Abstract Background We have previously developed a DNA-based vaccine, INO-4500, encoding the Lassa lineage IV glycoprotein precursor. INO-4500, when delivered with electroporation, elicited humoral and cellular responses, and conferred 100% protection in cynomolgus non-human primates. Here, we expanded the characterization of INO-4500 assessing immunogenicity and protective efficacy of lower doses and single immunization, and the durability of immune responses. Methods The study was divided into three arms evaluating INO-4500 vaccination: Arm 1 – Dosing regimen; Arm 2 – Single immunization; and Arm 3—Durability of immune responses and protective efficacy. Humoral and T cell responses were assessed by IgG binding ELISA, IFNγ ELISpot and flow cytometry-based T cell activation assays. NHPs were challenged with a lethal dose of Lassa lineage IV 8 weeks (Arms 1 and 2) or one year (Arm 3) after immunization. NHPs were assigned clinical scores and monitored for survival. Viremia, virus neutralization and release of soluble mediators were assessed post-challenge, as well as disease pathology following NHPs death or euthanasia. Results INO-4500 induces dose-dependent immune responses and protective efficacy. Animals receiving two doses of 2 mg of INO-4500 show complete short- and long-term LASV protection. NHPs receiving 1 mg of INO-4500 are protected from LASV challenge one year after vaccination but are only partially protected 8 weeks post-vaccination. LASV-specific memory T cells are present in vaccinated NHPs one year after vaccination. INO-4500 vaccination prevents NHPs from developing severe disease. Conclusions These studies demonstrate that INO-4500 can provide short- and long-term protection in NHPs from lethal LASV challenge.

Abstract Background Effective endodontic treatment requires the use of a root canal sealer with optimal properties to ensure a hermetic seal, prevent reinfection, and promote healing. Despite the availability of various sealers, a standardized evaluation system still needs to be improved. Objectives To develop a Clinical Utility Index (CUI) that systematically evaluates and ranks root canal sealers based on their sealing ability, antimicrobial efficacy, flow rate, and solubility. Methods The CUI was developed through a structured process involving expert identification, panel discussions, and the establishment of scoring criteria. Five sealers were evaluated (Sealers A, B, C, D, and E). Mean values for the core properties were calculated, and sealers were ranked accordingly. The total CUI for each sealer was computed based on the assigned scores for each property. Results Sealer B achieved the highest CUI at 95%, demonstrating superior performance across all core properties. Sealer C followed with a CUI of 80%, while Sealer A ranked third with 60%. Sealers D and E showed the lowest performance, with CUIs of 30% and 35%, respectively, highlighting deficiencies in multiple properties. Discussion The CUI provides a comprehensive evaluation framework for root canal sealers, facilitating informed decision-making by practitioners. Sealer B's high CUI underscores the importance of balancing sealing ability, antimicrobial effect, flow rate, and solubility. The results align with existing literature emphasizing the critical role of these properties in endodontic success. Conclusion The CUI offers a robust and balanced method for evaluating root canal sealers, aiding in selecting the most suitable sealer based on empirical data. Future research should refine the index and validate its applicability in diverse clinical scenarios to enhance endodontic treatment outcomes.

Abstract Backgroud Before fertilization, spermatozoa undergo a crucial maturation step called capacitation, which is a unique event regulates the sperm’s ability for successful fertilization. The capacitation process takes place as the spermatozoa pass through the female reproductive tract (FRT). Dihydrolipoamide dehydrogenase (DLD) protein is a post-pyruvate metabolic enzyme, exhibiting reactive oxygen species (ROS) production which causes capacitation. Additionally, other vital functions of DLD in buffalo spermatozoa are hyperactivation and acrosome reaction. DLD produces the optimum amount of ROS required to induce capacitation process in FRT. Depending on physiological or pathophysiological conditions, DLD can either enhance or attenuate the production of reactive oxygen species (ROS). Aim of this study was to investigate whether changes in the production of ROS in sperm cells can impact their ability to fertilize by triggering the capacitation and acrosome reaction. Results In this study, abundance of DLD protein was quantified between high (n = 5) and low fertile bull (n = 5) spermatozoa. Abstract Background This study aims to evaluate the compressive strength, solubility, radiopacity, and flow of Bromelain (BR)-modified Biodentine (BD) for direct pulp capping (DPC). This is suggested to determine the impact of BR on the physical properties of BD. Methods Eighty samples were prepared according to the ISO and ADA specifications and evaluated for compressive strength, solubility, radiopacity, and flow. The compressive strength was evaluated at 24 h and 21 days via a universal testing machine. The solubility was determined by weight loss after 24-hours immersion in deionized water. Radiopacity was assessed via X-ray with aluminum step-wedges, and flow was measured by the diameter of the discs under a standard weight. Independent sample t-tests were used to statistically assess the data. A significance level of 5% was considered. Results The compressive strength was 41.08 ± 1.84 MPa for BD and 40.92 ± 1.80 MPa for BR + BD after 24 h, and 88.93 ± 3.39 MPa for BD and 87.92 ± 3.76 MPa for BR + BD after 21 days, with no significant differences. Solubility was slightly greater in the BR + BD (2.75 ± 0.10%) compared to BD (2.62 ± 0.25%), but not significantly different. The radiopacity was similar between BD (2.82 ± 0.11 mm) and BR + BD (2.73 ± 0.10 mm). BR + BD resulted in significantly greater flow (9.99 ± 0.18 mm) than did BD (9.65 ± 0.27 mm) (p ≤ 0.05). Conclusion BR-modified BD maintains BD’s physical properties, with improved flow, making it a promising DPC agent that warrants further study.

Sahiwal cattle, known for their high milk yield, are propagated through artificial insemination (AI) using male germplasm, largely contingent on semen quality. Spermatozoa, produced in the testes, carry genetic information and molecular signals essential for successful fertilization. Seminal plasma, in addition to sperm, contains nano-sized lipid-bound extracellular vesicles (SP-EVs) that carry key biomolecules, including fertility-related miRNAs, which are essential for bull fertility. The current study focused on miRNA profiling of SP-EVs from high-fertile (HF) and low-fertile (LF) Sahiwal bulls. SP-EVs were isolated using size exclusion chromatography (SEC) and characterized by dynamic light scattering (DLS) and nanoparticle tracking analysis (NTA). Western blotting detected the EV-specific protein markers TSG101 and CD63. The DLS analysis showed SP-EV sizes of 170–180 nm in HF and 130–140 nm in LF samples. The NTA revealed particle concentrations of 5.76 × 1010 to 5.86 × 1011 particles/mL in HF and 5.31 × 1010 to 2.70 × 1011 particles/mL in LF groups, with no significant differences in size and concentration between HF and LF. High-throughput miRNA sequencing identified 310 miRNAs in SP-EVs from both groups, with 61 upregulated and 119 downregulated in HF bull. Further analysis identified 41 miRNAs with significant fold changes and p-values, including bta-miR-1246, bta-miR-195, bta-miR-339b, and bta-miR-199b, which were analyzed for target gene prediction. Gene Ontology (GO) and KEGG pathway analyses indicated that these miRNAs target genes involved in transcription regulation, ubiquitin-dependent endoplasmic reticulum-associated degradation (ERAD) pathways, and signalling pathways. Functional exploration revealed that these genes play roles in spermatogenesis, motility, acrosome reactions, and inflammatory responses. qPCR analysis showed that bta-miR-195 had 80% higher expression in HF spermatozoa compared to LF, suggesting its association with fertility status (p < 0.05). In conclusion, this study elucidates the miRNA cargoes in SP-EVs as indicators of Sahiwal bull fertility, highlighting bta-miR-195 as a potential fertility factor among the various miRNAs identified.

Key Clinical Message Left‐atrial septal pouch (LASP) is a newly described anatomical variant caused by incomplete fusion of the septa primum and secundum. This case visualizes LASP as a potential nidus for blood stasis and cardiac embolism and highlights the need for consideration of TEE in the work up of cryptogenic stroke.

Abstract Echocardiography frequently serves as the first-line treatment of diagnostic imaging for several pathological entities in cardiology. Artificial intelligence (AI) has been growing substantially in information technology and various commercial industries. Machine learning (ML), a branch of AI, has been shown to expand the capabilities and potential of echocardiography. ML algorithms expand the field of echocardiography by automated assessment of the ejection fraction and left ventricular function, integrating novel approaches such as speckle tracking or tissue Doppler echocardiography or vector flow mapping, improved phenotyping, distinguishing between cardiac conditions, and incorporating information from mobile health and genomics. In this review article, we assess the impact of AI and ML in echocardiography.

Background: Patient anxiety is known to influence various aspects of dental procedures, including endodontic treatments. However, the extent of its impact on the efficacy of such procedures remains uncertain. Understanding this influence is crucial for improving patient outcomes and optimizing treatment protocols. Materials and Methods: A prospective cohort study was conducted involving 100 patients undergoing endodontic procedures. Patient anxiety levels were assessed using standardized anxiety scales before and after treatment. The efficacy of endodontic procedures was evaluated based on posttreatment pain scores, success rates of root canal treatments, and patient-reported satisfaction levels. Results: The mean pre-treatment anxiety score was 45.7 (SD = 9.2), while the mean posttreatment anxiety score reduced significantly to 32.4 (SD = 7.6), indicating a significant reduction in anxiety levels following endodontic procedures (P < 0.001). Posttreatment pain scores were lower in patients with reduced anxiety levels, with an average pain score of 2.1 (SD = 1.3) compared to 3.8 (SD = 1.6) in patients with persistently high-anxiety levels (P = 0.002). Additionally, the success rate of root canal treatments was higher in patients with lower anxiety levels (85%) than those with higher anxiety levels (65%) (P = 0.015). Overall patient satisfaction was also significantly higher in the low-anxiety group (P < 0.001). Conclusion: Patient anxiety significantly influences the efficacy of endodontic procedures, affecting posttreatment pain, success rates of root canal treatments, and patient satisfaction levels. Efforts to reduce patient anxiety before and during endodontic procedures may lead to improved treatment outcomes and higher patient satisfaction.

Buffalo bulls are backbone of Indian dairy industry, and the quality of semen donating bulls determine the overall production efficiency of dairy farms. Seminal plasma harbor millions of lipid bilayer nanovesicles known as extracellular vesicles (EVs). These EVs carry a heterogenous cargo of essential biomolecules including fertility-associated proteins which contribute to fertilizing potential of spermatozoa. In this study, we explored size, concentration, and complete proteome profiles of SP EVs from two distinct fertility groups to uncover proteins influencing bull fertility. Through Dynamic Light Scattering (DLS) it was found that purified EVs were present in 7–14 size exclusion chromatographic (SEC) fractions with sizes ranging from 146.5 to 258.7 nm in high fertile (HF) and low fertile (LF) bulls. Nanoparticle Tracking Analysis (NTA) confirmed the size of seminal EVs up to 200 nm, and concentrations varying from 2.84 to 6.82 × 1011 and 3.57 to 7.74 × 1011 particles per ml in HF and LF bulls, respectively. No significant difference was observed in size and concentration of seminal EVs between two groups. We identified a total of 1,862 and 1,807 proteins in seminal EVs of HF and LF bulls, respectively using high throughput LC-MS/MS approach. Out of these total proteins, 1,754 proteins were common in both groups and about 87 proteins were highly abundant in HF group while 1,292 were less abundant as compared to LF bulls. Gene ontology (GO) analysis, revealed that highly abundant proteins in HF group were mainly part of the nucleus and involved in nucleosome assembly along with DNA binding. Additionally, highly abundant proteins in EVs of HF group were found to be involved in spermatogenesis, motility, acrosome reaction, capacitation, gamete fusion, and cryotolerance. Two highly abundant proteins, protein disulfide-isomerase A4 and gelsolin, are associated with sperm-oocyte fusion and acrosome reaction, respectively, and their immunolocalization on spermatozoa may indicate that these proteins are transferred through EVs. Our evidences support that proteins in EVs and subsequently their presence on sperm, are strongly associated with sperm functions. Altogether, our investigation indicates that SPEVs possess crucial protein repertoires that are essential for enhancing sperm fertilizing capacity.

Appendicular adenocarcinoma is a rare tumour, and the mucin variety is common. It is usually presented with symptoms such as abdominal pain, abdominal tenderness, nausea, vomiting, change in bowel habits, and unexplained weight loss. These symptoms can resemble those of other clinical conditions, making the diagnosis of appendicular adenocarcinoma challenging. Screening modalities such as Magnetic Resonance Imaging (MRI), Computed Tomography (CT) imaging, Contrast-Enhanced Computed Tomography (CECT), blood tests, and histopathological analysis can help confirm the diagnosis. This case is reported to highlight the unusual presentation and diagnostic difficulties associated with appendiceal adenocarcinoma. A 48 years old female patient presented with complaints of dull, continuous abdominal pain for the past six months, along with an abdominal lump and distension, without any other associated symptoms. Laboratory investigations for Cancer Antigen 125 (CA 125), β-Human Chorionic Gonadotropin (HCG), and α-fetoprotein were found to be within normal limits. Further, CECT imaging was suggestive of mucin-secreting adenocarcinoma of ovarian origin. The final diagnosis was confirmed by histopathological analysis as mucinsecreting adenocarcinoma of the appendix with pseudomyxoma peritonei and was managed through cytoreduction surgery.

The most common congenital gastrointestinal tract abnormality is Meckel's diverticulum, affecting 1% to 2% of the population. Meckel's diverticulum can sometimes house a gastrointestinal stromal tumor (GIST), which is a rare malignant tumor. A 63-year-old man presented with occult melena. It turns out to be a GIST developing from Meckel's diverticulum after an exploratory laparotomy. Occult melena is one of the causes of unexplained and intractable anemia. Every family and general physician should keep in mind the possibility of occult blood loss via the gastrointestinal tract when treating the patient.

Introduction: Root Canal Treatment (RCT) is the most common and effective treatment for saving natural tooth. Despite of modern technology and progress in the materials, failures in endodontic treatment still remains. Microbial infection majorly bacteria in the root canal is the prime cause of reinfection in root canal treatment. Endodontic instruments, solvents, lasers are the different techniques that can be used for removing gutta percha from the root canal. Aim: To compare the efficacy of orange oil, lemon oil, and xylene as solvents in removing gutta percha from curved root canals. Materials and Methods: This in-vitro study conducted in Sharad Pawar Dental College and Hospital, Sawangi Meghe, Wardha, Maharashtra, India, for three months and 24 freshly extracted teeth including incisors, premolars and molars having curved root will be taken. Then, RCT would be performed in all the extracted teeth by using cold lateral compression obturation method. Then all teeth would be divided in three groups (n=8). After one month, the Gutta Percha (GP) would removed by ProTaper Universal system. First group will use orange oil as solvent, second group will use lemon oil as solvent and the third group will use xylene as a solvent. The micro-Computed Tomography (CT) images prior to the application of solvent and after application of solvent will be taken and would be evaluated to check the percentage of filling material remaining. Results: The expected outcome is orange oil and lemon oil to act as a better root canal solvent, as compared to xylene. Conclusion: The determination and comparison of the efficacy of orange oil, lemon oil and xylene as GP solvents from the present planned study will help the clinicians during re-RCT treatment.

Introduction: Cocaine is a highly addictive substance that stimulates the sympathetic nervous system and cardiovascular system. A multitude of adverse cardiovascular events are associated with cocaine use including arrhythmia, congestive heart failure, coronary artery spasm and myocarditis. Case Report: We present a rare case of Takotsubo cardiomyopathy after recent use of cocaine in a female without any other identifiable risk factor. Discussion: Takotsubo cardiomyopathy (TCM) is a reversible cause of cardiomyopathy characterized by transient decrease in ejection fraction. Cocaine is a sympathomimetic that causes catecholamine surge and a variety of cardiovascular abnormalities. The association of cocaine use with Takotsubo cardiomyopathy is a rare occurrence, however the prognosis is generally good if diagnosis is made promptly. Conclusion: Physician vigilance is required to diagnose this rare cause of cardiomyopathy and reduce morbidity and mortality.

Introduction: Deep neck abscess in adults may occur as a result of local trauma, foreign body inoculation or immunosuppression. Method: We describe a case of deep neck abscess that developed after recent hospitalization and intubation for chronic obstructive pulmonary disease (COPD) exacerbation. Discussion: The incidence of deep neck abscesses has dramatically reduced due to the use of antibiotics. However, if present, they need prompt surgical drainage due to their proximity to the nearby vasculature of the neck. Conclusion: This case highlights the need for physician awareness of the clinical presentation of the unique complications of deep neck abscess developing after hospitalization.

This paper investigates the melting behaviour of phase change material (PCM) in a vertical thermal energy storage system with provision of thin rectangular fins of uniform and variable lengths on the heat transfer tube surfaces. The selected PCM and heat transfer fluid (HTF) are paraffin wax and water, respectively. The HTF is passed through the helically coiled copper tube of 10 mm diameter to melt the PCM. The time required to complete the melting of PCM in the system with fins is found to be five hours, whereas for the system without fins it is five hours and forty minutes, for the same conditions of constant water temperature of about 70°C and flow rate of 0.02 kg/s. HTF tube with fins is observed to be more effective with a 13.33% faster rate of melting when compared to that of the HTF tube without fins. Such a fast charging process will be helpful in storing maximum energy within a short period/duration of time shorter duration in for solar thermal and heat recovery applications during lean production times. ©2020. The outbreak of COVID- 19 disease has taken the world by surprise. The disease mainly affects the respiratory system and is caused by a strain of coronavirus named SARS-CoV-2. Due to its high recombination rates the virus has an immense potential to spread rapidly. Within months of the first case being reported in Wuhan China, the disease has turned rapidly into a pandemic. The world health experts are already actively looking for a definite cure or vaccine for the disease, but till that many of the countries across the globe are looking up onto mass quarantine measures as a method to control its spread. This article is an attempt to review the effectiveness of these measures and the possible adverse effects of the same. Other possible modes of prevention and controlling the spread of the disease have also been reviewed in the article.

Invasive cervical resorption is entirely uncommon entities and the etiology is poorly understood. A 19 year old patient presented with fractured upper left central incisor and sinus tract opening on the distobuccal aspect in cervical region. Radiographic examination shows irregular radiolucency over the coronal one-third and it extended externally towards the external invasive resorption. After sectional obturation, the defect was accessed surgically. The resorption area was chemomechanically debrided using irrigant solution. Fibre post placement using flowable composite resin and Mineral Trioxide Aggregate (MTA) was used to fill the resorptive defect, and the coronal access was temporarily sealed. Composite restoration was subsequently replaced with ceramic crown after 4 years. Radiographs at 1 and 4 years showed adequate repair of the resorption and endodontic success. Clinically and radiographically the tooth was asymptomatic, and no periodontal pocket was found after a 4-year followup.

Background Oral submucus fibrosis (OSMF) has a 0.2-0.5 prevalence rate in India. The malignancy rate of OSMF was found to be 7.6%. The focus of pathogenesis is currently on reactive oxygen species levels which can rise to extremely high levels, exceeding the human body’s antioxidant defense system and causing oxidative stress that goes beyond physiological limitations. Since oxidative stress plays such a significant part in carcinogenesis and other degenerative diseases, it is fair to believe that antioxidants will help to reduce or even stop these processes. Vitamin C is known for its antioxidant properties which act as a lipid-soluble free radical scavenger in cell membranes. But in some of studies a paradoxical effect of antioxidant doses was found. Thus, the present study will be conducted to evaluate the dose dependent efficacy of Vitamin C level in relation to oxidative stress level in subjects of OSMF. Methods After informed consent, the patient’s examination will be conducted. For each individual, 5ml of the blood sample will be withdrawn under all the aseptic precautions and oxidative stress level will be assessed depending on MDA & SOD level in a central research laboratory. Depending on oxidative stress level patients will be divided into two groups. Group I - Moderate oxidative stress in OSMF SI less than 240 but more than 192 Group II - High Oxidative Stress in OSMF SI more than 240 Single and double doses of Vitamin C will be given to patients accordingly and the MDA & SOD levels will be calculated on day 30,90 days and on 180 th day Conclusions Positive results of the study will give direction for choosing the correct dose of the antioxidant Vitamin C for OSMF depending on oxidative stress level.

Background: The aim will be to evaluate and compare the bonding of the total-etch adhesive to carious dentin in molars treated with and without 38% silver diamine fluoride using the universal testing machine and scanning electron microscope. Methods: 30 extracted teeth samples will be been randomly distributed into 2 groups (n=15). The test group will receive the intervention of 38% SDF whereas the control group will not receive any such intervention. Samples in both groups will be restored with resin composite. Assessment will be done based on the primary outcome, which will be determined by microtensile bond strength whereas the secondary outcome will be determined by fracture modes, i.e., cohesive, adhesive, or mixed fracture. Descriptive statistics will be used for the results of each group. Intergroup comparison will be done on the basis of independent sample t-test to detect the mean difference between the two groups. Independent t-test will be run to analyse the type of failure. In all tests, p-values below 0.05 will be considered statistically significant. Conclusion: The study may provide us with a new restorative technique that will not only be less invasive by preserving the remaining tooth structure but will also prevent the risk of caries progression beneath the restoration. Thus, this will contribute to the longevity of the tooth life by the maintenance of its pulpal health which will contribute to the durability and long-term successful outcome of the direct restoration procedure.

Background: The main factor contributing to surgical endodontic dissatisfaction is an inadequate seal at the apex. The material used for retrograde filling should prevent contaminants from penetrating periradicular tissue. So the objective of this study was to evaluate and compare the ability of Mineral Trioxide Aggregate (MTA), Zirconomer and bioactive bone cement to seal the root end as retrograde filling material with apices resected at 90° angles using dye penetration method under fluorescent microscope. Methods: 36 extracted teeth samples were cut at Cementoenamel junction horizontally. Following the customary protocols of, cleaning, shaping and obturation with gutta percha and AH Plus sealer, resection of samples was done 3mm at the apical end at 90 degree angle along the long axis of the tooth with the help of diamond disc. A depth of 3mm of root end cavity was made with diamond coated ultrasonic surgical tip. The samples were randomly allocated in 3 groups: Group I: MTA; Group II: Zirconomer; Group III: Bioactive Bone Cement. Coating of nail varnish was done with the roots except at the tip. Each material was compacted in the retrocavity using small pluggers. All the retrofilled samples were kept in acridine orange for 24 hours, following which cleaning & bucco-lingual sectioning (vertical) was carried out. Confocal laser scanning fluorescent microscope was used for checking the dye penetration in these samples thereby evaluating sealing ability. Results: Group III (Bioactive Bone Cement) had the highest root end sealing ability with mean (1.4 ± 0.51) followed by Group I (MTA) (2.3 ± 0.65) and lastly Group II (Zirconomer) (4.1 ± 0.57) as root end filling material with apices resected at 90°angle using dye penetration method under fluorescent microscopy which were statistically significant. Conclusions: Bioactive Bone cement can be recommended as an acceptable retrograde filling material.

Aim – The study aims to assess dentinal hypersensitivity after restoring the tooth with Cention N and resin modified glass ionomer cement (RMGIC) in class V cavities using the Schiff scale. Objective – This study will be conducted to assess the extent of dentin hypersensitivity to the patient's stimulus-response. Methods – A total of 36 patients reporting to the sharad pawar dental college and hospital will be taken having class V cavities. All the patients were divided into two groups. Using a high-speed flat-end straight diamond bur, a cavity with the dimensions two millimeters mmm in depth, three millimeters in width, and two millimeters in height was produced in the occlusal-gingival direction. In group A (n = 18) patients Cention N will be given as class V restoration and in group B (n = 18) patients RMGIC will be given. Follow-up will be done after seven days for evaluation of post-operative sensitivity. Informed written consent will be taken from the patients and the aims and methodology of the study will be explained before enrolment. Demographic data of the patient will be recorded. A statistical analysis will be done based on the recorded data using paired t-tests using Schiff Scale (SS). Expected results – Teeth restored with recently advanced restorative material Cention-N are anticipated to be more susceptible to microleakage and reduce postoperative dentinal hypersensitivity. Class V patients' postoperative sensitivity may be affected by the restorative material used and their method sensitivity. Conclusion – The overall microleakage in Cention N is expected to be minimal or absent after evaluating. Cention N will be superior mechanical properties in comparison to resin-modified glass ionomer cement (RMGIC). This substance will have a substitute restoring agent.
