1. CanvOI, an Oncology Intelligence Foundation Model: Scaling FLOPS Differently

Abstract

The rapidly evolving field of digital oncopathology faces significant challenges, including the need to address diverse and complex clinical questions, often involving rare conditions, with limited availability of labeled data. These limitations hinder the development of robust AI-driven tools in the biomedical space, where accuracy in probabilistic determinations is of utmost importance. To address this, digital pathology foundation models have begun to emerge, typically developed with the size and diversity of the pre-training dataset and model parameters in mind. Here, we present CanvOI, a ViT-g/10-based foundation model designed to enhance the capabilities of digital pathology by addressing these challenges through a different approach. Considering the unique nature of oncologic histopathological images and the requirements from the embeddings to provide meaningful representations for Multiple Instance Learning (MIL) downstream models, we chose to modify the input image characteristics. By introducing larger tile sizes (380 x 380 pixels) and smaller patch sizes (10 x 10 pixels), we were able to optimize the model's performance, pushing computational resources in a new direction and achieving state-of-the-art performance on cancer-related benchmarks. CanvOI demonstrated a 1.5-7.4% improvement in averaged AUC compared to other leading foundation models built for digital pathology. Moreover, our results demonstrate that CanvOI significantly outperformed the other models, with the performance gap widening substantially when trained on just 10% of the initial cohort. This work highlights an alternative approach that, if integrated with traditional development approaches, has the potential to advance Oncology Intelligence (OI), overcome some of the current barriers and ultimately improve the clinical outcome of cancer patients., Comment: 12 pages, 5 figures

Abstract

We are in the midst of a technological revolution that is providing new insights into human biology and cancer. In this era of big data, we are amassing large amounts of information that is transforming how we approach cancer treatment and prevention. Enactment of the Cancer Moonshot within the 21st Century Cures Act in the USA arrived at a propitious moment in the advancement of knowledge, providing nearly US$2 billion of funding for cancer research and precision medicine. In 2016, the Blue Ribbon Panel (BRP) set out a roadmap of recommendations designed to exploit new advances in cancer diagnosis, prevention, and treatment. Those recommendations provided a high-level view of how to accelerate the conversion of new scientific discoveries into effective treatments and prevention for cancer. The US National Cancer Institute is already implementing some of those recommendations. As experts in the priority areas identified by the BRP, we bolster those recommendations to implement this important scientific roadmap. In this Commission, we examine the BRP recommendations in greater detail and expand the discussion to include additional priority areas, including surgical oncology, radiation oncology, imaging, health systems and health disparities, regulation and financing, population science, and oncopolicy. We prioritise areas of research in the USA that we believe would accelerate efforts to benefit patients with cancer. Finally, we hope the recommendations in this report will facilitate new international collaborations to further enhance global efforts in cancer control.

Abstract

BackgroundThe genetic origins of chemotherapy resistance are well established; however, the role of epigenetics in drug resistance is less well understood. To investigate mechanisms of drug resistance, we performed systematic genetic, epigenetic, and transcriptomic analyses of an alkylating agent-sensitive murine lymphoma cell line and a series of resistant lines derived by drug dose escalation.MethodsDose escalation of the alkylating agent mafosfamide was used to create a series of increasingly drug-resistant mouse Burkitt's lymphoma cell lines. Whole genome sequencing, DNA microarrays, reduced representation bisulfite sequencing, and chromatin immunoprecipitation sequencing were used to identify alterations in DNA sequence, mRNA expression, CpG methylation, and H3K27me3 occupancy, respectively, that were associated with increased resistance.ResultsOur data suggest that acquired resistance cannot be explained by genetic alterations. Based on integration of transcriptional profiles with transcription factor binding data, we hypothesize that resistance is driven by epigenetic plasticity. We observed that the resistant cells had H3K27me3 and DNA methylation profiles distinct from those of the parental lines. Moreover, we observed DNA methylation changes in the promoters of genes regulated by E2a and members of the polycomb repressor complex 2 (PRC2) and differentially expressed genes were enriched for targets of E2a. The integrative analysis considering H3K27me3 further supported a role for PRC2 in mediating resistance. By integrating our results with data from the Immunological Genome Project (Immgen.org), we showed that these transcriptional changes track the B-cell maturation axis.ConclusionsOur data suggest a novel mechanism of drug resistance in which E2a and PRC2 drive changes in the B-cell epigenome; these alterations attenuate alkylating agent treatment-induced apoptosis.

Abstract

This study aims to examine the effect of competence and work motivation in improving the performance of extension workers through organizational commitment as an mediation variable. The object of this research is the Field Cooperative Extension Officer of the Banten Province Cooperatives and SMEs Office. The population in this study was 50 people, while the sampling used a survey technique or saturated sample, which means that the entire population was sampled. The results of this study indicate that competence directly has a significant impact on the performance of extension workers but does not have a significant impact on organizational commitment. Furthermore, work motivation has a significant effect on the performance of the extension workers and organizational commitment, while organizational commitment has a significant impact on the performance of the extension workers. On the other hand, organizational commitment can mediate between work motivation and performance of extension workers but cannot mediate on the competence and performance of extension workers.

Abstract

The research aims to study about the awareness and perceptions of the SMEs palm sugar in Lebak district, Banten Province-Indonesia on Fintech model such as Peer-to-Peer lending and crowd funding in developing their business. The researchers designed as many as 17 questions to conduct a survey related to the research variable. Descriptive analysis is applied by using Microsoft Office Excel data processing. Frequency distribution and central size are used for data analysis. The results show that awareness level of the most respondents is still insufficient on Peer-to-Peer lending and crowd funding as alternative financing instruments. The majority of them have a positive perception of FinTech development. It can be concluded that the level of awareness of most respondents is still very limited, especially on P2P loans, Crowdfunding and other FinTech services provided by non-bank financial institutions. They need to increase awareness level of palm sugar SMEs by related parties such as the government.

Abstract

To investigate the transition from non-cancerous to metastatic from a physical sciences perspective, the Physical Sciences-Oncology Centers (PS-OC) Network performed molecular and biophysical comparative studies of the non-tumorigenic MCF-10A and metastatic MDA-MB-231 breast epithelial cell lines, commonly used as models of cancer metastasis. Experiments were performed in 20 laboratories from 12 PS-OCs. Each laboratory was supplied with identical aliquots and common reagents and culture protocols. Analyses of these measurements revealed dramatic differences in their mechanics, migration, adhesion, oxygen response, and proteomic profiles. Model-based multi-omics approaches identified key differences between these cells' regulatory networks involved in morphology and survival. These results provide a multifaceted description of cellular parameters of two widely used cell lines and demonstrate the value of the PS-OC Network approach for integration of diverse experimental observations to elucidate the phenotypes associated with cancer metastasis.

Abstract

This study aims to determine the relationship between the variable person-job fit in improving employee performance and job satisfaction in mediating person-job fit in improving employee performance. Building a model on business phenomena and research gaps in improving employee performance using purposive sampling technique. Data analysis using SEM-PLS method. The results showed 1) Person-job fit had a positive and insignificant effect on employee performance, 2) Person-job fit had a positive and significant effect on job satisfaction, 3) Job satisfaction had a positive and significant effect on employee performance, 4) Job satisfaction mediate the effect of person-job fit significantly on employee performance. Penelitian ini bertujuan untuk mengetahui hubungan antar variabel person-job fit dalam meningkatkan kinerja pegawai serta kepuasan kerja dalam memediasi person-job fit dalam meningkatkan kinerja pegawai. Membangun model atas fenomena bisnis dan research gap dalam meningkatkan kinerja pegawai dengan teknik purposive sampling. Analisis data menggunakan metode SEM-PLS. Hasil penelitian menunjukan 1) Person-job fit berpengaruh positif dan tidak signifikan terhadap kinerja pegawai, 2) Person-job fit memiliki pengaruh positif dan signifikan terhadap kepuasan kerja, 3) Kepuasan kerja memiliki pengaruh positif dan signifikan terhadap kinerja pegawai, 4) Kepuasan kerja memediasi pengaruh person-job fit secara signifikan terhadap kinerja pegawai.

Abstract

The purpose of this study was to examine the effect of compensation on employee performance, the effect of compensation on job satisfaction, the effect of job satisfaction on employee performance, the effect of job placement on job satisfaction, at RSUD dr. Dradjat Prawiranegara. The population in this study were nurses with non-civil servant status or nurses with voluntary labor status (TKS) who were still working at RSUD dr. Dradjat Prawiranegara. The number of samples is determined based on the number of indicators of the 4 variables used multiplied by 5, the total of all indicators is 20 x 5, total of Sample is 100 respondents, the sampling method uses the Non Random Sampling technique, because the population and sample have been determined, namely Non-PNS Nurses.The results of this study indicate that there is a significant effect of compensation on employee performance, there is a significant effect of compensation on job satisfaction, there is a significant effect of job satisfaction on employee performance, there is a significant effect of job placement on job satisfaction, and there is a significant effect of job placement on employee performance.

Abstract

The research objective is to find out how Le Dian Hotel can carry out its commitment to provide quality service to every guest represented in an organizational culture so that it can make employees more motivated to achieve maximum work performance and performance assisted by optimal empowerment. The research object was the employees of Hotel Le Dian, Serang with a total sample of 100 people. Data analysis using SEM-PLS model. The results showed that there was a positive influence of organizational culture on employee performance and achievement motivation. There is a positive influence of achievement motivation on employee performance and there is a positive moderation of empowerment on the relationship between achievement motivation and employee performance. This research implies the importance of organizational culture at Le Dian Hotel to be able to improve employee performance which is strengthened by high achievement motivation and employee empowerment.

Abstract

This study aims to explore the economic impact, especially UMKM players in Banten on the spread of covid-19, and to identify the strength of entrepreneurial networks and the use of e-marketing by UMKM in Banten during the covid-19 pandemic. The method used in this study was analyzed by sampling using a questionnaire as a data collection tool. This questionnaire includes closed questions, measured on a nominal scale. The collected data were analyzed using the AMOS Statistical Package for the Social Sciences (SPSS) software. The results showed that there had been a decline in the marketing performance of UMKM due to the impact of the Covid-19 pandemic. And it is proven that the quality of entrepreneurial networks and e-marketing can improve the marketing performance of UMKM. From this research, a model for strengthening UMKM in dealing with the impact of the Covid-19 pandemic is obtained by strengthening entrepreneurial networks and the use of e-marketing, so that UMKM in Banten in particular and UMKM in Indonesia in general, do not fall into business collapse caused by the spread of Covid-19. The conclusion of this study is expected to contribute to the development of management science, especially marketing, entrepreneurship and the development of UMKM in dealing with the impact of the Covid-19 pandemic.

Abstract

Small and Micro Enterprises (SMEs) has an important role in supporting the Indonesian economy. On the other hand, the internet has developed rapidly. This internet media progress can be utilized by SMEs to develop their business. However, in Indonesia, the number and presentation of MSEs that have used digital media is still very low. The use of the internet for the business world is can be seen in the product marketing process, for example the increasing online business in Indonesia, including in Banten, and is applied to all business activities. However, the use of the internet in SMEs in Banten is still very small. According to the 2016 Economic Census, only around 37 thousand businesses or 3.84 percent use the internet both for product marketing processes and others and only around 2.55 percent of the internet is used for marketing, this fact become the basis for training and strengthening for the community, especially for SMEs in Banten to realize how important it is to become "internet literate" and encourage the use of the internet for marketing even higher.
