembling those found in other small canine Babesia spp. and displaying typical tetrads. Phylogenetic analyses with the 18S rRNA, cytb and mitogenome sequences revealed that the new Babesia sp. forms a monophyletic group, with a close phylogenetic relationship with the Babesia spp. that infect bears (Ursidae), raccoons (Procyonidae) and canids (Canidae). Notably, the mitogenome structure consisted of six ribosomal large subunit-coding genes (LSU1-6) and three protein-coding genes (cytb, cox3 and cox1) arranged linearly. Conclusions: Based on coupled morphological and genetic analyses, we describe a novel species of the genus Babesia, namely, Babesia ailuropodae n. sp., which infects giant pandas. The giant panda (Ailuropoda melanoleuca), as an endemic species of China, is also a flagship species of wildlife Conservation. However, the accuracy of individual identification of giant pandas is typically problematic due to the high error rate of microsatellite typing methods. In this study, a panel of 61 SNP makers was developed for individual identification. Of these, 55 polymorphic SNP markers were successfully genotyped. The observed heterozygosity (Ho), expected heterozygosity (He) and polymorphic information content (PIC) ranged from 0.056 to 0.611 (mean 0.352), 0.131–0.509 (mean 0.389) and 0.099–0.389(mean 0.305), respectively. No SNPs significantly deviated from Hardy–Weinberg equilibrium (P < 0.05). Livestock grazing and the collection of bamboo shoots are the main threats to giant panda (Ailuropoda melanoleuca) habitat in the Liangshan Mountains in China. It is important to clarify the effect of these disturbances to the giant panda to formulate targeted management policies. Based on species distribution models and daily activity models, we investigated the effects of livestock grazing and bamboo shoot collection on giant pandas from May 2021 to July 2022. Our results indicated the giant panda's suitable habitat in the reserve covered 51.83 km² (15.02% of the reserve area). Grazing and bamboo shoot collection led to losses of 19.08 km² and 7.68 km² of suitable habitat, respectively. Together, the 2 activities resulted in a loss of 28.35 km² of suitable habitat, which was more than half of the area of panda habitat. The areas of suitable habitat for giant pandas significantly overlapped with the areas affected by both disturbances. Giant pandas did not show significant differences in daily activity rhythms under a single disturbance, but the daily activity rhythms of giant pandas differed when we compared the area combining the 2 disturbances with the undisturbed area. Our study reveals that the anthropogenic disturbances in the reserve have varying effects on the suitable habitat range and daily activity rhythm of giant pandas and evidence of a synergistic effect. Therefore, when formulating relevant conservation policies, it is important to fully evaluate the extent and characteristics of anthropogenic disturbances in shaping the population distribution and habitat preferences of the giant panda and other wildlife to enhance the efficacy of conservation management practices. Responses of diverse gut microbiota affect the host's nutrient metabolism and disease immunity. However, giant pandas (Ailuropoda melanoleuca) have a low diversity of gut microbiota, and the reasons for this low diversity and its impact are poorly understood. In this study, we aimed to investigate the effects of diet on the diversity of gut microbiota in giant pandas. To analyze the functional genes of the gut microbiota, we employed PICRUSt (phylogenetic investigation of communities by reconstruction of unobserved states), a computational method for predicting the functional composition of genomes. A multispecies comparison was conducted, involving 66 individuals from 34 species including giant panda. Our comparative analysis revealed that a high-fiber diet significantly reduced the diversity of gut microbiota in giant pandas. Furthermore, we observed that the metabolic function of giant pandas, particularly in terms of energy families and amino acid metabolism, was significantly lower than in other species. During the high-fiber diet, the gut microbiota of giant pandas exhibited a greater abundance of genes involved in carbohydrate metabolism, amino acid metabolism, and transport functions. These findings suggest that a high-fiber bamboo (Poaceae) diet is associated with lower diversity and altered metabolic function of gut microbiota in giant pandas. Flagship species' conservation strategies hold significant prominence in biodiversity preservation. The giant panda, a globally recognized species, has drawn attention to its benefits and constraints as a flagship species. This study aimed to assess the potential benefits of a dual flagship strategy using both the giant panda and snow leopard, compared to an approach solely using the giant panda. We identified the number of potential beneficiary species based on their habitat overlap with the giant panda and snow leopard in Sichuan and Gansu, China. Subsequently, we examined public preferences for these two flagships and their influencing factors through questionnaire surveys within and outside China. The dual flagship strategy covered the habitats of more species and amplified existing protection for those species already benefiting from giant panda conservation efforts. The giant panda was commonly perceived as "Adorable", "Innocent", and "Rare", while perceptions of the snow leopard leaned towards "Mighty", "Mysterious", and "Rare". Though the giant panda is widely favored, the survey indicates a notable preference for snow leopards among a proportion of respondents. The dual flagship strategy offers expanded wildlife habitat coverage and benefits a broader range of species. Moreover, the combined appeal of the snow leopard and giant panda, each possessing unique charm and symbolism, holds the potential to garner broader societal interest and support. This study may serve as a reference for policy decisions in the Giant Panda National Park and other similar protected areas, optimizing conservation management and outreach initiatives for flagship species strategies. Analyses of animal social networks have traditionally been conducted on species that exhibit social behaviors such as group living, whereas relatively less work has been done on species that are thought of as solitary, are cryptic, and that communicate through scent-marking cues. We employed noninvasive fecal genetic sampling, conducted from March 2015 to February 2016, to identify individuals of one such species, the giant panda (Ailuropoda melanoleuca), across a study population in southwestern China. We then used spatiotemporal proximity thresholds to infer scent mark–based association networks and conduct social network analyses of the population. Results show social clustering in which cluster members preferentially associated with each other. Genetic relatedness was a positive predictor of associations outside the mating season but a negative predictor of associations in the mating season (Mar–Jun), potentially indicating a behavioral change that would reduce risk of inbreeding and kin competition that we term "kin-space recognition." Giant panda Ailuropoda melanoleuca exhibits are popular attractions for zoos and wildlife parks. However, it remains to be investigated whether such exhibits enhance visitor knowledge about pandas and broader conservation issues. We conducted questionnaire surveys at giant panda exhibits at three city zoos and five wildlife parks in China. Although visitors were generally interested in the giant panda, this was not reflected in their post-exhibit knowledge of giant panda biology. Socio-demographically, men were more knowledgeable of giant panda biology than women. Knowledge correlated positively with respondent level of education. Younger respondents (< 45 years) knew most about giant pandas and expressed an interest in learning more about them using social media. The most informed respondents had visited other giant panda exhibits previously. Respondents were generally satisfied with the giant panda exhibits (mean score 4.44/5). Wildlife parks delivered a better educational outcome than city zoos. We recommend approaches to improve the visitor experience further and to leverage public interest in broader conservation engagement and action in China. [ABSTRACT FROM AUTHOR]

An important tool for conserving species and their habitats and achieving sustainable development is the establishment of national parks. The population ecology of the species to be conserved, including their population size and suitable habitat, needs to be assessed prior to integrating protected areas into national parks. The Liangshan Mountains are currently the southernmost habitat of giant pandas and lie outside the newly established Giant Panda National Park (GPNP). The population is threatened with extinction due to severe isolation and human disturbance. However, there has been a ten-year gap in the census of giant pandas in this critical area. This means that conservation managers are unable to keep up to date with population and habitat dynamics. The Meigu Dafengding National Nature Reserve is the core area of the giant panda population in Liangshan and the link for gene exchange. The focus of this study is to assess the spatial distribution pattern of suitable habitat, habitat fragmentation and the habitat selection characteristics of giant pandas in the Meigu Dafengding Nature Reserve in Liangshan in order to lay the foundation for the future inclusion of Liangshan into the national park. A total of 151 giant panda fecal samples were collected in 2023. Using the distance–bamboo stem fragments method, this indicated that nearly 28 to 29 giant pandas exist within the reserve. Based on MaxEnt and FRAGSTATS, the giant panda population is concentrated in Wahei–Yiziyakou. They tend to choose gentle slopes of 10–30°, at altitudes between 2500 m and 3500 m, with average annual temperatures between 8.5 °C and 10 °C. They also select the areas close to roads and settlements for their frequent activities. Unsuitable habitat is widespread and surrounds low-, medium- and high-suitability habitats with poor habitat connectivity. In situ conservation measures are urgently needed due to the restricted distribution of populations and poor habitat suitability. This lays the foundation for the future inclusion of Liangshan into the national park, providing greater protection for the giant panda and other species in the region, and for other national parks to integrate all habitats into a single management unit. Ticks and tick-borne diseases have negative impacts on the health of wild animals including endangered and vulnerable species. The giant panda (Ailuropoda melanoleuca), a vulnerable and iconic flagship species, is threatened by tick infestation as well. Not only can ticks cause anemia and immunosuppression in the giant panda, but also bacterial and viral diseases. However, previous studies regarding tick infestation on giant pandas were limited in scope as case reports from sick or dead animals. In this study, an investigation focusing on the tick infestation of a reintroduced giant panda at the Daxiangling Reintroduction Base in Sichuan, China was conducted. Ticks were routinely collected and identified from the ears of the giant panda from March to September in 2021. A linear model was used to test the correlation between tick abundance and climate factors. All ticks were identified as Ixodes ovatus. Tick abundance was significantly different among months. Results from the linear model showed temperature positively correlated to tick abundance, while air pressure had a negative correlation with tick abundance. To the best of our knowledge, this study is the first reported investigation of tick species and abundance on a healthy giant panda living in the natural environment, and provides important information for the conservation of giant pandas and other species sharing the same habitat.

Signal detection theory propounds that animals choose the substrate and location of their scent marks so that the signals released are transmitted more widely and last longer. We believe that pandas trade‐off scent‐marking because they are an energetically marginal species and it is costly to generate and mark chemical signals. Existing studies only indicate where pandas mark more frequently, but their selection preferences remain unknown. This study investigates whether the marking behavior of pandas is consistent with signal detection theory. Feces count, reflecting habitat use intensity, was combined with mark count to determine the selection preference for marking. The results showed that pandas preferred to mark ridges with animal trails and that most marked tree species were locally dominant. In addition, marked plots and species were selected for lower energy consumption and a higher chance of being detected. Over 90% of the marks used were the longest‐surviving anogenital gland secretion marks, and over 80% of the marks were oriented toward animal trails. Our research demonstrates that pandas go out of their way to make sure their marks are found. Abstract There are only six isolated living giant panda populations, and a comprehensive understanding of their genetic health status is crucial for the conservation of this vulnerable species. Liangshan Mountains is one of the main distribution areas of living giant pandas and is outside the newly established Giant panda national park. In this study, 971 giant panda fecal samples were collected in the heartland of Liangshan Mountains (Mabian Dafengding Nature Reserve: MB; Meigu Dafengding Nature Reserve: MG; and Heizhugou Nature Reserve: HZG). Microsatellite markers and mitochondrial D‐loop sequences were used to estimate population size and genetic diversity. We identified 92 individuals (MB: 27, MG: 22, HZG: 43) from the three reserves. The giant panda (Ailuropoda melanoleuca) and the Chinese red panda (Ailurus styani) are distributed in the same region in the mountain forest ecosystem on the eastern edge of the Qinghai Tibet Plateau and share the same food sources. In order to understand how sympatric giant pandas and Chinese red pandas maintain interspecific relationships to achieve stable coexistence, we used species distribution models and diurnal activity rhythms to analyze the spatial and temporal niche characteristics of giant pandas and Chinese red pandas in the Daxiangling Mountain system based on 187 camera traps data. The results show that: (1) In the Daxiangling Mountains, the total area of suitable habitats for giant pandas and Chinese red pandas is 717.61 km2 and 730.00 km2, respectively, accounting for 17.78% and 18.25%, respectively, of the study area. (2) The top five environmental factors contributing to the model of giant panda and Chinese red panda are precipitation seasonality, temperature seasonality, distance to the road, and elevation and vegetation type. (3) The total overlapping area of suitable habitats for giant pandas and Chinese red pandas is 342.23 km2, of which the overlapping area of highly suitable habitats is 98.91 km2. The overlapping index of suitable habitats is 0.472, and the overlapping index of highly suitable habitats is 0.348, which indicates that the two achieve spatial niches are separated to achieve stable coexistence. (4) The overlapping index of the daily activity rhythm of giant panda and Chinese red panda is 0.87, which is significantly different (p <.05). The existence of Chinese red panda will significantly affect the daily activity rhythm of giant panda (p <.001). This research can provide scientific reference for the researches about population and habitat protection of giant pandas and Chinese red pandas, so as to understand the driving mechanism of resource allocation and population dynamics of sympatric species. Simple Summary: The giant panda is an endemic species in China and the flagship species of global wildlife conservation. Habitat studies of the giant panda corridor can reveal their survival status, habitat environment, and the threats they face, which is crucial for giant panda population recovery and habitat conservation. The results of this study show that due to the opening of the National Road 5 (G5) Niba Mountain tunnel and the completion of the Niba Mountain giant panda corridor plan, the recovery of vegetation within the Niba Mountain giant panda corridor has led to the emergence of giant panda activity in the area, which may have spread to the central part of the reserve through the corridor. The habitat selection characteristics of the giant pandas in the corridor were clarified by investigating the microhabitats of the giant panda corridor in Niba Mountain. These findings can provide a reference for scientists to formulate practical habitat conservation and management measures for giant pandas in the study area. Habitat reduction and increased fragmentation are urgent issues for the survival and recovery of the giant panda (Ailuropoda melanoleuca). However, changes in the distribution and microhabitat selection of giant panda habitats in different seasons in the same region have rarely been assessed. To further understand giant panda habitat requirements, this study analyzed the giant panda habitat selection characteristics and differences using the sample data of the giant panda occurrence sites collected during 2020–2022. The results showed that the giant panda in both seasons selected medium altitudes (2000–2400 m), southeastern slopes, slopes less than 15°, taller tree layers (8–15 m) with a larger diameter at breast height (17–25 cm) and medium density (25–55%), shorter shrub layers (<4 m) with sparse density (<30%), and taller bamboo (>2 m) with high density (>35%). The giant panda microhabitat survey in the Niba Mountain corridor clarified the characteristics of suitable habitat selection for the giant panda in the corridor. Simple Summary: Giant pandas are key protected animals in China. Baylisascaris schroederi, a parasitic nematode, is one of the main health risks threatening them. We used four anthelmintics—pyrantel pamoate (PYR), mebendazole (MBZ), albendazole (ABZ), and ivermectin (IVM)—on 22 enrolled giant pandas. The fecal egg count reduction (FECR) proportions were calculated using both the Markov chain Monte Carlo (MCMC) Bayesian mathematical model and the arithmetic mean based on fecal egg count data. Anthelmintic resistance (AR) was assessed based on the criteria recommended by the World Association for the Advancement of Veterinary Parasitology (WAAVP). We found that the nematode was suspected to be resistant to PYR. The number of eggs per gram in the feces of giant panda enrolled in the present study was increased near the end of the experiment. Baylisascaris schroederi is one of the main health risks threatening both wild and captive giant pandas. The administration of anthelmintics is a common method to effectively control B. schroederi infection, but there is a notable risk of anthelmintic resistance (AR) after long-term, constant use of anthelmintics. Four anthelmintics—pyrantel pamoate (PYR), mebendazole (MBZ), albendazole (ABZ), and ivermectin (IVM)—were each administered separately at intervals of 2 months to 22 enrolled giant pandas. The fecal egg count reduction (FECR) proportions were calculated by both the Markov chain Monte Carlo (MCMC) Bayesian mathematical model and the arithmetic mean. AR was assessed based on the criteria recommended by the World Association for the Advancement of Veterinary Parasitology (WAAVP). The estimated prevalence of B. schroederi infection was 34.1%. After treatment with PYR, MBZ, ABZ, and IVM, it was determined that MBZ, ABZ, and IVM were efficacious against B. schroederi, while nematodes were suspected to be resistant to PYR according to the fecal egg count reduction (FECR) proportions. This paper documents our comparative study on the community environments of giant pandas Ailuropoda melanoleuca between Wolong National Nature Reserve (Wolong NNR), Tangjiahe NNR, and Heizhugou NNR in Sichuan, China using infrared camera data collected from 2017 to 2021, in an attempt to explore the ecological relations of the giant panda with other animals. Results showed that the giant panda belonged to the species association networks of Tangjiahe lower altitude network (Tangjiahe LAN) and Wolong LAN. In Tangjiahe LAN, the giant panda had symmetric associations with two species: the Asiatic golden cat (Catopuma temminckii) and Temminck’s tragopan (Tragopan temminckii). In Wolong LAN, the panda was directly associated with six species. Three types of associations were found in Wolong NNR. The first type was mutually asymmetric association between the panda and the Sichuan snub-nosed monkey (Rhinopithecus roxellanae), in which the two species benefited from each other. The second type was a uni-directional asymmetric association between the giant panda and the red panda (Ailurus fulgens), in which the giant panda benefited from the red panda. The third type was symmetric association between the giant panda and other four species, including the Asiatic black bear (Ursus thibetanus), yellow-throated marten (Martes flavigula), sambar (Rusa unicolor), and tufted deer (Elaphodus cephalophus), in which they competed for similar ecological resources. In Heizhugou NNR, species didn’t form a network and the giant panda was not directly associated with any species. Accordingly, it is suggested that conservation resources be invested on Wolong NNR in the case of resources shortage, because it will ensure the giant panda to further survive easier and, at the same time, will also benefit more other species in surviving. It is also suggested that the state budget currently made for ecological restoration be made with priority to Heizhugou, helping the natural community to recover in the NNR.

Mature female giant pandas usually ovulate once a year. This is followed by an obligatory luteal phase, consisting of a long-lasting corpus luteum dormancy phase (CLD; primary increase in progestogens) and a much shorter active luteal phase (AL; secondary increase in progestogens). Varying duration of both the dormant (embryonic diapause) and AL (post-embryo reactivation) phases has hampered unambiguous pregnancy length determination in giant pandas until today. Additionally, progestogen profiles have been considered not to differ between pregnant and pseudopregnant cycles. Only ceruloplasmin, 13,14-dihydro-15-keto-PGF2α (PGFM) and – more recently – estrogens have been assigned diagnostic power so far. Our study investigated the competence of metabolic (fecal output) and Urinary Specific Gravity (USpG)-normalized urinary endocrine (progestogens, PGFM, glucocorticoids (GCM) and ceruloplasmin) markers for pregnancy monitoring including defining the duration of the AL phase length. Research on 24 (6 pregnant, 8 pseudopregnant and 10 non-birth) cycles of 6 giant pandas revealed a fixed AL phase length of 42 days in giant pandas, e.g. representing 6 weeks of post- diapause development in case of pregnancy. Progestogen concentrations were significantly higher in pregnant cycles throughout the majority of the AL phase, with significant higher values during the AL phase in healthy twin compared to singleton pregnancies. GCM concentrations were also markedly higher in giant pandas expecting offspring, with a clear increase towards birth in the final 2 weeks of pregnancy. This increase in GCM was running in parallel with elevating estrogen and PGFM concentrations, and decreasing progestogens. In addition, during the AL phase, a more pronounced decrease in fecal output was obvious for pregnant females. The combined profiles of non-invasive metabolic and endocrine markers, the latter normalized based on USpG, showed a true pregnancy signature during the AL phase. The findings of this study are applicable to retrospective evaluations of non-birth cycles facilitating categorizing those into pseudopregnant or lost pregnancies, with USpG-normalization of the urinary endocrine markers as a prerequisite.

Five metabolites (1–5), including two new sesquiterpenoids, designated ganodermanol L (1) and 4α,15-epoxyeudesmane-1β,6α,11-triol (2), together with three known structurally related compounds (3-5), have been isolated from the cultures of Streptomyces sp. XM17, a bacteria residing in the fresh feces of the giant panda Ailuropoda melanoleuca. The structures of 1–2 were established on the basis of extensive spectroscopic analyses, including 1D- and 2D-NMR (1H-1H COSY, HMQC, HMBC and NOESY) experiments. Furthermore, the absolute configuration of 1 was established by single-crystal X-ray crystallographic analyses. Of noted, these compounds were found to possessed antiviral activities using the 'pretreatment of virus' approach with IC50 values ranging from 4 to 30 nM, indicating that these sesquiterpenoids were potent in inhibiting the entry of influenza A virus. To achieve reproduction, male solitary mammals need to locate females using chemical communication with high levels of precision. In the case of giant pandas, the total estrus period of females was usually 15 days each year, however, successful mating activity is finished within 3 days from respective home range. The mating pattern of giant pandas, where multiple males compete for each female requires females employ efficient systems to communicate their estrus phases. To verifying whether the scent secretions of giant pandas changes by gender and estrus progression, the microbiota and compounds in 29 anogenital gland samples from 14 individuals during estrus were analyzed by 16S rRNA sequencing and GC-MS. We show that the microbiota communities covary by gender with 4 particular compounds of scent secretions. Among 597 genera, 34 were identified as biomarkers that could be used to distinguish between different estrus phases. By bacterialcompounds co-analysis, 3 fatty ester acids and squalene compounds covaried with the development of estrus in the bacterial communities of female giant pandas. This study helps clarify how a large, solitary mammal expresses accurate information to improve the likelihood of successful reproduction by changing the composition of microbiota and odor compounds of anogenital glands during estrus. An important tool for conserving species and their habitats and achieving sustainable development is the establishment of national parks. The population ecology of the species to be conserved, including their population size and suitable habitat, needs to be assessed prior to integrating protected areas into national parks. The Liangshan Mountains are currently the southernmost habitat of giant pandas and lie outside the newly established Giant Panda National Park (GPNP). The population is threatened with extinction due to severe isolation and human disturbance. However, there has been a ten-year gap in the census of giant pandas in this critical area. This means that conservation managers are unable to keep up to date with population and habitat dynamics. The Meigu Dafengding National Nature Reserve is the core area of the giant panda population in Liangshan and the link for gene exchange. The focus of this study is to assess the spatial distribution pattern of suitable habitat, habitat fragmentation and the habitat selection characteristics of giant pandas in the Meigu Dafengding Nature Reserve in Liangshan in order to lay the foundation for the future inclusion of Liangshan into the national park. A total of 151 giant panda fecal samples were collected in 2023. Using the distance–bamboo stem fragments method, this indicated that nearly 28 to 29 giant pandas exist within the reserve. Based on MaxEnt and FRAGSTATS, the giant panda population is concentrated in Wahei–Yiziyakou. They tend to choose gentle slopes of 10–30°, at altitudes between 2500 m and 3500 m, with average annual temperatures between 8.5 °C and 10 °C. They also select the areas close to roads and settlements for their frequent activities. Unsuitable habitat is widespread and surrounds low-, medium- and high-suitability habitats with poor habitat connectivity. In situ conservation measures are urgently needed due to the restricted distribution of populations and poor habitat suitability. This lays the foundation for the future inclusion of Liangshan into the national park, providing greater protection for the giant panda and other species in the region, and for other national parks to integrate all habitats into a single management unit. This will address conservation gaps and overlapping management, and promote the conservation of rare or endangered species.

A geriatric female giant panda developed grave signs of illness and was diagnosed with suspected hepatobiliary tract obstruction or other severe hepatic disease such as advanced cholangiohepatitis. The giant panda was euthanized and post mortem computed tomography was performed prior to necropsy. Common bile duct obstruction at the major duodenal papilla by a mineral attenuating calculus causing dilatation of common bile and gallbladder with concurrent multiple areas of liver abscess were detected by postmortem computed tomography. These were confirmed with gross necropsy. This is the first case report of common bile duct obstruction by mineral calculus with concurrent severe cholangiohepatitis in a giant panda. The giant panda (Ailuropoda melanoleuca) is a special rare and endangered species in China and is the flagship species for global biodiversity conservation. This study used different methods including Kernel density, statistical analysis, and logistic regression analysis to investigated the spatial distribution, migration in different seasons and different distribution areas, as well as the foraging patch selection strategies of wild giant pandas in Meigu Dafengding National Nature Reserve in the Liangshan Mountains. The results demonstrated that, in the study area, giant pandas were mainly distributed in the Wahei-Yizi Yakou Habitat Corridor with an activity area of 144.93 km2, which accounted for 28.59% of the whole reserve. The core distribution area of the species was 92.07 km2 and the population density was 0.24 pcs km–2. There were two significant seasonal domains for the activities of giant pandas: the low-altitude winter habitat of Yushania maculata Yi and Yushania ailuropodina Yi bamboo species, as well as the high-altitude summer habitat of Bashania fangiana species. With seasonal changes, giant pandas migrated among different bamboo distribution regions. Giant pandas were mainly distributed in high-altitude areas that had secondary forest, tall trees with a large diameter at breast height (DBH) and bamboo forest. The spatial distribution pattern and microhabitat selection of giant pandas in Meigu Dafengding National Nature Reserve were determined. The findings of this study provide scientific implications for the local conservation and management of the habitat of wild giant pandas.

Habitat reduction and increased fragmentation are urgent issues for the survival and recovery of the giant panda (Ailuropoda melanoleuca). However, changes in the distribution and microhabitat selection of giant panda habitats in different seasons in the same region have rarely been assessed. To further understand giant panda habitat requirements, this study analyzed the giant panda habitat selection characteristics and differences using the sample data of the giant panda occurrence sites collected during 2020–2022. The results showed that the giant panda in both seasons selected medium altitudes (2000–2400 m), southeastern slopes, slopes less than 15°, taller tree layers (8–15 m) with a larger diameter at breast height (17–25 cm) and medium density (25–55%), shorter shrub layers (2 m) with high density (>35%). The giant panda microhabitat survey in the Niba Mountain corridor clarified the characteristics of suitable habitat selection for the giant panda in the corridor. The findings of the study can provide scientific references for the development of practical habitat conservation and management measures for giant pandas in the study area.

The use of camera traps is prevalent in the ecological study of giant pandas (Ailuropoda melanoleuca). The reliability of camera-trap surveying results greatly depends on sampling designs that significantly influence the detection probability of the target species. Few studies have tested the efficacy of sampling designs on camera-trap surveys for monitoring giant pandas in a heterogeneous landscape. In this study, we conducted camera trapping of giant pandas based on two different sampling schemes in Changqing National Nature Reserve of China, and evaluated their outcomes based on three aspects: occupancy analysis, photographic rate, and activity pattern. The results demonstrated that both climate heterogeneity and distance to the nearest road had a strong positive influence on site occupancy, and slope and forest cover had a significant negative impact on site occupancy. Significant differences in the direction or magnitude of variables' influences indicated that there were apparently spatial-temporal dynamics of giant panda distribution between two sampling schemes. The low detection probabilities indicated that both sampling schemes were not robust to accurately monitor giant pandas in the whole study area. We recommended that more suitable sampling designs with local covariates be developed for camera-trap surveys monitoring giant pandas to account for temporal variability and small-scale variation in heterogeneous landscapes. Abstract Background Parasitic infections are among the important causes of death of giant pandas (Ailuropoda melanoleuca) that hamper their survival in the wild. There are about 35 species of parasites which have been identified in giant pandas, but no information is currently available regarding the infection of Babesia in giant pandas. Babesia spp. are common intraerythrocytic parasite in wildlife, transmitted by ixodid ticks, which cause babesiosis. Clinical signs of babesiosis include fever, hemolysis, anemia, jaundice and death. Methods A species of Babesia was detected in the blood of a giant panda based on morphology and PCR amplification of the 18S rRNA gene. The phylogenetic relationship of Babesia sp. infecting giant panda was assessed by gene sequence alignment and phylogenetic analysis. Results Our analysis revealed that the Babesia isolate detected was most similar to an unidentified species of Babesia identified in black bears (Ursus thibetanus japonicus) from Japan (Babesia sp. Iwate, AB586027.1) with a 99.56% sequence similarity, followed by Babesia sp. EBB (AB566229.1, 99.50%) and Babesia sp. Akita (AB566229.1, 99.07%). Conclusions To our knowledge, this is the first report of Babesia detected in the giant panda. The results indicate that this Babesia sp. may be a novel species, currently named Babesia sp. strain EBP01.

Abstract Reintroduction is a key approach in the conservation of endangered species. In recent decades, many reintroduction projects have been conducted for conservation purposes, but the rate of success has been low. Given the important role of gut microbiota in health and diseases, we questioned whether gut microbiota would play a crucial role in giant panda's wild‐training process. The wild procedure is when captive‐born babies live with their mothers in a wilderness enclosure and learn wilderness survival skills from their mothers. During the wild‐training process, the baby pandas undergo wilderness survival tests and regular physical examinations. Based on their performance through these tests, the top subjects (age 2–3 years old) are released into the wild while the others are translocated to captivity. After release, we tracked one released panda (Zhangxiang) and collected its fecal samples for 5 months (January 16, 2013 to March 29 2014). Here, we analyzed the Illumina HiSeq sequencing data (V4 region of 16S rRNA gene) from captive pandas (n = 24), wild‐training baby pandas (n = 8) of which 6 were released and 2 were unreleased, wild‐training mother pandas (n = 8), one released panda (Zhangxiang), and wild giant pandas (n = 18). Our results showed that the gut microbiota of wild‐training pandas is significantly different from that of wild pandas but similar to that of captive ones. The gut microbiota of the released panda Zhangxiang gradually changed to become similar to those of wild pandas after release. In addition, we identified several bacteria that were enriched in the released baby pandas before release, compared with the unreleased baby pandas. These bacteria include several known gut‐health related beneficial taxa such as Roseburia, Coprococcus, Sutterella, Dorea, and Ruminococcus. Therefore, our results suggest that certain members of the gut microbiota may be important in panda reintroduction.

Wild animals usually respond to different landscape features at different spatial scales. The adoption of multi-scale modeling frameworks in habitat suitability modeling studies have been shown to improve model performance and provide greater insights into relationships between species and habitat components. Although the advantage of multi-scale modeling, the implementation of this framework lagged considerably. In the present study, we used a multi-scale approach to assess the habitat suitability for the globally endangered giant panda (Ailuropoda melanoleuca) in Qionglai mountain range, Sichuan, China in an effort to provide improved species-environment relationships with the aim of informing conservation efforts. The occurrence data collected from the Fourth National Giant Panda Survey and a presence-only, multi-scale Maxent approach were used to model habitat suitability for giant pandas. Our results showed that the optimal scale identified for each environmental variable varied, and most variables were strongly related to giant panda habitat suitability at a relatively fine-scale (≤ 2000 m). Multi-scale models outperformed their analogous single-scale counterparts with respect to discrimination and predictive ability. Additionally, there were significant differences in spatial predictions between multi-scale and single-scale model. This study reveals the multi-scale response of giant pandas to environmental features and confirms the advantage of multi-scale habitat modeling. Therefore, it is necessary and beneficial to take scale dependence into consideration in future habitat suitability modeling for the giant panda.

Scientific research provides essential information for conservation of threatened species. Data deficiency due to insufficient research impedes the design of conservation plans, and research bias may mistakenly direct limited resources to low biodiversity regions or less threatened species. Here, we conducted a systematic review of published papers, grants, and graduate student training on carnivorans in China to identify species bias and research gaps. Furthermore, we collected intrinsic and extrinsic features of carnivorans, and identified features that impact research intensity using generalized linear models. We found that the amount of research on carnivorans increased markedly after 2000, but species bias existed. Bears and big cats received the greatest research attention, while most small- and medium-sized carnivorans received little attention, thus showing the 80-20 phenomenon. Species with a higher level of endemism and protection under Chinese law received more consideration. As an animal conservation icon in China, the giant panda (Ailuropoda melanoleuca) attracted more than 50% of overall carnivoran research resources. However, the giant panda also showed spillover effects, i.e., post-doctoral graduates who studied the giant panda shifted their research focus to other species after graduation, which may help improve research on other species. Thus, to improve and strengthen Carnivora research and conservation, we suggest investing greater effort in species of less concern, training of more graduate students, and reinforcing academic exchange. The female giant panda (Ailuropoda melanoleuca) has a characteristic reproductive physiology that involves a defined breeding season, embryonic diapause and potential for pseudopregnancy. The hormonal variation related to these reproductive characteristics may be a factor contributing to reproductive pathologies. In this case report, open pyometra was diagnosed in a giant panda. Clinical signs included lethargy, anorexia and mucopurulent vulvar discharge. Diagnostic tests performed included complete blood cell count, plasma biochemistry, protein electrophoresis, cytology, culture and sensitivity and abdominal ultrasonography. Treatment consisted of systemic antibiotics based on culture and sensitivity. Response to treatment was monitored via ultrasonography and visual examination of the vulvar region. Two weeks after treatment, the pyometra resolved and no subsequent complications were seen. This is the first case of pyometra reported in a giant panda. Pyometra should be considered as a differential diagnosis in entire female giant pandas presenting with abnormal clinical signs following breeding season. [ABSTRACT FROM AUTHOR]

Abstract Paradoxically, despite the growth in protected areas globally, many species remain threatened and continue to decline. Attempts to conserve species in suboptimal habitats (i.e., as refugee species) may in part explain this Protected Area Paradox. Refugee species yield poor conservation outcomes as they suffer lower densities and fitness. We suggest that the giant panda may serve as an iconic example, reflecting the contraction and shift in the giant panda's range, diet and habitat use over the past 3,500 years, coinciding with increasing human pressure, and now maintained by conservation efforts, this due to shifted baselines. The global bias of protected area location to less productive habitats indicates that this problem may be widespread. We urgently need efforts to identify victims of refugee species status to allow improved conservation management globally, reducing the paradoxical outcomes of our conservation efforts.

The Chinese government recently finalized a plan to establish a Giant Panda National Park in 2020, one of the first national parks in the country. The plan will extend protection status to a significant amount of areas that were previously unprotected; it will also bring many of the existing giant panda protected areas under one authority in order to improve effectiveness and reduce inconsistencies in management. We provide an overview of the history and status of giant panda conservation and the rationale for creating the park. We also give first-hand information on details of the park design, including its general objectives, geographic range, zone divisions, management and funding structure, as well as analysis of the challenges and opportunities ahead. As a new conservation model for China, the Giant Panda National Park has the ambitious goal of standardizing conservation across a large region. It is a major step toward significantly expanding the amount of area protected and establishing a cohesive conservation network for a sustainable giant panda population in the wild.

The giant panda (Ailuropoda melanoleuca) is a vulnerable species and a charismatic member of zoological collections worldwide. Despite its importance as a representative species for global wildlife conservation efforts, no studies to date have described normal cell morphology or cytoplasmic constituents by traditional techniques such as cytochemical staining and evaluation of ultrastructural features. The objective of this study was to accurately identify and characterize the leukocytes and platelets of clinically healthy giant pandas using routine Wright-Giemsa stain, eight cytochemical stains, immunocytochemistry (CD3), and transmission electron microscopy (TEM) to further the collective understanding of normal cellular morphological features, cytochemical reactivity, and cytoplasmic contents found in health. Voluntary venipuncture was performed on four healthy individual animals (two adults and two juveniles), as part of routine preventive health evaluation. Blood was collected for routine and cytochemical stains, and into 2.5% glutaraldehyde for TEM. On Wright-Giemsa-stained blood films, leukocytes were differentiated into granulocytes (neutrophils, eosinophils, basophils) and mononuclear cells (lymphocytes, monocytes). Cytochemical staining revealed similar leukocyte and platelet staining patterns to those reported in other mammals, with some notable differences. By TEM, leukocytes with nuclear and cytoplasmic features of mononuclear cells were readily differentiated from granulocytes, and platelets had similar ultrastructural features to those reported in other mammals. Neutrophils were the predominant cell type followed by lymphocytes, while basophils were rare. Rare large or reactive lymphocytes, rare reactive monocytes, and rare large platelets were noted in apparently healthy giant pandas of this study. A unique mononuclear cell, with a moderately indented nucleus and shared cytochemical and ultrastructural characteristics of lymphocytes and monocytes, was discovered in this species. The combined cytochemical, immunocytochemical (CD3), and ultrastructural features of these unique cells more closely resemble those of monocytes, but the definitive cell lineage remains unknown at this time. This study provides novel information on giant panda leukocyte morphology and cellular constituents in health, shows the importance of manual blood film review, has important implications for hemogram interpretation in future clinical cases and research, and provides a baseline for future characterization and understanding of hemogram changes in response to disease.

Background/Aims: Giant pandas, an endangered species, are a powerful symbol of species conservation. Giant pandas may suffer from a variety of diseases. Owing to their highly specialized diet of bamboo, giant pandas are thought to have a relatively weak ability to resist diseases. The spleen is the largest organ in the lymphatic system. However, there is little known about giant panda spleen at a molecular level. Thus, clarifying the regulatory mechanisms of spleen could help us further understand the immune system of the giant panda as well as its conservation. Methods: The two giant panda spleens were from two male individuals, one newborn and one an adult, in a non-pathological condition. The whole transcriptomes of mRNA, lncRNA, miRNA, and circRNA in the two spleens were sequenced using the Illumina HiSeq platform. EBseq and IDEG6 were used to observe the differentially expressed genes (DEGs) between these two spleens. Gene Ontology and KEGG analyses were used to annotate the function of DEGs. Furthermore, networks between non-coding RNAs and protein-coding genes were constructed to investigate the relationship between non-coding RNAs and immune-associated genes. Results: By comparative analysis of the whole transcriptomes of these two spleens, we found that one of the major roles of lncRNAs could be involved in the regulation of immune responses of giant panda spleens. In addition, our results also revealed that microRNAs and circRNAs may have evolved to regulate a large set of biological processes of giant panda spleens, and circRNAs may function as miRNA sponges. Conclusion: To our knowledge, this is the first report of lncRNAs and circRNAs in giant panda, which could be a useful resource for further giant panda research. Our study reveals the potential functional roles of miRNAs, lncRNAs, and circRNAs in giant panda spleen.

In November 1928, Theodore Jr. and Kermit Roosevelt led an expedition to China with the expressed purpose of being the first Westerners to kill the giant panda (Ailuropoda melanoleuca). The expedition lasted 8 months and resulted in the brothers shooting a giant panda in the mountains of Sichuan Province. Given the concurrent attention in the popular press describing this celebrated expedition, the giant panda was poised to be trophy hunted much like other large mammals around the world. Today, however, the killing of giant pandas, even for the generation of conservation revenue, is unthinkable for reasons related to the species itself and the context, in time and space, in which the species was popularized in the West. We found that the giant panda's status as a conservation symbol, exceptional charisma and gentle disposition, rarity, value as a nonconsumptive ecotourism attraction, and endemism are integral to the explanation of why the species is not trophy hunted. We compared these intrinsic and extrinsic characteristics with 20 of the most common trophy‐hunted mammals to determine whether the principles applying to giant pandas are generalizable to other species. Although certain characteristics of the 20 trophy‐hunted mammals aligned with the giant panda, many did not. Charisma, economic value, and endemism, in particular, were comparatively unique to the giant panda. Our analysis suggests that, at present, exceptional characteristics may be necessary for certain mammals to be excepted from trophy hunting. However, because discourse relating to the role of trophy hunting in supporting conservation outcomes is dynamic in both science and society, we suspect these valuations will also change in future. Paradoxically, despite the growth in protected areas globally, many species remain threatened and continue to decline. Attempts to conserve species in suboptimal habitats (i.e., as refugee species) may in part explain this Protected Area Paradox. Refugee species yield poor conservation outcomes as they suffer lower densities and fitness. We suggest that the giant panda may serve as an iconic example, reflecting the contraction and shift in the giant panda's range, diet and habitat use over the past 3,500 years, coinciding with increasing human pressure, and now maintained by conservation efforts, this due to shifted baselines. The global bias of protected area location to less productive habitats indicates that this problem may be widespread. We urgently need efforts to identify victims of refugee species status to allow improved conservation management globally, reducing the paradoxical outcomes of our conservation efforts. Interspecific killing is a primary reason for the low survival rates of some animal species. The giant panda (Ailuropoda melanoleuca) is an altricial eutherian mammal and thus, in comparison to other infants, panda cubs are highly vulnerable, which may significantly influence the selection of breeding sites by females. Here, we used infrared camera traps to monitor giant panda dens for 5.5 years in Foping National Nature Reserve (FNNR) to determine how interspecific factors affect den selection by wild female pandas. Results indicated that Asian black bears (Ursus thibetanus), yellow-throated martens (Martes flavigula), leopard cats (Prionailurus bengalensis), and masked palm civets (Paguma larvata) visited the dens frequently, and the presence of these species negatively influenced den selection by female pandas. Interestingly, the presence of rodents and terrestrial birds appeared to indicate den safety, and female giant pandas were not averse and even preferred dens with a high abundance index of rodents and terrestrial birds. The den suitability index (DSI) was a reliable tool for evaluating whether dens were suitable for female giant pandas to give birth to and rear cubs, with preference for dens with high DSI values. This study increases our understanding of the den selection criteria of female giant pandas and the main threats to the survival of their cubs, thus providing important guidance for the conservation and management of this species. Although several mammals impregnate their fur with environmental odors, a phenomenon termed scent anointing or rubbing, the functional relevance of this behavior often is unclear. One theory is that scent anointing could be a form of scent matching with environmental odors to signal competitiveness and home range occupation. In this study we presented giant pandas with a range of odors to determine whether scent matching could provide a functional explanation for scent anointing in this species. We found that only a musk-based perfume elicited significantly more scent-anointing and scent-marking behavior than control. Males were also significantly more likely to scent-anoint and scent-mark than females. A preference for anointing, but not scent marking, when presented with peppermint (an insecticide) also was revealed. Our results suggest that giant pandas differentially scent-anoint with foreign odors to signal home range occupation, and possibly to repel ectoparasites. We also highlight how chemical signaling of resource-holding potential is likely to play an important role in determining competitive interactions between adult male giant pandas. A review of anesthetic procedures used in Andean bears (Tremarctos ornatus), sloth bears (Melursus ursinus), and giant pandas (Ailuropoda melanoleuca) housed at the Smithsonian Institution's National Zoological Park (NZP) from 1995 to 2016 was performed. A total of 146 anesthetic procedures (55 procedures on 12 Andean bears, 38 procedures on nine sloth bears, and 53 procedures on five giant pandas) occurred at NZP during this time frame. Induction protocols involved some combination of ketamine (K; n = 121), tiletamine--zolazepam (TZ; n=91), medetomidine (M; n=67), xylazine (X; n=42), midazolam (Mid; n=9), and butorphanol (B; n = 1). The most commonly used protocols were TZKM for both Andean bears (n = 29) and sloth bears (n = 12), and KX in giant pandas (n = 17). Supplemental injections of K, TZ, Mid, or M were required to complete inductions in 66 cases (45%). Anesthetic maintenance was most often completed with isoflurane, ketamine, or propofol. The most commonly reported complications included perianesthetic emesis (n=47), ptyalism (n=16), and seizure-like activity (n=11). The most frequent physiologic findings included low pulse oximetry values (n= 95), bradycardia (n=95), hypothermia (n=74), and hypertension (n=55). Reintroduction is a key approach in the conservation of endangered species. In recent decades, many reintroduction projects have been conducted for conservation purposes, but the rate of success has been low. Given the important role of gut microbiota in health and diseases, we questioned whether gut microbiota would play a crucial role in giant panda's wild‐training process. The wild procedure is when captive‐born babies live with their mothers in a wilderness enclosure and learn wilderness survival skills from their mothers. During the wild‐training process, the baby pandas undergo wilderness survival tests and regular physical examinations. Based on their performance through these tests, the top subjects (age 2–3 years old) are released into the wild while the others are translocated to captivity. After release, we tracked one released panda (Zhangxiang) and collected its fecal samples for 5 months (January 16, 2013 to March 29 2014). Here, we analyzed the Illumina HiSeq sequencing data (V4 region of 16S rRNA gene) from captive pandas (n = 24), wild‐training baby pandas (n = 8) of which 6 were released and 2 were unreleased, wild‐training mother pandas (n = 8), one released panda (Zhangxiang), and wild giant pandas (n = 18). Our results showed that the gut microbiota of wild‐training pandas is significantly different from that of wild pandas but similar to that of captive ones. The gut microbiota of the released panda Zhangxiang gradually changed to become similar to those of wild pandas after release. In addition, we identified several bacteria that were enriched in the released baby pandas before release, compared with the unreleased baby pandas. These bacteria include several known gut‐health related beneficial taxa such as Roseburia, Coprococcus, Sutterella, Dorea, and Ruminococcus. Abstract Background Hookworms belonging to the genus Ancylostoma (Dubini, 1843) cause ancylostomiasis, a disease of considerable concern in humans and domestic and wild animals. Molecular and epidemiological data support evidence for the zoonotic potential among species of Ancylostoma where transmission to humans is facilitated by rapid urbanization and increased human-wildlife interactions. It is important to assess and describe these potential zoonotic parasite species in wildlife, especially in hosts that have physiological similarities to humans and share their habitat. Moreover, defining species diversity within parasite groups that can circulate among free-ranging host species and humans also provides a pathway to understanding the distribution of infection and disease. In this study, we describe a previously unrecognized species of hookworm in the genus Ancylostoma in the giant panda, including criteria for morphological and molecular characterization. Methods The hookworm specimens were obtained from a wild giant panda that died in the Fengtongzai Natural Reserve in Sichuan Province of China in November 2013. They were microscopically examined and then genetically analyzed by sequencing the nuclear internal transcribed spacer (ITS, ITS1-5.8S-ITS2) and mitochondrial cytochrome c oxidase subunit 1 (cox1) genes in two representative specimens (one female and one male, FTZ1 and FTZ2, respectively). Results Ancylostoma ailuropodae n. sp. is proposed for these hookworms. Morphologically the hookworm specimens differ from other congeneric species primarily based on the structure of the buccal capsule in males and females, characterized by 2 pairs of ventrolateral and 2 pairs of dorsolateral teeth; males differ in the structure and shape of the copulatory bursa, where the dorsal ray possesses 2 digitations. Pairwise nuclear and mitochondrial DNA comparisons, genetic distance analysis, and phylogenetic data strongly indicate that A. ailuropodae from giant pandas is a separate species which shared a most recent common ancestor with A. ceylanicum Looss, 1911 in the genus Ancylostoma (family Ancylostomatidae). Conclusion Ancylostoma ailuropodae n. sp. is the fourth species of hookworm described from the Ursidae and the fifteenth species assigned to the genus Ancylostoma. A sister-species association with A. ceylanicum and phylogenetic distinctiveness from the monophyletic Uncinaria Frölich, 1789 among ursids and other carnivorans indicate a history of host colonization in the evolutionary radiation among ancylostomatid hookworms. Further, phylogenetic relationships among bears and a history of ecological and geographical isolation for giant pandas may be consistent with two independent events of host colonization in the diversification of Ancylostoma among ursid hosts. A history for host colonization within this assemblage and the relationship for A. ailuropodae n. sp. demonstrate the potential of this species as a zoonotic parasite and as a possible threat to human health. The cumulative morphological, molecular and phylogenetic data presented for A. ailuropodae n. sp. provides a better understanding of the taxonomy, diagnostics and evolutionary biology of the hookworms.

Livestock grazing occurs in many protected areas for wildlife and has become a threat to wildlife worldwide. Livestock grazing within protected areas causes negative effects to rare wildlife (e.g., giant panda [Ailuropoda melanoleuca]) and their habitat. We used the 2,000‐km2 Wolong National Nature Reserve, Sichuan Province, southwestern China, to document the effects of livestock on the giant panda and its habitat. We monitored arrow bamboo (Bashania fangiana), wildlife sign (i.e., feces and tracks), and characteristics of plant communities in intact habitat (IH; limited livestock grazing) and disturbed habitat (DH; with grazing disturbance) to assess the effects of livestock grazing and the responses of giant pandas and sympatric species across spatial and temporal scales. Bamboo coverage and the height and basal diameter of bamboo in IH were greater than those in DH, whereas the number of herbaceous species and herbaceous coverage in IH were lower than those in DH. Wildlife signs in IH were greater than those in DH; specifically, giant panda and red panda (Ailurus fulgens) signs were greater, whereas signs of sambar (Rusa unicolor) and tufted deer (Elaphodus cephalophus) in IH were similar to those in DH. Livestock grazing reduced bamboo, which may threaten the long‐term survival of the giant panda. Our results have implications for understanding and management of livestock grazing in the Wolong National Nature Reserve and elsewhere. © 2019 The Wildlife Society. Plant communities in areas disturbed by livestock in Wolong Nature Reserve differed from nearby areas with limited disturbance by livestock. Repeat number variations of coding simple sequence repeats (coding SSRs) generate phenotype diversity for environmental adaptation and rapid adaptive evolution. Despite the positive potential of coding SSR mutations, the repeat variations have also been shown to cause hereditary disorders and severe diseases. However, there have been few reports on the analysis of SSR polymorphism in genome-wide coding regions. We identified 1016 coding SSRs, of which 283 SSRs (27.9%) were polymorphic with 2 to 8 alleles, using 33 re-sequenced giant panda genomes. The 283 polymorphic coding SSRs were mapped on 260 genes, which were assigned into 547 GO terms. Functional analysis of 260 genes showed that four significantly enriched pathways: thyroid hormone signaling pathway, pancreatic secretion, gastric acid secretion, and thyroid hormone synthesis, were relevant to digestion and metabolism. The comparison analysis of gene functions exhibited positive regulation of transcription, DNA-templated term (GO:0045893), the poly(A) RNA-binding term (GO:0008270), and zinc ion–binding term (GO:0044822) which were only significantly enriched in genes containing polymorphic coding SSRs. This indicates coding SSR polymorphism may be involved in binding and regulation functions that can affect the expression of their downstream genes and contribute to diverse phenotypes. Our results provide a solid foundation for further study regarding the effect of coding SSR polymorphism on the adaptation, adaptive evolution, and abnormal traits of the giant panda. The first report of giant pandas (Ailuropoda melanoleuca) infected with a novel Hepatozoon species is presented. An intraleukocytic parasite was detected via routine blood smear from a zoo-housed giant panda at the National Zoological Park. Ribosomal DNA sequences indicated a previously undescribed Hepatozoon species. Phylogenetic and distance analyses of the sequences placed it within its own branch, clustered with Old World species with carnivore (primarily ursid and mustelid) hosts. Retrospective and opportunistic testing of other individuals produced additional positive detections (17/23, 73.9%), demonstrating 100% prevalence (14/14) across five institutions.
