In mammals, RNA editing events involve the conversion of adenosine (A) in inosine (I) by ADAR enzymes or the hydrolytic deamination of cytosine (C) in uracil (U) by the APOBEC family of enzymes, mostly APOBEC1. RNA editing has a plethora of biological functions, and its deregulation has been associated with various human disorders. While the large-scale detection of A-to-I is quite straightforward using the Illumina RNAseq technology, the identification of C-to-U events is a non-trivial task. This difficulty arises from the rarity of such events in eukaryotic genomes and the challenge of distinguishing them from background noise. Direct RNA sequencing by Oxford Nanopore Technology (ONT) permits the direct detection of Us on sequenced RNA reads. Surprisingly, using ONT reads from wild-type (WT) and APOBEC1-knock-out (KO) murine cell lines as well as in vitro synthesized RNA without any modification, we identified a systematic error affecting the accuracy of the Cs call, thereby leading to incorrect identifications of C-to-U events. To overcome this issue in direct RNA reads, here we introduce a novel machine learning strategy based on the isolation Forest (iForest) algorithm in which C-to-U editing events are considered as sequencing anomalies. Using in vitro synthesized and human ONT reads, our model optimizes the signal-to-noise ratio improving the detection of C-to-U editing sites with high accuracy, over 90% in all samples tested. Our results suggest that iForest, known for its rapid implementation and minimal memory requirements, is a promising tool to denoise ONT reads and reliably identify RNA modifications.

ABSTRACT: This international survey provides insights into public awareness of the importance of iodine as an essential trace mineral in human health along with knowledge of iodine dietary sources. The online questionnaire included sociodemographic aspects and dietary iodine consumer awareness on 7-point Likert-type questions. A total of 4,704 questionnaires from 16 countries were considered. Answers were analyzed through a multiple regression linear model including country, gender, age, education level, and employment status as fixed effects. Respondents were moderately aware of the importance of fish (4.86) and seafood (4.90) as dietary iodine sources, but less aware of milk as a primary iodine source (3.32). Respondent awareness varied considerably across countries. Age, education level, and employment status only modified their perception when asked about fish and seafood as a source of iodine, with elderly respondents, those highly educated and of working age being more aware of their relevance as dietary iodine sources. Respondent knowledge did not vary by age, education level, employment status, or gender when asked about cereals, vegetables and fruits, meat, and milk as iodine-rich food sources. Consequently, labeling milk and dairy products as an iodine-rich food source should be considered. Public authorities can consider the results from this survey in promotional campaigns to improve the awareness of different iodine sources and their beneficial effect on health.

Intermittent hemodialysis (IH) is an important therapy in the context of kidney dysfunction in dogs. However, its impact on pro-inflammatory cytokines is unclear. This study assessed IL-1β, IL-6, and TNF-α serum concentrations in dogs with chronic kidney disease (CKD) undergoing one session of IH without bypass (IH group, n = 4) and with bypass (IH + bypass group, n = 4). The control group (CG) included four healthy dogs. Cytokine levels were measured before, during, and after the first IH session. Comparative analyses of each cytokine within each group and time point were performed, along with a global comparison between groups. No significant changes were observed in cytokines across evaluation times in the IH groups. IL-1β was significantly higher post-session in the IH and IH + bypass groups compared to CG. Globally, IL-1β and TNF-α concentrations were significantly higher in the IH (11.41 pg/mL (10–16.17) and 2 pg/mL (2–88.54), respectively) and IH + bypass groups (10 pg/mL (10–10) and 2 pg/mL (2–215.5), respectively) compared to CG (0.96 pg/mL (0–3.56) and 0 pg/mL (0–0.003), respectively). The IH group also showed elevated IL-6 concentration (0.1 ng/mL (0.1–0.5)) compared to CG (0 ng/mL (0–0.1)). Higher IL-1β and IL-6 concentrations were observed in the IH group (11.41 pg/mL (10–16.17) and 0.1 ng/mL (0.1–0.1), respectively) compared to the IH + bypass group (10 pg/mL (10–10) and 0.1 ng/mL (0.1–0.5), respectively). In conclusion, a single IH session, with or without bypass, did not increase pro-inflammatory cytokines in CKD dogs with uremic syndrome but maintained the inflammatory state. Dogs undergoing IH without bypass may receive a stronger stimulus for cytokine release.

It includes new records, exclusions, and status changes from casual to naturalized or invasive for Italy or for Italian administrative regions for taxa in the genera Acacia, Akebia, Allocasuarina, Aloë, Brachychiton, Convolvulus, Freesia, Gleditsia, Hesperocyparis, Kalanchoë, Liriope, Mazus, Melia, Phyla, Platycladus, Prunus, and Retama. Abstract Radon originating from meta-volcanics located at the bottom of Bossea karstic mountain aquifer is subject to transport within the cave through both advective and diffusive processes. According to the double convective cell model of circulation referable to this hypogeal atmosphere, it is possible to correlate radon fluctuations with hydrodynamics, excluding the influence of external meteo-climatic factors. Transfer of gas from underground water flows presents delayed concentration maxima compared to flood peaks, amounting to 45 h in the upper meta-carbonate canyon and 66 h in the collapse chambers located at the contact with the meta-volcanics. Furthermore, different radon concentrations in water can be detected between the main creek and secondary inflows. Although both are subjected to a piston flow hydrodynamic response, the former shows an increase in radon during floods (with a lag of 46 h) due to the mobilization of long-resident water volumes in karst fractures, while in the latter case, there is a dilution of water in contact with radioactive elements of the impermeable basement (delayed by 6 h and caused by volumes of neo-infiltration). The analysis of radon emissions related to the site seismicity reveals no relationship, likely due to the low magnitudes and the predominant effect of hydrodynamics.

Abstract Background Understanding environmental correlates of sedentary behaviour (SB) among young people is important as such data can identify approaches to limit sedentary time. This paper estimates associations of parent-reported neighbourhood and adolescent-reported home environments with SB among adolescents aged 11–19 years from 14 countries. Methods In the International Physical activity and the Environment Network (IPEN) Adolescent Study (an observational, cross-sectional multi-country study), adolescents wore a triaxial accelerometer for seven days that assessed sedentary time (ST). Adolescents completed survey measures of sedentary behaviour (SB) related to recreational screen time and sitting time in motor vehicles. Parents and adolescents completed surveys assessing neighbourhood and home environments. Accelerometer based ST was available in 3,982 adolescents while survey data were available for 6,302 dyads. We estimated the total and direct effects of each environmental attribute on ST and SB. Sex of the adolescent and city/country were examined as moderators. Results The average ST in adolescents from 14 countries ranged from 7.8 to 10.5 h/day. Personal social media was the only significant correlate of total ST across both sexes. With respect to self-reported SB, adolescents accumulated an average of 3.8 h of non-school screen time per day and nearly 40 min of transport-related sitting time. Screen time was associated with all home environment variables, including social media account, as well as land use mix—diversity, traffic safety, and crime safety. Transport-related sitting time was related to land use mix—diversity, recreation facilities, walking facilities, and pedestrian infrastructure, but no home environment variables. City/country and sex were significant moderators of several associations. Conclusions Both home and neighbourhood environment features were related to ST and SB. Having social media accounts emerged as a major contributor towards sedentarism in adolescents.

Purpose – This teaching case aims to analyze the business model of Amaro, a company that directly sells lifestyle goods to end consumers (B2C), focused on the women's market. Amaro's original business model is introduced and a challenging dilemma is proposed: could Amaro innovate its business model, moving from a digitally native vertical brand (DNVB)-type company to a platform, specifically a vertical marketplace focused on the female audience? Would Amaro be prepared for this evolution or would it be more appropriate to focus on or strengthen the DNVB model? Design/methodology/approach – This teaching case was developed based on in-depth interviews with Amaro's leadership. The teaching notes were proposed based on business model innovation, competitive positioning and market trends concepts. The teaching case considers a new type of business model called DNVB. Students can review the concepts, create analyses and recommend which strategic options can leverage the company for a new growing cycle. Findings – Using the case study in the classroom should promote the discussion and reflections on business model innovation and the future of retail in omnichannel contexts - Amaro offers products online (on an e-commerce platform and native mobile applications) and physically in locations called guide shops. The authors suggested the adoption of frameworks and tools (e.g. the competitive positioning map to allow students to visualize ways to compare strategies and make decisions). Research limitations/implications – The case introduces a fictional dilemma related to the decision to maximize offline or online investments or completely change the company's business model by adding a new vertical marketplace approach. Practical implications – This teaching case contributes to the student's learning about business model innovation and evolution. Case discussions could explore contemporary concepts such as value proposition, disintermediation and omnichannel commerce. Originality/value – Offering goods directly to the consumers by using modern technological architecture through vertical integration within the supply chain makes the DNVB business model an original topic in the start-up segment.

Abstract Background The cultivation of maize (Zea mays L.), one of the most important crops worldwide for food, feed, biofuels, and industrial applications, faces significant constraints due to Fusarium verticillioides, a fungus responsible for severe diseases including seedling blights, stalk rot, and ear rot. Its impact is worsened by the fact that chemical and agronomic measures used to control the infection are often inefficient. Hence, genetic resistance is considered the most reliable resource to reduce the damage. This study aims to elucidate the genetic basis of F. verticillioides resistance in maize. Results Young seedlings of eight divergent maize lines, founders of the MAGIC population, were artificially inoculated with a F. verticillioides strain. Phenotypic analysis and transcriptome sequencing of both control and treated samples identified several hundred differentially expressed genes enriched in metabolic processes associated with terpene synthesis. A WGCNA further refined the pool of genes with potential implications in disease response and found a limited set of hub genes, encoding bZIP and MYB transcription factors, or involved in carbohydrate metabolism, solute transport processes, calcium signaling, and lipid pathways. Finally, additional gene resources were provided by combining transcriptomic data with previous QTL mapping, thereby shedding light on the molecular mechanisms in the maize-F. verticillioides interaction. Conclusions The transcriptome profiling of eight divergent MAGIC maize founder lines with contrasting levels of Fusarium verticillioides resistance combined with phenotypic analysis, clarifies the molecular mechanisms underlying the maize-F. verticillioides interaction.

Objetivo:Comparar a prevalência da síndrome metabólica (SM) com diferentes critérios de classificação e perfil lipídico em idosos remanescentes dos quilombos. Métodos: Foram avaliados 110 idosos, 54 remanescentes dos quilombos (IRQ), média de idade 71,1 ± 9,6 anos. A classificação para SM ocorreu pela International Diabetes Federation (IDF) e National Cholesterol Education Program’s Adult Treatment Panel III (NCEP/ATPIII). Os IRQ foram pareados e comparados a um grupo controle de 56 idosos não remanescentes dos quilombos (INRQ) com idade média 68,8 ± 5,7 anos. Foram submetidos a uma entrevista, avaliação clínica, antropométrica e bioquímica. Resultados: A prevalência de SM entre os IRQ foi de 18,5% e 38,9%, e nos INRQ foi de 34% e 64,3%, pelos critérios do NCEP/ATPIII e da IDF. Pelo NCEP/ATPIII, o critério de SM mais prevalente foi baixo HDL-c. No perfil lipídico dos grupos, houve diferença de colesterol total (p ≤ 0,037), (VLDL p ≤ 0,001) e triglicerídeos (p ≤ 0,001). Os IRQ apresentaram redução na glicemia de jejum e triglicerídeos. Conclusão: Os IRQ apresentaram menor prevalência de SM em ambas as definições e valores significativamente menores de triglicerídeos, colesterol total e VLDL-c.

Introdução: Os idosos representam uma população crescente no Brasil e no mundo. As alterações fisiológicas decorrentes da idade, as comorbidades e a necessidade de polifarmacoterapia os tornam altamente susceptíveis à ocorrência de eventos adversos relacionados a medicamentos. A conciliação medicamentosa mostra-se estratégia útil para reduzir esses eventos, quando o paciente transita entre diferentes níveis de cuidado à saúde. Objetivo: Realizar uma revisão de literatura sobre conciliação medicamentosa em idosos na transição do cuidado à saúde. Metodologia: Trata-se de revisão da literatura realizada na base de dados PubMed. Os critérios de inclusão foram: Enfoque na população idosa; idioma inglês, espanhol ou português; publicação nos últimos 5 anos. Dois revisores examinaram títulos e resumos e procederam à leitura integral e à inclusão dos artigos relevantes. Resultados: Doze estudos foram incluídos, dentre os quais a metade descreveu as discrepâncias de medicamentos nos pontos de transição, revelando como principal a omissão de algum fármaco; 3 avaliaram a contribuição do farmacêutico para o processo, evidenciando maior qualidade com o envolvimento desse profissional; e 5 analisaram a consistência das informações prestadas na continuidade do cuidado e o impacto da conciliação de medicamentos sobre os erros de medicação, demonstrando a importância da coleta de uma história medicamentosa inicial precisa para redução de eventos adversos, polifarmácia e erros de medicação. Conclusão: A conciliação medicamentosa revelou-se importante para a segurança do paciente idoso na transição do cuidado à saúde. A atuação do farmacêutico e a integridade da história farmacoterápica relacionaram-se a uma melhoria no processo, aprimorando a assistência prestada ao paciente.

Objetivo: Este estudo buscou compreender o significado e os sentimentos que envolvem o processo de aprendizagem, o vínculo com o computador e a associação das palavras tecnologia, computador e informática para participantes de um grupo de convivência. Método: Tratou-se de uma pesquisa quantiqualitativa com 20 sujeitos com idade igual ou superior a 50 anos. Os dados quantitativos foram descritos por meio de estatística descritiva. Os de cunho qualitativo foram analisados pelo método de análise de conteúdo. Resultados: Constatou-se que o processo de aprendizagem, mediado pelo computador, proporcionou mudanças positivas na vida desses sujeitos, promovendo o aprendizado, a interação, a formação de círculo de amizades e elevação da autoestima. Além disso, revelou- se que a associação das palavras tecnologia, computador e informática representam um grande avanço e facilidade na vida das pessoas. Conclusão: Da interação com o computador se promoveu a inclusão digital, a conquista de um espaço sociodigital, de novas amizades e a valorização do idoso enquanto pessoa.

Abstract Seed quality is the set of physical, genetic, and physiological characteristics, reflecting the overall germination potential. Maintaining an optimal seed quality is essential for agriculture and seed banks to preserve genetic diversity. Compared to conventional methods (e.g., germination tests), non-invasive approaches allow a more sustainable and rapid evaluation of seed quality but this is limited by high costs. The measurement of ultra-weak photon emission (UPE) and delayed fluorescence (DL), defined as biological phenomena potentially related to the physiological status of living systems, may represent a suitable approach to estimate seed quality. To test this hypothesis, seeds of five agriculturally relevant legume species (Phaseolus vulgaris L., Lathyrus sativus L., Cicer arietinum L., Pisum sativum L., and Vicia faba L.), stored at different conditions (room temperature or -18 °C) for several years, were analysed using a LIANA© prototype to collect data regarding DL and UPE occurring after UV excitation. The obtained data were integrated with germination parameters which underline species-specific behaviours in response to storage conditions. The prediction models show variable efficiency in classifying seeds based on germination which underline species-dependent links between photon emission and seed quality. Therefore, these measurements represent novel, non-invasive, and rapid approaches to evaluate seed quality.

Abstract The aim of this pilot study was to assess the compliance of breast cancer (BC) patients with fitness tracker (FT) monitoring program during radiotherapy (RT) and to characterize radiation-induced fatigue (RIF) status through objective evaluation using FT-collected parameters. Thirty-six BC patients were invited to wear FT during their RT course for continuous monitoring of heart rate (HR) and step counts (STP). RIF assessment was performed weekly, according to CTCAE v5.0 and dichotomized into G0 vs. any-grade. A novel concept based on patient Repeated Activity Window (RAW) was introduced to evaluate HR and STP variations during RT. Several Machine Learning (ML) methods were trained to characterize RIF on the basis of HR and STP collected data. RIF of any grade was reported by 17 out of 36 patients (47%) included in the study. None of patient clinical variables were significantly correlated with RIF. All patients accepted the FT monitoring program, and for 32 patients FT collection efficiency was greater than 60%. For each patient, a distinct distribution of RAWs was identifiable over RT and across the entire patient cohort, with a total of 7950 RAWs processed. Six features related to RAWs, HR and STP were identified as associated with RIF. The best-performing classifier was the Bagged Trees model, showing a cross-validated ROC-AUC of 89% (95% CI 88–90%). This study confirms the feasibility of continuous biomedical monitoring of BC patients by FT. We successfully identify objective indicators of RIF through HR and STP variation measures within each patient’s RAW, thus providing a novel and practical approach to assess and manage RIF. This can significantly aid medical staff in evaluating RIF trajectories, potentially leading to better individualized care strategies and improved patient outcomes.

Abstract Agonist-stimulated GPCR endocytosis typically occurs via the multi-faceted adaptor proteins known as βarrestins. However, endocytosis of several GPCRs occurs independently of β-arrestins, suggesting an additional mode of GPCR endocytosis, but the mechanisms remain unknown. Here we provide evidence that sorting nexin 9 (SNX9), a previously described endocytic remodeling protein, functions as a novel cargo adaptor that promotes agonist-stimulated GPCR endocytosis. We show that SNX9 and SNX18, but not β-arrestins, are necessary for endocytosis of the chemokine receptor CXCR4. SNX9 is recruited to CXCR4 at the plasma membrane and interacts directly with the carboxyl-terminal tail of the receptor in a phosphorylation-dependent manner. We also provide evidence that some receptors do not require SNX9 and SNX18 nor β-arrestins for endocytosis, suggesting additional modes for GPCR endocytosis. These results provide novel insights into the mechanisms regulating GPCR trafficking and broaden our overall understanding of GPCR regulation.

Abstract Background The impact of the COVID-19 pandemic on breast cancer care across Brazilian regions with varying Human Development Index (HDI) levels remains unclear. This study evaluates the pandemic’s effects on screening mammograms, tumor staging at diagnosis, and treatment initiation in the Brazilian Public Health System between 2017 and 2022, focusing on regions with different HDI levels. Methods This ecological time series study uses an inflection point regression model and monthly percentage change (MPC) to analyze an open-access Brazilian Public Healthcare System dataset. The study focuses on trends and variations in these variables among women aged 50–69 in three state groups classified by HDI: Group A (very high), Group B (medium), and Group C (high). Results The average monthly rate of screening mammograms was highest in Group A (10.70) and lowest in Group B (8.38). At the onset of the COVID-19 pandemic, screening rates dropped significantly, with the most significant decline in Group B (58.6% decrease) and the smallest in Group A (45.7% decrease), lasting for three months. Subsequently, this variable recovered until December 2022 but was insufficient to restore the total series MPC to pre-pandemic levels. Group B had the lowest average rate of early-stage (0-II) diagnoses (2.88), while Group C had the highest (3.68). Early-stage diagnoses declined in the first three months of the pandemic, followed by a partial recovery that was insufficient to restore the pre-pandemic MPC levels. The proportion of advanced-stage diagnoses was highest in Group B (49.02%) and lowest in Group A (45.97%). The pandemic maintained the upward trend of advanced-stage (III-IV) diagnoses across all groups in the total time series. The average proportion of treatments initiated after 60 days of diagnosis was above 60% across all groups, with Group B at 64.50%. This variable began to rise 3 to 4 months after the pandemic and continued increasing until December 2022, with a greater intensity than the pre-pandemic period across all groups. Conclusion The COVID-19 pandemic reduced breast cancer screening and early diagnosis in Brazil, leading to more advanced cases and treatment delays across all regions, with varying impacts according to regional HDI levels.

O presente artigo tem por objetivo explorar o desenvolvimento do direito de propriedade no Brasil, do ponto de vista jurídico e histórico. A partir da vertente metodológica jurídico-dogmática, foi possível investigar as relações normativas interdisciplinares referentes ao direito de propriedade, analisando a sua ideal aplicabilidade e hermenêutica no ordenamento jurídico pátrio. Desenvolveu-se a pesquisa por meio do método dialético e da pesquisa histórica, a fim de alcançar o objeto pela formulação de contradição à realidade e criação do conflito. Tendo em vista todo o desenvolvimento do direito analisado no Brasil, ficará evidenciado que a função social da propriedade, atualmente, representa um elemento estruturante do direito, que ainda deve ser respeitado e elencado como um dos pilares das legislações internas.

Abstract Background The COVID-19 pandemic has overwhelmed the healthcare systems of many countries and negatively impacted the care of other diseases. Objective To evaluate the trend of screening mammograms, oncological breast surgeries, and breast cancer hospitalizations in Brazil’s public and private healthcare system between 2014 and 2022. Method This ecological time series study uses the inflection point regression model and semester percentage change (SPC). We use the open-access dataset of the different healthcare systems in Brazil. We analyzed the trend of the variables in the pre-pandemic and the effect of the pandemic on the total time series. Results In 2020, compared to 2019, the decrease in screening mammograms, oncological breast surgeries, and breast cancer hospitalization was − 41.44%, -23.13%, and − 10.52% (public health system) and − 29.49%, -18.96%, and − 15.35% (private healthcare system). In the public healthcare system, the pandemic has enhanced the decreasing trend of mammograms (SPC − 1.6% before and − 3.4% after), has reverted the stationary trend of oncological breast surgeries to decreasing (SPC − 1.0%), has slowed the increasing trend of breast cancer hospitalization (SPC 1.8% before and 0.9% after). In the private healthcare system, the pandemic has reverted the stationary trend of mammograms to decreasing (SPC − 1.0%), has slowed the increasing trend of breast cancer surgeries (SPC 2.3% before and 0.8% after), has reverted the growing trend of breast cancer hospitalization (SPC 3.9%) to stationary. Conclusion During the COVID-19 pandemic, there was an increase in inequalities between healthcare systems, especially in breast cancer screening.

O racismo estrutural e o projeto moderno-colonizador em curso na América Latina não seriam os mesmos sem as contribuições da ciência moderna ocidental. Contraditoriamente, é também por meio das ciências — sobretudo das ciências sociais — que as colonialidades têm sido contestadas. Inspirados pela crítica decolonial, contamos a história de vida de Waxihô Karajá, um homem indígena Karajá Xambioá que ingressou e abandonou um curso de Biologia. Os resultados ilustram como a educação indígena Iny contribuiu para o desenvolvimento do interesse de Waxihô Karajá por Biologia. Nossa análise também revela que a baixa qualidade da educação básica ofertada em comunidades indígenas; o despreparo das instituições de educação superior para receber a diversidade; as dificuldades experimentadas por jovens oriundos de realidades de baixa escolarização e a energia dispensada por minorias culturais para combater uma miríade de violências coloniais por meio do ativismo estudantil podem comprometer o sucesso acadêmico. Os resultados indicam que a aproximação e o afastamento de cursos de ciências da natureza por jovens indígenas parecem ter motivos ora similares ora distintos daqueles elencados pela pesquisa com jovens da sociedade envolvente. Urge reorientar as políticas de permanência universitária para fomentar o sucesso acadêmico de estudantes indígenas em cursos de ciências da natureza.

Structural racism and the ongoing modern-colonizing project in Latin America would not be the same without the contributions of Modern Western Science. Contradictorily, it is also through the sciences — especially social sciences — that colonialities have been challenged. Inspired by decolonial criticism, we tell the life story of Waxihô Karajá, an indigenous Karajá Xambioá man who entered and abandoned a Biology course. The results illustrate how Iny indigenous education contributed to the development of Waxihô Karajá's interest in Biology. Our analysis also reveals that the low quality of basic education offered in indigenous communities; the unpreparedness of higher education institutions to receive diversity; the difficulties experienced by young people with low-education backgrounds and the energy expended by cultural minorities to combat a myriad of colonial violences through student activism can compromise academic success. Results indicate that the approach and the departure from natural science courses by young indigenous people seem to have reasons that are sometimes similar and sometimes different from those listed in the research with young people from the national society. There is an urgent need to reorient university retention policies to promote the academic success of indigenous students in natural sciences courses.

Abstract We presented a method to find potential cancer attractors using single-cell RNA sequencing (scRNA-seq) data. We tested our method in a Glioblastoma Multiforme (GBM) dataset, an aggressive brain tumor presenting high heterogeneity. Using the cancer attractor concept, we argued that the GBM’s underlying dynamics could partially explain the observed heterogeneity, with the dataset covering a representative region around the attractor. Exploratory data analysis revealed promising GBM’s cellular clusters within a 3-dimensional marker space. We approximated the clusters’ centroid as stable states and each cluster covariance matrix as defining confidence regions. To investigate the presence of attractors inside the confidence regions, we constructed a GBM gene regulatory network, defined a model for the dynamics, and prepared a framework for parameter estimation. An exploration of hyperparameter space allowed us to sample time series intending to simulate myriad variations of the tumor microenvironment. We obtained different densities of stable states across gene expression space and parameters displaying multistability across different clusters. Although we used our methodological approach in studying GBM, we would like to highlight its generality to other types of cancer. Therefore, this report contributes to an advance in the simulation of cancer dynamics and opens avenues to investigate potential therapeutic targets.

Abstract Background Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disease that results in death within a short time span (3-5 years). One of the major challenges in treating ALS is its highly heterogeneous disease progression and the lack of effective prognostic tools to forecast it. The main aim of this study was, then, to test the feasibility of predicting relevant clinical outcomes that characterize the progression of ALS with a two-year prediction horizon via artificial intelligence techniques using routine visits data. Methods Three classification problems were considered: predicting death (binary problem), predicting death or percutaneous endoscopic gastrostomy (PEG) (multiclass problem), and predicting death or non-invasive ventilation (NIV) (multiclass problem). Two supervised learning models, a logistic regression (LR) and a deep learning multilayer perceptron (MLP), were trained ensuring technical robustness and reproducibility. Moreover, to provide insights into model explainability and result interpretability, model coefficients for LR and Shapley values for both LR and MLP were considered to characterize the relationship between each variable and the outcome. Results On the one hand, predicting death was successful as both models yielded F1 scores and accuracy well above 0.7. The model explainability analysis performed for this outcome allowed for the understanding of how different methodological approaches consider the input variables when performing the prediction. On the other hand, predicting death alongside PEG or NIV proved to be much more challenging (F1 scores and accuracy in the 0.4-0.6 interval). Conclusions In conclusion, predicting death due to ALS proved to be feasible. However, predicting PEG or NIV in a multiclass fashion proved to be unfeasible with these data, regardless of the complexity of the methodological approach. The observed results suggest a potential ceiling on the amount of information extractable from the database, e.g., due to the intrinsic difficulty of the prediction tasks at hand, or to the absence of crucial predictors that are, however, not currently collected during routine practice.

Abstract Liquid-liquid phase separation (LLPS) in living cells provides innovative pathways for synthetic compartmentalized catalytic systems. While LLPS has been explored for enhancing enzyme catalysis, its potential application to catalytic peptides remains unexplored. Here, we demonstrate the use of coacervation, a key LLPS feature, to constrain the conformational flexibility of catalytic peptides, resulting in structured domains that enhance peptide catalysis. Using the flexible catalytic peptide P7 as a model, we induce reversible biomolecular coacervates with structured peptide domains proficient in hydrolyzing phosphate ester molecules and selectively sequestering phosphorylated proteins. Remarkably, these coacervate-based microreactors exhibit a 15,000-fold increase in catalytic efficiency compared to soluble peptides. Our findings highlight the potential of a single peptide to induce coacervate formation, selectively recruit substrates, and mediate catalysis, enabling a simple design for low-complexity, single peptide-based compartments with broad implications. Moreover, LLPS emerges as a fundamental mechanism in the evolution of chemical functions, effectively managing conformational heterogeneity in short peptides and providing valuable insights into the evolution of enzyme activity and catalysis in prebiotic chemistry.

Abstract Mutations in GBA1 encoding the lysosomal enzyme β-glucocerebrosidase (GCase) are among the most prevalent genetic susceptibility factors for Parkinson’s disease (PD), with 10–30% of carriers developing the disease. To identify genetic modifiers contributing to the incomplete penetrance, we examined the effect of 1634 human transcription factors (TFs) on GCase activity in lysates of an engineered human glioblastoma line homozygous for the pathogenic GBA1 L444P variant. Using an arrayed CRISPR activation library, we uncovered 11 TFs as regulators of GCase activity. Among these, activation of MITF and TFEC increased lysosomal GCase activity in live cells, while activation of ONECUT2 and USF2 decreased it. While MITF, TFEC, and USF2 affected GBA1 transcription, ONECUT2 might control GCase trafficking. The effects of MITF, TFEC, and USF2 on lysosomal GCase activity were reproducible in iPSC-derived neurons from PD patients. Our study provides a systematic approach to identifying modulators of GCase activity and deepens our understanding of the mechanisms regulating GCase.

There is evidence that anxiety and avoidance toward close relationships (i.e., insecure attachment orientations), as well as posttraumatic stress symptoms (PTSSs), are linked to a poor quality of life. The current study aimed to investigate the potential mediating effects of PTSSs on the associations between insecure attachment orientations and domains of quality of life. A convenience sample of 497 adults (375 females, 75.5%), ranging in age between 18 and 65 years old (M = 32.48, SD = 13.26), was recruited. Participants were administered self-report instruments assessing attachment anxiety and avoidance, PTSSs, and domains of quality of life, including physical health, psychological status, social relationships, and environment. A series of mediation analyses were performed to test the mediating role of PTSSs in the relationships between attachment orientations and domains of quality of life. Results showed that attachment anxiety was related to decreased levels of quality of life in all domains, and that their associations were mediated by PTSSs. Also, attachment avoidance was related to a worse quality of psychological status and social relationships, and PTSSs were a significant mediating variable in these associations. Prevention programs and clinical interventions focused on promoting effective strategies for managing distress might be critical in reducing the impact of distressing events on the quality of life of individuals with insecure attachment.

The study aimed to select rice genotypes based on agronomic traits and commercial grain quality. The study was carried out in the 2014/15 and 2015/16 harvests in eight growing environments, located in the state of Rio Grande do Sul, Brazil. Fourteen rice genotypes were sown in each environment. The experiment was conducted in a randomized block design with four replications. The following agronomic traits were evaluated: number of days to flowering and grain yield as well as commercial grain quality traits: percentage of whole grains and percentage of chalky area in the grains. AMMI, GGE and MGIDI methodologies were used to select the best performing genotypes. Using the AMMI, GGE and MGDI methods, genotypes G9 and G1 performed close to the ideal ideotype for the number of days to grain flowering, yield and quality.

Knowledge of the family Assamiidae in the Eastern Himalayas is primarily concentrated from Nepal, while Bhutan, Xizang, and northeastern India remain much less studied. Herein, a new genus, Linzhiassamia gen. nov., is described from Xizang, China, along with two new species, Linzhiassamia medogensis sp. nov. and Linzhiassamia zayuensis sp. nov. These represents the first records of the family Assamiidae from Xizang. A comparative analysis with other continental Asian Assamiidae is undertaken, with a focus on genital morphology. Potential closest relatives are identified, including the genus Dhaulagirius Martens, 1977 from Nepal.

Within a practical course of cytotaxonomy organized in Pisa (Italy) on February 2024 by the Group for Floristics, Systematics and Evolution of the Italian Botanical Society, we tested whether relevant differences in karyotype asymmetry and structure occur in four tetraploid European taxa from Paeonia sect. Paeonia (P. mascula subsp. russoi, P. officinalis subsp. huthii, P. officinalis subsp. italica, and P. peregrina). Our results point towards a homogeneous karyotype asymmetry and structure among studied tetraploid species, with no statistically significant difference among taxa and high overlap in variation highlighted by PCA.

Abstract We describe the geographical variation in tree species composition across Amazonian forests and show how environmental conditions are associated with species turnover. Our analyses are based on 2023 forest inventory plots (1 ha) that provide abundance data for a total of 5188 tree species. Within-plot species composition reflected both local environmental conditions (especially soil nutrients and hydrology) and geographical regions. A broader-scale view of species turnover was obtained by interpolating the relative tree species abundances over Amazonia into 47,441 0.1-degree grid cells. Two main dimensions of spatial change in tree species composition were identified. The first was a gradient between western Amazonia at the Andean forelands (with young geology and relatively nutrient-rich soils) and central–eastern Amazonia associated with the Guiana and Brazilian Shields (with more ancient geology and poor soils). The second gradient was between the wet forests of the northwest and the drier forests in southern Amazonia. Isolines linking cells of similar composition crossed major Amazonian rivers, suggesting that tree species distributions are not limited by rivers. Even though some areas of relatively sharp species turnover were identified, mostly the tree species composition changed gradually over large extents, which does not support delimiting clear discrete biogeographic regions within Amazonia.

Abstract Objective This study introduces the complete blood count (CBC), a standard prenatal screening test, as a biomarker for diagnosing preeclampsia with severe features (sPE), employing machine learning models. Methods We used a boosting machine learning model fed with synthetic data generated through a new methodology called DAS (Data Augmentation and Smoothing). Using data from a Brazilian study including 132 pregnant women, we generated 3,552 synthetic samples for model training. To improve interpretability, we also provided a ridge regression model. Results Our boosting model obtained an AUROC of 0.90±0.10, sensitivity of 0.95, and specificity of 0.79 to differentiate sPE and non-PE pregnant women, using CBC parameters of neutrophils count, mean corpuscular hemoglobin (MCH), and the aggregate index of systemic inflammation (AISI). In addition, we provided a ridge regression equation using the same three CBC parameters, which is fully interpretable and achieved an AUROC of 0.79±0.10 to differentiate the both groups. Moreover, we also showed that a monocyte count lower than $$490 /mm^{3}$$ 490 / m m 3 yielded a sensitivity of 0.71 and specificity of 0.72. Conclusion Our study showed that ML-powered CBC could be used as a biomarker for sPE diagnosis support. In addition, we showed that a low monocyte count alone could be an indicator of sPE. Significance Although preeclampsia has been extensively studied, no laboratory biomarker with favorable cost-effectiveness has been proposed. Using artificial intelligence, we proposed to use the CBC, a low-cost, fast, and well-spread blood test, as a biomarker for sPE.

Thermal modification is a process capable of improving properties affecting wood performance, such as biological durability. This study aimed to assess the potential of thermal modification in enhancing the resistance of Couratari sp. wood to deterioration by Trametes versicolor, Nasutitermes corniger, and Cryptotermes brevis. Five treatments were analyzed, represented by the untreated and thermally modified wood at 180, 190, 200, and 210 °C. The wood’s chemical composition and resistance to biodeterioration in laboratory tests were evaluated. Thermal modification, especially at 210 °C, altered the chemical composition and increased the wood’s durability class against the fungus. However, the process did not affect termite attack. There was a significant positive correlation between corrected mass loss and hemicellulose and total extractive contents, as well as mass loss caused by T. versicolor and lignin and hemicellulose contents. The use of thermal modification in 190 °C is recommended for Couratari sp. wood due to its enhanced biological durability, with 210 °C being particularly effective.

The potential of spaceborne global navigation satellite system reflectometry (GNSS-R) to retrieve a variety of geophysical parameters has already been demonstrated in numerous studies. In 2021, China successfully launched the Fengyun-3E (FY-3E) polar orbit satellite. It carries the GNSS occultation sounder-II (GNOS-II) that can simultaneously receive reflected signals from the global positioning system, BeiDou, and Galileo constellations. Multiconstellation measurement significantly reduces the revisit time of the spaceborne GNSS-R data. This offers the possibility to study the sensitivity of the reflected signals from multi-GNSS constellations to surface parameters at global scale. The main objective of this article is to analyze the sensitivity of FY-3E GNOS-II surface reflectivity (SR) to soil moisture (SM), vegetation, and surface roughness for different GNSS constellations, and different incidence angles. FY-3E data from May 2023 to October 2023 were collected along with SM data from the soil moisture active passive (SMAP). The SMAP static auxiliary surface roughness parameter SMAP-h was also collected for subsequent comparisons. Furthermore, the effect of vegetation in the reflected signal was accounted for using the SM and ocean salinity INRA-CESBIO L-band vegetation optical depth. Afterward, for each GNSS constellation, the SR was binned as a function of SM, SMAP-h, and incidence angle. The results indicate that the sensitivity for the different GNSS constellations is consistent, showing similar behavior. Moreover, this study also reports for the first time the experimentally computed sensitivity of FY-3E SR to SM under different SMAP-h values. The sensitivity of GNOS-II SR to SM is in general agreement with the values in the previous studies, demonstrating the feasibility of using single-pass multi-GNSS constellation GNSS-R data to retrieve surface parameters, such as SM, with shorter revisit times. Furthermore, the results of this study re-emphasize the nonlinear sensitivity of SR to SM. As compared with higher SM, SR is more sensitive at lower SM. This highlights the shortcomings of using linear models to retrieve SM. Meanwhile, another important finding is that, for spaceborne GNSS-R data, SMAP-h may have underestimated the effective surface roughness.

Goal: Organ perfusion is a vast subject with several techniques, the most common being Hypothermic Machine Perfusion (HMP) and Normothermic Machine Perfusion (NMP). In this paper, a new machine developed at the Biolab of the University of Florence has been tested to validate its capability to perform a well-controlled Oxygenated Rewarming (COR) phase and maintain stability during the NMP phase. Methods: The tests (n = 5) were conducted on fresh porcine livers and evaluated according to liver perfusion standards. The developed machine is based on a previous version, with an integrated control and sensor system and a complete mechanical and electronic redesign. Results: The results demonstrate the excellent usability of the machine and its ability to effectively maintain the organs in good condition. The new system performed well, and the measures made on the livers were satisfactory for good preservation of the organ. Conclusion: This study showed the effectiveness of the developed machine. Future development of the system will include a more sophisticated control system to ensure the correct parameters for perfusion.

Abstract Introduction Photobiomodulation (PBM) has become a promising approach for slowing the progression of early and intermediate dry age-related macular degeneration (dAMD) to advanced AMD. This technique uses light to penetrate tissues and activate molecules that influence biochemical reactions and cellular metabolism. This preliminary analysis is aimed at assessing the safety, tolerability, and short-term effectiveness of the EYE-LIGHT®PBM treatment device in patients with dAMD. Methods The EYE-LIGHT® device employs two wavelengths, 590 nm (yellow) and 630 nm (red), in both continuous and pulsed modes. Patients over 50 years of age with a diagnosis of dAMD in any AREDS (Age-Related Eye Disease Study) category were randomly assigned to either the treatment group or the sham group. The treatment plan consisted of an initial cycle of two sessions per week for 4 weeks. Safety, tolerability, and compliance outcomes, along with functional and anatomical outcomes, were assessed at the end of the fourth month. Results This preliminary analysis included data from 76 patients (152 eyes). All patients were fully compliant with treatment sessions, and only one fifth of patients treated with PBM reported mild ocular adverse events, highlighting exceptional results in terms of tolerability and adherence. Changes in best-corrected visual acuity (BCVA) from baseline to month 4 differed significantly between the sham and PBM-treated groups, favoring the latter, with a higher proportion achieving a gain of five or more letters post-treatment (8.9% vs. 20.3%, respectively; p = 0.043). No significant differences in central subfield thickness (CST) were observed between the two groups over the 4-month period. The study also found a statistically significant disparity in mean drusen volume changes from baseline to month 4 between the groups in favor of patients treated with PBM (p = 0.013). Conclusion These preliminary results indicate that PBM treatment using the EYE-LIGHT® system is safe and well tolerated among patients with dAMD. Furthermore, both functional and anatomical data support the treatment’s short-term efficacy. Trial Registration ClinicalTrials.gov identifier NCT06046118.

Background: Increasingly valuable, sophisticated, and powerful aircraft are developed and incorporated into the armed forces. Technological advances are notable for increasing physiological load during the flight, making human resources a limiting factor in such operations. It is necessary to understand the physiological demands to which aircraft pilots are exposed, especially combat pilots. Objective: This study aimed to present: i. the physiological consequences on the human body due to high accelerative forces during high-performance flights; ii. the efficiency of the Anti-G Straining Maneuver (AGSM) countermeasure to such consequences; and iii. the state of the art about surface electromyography's uses (sEMG) in AGSM. Method: It was conducted a bibliographic search in the PubMed database using the keywords “anti-G effort maneuver” and “electromyography” and their synonyms. After this, a narrative review of the results was performed. This research model allows a comprehensive discussion about the topic, enabling an in-depth contextualization in a prosperous field of study. These characteristics are suitable for detecting literature gaps and directing the research discussion. Discussion: There is a consensus in the literature that AGSM is the most effective resource to prevent loss of consciousness induced by G-load (G-LOC). The sEMG is an affordable and useful tool for AGSM training, especially given the unavailability of human centrifuges. The current literature on the application of sEMG in the context of AGSM training is limited, presenting a diversity of objectives, methodology, and analyzed parameters. Conclusion: The use of sEMG as real-time biofeedback can improve the pilot's muscle control skills during AGSM.

Muitos são os riscos ocupacionais inerentes a aviação que podem estar associados ao desenvolvimento de desconfortos musculoesqueléticos e lesões. A atividade operacional do piloto na cabine da aeronave é complexa, é necessário que ele execute tarefas simultâneas, expondo-os a riscos físicos além das demandas inerentes da aviação operacional militar. Objetivos: Descrever a prevalência de desconforto musculoesquelético em pilotos militares de transporte da Força Aérea Brasileira e suas possíveis associações com os parâmetros biométricos, horas de voo e frequência de prática de atividade física. Método: Trata-se de um estudo observacional descritivo, com amostra composta por pilotos militares de aeronave de transporte. Como instrumentos foram utilizados questionários de autopreenchimento para caracterização da história ocupacional, perfil de atividade física e histórico de desconforto físico. Resultados: Um total de 26 pilotos foram incluídos no estudo. A frequência de atividade física em média 4,3 ± 1,59 vezes por semana, 46,15% dos sujeitos foram classificados como muito ativos. Na avaliação de desconforto, os segmentos corporais mais frequentes foram: costas inferior com 73,1%, seguido da cervical com 42,2 % e pescoço com 34,6 % dos resultados. Conclusão: Os desconfortos da coluna vertebral, cervical e lombar, foram os com maior prevalência observados na população analisada, com a intensidade dor variando de leve a desconforto intenso. Apesar da amostra ter sido caracterizada como jovem e ativa quanto a prática de atividade física, a presença de desconforto se fez presente.

Background: Air traffic controllers (ATC) are subject to high cognitive and operational demands, exposing them to stress, anxiety and depression. Temporomandibular disorder (TMD) is the main disorder of non-odontogenic etiology for chronic facial pain that affects muscles and joints, and has been related to psychological distress. Therefore, ATC could be susceptible to TMD. Objective: This study verified the prevalence of TMD and its relationship with psychological distress in ATC. Methods: Brazilian ATC of both sexes participated in this study (n: 121; 27.9 ± 4.2 years old). Participants were asked to complete questionnaires about signs and symptoms of temporomandibular disorders: FAI; and about psychological distress: PSS-10; GAD-7; and PHQ-9. Inferential statistical analyzes for comparison between factors and correlation between dependent variables were adopted with 5.0% significance. Results: TMD was highly prevalent among ATC. More than half of ATC have at least one psychological disturbing condition; they include anxiety (n:13; 13.7%), stress (n: 58; 47.9%), depression (n: 24; 19.8%), and comorbidities (n: 22; 18.2%) among these psychological distressing conditions. Abstract PDZ domain mediated interactions with voltage-gated calcium (CaV) channel C-termini play important roles in localizing membrane Ca2+ signaling. The first such interaction was described between the scaffolding protein Mint-1 and CaV2.2 in mammals. In this study, we show through various in silico analyses that Mint is an animal-specific gene with a highly divergent N-terminus but a strongly conserved C-terminus comprised of a phosphotyrosine binding domain, two tandem PDZ domains (PDZ-1 and PDZ-2), and a C-terminal auto-inhibitory element that binds and inhibits PDZ-1. In addition to CaV2 chanels, most genes that interact with Mint are also deeply conserved including amyloid precursor proteins, presenilins, neurexin, and CASK and Veli which form a tripartite complex with Mint in bilaterians. Through yeast and bacterial 2-hybrid experiments, we show that Mint and CaV2 channels from cnidarians and placozoans interact in vitro, and in situ hybridization revealed co-expression in dissociated neurons from the cnidarian Nematostella vectensis. Unexpectedly, the Mint orthologue from the ctenophore Hormiphora californiensis strongly bound the divergent C-terminal ligands of cnidarian and placozoan CaV2 channels, despite neither the ctenophore Mint, nor the placozoan and cnidarian orthologues, binding the ctenophore CaV2 channel C-terminus. Altogether, our analyses suggest that the capacity of Mint to bind CaV2 channels predates bilaterian animals, and that evolutionary changes in CaV2 channel C-terminal sequences resulted in altered binding modalities with Mint.

Abstract The cellular prion protein (PrPC) plays many roles in the developing and adult brain. In addition, PrPC binds to several amyloids in oligomeric and prefibrillar forms and may act as a putative receptor of abnormal misfolded protein species. The role of PrPC in tau seeding and spreading is not known. In the present study, we have inoculated well-characterized sarkosyl-insoluble fractions of sporadic Alzheimer’s disease (sAD) into the brain of adult wild-type mice (Prnp +/+), Prnp 0/0 (ZH3 strain) mice, and mice over-expressing the secreted form of PrPC lacking their GPI anchor (Tg44 strain). Phospho-tau (ptau) seeding and spreading involving neurons and oligodendrocytes were observed three and six months after inoculation. 3Rtau and 4Rtau deposits from the host tau, as revealed by inoculating Mapt 0/0 mice and by using specific anti-mouse and anti-human tau antibodies suggest modulation of exon 10 splicing of the host mouse Mapt gene elicited by exogenous sAD-tau. However, no tau seeding and spreading differences were observed among Prnp genotypes. Our results show that PrPC does not affect tau seeding and spreading in vivo.

Abstract Background Ceramides have recently been identified as novel biomarkers associated with diabetes mellitus (DM) and major adverse cardiac and cerebrovascular events (MACCE). This study aims to explore their utility in diagnosing microvascular disease. Methods This study prospectively enrolled 309 patients from 2018 to 2020 into three groups: healthy controls (Group 1, N = 51), DM patients without acute myocardial infarction (AMI) (Group 2, N = 150), and DM patients with AMI (Group 3, N = 108). We assessed outcomes using stress perfusion cardiac magnetic resonance (CMR) imaging for coronary microvascular disease (CMD) (Outcome 1), retinography for retinal microvascular disease (RMD) (Outcome 2), both CMD and RMD (Outcome 3), and absence of microvascular disease (w/o MD) (outcome 4). We evaluated the classification performance of ceramides using receiver operating characteristic (ROC) analysis and multiple logistic regression. 11-ceramide panel previously identified by our research group as related to macrovascular disease were used. Results Average glycated hemoglobin (HbA1c) values were 5.1% in Group 1, 8.3% in Group 2, and 7.6% in Group 3. Within the cohort, CMD was present in 59.5% of patients, RMD in 25.8%, both CMD and RMD in 18.8%, and w/o MD in 38.5%. The AUC values for the reference ceramide ratios were as follows: CMD at 0.66 (p = 0.012), RMD at 0.61 (p = 0.248), CMD & RMD at 0.64 (p = 0.282), and w/o MD at 0.67 (p = 0.010). In contrast, the AUC values using 11-ceramide panel showed significant improvement in the outcomes prediction: CMD at 0.81 (p = 0.001), RMD at 0.73 (p = 0.010), CMD & RMD at 0.73 (p = 0.04), and w/o MD at 0.83 (p = 0.010). Abstract Background Several health institutions developed strategies to improve health content on Wikimedia platforms given their unparalleled reach. The objective of this study was to compare an online volunteer-based Wikimedia outreach campaign with university course Wikipedia assignments (both focused on improving hearing health content in Wikimedia’s public digital knowledge archives), in terms of the reach of the contributions and the extent of the participants’ input. A secondary objective was to examine the feasibility and the implementation of the different strategies. Methods The research team partnered for the (1) coordination of improvements in hearing and healthcare content through educational programs using Wikimedia platforms, (2) participation in the global campaign Wiki4WorldHearingDay2023 and (3) evaluation of the proposed strategies. Metrics used in the comparison of the two strategies included the number of articles edited, number of views of the edited articles (as reach) and the extent of edits, captured as the number of words. The feasibility evaluation included assessing recruitment success and the implementation of the proposed plan among faculty, students from various university programs, and volunteers representing different constituencies. Results The effort increased the availability of quality plain language information on hearing conditions and hearing care. Both strategies demonstrated to be feasible by their success in recruiting participants who contributed to the effort and by measurable outputs as edits. The contribution of content to Wikimedia platforms as part of education activities provided a more robust result. Wiki4WorldHearingDay2023 145 participants (78 from educational programs) contributed 167,000 words, 258 + references and 140 images to 322 Wikipedia articles (283 existing and 39 new ones), which were viewed 16.5 million times. Contributions occurred in six languages. Edits in Portuguese, mainly by those involved in educational programs, led the number of articles (226 or 70.2%) that were expanded or created during the 5-month tracking period. Conclusions The elements that contributed to the success of the studied strategies include an impact topic, coordination with educational programs, international multidisciplinary collaborations, the dissemination of the initiative in several platforms, connection with a robust local Wikimedia affiliate, and the use of a technical infrastructure that provides metrics and coordination mechanisms. The inventory of the taxa collected in 2021 during the annual field trip of the Working Group for Floristics, Systematics, and Evolution of the Italian Botanical Society is reported. The field trip was held in the Mount Pennino massif, between Umbria and Marche administrative regions (central Italy). The flora documented for the study area amounts to 413 specific and subspecific taxa (including one hybrid), belonging to 234 genera and 64 families. Thirty-seven taxa are endemic to Italy; 42 are included in the IUCN Red List of the Italian Flora. Only three alien taxa were found. Ten taxa and one hybrid are considered floristic novelties because either new or confirmed for the regional flora of Umbria and/or Marche. Particularly, 6 taxa are new and 4 are confirmed for Umbria; one taxon and one hybrid are new for Marche.

Within a practical course of cytotaxonomy organized in Pisa (Italy) on February 2024 by the Group for Floristics, Systematics and Evolution of the Italian Botanical Society, we tested whether using image analysis softwares possible biases are still introduced by different observers. We conclude that observer bias selectively applies in possibly overestimating the length of short arms in a karyotype. As a consequence, the parameters most sensitive to these possible errors are CVCI and CVCL, and to a less degree MCA and THL. To achieve more stable results among observers, a still lacking standardized measurement protocol could be helpful.

Access to higher education depends on the interaction between social origins and academic performance: background resources boost academic skills; but even when controlling for performance, privileged students are more likely to make ambitious choices and further transitions. Recent literature has shown that inequality in educational choices is heterogeneous across countries. However, it is still not well understood how different institutional designs within countries may affect the workings of those effects and how they can strengthen or weaken the inequality of educational opportunities. Using high-quality register data from the Brazilian higher education system, our work contributes to this understanding by investigating how SES and performance interact and drive students' choice between three different tracks: not entering higher education, entering the private system, or entering the public system. We developed a strategy to encompass multinomial choices and decompose the inequalities into primary and secondary effects. Using the Shapley Value decomposition strategy, we correct an intrinsic asymmetry that biased previous results. Our findings suggest affluent students enjoy dual advantages: high exam performance amplifies access to public universities (indirect effect) and family resources offset subpar performance, ensuring private university access (direct effect). We found no signs of multiplicative advantages.

This study aims to register part of the collection of body culture in the city of Tocantinópolis, Tocantins, located in the North of Brazil, based on the play reported by the older adult participants in the Health and Cognition Project for Seniors at the FUNT (Federal University of Northeast of Tocantins). Thus, we carried out exploratory fieldwork and qualitative analysis. The field information was gathered through semi-structured interviews with 42 (forty-two) participants. The script of the interviews encompassed their favourite places to play, partnerships, toys, and general characteristics. The analysis of the interviews indicated 64 (sixty-four) manifestations, divided into 8 (eight) categories, according to their particularities. The reports also revealed some portraits of a childhood suffering from poverty and early employment. Furthermore, the rural environment was the setting of some records, using toys manufactured with natural resources.

Abstract In this study, lactobionic acid (LBA) was incorporated into poly(vinyl alcohol) (PVA) and poly(ε-caprolactone) (PCL) by electrospinning. The antimicrobial effects of the nanofibers were tested using the agar diffusion method. Only the PVA formulations showed antimicrobial activity against Staphylococcus aureus. The PVA and PCL nanofibers containing LBA showed antioxidant activity ranging from 690.33 to 798.67 µM TEAC when tested by the ABTS method. The characterization of nanofibers was performed by scanning electron microscopy, Fourier transform infrared spectroscopy, thermogravimetric analysis, differential scanning calorimetry, and mechanical analysis. The nanofibers showed a uniform morphology and their average diameters ranged from 295.5 to 2778.2 nm. The LBA addition induced a decrease in the enthalpy of fusion (ΔHm) of PVA and PCL nanofibers, while the Young’s modulus was reduced from 20 to 10 MPa in PCL and PCL-LBA nanofibers, respectively. No relevant differences were observed between the FTIR spectra of the control nanofibers and the nanofibers containing LBA. All nanofibers presented hemolysis rate below 2%, thus can be considered as non-hemolytic materials. Further toxicological assessment was performed with the selected formulation PVA10 + LBA. The evaluations by mutagenicity assay, cell survival measurement, cell viability analysis and agar diffusion cytotoxicity test indicated that there are no significant toxic effects. Electrospun nanofibers PVA-LBA and PCL-LBA were successfully produced, showing good thermal and mechanical properties and non-toxic effects. Furthermore, the nanofibers showed antimicrobial activity and antioxidant activity. The findings of this study indicate that PVA and PCL electrospun nanofibers incorporating LBA are promising for use in packaging applications.

Abstract Automatic disease progression prediction models require large amounts of training data, which are seldom available, especially when it comes to rare diseases. A possible solution is to integrate data from different medical centres. Nevertheless, various centres often follow diverse data collection procedures and assign different semantics to collected data. Ontologies, used as schemas for interoperable knowledge bases, represent a state-of-the-art solution to homologate the semantics and foster data integration from various sources. This work presents the BrainTeaser Ontology (BTO), an ontology that models the clinical data associated with two brain-related rare diseases (ALS and MS) in a comprehensive and modular manner. BTO assists in organizing and standardizing the data collected during patient follow-up. It was created by harmonizing schemas currently used by multiple medical centers into a common ontology, following a bottom-up approach. As a result, BTO effectively addresses the practical data collection needs of various real-world situations and promotes data portability and interoperability. BTO captures various clinical occurrences, such as disease onset, symptoms, diagnostic and therapeutic procedures, and relapses, using an event-based approach. Developed in collaboration with medical partners and domain experts, BTO offers a holistic view of ALS and MS for supporting the representation of retrospective and prospective data. Furthermore, BTO adheres to Open Science and FAIR (Findable, Accessible, Interoperable, and Reusable) principles, making it a reliable framework for developing predictive tools to aid in medical decision-making and patient care. Although BTO is designed for ALS and MS, its modular structure makes it easily extendable to other brain-related diseases, showcasing its potential for broader applicability. After tooth loss, particularly in the posterior maxilla, the alveolar ridges undergo bone resorption. Therefore, ensuring the appropriate quantity and quality of alveolar bone is crucial for accurate implant positioning and achieving optimal esthetic and functional results. This study aimed to evaluate biomechanical parameters (insertion torque: IT, removal torque: RT, and implant stability quotient: ISQ) of distinct truncated cone implant designs (Sinus-plant and SLC) on polyurethane blocks simulating type D3 and D4 bone. SLC implants exhibited significantly higher IT, RT, and ISQ values compared to Sinus-plant implants, except in the 10 pounds per cubic foot (PCF) density block with a cortical layer for the IT (24.01 ± 0.91 vs. 23.89 ± 1.66 Ncm). The IT values for SLC implants ranged from 13.95 ± 0.19 Ncm in the lowest density block to 37.94 ± 0.45 Ncm in the highest density block, consistently providing significantly higher primary stability with an ISQ of approximately 70 in the highest density block. Despite lower ISQ in the lowest density block (48.60 ± 0.52 and 48.80 ± 0.42 in buccolingual and mesiodistal directions), it was deemed acceptable considering the inadequate bone densities in the maxillary region. These findings on SLC suggest potential clinical advantages, including reduced procedure duration and costs, improved stability, and the possibility of immediate implant placement following sinus augmentation, thereby streamlining the rehabilitation process.
