A cataract is an eye disease that causes visual distortion and the late stage of this disease can lead to blindness. It is considered a silent disease that can occur without the appearance of symptoms. Therefore, the most effective way to detect cataracts is through accurate and timely detection to prevent hurting, expensive operations, and to stop blindness. The purpose of this paper is to propose an automated system based on the wavelet scattering network which categorizes the patients into four classes: early cataract, intermediate cataract, late cataract, and non-cataract conditions using 512 images of the ODIR dataset (212 Cataract and 300 of Normal). The first step in this technique is the preprocessing step for the retinal image was a mean filter, which was utilized to reduce the image's noise. The limited contrast adaptive histogram equalization (CLAHE) method was then employed to improve the image's contract level. Then, Low-variance characteristics can be extracted from image data using a wavelet scattering network for use in deep learning applications. In this network, lowpass scaling filters and predefined wavelets are employed. The average accuracy of the suggested method was 100% for four classes (Normal, Early, Moderate, and Severe). The results are promising compared with other similar works.

Di era teknologi saat ini, penggunaan dari Artificial Intelligence sangat berkembang pesat. Banyak teknologi yang sudah menerapkan AI untuk menghasilkan suatu informasi. Akan tetapi, tidak semua teknologi yang diciptakan menggunakan AI membawakan dampak positif. Salah satu teknologi AI yang dapat menimbulkan dampak negatif adalah deepfake. Salah satu dampak negatif yang dapat disebabkan oleh teknologi deepfake adalah membuat wajah manusia buatan yang menyerupai wajah aslinya. Oleh karena itu, deepfake merupakan salah satu kasus yang perlu diperhatikan lebih lanjut. Penggunaan deepfake yang salah ini apabila dibiarkan dapat merugikan banyak pihak, baik untuk orang lain maupun diri sendiri. Penelitian ini dibuat dengan tujuan agar dapat membuat sebuah model bersifat kopnseptual yang dapat melakukan identifikasi pada wajah asli maupun wajah buatan deepfake. Metode yang digunakan adalah metode CNN dengan pembuatan arsitektur 6 convolutional layer dan 3 max pooling serta tambahan layer Batch Normalization. Metode CNN dipilih karena dinilai baik dalam melakukan klasifikasi suatu citra. Setelah itu, metode CNN akan ditambahkan dengan arsitektur menyerupai VGG dan layer tambahan yaitu Batch Normalization. Hasil yang didapatkan pada penelitian ini adalah model yang dibuat dapat dinyatakan berhasil untuk melakukan identifikasi wajah buatan deepfake dengan wajah asli. Hal ini dibuktikan dengan tingkat akurasi, presisi, recall, dan F1-Score sebesar 91%.

The vision system of humanoid robots is one of the essential components that allow them to do complex activities. This has led to numerous studies on artificial gaze stabilization, many of which are based on biological vision system reflexes. The gaze stabilization solution includes image capture and processing, inverse kinematics, and feedback control to achieve the desired gaze behavior. Generally, the multi-solution problem of finding the desired gaze direction and the inverse kinematics problem of finding the desired joints are solved by using cost functions and numerical solvers. This results in disadvantages that include long processing time, uncertain number of iterations, and risk of numerical instability. In this work, an alternative algebraic method is introduced by exploiting the cascading structure of the neck and eye. By using direct equations instead of numerical solving, the multi-solution and inverse kinematics problems are solved utilizing the geometrical structure and the proposed virtual configurations. Moreover, a sliding mode controller is designed to move the neck-eye joints to their desired positions. The developed scheme is simulated in the MATLAB/SIMULINK environment. Results for various scenarios show that the system can effectively gaze at different stationary and moving targets, showing human-like gaze behavior. In terms of timing, the desired gaze direction is quickly achieved and stabilized, whereas the neck-eye system dynamics stay longer. For one of the simulation cases presented, it took 0.3 seconds to achieve the desired gaze direction for the dynamics of more than 1 second.

Color information is useless for distinguishing significant edges and features in numerous applications. In image processing, a gray image discards much-unrequired data in a color image. The primary drawback of colour-to-grey conversion is eliminating the visually significant image pixels. A current proposal is a novel approach for transforming an RGB image into a grayscale image based on singular value decomposition (SVD). A specific factor magnifies one of the color channels (Red, Green, and Blue). A vector of three values (Red, Green, Blue) of each pixel in an image is decomposed using SVD into three matrices. The norm of the diagonal matrix was determined and then divided by a specific factor to obtain the grey value of the corresponding pixel. The contribution of the proposed method gives the user high flexibility to produce many versions of gray images with varying contrasts, which is very helpful in many applications. Furthermore, SVD allows for image reconstruction by combining the weighting of each channel with the singular value matrix. This results in a grayscale image that more accurately captures the actual intensity values of the image and preserves more color information than traditional grayscale conversion methods, resulting in loss of color information. The proposed method was compared with a similar method (converting the color image into grayscale) and was found to be the most efficient.

Brucellosis is an endemic zoonotic disease caused by intracellular gram-negative coccobacilli called Brucella. This infectious disease may implicate many farm animals and is transmissible to exposed humans. Brucellosis is potentially life-threatening and may lead to granulomatous multi-organ involvement with tendency to chronicity and recurrence. The treatment of brucellosis requires combined and protracted antimicrobial therapies to eliminate the disease and to avoid its relapse. Genitourinary brucellosis is common among infected humans in endemic areas and is considered the second-most affected focal site, which commonly manifests as epididymo-orchitis. Testicular abscess, however, is an extremely rare complication of brucellosis. To the best of our knowledge, in the literature, there are to date only 16 previously published case reports, including 22 patients of brucellar testicular abscesses, emphasizing the extreme rarity of this condition. Most of these cases harbored small abscesses, which were treated conservatively using antibiotics therapy only, or with added drainage of the abscesses. Larger abscesses were reported to necessitate orchiectomy. In some cases, the abscesses were mimicking tumors, and surgeries for orchiectomy were done accordingly. A summary of the previously reported cases in the literature is presented. Here, we present the 17th case report of a 34-year-old man with a right-side huge multilocular brucellar testicular abscess apparently replacing the entire testicle, who was successfully treated with organ-sparing management by incision-drainage of the large abscess with antibiotics, to eventually preserve his testis. In conclusion, brucellosis should be considered among the differential diagnoses of any testicular swelling, especially in endemic areas. Drainage of brucellar testicular abscess with appropriate medical treatment is feasible and may preserve the testicle, even with large abscess apparently replacing the entire testicle.

Background: Doxorubicin (Dox) is a potent chemotherapeutic agent, but its usage is limited by dose-dependent cardiotoxicity. Intracellular calcium dysregulation has been reported to be involved in doxorubicin-induced cardiomyopathy (DICM). The cardioprotective role of RyR stabilizer dantrolene (Dan) on the calcium dynamics of DICM has not yet been explored. Objective: To evaluate the effects of dantrolene on intracellular calcium dysregulation and cardiac contractile function in a DICM model. Methods: Adult male C57BL/6 mice were randomized into 4 groups: (1) Control, (2) Dox Only, (3) Dan Only, and (4) Dan + Dox. Fractional shortening (FS) and left ventricular ejection fraction (LVEF) were assessed by echocardiography. In addition, mice were sacrificed 2 weeks after doxorubicin injection for optical mapping of the heart in a Langendorff setup. Results: Treatment with Dox was associated with a reduction in both FS and LVEF at 2 weeks (P < .0001) and 4 weeks (P < .006). Dox treatment was also associated with prolongation of calcium transient durations CaTD50 (P = .0005) and CaTD80 (P < .0001) and reduction of calcium amplitude alternans ratio (P < .0001). Concomitant treatment with Dan prevented the Dox-induced decline in FS and LVEF (P < .002 at both 2 and 4 weeks). Dan also prevented Dox-induced prolongation of CaTD50 and CaTD80 and improved the CaT alternans ratio (P < .0001). Finally, calcium transient rise time was increased in the doxorubicin-treated group, indicating RyR2 dyssynchrony, and dantrolene prevented this prolongation (P = .02). Conclusion: Dantrolene prevents cardiac contractile dysfunction following doxorubicin treatment by mitigating dysregulation of calcium dynamics.

We present an interesting case and images of a patient with identified as having Brugada syndrome due to dynamic electrocardiographic (ECG) changes seen during high-grade fever in the context of an associated COVID-19 infection. We show serial ECGs demonstrating dynamic but unusual change from a Brugada Type-1 to a non-Type-1 Brugada pattern, then ultimately having ECG normalisation once the fever had been corrected with antipyretic medication. This case and set of images illustrate how ECG was helpful in successfully identifying a COVID-19 patient with underlying Brugada syndrome, thus enabling simple but appropriate management and risk-stratification.

The technological development in the field of information and communication has been accompanied by the emergence of security challenges related to the transmission of information. Encryption is a good solution. An encryption process is one of the traditional methods to protect the plain text, by converting it into inarticulate form. Encryption implemented can be occurred by using some substitute techniques, shifting techniques, or mathematical operations. This paper proposed a method with two branches to encrypt text. The first branch is a new mathematical model to create and exchange keys, the proposed key exchange method is the development of Diffie-Hellman. It is a new mathematical operations model to exchange keys based on prime numbers and the possibility of using integer numbers. While the second branch of the proposal is the multi-key encryption algorithm. The current algorithm provides the ability to use more than two keys. Keys can be any kind of integer number (at least the last key is a prime number), not necessarily to be of the same length. The Encryption process is based on converting the text characters to suggested integer numbers, and these numbers are converted to other numbers by using a multilevel mathematical model many times (a multilevel process depending on the number of keys used), while the decryption process is a one-level process using just one key as the main key, while the other keys used as secondary keys. The messages are encoded before encryption (coded by ASCII or any suggested system). The algorithm can use an unlimited number of keys with a very large size (more than 7500 bytes), at least one of them a prime number. Exponentiation is also used for keys to increase complexity. The experiments proved the robustness of the key exchange protocol and the encryption algorithm in addition to the security. Comparing the suggested method with other methods ensures that the suggested method is more secure and flexible and easy to implement.

Mobile communication systems are always in continuous evolution due to the demands of the end-users using this technology. Therefore, before the possible launch of 5G, some technologies have opened the way to the new mobile communication system. The need for technologies that provide more comfort to users has led to the construction of complex communication systems that were only science fiction decades ago. The information society in which we are now immersed has been the result of constant progress over time. In this paper, a survey of multiple access schemes for next-generation wireless communication systems is presented. Multiple access schemes are reviewed for possible use in next-generation wireless communication systems such as orthogonal multiple access (OMA), non-orthogonal multiple access (NOMA), and delta-orthogonal multiple access (D-OMA), etc. General comparisons of 1G to 6G are presented. Different types of OMA are explained, and then orthogonal frequency division multiple access (OFDMA) is chosen as an example of the OMA scheme to compare with NOMA and D-OMA. There are two types of NOMA: power-domain and code-domain, which are discussed and compared. Simulation results are presented, and a comparison among different access schemes is provided.

Purpose: Most common viruses causing ocular infections are Herpes Simplex Viruses (HSV) type 1 and type 2, Cytomegalovirus (CMV), Varicella-zoster Virus (VZV), and few strains of Adenovirus. Diagnosis of these infections through clinical manifestations and using conventional methods has a number of limitations. The purpose of this study was to develop a multiplex Polymerase Chain Reaction (PCR) for simultaneous detection of all pathogenic viruses from ocular infections. Methods: Ten uniplex PCRs were standardized, two each for HSV type 1 (HSV-1) and type 2 (HSV-2), CMV, VZV, and Adenovirus. Various multiplexing combinations of above PCRs were put to finalize targets and reaction conditions enabling diagnosis of all in a single reaction. The uniplex and multiplex PCRs were run for known positive and negative controls, and samples from clinically suspected patients and healthy controls. Results: Out of the 170 samples from suspected ocular infections, 24.7% were positive by uniplex PCR and 22.9% were correctly identified by multiplex PCR. None of the samples negative by uniplex PCRs was positive by the multiplex PCR. The sensitivity and specificity of multiplex PCR compared to the commonly used uniplex PCRs as gold standard was 92.86% and 100%, respectively. The prevalence of different viral pathogens was 13.5% for HSV-1, followed by 5.9% for Adenovirus, 2.4% for VZV, 1.8% for HSV-2, and 1.2% for CMV. Conclusion: The establishment of multiplex PCR has found immediate application in diagnosing ocular viral pathogens in a single reaction, thus saving time, manpower, and resources by fivefold.

Magnesium (Mg) is an essential plant macronutrient responsible for modulating many physiological or biochemical processes such as photosynthetic activity, amino acid synthesis and nucleotide metabolism. Agricultural soils with a more-than-adequate availability of calcium (Ca) have inherent Mg deficiency, potentially resulting in overall reduced soil productivity and crop yield potential. We conducted a field experiment to investigate the optimum soil application of Mg to increase crop growth and productivity under calcareous soil conditions. In addition to recommended soil application of mineral fertilizers, we applied the following four levels of Mg to the soil in the form of anhydrous MgSO4: control, 4 kg Mg ha−1 (Mg4), 8 kg Mg ha−1 (Mg8) and 16 kg Mg ha−1 (Mg16). Results showed that Mg16 application enhanced the plant height (21%), number of grains (18%), 1,000 grains weight (20%), grain yield (20%) and biological yield (9%) over control (p ≤ 0.05). Chlorophyll a, chlorophyll b and total chlorophyll were generally higher at the Mg8 and Mg16 levels than at the control level. Contrasting to increases in growth traits, the concentration of K significantly decreased in grains, leaves and shoots of maize along the soil’s Mg gradient (p ≤ 0.05). We suggest that Mg16 overcomes the deficiency of soil Mg and can increase the crop yield traits in calcareous soils. More investigations of the effect of soil Mg on various crops grown in calcareous soils may add to our knowledge related to the stressing impact of soil Mg on plant K concentration.

Mango orchards in Pakistan are deficient in soil micronutrients. Multan is one of the prime regions for mango production in Pakistan; therefore, this study was conducted to evaluate the micronutrient status of mango orchards in the Multan region. Soil samples from four different depths (0–30, 30–60, 60–90, and 90–120 cm) and leaf samples were collected from thirteen different locations of Multan. Depth-wise variations in the micronutrient status and the levels of pH, EC, CEC, SOM, and CaCO3 were determined. All data collected from the field and laboratory work of mango orchards under study were analyzed statistically by applying the RCBD design. It was observed that pH and ECe of soil under study were significantly higher in upper depths when compared with lower depths whereas CaCO3 content was contrary to pH and EC as it was observed to be higher from the lower depth of the soil. Moreover, mango leaves from the majority of locations were deficient in total micronutrients due to poorly available micronutrients status of the soil. Thus, there is a serious need to improve the chemical properties of the soil, and the proper dose of micronutrients should be applied every season for sufficient supply throughout the growing cycle of mango in and around the Multan region.

ObjectivesTo quantify SARS-CoV2 IgG antibody titers over time and assess the longevity of the immune response in a multi-ethnic population setting.SettingThis prospective study was conducted in a tertiary hospital in Abu Dhabi city, UAE, among COVID-19 confirmed patients. The virus-specific IgG were measured quantitatively in serum samples from the patients during three visits over a period of 6 months. Serum IgG levels ≥15 AU/ml was used to define a positive response.Participants113 patients were analyzed at first visit, with a mean (SD) age of participants of 45.9 (11.8) years 87.5% of the patients were men. 63 and 27 participants had data available for visits 2 and 3, respectively.Primary outcomeChange in SARS-CoV2 IgG antibody titers over the visits.ResultsNo mortality or re-infection were reported. 69% of the patients developed positive IgG response within the first month after the onset of symptoms. The levels of IgG showed a consistent increase during the first three months with a peak level during the third month. Increasing trend in the levels of IgG were observed in 82.5%, 55.6% and 70.4% of patients between visit 1 to visit 2, visit 2 to visit 3, and from visit 1 to visit 3, respectively. Furthermore, about 64.3% of the patients showed sustained increase in IgG response for more than 120 days.ConclusionsOur study indicates a sustained and prolonged positive immune response in COVID-19 recovered patients. The consistent rise in antibody and positive levels of IgG titers within the first 5 months suggest that immunization is possible, and the chances of reinfection minimal.

In the current analysis, the effects of circumferential scratches along the inner surface of a 170ᵒ -arc partial journal bearing has been numerically investigated. Their impact on the thermo-elasto-hydrodynamic performance characteristics, including maximum pressure, temperature, deformation, and stress, has been examined thoroughly. The ANSYS Fluent CFD commercial code was employed to tackle the iterative solution of flow and heat transfer patterns in the fluid film domain. They are then applied to the ANSYS Static Structure solver to compute the deformation and stress resulted in the solid bearing zone. A wide range of operating conditions has been considered, including the eccentricity ratio ( ) and scratch depth ( ). In contrast, the bearing length-diameter ratio (L/D) and the rotation speed (N) have been fixed at 0.77 and 1500 rpm, respectively. The thermo-hydrodynamic pressure, temperature, stress, and deformation have all been computed. It was found that the scratch depth has a direct effect on the thermo-hydrodynamic performance of the partial bearings. Meanwhile, the deep central scratches are important, especially at scratch depth equal to 0.224 mm.

Time synchronization is a cooperative work's foundation among underwater sensor networks' nodes; it takes a crucial role in the application and the study of underwater sensor networks. Several time synchronization protocols have been presented for terrestrial radio networks, but they cannot be instantly utilized to real-time underwater sensor networks due to the slow propagation speed of the underwater acoustic signals, the mobility among sensor nodes, and the energy limitation in the underwater wireless sensor networks. In this paper, we propose an adaptive power-efficient time synchronization for mobile underwater sensor networks, called APE-Sync. The proposed scheme takes into account the time-varying clock skew and combines the Doppler-Enhanced synchronization protocol (DE-Sync) and the Kalman filter tracking the clock skew to achieve time synchronization. The proposed scheme also helps in reducing the energy consumption of the nodes by guaranteeing accurate time synchronization. The simulation results are presented, which show that APE-Sync outperforms existent time synchronization schemes in both power efficiency and precision.

This article looks into solutions offered by the international community to the countries suffering from a constant influx of refugees and which require international assistance. Refugee migration is a type of forced migration. Since the introduction of international refugee law, the international community has been using one of the three solutions to the refugee problem: voluntary repatriation, local integration and resettlement in a third country. The article regards resettlement in a third country as the main solution to the global refugee problem, which should be used on a large scale. The article also reviews the cases of interference in the developing countries’ internal affairs and the role of such interference in aggravating the refugee problem. Despite the benevolence of the intentions, such as the protection of the refugees’ human rights, this intervention demonstrated the opposite effect in certain cases in Iraq, Afghanistan and Libya. Consequently, the author touches upon the importance of the peaceful ways in solving and preventing the problem of refugees. Finally, the article points out the international community’s new approach to the problem: integration of development projects and refugee support programs in countries needing this type of assistance.

Members of the genus Pulicaria have been used in traditional medicine for alleviating several complaints as they have a rich pool of biometabolites. Acute lung injury (ALI) is a serious disease with an elevated mortality rate. The present investigation aimed to evaluate the total phenolic and flavonoid contents and antioxidant capacity of the methanolic extract of P. petiolaris Jaub. and Spach. (PP) (Asteraceae). Moreover, the potential protective potential of PP against lipopolysaccharide-(LPS)-induced ALI was assessed. PP is a rich source of phenolics and flavonoids. The total phenolic content (TPC) was 68.05 mg gallic acid equivalent (GAE)/g dried extract, and the total flavonoid content (TFC) was 45.86 mg quercetin equivalent (QE)/g dried extract. Additionally, PP possessed a promising DPPH-scavenging activity, with an IC50=27 μg/mL. Our results showed that PP lessened LPS-induced lung injury. PP effectively reduced pulmonary edema as it lowered total protein and the lung wet/dry weight (W/D) ratio in the bronchoalveolar lavage fluid (BALF). It also significantly ameliorated the level of lactate dehydrogenase (LDH) in the BALF and improved the histopathological lesions in the lung tissue. LPS-induced inflammatory cell infiltration was greatly depressed in PP-treated animals. PP showed antioxidant capacity as it reduced the LPS-induced increase in the lipid peroxidation marker, malondialdehyde (MDA). It also increased the activity of superoxide dismutase (SOD) and the content of reduced glutathione (GSH). This study indicates that PP significantly decreased LPS-induced inflammation in the LPS-mediated ALI murine model, suggesting that it may become a significant preventive strategy for treating nonspecific inflammation of the lungs.

El terrorismo unido al extremismo es un fenómeno global, del cual las Maldivas no tuvieron experiencia hasta el año 2007. Campañas terroristas sostenidas y repetidas pueden hacer descarrilar la economía de un país, que depende principalmente del turismo y las industrias relacionadas. Un repaso a la literatura y un análisis de las estadísticas muestran que la industria del turismo es a la vez vulnerable y resistente. Incidentes tales como los del atentado en el Parque Sultan Malé y el enfrentamiento en la Mezquita Himandhoo en las Maldivas sugieren que la sociedad está perdiendo su naturaleza tolerante y moderada hacia otras culturas. Estos sucesos implican la necesidad de desarrollar medidas políticas para hacer frente al extremismo en el país. Para un determinado grupo terrorista la industria turística de las Maldivas es un objetivo fácil. Se puede atacar o hacer rehenes en hoteles o a bordo de un crucero. Tal incidente puede provocar miedo entre los turistas y dañar la imagen asociada con las Maldivas. Como el terrorismo y el extremismo tienen una dimensión internacional, las soluciones sólo pueden alcanzarse a través de la cooperación global. A nivel local la ideología o el pensamiento que lleva a extremismo debería ser desafiado y moderado.

Abstract Salmonella Typhi is a human restricted pathogen with a significant number of individuals as asymptomatic carriers of the bacterium. Salmonella infection can be effectively controlled if a reliable method for identification of these carriers is developed. In this context, the availability of whole genomes of carrier strains through high- throughput sequencing and further downstream analysis by comparative genomics approaches is very promising. Herein we describe the genome sequence of a Salmonella Typhi isolate representing an asymptomatic carrier individual during a prolonged outbreak of typhoid fever in Kelantan, Malaysia. Putative genomic coordinates relevant in pathogenesis and persistence of this carrier strain are identified and discussed.

Abstract Background By measuring the quality of the organisation of palliative care with process and structure quality indicators (QIs), patients, caregivers and policy makers are able to monitor to what extent recommendations are met, like those of the council of the WHO on palliative care and guidelines. This will support the implementation of public programmes, and will enable comparisons between organisations or countries. Methods As no European set of indicators for the organisation of palliative care existed, such a set of QIs was developed. An update of a previous systematic review was made and extended with more databases and grey literature. In two project meetings with practitioners and experts in palliative care the development process of a QI set was finalised and the QIs were categorized in a framework, covering the recommendations of the Council of Europe. Results The searches resulted in 151 structure and process indicators, which were discussed in steering group meetings. Of those QIs, 110 were eligible for the final framework. Conclusions We developed the first set of QIs for the organisation of palliative care. This article is the first step in a multi step project to identify, validate and pilot QIs.

Abstract Mycobacterium avium subsp. paratuberculosis (MAP) is a zoonotic pathogen, a very slow growing bacterium which is difficult to isolate and passage in conventional laboratory culture. Although its association with Johne’s disease or paratuberculosis of cattle is well established, it has been only putatively linked to Crohn’s disease in humans. Further, MAP has been recently suggested to be a trigger for other autoimmune diseases such as type-1 diabetes mellitus (T1DM). Recently, some studies have indicated that exposure to MAP is associated with elevated levels of antibodies against MAP lysate although the exact mechanism and significance of the same remains unclear. Further, the cytokine profiles relevant in MAP associated diseases of humans and their exact role in the pathophysiology are not clearly known. We performed in vitro cytokine analyses after exposing different cultured human cells to the whole cell lysate of MAP and found that MAP lysate induces secretion of cytokines IL-1β, IL-6, IL-8, IL-10 and TNF-α by human peripheral blood mononuclear cells (PBMCs). Also, it induces secretion of IL-8 by cultured human stomach adenocarcinoma cells (AGS) and PANC-1(human pancreatic carcinoma cell line) cells. We also found that MAP lysate induced cytotoxicity in PANC-1cells. Collectively, these results provide a much needed base-line data set of cytokines broadly signifying a MAP induced cellular response by human cells.

Abstract Background In recent years, many studies have focused on adolescent's sex-related issues in China. However, there have been few studies of unmarried migrant females' sexual knowledge, attitudes and behaviors, which is important for sexual health education and promotion. Methods A sample of 5156 unmarried migrant female workers was selected from three manufacturing factories, two located in Shenzhen and one in Guangzhou, China. Demographic data, sexual knowledge, attitudes and behaviors were assessed by self-administered questionnaires. Multivariate logistic regression analysis was conducted to examine the factors associated with premarital sexual intercourse. Results The average age of the unmarried female workers included in the sample was 20.2 years, and majority of them showed a low level of sex-related knowledge. Females from the west of China demonstrated a significant lower level of sex-related knowledge than those from the eastern or central provinces (p < 0.05). Approximately 13% of participants held a favorable attitude towards premarital sexual intercourse, and youths from the east/central were more likely to have favorable attitudes compared with those from the west (p < 0.05). About 17.0% of the unmarried female workers reported having engaged in premarital sexual intercourse, and females from the east/central were more likely to have experienced premarital sexual intercourse than those from the west (p < 0.05). Multivariate analysis revealed that age, education, current residential type, dating, sexual knowledge, attitudes, and pattern of communication were significantly associated with premarital sexual intercourse. Conclusion The unmarried migrant female workers lack sexual knowledge and a substantial proportion of them are engaged in premarital sexual behaviors. Interventions aimed at improving their sexual knowledge and related skills are needed.

Abstract Type 1 diabetes mellitus (T1DM) is a multifactorial autoimmune disease in which the insulin producing β cell population is destroyed by the infiltrated T lymphocytes. Even though the exact cause of T1DM is yet to be ascertained, varying degree of genetic susceptibility and environmental factors have been linked to the disease progress and outcome. Mycobacterium avium subsp. paratuberculosis (MAP) is an obligate zoonotic pathogen that causes chronic infection of intestines in ruminants, the Johne's disease. MAP that can even survive pasteurization and chlorination has also been implicated to cause similar type of enteritis in humans called Crohn's disease. With the increasing recognition of the link between MAP and Crohn's disease, it has been postulated that MAP is an occult antigen which besides Crohn's could as well be thought to trigger T1DM. Epitope homologies between mycobacterial proteins (Hsp 65) and pancreatic glutamic acid decarboxylase (GAD 65) and infant nutrition studies implicate MAP as one of the triggers for T1DM. PCR and ELISA analyses in diabetic patients from Sardinia suggest that MAP acts as a possible trigger for T1DM. Systematic mechanistic insights are needed to prove this link. Unfortunately, no easy animal model(s) or in-vitro systems are available to decipher the complex immunological network that is triggered in MAP infection leading to T1DM.

Abstract Helicobacter pylori is a well known inhabitant of human stomach which is linked to peptic ulcer disease and gastric adenocarcinoma. It was recently shown in several studies that H. pylori can be harnessed as a surrogate marker of human migration and that its population structure and stratification patterns exactly juxtapose to those of Homo sapiens. This is enough a testimony to convey that H. pylori may have coevolved with their host. Several protective effects of H. pylori colonization have been considered as evidence of a presumed symbiotic relationship. Contrary to this assumption is the presence of a strong virulence apparatus within H. pylori; why a co-evolved parasite would try inflicting its host with serious infection and even causing cancer? The answer is perhaps embedded in the evolutionary history of both the bacterium and the host. We discuss a hypothetical scenario wherein H. pylori may have acquired virulence genes from donors within its environment that varied with change in human history and ecology. The H. pylori genomes sequenced to date portray fairly high abundance of such laterally acquired genes which have no assigned functions but could be linked to inflammatory responses or other pathogenic attributes. Therefore, the powerful virulence properties and survival strategies of Helicobacter make it a seasoned pathogen; thus the efforts to portray it as a commensal or a (harmless) 'bacterial parasite' need rethinking.

Abstract Anaerobic spirochaetes of the genus Brachyspira have long been known as important gut pathogens of pigs, but increasingly they are recognised as causing disease in birds and other animal species, including human beings. The genome sequence of the major swine pathogen Brachyspira hyodysenteriae was recently published, and this revealed extensive genome optimisation that leads to adaptation to the complex environment of the colon. The genome sequences of other pathogenic and non-pathogenic Brachyspira species are becoming available, and this data will help to reveal how these species have evolved and adapted to varied lifestyles in the large intestines of different species, and why some but not others can induce colitis and diarrhoea.

Abstract Background The role of pathogenic mycobacteria in diabetes has been a focus of speculation since a decade without any meaningful insights into the mechanism of diabetes causation vis a vis mycobacterial factors. Two of our studies based on PCR identification of mycobacterial DNA and detection of antibodies specific to the recombinant antigens and whole cell lysates of the Mycobacterium avium subsp. paratuberculosis (MAP) shown a clear association of MAP with the presence of type 1 diabetes mellitus (T1DM). Methods In this study, we sought to investigate if or not type 2 diabetes (T2DM) patients harbour humoral responses to MAP. Using three different MAP antigen preparations, humoral antibody profiles were estimated for 57 T2DM patients and 57 healthy controls. Statistical analysis was performed with the Chi-square test with Yates' corrections. Results We observed insignificant levels of humoral antibodies against recombinant heparin binding haemagglutinin (HbHA), glycosyl transferase (Gsd) and MAP whole cell lysate in the blood of subjects with T2DM as compared to healthy controls. Conclusion We found no obvious association of MAP with the incidence of T2DM in Sardinian patients.

Abstract Background The human gastric pathogen Helicobacter pylori is co-evolved with its host and therefore, origins and expansion of multiple populations and sub populations of H. pylori mirror ancient human migrations. Ancestral origins of H. pylori in the vast Indian subcontinent are debatable. It is not clear how different waves of human migrations in South Asia shaped the population structure of H. pylori. We tried to address these issues through mapping genetic origins of present day H. pylori in India and their genomic comparison with hundreds of isolates from different geographic regions. Results We attempted to dissect genetic identity of strains by multilocus sequence typing (MLST) of the 7 housekeeping genes (atpA, efp, ureI, ppa, mutY, trpC, yphC) and phylogeographic analysis of haplotypes using MEGA and NETWORK software while incorporating DNA sequences and genotyping data of whole cag pathogenicity-islands (cagPAI). The distribution of cagPAI genes within these strains was analyzed by using PCR and the geographic type of cagA phosphorylation motif EPIYA was determined by gene sequencing. All the isolates analyzed revealed European ancestry and belonged to H. pylori sub-population, hpEurope. The cagPAI harbored by Indian strains revealed European features upon PCR based analysis and whole PAI sequencing. Conclusion These observations suggest that H. pylori strains in India share ancestral origins with their European counterparts. Further, non-existence of other sub-populations such as hpAfrica and hpEastAsia, at least in our collection of isolates, suggest that the hpEurope strains enjoyed a special fitness advantage in Indian stomachs to out-compete any endogenous strains. These results also might support hypotheses related to gene flow in India through Indo-Aryans and arrival of Neolithic practices and languages from the Fertile Crescent.

Abstract Helicobacter pylori is an important yet unproven etiological agent of gastric cancer. H. pylori infection is more prevalent in developing Asian countries like India and it is usually acquired at an early age. It has been two decades since Marshall and Warren (1984) first described curved bacilli in the stomach of ulcer and gastritis patients. This discovery has won them the Nobel Prize recently, but the debate whether H. pylori is a pathogen or a commensal organism is still hot. Associations with disease-specific factors remain illusive years after the genome sequences were made available. Cytotoxin-associated antigen A (CagA) and the so-called plasticity region cluster genes are implicated in pathogenesis of the carcinoma of stomach. Another virulence factor VacA whose role is still debatable, has recently been projected in pathology of gastric cancer. Studies of the evolution through genetic variation in H. pylori populations have provided a window into the history of human population migrations and a possible co-evolution of this pathogen with its human host. Possible symbiotic relationships were seriously debated since the discovery of this pathogen. The debate has been further intensified as some studies proposed H. pylori infection to be beneficial in some humans. In this commentary, we attempt to briefly discuss about H. pylori as a human pathogen, and some of the important issues linked to its pathophysiology in different hosts. Abstract Background Leptospira are the parasitic bacterial organisms associated with a broad range of mammalian hosts and are responsible for severe cases of human Leptospirosis. The epidemiology of leptospirosis is complex and dynamic. Multiple serovars have been identified, each adapted to one or more animal hosts. Adaptation is a dynamic process that changes the spatial and temporal distribution of serovars and clinical manifestations in different hosts. Serotyping based on repertoire of surface antigens is an ambiguous and artificial system of classification of leptospiral agents. Molecular typing methods for the identification of pathogenic leptospires up to individual genome species level have been highly sought after since the decipherment of whole genome sequences. Only a few resources exist for microbial genotypic data based on individual techniques such as Multiple Locus Sequence Typing (MLST), but unfortunately no such databases are existent for leptospires. Results We for the first time report development of a robust MLST method for genotyping of Leptospira. Genotyping based on DNA sequence identity of 4 housekeeping genes and 2 candidate genes was analyzed in a set of 120 strains including 41 reference strains representing different geographical areas and from different sources. Of the six selected genes, adk, icdA and secY were significantly more variable whereas the LipL32 and LipL41 coding genes and the rrs2 gene were moderately variable. The phylogenetic tree clustered the isolates according to the genome-based species. Conclusion The main advantages of MLST over other typing methods for leptospires include reproducibility, robustness, consistency and portability. The genetic relatedness of the leptospires can be better studied by the MLST approach and can be used for molecular epidemiological and evolutionary studies and population genetics.

Abstract Background The study was conducted between 2000 and 2003 on 750 human subjects, yielding 850 strains of staphylococci from clinical specimens (575), nasal cultures of hospitalized patients (100) and eye & nasal sources of hospital workers (50 & 125 respectively) in order to determine their epidemiology, acquisition and dissemination of resistance genes. Methods Organisms from clinical samples were isolated, cultured and identified as per the standard routine procedures. Susceptibility was measured by the agar diffusion method, as recommended by the Nat ional Committee for Clinical Laboratory Standards (NCCLS). The modified method of Birnboin and Takahashi was used for isolation of plasmids from staphylococci. Pulsed-field gel electrophoresis (PFGE) typing of clinical and carrier Methicillin resistant Staphylococcus aureus (MRSA) strains isolated during our study was performed as described previously. Results It was shown that 35.1% of Staphylococcus aureus and 22.5% of coagulase-negative staphylococcal isolates were resistant to methicillin. Highest percentage of MRSA (35.5%) was found in pus specimens (n = 151). The multiple drug resistance of all MRSA (n = 180) and Methicillin resistant Coagulase-negative Staphylococcus aureus (MRCNS) (n = 76) isolates was detected. In case of both methicillin-resistant as well as methicillin-sensitive Saphylococcal isolates zero resistance was found to vancomycin where as highest resistance was found to penicillin G followed by ampicillin. It was shown that the major reservoir of methicillin resistant staphylococci in hospitals are colonized/infected inpatients and colonized hospital workers, with carriers at risk for developing endogenous infection or transmitting infection to health care workers and patients. The results were confirmed by molecular typing using PFGE by SmaI-digestion. It was shown that the resistant markers G and T got transferred from clinical S. aureus (JS-105) to carrier S. aureus (JN-49) and the ciprofloxacin (Cf) and erythromycin (E) resistance seemed to be chromosomal mediated. In one of the experiments, plasmid pJMR1O from Staphylococcus aureus coding for ampicillin (A), gentamicin (G) and amikacin (Ak) resistance was transformed into Escherichia coli. The minimal inhibitory concentrations (MICs) for A and G were lower in E. coli than in S. aureus. However, the MIC for Ak was higher in E. coli transformants than in S. aureus. Conclusion There is a progressive increase in MRSA prevalence and multi-drug resistance in staphylococci. Vancomycin is still the drug of choice for MRSA infections. The major reservoir of methicillin resistant staphylococci in hospitals is colonized/infected inpatients and colonized hospital workers. Resistance transfer from staphylococci to E. coli as well as from clinical to carrier staphylococci due to antibiotic stress seemed to be an alarming threat to antimicrobial chemotherapy.

Abstract Background Helicobacter pylori is presumed to be co-evolved with its human host and is a highly diverse gastric pathogen at genetic levels. Ancient origins of H. pylori in the New World are still debatable. It is not clear how different waves of human migrations in South America contributed to the evolution of strain diversity of H. pylori. The objective of our 'phylogeographic' study was to gain fresh insights into these issues through mapping genetic origins of H. pylori of native Peruvians (of Amerindian ancestry) and their genomic comparison with isolates from Spain, and Japan. Results For this purpose, we attempted to dissect genetic identity of strains by fluorescent amplified fragment length polymorphism (FAFLP) analysis, multilocus sequence typing (MLST) of the 7 housekeeping genes (atpA, efp, ureI, ppa, mutY, trpC, yphC) and the sequence analyses of the babB adhesin and oipA genes. The whole cag pathogenicity-island (cagPAI) from these strains was analyzed using PCR and the geographic type of cagA phosphorylation motif EPIYA was determined by gene sequencing. We observed that while European genotype (hp-Europe) predominates in native Peruvian strains, approximately 20% of these strains represent a sub-population with an Amerindian ancestry (hsp-Amerind). All of these strains however, irrespective of their ancestral affiliation harbored a complete, 'western' type cagPAI and the motifs surrounding it. This indicates a possible acquisition of cagPAI by the hsp-Amerind strains from the European strains, during decades of co-colonization. Conclusion Our observations suggest presence of ancestral H. pylori (hsp-Amerind) in Peruvian Amerindians which possibly managed to survive and compete against the Spanish strains that arrived to the New World about 500 years ago. We suggest that this might have happened after native Peruvian H. pylori strains acquired cagPAI sequences, either by new acquisition in cag-negative strains or by recombination in cag positive Amerindian strains.

Abstract Background An observational case series was used to study the virulence characteristics and genotypes of paired Staphylococcus epidermidis isolates cultured from intraocular samples and from periocular environment of patients with postcataract surgery endophthalmitis. Methods Eight S. epidermidis isolates were obtained from three patients (2 from patients #1 and 2 and 4 from patient #3) whose vitreous and/or anterior chamber (AC) specimens and preoperative lid/conjunctiva samples were culture positive. Cultures were identified by API-Staph phenotypic identification system and genotypically characterized by Fluorescent Amplified Fragment Length Polymorphism (FAFLP) and checked for their antimicrobial susceptibility. The isolates were tested for biofilm-production and methicillin-resistance (MR) by PCR amplification of icaAB and mecA gene respectively. Results Four out of eight S. epidermidis strains showed multiple drug resistance (MDR). All the eight strains were PCR positive for mecA gene whereas seven out of eight strains were positive for icaAB genes. In all three patients FAFLP typing established vitreous isolates of S. epidermidis strains to be indistinguishable from the strains isolated from the patient's conjunctival swabs. However, from patient number three there was one isolate (1030b from lid swab), which appeared to be nonpathogenic and ancestral having minor but significant differences from other three strains from the same patient. This strain also lacked icaAB gene. In silico analysis indicated possible evolution of other strains from this strain in the patient. Conclusion Methicillin-resistant biofilm positive S. epidermidis strains colonizing the conjunctiva and eyelid were responsible for postoperative endophthalmitis (POE).

Abstract Background Investigation of two independent outbreaks of post cataract surgery endophthalmitis identified the reservoir of epidemic strains of P. aeruginosa. Methods Patient isolates cultured from vitreous fluid of all the nine cases and from the peripheral devices of phacoemulsification machine were subjected to high-resolution Fluorescent Amplified Fragment Length Polymorphism (FAFLP) analysis. Results FAFLP based genotyping of the isolates confirmed nosocomial transmission. Although biochemical characterization and antibiotic susceptibility profiles grouped all the isolates together, FAFLP based genotyping revealed that, all the outbreak isolates were derived from 2 different strains, with independent origins. One group of isolates was traced to phacoprobe and the second one to the internal tubing system of the phacoemulsification machine used in cataract surgery. In silico analysis indicated possible evolution in both the clusters of P. aeruginosa isolates due to genetic polymorphisms. The polymorphisms were mapped to gene products (cell envelope, outer membrane proteins) possibly having significant role in pathogenesis. Conclusion The present study is probably the first one to apply FAFLP typing successfully to investigate outbreaks of postoperative endophthalmitis (POE) in an ophthalmic setting, which was able to identify the source, and helped to make rational decisions on sterilization procedures that halted more cases of infection in these hospitals.

Abstract Background Eradication of Helicobacter pylori is an important objective in overcoming gastric diseases. Many regimens are currently available but none of them could achieve 100% success in eradication. Eugenol and cinnamaldehyde that are commonly used in various food preparations are known to possess antimicrobial activity against a wide spectrum of bacteria. Aim The present study was performed to assess the in vitro effects of eugenol and cinnamaldehyde against indigenous and standard H. pylori strains, their minimum inhibitory concentrations (MICs) and time course lethal effects at various pH. Methods A total of 31 strains (29 indigenous and one standard strain of H. pylori ATCC 26695, one strain of E. coli NCIM 2089) were screened. Agar dilution method was used for the determination of drug sensitivity patterns of isolates to the commonly used antibiotics and broth dilution method for the test compounds. Results Eugenol and cinnamaldehyde inhibited the growth of all the 30 H. pylori strains tested, at a concentration of 2 μg/ml, in the 9th and 12th hours of incubation respectively. At acidic pH, increased activity was observed for both the compounds. Furthermore, the organism did not develop any resistance towards these compounds even after 10 passages grown at sub-inhibitory concentrations. Conclusion These results indicate that the two bioactive compounds we tested may prevent H. pylori growth in vitro, without acquiring any resistance.

Abstract The Gram negative curved bacillus H. pylori has become the prize bug of all times. Barry Marshall and Robin Warren the two discoverers of this organism have been awarded with this year's Nobel Prize. The Nobel committee at the Karolinska Institute of Sweden has selected this paradigm shift discovery of 1982 as the most impacting in medical sciences. This award has surprised many as the Nobel assembly has selected this 'Robert Koch styled medical detective work' for the prize as compared to many outstanding basic research stories on the waitlist. This editorial briefly touches the significant impact of H. pylori on gastroduodenal management and the path forward as the bug has become quite controversial in recent times.

Abstract Background Genomic diversity of H. pylori from many different human populations is largely unknown. We compared genomes of 65 H. pylori strains from Nottingham, England. Molecular analysis was carried out to identify rearrangements within and outside the cag-pathogenicity-island (cag PAI) and DNA sequence divergence in candidate genes. Phylogenetic analysis was carried out based on various high-resolution genotyping techniques. Results Analyses of virulence genes (cagT, cagE, cagA, vacA, iceA, oipA and babB) revealed that H. pylori strains from England are genetically distinct from strains obtained from other countries. The toxigenic vacA s1m1 genotype was found to be less common and the plasticity region cluster was found to be disrupted in all the isolates. English isolates showed a predominance of iceA1 alleles and a functional proinflammatory oipA gene. The English H. pylori gene pool revealed several Asian/oriental features. This included the predominance of cagA – glr (cagA right junction) motif types III and II (up to 42%), presence of vacA m1c alleles and phylogenetic affinity towards East Asian / Amerindian gene pools based on fluorescent amplified fragment length polymorphism (FAFLP) analysis and glmM sequence analysis. Conclusion Overall, our results demonstrated genetic affinities of H. pylori in England with both European and the Asian gene pools and some distinctive genetic features of virulence genes that may have evolved in this important European population.

Abstract The human gastric pathogen Helicobacter pylori causes chronic gastritis, peptic ulcer disease, gastric carcinoma, and mucosa-associated lymphoid tissue (MALT) lymphoma. It infects over 50% of the worlds' population, however, only a small subset of infected people experience H. pylori-associated illnesses. Associations with disease-specific factors remain enigmatic years after the genome sequences were deciphered. Infection with strains of Helicobacter pylori that carry the cytotoxin-associated antigen A (cagA) gene is associated with gastric carcinoma. Recent studies revealed mechanisms through which the cagA protein triggers oncopathogenic activities. Other candidate genes such as some members of the so-called plasticity region cluster are also implicated to be associated with carcinoma of stomach. Study of the evolution of polymorphisms and sequence variation in H. pylori populations on a global basis has provided a window into the history of human population migration and co-evolution of this pathogen with its host. Possible symbiotic relationships were debated since the discovery of this pathogen. The debate has been further intensified as some studies have posed the possibility that H. pylori infection may be beneficial in some humans. This assumption is based on increased incidence of gastro-oesophageal reflux disease (GERD), Barrett's oesophagus and adenocarcinoma of the oesophagus following H. pylori eradication in some countries. The contribution of comparative genomics to our understanding of the genome organisation and diversity of H. pylori and its pathophysiological importance to human healthcare is exemplified in this review.

Considerable recent research in neurosciences has dealt with the topic of consciousness, even though there is still disagreement about how to identify and classify conscious states. Recent behavioral work on the topic also exists. We survey recent behavioral and neuroscientific literature with the aims of commenting on strengths and weaknesses of the literature and mapping new directions and recommendations for experimental psychologists. We reconcile this literature with a view of human information processing (Cowan, 1988; Cowan et al., 2024) in which a capacity-limited focus of attention is embedded within the activated portion of long-term memory, with dual bottom-up and top-down control of the focus of attention. None of the many extant theories fully captures what we propose as the organization of conscious thought at cognitive and neural levels. It seems clear that information from various cognitive functions, based on signals from various brain areas, is integrated into a conscious whole. In our new proposal, the integration involves funneling information to a hub or focus of attention neurally centered in the parietal lobes and functionally connected to areas representing the currently attended information. This funneling process (bringing information from diverse sensory and frontal sources to contact a small parietal area where attended information is coordinated and combined) may be the converse of global broadcasting, from other proposals (Baars et al., 2021; Baars & Franklin, 2003; Dehaene & Changeux, 2011). The proposed system incorporates many principles from previous research and theorization and strives toward a resolution of the relation between consciousness and attention. Spiny projection neurons (SPNs) of the striatum are critical in integrating neurochemical information to coordinate motor and reward-based behavior. Mutations in the regulatory transcription factors expressed in SPNs can result in neurodevelopmental disorders (NDDs). Paralogous transcription factors Foxp1 and Foxp2, which are both expressed in the dopamine receptor 1 (D1) expressing SPNs, are known to have variants implicated in NDDs. Utilizing mice with a D1-SPN-specific loss of Foxp1, Foxp2, or both and a combination of behavior, electrophysiology, and cell-type-specific genomic analysis, loss of both genes results in impaired motor and social behavior as well as increased firing of the D1-SPNs. Differential gene expression analysis implicates genes involved in autism risk, electrophysiological properties, and neuronal development and function. Viral-mediated re-expression of Foxp1 into the double knockouts is sufficient to restore electrophysiological and behavioral deficits. Long-range glutamatergic inputs originating from the cortex and thalamus are indispensable for striatal development, providing the foundation for motor and cognitive functions. Despite their significance, transcriptional regulation governing these inputs remains largely unknown. We investigated the role of a transcription factor encoded by a high-risk autism-associated gene, FOXP1 , in sculpting glutamatergic inputs onto spiny projection neurons (SPNs) within the striatum. We find a neuron subtype-specific role of FOXP1 in strengthening and maturing glutamatergic inputs onto dopamine receptor 2-expressing SPNs (D2 SPNs). We also find that FOXP1 promotes synaptically driven excitability in these neurons. Using single-nuclei RNA sequencing, we identify candidate genes that mediate these cell-autonomous processes through postnatal FOXP1 function at the post-synapse. Last, we demonstrate that postnatal FOXP1 reinstatement rescues electrophysiological deficits, cell type-specific gene expression changes, and behavioral phenotypes. Together, this study enhances our understanding of striatal circuit development and provides proof of concept for a therapeutic approach for FOXP1 syndrome and other neurodevelopmental disorders.

We investigated the role of a transcription factor and high-risk autism-associated gene, FOXP1, in the development of glutamatergic inputs onto spiny projection neurons (SPNs) in the striatum. We find that FOXP1 robustly drives the strengthening and maturation of glutamatergic input onto dopamine receptor 2-expressing SPNs (D2 SPNs) but has a comparatively milder effect on D1 SPNs. This process is cell-autonomous and is likely mediated through postnatal FOXP1 function at the postsynapse. We identified postsynaptic FOXP1-regulated transcripts as potential candidates for mediating these effects. Postnatal reinstatement of FOXP1 rescues electrophysiological deficits, reverses gene expression alterations resulting from embryonic deletion, and mitigates behavioral phenotypes. These results provide support for a possible therapeutic approach for individuals with FOXP1 syndrome.

Autism Spectrum Disorder (ASD) has been associated with a complex interplay between genetic and environmental factors. Prenatal stress exposure has been identified as a possible risk factor, although most stress-exposed pregnancies do not result in ASD. The serotonin transporter (SERT) gene has been linked to stress reactivity, and the presence of the SERT short (S)-allele has been shown to mediate the association between maternal stress exposure and ASD. In a mouse model, we investigated the effects of prenatal stress exposure and maternal SERT genotype on offspring behavior and explored its association with maternal microRNA (miRNA) expression during pregnancy. Pregnant female mice were divided into four groups based on genotype (wildtype or SERT heterozygous knockout (Sert-het)) and the presence or absence of chronic variable stress (CVS) during pregnancy. Offspring behavior was assessed at 60 days old (PD60) using the three-chamber test, open field test, elevated plus-maze test, and marble-burying test. We found that the social preference index (SPI) of SERT-het/stress offspring was significantly lower than that of wildtype control offspring, indicating a reduced preference for social interaction on social approach, specifically for males. SERT-het/stress offspring also showed significantly more frequent grooming behavior compared to wildtype controls, specifically for males, suggesting elevated repetitive behavior. We profiled miRNA expression in maternal blood samples collected at embryonic day 21 (E21) and identified three miRNAs (mmu-miR-7684-3p, mmu-miR-5622-3p, mmu-miR-6900-3p) that were differentially expressed in the SERT-het/stress group compared to all other groups. These findings suggest that maternal SERT genotype and prenatal stress exposure interact to influence offspring behavior, and that maternal miRNA expression late in pregnancy may serve as a potential marker of a particular subtype of ASD pathogenesis.
