The adventitious bud development was induced in epicotyl segments of Valencia sweet orange (Citrus sinensis L. Osbeck). Seeds were cultured in vitro for three weeks in the dark, followed by one week at a 16-h photoperiod. Epicotyl segments were cultured horizontally for the induction of organogenesis in Murashige and Tucker (1969, MT) culture medium supplemented with 1.0 mg dm−3 benzylaminopurine. Samples were observed by light and scanning electron microscopy from day zero to day 25, when buds were well grown. It was shown that the adventitious buds originated directly from the cambial region on the cut ends of the explants.

A homolog of FLORICAULA/LEAFY, CfLFY (for Cedrela fissilis LFY), was isolated from tropical cedar. The main stages of the reproductive development in C. fissilis were documented by scanning electron microscopy and the expression patterns of CfLFY were studied during the differentiation of the floral meristems. Furthermore, the biological role of the CfLFY gene was assessed using transgenic Arabidopsis plants. CfLFY showed a high degree of similarity to other plant homologs of FLO/LFY. Southern analysis showed that CfLFY is a single-copy gene in the tropical cedar genome. Northern blot analysis and in situ hybridization results showed that CfLFY was expressed in the reproductive buds during the transition from vegetative to reproductive growth, as well as in floral meristems and floral organs but was excluded from the vegetative apex and leaves. Transgenic Arabidopsis lfy26 mutant lines expressing the CfLFY coding region, under the control of the LFY promoter, showed restored wild-type phenotype. Taken together, our results suggest that CfLFY is a FLO/LFY homolog probably involved in the control of tropical cedar reproductive development.

Flowering initiation depends on the balanced expression of a complex network of genes that is regulated by both endogenous and environmental factors. The timing of the initiation of flowering is crucial for the reproductive success of plants; therefore, they have developed conserved molecular mechanisms to integrate both environmental and endogenous cues to regulate flowering time precisely. Extensive advances in plant biology are possible now that the complete genome sequences of flowering plants is available and plant genomes can be comprehensively compared. Thus, association studies are emerging as powerful tools for the functional identification of genes involved on the regulation of flowering pathways. In this paper we report the results of our search in the Eucalyptus Genome Sequencing Project Consortium (FORESTS) database for expressed sequence tags (ESTs) showing sequence homology with known elements of flowering-time pathways. We have searched the 33,080 sequence clusters in the FORESTS database and identified Eucalyptus sequences that codify putative conserved elements of the autonomous, vernalization-, photoperiod response- and gibberellic acid-controlled flowering-time pathways. Additionally, we have characterized in silico ten putative members of the Eucalyptus homologs to the Arabidopsis CONSTANS family of transcription factors.

The rubber tree (Hevea brasiliensis Muell. Arg.) is an important source of natural rubber in tropical regions and, as with many woody species, shows a long juvenile phase. To understand the genetic and molecular mechanisms underlying the reproductive process in rubber trees, H. brasiliensis RRIM600 flower and inflorescence development have been characterized, the rubber tree FLORICAULA/LEAFY (FLO/LFY ) orthologue, HbLFY, cloned, and its expression patterns were analysed during vegetative and reproductive development. The rubber tree, similar to other Euphorbiaceae species, produces lateral inflorescences containing male, female, and bisexual flowers. HbLFY is expressed in lateral meristems that give rise to inflorescences and in all flower meristems, consistent with a role in reproductive development. Complementation studies using Arabidopsis lfy mutants indicated that the biological function of LFY might be conserved among Brassicaceae and Euphorbiaceae species.

The objective of this work was to evaluate leaf epidermis morphological characteristics of three citrus somatic hybrids, compared to their parents. Parental and somatic hybrid young leaves were collected and processed for scanning electron microscope observations. Citrus polyploid hybrids have fewer stomata per area and these are larger compared to their diploid parental parents. No differences in internal arrangement of the stomatal cells were detected between parental plants and somatic hybrids. Additional studies may determine if these differences will influence physiological behavior of the plants in the field. O objetivo deste trabalho foi avaliar as características morfológicas de três híbridos somáticos de citros, comparando-as com as de seus respectivos parentais. Folhas jovens dos híbridos somáticos e seus respectivos parentais foram coletadas e preparadas para observações em microscópio eletrônico de varredura. Híbridos poliplóides de citros apresentam menor número de estômatos por área, com maior tamanho individual quando comparados com aqueles das plantas diplóides parentais. Não foram observadas diferenças no arranjo interno das células estomáticas entre as plantas parentais e os híbridos somáticos. Investigações adicionais poderão determinar se essas diferenças poderão influenciar o comportamento fisiológico dessas plantas no campo.

An ultrasound extraction procedure for Ca, K and Mg from in vitro plant cultures is proposed, comparing cultures of different embryogenic levels of Citrus sinensis and Citrus limonia, employing ultrasound energy. Parameters related to metals extraction, such as plant material sampling, acid concentration and sonication time were investigated. For accuracy check, the proposed ultrasound extraction procedure was compared with a microwave-assisted digestion procedure and no differences in the results were verified at 95% of the confidence level. With this simple and accurate extraction procedure, it was possible to determine differences in Ca, K and Mg concentrations during Citrus embryo formation/development and between cultures (embryogenic and non-embryogenic). Finally, the ultrasound extraction method demonstrated to be an excellent alternative for handless sampling and operational costs.

Genetic transformation using mature tissue as explant is important for the precocious evaluation of the genetically modified characteristic. This research presents a protocol for adventitious organogenesis and genetic transformation of sweet orange varieties ( Citrus sinensis L. Osbeck) using as explants internodal segments from adult plants maintained in the greenhouse. Organogenesis was induced in DBA3 medium supplemented with benzilaminopurine (1.0 mg l −1 )+naphthaleneacetic acid (0.5 mg l −1 ), in the dark at 27±2 °C until adventitious buds had formed and then transferred to a 16-h photoperiod. Histological analysis showed an indirect formation of the adventitious buds from callus that had proliferated at the cut ends of the explants. Transformation was done using the Agrobacterium tumefaciens strain EHA-105 containing the plasmid p35SGUSINT as a vector. The explants were co-cultivated with Agrobacterium for 24 h, at 24 °C. Adventitious shoots were tested histochemically for GUS expression and micrografted into plantlets of the rootstock Carrizo citrange ( C. sinensis L. Osbeck× Poncirus trifoliata L. Raf.). Genetic transformation was confirmed by PCR and Southern blot analysis of the recovered plants.

Genetic transformation allows the release of improved cultivars with desirable characteristics in a shorter period of time and therefore may be useful in citrus breeding programs. The objective of this research was to establish a protocol for genetic transformation of Valencia and Natal sweet oranges (Citrus sinensis L. Osbeck) and Rangpur lime (Citrus limonia L. Osbeck). Epicotyl segments of germinated in vitro plantlets (three weeks in darkness and two weeks in a 16-h photoperiod) were used as explants. These were co-cultivated with Agrobacterium tumefaciens strain EHA-105 and different experiments were done to evaluate the transformation efficiency: explants were co-cultivated with Agrobacterium for one, three or five days; explants were incubated with Agrobacterium suspension for 5, 10, 20 or 40 minutes; co-cultivation medium was supplemented with acetosyringone at 0, 100 or 200 µmol L-1; Explants ends had a longitudinal terminal incision (2-3 mm); co-cultivation temperatures of 19, 23 or 27°C were imposed. The experimental design was completely randomized in all experiments with five replications, each consisted of a Petri dish (100 x 15 mm) with 30 explants and resulted in a total of 150 explants per treatment. Longitudinal terminal incision in the explant ends did not improve shoot regeneration. However, transgenic plants of all three cultivars were confirmed from explants that had been subjected to inoculation time of 20 minutes, co-culture of three days at 23-27°C, in the absence of acetosyringone. A transformação genética permite produzir cultivares com características específicas e pode, dessa forma ser associada a programas de melhoramento de citros. O objetivo deste trabalho foi estabelecer protocolos de transformação genética para as laranjas doce 'Valência' e 'Natal' (Citrus sinensis L. Osbeck), bem como para o limão 'Cravo'(Citrus limonia L. Osbeck). Segmentos de epicótilo de plântulas germinadas in vitro (três semanas no escuro e duas semanas sob fotoperíodo de 16h) foram utilizados como explantes. Estes foram co-cultivados com Agrobacterium tumefaciens (EHA-105), realizando-se vários experimentos para avaliar a eficiência do processo de transformação genética: explantes co-cultivados por um, três e cinco dias; tempo de inoculação com a bactéria de 5, 10, 20 e 40 minutos; co-cultivo em meio de cultura contendo 0, 100 e 200 mimol L-1 de acetoseringona; Incisão longitudinal (2-3 mm) nas extremidades do explante; temperatura de co-cultivo 19, 23 e 27°C. Todos os experimentos consistiram de cinco repetições por tratamento, sendo cada repetição representada por uma placa de Petri contendo 30 explantes, perfazendo um total de 150 explantes por tratamento. Plântulas transgênicas dos três cultivares foram obtidas utilizando-se tempo de inoculação de 20 minutos, co-cultivo com Agrobacterium tumefaciens (EHA-105) por três dias, na ausência de acetoseringona no meio de cultura de co-cultivo e temperatura de co-cultivo de 23-27°C. A incisão longitudinal na extremidade do explante favoreceu à organogênese in vitro, mas quando co-cultivado com Agrobacterium não houve regeneração de brotações.

Histological changes in banana explants, cv. Nanicao (Musa spp. Group AAA), submitted to different auxins for induction of somatic embryogenesis). The present work analyzes the behavior of banana explants, cv. Nanicao (Musa spp. Group AAA) regarding somatic embryogenesis induction treatments with several auxins. Longitudinal segments of shoot meristematic apices of micropropagated banana plantlets cultivated and rooted in vitro were introduced in culture medium containing dicamba, picloram, 2,4-D or NAA in different concentrations. Explant samples were collected at 0, 7 and 10 days and prepared for light microscopy. Histological sections were used for comparison of the histological changes occurring after induction treatment with different auxins. Embryogenic response was observed only in treatments with picloram or dicamba, with distinct embryogenic regions observed at 14 and 21 days in culture, respectively. Histological sections of embryogenic regions of the explant at 26 days in culture revealed the formation of meristematic regions, structures with multiple root meristems, and somatic embryos at early globular stages. Embryo-like structures morphologically similar to Musa balbisiana zygotic embryos were sectioned and showed a lack of apical meristems and absence of procambial differentiation. These results indicate the induction of non-functional somatic embryos and the need for more studies on developmental aspects and maturation treatments for optimization of the process. RESUMO - (Alteracoes histologicas em explantes de bananeira, cv. Nanicao (Musa spp. Grupo AAA) submetidos a diferentes auxinas para inducao de embriogenese somatica). O presente trabalho analisa o comportamento de explantes de bananeira 'Nanicao' (Musa spp. Grupo AAA) a tratamentos de inducao com diferentes auxinas, visando a obtencao de embriogenese somatica. Segmentos longitudinais de apices meristematicos caulinares de plântulas de bananeira micropropagadas e enraizadas in vitro foram introduzidos em meio de cultura contendo dicamba, picloram, 2,4-D ou NAA. Amostras dos explantes aos 0, 7 e 10 dias foram coletadas e preparadas para microscopia optica. Cortes histologicos foram realizados nesses explantes, comparando-se as alteracoes observadas apos os tratamentos com diferentes auxinas. Respostas embriogenicas foram observadas somente nos tratamentos com picloram e dicamba, sendo possivel a observacao visual de regioes embriogenicas aos 14 e 21 dias de cultivo, respectivamente. Cortes histologicos de regioes embriogenicas dos explantes, aos 26 dias, revelaram a formacao de regioes meristematicas compostas aparentemente por multiplos meristemas radiculares e embrioes nas fases iniciais do estagio globular. Cortes histologicos de estruturas morfologicamente semelhantes a embrioes zigoticos de Musa balbisiana revelaram ausencia dos meristemas apicais e nao diferenciacao procambial. Os resultados indicam a inducao de embrioes somaticos nao-funcionais e, consequentemente, a necessidade de maiores estudos visando otimizar o processo de desenvolvimento e maturacao dos embrioes somaticos.

Somatic embryogenesis from nucellus-derived callus cultures of five cultivars, including three (Caipira, Seleta Vermelha, and Valencia) of sweet oranges (C. sinesis L. Osbeck), Rangpur lime (C. limonia L. Osbeck), and Cleopatra mandarin (C. reticulata Blanco) (lines I and II), were studied. Callus lines maintained on MT medium supplemented with 50 g l−1 sucrose were transferred to MT medium supplemented with different carbohydrate sources: galactose, glucose, lactose, maltose, or sucrose at 18, 37, 75, 110, or 150 mM, or glycerol at 6, 12, 24, 36, or 50 mM. Globular embryos were observed after approximately 4 wk, in several treatments. Cultures of Valencia and Caipira sweet oranges and Cleopatra mandarin (line I) showed high numbers of embryos on medium containing galactose, lactose, and maltose. Histological studies showed somatic embryos in all developmental stages with a normal histodiffeentiation pattern. The other two cultivars (Rangpur lime and Cleopatra mandarin, line II) formed very few embryos, which did not develop further following the globular stage. Some of the abnormalities observed were lack or dedifferentiation of protoderm and absence of apical meristems and procambial strands. Embryos that followed the normal sequence of development were easily converted into plants. Non-embryogenic cultures continued as proliferating callus cultures, eventually forming a few embryos which did not convert into plants. Statistical analyses of the callus response to carbohydrate treatments was done using an overdispersion Poisson model.

A embriogênese somática pode ser uma alternativa para o melhoramento de espécies vegetais, quando associada à transformação genética. A regeneração de plântulas a partir de embriões somáticos é dependente de vários fatores, entre eles, a qualidade do embrião somático. Neste trabalho embriões somáticos com diferentes características morfológicas são descritos. Embriogênese somática de bananeira, cv. Grand Nain e Nanicão Jangada foi induzida a partir de inflorescências funcionalmente masculinas, em meio de indução (MI) seguido de subcultivo em meio de germinação (MG), em ausência de luz. A presença de luz na indução, não favoreceu a resposta embriogênica. Em ausência de luz, os explantes da cv. Nanicão Jangada, durante os seis meses de cultivo, sofreram diversas alterações, como a formação de calos embriogênicos e não embriogênicos, formação de embriões somáticos, continuidade no desenvolvimento do botão floral ou oxidação do explante. A freqüência de resposta embriogênica foi baixa, com apenas 6% dos explantes formando embriões somáticos e 23% com formação de calos embriogênicos. O acompanhamento histológico e morfológico das alterações ocorridas nos explantes permitiu a identificação de embriões somáticos com características morfológicas distintas: com maior freqüência observou-se um tipo de embrião com morfologia semelhante ao embrião zigótico de Musa acuminata; em menor freqüência desenvolveram-se embriões somáticos semelhantes a embriões zigóticos de outras monocotiledôneas, indicando que em um único protocolo de embriogênese somática, é possível ocorrer a formação de diferentes padrões morfológicos de embriões somáticos.

In vitro morphogenesis of C. melo L. var. inodorus was studied by the induction of adventitious buds and somatic embryos. Organogenesis was obtained from cotyledon segments and leaf discs in culture medium supplemented with benzylaminopurine (1 mg l−1) and somatic embryogenesis was induced in medium containing 2,4-dichlorophenoxyacetic acid (5 mg l−1) + thidiazuron (1 mg l−1). Through histological analysis it was possible to verify that in cotyledonary explants, protuberances that do not develop into well-formed shoot buds and leaf primordia are more frequently formed than complete shoot buds, resulting in a low frequency of plant recovery in the organogenic process. A high percentage of explants responded with the formation of somatic embryos; the microscopical analysis showed that the somatic embryos lacking well developed apical meristems had a low conversion rate into plants. Plant recovery was not obtained from leaf-disc explants, with high rates of contamination and formation of protuberances which did not develop into shoot buds. Histological sections showed the development of epidermis and leaf hairs, indicating those structures could be leaf primordia; however, these were not associated with a shoot apical meristem.

Organogenesis induction from internodal segments of yellow passionfruit (Passiflora edulis f. flavicarpa) was obtained in a medium supplemented with 1.0-4.0 mg L-1 benzylaminopurine (BAP). Callus proliferation and adventitious shoot development were analyzed by light and scanning electron microscopy. Callus proliferation and adventitious shoots were observed simultaneously and continuously, with asynchronous development of shoots in the explants. Proliferation of callus was observed in the explant cut ends in all treatments, including the control. The results show that concentrations of 1.0 or 2.0 mg L-1 BAP gave better rate of organogenesis. Higher concentrations were efficient for the induction of adventitious buds and, were, however, detrimental for subsequent shoot development. Growth and rooting of shoots were obtained in half strength medium without growth regulators.

Mineral deficiency symptoms were observed in leaves of yellow passionfruit plantlets grown in MS medium (Murashige and Skoog, 1962) with 1.0 mg l−1 (3.0 μM) gibberellic acid. Initially, leaves showed interveinal chlorosis, followed by bleaching of the leaves and retarded growth. Leaf mineral analysis was done and compared to mineral requirements suggested for passionfruit in the literature. Several modifications were made to the inorganic composition of MS medium, according to mineral deficiencies, mainly of Fe and Ca, and possible toxicity of Cl. The concentration of the elements in the new medium (MSM) was based on the mineral composition of leaves of healthy plants. The chemical equilibrium was checked using the software Geochem (Sposito and Mattigod, 1980) and final adjustments were made to ensure good availability of nutrients. To test the efficiency of the modified medium nodal segments were cultured in both MS and MSM supplemented with 3.0 mg l−1 (13.3 μM) 6-benzyladenine. After three subcultures mineral analysis of the leaves was done. Severe mineral deficiency was observed on the leaves of plantlets cultured in MS, while plantlets cultivated in MSM had green leaves. A comparison of the mineral analysis of plantlets in both media showed a fairly large increase in Ca, Cu, Fe, Mg and S and decrease in levels of B and Cl in plantlets cultivated in MSM. A slight increase or decrease in other elements was also observed. Subculture of the chlorotic plantlets into MSM showed that the visual symptoms of mineral deficiency disappeared in 2–4 wk.

The use of micropropagation to obtain large numbers of high-quality planting material has increased in recent years. Behavior in culture, mainly in terms of multiplication rate, varies among genotypes, directly affecting plant production planning. To study multiplication rates over time, suckers of banana, Musa spp., cv. Maca, were collected in the field and the shoot apex introduced in vitro for micropropagation. The number of new shoots produced in each of the six multiplication cycles was recorded and the data analyzed statistically. Variability in total shoot production and differences in multiplication rates was considerable among families, which consisted of the initial explant and its progeny. Moreover, the adjusted Poisson regression models for the number of shoots showed that the multiplication rate in this cultivar tends to decrease with time: after the seventh subculture, new shoots may form at a very low rate. Interpretation of the first and second derivatives of the regression model allowed determination of the maximum speed of multiplication and the time at which the multiplication rate begins to decline.

A regeneração in vitro de plantas de Passiflora suberosa foi obtida a partir de discos foliares. Folhas foram retiradas de plantas germinadas em casa de vegetação, imersas em solução comercial de hipoclorito de sódio (3:1), durante 20 minutos. Discos foliares de 0,5 cm de diâmetro, com a face adaxial em contato com o meio de cultura, foram introduzidos em placa de petri. Organogênese foi obtida utilizando-se meio MS acrescido de 0,5 ou 1,0 mg L-1 de BAP (6-benzilaminopurina). Após 4 a 8 semanas, observou-se formação de calos nas bordas dos discos foliares. Os calos formados foram transferidos para meio MSM, acrescido de 1,0 mg L-1 de GA3 (ácido giberélico), sob fotoperíodo de 16 horas. Desenvolvimento de gemas adventícias foi obtido a partir dos calos, sendo estas alongadas e enraizadas no mesmo meio de cultura, após periódicas repicagens, e aclimatadas em casa de vegetação.

† Background and Aims Floral development of Cedrela and Toona, the genera comprising the basal tribe Cedreleae of the sub-family Swietenioideae of Meliaceae, is described. The focus was on three endangered, ecologically and economically important species: Cedrela fissilis, Cedrela odorata and Toona ciliata. The aims of the study were to characterize the patterns of floral development in the tribe and to establish apomorphic and plesiomorphic floral characters in relation to other taxa within the family based on the current molecular phylogeny of Meliaceae. † Methods A detailed floral structural and developmental study was completed using both scanning electron microscopy and visualization of microtome sections with a light microscope. † Key Results Twelve floral developmental stages were identified. The initial development of the pentamerous flowers of both Toona and Cedrela is strikingly similar. The morphological differences observed between them are due to differential patterns of organ elongation and adnation/connation occurring late in development. Additionally, the formation of functionally male and female flowers was found to occur at specific positions within the inflorescence. † Conclusions Due to the basal position of the tribe Cedreleae in the phylogeny of Meliaceae, functionally either male or female pentamerous flowers and the presence of (at least partially) free stamens may be considered plesiomorphic traits within the family. In contrast, sympetaly and the absence of nectaries in Cedrela species are synapomorphies.

The objective of this work was to study the in vitro organogenesis of Citrullus lanatus, by the induction of adventitious buds in cotyledon segments cultured in medium supplemented with cytokinin. Explants were collected from one, three and five-day-old in vitro germinated seedlings, considering the distal and proximal cotyledon regions. The data obtained showed that in vitro organogenesis of watermelon occurred with higher efficiency, when cotyledon segments from the proximal region collected from three-day-old seedlings were cultivated in medium MS, supplemented with BAP (1 mg L-1) and coconut water (10%). The histological study showed that the organogenesis occurs directly, without callus formation, on epidermal and subepidermal layers of the explants. Adventitious shoots were characterized by the development of shoot apical meristem and leaf primordia. The formation of protuberances, that do not develop into adventitious buds, was also observed. O objetivo do trabalho foi estudar a organogênese in vitro de C. lanatus, pela indução de gemas adventícias, em segmentos de cotilédones, cultivados em meio de cultura suplementado com citocinina. Os explantes consistiram de segmentos das regiões distal e proximal de cotilédones, coletados de plantas germinadas in vitro com um, três e cinco dias de idade. Os dados obtidos mostram que a organogênese de melancia, in vitro, ocorre com maior eficiência em segmentos da região proximal dos cotilédones, coletados de plântulas com três dias de idade e cultivados em meio de cultura MS, suplementado com a combinação BAP (1 mg L-1) e água de coco (10%). Pelo estudo histológico, verificou-se que a organogênese ocorre diretamente, sem a formação de calo, na epiderme e subepiderme do explante. As gemas adventícias foram caracterizadas pela presença de meristema apical e primórdios foliares. Observou-se, também, o desenvolvimento de protuberâncias que não se desenvolvem em gemas adventícias.

During the last thirty years plant micropropagation has been one of the most significant contributions of biotechnology to the horticultural industry (Giles and Friesen, 1994). The possibility of mass propagation of elite, endangered, or difficult-to-propagate genotypes, in relatively small areas and time period, is the most important characteristic of micropropagation. In association with breeding programs micropropagation can have a significant impact not only in the multiplication of important genetic material, but also for thein vitromaintenance of germplasm.

Exogenous genes can be introduced in plants by genetic transformation techniques. However, an efficient tissue culture system with high rates of plant recovery is necessary for gene introduction. This work aimed to define organogenesis and plant regeneration protocols for sweet orange varieties Natal, Valencia and Hamlin (Citrus sinensis L. Osbeck) and Rangpur lime (Citrus limonia L. Osbeck) which can be used in plant transformation experiments. Seeds of which teguments were removed, were germinated in vitro and maintained in the dark for three weeks, followed by one week at 16-h photoperiod (40 µmol m-2 s-1) and 27 ± 2°C. Organogenesis induction was done by introducing epicotyl segments in MT medium with 25 g L-1 sucrose and different BAP concentrations. After adventitious bud growth, the shoots were transferred to MT medium with either NAA or IBA (1 mg L-1), or absence of auxin, for rooting. The best results were obtained with 1 mg L-1 BAP for bud induction and 1 mg L-1 IBA for rooting for all three sweet orange cultivars. The use of 0.5-2.5 mg L-1 BAP, followed by 1 mg L-1 IBA were the best growth regulator combinations for bud induction and rooting, respectively, for 'Rangpur' lime. The protocols presented in this work are suitable for associations with genetic transformation experiments for these cultivars. Com o desenvolvimento de novas técnicas biotecnológicas, é possível a introdução no genoma vegetal de genes de outras plantas ou outros organismos pela transformação genética. Entretanto, um dos principais requisitos para o sucesso desta técnica é a existência de uma metodologia eficiente de regeneração de plantas in vitro. O objetivo deste trabalho foi definir protocolos de regeneração de plantas, via organogênese, para laranjas 'Natal', 'Valência' e 'Hamlin' (Citrus sinensis L. Osbeck), e limão 'Cravo' (Citrus limonia L. Osbeck), para obtenção de plantas transgênicas nestas variedades. Sementes sem tegumentos foram germinadas in vitro durante três semanas no escuro e uma semana sob fotoperíodo de 16 h (40 µmol m-2 s-1), a 27 ± 2°C. Para indução das brotações, segmentos de epicótilo foram cultivados no meio de cultura MT (25 g L-1 de sacarose) suplementado com diferentes concentrações de benzilaminopurina (BAP). Para enraizamento, utilizou-se meio MT, com ou sem a adição de NAA ou IBA (1,0 mg L-1). A combinação 1,0 mg L-1 BAP, utilizada na fase de indução de brotações e 1,0 mg L-1 IBA, utilizada na fase de enraizamento, permitiu regeneração de plantas com bom índice de enraizamento nas três variedades de laranjas. A utilização de 0,5-2,5 mg L-1 BAP na fase de indução de brotações, com 1,0 mg L-1 IBA na fase de enraizamento, assegurou regeneração de plantas com alto índice de enraizamento para o limão 'Cravo'. Os protocolos de organogênese desenvolvidos neste trabalho podem ser utilizados em experimentos de transformação genética envolvendo essas variedades.

The EgLFY gene cloned from Eucalyptus grandis has sequence homology to the floral meristem identity gene LEAFY (LFY) from Arabidopsis and FLORICAULA (FLO) from Antirrhinum. EgLFY is preferentially expressed in the developing eucalypt floral organs in a pattern similar to that described previously for the Arabidopsis LFY. In situ hybridization experiments have shown that EgLFY is strongly expressed in the early floral meristem and then successively in the primordia of sepals, petals, stamens and carpels. It is also expressed in the leaf primordia of adult trees. The expression of the EgLFY coding region under control of the Arabidopsis LFY promoter could complement strong lfy mutations in transgenic Arabidopsis plants. These data suggest that EgLFY plays a similar role to LFY in flower development and that the basic mechanisms involved in flower initiation and development in Eucalyptus may be similar to those occurring in Arabidopsis. O gene EgLFY, clonado de Eucalyptus grandis, possui homologia de seqüência com o gene de identidade meristemática LEAFY (LFY) de Arabidopsis e FLORICAULA (FLO) de Antirrhinum. EgLFY é, preferencialmente, expresso nos órgãos florais em desenvolvimento de eucalipto, obedecendo a um padrão similar ao descrito para o gene LFY de Arabidopsis. Experimentos de hibridização in situ mostraram que o gene EgLFY é fortemente expresso em meristemas florais no início de seu desenvolvimento e, então, sucessivamente, durante a formação dos primórdios de sépalas, pétalas, estames e carpelos. Há expressão também nos primórdios foliares de árvores adultas. A expressão da região codificadora de EgLFY sob o controle do promotor do LFY de Arabidopsis pôde complementar mutantes lfy nulos em plantas de Arabidopsis transgênicas. Essas observações sugerem que o gene EgLFY possui um papel similar ao de LFY no desenvolvimento floral e que os mecanismos básicos envolvidos na iniciação e no desenvolvimento floral em Eucalyptus podem ser semelhantes aos de Arabidopsis.

A família Meliaceae compreende cerca de 51 gêneros e 550 espécies distribuídas principalmente na região Neotropical. Incluídas nesta família, estão espécies de elevado interesse comercial para a produção de madeiras nobres. Há carência de informações quanto à biologia floral dessas espécies, que aliada a problemas silviculturas dificulta a elaboração de programas efetivos de melhoramento genético das espécies de interesse econômico da família. O presente trabalho visa contribuir para o entendimento do desenvolvimento floral na família Meliaceae, com destaque para sete espécies de interesse econômico e/ou ecológico: Cedrela fissilis L., Cedrela odorata L., Swietenia macrophylla R. A. King, Trichilia claussenii C. DC., Guarea guidonea (L.) Sleumer, Toona ciliata M. J. Roem e Melia azedarach L. Para os estudos morfo-anatômicos utilizaram-se técnicas de microscopia óptica e microscopia eletrônica de varredura. Analisou-se igualmente o padrão de expressão de genes do modelo ABC, relacionados ao desenvolvimento floral, via hibridização in situ. Os estudos morfo-anatômicos permitiram caracterizar o desenvolvimento dos primórdios dos órgãos florais e o estabelecimento de estágios arbitrários de desenvolvimento, auxiliando na caracterização da expressão gênica. Estes resultados permitiram ainda a identificação de flores funcionalmente femininas ou masculinas em S. macrophylla C. fissilis, C. odorata e T. ciliata, as quais apresentam dimorfismo sexual. O padrão de expressão dos homólogos dos genes: APETALA1 (AP1), APETALA3 (AP3) e AGAMOUS (AG) foram diferentes daqueles previstos pelo modelo ABC. A expressão dos homólogos de AP1 foi verificada em todo o meristema floral nos estágios iniciais de desenvolvimento, em todas as espécies estudadas. O sinal de expressão concentrou-se nos primórdios das sépalas e pétalas em estágios mais avançados do desenvolvimento, porém um fraco sinal de hibridização de AP1 foi verificado em todos os verticilos. A expressão dos homólogos de AP3 foi verificada nas regiões previstas pelo modelo ABC, correspondentes ao segundo e terceiro verticilos. Porém um fraco sinal de AP3 foi também observado no quarto verticilo, o que não é previsto pelo modelo teórico. A expressão dos homólogos de AG foi restrita à região central do meristema floral, correspondente ao terceiro e quarto verticilos, em todos os estágios do desenvolvimento floral de todas as espécies estudadas. Entretanto, sinais adicionais de hibridização dos homólogos de AG foram visualizados na região abaxial dos primórdios de sépalas e pétalas em C. fissilis, C. odorata e T. ciliata. Os estudos da expressão dos genes do modelo ABC em Meliaceae revelaram padrões de expressão que não concordam com o modelo teórico vigente do controle molecular da determinação da identidade dos órgãos florais._________________________________________________________________________________________ ABSTRACT: The Meliaceae family comprises approximately 51 genus and 550 species mainly distributed in the Neotropical region. In this family there are many species of commercial interest for the production of noble wood. The lack of information on the floral biology associated with silvicultural problems limits the elaboration of effective breeding programs with species of economic interest. The present work aims to contribute with the understanding of floral development in Meliaceae, focusing in seven species of economical or ecological interest: Cedrela fissilis L., Cedrela odorata L., Swietenia macrophylla R. A. King, Trichilia claussenii C. DC., Guarea guidonea (L.) Sleumer, Toona ciliata M. J. Roem e Melia azedarach L. Morpho-anatomical analyses were done by light and scannning electron microscopy. The expression pattern of the ABC model genes, which are related to floral development, was analyzed by in situ hybridization. The characterization of the development of floral organ primordia and the establishment of arbitrary stages of floral development was done and was important for the characterization of gene expression. These results allowed for the identification of functionally female and male flowers in Swietenia macrophylla, Cedrela fissilis, Cedrela odorata and Toona ciliata, characterizing sexual dimorphism. The patterns of gene expression of the homologous of APETALA 1, APETALA 3 and AGAMOUS were different from those predicted by the ABC model. AP1 expression was observed in the entire floral meristem in the initial stages of development, in all seven species. The expression signal was more concentrated in sepal and petal primordia in further stages of development, although a weak hybridization signal of AP1 was verified in all four whorls. The expression of AP3 homologs was observed in the second and third whorls, as predicted by the ABC model. However, a weak signal of AP3 was also observed in the forth whorl, which was not predicted by the ABC model. AG homolog expression was restricted to the central region of the floral meristem, corresponding to the third and forth whorls, in all stages of development of all seven species. However, additional hybridization signal of AG homologs were seen in the abaxial region of sepal and petal primordia in Cedrela fissilis, Cedrela odorata and Toona ciliata. The analyses of the ABC model genes in Meliaceae revealed that the expression patterns do not agree with the theoretical currently accepted model that determines the molecular control of the identity of the floral organs.

As plantas apresentam diversas adaptações fisiológicas à baixa disponibilidade de fósforo (Pi) do solo. Este trabalho discute os custos fisiológicos e energéticos associados com essas estratégias, focado nas respostas da cana-de-açúcar (Saccharum spp.) à disponibilidade de Pi durante a simbiose com micorrizas arbusculares (Glomus clarum). Esses custos são importantes componentes para a adaptação a solos com baixo Pi, afetando a aquisição e conteúdo de fósforo; o crescimento e concentração de açúcares em tecidos vegetais. Plantas de cana-de-açúcar foram cultivadas em vasos com ou sem micorrizas (Glomus clarum), e sob a disponibilidade de baixo (20 mg kg-1) ou alto (202 mg kg-1) fósforo. Raízes e parte-aérea foram coletadas para as análises após 14, 30, 44 e 58 dias pós-inoculação (dpi) com Glomus clarum . A condição de BP causou a deficiência de Pi nas plantas, micorrízicas ou não. As plantas sob AP continham um teor foliar de Pi adequado, e partir dos 44 dpi acumularam pelo menos 6 vezes mais Pi parte-aérea, do que as cultivadas sob BP, efeito mais evidente nas micorrízicas. A eficiência de absorção, indicada pelo acúmulo de Pi na parte-aérea a uma dada biomassa da raiz, foi igual para todos os tratamentos, sugerindo que as eficiências radicular e micorrízica da absorção de Pi foram similares, independentemente da doses de Pi. A disponibilidade de fósforo não afetou a biomassa total das plantas, sendo as cultivadas sob BP mais eficientes na utilização deste nutriente. Por outro lado, as plantas micorrízicas suplementadas com BP apresentaram maior crescimento da raiz e redução na parte-aérea, resultando no aumento da proporção raiz:parte-aérea. Aos 58 dpi, a glicose, frutose e sacarose presente nas folhas de plantas micorrízicas foi 3,8, 2,3 e 2,4 vezes respectivamente mais concentrada do que nas não micorrízicas. Esses resultados sugerem que, nestas condições experimentais, o estabelecimento da simbiose não foi uma associação mutualística típica, afetando o perfil de crescimento e a alometria da cana cultivada com BP. As concentrações de fotoassimilados na folhas de planta micorrízicas indicam que houve aumentos nas taxas fotossintéticas, mas isso não resultou no maior crescimento do macrosimbionte. A tecnologia de amplificação quantitativa de transcritos reversos (RT-PCR) se tornou uma opção para a validação funcional de genes, com alta sensibilidade, acurada quantificação e eficácia. A quantificação relativa da expressão gênica é conseqüentemente fácil e determina a expressão de um gene em relação a outro expresso e relativamente constante. Neste trabalho foi analisada a variabilidade de expressão dos genes de cana-de-açúcar codificando a actina (Actina), gliceraldeido fosfato desidrogenase (GAPDH), tubulina (Tubulina), e ubiquitinas (UbiQ1 e UbiQ2) em diversos tecidos, e comparou-se a variabilidade obtida utilizando os programas Genorm e NormFinder. Em seguida, foram realizadas análises de expressão gênica utilizando o programa REST para a validação estatística da expressão de genes de cana-de-açúcar. O gene UbiQ1 foi mais estável nos tecidos ou órgãos testados: meristema, inflorescência, folha, colmo e raízes tratadas com alto e baixo fósforo. Tendo o gene UbiQ1 como referência, a expressão relativa dos genes transportadores de fosfato de alta afinidade de cana-de-açúcar PT7 e PT8 foi avaliada em amostras de raiz fertilizadas com alto ou baixo Pi, inoculadas ou não com fungo micorrízico e coletadas aos 58 dpi. Esses dois genes pertencem à família Pht1 de transportadores de fosfato e são similares aos ortólogos de arroz ORYsa;tPht1;7 e ORYsa;Pht1;8. Sob a deficiência de Pi, o transportador de fosfato PT7 foi induzido em raízes não colonizadas; já nas micorrízicas foi pouco expresso, sendo que altas taxas de colonização radicular suprimiram a expressão do PT7. O gene PT8 pouco variou sua expressão, sendo sutilmente mais expresso em plantas micorrízicas do que nas não micorrízicas sob o suprimento de BP. Estes resultados indicam que o PT7 é induzido em raízes sob estresse por fósforo e provavelmente associado à absorção radicular de Pi. Enquanto o gene PT8 possui uma modulação pouco variável provavelmente envolvido na manutenção do fluxo ou homeostase de Pi, possivelmente associado com a absorção radicular e micorrízica de fosfato. O PT7 e o PT8 foram expressos em tratamentos de médio/longo prazo, apresentando expressão ou indução em resposta a privação por Pi, o que é consistente com a função proposta de aquisição e mobilização de Pi para esta família de transportadores. This work discussed physiological and energetic costs associated with these strategies, focusing on sugarcane responses to Pi availability during the development of arbuscular mycorrhizal (AM) symbiosis. Such costs are important components of adaptation to low phosphorus soils affecting phosphorus acquisition and leve, growth and soluble sugars concentration in plant tissues. Sugarcane plants were grown in pots, with or without AM (Glomus clarum), and with low (20 mg kg-1) or high (200 mg kg-1) phosphorus supply. Roots and shoots were harvest for analysis after 14, 30, 44 and 58 days post-inoculation (dpi) with the fungus Glomus clarum,. The low Pi supply caused Pi deficiency in mycorrhizedl or non-mycorrhizedl plants. The efficiency of Pi absorption, indicated by shoot Pi accumulation in correlation to root biomass, suggested that root and mycorrhizal Pi absorption were similar, regardless of the Pi doses. Phosphorus availability did not affect the whole-plant biomass, and plants under low Pi supply, used more efficiently this nutrient. On the other hand, mycorrhized plants supplemented with low Pi presented the highest root growth and shoot reduction, resulting in high root:shoot ratio. At 58 dpi, glucose, fructose and sucrose concentrations in leaves of mycorrhized plants were 3.8, 2.3 and 2.4-fold higher respectively, than in non-mycorrhizedl plants. These results suggested that, under these experimental conditions, mycorrhizal symbiosis establishment was not a typical mutualistic association affecting sugarcane growth profile and allometry, when cultivated under low Pi. The photosynthate levels of leaves from mycorrhizd plants indicated an increase in photossynthetic rate but withut resulting in higher macrobiont growth. The quantitative amplification of reversed transcripts (RT-PCR) technology has become a method of choice for functional gene validation, with sensitiveness, accurate quantification and high-throughput. The relative quantification is easier determined by relative expression in comparison to a constitutively expressed refernce gene. Here, the expression variability of a sugarcane actin gene (Actin) glyceraldehydo phosphate dehydrogenase (GAPDH), tubulin (Tubulina), and ubiquitins (UbiQ1 and UbiQ2) from various tissues were analysed and compared based on the ranking list from the Genorm and NormFinder softwares. Expression analysis based on the REST software gave the proper statistic validation. UbiQ1 was the most stable gene among the candidate gen-references along the various tissues or organs tested: meristem, inflorescence, leaf, stem and roots treated with high and low phosphorus. Considering UbiQ1 as the reference gene, the relative expression of the sugarcane high-affinity phosphate transporters genes PT7 and PT8 were assessed from roots fertilized with low Pi or high Pi , inoculated or not with the mycorrhizal fungus, harvest 58 dpi. Both genes belong to the Pht1 Pi transporter and share similarity with the rice orthologs ORYsat;Pht1;7 and ORYsat;Pht1;8. Under Pi deficiency, the phosphate transporter PT7 was induced in non-colonized roots, but less expressed in mycorrhizedl ones, with high root colonization rate suppressing PT7 expression. PT8 showed low variability in expression, slightly more expressed in mycorrhized plants than in non-mycorhized plants under low Pi supply. These results indicated that PT7 was induced in Pi stressed roots, and possibly associated with the root Pi uptake, while PT8 had limited modulation in expression and probably involved on Pi fluxes or homeostasis, likely associated with both root and mycorrhizal phosphate uptake pathways. PT7 and PT8 were induced during medium/long-term treatments, showing induction or constant expression in both acquisition and mobilization of Pi in response to Pi deprivation, which is consistent with the proposed role of this tranporter family. O crescimento cada vez maior da industrialização e a existência de um mercado globalizado vêm exigindo dos produtores rurais à utilização de modernas tecnologias ligadas à produção animal. O baixo custo de produção e o aumento da qualidade do produto final gerado têm sido a grande meta dos pecuaristas e centros de pesquisa voltados à agropecuária. Devido à necessidade de aumento na produção, propriedades agropecuárias têm recorrido ao uso de todo e qualquer recurso disponível que possa promover melhorias na atividade produtiva.O uso indiscriminado dos antimicrobianos com a intenção de aumentar a produção animal pode resultar em concentrações residuais nos produtos como carne, ovos e leite acima das doses aceitáveis para o consumo humano. Inicialmente, os antibióticos eram usados como medida terapêutica, mas com o avanço do conhecimento e desenvolvimento de novos compostos, passaram a ser utilizados também como medida preventiva e como promotores do crescimento.O que se deve questionar não é a presença destes resíduos, mas sim o tipo e as concentrações dos mesmos. De fato, as modernas tecnologias analíticas têm permitido a detecção de partes por bilhão de substâncias químicas, componentes ou metabólitos de medicamentos de uso veterinário em alimentos e, nestas quantidades, dificilmente elas representariam perigo à saúde dos consumidores. Portanto, há necessidade de se estabelecer metodologias analíticas mais eficientes que possam auxiliar os órgãos de fiscalização da agricultura no controle do uso desses antimicrobianos na produção animal. Assim como auxiliar no controle de qualidade, principalmente em indústrias exportadoras que almejam expansão de mercado, onde essa prática é quase uma imposição no contexto do comércio internacional de produtos pecuários \"in natura\" e processados. Devido a essas necessidades, o objetivo do presente foi padronizar e desenvolver reagentes imunoenzimáticos para a pesquisa da ciprofloxacina em produtos de origem animal. Os resultados obtidos no desenvolvimento desses imunoreagentes foram positivos quando correlacionado (r2 = 0,9588) com o kit referência RIDASCREEN® Enro/Cipro da R-Biopharm. The low production cost and the increasing of obtained final product quality have been the great aim of cattle farmers and cattle farming research centers. Due to the necessity of increasing the production, rural properties have required the use of any available source able to promote better productive activities. The indiscriminated antibiotics use aiming to increase animal production may result in residual concentrations found in products such as meat, eggs and milk over acceptable doses to the human consumption. Firstly, antibiotics were used as therapeutic prescriptions although the advances on knowledge and development of new compounds were also used as a preventive way and as growth promoters. What must be questionable its not the presence of these residuals but surely what kind of residual and their concentrations. In fact, the modern analytic technologies make possible the detection of parts per billions (ppb) of chemical substances, compounds or medicine metabolites of veterinary use in food and in these quantities, hardly they would be dangerous to the consumer health. However, it\'s necessary to establish more efficient analytic methodologies to help on agriculture supervising organs in the control of these antibiotics use in the animal production. Thus, how to help in the quality control, mainly in industries that export to get the market expansion, where this activity is almost an imposition at the international trade context of farming products \"in natura\" and industrialized ones. Due to these necessities, the goal of our present work was to standard and develop immunoenzymatics reagents to the ciprofloxacin researching from animal products. The results obtained on development of these immunoreagents were positive when correlated (r2= 0,9588) to the kit RIPASCREEN® Enro/Cipro from R-Biopharm reference. At the light of these knowledge, it?s possible to conclude that Brazil has adequate proper-structure to standardize these quick tests, to avoid in this way a high cost to import these kits.

A embriogênese somática é uma técnica alternativa com potencial aplicação na propagação clonal de plantas, além de ser uma excelente ferramenta para estudos básicos e análise dos eventos moleculares e bioquímicos que ocorrem durante a embriogênese vegetal. Contudo, apesar de várias pesquisas relativas a embriogênese somática, a falta de conhecimento sobre os fatores que controlam o fenômeno, comprova que existem ainda muitos pontos a serem entendidos sobre o processo. Deste modo, o presente estudo foi concebido com o intuito de estudar a embriogênese somática de duas espécies de citros, as quais apresentam diferentes graus de eficiência na produção de embriões somáticos. Inicialmente, buscou-se avaliar comparativamente o processo de embriogênese somática de duas espécies de citros, observando-se as diferenças estruturais através de análises histológicas e histoquímicas, e as diferenças na expressão do gene AtSERK1 nos calos, através de hibridização in situ. Para instalação dos experimentos, foram utilizados calos provenientes de nucelos de duas espécies, Citrus sinensis L. Osbeck, variedade Valência, e Citrus limonia Osbeck, variedade limão ‘Cravo’. Amostras de calos em meio de multiplicação, em meio de obtenção de embriões, e embriões somáticos nas diferentes fases de desenvolvimento foram coletados para observações anatômicas e histoquímicas através de microscopia óptica e realização de estudo da expressão gênica através de hibridização in situ. Com relação à morfologia, os principais resultados obtidos demonstraram que existem diferenças anatômicas e histoquímicas entre calos de limão ‘Cravo’ e ‘Valência’. Em relação à expressão gênica, os resultados evidenciaram a expressão do gene AtSERK1 em calos das duas espécies de citros, tanto em meio de multiplicação, com sacarose, como em meio de indução, com maltose, indicando que o potencial embriogênico já está instalado no calo em meio de multiplicação. Pode-se concluir também que este gene é conservado entre Arabidopsis e Citrus. Desenvolveram-se, também, experimentos para estudar os efeitos de alguns tratamentos (frio, dessecação, agrupamentos celulares) na otimização da indução e sincronização da embriogênese somática de C. sinensis, var. Valência. Neste sentido, calos nucelares de laranja ‘Valência’, procedentes do meio de multiplicação foram mantidos no mesmo meio líquido em agitador orbital a 200 rpm durante 24h e posteriormente peneirados de acordo com os tratamentos. As peneiras utilizadas apresentaram malhas de 150 µm (P1), 300 µm (P2) e 600 µm (P3). Além do efeito das peneiras também foram observados os efeitos de exposição ao frio (4 o C por quatro semanas, ausência de luz) e dessecação (27 o C, em placas de Petri na ausência de meio de cultura, seis dias, ausência de luz), sendo posteriormente mantidos em B.O.D., a 26 ± 1 o C e intensidade luminosa de 300 lux. Os resultados obtidos permitiram concluir que em relação ao desenvolvimento e produção de embriões somáticos o fator peneira foi superior ao fator estresse na freqüência embriogênica. O desenvolvimento de embriões somáticos em Citrus sinensis ocorre mais eficientemente a partir de agrupamentos celulares de tamanhos específicos. Somatic embryogenesis is an alternative technique with potential application for clonal propagation of plants, and an excellent tool for basic studies and analysis of the molecular and biochemical events that occur during embryogenesis. However, although many studies have been done, the factors that control somatic embryogenesis are not completely understood. Thus, the present study proposes to evaluate aspects of somatic embryogenesis of two citrus species, which differ in efficiency for the production of somatic embryos. Initially, a comparison of the embryogenic process was done in terms of structural and histochemical analysis and evaluation of the expression of the gene AtSERK1 in callus cultures through in situ hybridization. For the experiments, callus cultures obtained from nucelli of two species, Citrus sinensis L. Osbeck, cv. Valencia, and Citrus limonia Osbeck, cv. Rangpur lime were used for anatomical and histochemical observations, callus samples from cultures in multiplication medium, in embryo induction medium, and somatic embryos in different stages of development were collected. Callus and embryo samples were also collected for analysis of gene expression through in situ hybridization. The anatomical and histochemical analysis showed differences between Rangpur lime and Valencia callus cultures. The in situ hybridization results evidenced the expression of the gene AtSERK1 in cultures of both citrus species, and also in both culture conditions: multiplication medium, with sucrose, and embryo induction medium, with maltose, indicating that the embriogenic potential is already installed in the callus cultures in multiplication medium. The results also lead to the conclusion that the gene is conserved between Arabidopsis thaliana and Citrus. Experiments aiming to synchronize the somatic embryogenesis formation in C. sinensis, cv. Valencia, were installed testing the effect of cold, desiccation and cell cluster size on somatic embryo formation. For that, callus cultures of ‘Valência’ sweet orange were cultivated in liquid medium in an orbital shaker, at 200 rpm, for 24h, and sieved through the following screens: 150 µm (P1), 300 µm (P2) and 600 µm (P3). The cell aggregates were then exposed to cold (4 o C, for four weeks, in the dark) and desiccation (27 o C, in Petri plates without culture medium, six days, in the dark) treatments, and cultured in incubators at 26 ± 1 o C and 300 lux of light intensity. The results lead to the conclusion that the separation of cultures in clusters of different sizes and the stress treatments were effective for synchronization and embryogenesis frequency. The size of cell clusters was the most important factor.

A Cryptomeria japonica D. Don. "elegans" é uma conífera pertencente à família Taxodiaceae, que apresenta crescimento rápido e boas respostas à fertilização. Dez variações nas concentrações de nitrato e amônio presentes no meio de cultura MS foram realizadas no cultivo 'in vitro' de brotações obtidas a partir de explantes primários de ápices caulinares durante 90 dias. A cada 30 dias foram avaliados, a taxa de crescimento relativo e o incremento em massa vegetal seca. Ao final de 90 dias, foram realizadas análises dos teores de proteínas solúveis totais, aminoácidos solúveis totais, carboidratos não-estruturais totais e eletroforese de proteínas totais em gel de poliacrilamida. Os tratamentos que exibiram resultados mais satisfatórios foram utilizados em um experimento com diferentes relações auxina/citocinina, onde foram avaliadas a produção de brotações e a altura média das mesmas aos 30, 60 e 90 dias de cultivo. As concentrações de nitrato e amônio entre 25 e 31mmol.L -1 e até 5mmol.L -1, respectivamente, foram as mais efetivas para o crescimento e desenvolvimento do material vegetal cultivado 'in vitro'. Durante o experimento com diferentes concentrações de ANA e BAP, foi possível observar que a produção de brotações, etapa essecial para um processo de micropropagação eficiente, foi superior quando as combinações entre nitrato e amônio foram 22,5 + 2,5mmol.L -1 e 25,0 + 5,0mmol.L -1. Cryptomeria japonica D. Don. "elegans" is a fast-growing tree belonging to the Taxodiaceae and it is considered a responsive species 'in vitro'. The aim of this work was to evaluate the effect of ten different concentrations of nitrate and ammonium in morphogenesis 'in vitro' of Cryptomeria japonica D. Don. "elegans" explants. Analysis of relative growth rate, total soluble proteins, total soluble aminoacids, total non structural carbohydrates and electrophoresis of total proteins was performed in order to observe the role of different nitrogen sources on the growth and development of the explants cultivated 'in vitro'. The concentratios of nitrate and ammonium that showed the best results were selected and used in an experiment with different concentrations of NAA and BAP to observe the shoot production. Each experiment was analised at the 30 th , 60 th and 90 th days of culture. The biochemical analysis was performed only at 90 th day of cultive. Concentrations from 26 to 31mmol.L -1 of NO3 - and lower than 5mmol.L -1 of NH4 + were the most effective for growth and development of the explants. However, the experiment with different concentrations of NNA and BAP showed that the best shoot production was achieved on 22,5mmol.L -1 of NO3- + 2,5mmol.L -1 of NH4 + and on 25,0mmol.L -1 of NO3 - + 5,0mmol.L -1 of NH4 + and with 2,0mg.L -1 of BAP + 1,0mg.L -1 of NNA.

O trabalho teve como objetivo otimizar a metodologia de transformação de plantas de Eucalyptus grandis; uma importante espécie florestal amplamente cultivada no Brasil. Foram estudadas a transformação por agrobiobalística de explantes com acúmulo de gemas axilares e do SAAT ('sonication-assisted Agrobacterium mediated transformation'). Ambas as técnicas causam microferimentos nos explantes aumentando a penetração da Agrobacterium nos tecidos vegetais. Na agrobiobalística o explante é submetido ao bombardeamento com micropartículas seguida por inoculação com a bactéria; enquanto que no SAAT o explante é submetido à sonicação por breves períodos de tempo na presença da bactéria. Os experimentos de transformação foram avaliados pela analise da expressão transitória do gene repórter uidA. Os parâmetros estudados na transformação por agrobiobalística foram: pressão de disparo ( pressão do gás hélio), distância de vôo das micropartículas, estabilidade da expressão do gene uidA e temperatura de co-cultivo. As freqüências mais elevadas da expressão da ß-glucuronidase foram observadas utilizando 1350 PSI de gás hélio, 9,5 cm de distância de vôo dos microprojéteis e temperatura de 26ºC durante o co-cultivo. Para otimizar a transformação de explantes com acúmulo de gemas axilares foi realizado um estudo histológico, o qual permitiu constatar que as áreas meristemáticas estão localizadas na superfície do explante. O meristema é formado pela rediferenciação de células da epiderme e das primeiras camadas do parênquima cortical caracterizando sua origem exógena. A localização histológica do produto da expressão do gene repórter uidA mostrou que esta expressão ocorre apenas em células já diferenciadas, não sendo encontrada durante os tratamentos com células meristemáticas. Para realizar os ensaios com SAAT foi selecionado previamente um clone com características favoráveis à transformação: alta taxa de regeneração, susceptibilidade a agentes seletivos e à transformação por Agrobacterium. A maior taxa de plantas que apresentaram atividade da ß-glucuronidase foram observadas utilizando-se 60 segundos de sonicação; tempos superiores a este causaram um comprometimento da regeneração do explante. A sonicação adicional, após a sonicação preliminar do explante, juntamente com a bactéria, melhora a eficiência do processo de transformação. Os resultados demonstram que o uso de SAAT é viável na transformação de eucalipto com Agrobacterium. The research project aimed the establishment of a method for genetic transformation of Eucalyptus grandis; an important and widely planted forestry tree in Brazil. Two techniques of transformation were tested: agrobiolistic of explants with accumulation of meristematic cells and SAAT ('sonication-assisted Agrobacterium mediated transformation'). Both systems of transformation aim to produce micro wounds in the explants in order to increase the Agrobacterium penetration in the tissues. In the case of agrolistica the explant was previously submitted to micro projectile bombardment, followed by bacteria inoculation. On the other hand in the SAAT technique the explante is submitted to short periods (few seconds) of sonication together with the bacteria. Transformation was evaluated by measuring the expression of the reporter gene uidA which codes the ß-glucuronidase enzyme. Several parameters were tested for agrobiolistic such as, gas pressure (helium), flight distance of micro projectiles, gene expression of the uidA, co-cultivation temperature. The highest values of ß-glucuronidase were observed for 1350 PSI, 9.5 cm from target and 26O C for co-cultivation. A histological analyze was carried out to check it the meristematic tissues were being transformed. The results showed that the meristematic tissues were localized in the surface of the explante. The meristem is formed by the dedifferentiation of epidermal cells and the first layers of the cortical parenchyma indicating its exogenous origin. The histological location of the reporter gene uidA showed that the expression occurs only in cells already differentiated, and not in meristematic cells. For SAAT experiments a clone was previously selected for favorable characteristic of transformation: high rate of regeneration, susceptibility of selective agents and to Agrobacterium transformation. The highest rate of ß-glucuronidase activity were observed for 60 s of sonication; higher sonication time reduced the efficiency of regeneration. Additional sonication, following a preliminary sonication of explante , in the presence of the bacteria, improved the efficiency of transformation. The results provided evidence the SAAT is a viable technique for Eucalyptus transformation via Agrobacterium.

Este trabalho teve como objetivo a obtenção de embriogênese somática em cultivares brasileiras de bananeira e subsequente conversão dos embriões somáticos em plantas. Estudos desta natureza visam possibilitar trabalhos futuros que envolvam a manipulação genética, para melhoramento desta cultura. O projeto consistiu, primeiramente, na indução de embriogênese somática a partir de ápices vegetativos testando-se: diferentes auxinas (dicamba, picloram, 2,4-D e NAA) em meio MS (Murashige & Skoog, 1962); três meios de cultura com diferentes razões entre nitrogênio nítrico e amoniacal (NO3-: NH4+); e tempo necessário para permanência do explante no meio de indução. Indução foi obtida, porém, embriões somáticos não converteram-se em plantas. Em seguida, estudou-se o efeito do tratamento de maturação, através de cultivo contendo concentração mais elevada de sacarose (60 g/L) ou PEG (polietilenoglicol), na conversão e germinação de embriões somáticos a partir de ápices vegetativos. Alternadamente, foram utilizados explantes de inflorescências masculinas na indução de embriogênese somática. Em todas as etapas foram realizadas análises morfo-anatômicas através de microscopia ótica e eletrônica de varredura. Na indução a partir de ápices vegetativos, as auxinas dicamba e picloram apresentaram formação de estruturas embriogênicas e embriões somáticos, enquanto que as demais apresentaram ausência de resposta embriogênica. Cortes histológicos dos explantes em meio com dicamba, mostraram regiões embriogênicas com formação de pró- embriões e regiões meristemáticas não organizadas, que possivelmente, originaram estruturas monopolares. Os embriões somáticos eram morfologicamente semelhantes aos embriões zigóticos de Musa acuminata, porém, diferenciação de protoderme, ápices meristemáticos e procâmbio, não foram observados. Entre os três diferentes meios de cultura testados na indução (SH, FN-Lite ou 1/2 MS), o meio FN-Lite e 1/2 MS, com razão nitrato:amônio de 4:1 e 2:1, respectivamente, apresentaram melhor qualidade dos embriões somáticos, do que os embriões em meio SH com razão de 9,5:1, e o melhor tempo de indução foi de 4 semanas. Cortes histológicos foram realizados para observação das características dos embriões somáticos formados. O tratamento de maturação dos embriões somáticos favoreceu a conversão dos embriões em plântulas, porém, em baixo percentual. Frequentemente foram observadas, tanto em meio de indução, como de maturação, uma alteração de desenvolvimento dos embriões somáticos, com um crescimento preferencial do ápice radicular, e ausência de desenvolvimento do ápice caulinar. Em explantes de inflorescências masculinas, identificaram-se através de análises morfo-anatômicas, dois tipos de embriões somáticos: um tipo semelhante ao embrião zigótico de Musa acuminata e, em menor frequência, desenvolveram-se embriões somáticos semelhantes a embriões zigóticos de outras monocotiledôneas, indicando que em um único protocolo de embriogênese somática, é possível ocorrer a formação de diferentes padrões morfológicos de embriões somáticos. The present work aimed to obtain somatic embryogenesis in Brazilian cultivars of banana and their subsequent conversion into plants. These studies are important for the development of protocols involving genetic manipulation techniques for crop improvement of this species. Initially, the induction of somatic embryogenesis from shoot apices was tested using MS medium (Murashige & Skoog, 1962) supplemented with different auxins (dicamba, picloram, 2,4-D and NAA); three culture media with different ratios of nitric:amoniacal nitrogen (NO3-: NH4+); and length of time in the induction medium. Induction was obtained but the somatic embryos did not convert into plants. Maturation treatments with higher sucrose levels or PEG (polyethylene glicol) were then tested. In addition, the use of male inflorescences were used as explants for somatic embryo induction. During the experiments, morpho-anatomic analyses were performed through light and scanning electron microscopy. When shoot apices were used as explants, the auxins dicamba and picloram showed the formation of embryogenic structures and somatic embryos, while 2,4-D and NAA were not. Explants in medium containing dicamba presented both embryogenic regions with pro-embryos and non-organized meristematic regions, which may have formed unipolar structures. The somatic embryos were morphologically similar to the zygotic embryos of Musa acuminata, however lacked the differentiation of protoderm, meristematic apices and procambium. These somatic embryos did not convert into plants. Among the three different culture media tested for induction (SH, FN-Lite or 1/2 MS), FN- Lite and 1/2 MS provided better quality embryos and four weeks was considered the best period for induction. These two media have a 4:1 and 2:1 NO3-: NH4+ ratio, while SH has a 9,5:1 ratio. Histological sections were done to evaluate the somatic embryos formed. Somatic embryo maturation treatments favored the conversion of embryos into plants, however at low frequency. Frequently, somatic embryos showed the development of the root pole only. This behavior was observed both during induction and maturation. Male inflorescence explants formed two types of somatic embryos: one morphologically resembling the zygotic embryo of Musa acuminate, and less frequently somatic embryos with morphological characteristics of other monocots. This behavior indicates that in one protocol of somatic embryogenesis it is possible to obtain different morphological patterns of somatic embryos.

Estudos in vitro foram realizados com Passiflora edulis f. flavicarpa, P. alata eP. suberosa visando obtenção de plântulas através da organogênese ou embriogênese somática. Inicialmente, estudos foram feitos para adequar o meio de cultura visando melhor crescimento in vitro de P. edulis f. flavicarpa. O novo meio de cultura (MSM), foi desenvolvido com base nos níveis nutricionais considerados adequados para plantas desta espécie, cultivadas em campo. Segmentos nodais de P. edulis f. flavicarpa, em casa de vegetação, foram cultivados em meio de cultura MS ou MSM contendo 3,0 mg/L de BAP, durante 3 subcultivos. Foi feita análise foliar das plântulas e observou-se que aquelas cultivadas em meio MSM apresentaram maiores níveis de Ca, Cu, Fe, Mg e S e baixos teores de B e CI. Plântulas cultivadas em meio MS apresentaram sintomas de deficiência mineral, enquanto plântulas cultivadas em meio MSM apresentaram folhas sadias. Organogênese a partir de discos foliares de P. edulis f. flavicarpa P. alata e P. suberosa foi obtida. Benzilaminopurina (BAP), nas doses de 0,5 ou 1,0 mg/L foi eficiente para produção gemas adventícias em P. suberosa . A proliferação de calos foi observada nas bordas dos discos, como fase intermediária da organogênese. BAP (0,6; 0,8 ou 1,0 mg/L) ou thidiazuron (TOZ) (0,25; 0,50; 0,75 ou 1,0 mg/L) induziram a formação de gemas adventícias nas espécies P. edulis f. flavicarpa P. alata. Cortes histológicos mostraram que as gemas formam formadas pela proliferação de células nas bordas dos discos. Essas células diferiram daquelas formadas nos discos de P. suberosa, sendo estas, mais friáveis, com proliferação não organizada. TDZ promoveu a formação de um grande número de gemas em relação ao BAP, nos discos de P. edulis f. flavicarpa P. alata. Entretanto, as gemas induzidas com BAP se desenvolveram mais rapidamente. Plântulas de P. suberosa e P. edulis f. flavicarpa foram alongadas e enraizadas in vitro e aclimatadas em casa de vegetação. Gemas de P. alata não se desenvolveram em meio de alongamento. Estruturas globulares foram formadas juntamente com gemas adventícias nos discos de P. edulis f. flavicarpa P. alata., quando cultivados em meio contendo BAP ou TDZ. Cortes histológicos e observações em microscópio eletrônico de varredura mostraram que essas estruturas eram embriões somáticos em estádios iniciais de desenvolvimento In vitro studies were done with Passiflora edulis f. Flavicarpa, P. alata and P. suberosa to obtain plantlets by organogenesis or somatic embryogenesis. Initially studies were done to adequate the culture medium for better growth of P. edulis f. flavicarpa plants in vitro. A new medium (MSM) was developed based on the leaf nutrients levels considered adequate for this species in the field. Cultures were initiated with nodal segments of P. edulis f. flavicarpa plantlets grown in the greenhouse, in MS or MSM with 3.0 mg/L BAP, during 3 subcultures. Leaf mineral analysis was done and it was observed that plantlets grown in MSM medium had higher Ca, Cu, Fe, Mg and S levels and lower B and CI levels. Plantlets grown in MS medium showed visible mineral deficiency symptoms, while plantlets grown in MSM had green healthy leaves. Organogenesis from leaf discs of P. edulis f. flavicarpa, P. alata and P. suberosa was obtained. Benzylaminopurine (BAP) at 0.5 or 1.0 mg/L was efficient to produce adventitious buds in P. suberosa. Proliferation of callus was observed on the edge of the leaf discs as an intermediary step. BAP (0,6; 0,8 or 1,0 mg/L) or thidiazuron (TDZ) (0,25; 0,50; 0,75 ou 1,0 mg/L) induced the formation of adventitious buds in both Passiflora edulis f. flavicarpa and P. alata. Histological sections showed that buds are formed from cells proliferating on the edge of the leaf discs. These cells were different from those formed in P. suberosa in that those were more friable, more characteristic of unorganized proliferation. TDZ seemed to form larger number of buds, compared to BAP both Passiflora edulis f. flavicarpa and P. alata, however buds induced in BAP grew faster. Plantlets of P. suberosa and P. edulis f. flavicarpa were rooted ,in vitro and acclimatized in the greenhouse. P. alata buds did not develop to form plantlets in vitro. Globular structures formed interspersed with buds in P. edulis f. flavicarpa and P. alata when leaf discs were introduced in medium containing TDZ ou BAP. Histological sections and observations under scanning electron microscopy showed that these structures were early stages of somatic embryos.
