We examined whether urban environmental change drives parallel evolution by sampling 110,019 white clover plants from 6169 populations in 160 cities globally. Plants were assayed for a Mendelian antiherbivore defense that also affects tolerance to abiotic stressors. Urban-rural gradients were associated with the evolution of clines in defense in 47% of cities throughout the world. Variation in the strength of clines was explained by environmental changes in drought stress and vegetation cover that varied among cities. Sequencing 2074 genomes from 26 cities revealed that the evolution of urban-rural clines was best explained by adaptive evolution, but the degree of parallel adaptation varied among cities. Our results demonstrate that urbanization leads to adaptation at a global scale. Fecundity and mating outcomes commonly differ among plant populations occupying contrasting environments. If self-pollination occurs primarily among flowers within plants, contrasting reproductive outcomes among populations must reflect environmental effects on plant-pollinator interactions. Specifically, local conditions could affect features of plant phenotypes that influence pollinator behavior, in turn modifying plant reproductive outcomes.We compared phenotypes, pollinator abundance and behavior, and female fecundity and mating in two meadow populations and two forest populations of Aconitum kusnezoffii within 3 km of each other. Mating outcomes were assessed using microsatellites.Meadow plants generally produced more, shorter ramets with more, larger flowers, but less nectar per flower than forest plants. These differences likely largely represent phenotypic plasticity. Individual bumble bees visited more flowers on forest plants, likely because the more abundant bees in the meadows depleted nectar availability, as indicated by briefer visits to individual flowers. Despite similar fruit set in both habitats, forest plants set more seeds per fruit. Nevertheless, meadow plants produced more seeds overall, owing to sevenfold greater flower production. Consistent with individual bees visiting fewer flowers on meadow plants, more of their seeds were outcrossed. However, the outcrossed seeds of forest plants included more male mates.Reproductive outcomes can vary among populations of animal-pollinated plants as a result of differences in the availability of effective pollinators and environmental effects on plant phenotypes, and their functional consequences for pollinator behavior that governs pollen dispersal.

Summary Interactions between mutualists, competitors, and antagonists have contrasting ecological effects that, sustained over generations, can influence micro‐ and macroevolution. Dissimilar benefits and costs for these interactions should cause contrasting co‐diversification patterns between interacting clades, with prevalent co‐speciation by mutualists, association loss by competitors, and host switching by antagonists.We assessed these expectations for a local assemblage of 26 fig species (Moraceae: Ficus), 26 species of mutualistic (pollinating), and 33 species of parasitic (galling) wasps (Chalcidoidea). Using newly acquired gene sequences, we inferred the phylogenies for all three clades. We then compared the three possible pairs of phylogenies to assess phylogenetic congruence and the relative frequencies of co‐speciation, association duplication, switching, and loss.The paired phylogenies of pollinators with their mutualists and competitors were significantly congruent, unlike that of figs and their parasites. The distributions of macroevolutionary events largely agreed with expectations for mutualists and antagonists. By contrast, that for competitors involved relatively frequent association switching, as expected, but also unexpectedly frequent co‐speciation. The latter result likely reflects the heterogeneous nature of competition among fig wasps.These results illustrate the influence of different interspecific interactions on co‐diversification, while also revealing its dependence on specific characteristics of those interactions.

Fairy rings (FRs) are common ecological grassland landscapes that have been studied for a long time. However, little is known about their interactions with soil physicochemical properties and bacterial communities. This study performed high-throughput sequencing of the 16S rRNA V3-V4 variable regions of soil bacteria in the three concentric zones of chosen FR, namely, the ON zone, on the ring; IN zone, inside the ring; and OUT zone, outside the ring. Also, the change in physicochemical properties and enzyme activities of the soil were determined. This study found that the nutrients and enzyme activities on the ring were higher than inside and outside of the ring. The activities of microorganisms were frequent and the plant grew splendidly. The bacterial species diversity was the lowest on the ring with the main genera Pseudonocardia, Streptosporangium, Kribbella and Promicromonospora. The imbalance of the microbial community structure at different ring zones may be the driving factor for the continuous outward expansion of FRs. Soil available phosphorus, electrical conductivity, total nitrogen and organic matter positively correlated with the distribution of FR soil bacteria.

Background: Verticillium dahliae, the causal agent of Verticillium wilt, is notoriously invasive in many crops and has been involved in numerous epidemics worldwide. Bacillus species, as representatives of biocontrol bacteria, produce a variety of lipopeptides (LPs), which are useful as biofungicides to many pathogenic fungi, including Verticillium dahliae. This study will explore the mechanism of resistance of V. dahliae to Bacillus and biocontrol bacteria.Results: By using in vitro confrontation bioassays, we found that under the stress induced by Bacillus, the spore vitality of V. dahliae with larger colonies was higher, and more abundant microsclerotia were formed. Then, according to the RNA-Seq analysis, the target of rapamycin (TOR) and mitophagy pathways were enriched among the significantly upregulated 542 genes observed in two co-culture groups with different colony sizes. In addition, in the group of V. dahliae with large colonies, the pathways related to cell wall synthesis, microsclerotia formation and the clearance of reactive oxygen species were regulated, and the expression of genes was up-regulated.Conclusion: This study found that the larger colonies of V. dahliae were more resistant to the antagonistic actions of Bacillus and the likelihood of the formation of homeostasis. Therefore, the prevention of Verticillium wilt by Bacillus is more effective than the treatment of an active fungal infection. These transcriptomic insights provide direction for the use of fungicides in the prevention and treatment of diseases such as Verticillium wilt.

The frequently observed forest decline in water-limited regions may be associated with impaired tree hydraulics, but the precise physiological mechanisms remain poorly understood. We compared hydraulic architecture of Mongolian pine (Pinus sylvestris var. mongolica) trees of different size classes from a plantation and a natural forest site to test whether greater hydraulic limitation with increasing size plays an important role in tree decline observed in the more water-limited plantation site. We found that trees from plantations overall showed significantly lower stem hydraulic efficiency. More importantly, plantation-grown trees showed significant declines in stem hydraulic conductivity and hydraulic safety margins as well as syndromes of stronger drought stress with increasing size, whereas no such trends were observed at the natural forest site. Most notably, the leaf to sapwood area ratio (LA/SA) showed a strong linear decline with increasing tree size at the plantation site. Although compensatory adjustments in LA/SA may mitigate the effect of increased water stress in larger trees, they may result in greater risk of carbon imbalance, eventually limiting tree growth at the plantation site. Our results provide a potential mechanistic explanation for the widespread decline of Mongolian pine trees in plantations of Northern China.

Direction of Arrival (DOA) estimation is of big importance in Sonar, Radar and satellite systems. In this work, we estimated the 2-D DOA (two-dimensional DOA) of coherent wide-band sources by using a broadband focusing method, which was based on the separation of angle and frequency of the array manifold matrix. At first, we transform the element position of the plane array into the spherical coordinates. Then we adopted a generalized Fourier series expansion based on the spherical harmonics and the first kind of spherical Bessel functions to get the arbitrary plane array steering vector. In this vector, the angle and frequency of array manifold matrix was separated, so we could get the matrix of focusing transformation. Finally, we used the traditional narrow-band algorithm to estimate the 2-D DOA. The design is suitable for arbitrary array geometry models of coherent broadband signals, and avoids the angle pre-estimation in traditional methods. Simulation results demonstrate its validity and better performance.

Ommochrome is the major source for eye coloration of all insect species so far examined. Phenoxazinone synthetase (PHS) has always been regarded as the terminal step enzyme for ommochrome formation, which is encoded by cardinal or karmoisin genes. Our previous study indicated that the karmoisin ortholog gene (Nl-karmoisin) product in the brown planthopper (BPH) was a monocarboxylate transporter, while not a PHS. Here, based on full-length complementary DNA, the cardinal ortholog gene in BPH (Nl-cardinal) product was predicted to be a haem peroxidase rather than a PHS. We suggest for the first time that neither karmoisin nor cardinal encodes the PHS, but whether PHS participates in BPH eye pigmentation needs further research. Nymphal RNA interference (RNAi) experiments showed that knockdown Nl-cardinal transcript led the BPH ocelli and compound eye to color change from brown to red, while knockdown Nl-karmoisin only made the ocelli present the red phenotype. Notably, not only the Nl-cardinal transcript, dscd injection (Nl-cardinal targeting double-stranded DNA (dsRNA)) also significantly reduced the Nl-karmoisin transcript by 33.7%, while dska (Nl-karmoisin targeting dsRNA) injection did not significantly change the Nl-cardinal transcript. Considering the above RNAi and quantitative real-time polymerase chain reaction results, we propose that Nl-cardinal plays a more important role in ommochrome synthesis than Nl-karmoisin, and it may be an upstream gene of Nl-karmoisin. The present study suggested that both karmoisin and cardinal ortholog genes play a role in ommochrome synthesis in a hemimetabolous insect.

To examine the effects of dietary lipids on the growth and feed utilization of jade perch juveniles, Scortum barcoo, diets containing 36.3% crude protein supplemented with increasing lipid levels (6, 9, 12, and 15% of the dry matter) were used to feed triplicate groups of 30 fish for 60 d. At the end of the experiment, more than 95% fish survived well from all diet groups (P > 0.05). Measurements on the weight gains and the daily specific growth rates indicated that fish fed with diets of 12 and 15% lipids exhibited higher growth rates (P 0.05). Together, increasing lipid levels in fish diets was effective to improve fish growth, feed efficiency, and protein utilization.

To improve the diagnosticability of selective enteroclysis in the area of intestinal obstruction, by reviewing the cases of selective intestinal enteroclysis through a naso-intestinal decompression tube, and evaluating its diagnostic concordance level and the influence factors.The selective enteroclysis was different from traditional enteroclysis; it utilized a naso-intestinal decompression long tube to perform local intestinal opacification. In the four years between Apr. 2008 and Apr. 2012, 98 cases of selective intestinal enteroclysis were analyzed retrospectively, and diagnostic concordance value was made between the radiologic diagnosis and the final clinical diagnosis recorded in the medical history according to an evaluating criterion. Five scores were used in the evaluating system: 0 meant that the enteroclysis had no valuable information to provide; 0.25 meant that the enteroclysis could prompt the occurring of obstruction, but could not provide the information of location and cause; 0.50 meant that the enteroclysis could find the location of obstruction, but could not determine the cause; 0.75 meant that the enteroclysis could provide some valuable analysis of the causes of the obstruction, and very close to the final clinical diagnosis; 1.00 meant that the complete concordance between the enteroclysis diagnosis and the final clinical diagnosis. The influence factors would also be considered to improve the competence of the selective enteroclysis in the diagnosis of intestinal obstruction.In the 98 cases, 53.1% had definite abdominal operative history. The most common cause of obstruction was conglutination between intestinal loops when discharged from hospital, almost occupying 50%. In this group of cases, the most common types were ileum obstruction (47.6%), multiple location obstruction (41.7%) and incomplete obstruction (59.5%). Conglutination and stricture of the intestinal was the common radiologic appearance (61.9%). There were 50 cases with higher concordance scoring 0.75 or 1.00, in comparison, there were 25 cases with lower concordance scoring 0 or 0.25. The difference between the two groups had statistical significance.As a combination of traditional enteroclysis and naso-intestinal decompression tube, selective intestinal enteroclysis could exert higher diagnostic ability than that of traditional enteroclysis and also could break through the limitations of traditional enteroclysis in the condition of intestinal obstruction. This method has higher diagnostic concordance and could provide valuable information in obstruction location, extent, severity and possible causes. The main factor influencing the effectiveness of the examination is the location of the decompression tube and dynamic observation would be very helpful and important.

It is widely accepted that intersexual differences occur in cognitive domains, e.g., in spatial learning and memory. The hippocampus plays important roles in the consolidation of information from short-term memory to long-term memory and spatial navigation. However, it still remains unknown whether the hippocampal proteomic profiling differs between males and females. In this study, we investigated the intersexual differences in protein expression of hippocampi using the two-dimensional electrophoresis analysis. In all, 33 differentially expressed proteins were characterized by matrix-assisted laser desorption and ionization time-of-flight mass spectrometry and validated by Western-blotting analysis. In line with Western-blotting validation, the proteomic identification revealed the overexpression of glial fibrillary acidic protein in female rats' hippocampi, and the overexpression of both creatine kinase B-type and DRP-2 in male rats' hippocampi. The intersexual differences in hippocampal proteomic profiling are probably closely related to those in spatial learning and memory abilities.

To investigate the expression of apoptosis-related protein Bcl-w in adenocarcinoma of the small intestine, and the apoptotic effect of Bcl-w siRNA on small intestinal adenocarcinoma cells HuTu-80.Forty-two tissue samples were examined in our study, including 7 cases from human small intestinal adenocarcinoma, and 35 cases from normal small intestine served as control. The expression of Bcl-w was detected by immunohistochemistry (IHC). Western blot analysis was performed to confirm whether Bcl-w siRNA could effectively down-regulate Bcl-w protein after HuTu-80 cells were transfected with Bcl-w siRNA. The cells were treated with chemotherapeutic agent 5-Fu to observe whether Bcl-w protein-silecing affects the pro-apoptotic effect of 5-Fu. Flow cytometry analysis was used for assessment of apoptotic rate of HuTu-80 cells cultured with Bcl-w siRNA alone, with 5-Fu alone, and with combination of Bcl-w siRNA and 5-Fu, using untreated HuTu-80 cells as control.The positive rate of Bcl-w expression was significantly higher in small intestinal adenocarcinoma than that in normal tissue (85.7% vs. 25.7%, P=0.005). Compared with the control group, Bcl-w siRNA transfection effectively down-regulated the expression of Bcl-w protein (P0.05). The apoptosis in HuTu-80 cells was not increased significantly in Bcl-w-/-cells compared with that of control group (12.4±2.2)% vs. (8.6±1.7)% (P0.05). However, compared with the 5-Fu group, the apoptosis in HuTu-80 cells was effectively enhanced after combination treatment with Bcl-w siRNA and 5-Fu (45.7±2.1)% vs. (71.6±3.2)% (P0.05).Bcl-w protein plays a significant role in the carcinogenesis of human small intestinal adenocarcinoma. Down-regulation of Bcl-w protein in small intestine adenocarcinoma HuTu-80 cells leads them susceptible to 5-Fu.

Given that the studies on human small intestinal stromal tumors are very limited, our study was designed to determine the significance of apoptosis, the role of some apoptosis-related proteins, including Bax, Bad, Bid, Bcl-2, Bcl-Xl, and Bcl-W, in jejunal stromal tumors (JST). For this purpose, 52 samples were examined by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling and immunohistochemistry, including 40 cases from normal jejunum and 12 cases from the JST. The results showed that a significantly decreased apoptosis was observed in JST compared with normal jejunum (median of apoptotic index, 0.13% vs 15.17%, p0.001). In univariate analysis, statistically significant differences were shown in percentage of positive-expression of Bid (0.0% vs 77.5%, p0.001), Bcl-2 (91.7% vs 42.5%, p = 0.003), and Bcl-Xl (75.0% vs 35.0%, p = 0.014), but not in Bax, Bad, and Bcl-W. Moreover, Bcl-Xl was an independent parameter associated with JST by multivariate Logistic analysis. For the role of ratios of anti-apoptotic proteins to pro-apoptotic proteins, Bcl-Xl/Bid ratio was the most preferable index. In conclusion, decreased apoptosis and expression of Bid, increased level of Bcl-Xl may play some important roles in human JST, and Bcl-Xl/Bid ratio may be as a new potentially associated index, although they should be validated in more studies.

To investigate the roles of STAT1 and STAT2 in growth inhibition induced by phosphatidylethanolamine (PE) in human hepatoma HepG2 cells.The growth of HepG2 cells exposed to 0.125, 0.25, 0.5 and 1.0 mmol/L PE was assessed by MTT assay, and the expressions of STAT1 and STAT2 were analyzed using immunocytochemical assay.PE inhibited the growth of HepG2 cells in a dose-dependent manner and increased the expression of STAT1 and STAT2 in comparison with those in the control group. AG490, an inhibitor of JAKs, partially reversed PE-induced growth inhibition of HepG2 cells.STAT1 and STAT2 are involved in the growth inhibition of human hepatoma HepG2 cells induced by PE.

The IR (3200–30 cm−1) spectra have been recorded for S-methyl-N,N-dimefhylthiocarbamate, (CH3)2NC(O)SCH3, and its isotopomers, S-d3, N-d6 and N-d9, for the gas and liquid. Additionally, the Raman spectra (3200–10 cm−1) for the solid and liquid, with qualitative depolarization ratios, have been obtained for all the isotopes. These data are interpreted on the basis that the s-cis conformer (the S-methyl group oriented eis to the carbonyl group) with Cs symmetry is the only form existing in all three phases for this molecule. A complete vibrational assignment proposed for the -d0 molecule is facilitated by the availability of spectral data for five different isotopomers. A normal coordinate analysis has been carried out utilizing ab initio calculations with the 3–21G* basis set. The potential energy distributions and ab initio calculated frequencies have allowed a clarification of some of the corresponding results obtained from experiment. Structural optimizations and potential surface scan have also been carried out by ab initio calculations with the 3–21G* basis set. These results are compared with some previous studies on this molecule as well as on similar molecules.

Because different authors construct composite torsional coordinates differently, torsional force constant values reported for the same or very similar internal rotors differ from each other by as much as one or occasionally two orders of magnitude. Since these coordinates are not always precisely defined, the source of the discrepancies cannot always be delineated. Examples are given which show how the recommended torsional coordinate (IUPAC Commission on Molecular Spectroscopy) ensures that the G and F matrix values for the same internal rotor in different surroundings stay comparable. The other types of torsional coordinates which are frequently used lead to inconsistent results even with the same molecule. A similar problem can also exist for the twisting coordinate.

Apoptosis is a physiological process that an organism selectively eliminates cells that are no longer needed, or have been damaged, or are dangerous. Bcl-xL, an important member of the Bcl-2 family that plays indispensable roles in regulating cell survival and apoptosis, is frequently over-expressed in various kinds of human cancers. The inhibition of this molecule is associated with decreased tumorigenesis and resistance to conventional chemotherapy. This article briefly reviews some progresses in the study of Bcl-xL in the past few years.

The far infrared spectrum of gaseous 2‐fluoropropenoyl fluoride CH2CFCFO, has been recorded at a resolution of 0.10 cm−1 in the 350–35 cm−1 region. The asymmetric torsional fundamental of the more stable s‐trans (two double bonds oriented trans to one another) and the high energy s‐cis conformations have been observed at 84.1 and 68.5 cm−1, respectively, each with excited states falling to lower frequencies. From these data, the asymmetric torsional potential function governing internal rotation about the C–C bond has been determined. The potential coefficients are V1=−201±4, V2=2106±16, V3=382±6, V4=−41±5, and V5=−74±3 cm−1. The s‐trans to s‐cis and s‐cis to s‐trans barriers have been determined to be 2232 and 2124 cm−1, respectively, with an enthalpy difference between the conformations of 108±26 cm−1 (309±74 cal/mol). From variable temperature studies of the Raman spectrum, the conformational enthalpy difference has been determined to be 187±39 cm−1 (535±112 cal/mol) and 370±82 cm−1 (1058±234 cal/mol) ...
