Abstract

Over the last decades, impressive progresses have been made in many experimental domains, e.g. microscopic techniques such as single-particle tracking, leading to plethoric amounts of data. In a large variety of systems, from natural to socio-economic, the analysis of these experimental data conducted us to conclude about the omnipresence of power-laws. For example, in living systems, we are used to observing anomalous diffusion, e.g. in the motion of proteins within the cell. However, estimating the power-law exponents is challenging. Both technical constraints and experimental limitations affect the statistics of observed data. Here, we investigate in detail the influence of two essential constraints in the experiment, namely, the temporal-spatial resolution and the time-window of the experiment. We study how the observed distribution of an observable is modified by them and analytically derive the expression of the power-law distribution for the observed distribution through the scope of the experiment. 10. Blood-Based Nanoparticle-Enhanced Quaking-Induced Conversion (Nano-QuIC): Inhibitor-Resistant Detection of Seeding Activity in Patients Diagnosed with Parkinson’s Disease

Abstract

A hallmark of α-synucleinopathies (e.g., Parkinson’s disease) is the misfolding and aggregation of α-synuclein in tissues and biological fluids. Protein amplification assays like real-time quaking-induced conversion (RT-QuIC) are sensitive yet currently limited to semi-invasive sample types such as cerebrospinal fluid because more accessible samples, such as blood, contain inhibitors. Here, we show that Nanoparticle-enhanced Quaking-induced Conversion (Nano-QuIC) can double the speed of reactions spiked with misfolded α-synuclein while increasing sensitivity 100-fold in human plasma. Nano-QuIC detected spike concentrations down to 90 pg/mL in lysed whole blood, while reactions without nanoparticles (RT-QuIC) failed to have any detection due to the presence of strong inhibitors. Moreover, Nano-QuIC showed increased seeding activity in plasma samples from Parkinson’s patients (n = 4) versus healthy controls (n = 4). This sets the groundwork for the noninvasive diagnostic use of Nano-QuIC, potentially enabling early disease detection and management through blood-based testing.

Abstract

The spread of coronavirus disease 2019 (COVID-19) is causing an unprecedented crisis around the world, with South Korea being no exception. South Korea experienced a surge of COVID-19 cases on February 19, 2020, but was able to flatten the COVID-19 curve in only 20 days without enforcing lockdown measures that restrict the freedom and movement of people. The actual field experience of the Korean government's COVID-19 response team, who participated in drafting the document "Tackling COVID-19: Health, Quarantine, and Economic Measures: Korean experience," was examined and an in-depth analysis was conducted of the South Korean government's experience dealing with the Middle East respiratory syndrome of 2015. Three prominent factors were identified behind Korea's responses against COVID-19. First, it was crucial that the government responded as 1 team efficiently to cooperate and respond to COVID-19. Second, a novel approach was needed to combat COVID-19 spread. Information and communication technology had a key role in South Korea's strategy to contain COVID-19. Third, an effective response to COVID-19 was possible due to the successful partnership between the general public and state authorities., (© The Author(s) 2020.
