This laboratory experiment introduces the fundamentals of electrochemical techniques to the undergraduate students by developing a simple and cost-effective miniaturized electrochemical setup for detecting UV-induced DNA damage. This is an imperative experiment, as it provides students first-hand experience with electrochemical techniques while reinforcing lecture material on the biosensing applications. In this experiment, a simple and daily use material such as an ordinary pencil lead acts as a platform for an electrochemical DNA sensor. A three-electrode cell setup is utilized to monitor UV-induced DNA damage. Students will test the hypothesis about UV-induced DNA damage by measuring the redox signal of guanine under various experimental conditions. The simple, fast, and inexpensive setup allows students to examine the factors that would influence the electrochemical signals. This experimental work provides an opportunity for students to learn how to design a simple electrochemical experiment for research purposes as well as handling the biological samples. Proposing a hypothesis and testing it by designing an experiment are the experiences that will be gained by the students while learning about the optimization and validation of an analytical technique.

Carbon dots (CDs) are versatile, easy to fabricate, chemically stable, and ecofriendly materials. This work presents the impact of different post-treatments, such as centrifugation and filtration, on the photoluminescence (PL) and particle-size distribution of electrochemically synthesized CDs. Because organic envelope surfactants have been reported to influence PL, bare CDs were compared with CDs modified with amphiphilic and nonamphiphilic surfactants. The CD colloidal solution exhibited stabilization in the case of bare CDs and CDs modified with amphiphilic sodium dodecyl sulphate or non-amphiphilic surfactant sodium citrate, as indicated by the zeta potential. The experimental results also revealed a minor influence of the organic envelope on the absorbance or PL spectrum, whereas filtration of the colloid solution had a significant impact, revealing an effect not previously observed. The change in the absorbance spectrum was attributed to sp3 hybridization due to increased disorder and surface states during the filtration process. Absorbance deconvolution also disclosed two energy substates not mentioned elsewhere. The PL spectra of the filtered CD solutions revealed that the emission peaks shifted according to the incident wavelength, and the centrifuged CDs synthesized with the sodium citrate surfactant lacked PL due to the absence of sp3 states. Oxytetracycline (OTC) is a widely employed antibiotic in veterinary treatment and in the prevention of infections, potentially leaving residues in animal-derived food products, such as milk, that are consumed by humans. Given the detrimental effects of prolonged human exposure to antibiotics, it has become imperative to develop precise and sensitive methods for monitoring the presence of OTC in food. Herein, we describe the development and results of a preliminary label-free electrochemical aptasensor with antifouling properties designed to detect OTC in milk samples. The sensor was realized by modifying a gold screen-printed electrode with α-lipoic acid–NHS and an amine-terminated aptamer. Different electrochemical techniques were used to study the steps of the fabrication process and to quantify OTC in the presence of the Fe(CN)64−/Fe(CN)63− redox couple The detectable range of concentrations satisfy the maximum residue limits set by the European Union, with an limit of detection (LOD) of 14 ng/mL in phosphate buffer (BP) and 10 ng/mL in the milk matrix, and a dynamic range of up to 500 ng/mL This study is a steppingstone towards the implementation of a sensitive monitoring method for OTC in dairy products. Electrochemical biosensors are valued for their sensitivity and selectivity in detecting biological molecules. Having the advantage of generating signals that can be directly or indirectly proportional to the concentration of the target analyte, these biosensors can achieve specificity by utilizing a specific biorecognition surface designed to recognize the target molecule. Electrochemical biosensors have garnered substantial attention, as they can be used to fabricate compact, cost-effective devices, making them promising candidates for point-of-care testing (POCT) devices. This study introduces a label-free electrochemical biosensor employing a gold screen-printed electrode (SPE) to detect lysophosphatidic acid (LPA), a potential early ovarian cancer biomarker. We employed the gelsolin–actin system, previously introduced by our group, in combination with fluorescence spectrometry, as a biorecognition element to detect LPA. By immobilizing a gelsolin–actin complex on an SPE, we were able to quantify changes in current intensity using cyclic voltammetry and differential pulse voltammetry, which was directly proportional to the LPA concentration in the solution. Our results demonstrate the high sensitivity of the developed biosensor for detecting LPA in goat serum, with a limit of detection (LOD) and a limit of quantification (LOQ) of 0.9 µM and 2.76 µM, respectively, highlighting its potential as a promising tool for early-stage diagnosis of ovarian cancer.
