In this article, the problem of real‐time estimation of the fermentative ethanol process is tackled. The considered observer is a model‐based technique that is robust regarding the model parameter uncertainties and inline noisy measurements. An unstructured kinetic model was used to describe the production of ethanol in a batch bioreactor for Saccharomyces cerevisiae. The biomass concentration was selected as the measured bioreactor´s output via an inline device, where the estimate variables were the substrate and ethanol concentrations. An experimental prototype was constructed to demonstrate the observer's real‐time performance. The experimental results show that the robust, smooth sliding mode observer performs better than the standard proportional sliding mode observer. [ABSTRACT FROM AUTHOR]

In this contribution, a class of observer-based optimal feedback control is designed. The proposed feedback control is based on the Euler–Lagrange theoretical framework, and it is motivated by the productivity intensification from the chemical reactors, which is optimally increased. A Lagrangian is computed by employing the corresponding mass balance equation of a specifically selected biochemical compound. The resulting optimal controller is coupled with a novel uncertainty estimator with bounded feedback to derive an accurate estimation of the unknown terms and functions, mostly related to the reaction rate. Via Lyapunov analysis, it was shown that the proposed observer is asymptotically stable. The estimation of the unknown terms and functions is used by the proposed controller. The proposed methodology is applied to a generic model of an enzymatic biochemical continuous reactor with complex oscillatory dynamic behavior described by mass balance equations, so, in general, the proposed controller may be applied to any continuous stirred tank bioreactor; that is, the controller is independent of the specific kinetic functions. This study deals with the problem of estimating the amount of biomass and lactic acid concentration in a lactic acid production process. A continuous stirred tank bioreactor was used for the culture of Lactobacillus helveticus. A nonlinear sliding mode observer is proposed and designed, which gives an estimate of both the biomass and lactic acid concentrations as a function of glucose uptake from the culture medium. Numerical results are given to illustrate the effectiveness of the proposed observer against a standard sliding-mode observer. It was found that the proposed observer worked very well for the benchmark bioreactor model. Also, the numerical results indicated that the proposed estimation methodology was robust to the uncertainties associated with un-modelled dynamics. A non-segregated kinetic model is proposed to describe a fermentation process of agro-industrial residues derived via cocoa (mucilage juice) by Pichia kudriavzevii. The novel proposed hybrid model is based on a multiple coupling reaction mechanisms (structured) to describe the kinetics of substrate consumption, biomass, carbon dioxide, and ethanol, coupled to an unstructured model for the activity enzyme. The parameters of the kinetic model are estimated by non-linear least-squares curve fitting using the Marquardt-Levenberg algorithm. In addition, numerical simulations were compared with the experimental data via residual graphs. The effectiveness of the model was statistically evaluated using dimensionless efficiency coefficients under different initial conditions. A global sensitivity analysis was applied (Fisher's information matrix). The experimental results of the batch reactor showed a maximum ethanol concentration of 29 g/L, with a yield of 0.48 g-ethanol/g-glucose and a productivity of 0.30 g/L h. The method determined that the cell formation coefficient and the specific substrate consumption rate ( θ 1 and θ 2 ) directly influence most of the states of our system. The proposed scheme is particularly suitable to assist in the rational design of cell factory properties or fermentation processes because it can represent the complex biochemistry in more detail and under different initial experimental conditions; the above reveals that the generated model is robust and can be considered for control and optimization purposes.
