3. Metabolic alterations caused by smoking: the use of 1H-NMR in blood plasma analysis to unravel underlying mechanisms of lung cancers leading risk factor

Abstract

Nowadays, the screening methods for the early detection of lung cancer struggle with several limitations such as many false positive results and low sensitivity. The detection of specific biomarkers is of high interest to complement these conventional screening methods. The objective of this study is to prove the power of 1H-NMR in metabolomics for the detection of smoking behavior, which is the leading risk factor for lung cancer, and as such gain more insights in the metabolic alterations that are caused by smoking. In this research, 1H-NMR spectra of human blood plasma samples were divided in 110 integration regions, from which the integration values were used to train an OPLS-DA classification model that underwent further data reduction. Results show that a classification model could discriminate between individuals based on their smoking status with a sensitivity of 96 percent and a specificity of 94 percent. This study also demonstrates that by performing a pathway-specific variable reduction of almost 50 percent, the sensitivity and specificity of the model almost remains the same. To conclude, 1H-NMR analyses show that the bloods metabolic profile of a smoker is altered compared to that of a non-smoker. Also, pathway-specific variable reduction shows great potential to perform overall data reduction. This workflow could be interesting to apply in the identification of lung cancer, to potentially detect specific biomarkers.

Abstract

Grafting organophosphonic acids (PAs) on metal oxides has shown to be a flexible technology to tune the surface properties of metal oxides for various applications. The solvents applied in the commonly used synthesis method have associated impeding effect on tailoring the resulting modification degree. In this work, an alternative solid-phase manual grinding method is proposed that (i) is straightforward, (ii) can achieve controllable and higher modification degree, and (iii) excludes the use of solvent during the synthesis. Specifically, propylphosphonic acid (3PA) was grafted onto titania by manual grinding, and different modification degrees were obtained by varying the duration of the post-synthetic thermal treatment. Importantly, the solid-phase method can achieve a modification degree that is 25.0% higher than the maximal modification degree reached by the liquid-phase method, while its atom utilization efficiency is 4.8 times (toluene-based) or 7.5 times (water-based) that of the liquid-phase method. Abstract

An ideal wound dressing not only needs to absorb excess exudate but should also allow for a moist wound-healing environment as well as being mechanically strong. Such a dressing can be achieved by combining both a natural (alginate) and synthetic (poly(ethylene glycol) polymer. Interestingly, using an electron beam on (meth)acrylated polymers allows their covalent crosslinking without the use of toxic photo-initiators. The goal of this work was to crosslink alginate at different methacrylation degrees (26.1 and 53.5% of the repeating units) with diacrylated poly(ethylene glycol) (PEGDA) using electron-beam irradiation at different doses to create strong, transparent hydrogels. Infrared spectroscopy showed that both polymers were homogeneously distributed within the irradiated hydrogel. Rheology showed that the addition of PEGDA into alginate with a high degree of methacrylation and a polymer concentration of 6 wt/v% improved the storage modulus up to 15,867 ± 1102 Pa. Gel fractions > 90% and swelling ratios ranging from 10 to 250 times its own weight were obtained. It was observed that the higher the storage modulus, the more limited the swelling ratio due to a more crosslinked network. Finally, all species were highly transparent, with transmittance values > 80%. This may be beneficial for the visual inspection of healing progression. Furthermore, these polymers may eventually be used as carriers of photosensitizers, which is favorable in applications such as photodynamic therapy.

Abstract

The initial changes in the microstructure of alkali‐activated slag cement (AAS) characterize the rheological behavior, initial structuration, and setting times of the mixture. In this study, the relationships among the rheological behavior, solidification process, and nanostructure changes of sodium hydroxide‐activated slag (NH–AAS) and sodium silicate‐activated slag (SS–AAS) pastes over time are investigated by small amplitude oscillation shear test, isothermal calorimetry, scanning electron microscopy analyses, and 1H–29Si cross‐polarization magic‐angle spinning nuclear magnetic resonance spectroscopy (CP MAS NMR) and 29Si MAS NMR spectroscopy. Test results showed that the NH–AAS reaches its initial setting time following a continuously proceeding structuration process, whereas the SS–AAS reaches its initial setting with an abrupt increase in the structuration process. At their initial setting times, the NH–AAS and SS–AAS mixtures release similar heat and reach a similar reaction degree. The SS–AAS mixture had more N–A–S–H formation than C–A–S–H at the initial setting time. On the other hand, the amount of C–A–S–H was similar to N–A–S–H in the NH–AAS mixture. 45. A New Cost-Benefit Economic Model Approach of Hemodialyzer Reuse

Abstract

In this study, a new cost-benefit economic model for hemodialyzer reuse has been developed considering all of the direct costs (dialyzer price, disinfection fluid price, reverse osmosis water cost, personnel, and miscellaneous) as well as the number of disinfections applied to the hemodialyzer. The maximum number of disinfections/reuses for different models of hemodialyzer was estimated using statistical analysis based on the information obtained from a total of 60 adult patients on maintenance hemodialysis for approximately 4 years; from a hospital in Santiago de Cuba province, Cuba. An equal number of treatments (100) was evaluated for each hemodialyzer including 2,800 total reuses. The total cost savings for reuse/disinfection using the new economic approach is compared with the single-use modality. Obtained results by applying the proposed model indicated that the correlation between the economic advantages of the reuse/disinfection process in the total cost of the hemodialysis treatment significantly depends on the type of hemodialyzer used for the treatment, the disinfection price, the virgin hemodialyzer price, the disposal cost and its price reduction because of the number of reuses and the maximum possible reuses that could be applied to the hemodialyzer. ispartof: Engineering Science & Technology vol:3 issue:2 pages:169-187 status: accepted

47. Cell-Interactive Gelatin-Based 19F MRI Tracers: An In VitroProof-of-Concept Study

Abstract

Cross-linked gelatin-based hydrogels are highly promising cell-interactive, biocompatible, and biodegradable materials serving tissue engineering. Moreover, gelatins with covalently bound methacrylamide (gel-MA) and 2-aminoethyl methacrylate moieties (gel-AEMA) can be cross-linked through ultraviolet (UV) irradiation, which allows light-based three-dimensional (3D)-printing of such hydrogels. Furthermore, the physicochemical and biological properties of these hydrogels can be broadly tuned by incorporating various comonomers into the polymer chains, which makes these hydrogels a widely applicable platform in tissue engineering and reconstructive surgery. However, monitoring the degradation rate of hydrogel-based implants in vivois challenging, thereby prohibiting their broad clinical transition and further research. Therefore, herein, we describe the synthesis of 3D-printable gelatin-based hydrogels with N-(2,2-difluoroethyl)acrylamide (DFEA), detectable with the chemical shift of −123 ppm, which enables us to monitor these implants in vivowith 19F magnetic resonance imaging (MRI) and assess their degradation kinetics. Next, we describe the physicochemical and biological properties of these hydrogels. Adding DFEA monomers into the reaction mixture accelerates their cross-linking kinetics. Moreover, increasing the DFEA content within the hydrogels increases their swelling ratio and 19F MRI signal. All hydrogels were detectable at small quantities (<16 mg) using 19F MRI. Moreover, our hydrogels supported the cell proliferation of adipose tissue-derived stem cells (ASCs) and had tunable biodegradation rates. Finally, we present a strategy for increasing the DFEA content without affecting the mechanical properties. Our results may be implemented in the future development of hydrogel implants, whose fate and biodegradation rate can be monitored via19F MRI.
