Abstract

In the contemporary era, social media platforms amass an extensive volume of social data contributed by their users. In order to promptly grasp the opinions and emotional inclinations of individuals regarding a product or event, it becomes imperative to perform sentiment analysis on the user-generated content. Microblog comments often encompass both lengthy and concise text entries, presenting a complex scenario. This complexity is particularly pronounced in extensive textual content due to its rich content and intricate word interrelations compared to shorter text entries. Sentiment analysis of public opinion shared on social networking websites such as Facebook or Twitter has evolved and found diverse applications. However, several challenges remain to be tackled in this field. The hybrid methodologies have emerged as promising models for mitigating sentiment analysis errors, particularly when dealing with progressively intricate training data. In this article, to investigate the hesitancy of COVID-19 vaccination, we propose eight different hybrid deep learning models for sentiment classification with an aim of improving overall accuracy of the model. The sentiment prediction is achieved using embedding, deep learning model and grid search algorithm on Twitter COVID-19 dataset. According to the study, public sentiment towards COVID-19 immunization appears to be improving with time, as evidenced by the gradual decline in vaccine reluctance. Through extensive evaluation, proposed model reported an increased accuracy of 98.86%, outperforming other models. Specifically, the combination of BERT, CNN and GS yield the highest accuracy, while the combination of GloVe, BiLSTM, CNN and GS follows closely behind with an accuracy of 98.17%. Abstract

Intron selection during the formation of prespliceosomes is a critical event in pre-mRNA splicing. Chemical modulation of intron selection has emerged as a route for cancer therapy. Splicing modulators alter the splicing patterns in cells by binding to the U2 snRNP (small nuclear ribonucleoprotein)-a complex chaperoning the selection of branch and 3' splice sites. Here we report crystal structures of the SF3B module of the U2 snRNP in complex with spliceostatin and sudemycin FR901464 analogs, and the cryo-electron microscopy structure of a cross-exon prespliceosome-like complex arrested with spliceostatin A. The structures reveal how modulators inactivate the branch site in a sequence-dependent manner and stall an E-to-A prespliceosome intermediate by covalent coupling to a nucleophilic zinc finger belonging to the SF3B subunit PHF5A. These findings support a mechanism of intron recognition by the U2 snRNP as a toehold-mediated strand invasion and advance an unanticipated drug targeting concept.

24. Exploring the influence of fiber orientation on the mechanical characteristics of polymer composites reinforced with banana and corn fibers.

Abstract

In recent decades, the utilization of polymer composites reinforced with natural fibers has seen a significant increase due to their durability, eco-friendliness, and favorable composite properties. This study investigates the influence of unidirectional and cross-directional fiber orientation on the mechanical properties of polymer composites reinforced with banana and corn fibers, fabricated through a hand-lay-up process. The research assesses the impact of fiber orientation on various mechanical properties, including density, porosity, tensile strength, flexural strength, and impact strength. The findings reveal that the higher densities of banana and corn fibers, in comparison to the epoxy matrix, contribute to increased overall weight density in the composites, with cross-directional fiber orientation leading to higher porosity. Moreover, cross-directional reinforcement orientation enhances tensile strength, resulting in a robust bond with the matrix. Composites with cross-directional corn fibers exhibit the highest ultimate tensile strength of 49.57 MPa, marking a significant improvement over other fiber configurations. Notably, unidirectional corn fibers outperform in flexural strength of 14.07 MPa, surpassing banana–corn, and banana–banana configurations by 268.32% and 32.73%, respectively, and cross-directional banana–corn hybrid composites exhibit superior impact strength measuring 5.31 kJ/m2 due to their ability to resist crack propagation. Abstract

Hotspot mutations in splicing factor genes have been recently reported at high frequency in hematological malignancies, suggesting the importance of RNA splicing in cancer. We analyzed whole-exome sequencing data across 33 tumor types in The Cancer Genome Atlas (TCGA), and we identified 119 splicing factor genes with significant non-silent mutation patterns, including mutation over-representation, recurrent loss of function (tumor suppressor-like), or hotspot mutation profile (oncogene-like). Furthermore, RNA sequencing analysis revealed altered splicing events associated with selected splicing factor mutations. In addition, we were able to identify common gene pathway profiles associated with the presence of these mutations. Our analysis suggests that somatic alteration of genes involved in the RNA-splicing process is common in cancer and may represent an underappreciated hallmark of tumorigenesis.

Abstract

Heat treatment of steel is one of most essential variables in engineering metallurgy because it improves various physical & mechanical characteristics that are useful in a variety of engineering applications. Heat treatment is a set of processes that involves heating & cooling a metal or alloy in a solid state in order to get desired microstructures by refining grain size & a combination of characteristics. The main goal of this study is to demonstrate how heat treatment affects low-carbon steel (A36) in order to reveal its microstructural and mechanical (impact, tensile, and hardness) characteristics. A36 steel specimens that were cylindrical and rectangular were used for this purpose. The samples were polished by using emery paper, grades were following 80, 220, 400, 600, 800, 1000, and 2000. After polishing the specimen numbering was done and the specimen numbering was S0, S1, S2...S8. These all specimens were ready for different types of heat treatment processes like tempering, quenching, annealing, and normalizing. For this purpose, samples were heat treated in a muffle furnace at approximately 7750 C for almost two hours and then cooled by different quenching media (air, water, and oil). After heat treatment (quenching, normalizing, tempering, and annealing) Brinell hardness test was done by the Brinell hardness testing machine. After this test, there were two tests more done, tensile and impact. It was observed that there were two types of the quenching process, oil, and water After - this process hardness test was done, and observed that the oil quenching sample's hardness was higher than water quenching sample. Abstract

Impaired retinal blood flow is associated with ocular diseases such as glaucoma, macular degeneration, and diabetic retinopathy. Among several ocular imaging techniques developed to measure retinal blood flow both invasively and non-invasively, adaptive optics (AO)-enabled scanning laser ophthalmoscopy (AO-SLO) resolves individual red blood cells and provides a high resolution with which to measure flow across retinal microvasculature. However, cross-validation of flow measures remains a challenge owing to instrument and patient-specific variability in each imaging technique. Hence, there is a critical need for a well-controlled clinical flow phantom for standardization and to establish blood-flow measures as clinical biomarkers for early diagnosis. Here, we present the design and validation of a simple, compact, portable, linear flow phantom based on a direct current motor and a conveyor-belt system that provides linear velocity tuning within the retinal microvasculature range (0.5–7 mm/s). The model was evaluated using a sensitive AO-SLO line-scan technique, which showed a <6% standard deviation from the true velocity. Further, a clinical SLO instrument showed a linear correlation with the phantom's true velocity (r2 > 0.997). This model has great potential to calibrate, evaluate, and improve the accuracy of existing clinical imaging systems for retinal blood flow and aid in the diagnosis of ocular diseases with abnormal blood flow. Abstract

Photoreceptors (PRs) and retinal pigment epithelial (RPE) cells form a functional unit called the PR-RPE complex. The PR-RPE complex plays a critical role in maintaining retinal homeostasis and function, and the quantification of its structure and topographical arrangement across the macula are important for understanding the etiology, mechanisms, and progression of many retinal diseases. However, the three-dimensional cellular morphology of the PR-RPE complex in living human eyes has not been completely described due to limitations in imaging techniques. We used the cellular resolution and depth-sectioning capabilities of a custom, high-speed Fourier domain mode-locked laser-based adaptive optics–optical coherence tomography (FDML-AO-OCT) platform to characterize human PR-RPE complex topography across the temporal macula from eleven healthy volunteers. With the aid of a deep learning algorithm, key metrics were extracted from the PR-RPE complex of averaged AO-OCT volumes including PR and RPE cell density, PR outer segment length (OSL), and PR/RPE ratio. We found a tight grouping among our cohort for PR density, with a mean (±SD) value of 53,329 (±8106) cells/mm2 at 1° decreasing to 8669 (±737) cells/mm2 at 12°. We observed a power function relationship between eccentricity and both PR density and PR/RPE ratio. We found similar variability in our RPE density measures, with a mean value of 7335 (±681) cells/mm2 at 1° decreasing to 5547 (±356) cells/mm2 at 12°, exhibiting a linear relationship with a negative slope of −123 cells/mm2 per degree. OSL monotonically decreased from 33.3 (±2.4) µm at 1° to 18.0 (±1.8) µm at 12°, following a second-order polynomial relationship. PR/RPE ratio decreased from 7.3 (±0.9) µm at 1° to 1.5 (±0.1) µm at 12°. The normative data from this investigation will help lay a foundation for future studies of retinal pathology. Abstract

In this study, the microstructural and mechanical properties of Al-Si3N4/TiB2 hybrid reinforced composites are examined. The aim is to investigate the impact of a novel combination of reinforcements (Si3N4/TiB2) on the mechanical behavior of hybrid metal matrix composites (HMMCs). The composites are produced by adding a fixed amount of Si3N4 (4 wt.%) and 2, 4, and 6 wt.% of TiB2, respectively, into molten AA7075 aluminum alloy using a liquid state stir casting process. The physical and mechanical properties of fabricated HMMCs are determined through the measurement of density, porosity, tensile strength, Brinell hardness, flexural, and impact strength. The microstructural study of HMMCs reveals a consistent distribution of microparticles within the matrix. Evidence of reinforcement particles in the HMMCs is confirmed through field emission scanning electron microscopy with energy-dispersive spectroscopy and X-ray diffraction analysis. The fabricated HMMCs with 4 wt.% Si3N4 and 6 wt.% TiB2 particulates demonstrated the greatest strength among others. The ultimate tensile strength, Brinell hardness, and flexural strength are improved by 59.36%, 95.59%, and 49.33%, respectively, compared to the base Al-Zn-Mg alloy. However, the increment in strength of HMMCs comes with the tradeoff of reduced ductility and impact resistance measured through the percentage elongation and energy absorbed by the samples.
